本発明は血液を各成分に分離し、保存するための血液バッグ及び血液分離方法に関する。

【従来の技術】
プラスチック製のバッグに入れられた血液を遠心し、赤血球、白血球、血小板などの比重の差を利用して沈殿させ、各々の成分を取り出す血液成分の分離方法は、血液事業の分野において従来より行われていた。

従来血液成分分離に用いられる血液バッグは図5に示したように、プラスチック製の略長方形のバッグ本体1と、バッグ本体1に接続されたポート2と、同じくバッグ本体1と連通するよう接続された送液チューブ3、31、32により構成されている。また、送液チューブ31、32の末端には、分離した血液成分(血漿成分、白血球成分)を貯留するための子バッグ(不図示)がそれぞれ接続されてなる。

血液分離方法としては、採取した血液は送液チューブ3を通過してバッグ本体1に貯留された後、遠心され、図5に示したような赤血球層Aと、血漿層Bと、血小板を含んだ白血球層Cとに分離する。そこで、バッグ本体1を加圧し、血漿層B及び白血球層Cを、送液チューブ31あるいは32を通じて各送液チューブ末端に接続された各子バッグに移送して各血液成分の分離を行う方法が一般的であった。

本発明は上記事情について鑑み、他の血液成分を混入することなく、純粋な血液成分を得ることが容易に可能となり、バッグ形状が簡単でその製造工程が簡素化することが可能な血液成分分離用血液バッグと血液成分分離方法を提供する。

上述の課題を解消するため、本発明にかかる血液成分分離用血液バッグは、血液を貯留するためのバッグ本体と、血液をバッグ本体内に移送するための送液チューブが接続されてなる血液成分分離用血液バッグであって、バッグ本体が第1バッグ部と第2バッグ部と第3バッグ部からなり、各バッグ部には内部の血液を送出するためのポートがそれぞれ設けられてなり、第1バッグ部と第2バッグ部はバッグ本体の両端部に、第3バッグ部はその中間部に位置し、第3バッグ部の幅の長さが第1バッグ部及び第2バッグ部の幅の長さより短い関係であることを特徴とする血液成分分離用血液バッグであって、

【発明の実施の形態】
以下に添付図面を参照して、本発明である血液成分分離用血液バッグの詳細について説明する。まず、図1を参照して、概略的な構成について説明する。図1は本発明にかかる血液成分分離用血液バッグの概略図である。本発明にかかる血液成分分離用血液バッグは、全血を貯留するバッグ本体1と、採血した血液をバッグ本体1に移送するようバッグ本体1と接続された送液チューブ3より構成される。バッグ本体1は第1バッグ部11、第2バッグ部12、第3バッグ部13よりなり、第1バッグ部11と第2バッグ部12はそれぞれバッグの両端部に位置しており、第3バッグ部13は第1バッグ部11と第2バッグ部12の中間部に位置している。また、図1に示したように、第3バッグ部13の幅は第1バッグ部11及び第2バッグ部12の幅よりも短くなるような関係で構成されており、バッグ本体1の血液が貯留される箇所は略ダンベル型の形状に形成されている。また、各バッグ部には血液成分分離後にその内容物を取り出すためのポート21、22、23が各バッグ部に設けられている。

上記構成によれば、全血を貯留したバッグ本体1を血液成分分離処理後に第1バッグ部11と第3バッグ部13の境界部、第3バッグ部13と第2バッグ部12の境界部(いずれも図1中の点線で示した2箇所)をシールした後、各バッグ部を切り離すことができる。これにより、他の血液成分が混入することなく純粋な各血液成分に分離することが可能となる。

また、本発明にかかる血液成分分離用血液バッグにおいて、第3バッグ部に予め設けられたシール部を有する構成とするのが好ましい。図6が示すように、第3バッグ部13の中央部には第1バッグ部11と第2バッグ部12の方向にシール部5が複数設けられ、第3バッグ部13を複数の連通する区画となるよう構成されている。これはバッグの幅をより短くすることで、各区画の貯留可能な容量は少なくなるために、結果として、白血球成分の層をより厚くさせて存在させることが可能となる。これにより白血球層を取り扱うのがより容易となり、他の血液成分との分離操作がより容易に可能となる。

また、これらのシール部5は、血液の分離工程において血液成分の沈降を妨げないような形状であるのが望ましい。図6に示したようなバッグの縦方向、即ち第1バッグ部11と第2バッグ部12の方向に、シール部5が少なくとも1つ設けられているのが望ましい。

血液成分分離後、バッグ本体1の外側よりシールされるシール部分4のシール方法については、公知であるいずれの技術を採用しても構わない。具体的なシール方法は、バッグ本体1の材質によって採用する方法が異なる。材質がポリエチレンやポリプロピレン等のような場合は、熱シール方法を採用するのが好ましく、材質がポリ塩化ビニルや酢酸ビニル等のような場合は、高周波ウェルダーによるシール方法を採用するのが好ましく、それぞれの材質のシール特性に応じたシール方法を採用することが好ましい。

