Description
a)ピペットが適切ではない、
b)転移が水滴を経由するとき、液体密度、ターゲット領域の動作、転移されるべき液体の速度などが、液体がディスク表面に命中するように複雑に決定される、
c)遠心力は、ターゲット領域に転移された液体をターゲット領域の外へ動かしディスクを置き去りにする傾向がある、
d)その他、
などである。
第1の目的は、
(a)ターゲット領域を分配装置に物理的に相互接続する必要、
(b)ディスクの静止の間に、液体がロードされるターゲット領域からの差動の蒸発により生じる変化、
これら(a)(b)無しに、ディスク形状のマイクロシステムの個別のターゲット領域に液体水滴を分配できる、器具セットアップ(構成)及び方法を提供することである。

本発明の第1の実施形態は、表面にターゲット領域(TA1)を含むディスクの形態のマイクロシステムに液体を分配するための方法である。ディスクは、構造のターゲット領域(TA1)であり入口ポートを伴うマイクロチャネル構造を含むマイクロ流体ディスクであるのが好ましい。上記方法は、以下のステップ(i)−(iii)を含むことを特徴とする。
(1)一つ又はそれ以上の分離したターゲット領域と、ディスクの他のあらゆる部分の角位置が定義される起点となるトリガマークとを有する、本明細書の他の箇所で定義されるディスクと、
(2)本明細書で記述される斬新なディスペンサ構成とを提供する。
“マイクロシステム”という用語は、水滴分配のためのターゲット領域がディスク表面に存在するシステムを含む。上記用語は、マイクロ流体システムと、ターゲット領域がどの流体システムの一部でもないシステムとを含む。
上記構成は、本発明の第2の実施形態である。
ディスペンサ構成は、
a)ディスク(103)を軸周りに回転するスピナ(113)と、
b)ディスペンサオリフィス(108)を経由してターゲット領域(TA1、TA2、TA3など)に水滴を分配し得る水滴ディスペンサ(110)と、
c)検出部に伴う固定のトリガ位置(105)と、
d)トリガ位置(105)を通過するトリガマーク(104)の機能として分配信号によりターゲット領域(TA)の中に水滴を分配することを始め得るコントローラ(114)を含む。
a)スピナ
スピナ(113)は、モータ(115)と、ディスクを軸周りに回転するシャフト(117)を伴うディスクホルダ(116)を含む。エンコーダ(118)はシャフトに連結され、例えば、20000以上の階調や30000以上の階調のように、10000以上の階調に、回転を小部分に分ける。他方で、エンコーダはディスクと結合してもよい。
水滴ディスペンサ(110)は、制御可能な周波数で、制御可能な体積と速度(m/s)で、ディスペンサオリフィス(108)を経由して、水滴をターゲット領域(TA)に分配することができる。
トリガ位置(105)は、トリガマーク(104)がトリガ位置を通過する毎にディスク(103)上のトリガマーク(104)を検出し得る検出器を含む。トリガマークが通過すると、通常はコントローラにより、分配信号が開始され得る。通過と実際の分配との間の所与の遅延(Telec)のためにシステムが提示されてもよい。この遅延は調整可能となり得る。
コントローラ(114)は、例えば、オペレータのインタフェースとソフトウエアを伴う電子工学のプログラム可能な制御のための手段であり、ここではこれ以上は説明しない。コントローラは、構成の範囲内の独立の物理的部位でもよく、及び/若しくは、信号を送受信して通信するユニットと物理的に結合する部位を有していてもよい。コントローラ(114)は、スピナ(113)、水滴ディスペンサ(110)、及び/又は、例えばトリガ位置(105)の検出器と結合する。
ステップ(ii)対して、プログラムされるべき主要な分配のパラメータは、上述の式により定義され、更に/又は、液体の物理化学特性に依存する。これらパラメータは、以下のものを含む。
