Description
制御や種々の細胞機能の調節に重要な役割を果たしていることが明らかにされている(非特許文献1〜3)。CaMKIIの大きな特徴として、1)脳に多量に存在する。2)可溶型と顆粒結合型がある。3)基質特異性が広い。4)臓器特異性アイソフォームが存在する。5)自己リン酸化によって活性を制御する、などの際だった特徴を備えている。特にCaMKIIは脳神経系では、神経伝達物質の生合成、神経伝達物質の放出、記憶の素過程と考えられている長期増強作用、記憶特に空間記憶、などに関与していることが示唆されている。
メイン (catalytic domain)、自己阻害ドメイン (autoinhibitory domain, AID)、Ca2+/calmodulin結合ドメイン (Ca2+/calmodulin-binding domain, CaM)、variable region (V)、subunit association region (Association)からなっていることが判明している(図1
)。CaMKIIの各サブユニットの分子量は約50-60kDaであり、subunit association region
が互いに相互作用し、8-12の複数のサブユニットからなるオリゴマー構造をとっている
。またCa2+/calmodulinが存在しない場合、自己阻害ドメインと触媒ドメインが相互作用
し、不活性な状態をとっている(非特許文献4)。一方、Ca2+/calmodulin存在下では、Ca2+/calmodulinがCa2+/calmodulin結合ドメインに結合して、CaMKIIのコンフォメーション変化を誘導し、自己阻害ドメインと触媒ドメインの相互作用を分断する結果、CaMKIIのリン酸化酵素の活性(Ca2+/calmodulin依存性活性)を誘導する。CaMKIIの活性化が起こる
と自己阻害ドメイン内にあるThr286 /Thr287(αサブユニットの場合Thr286、 β・γサブユニットの場合Thr287)の自己リン酸化が速やかに起こり、Ca2+/calmodulin非依存性の持続的な活性(Ca2+/calmodulin非依存性活性)が出現する。
パク質をCa2+/calmodulin非依存的にリン酸化できるので、CaMKIIの基質特異性、ターゲ
ットとなる基質タンパク質の解析、CaMKIIの生理機能の解明、リン酸化したタンパク質の調製などを行うために、活性化などの面倒な操作の要らない、非常に有用なリン酸化試薬として利用されている(非特許文献6) 。しかしながらこれまで30k-CaMKIIを調製する方法として、ラットなどの脳から精製したCaMKIIをタンパク質分解酵素で消化して得る方法や、CaMKIIの豊富に含まれるシナプス後肥厚(PSD)画分をタンパク質分解酵素で消化して
得る方法が使われてきたが、操作が煩雑で大量に得ることは難しかった(非特許文献5, 7)。そこで大腸菌の発現系を用いた30k-CaMKIIの調製法を検討することにした。

項1. カルモジュリン依存性リン酸化酵素IIの触媒ドメインのN末端側に、宿主で大量
発現可能なアミノ酸領域を有する、カルモジュリン非依存性キナーゼ活性を有するポリペプチド。
項2. 前記アミノ酸領域が、宿主で発現可能なタンパク質のN末端側のアミノ酸配列を含む項1に記載のポリペプチド
項3. 前記アミノ酸領域が、アフリカツメガエルCaMKIxのN末端側から10個以上のアミノ酸配列を含む項1又は2に記載のポリペプチド。

項5. 前記領域が、アフリカツメガエルCaMKIxのN末端の少なくとも10個のアミノ酸をコードするポリヌクレオチドを含む、項4に記載のDNA。
項6. カルモジュリン依存性リン酸化酵素IIの触媒ドメインをコードするDNAを発現可能に含む、発現ベクター。
項7. 項6に記載の発現ベクターを細菌に導入してなる形質転換体。
項8. 項7に記載の形質転換体を培地中で培養する工程、
培養液の菌体を破砕処理し、不溶性画分を回収する工程;
得られた不溶性画分の蛋白質を可溶化する工程、
