従来から、シミ、ソバカス等の予防又は治療を目的として美白用化粧料が提供、使用されてきた。 この美白用化粧料中の美白成分としては、ハイドロキノンおよびその配糖体、コウジ酸およびその誘導体、アスコルビン酸およびその誘導体、チオール系化合物の物質の他、種々の動植物抽出物等の物質が知られており、これらを配合することが提案されている。

しかし、従来より知られている美白成分には種々の問題点が残されており、美白用化粧料組成物の配合成分として十分満足の行くものではなかった。 例えばハイドロキノンは安全性に問題があり、ハイドロキノン配糖体、コウジ酸やその誘導体、アスコルビン酸やその誘導体等には、化粧料として使用するには極性が高すぎるという問題点があった。 さらに、グルタチオン、システイン等のチオール系化合物には、配合後の安定性に問題が残っている。 その他、現在知られている動植物抽出物、例えば胎盤エキス、アロエエキス等はその美白効果が満足出来るものではなかった。

【課題を解決するための手段】
本発明者等は、上記課題を解決すべく、種々の植物抽出物についてその美白作用を鋭意スクリーニングした結果、ウリ科スズメウリ属(Cucurbitaceae Melothria)植物の溶媒抽出物はメラニン生成を強く抑制すると共に細胞毒性が極めて弱いことを見出した。
そして、この抽出物を化粧料組成物に配合すれば、安全で効果の高い美白用化粧料組成物が得られることを見出し、本発明を完成した。

すなわち、本発明はウリ科スズメウリ属植物の溶媒抽出物を有効成分として含有する美白用化粧料組成物を提供するものである。

【発明の実施の形態】
本発明において用いられるウリ科スズメウリ属の植物の例としては、中国名で「バハクジ (馬▲はく▼児)」(Melothria indica)と呼ばれる植物や、同じく「ボウカ(芽瓜)」(Melothria heterophylla)と呼ばれる植物、あるいはスズメウリ(Melothria japonica)が挙げられる。
このバハクジやボウカは、中国で薬用にされる植物であり、特にそれらの塊根は熱性の咳、下痢、淋病、尿路感染、のどの痛みや湿疹に対して用いられる。 また、これら植物はインドでも薬用にされ、更に若芽や果実が食用にされるといわれている。

これらの植物からの有効成分の抽出は、その植物体、特にその塊根を使用し、適当な溶媒を用いて抽出すれば良い。 利用される溶媒としては、有効成分が効果的に抽出される溶媒であれば特に限定されるものではないが、親水性溶媒、例えば水またはメタノール、エタノール、プロパノール等の低級アルコールまたはそれらの混液が好ましく、特に安全性の面から、水もしくはエタノールまたはそれらの混液を用いるのが好ましい。

なお、有効成分を高濃度に配合するために、得られた抽出物を、更に濃縮、液液分配、吸着クロマトグラフィー等の手段により精製した後配合することも可能である。

本発明の美白用化粧料組成物は、上記のようにして得られたウリ科スズメウリ属植物の抽出物を、化粧料基剤中に配合することにより調製される。化粧料用基剤に対する抽出物の配合量は、抽出物として0.1〜10重量%(以下、単に「%」で示す)とすることが好ましく、更に1〜5%とすることがより好ましい。また、化粧料基剤としては、化粧料に一般に用いられている油性成分、界面活性剤、紫外線吸収剤、低級アルコール、防腐剤、殺菌剤、色剤、粉末、香料、水溶性高分子、緩衝剤などその他の成分が挙げられ、本発明の効果を損なわない範囲で適宜配合できる。

かくして得られる本発明の美白用化粧料組成物は、シミ、ソバカス等の予防又は治療に有効な皮膚用化粧料組成物として利用することができ、より具体的には医薬部外品としてのローション、乳液、クリーム、パック剤、石鹸等の薬用化粧品、および医薬品としてのローション、乳液、クリーム、軟膏等の皮膚外用剤として利用することが可能である。

次に、参考例、試験例及び実施例を挙げ、本発明を更に詳しく説明するが、本発明はこれら実施例等に何ら制約されるものではない。

参 考 例 1
スズメウリ属植物の抽出物の調製:
(1)バハクジ抽出物の調製;
バハクジ(Melothria indica) 20gを、50%エタノール 200mlを用いて、室温で5日間抽出した。 ろ過により固形物を除き、抽出液 188mlを得た。 この抽出液を減圧濃縮することにより溶液を留去し、抽出物を得た。 抽出物の収量は、1.96gであった(抽出物1)。

(2)ボウカ抽出物の調製;
ボウカ(Melothria heterophylla) 20gを、50%エタノール 200mlを用いて、室温で5日間抽出した。 ろ過により固形物を除き抽出液 185mlを得た。 この抽出液を減圧濃縮することにより溶液を留去し、抽出物を得た。 抽出物の収量は、1.64gであった(抽出物2)。

試 験 例
メラニン生成抑制試験
上記参考例1の(1)及び(2)で得た抽出物について、培養細胞を用いてその脱色効果を調べた。 すなわち、マウスのメラノーマ細胞を用いて、次の様にメラニン生成抑制試験を行った。 先ず、1×105個のB16メラノーマ細胞を、10%(v/v)牛胎児血清を含むイーグル最少栄養培地 5mlを入れた直径60mmのシャーレに播種し、5%(v/v)炭酸ガスに調整した炭酸ガスインキュベーターで37℃で24時間培養した。

次いでこのシャーレに、表1に記載の濃度となる量の各抽出物を50%メタノールに溶解した試料を50μl添加した。 同条件でさらに5日間培養した後、トリプシン処理により細胞を回収し、その白色化度を肉眼で評価した。 コントロールとしては、50%エタノール単独のものを用いた。 また、白色化度の判定は、従来より美白成分として知られているアルブチンを指標とし、13.6μg/ml(50%エタノール中)の時の白色化度を「++」(白色化大)、6.8μg/mlの時の白色化度を「+」(白色化中)、50%メタノールのみの白色化度を「−」(白色化せず)と判定した。 更に、「+」ほどは白色化していないが、白色化が起こっているものを「±」(やや白色化)と判定した。 同時に、細胞塊体積の変化を肉眼で判定し、細胞毒性の指標とした。