(a)ディスクの回転の速度(角速度ω)、
(b)分配が発生すべき回転、及び/又は、ターゲット領域(TA)への水滴の分配の周波数f’、
(c)例えば大きさなどの、分配信号の形状、及び/又は分配パルスの周波数f、
(d)トリガ位置からの信号と水滴の実際の分配との間の遅延Telec、
(e)ディスペンサオリフィスとディスクとの間の距離h、
(f)放射動作、及び/又は、ディスペンサオリフィスの放射位置。
パラメータ(a)−(f)の値は、予め選択されたターゲット領域(TA)に水滴を分配するために、選択される。
生化学、化学、生物物理学、微生物学、医学、動物学、分子生物学などを含む化学及び生物学フィールドの範囲内で、種々の微細化された化学及び生物学の実験、即ちアッセイプロトコル、合成プロトコル、細胞培養プロトコルなどを実施するために、個々のマイクロチャネル及び/又はターゲット領域で液体が処理される。処理は、種々の化学反応及び/又は生物化学反応及び/又は生物学反応などが発生する、ということを含む。通常のプロトコルは、互いに相互の親和性を有する反応体の間の特有の反応を利用する。そして以下のものが形成される。
(a)検出及び/又は反応マイクロキャビティ内で固体相に固定される親和性複合体の形成、又は、
(b)検出マイクロキャビティ内の溶解性若しくは非溶解性の一つ又は複数の反応生成物。
通常の検出原理は、例えば、対応する属性を与えるグループ、若しくはこれらのグループの一つに変容され得るグループを提示する反応体を利用することにより、放射能、蛍光発光、化学ルミネセンス、生物ルミネセンス、酵素活性、クロモゲン、光散乱(タービジメトリ)などを基にする。
利用されるマイクロ流体ディスクは注入モールディングにより透明プラスチック部材で製造され、“マイクロ流体ディスク及び実行される処理”の項で示されたようにふたで覆われた。
水滴がマイクロ流体装置内で集められなかったこと以外は、実験セットアップは図1のものである。水性のチバクロンダイ(ブリリアントレッド4B−E、チバ)による比色定量分析が、異なる流速(0.1及び0.5ml/分)の(水から、1、2又は3分以上利用された70%のチバクロン溶液までの)勾配を調査するため、利用された。500水滴を所定の間隔で収集し収集サンプルの色強度を計測することにより、処理がモニタされた。
図3はディスペンサから得られた4つの勾配を示す。示される結果は、0.3ml/分の流速で1分の勾配の再生を照明している。勾配が90秒後に得られ、流れ構成の内部で30秒のラグタイムを示唆することを、このグラフは示している。
水滴がマイクロ流体装置内で集められなかったこと以外は、実験セットアップは図1のものである。アセトニトリルの勾配(0−80%)若しくは塩の勾配250mM Tris−HCl pH8(0−1.5M NaCl)の、水滴速度v(m/秒)の変動が行われた。インクジェットプリントヘッドの評価のために開発されたコンピュータIRカメラシステム(Sydat Automation,スエーデン)を利用して、速度が計測された。
図4を参照されたい。分配された液体の物理的属性は、勾配プロファイルと共に変動し、調整不良を起こすような、水滴の速度への明確な影響を与える。しかしながら、この調整不良は種々のパラメータを調節することにより補償され得る。例えば、トリガ遅延又はディスク角速度(式2及び式3参照)が変更可能である。但し、角速度を調節するとフロー制御に影響し得る(図5も参照されたい)。
水滴がマイクロ流体装置内で集められなかったこと以外は、実験セットアップは図1のものである。水滴速度v(m/秒)が、通常の高塩分緩衝剤溶液のための、パルス大きさの関数として実施された。
パルス大きさを調整することにより、水滴速度を調節し、調整不良問題を解決することが可能になる。