可溶化された蛋白質を再生溶液中で処理して変性されたカルモジュリン非依存性キナーゼの不活性型を活性型に変換する工程、
を有するカルモジュリン非依存性キナーゼ活性を有する断片の製造方法。
項9. 可溶化を6Mグアニジン塩酸塩又は8M尿素、50mMトリス塩酸(pH7.5)、10 mM β-メルカプトエタノールを含む可溶化溶液中で行う、項8に記載の方法:
項10. 再生を50 mM トリス塩酸(pH 7.5)、0.05% Tween 40、150 mM NaCl、10% ethylene glycol、1 mM DTTを含む再生溶液中で行う、項8又は9に記載の方法。
以下、本発明をより詳細に説明する。
(1)大腸菌等の宿主で発現させた場合に、活性を有するカルモジュリン非依存性キナーゼ断片が発現されること
これは、カルモジュリン依存性リン酸化酵素IIの触媒ドメインのN末端側に、宿主で大量発現可能なアミノ酸領域を導入することで達成することができる。
(2)不溶性画分に発現された不活性なカルモジュリン非依存性キナーゼ断片を可溶化および再生溶液を使用して活性型に導くこと
これは、例えば可溶化溶液および再生溶液として以下の溶液を使用することにより達成できる。
・可溶化溶液(6Mグアニジン塩酸塩又は8M尿素、50mMトリス塩酸(pH7.5)、10 mM β-メルカプトエタノール)
・再生溶液(50 mM トリス塩酸(pH 7.5)、0.05% Tween 40、150 mM NaCl、10% ethyle
ne glycol、1 mM DTT)
本明細書において、「N末端側から」とは、「開始メチオニンから」と同義である。
少なくしかもすべて不溶性画分に検出された(図2、3)。

高い30k-CaMKIIは得られるが、量的にはごくわずかに過ぎなかった。
端40アミノ酸に相当するcDNA部分)をつないで、CX-30k-CaMKIIの発現ベクターを構築し
た(図4)。CaMKIxは、アフリカツメガエルcDNAライブラリー中から見出した新規リン酸
化酵素であり、分子量は約38000、rat CaMKIと75%のホモロジーをもつタンパク質で、大
腸菌での大量発現が確認されている酵素である(CaMKIx, Acc. No. AB098710, Xenopus laevis 由来 Ca2+/calmodulin-dependent protein kinase I-like protein)。
きたが、ほとんど不溶性画分として得られた(図3)。そこでCX-30k-CaMKIIを完全にグアニジン塩酸又は8M尿素で変性した後に、酵素活性を再生する条件を検討した。透析処理では酵素活性の回復が見られなかったのに対し、DTT, Tween40, NaClを含む再生溶液で希釈
し、0℃で静置すると時間依存的に顕著な活性回復が見られた。こうして得られた活性化CX-30k-CaMKIIを用いて、さまざまな基質タンパク質をリン酸化できることが確認された。従って、大腸菌を用いた30k-CaMKIIの大量発現と可溶化/再活性化は、タンパク質リン酸化用の酵素試薬の簡便な調製法として有用である。
上流に連結した例である。該領域としては、アフリカツメガエルCaMKIxの5'配列(N末端
1個以上、好ましくは5個以上、より好ましくは10個以上のアミノ酸配列に相当するcDNA部分)の他に、大腸菌中で大量発現可能なタンパク質の1個以上好ましくは5個以上、より好ましくは10個以上のアミノ酸配列、特にN末端側のアミノ酸部分配列に相当するcDNA部分が挙げられる。この領域には、大腸菌中で大量発現可能なタンパク質の全配列を用いてもよい。
・ホモジナイズ溶液(20 mM トリス塩酸(pH 7.5), 150 mM NaCl, 0.05% Tween 40)
・可溶化溶液(6Mグアニジン塩酸塩、50mMトリス塩酸(pH7.5)、10 mM β-メルカプトエタノール)
・再生溶液(50 mM トリス塩酸(pH 7.5)、0.05% Tween 40、150 mM NaCl、10% ethylene glycol、1 mM DTT)
