生物学及び化学の発展を容易にするために、流体マイクロチップ技術が、調整された微細製造された環境下で従来の科学研究所の機能を実行するためにますます利用されている。微細製造された化学的計測法は、ラボオンチップ技術としても知られ、複数の微細製造された機能エレメント又はマイクロチップ上で協働するようにリンクされた単位プロセスを開発して、小体積の化学/生物学測定法を開発する必要がある。

本発明の1つの態様によれば、動電的に駆動された流体材料ストリームを基板上に同時に空間的に閉じ込めるようになっている流体マイクロチップが提供される。装置は、基板の表面に形成された収束チャンバと、サンプルの流体ストリームを自身の中で搬送する前記の基板の表面に形成された2つのサンプルチャネルとを含んでいる。

本発明による流体マイクロチップを、その2つの好ましい実施形態と関連させて説明する。一つの実施形態では、収束チャンバは、2つのサンプル材料ストリームが、3つの収束チャネル中で提供される収束流体を用いて横方向に閉じ込められるすなわち収束されるマイクロチップ上に提供される。第2の実施形態では、本発明によるマイクロチップは、収束されたサンプル材料ストリームを受容する収集チャネルと、収束済みサンプル材料ストリームの移送をこの収集チャネル中に選択的に方向付けするためのバッファ流体を提供するバッファチャネルと、を含んでいる。本発明に従ってマイクロチップを用いて流体サンプル材料を分析する方法もまた、移送細胞測定法の動作例と関連して説明する。

本発明を実現するデバイスは、動電力を用いて、流体材料のストリームを微細製造されたチャネルを介して駆動する。この微細製造されたデバイスは、内部を同時に走行している流体材料のストリームを閉じ込めるための収束チャンバと流体連通している複数のサンプルチャネルと収束チャネルを含んでいる。この収束チャンバは、マイクロチップの基板に形成されているサンプルチャネルと収束チャネルの合流点に形成されている。

収束されたストリーム幅は収束チャンバの出口(排水チャネルの取り入れ口)の幅に比例する。例えば、図2Aと2Bに示す実施形態では、収束チャネル40及び32並びにサンプルチャネル28の中の流体ストリームは排水チャネル48に集中し、また、収束チャネル32及び42並びにサンプルチャネル36中の流体ストリームは排水チャネル50に集中する。狭い排水チャネル48と50は物理的バリヤ、すなわち移送の障害物と成るが、これによって、動電的な収束の閉じ込め性が向上する。これは、単一サンプルと複数サンプルの双方の場合の動電的収束チャンバに応用される。したがって、収束チャンバと排水チャネルを狭くすることによってより緊密な横方向の閉じ込めが達成される。

本書に述べるようなマイクロチップデバイス10によって提供される多重動電的収束効果は、撮像用の電荷結合素子(CCD)と共にレーザー誘導の蛍光(LIF)を用いると観察された。アルゴンイオンレーザービーム(514.5nm、100mW)がレンズを用いて微細製造デバイス10の表面のところに直径で約5mmに拡大された。蛍光信号が光顕微鏡を用いて収集され、スペクトル的にフィルタリングされ(550nmカットオン(cut−on))、CCDで測定された。

前記基板の表面に形成された収束チャンバと、
前記基板の前記表面に形成されて、サンプル流体ストリームを搬送する第1及び第2のサンプルチャネルであって、夫々、サンプル流体発生源と流体連通した第1の端部と、前記収束チャンバと流体連通された第2の端部とを有する第1及び第2のサンプルチャネルと、