(30及び35ナノリットル体積の)コラムが、ディスクマイクロ構造への遠心分離により、SOURCE 15RPC(Amersham Pharmacia Biotech、スエーデン)をパックされた。利用されたマイクロチャネル構造は図2で示される。ビーズ部材の懸濁液を入口ポート(210)の中に詰めることによって、パックは完了した。遠心分離において、ビーズは、Y形状構造の下方シャンク(2149のより深い部分に収集する。ペプチド(アンギオテンシンI II)は、Cy3又はCy5のラベルキット(Amersham Pharmacia Biotech、スエーデン)を利用して、ラベルされた。コラムは、2×500nl 50%アセトニトリル、0.1%TFAを塗布することでコンディションされ、入口ポート(203)を介して共通配布チャネル(202)を充填し各々の溶液につきスピンすることにより、制御されたスピン(1500rpm)条件の下で0.1%TFAの500nlで洗浄された。Cy5ラベルのアンギオテンションI(110nM−Sigma)とCy3ラベルのアンギオテンションII(880nM−Sigma)の500nlの混合物が、入口ポート(203)を介して共通配布チャネル(202)にロードされ、スピン速度が段階的に増加する。バインドされた成分は、12.5−37.5%アセトニトリルの間の段階的勾配で溶離された。アセトニトリルの濃度の増加に対応する部分は、ピペットにより共通配布チャネル(202)に充填され、ディスクの回転によりコラムを通された。アセトニトリル濃度は、それぞれの200nlの段階で2.5%増加し、制御されたスピンフローで付加された。コラム内の分離は、蛍光発光マイクロスコープを利用して、モニタされた。
ラベルされたペプチドとフリーダイの分離が、ディスク上の同時処理の16コラム上でうまく再生された(結果は示さない)。この結果は、ピペットによる溶液の分配とは関連しない。入口ポート(210)へ水滴を分配することにより本発明に係る分配も生じうることも想定できる。
実験セットアップは図1に従った。(34及び38ナノリットル体積の)コラムが、実験4で記したようにパックされコンディションされた。Cy3(700nM)/Cy5(300nM)混合物が、スピン速度の段階的な増加によりロードされた。連続勾配の溶離が、直径60μmのノズルと、幅1mm深さ50μmのフロースルーチャネルとを備えるディスペンサを利用して、なされる。勾配の分配は本発明に係る入口ポート(210)を介して為された。1kHzの周波数と2500〜1800rpmのスピンのディスペンサを利用して、種々の溶離プロファイルがテストされた。分離は、蛍光発光マイクロスコープによりコラム内でモニタされた。
1分以上勾配0−40%アセトニトリル、0.1%TFAを利用したCy3とCy5ダイの分離の開始を、結果は示した。他の実験は、水、アセトニトリル混合物、トリス緩衝剤などの種々の液体を利用して14のマイクロ構造の中にスピンして、勾配を分配する可能性を示した。
Claims (26)
- マイクロシステムを画定するディスク表面内に存在するターゲット領域TA1に液体を分配するための方法であって、
上記ディスクが、TA1である入口ポートの形態でターゲット領域を伴うマイクロチャネル構造を含むマイクロ流体ディスクであり、
i)(A)ターゲット領域が分離しており周辺にトリガマークが存在するディスクと、(B)ディスペンサ構成とを提供するステップであって、該ディスペンサ構成が、a)軸周りにディスクを回すスピナと、b)水滴を上記ターゲット領域TA1に分配できる水滴ディスペンサと、c)ディスク外部の固定されたトリガ位置と、d)分配信号により、トリガ位置を通過するトリガマークの関数としてTA1の中への水滴の分配をトリガできるコントローラとを含む、ステップと、
ii)ディスクをスピナ内に配置し、水滴の分配をTA1に与える分配パラメータのための値をコントローラにプログラムするステップと、