本発明のポリペプチドは、GSTタグ、カルボキシ末端タグ、Hisタグ、多重荷電ペプチド、グルタチオンS−トランスフェラーゼ(GSTタグ)、抗体のFc部分、免疫グ
ロブリン結合ドメイン、プロテインA若しくはプロテインG若しくはこれらの一部、レシチン、核酸結合部位、ヘパリン結合部位、特異性リガンド、特異性受容体、インテグリン、サイトカイン又はそれらの受容体結合ドメインなどを触媒ドメインのC末端側にさらに結合してもよい。
<実施例1>発現ベクター(pET-30k-CaMKIIγ)の構築
まず、EST database中からラットCaMKIIγの全塩基配列(Acc. No. S71571)を見出し、こ
の配列をもとに以下の2本のプライマーを合成した。
UrCaMKIIγ/Nhe 5'-AAAGCTAGCATGGAGACCACCGCCACCTG-3' (下線部 NheI site)
LrCaMKIIγ30/Xho 5'-TTTCTCGAGGACCCATGGGTGCTTGAGAGC-3' (下線部 XhoI site)
次に、5'-RACE Ready cDNA library (Rat brain)(クロンテック社製)を鋳型にしてPCR (ABI社製 GeneAmp PCR System 2700を使用)を行った。PCRにはタカラバイオ社製のPyrobest
DNA polymeraseを用い、プログラムは96℃ 2 min, (96℃ 10 sec, 68℃ 2 min) x 30, 68℃ 7 minで増幅した。増幅産物を電気泳動後、ゲルから精製し(フナコシ社製 Gene Clean III kitを使用)、NheIおよびXhoIで消化した。引き続き、pET23a(Novagen社製)のNheI/XhoI siteに組み込んで発現ベクターpET-30k-CaMKIIγを構築した(図2)。
<実施例2>発現条件の検討1
発現ベクターpET-30k-CaMKIIγを大腸菌BL21(DE3)(Novagen社製)に電気穿孔法にて導入し(Harvard Apparatus社、BTX ECM-600を使用)、100 mg/mlのアンピシリンを含むLB寒天
培地で37℃、16 h培養した。生えてきたコロニーを3 mlのLB液体培地(100 mg/mlアンピ
シリン)に植菌し、37℃で16 h振とう培養した。次に、培養液の一部を新鮮な5 mlのLB液体培地(100 mg/mlアンピシリン)に植菌し、37℃で16h振とう培養した。培養後の菌体をSDS-PAGEサンプル溶液(63 mM トリス塩酸(pH 6.8), 2% SDS, 10% glycerol, 1% β-メルカプトエタノール, 0.001% bromophenol blue)中で超音波破砕し(Branson社のBranson sonifier 450を使用)、SDS-PAGEにて発現の有無を確認したところ、わずかに30k-CaMKIIγの発現が認められた(図3)。培養条件及び大腸菌株をいろいろ変えても発現量は改善さ
れなかった。
<実施例3>発現ベクター(pET-CX-30k-CaMKIIγ)の構築
上記実施例2で30k-CaMKIIγをそのままの状態で発現させても十分な発現量が得られなか
ったのは、ラットCaMKIIの開始メチオニン付近の配列がコドン使用頻度の問題で大腸菌に認識されにくくなっているためと考えられる。これを解決するため、既に大腸菌にて可溶性画分に大量発現することが確認されているアフリカツメガエルCaMKIxのN末端40アミノ
酸に相当する配列(配列番号7)を30k-CaMKIIγのN末端に挿入し、発現ベクターpET-CX-30k-CaMKIIgを構築した。新たに合成したプライマーUrCaMKIIγ/EcoRIと実施例1で示したプライマーLrCaMKIIγ30/Xhoを用い、5'-RACE Ready cDNA library (Rat brain)(クロンテック社製)を鋳型にしてPCRを行った。

