Claims (3)
- 担体上の複数の領域に固定されたオリゴヌクレオチドプローブに標識物質で標識されたポリヌクレオチドをハイブリダイズさせた後、前記オリゴヌクレオチドプローブと二本鎖を形成していない1本鎖部分の標識ポリヌクレオチドを、1本鎖のDNAに末端から特異的に作用して1本鎖ポリヌクレオチドを分解することができる第1の制限酵素を作用させて、前記オリゴヌクレオチドプローブと前記標識ポリヌクレオチドとで形成された二本鎖から切り離し、その後、標識ポリヌクレオチドの標識物質からの信号を前記領域毎に検出して第1検出データを得、該第1検出データを得た後、単鎖のRNAまたはDNAを5′−モノヌクレオチドに分解することができる第2の制限酵素を作用させて前記オリゴヌクレオチドプローブと前記標識ポリヌクレオチドがミスマッチしている部分を切り離し、その後、標識ポリヌクレオチドの標識物質からの信号を前記領域毎に検出して第2検出データを得、前記第1検出データと前記第2検出データとを比較してミスマッチポリヌクレオチドが結合していた領域を特定することを特徴とするミスマッチ領域検出方法。
- 前記第1の制限酵素がエキソヌクレアーゼVIIであることを特徴とする請求項1記載のミスマッチ領域検出方法。
- 前記第2の制限酵素がS1ヌクレアーゼおよび/またはMung Bean ヌクレアーゼであることを特徴とする請求項1または2記載のミスマッチ領域検出方法。