iii)ディスクがスピンする間に、水滴を分配するステップを含むことを特徴とする方法。 - (a)ディスク回転速度(各速度ω)と、
(b)分配が発生する回転、及び/又は、水滴をターゲット領域TA1に分配する周波数f’と、
(c)例えば大きさなどの分配信号の形状、及び/又は分配パルスの周波数fと、
(d)トリガ位置からの信号と水滴の実際の分配との間の遅延Telecと、
(e)ディスペンサオリフィスとディスクの間の距離hと、
(f)ディスペンサオリフィスの放射方向動作、及び/又は放射方向位置
から、上記パラメータが選択されることを特徴とする請求項1に記載の方法。 - 液体が、成分の少なくとも一つに関して勾配を含み、該勾配が連続の勾配か、又は、例えば一つ、二つ若しくはそれ以上の段階などの段階的勾配であることを特徴とする請求項1又は請求項2に記載の方法。
- パラメータ(a)、(b)、(c)及び(d)のうちの少なくとも一つのための値が、勾配により生ずる水滴速度の変化を補償するために分配の間に調整され、その調整はコントローラにより処理されることを特徴とする請求項3に記載の方法。
- ディスクが、マイクロ流体ディスクであり、
上記マイクロチャネル構造が、TA1である入口ポートと、TA1下流に配置され化学的又は生物学的実験を実行するのに利用されるマイクロキャビティとを、含むことを特徴とする請求項1から請求項4のうちのいずれか一つに記載の方法。 - 上記液体が、塩分濃度、塩の種類、pH、溶媒の構成、及び/又は、マイクロキャビティ内で実行される実験を干渉する他の要素における、変動として定義される勾配を含むことを特徴とする請求項1から請求項5のうちのいずれか一つに記載の方法。
- ビーズ形態、且つ/又は、モノサイズ(単分散)でもポリサイズ(多分散)であり、例えば、多孔モノリス、多孔粒子や非多孔粒子のパックされた床である多孔性床の形態の分離媒質を、マイクロキャビティが含むことを特徴とする請求項5又は請求項6に記載の方法。
- a)液体サンプル(液体1)を上記マイクロチャネル構造のサンプル入口ポートに分配するステップであって、サンプルが床を通過するときに床にバインドし得る少なくとも一つの物質を含むステップと、
b)分離媒質から上記物質の少なくとも一部をリリースするため、溶離剤(液体2)を上記マイクロチャネル構造の入口ポートに続いて分配するステップを含み、
・上記入口ポートの少なくとも一つがTA1であり、
・分配パラメータに対する上記のプログラムされた値を利用して、上記水滴ディスペンサを介して水滴として、液体1及び/又は液体2がTA1に分配されることを特徴とする請求項7に記載の方法。 - 溶離剤が成分の一つに関して勾配を含み、分配パラメータに対する上記のプログラムされた値を利用して上記水滴ディスペンサを介して水滴としてTA1に分配されることを特徴とする請求項8に記載の方法。
- a)スピナが、回転毎に少なくとも10000階調を与えるエンコーダに繋がり、
b)分配信号が与えられるタイミングが、トリガマークとトリガ位置との間のエンコーダ階調数により決定されることを特徴とする請求項1から請求項9のうちのいずれか一つに記載の方法。 - 分配信号が与えられるタイミングが、回転速度(角速度)と、トリガマークがトリガ位置を通過するタイミングとから計算されることを特徴とする請求項1から請求項9のうちのいずれか一つに記載の方法。
- 分配信号に従い動作される圧電駆動アクチュエータが水滴ディスペンサを駆動することを特徴とする請求項1から請求項11のうちのいずれか一つに記載の方法。
- 水滴ディスペンサがフロースルーディスペンサであることを特徴とする請求項1から請求項12のうちのいずれか一つに記載の方法。
- ディスクが、ディスク中心から同じ半径距離にある2つ又はそれ以上の離散ターゲット領域TA1、TA2、TA3を含み、コントローラが、これらターゲット領域に水滴の分配を与える分配パラメータに対する値をプログラムされていることを特徴とする請求項1から請求項13のうちのいずれか一つに記載の方法。