PCRにはタカラバイオ社製のPyrobest DNA polymeraseを用い、プログラムは96℃ 2 min, (96℃ 10 sec, 68℃ 2 min) x 30, 68℃ 7 minで増幅した。増幅産物を電気泳動後、ゲルから精製し、EcoRIおよびXhoIで消化した。一方、pET-CaMKIxも同様にEcoRIおよびXhoIで消化することでCaMKIxのN末端40アミノ酸以外の部分を除去しておき、代わりに前述のPCR増幅産物を組み込むことで、発現ベクターpET-CX-30k-CaMKIIγを構築した(図4)。

<実施例4>発現条件の検討3
発現ベクターpET-CX-30k-CaMKIIγを大腸菌BL21(DE3), BL21(DE3) pLysS, BL21(DE3) pLysE(Novagen社製)に電気穿孔法にて導入し、100 mg/mlのアンピシリンを含むLB寒天培地で37℃、16 h培養した。生えてきたコロニーを3 mlのLB液体培地(100 mg/mlアンピシリ
ン)に植菌し、37℃で16 h振とう培養した。次に、培養液の一部を新鮮な5 mlのLB液体培地(100 mg/mlアンピシリン)に植菌し、0.1 mMのIPTG存在・非存在下で37℃(or 25℃)で16 h振とう培養した。培養後の菌体をSDS-PAGEサンプル溶液中で超音波破砕し、SDS-PAGEにて発現の有無を確認した。BL21(DE3)で発現させた場合、37℃で培養するとIPTGの有
無に関わらず大量に発現したが、25℃では菌の生育が悪く、発現量も非常に少なかった。一方BL21(DE3) pLysS及びBL21(DE3) pLysEで発現させると、いずれの条件においても全く発現が認められなかった。最終的にBL21(DE3)を宿主とし、IPTG非存在下で37℃、24時間
培養するのが最も発現に適していた(図3)。
<実施例5>CX-30k-CaMKIIγの大量発現
発現ベクターpET-CX-30k-CaMKIIγを導入した大腸菌BL21(DE3)を3 mlのLB液体培地(100 mg/mlアンピシリン)に植菌し、37℃で16 h振とう培養した。次に、培養液の一部を新鮮
な100 mlのLB液体培地(100 mg/mlアンピシリン)に植菌し、37℃で24 h振とう培養した
。培養液を8,000 rpmで10 min遠心分離することで菌体を回収し、10 mlのホモジナイズ溶液(20 mM トリス塩酸(pH 7.5), 150 mM NaCl, 0.05% Tween 40)に懸濁して超音波破砕し
た。遠心分離にて沈殿を回収し(20,000g, 10分, 2℃)、同様に2回超音波破砕を繰り返す
ことで沈殿を洗浄し、不溶性画分を回収した。
<実施例6>CX-30k-CaMKIIγの可溶化
不溶性CX-30k-CaMKIIγを大量に含む画分は、可溶化溶液(6Mグアニジン塩酸塩、50mMト
リス塩酸(pH7.5)、10 mM β-メルカプトエタノール)中で超音波処理をすることにより完全に可溶化された。この時の可溶化サンプルの蛋白量は3 mg/ml以上になるように調整
した。
<実施例7>CX-30k-CaMKIIγの再生と精製
可溶化したCX-30k-CaMKIIγは、そのままでは活性が見られないので、酵素の再生法を検
討した。変性剤であるグアニジン塩酸を除くために透析処理を行っても活性の回復は全く見られなかったが、再生溶液(50 mM トリス塩酸(pH 7.5)、0.05% Tween 40、150 mM N
aCl、10% ethylene glycol、1 mM DTT)により急激に希釈すると活性が徐々に回復した。再生溶液により200倍に希釈して、氷上ならびに16℃に静置し、一定時間ごとにSyntide-2(PLARTLSVAGLPGKK)を基質に用いてプロテインキナーゼ活性を測定した(図5)。
間あたりの[32P]リン酸の取り込み量をホスホセルロースペーパーを用いるRoskoskiの方
法(Roskoski, R. (1983) Assay of protein kinases. Methods in Enzymology 99, 3-6
)で測定することにより算出した。タンパク質リン酸化酵素の活性測定反応液の組成は以下の通りである。