一方、シール後、シール部4より各バッグ部を切り離すことがあるため、切り離し操作が容易となるよう、シール部4の中心部に薄肉部分を形成する等の処置が施されていれば好適である。また、シール器具に切り離す工程を行うことが可能な手段を設けて、実質的にシール工程と同時にシール器具により各バッグ部を切り離す工程を行っても構わない。

尚、従来の血液成分分離用血液バッグにおいては、血液を劣化させないような血液保存液が予め充填されているが、本発明にかかる血液成分分離用血液バッグにおいても、血液を貯留する前に予め一般的に用いられる血液保存液がバッグ本体1中に充填されていることが好ましい態様である。

各バッグ部における貯留容量については、バッグ本体1に貯留する全血の容量より算出される各血液成分の容量となるよう各バッグ部を形成すればよく、バッグ本体全体に貯留する全体の血液量に応じて、適宜バッグ形状(各バッグ部の幅、第1、第2バッグ部の両端部から第3バッグ部の両端部への傾斜角度等)を形成すればよい。

また、本発明の実施形態においてバッグ本体1全体の形状を通常の血液バッグと同様の略長方形にし、バッグ本体1の血液貯留部分を略ダンベル形状となるように形成した実施形態を示した。しかしながら、別の実施形態としてバッグ本体1全体の形状を略ダンベル形状となるように形成しても構わない(不図示)。

次に、図2から図4に本発明における血液成分分離用血液バッグの血液成分分離工程の詳細について示した。

採血した血液を送液チューブ3を通じてバッグ本体1に貯留した後、血液成分分離工程(通常遠心分離による血液成分分離工程)を経て、図2に示したようにバッグ本体1内にて各血液成分、即ち赤血球成分、白血球成分、血漿成分が比重の差により分離し、各層を形成する。この時、最下部に位置する第1バッグ部11には赤血球層が、最上部に位置する第2バッグ部12には血漿層が、そして中間部に位置する第3バッグ部13には白血球層がそれぞれ形成される。

通常血液成分としての白血球は他の血液成分に比して、全血に占める割合が少なく、図5に示した従来の血液バッグにおいては、バッグ本体1内でごく薄い層として形成されることとなる。しかしながら、本発明にかかる血液成分分離用血液バッグにおいては最下部に位置する第1バッグ部11の幅よりも中間部に位置する第3バッグ部13の幅が短い関係にある。同様に最上部に位置する第2バッグ部12の幅よりも中間部に位置する第3バッグ部13の幅が短い関係にあり、バッグ本体1の形状は中間部がくびれた略ダンベル型の形状をなしている。これによれば容量の少ない白血球成分であっても、バッグ本体1内で比較的厚い層として存在させることが可能となる。

これにより従来バッグ本体1内で薄い層として存在していた白血球層を取り扱うのが比較的容易となり、他の血液成分との分離操作が容易に可能となる。

次に図3に示したように、第1バッグ部11と第3バッグ部13との境界部、第2バッグ部12と第3バッグ部13との境界部にそれぞれ位置するシール部4をバッグ本体1の外側より、別のシール器具(不図示)によりシールする。これらのシール部4はそれぞれ赤血球層と白血球層、白血球層と血漿層との境界部と一致する。そのためシール器具によりバッグ本体1の外側よりシールする際、目視にてその境界部を確認しながら、他の血液成分が混入しないようにシールし、各血液成分を分離することが容易に可能となる。このシールの工程により、各バッグ部はそれぞれ連通することのない独立した区域を形成することとなる。

これにより、バッグ本体内1で分離した各血液成分は、一つのバッグ内で層構造として貯留されているのではなく、それぞれシール部4により物理的に遮断されて独立したバッグ部に貯留されることとなり、各血液成分が混合されることがなくなる。

さらに、図4に示したようにシールの工程により形成されたシール部4より各バッグ部を切り離して、各バッグ部をそれぞれ別のバッグ体とする。各血液成分がそれぞれ別のバッグ体に貯留されていることにより、各バッグ体をそれぞれに応じて使用あるいは保存したりすることが可能となる。

【発明の効果】
本発明の血液成分分離用血液バッグによれば、血液成分を分離した際に白血球層に位置する第2バッグ部が他のバッグ部の幅より短く形成することにより、白血球層を比較的厚い層にて存在させることが可能となる。これによって、白血球層の存在する第3バッグ部の操作が比較的容易となる。また、各バッグ部との境界部をバッグ本体1の外側からシールし、各バッグ部内をお互いに連通しないように独立させることで、各血液成分同士が混入することがなくなり、純粋な血液成分を得ることが可能となる。さらには該シール部において切り離すことにより、各成分を別のバッグ体として、その血液成分に応じて使用することが可能となる。また、上記構成のような簡単な形状のバッグとすることによりバッグの製造工程を簡素化することが可能となる効果を奏する。