前記微小粒子の物理的特性値を測定する工程と、
測定された物理的特性値を前記物理的特性値の基準値と比較する工程と、
前記測定された物理的特性値が前記基準値に対して第1の量的関係を有する時に、前記サンプル流体ストリームを第1のリザーバへ向けさせ、或いは、前記測定された物理的特性値が前記基準値に対して第2の量的関係を有する時に、前記サンプル流体ストリームを第2のリザーバへ向けさせる工程とを含む、マイクロチップ上で流体媒体中の微小粒子を分析する方法。 - 前記サンプル流体ストリームの幅を狭める前記工程が、前記サンプル流体チャネルの幅を狭める工程を更に含んでいる、請求項6に記載の方法。
- 基板を有するマイクロチップ上で流体媒体中の微小粒子を分析する装置であって、
前記基板の表面に形成された収束チャンバと、
前記基板の前記表面に形成されて、サンプル流体ストリームを搬送するサンプルチャネルであって、微小粒子を含むサンプル流体発生源と流体連通された第1の端部と、前記収束チャンバと流体連通された第2の端部とを有するサンプルチャネルと、
前記基板の前記表面に形成されて、収束流体ストリームを搬送する収束チャネルであって、収束流体発生源と流体連通された第1の端部と、前記収束チャンバと流体連通された第2の端部とを有する収束チャネルと、
前記基板の前記表面に形成されて、前記サンプル流体を受けるために前記収束チャンバと流体連通された第1の端部を有する検出チャネルと、
前記基板の前記表面に形成されて、前記検出チャネルと流体連通された第1の端部を有する収集チャネルと、
前記基板の前記表面に形成されて、前記検出チャネルと流体連通された第1の端部を有する排水チャネルと、
前記収束流体ストリームが前記サンプル流体ストリームを前記収束チャンバ及び前記検出チャンバ内に空間的に閉じ込めるように、前記サンプル流体ストリームと前記収束流体ストリームとを前記各チャネルを介して前記収束チャンバ内へ追い遣る手段と、
前記検出チャネル内に空間的に閉じ込められたサンプル流体の微小粒子を検出する手段と、
検出された微小粒子の物理的特性値を測定する分析手段と、
測定された物理的特性値を前記物理的特性値の基準値と比較する比較手段と、
前記測定された物理的特性値が前記基準値に対して第1の量的関係を有する時に、前記サンプル流体ストリームを前記収集チャネルへ向けさせ、前記測定された物理的特性値が前記基準値に対して第2の量的関係を有する時に、前記サンプル流体ストリームを前記排水チャネルへ向けさせる方向転換手段とを有する装置。 - 前記検出チャネルが、前記サンプルチャネルの幅よりも狭い幅を有している、請求項8に記載の装置。
- 前記サンプルチャネルが、前記検出チャネルの幅よりも狭い幅を有している、請求項8に記載の装置。
- 前記サンプルチャネルと前記検出チャネルが、前記収束チャネルの幅よりも狭い幅を夫々有している、請求項8に記載の装置。
- 前記基板の前記表面に形成されて、第2の収束流体ストリームを搬送する第2の収束チャネルを更に有し、前記第2の収束チャネルが、収束流体発生源に流体連通された第1の端部と、前記収束チャンバに流体連通された第2の端部とを有し、それにより、前記サンプル流体ストリームが前記第1及び第2の収束流体ストリームによって空間的に閉じ込められるようになっている、請求項8〜11のいずれか一項に記載の装置。
- 前記基板に形成されて、バッファー流体を搬送するバッファーチャネルであって、バッファー流体発生源に流体連通された第1の端部と前記収集チャネルと前記排水チャネルに流体連通された第2の端部を有するバッファーチャネルと、
前記バッファー流体発生源と前記収集チャネルと前記排水チャネルに作動的に連結されて、前記バッファーチャネルを介して前記バッファー流体を駆り立てて、前記サンプル流体ストリームを前記排水チャネル又は前記収集チャネルに向けさせ又は前記排水チャネルと前記収集チャネルとの双方へ向けさせるための手段とを更に有している、請求項8又は12に記載の装置。 - 材料ストリームを空間的に閉じ込める装置であって、