16℃では、希釈後数時間で活性の回復がピークに達し、その後時間経過とともに活性の低下がみられた。これに対し、氷上での酵素再生処理では、24時間後までゆっくりと活性の回復が観察された(図5)。再生した酵素は、そのままでも使用可能であるが、酵素のC末端にhistidine-tagがついているので、HiTrap Chelating HPカラム(アマシャムバイオサイエンス社)を通して、容易に精製することが可能である。
<実施例8〜10>発現条件の検討2
実施例3のCaMKIxのN末端40アミノ酸に代えて、CaMKIxのN末端の10アミノ酸(配列番号8)および30アミノ酸(配列番号9)を30k-CaMKIIγのN末端に挿入した以外は、実施
例3〜7の操作を繰り返した。
用した場合には、比活性が高くなったが、キナーゼタンパク質の発現量は低下した。
<実施例11>活性型CX-30k-CaMKIIγを用いたタンパク質のリン酸化反応
再生した酵素が、native CaMKIIと同様にさまざまな基質タンパク質のリン酸化反応に
使えるかどうかを調べてみた。図6AはCa2+/calmodulin存在下でのCaMKIIによるリン酸化
反応を、図6BにはCX-30k-CaMKIIγによるリン酸化反応を示した。用いるタンパク質基質
は、ミエリン塩基性タンパク質(myelin basic protein, MBP)(Sigma-Aldrich社製)、ヒストン(histone)(Sigma-Aldrich社製)、ミオシン軽鎖(myosin light chain, MLC)(ニ
ワトリの砂嚢から精製)、シナプシンI(Synapsin I)(ウシ脳から精製)の4種のタンパク質を用いた。
以下の通りである。またCX-30k-CaMKIIγによるリン酸化反応の場合、0.2 mM CaCl2と1 mM calmodulinは反応液から除いた。
CaMKIIを用いた場合、いずれの場合にも50 kDaと60 kDa付近にCaMKIIの自己リン酸化のバンドが認められた(図6A)。一方、CX-30k-CaMKIIγを用いたリン酸化反応では、Ca2+/calmodulinが不要であり、CX-30k-CaMKIIγにはCaMKIIの自己阻害ドメイン及びそれより
C末端側の領域を欠いているので、50 kDaと60 kDa付近のCaMKIIの自己リン酸化由来のバンドが認められなかった(図6B)。CaMKII及びCX-30k-CaMKIIγを用いた場合いずれも、M
BP、ヒストン、MLC、シナプシンIのリン酸化によるバンド(MBPは分子質量18 kDa 、ヒストンは14-15 kDa 、MLCは15-20 kDa、シナプシンIは80-86 kDa)が認められた。つまり、CX-30k-CaMKIIγを用いたリン酸化反応はnative CaMKIIを用いた場合と同程度に基質のリン酸化が認められた。

- カルモジュリン依存性リン酸化酵素IIの触媒ドメインのN末端側に、アフリカツメガエルCaMKIxのN末端側10〜40個のアミノ酸配列を有し、
前記アフリカツメガエルCaMKIxのN末端側10〜40個のアミノ酸配列は、配列番号7で表されるアミノ酸配列中のものである、
カルモジュリン非依存性キナーゼ活性を有するポリペプチド。 - カルモジュリン依存性リン酸化酵素IIの触媒ドメインをコードするDNAの5'末端側に
アフリカツメガエル由来の、アフリカツメガエルCaMKIxのN末端側10〜40個のアミノ酸配列をコードする領域
又はアフリカツメガエルCaMKIxのN末端側10〜40個のアミノ酸配列をコードし、コドンユーセージが大腸菌での発現に適したコドンユーセージである領域
を有する、カルモジュリン非依存性キナーゼ活性を有する断片をコードするDNA。 - 前記領域が、アフリカツメガエルCaMKIxのN末端の少なくとも10個のアミノ酸をコードするポリヌクレオチドを含む、請求項2に記載のDNA。
- 請求項2又は3に記載のDNAを発現可能に含む、発現ベクター。
- 請求項4に記載の発現ベクターを大腸菌に導入してなる形質転換体。