- ディスクが2つ又はそれ以上のマイクロチャネル構造I、II、IIIを含むマイクロ流体ディスクであり、マイクロチャネル構造の各々は、構造Iに対してTA1、構造IIに対してTA2、構造IIIに対してTA3の入口ポートを有することを特徴とする請求項14に記載の方法。
- 相互に隣接して位置するターゲット領域TA1、TA2、TA3の間の角距離が同じ若しくは異なることを特徴とする請求項14又は請求項15に記載の方法。
- 分配信号の形状が、各々が水滴の出射を生じるパルスの波数を含むようにプログラムされ、残りのパラメータ(a)−(f)のための値が、分配信号につきターゲット領域の中に回転当たりせいぜい多くとも1水滴を、好ましくはせいぜい多くとも1水滴を上記ターゲット領域TA1、TA2、TA3の各々に分配するようにプログラムされていることを特徴とする請求項14から請求項16のうちのいずれか一つに記載の方法。
- 上記水滴ディスペンサが、コントローラのコントロール下にある水滴ディスペンサのアレイの一部であることを特徴とする請求項1から請求項17のうちのいずれか一つに記載の方法。
- 表面に一つ又は複数の離散ターゲット領域TA1、TA2、TA3を含むディスクの形態のマイクロシステムの一部である上記離散ターゲット領域に液体を分配する装置であって、
上記ディスクは一つ又はそれ以上のマイクロチャネル構造I、II、IIIを含むマイクロ流体ディスクであり、そのマイクロチャネル構造の各々は、構造Iに対してTA1、構造IIに対してTA2、構造IIIに対してTA3の入口ポートを有する、装置において、
a)軸周りにディスクを回すスピナと、
b)水滴をTA1、TA2、TA3に分配できる水滴ディスペンサと、
c)ディスク外部に配置され、ディスクがスピナ内に配置され回転するときにトリガ位置を通過するトリガマークを検出し得る検出器を含む、固定されたトリガ位置と、
d)分配信号により、トリガ位置を通過するトリガマークの関数としてTA1、TA2、TA3の中への水滴の分配をトリガできるコントローラとを含むことを特徴とする装置。 - a)スピナが、回転毎に少なくとも10000階調を与えるエンコーダに繋がり、
b)分配信号が水滴ディスペンサに送られるタイミングが、トリガマークとトリガ位置との間のエンコーダ階調数により決定されることを特徴とする請求項19に記載の装置。 - 上記の一つ又は複数の分配信号が水滴ディスペンサに送られるタイミングが、回転速度(角速度)と、トリガマークがトリガ位置を通過するタイミングとから計算され得ることを特徴とする請求項19に記載の装置。
- 分配信号に従い動作される圧電駆動アクチュエータがディスペンサを駆動することを特徴とする請求項19から請求項21のうちのいずれか一つに記載の装置。
- ディスペンサがフロースルーディスペンサであることを特徴とする請求項19から請求項22のうちのいずれか一つに記載の装置。
- 水滴ディスペンサが、コントローラのコントロール下にある水滴ディスペンサのアレイの一部であることを特徴とする請求項19から請求項23のうちのいずれか一つに記載の装置。
- コントローラが、
a)各々が水滴ディスペンサから一つ又はそれ以上の水滴の出射を生じるパルスの波数を含む分配信号を送ることができ、
b)分配信号につきターゲット領域の中に回転当たりせいぜい多くとも1水滴を、好ましくは回転当たりせいぜい多くとも1水滴を上記ターゲット領域TA1、TA2、TA3の各々に分配する分配パラメータのための値をプログラムされていることを特徴とする請求項19から請求項23のうちのいずれか一つに記載の装置。 - 水滴ディスペンサが、装置内に配置されたディスクに平行な平面にて、側方に移動し得ることを特徴とする請求項19から請求項25のうちのいずれか一つに記載の装置。