基板の表面に形成された収束チャンバと、
前記基板の前記表面に形成されて、第1のサンプル流体ストリームを誘導する第1のサンプルチャネルであって、サンプル流体発生源と流体連通された第1の端部と、前記収束チャンバと流体連通された第2の端部とを有する第1のサンプルチャネルと、
前記基板の前記表面に形成されて、収束流体ストリームを誘導する収束チャネルであって、収束流体発生源と流体連通された第1の端部と、前記収束チャンバと流体連通された第2の端部とを有する収束チャネルと、
前記基板の前記表面に形成され、前記収束チャンバと流体連通された第1の端部と排水リザーバと流体連通された第2の端部とを有する第1の排水チャネルと、
前記サンプル流体発生源と前記収束流体発生源とに作動的に接続されて、前記収束流体ストリームが前記第1のサンプル流体ストリーを前記収束チャンバ内に空間的に閉じ込めるようにするために、前記サンプル流体ストリームと前記収束流体ストリームとを前記各チャネルを介して前記収束チャンバ内へ駆り立てる手段とを有し、
前記第1の排水チャネルが前記収束チャネルの幅よりも狭い幅を有している装置。 - 前記基板の前記表面に形成されて、第2のサンプル流体ストリームを誘導する第2のサンプルチャネルを更に有し、前記第2のサンプルチャネルが、第2のサンプル流体発生源と流体連通された第1の端部と前記収束チャンバと流体連通された第2の端部とを有し、前記サンプル流体ストリームと前記収束流体ストリームとを駆り立てる前記手段が、前記第2のサンプル流体ストリームを前記収束チャンバへ駆り立てるよう構成されて、前記収束流体ストリームが前記第1及び第2のサンプル流体ストリームを前記収束チャンバ内に空間的に閉じ込めるようになっている、請求項14に記載の装置。
- 前記収束チャネルが、前記第1のサンプルチャネルと前記第2のサンプルチャネルとの間に配置された前記収束チャンバに接続されて、前記排水チャネルと実質的に向き合っている、請求項15に記載の装置。
- 前記基板の前記表面に形成された第2の排水チャネルを更に有し、前記第2の排水チャネルが、前記収束チャンバに流体連通された第1の端部と第2の排水リザーバに流体連通された第2の端部とを有し、前記収束流体ストリームが前記収束チャンバ内に入った時に前記第1及び第2のサンプル流体ストリームを前記収束チャンバ内に閉じ込めるように、前記収束チャネルが前記第1のサンプルチャネルと前記第2のサンプルチャネルとの間に配置された前記収束チャンバに接続されている、請求項15に記載の装置。
- 前記第1のサンプルチャネルが、前記収束チャネルの幅よりも狭い幅を有している、請求項14に記載の装置。
- 前記基板の前記表面に形成されて、第2の収束流体ストリームを誘導する第2の収束チャネルを更に有し、前記第2の収束チャネルが、収束流体発生源に流体連通された第1の端部と前記収束チャンバに流体連通された第2の端部とを有し、前記第1のサンプルチャネルが、前記第1の収束チャネルと前記第2の収束チャネルとの間に配置され、前記第1及び第2の収束流体ストリームが前記第1のサンプル流体ストリームを前記収束チャンバ内に閉じ込めるように、サンプル流体ストリームと収束流体ストリームを駆り立てる前記手段が、前記第1のサンプル流体ストリームを前記収束チャンバへ駆り立てるように構成されている、請求項14又は18に記載の装置。
- 前記第1及び第2のサンプルチャネルが、夫々、前記収束チャネルの幅よりも狭い幅を有している、請求項15又は16に記載の装置。
- 前記第1及び第2のサンプルチャネルが、夫々、前記収束チャネルの幅よりも狭い幅を有している、請求項17に記載の装置。
- 前記第1及び第2の排水チャネルが、夫々、前記収束チャネルの幅よりも狭い幅を有している、請求項21に記載の装置。
