Description
超音波画像形成は治療方法を助けるための診断手段として用いられる。それは音波エネルギーの焦点を標的領域に集めそして反射させて画像を作ることができるという原理に基づいている。一般に、超音波変換器を画像を形成しようとする領域の上部の身体の表面に置き、そして音波を送受信することにより生ずる超音波エネルギーが伝播される。超音波エネルギーは変換器にまで反射され、そこで超音波画像に翻訳される。反射されたエネルギーの量及び特性は組織の音響的性質に依存する。そして標的部位で超音波エネルギーを発生させそして受ける画像を改善するため、反響を発生させる造影剤(contrast agents)を使用することが好ましい。
超音波画像形成では、造影剤の注入後に得られるビデオテープ画像をデジタル化し、心臓の30周期に対して1から225のグレースケール(gray scale)単位でグレースケールを定量化することができる。このコントラストの強度は水平軸上にとった時間に対して縦軸上にプロットされる。(ベースライン強度に対して補正された)ビデオ強度のピークは時間強度曲線上の最高点として決定される。

コントラスト超音波心臓検査法は心臓内部の構造を描くため、弁の能力を評価するため、そして心臓内シャントを証明するために用いられてきた。心筋コントラスト超音波心臓検査法(MCE)はヒトの冠状血流予備力(coronary blood flow reserve)を測定するために用いられてきた。MCEは心筋灌流の相対的変化を測定するためまた危険な部位を描出するために安全かつ有用な技術であることが明らかになってきた。
潜在的超音波画像形成剤の多重性(multiplicity)がコントラスト超音波心臓検査法について報告されてきた。そのような画像形成剤でも末梢の静脈内注射の後の心筋の取り込みを通常の検査で視覚的に区別できるようにするものはない。静脈注射の後における超音波造影剤の肺臓を横切る移動について多くの報告があるけれども、そして心エコー図(echocardiogram)上での心筋の混濁形成(opacification)がこのような造影剤の左側注射により生じ得るという事実にもかかわらず、心筋コントラストの映像化は音波処理された微小気泡の静脈内投与によっては達成されなかった。

発明の概要
本発明によれば、微小気泡造影剤の注入後に超音波の伝播を中止して造影剤が標的器管を灌流することを可能とし、ついで超音波の伝播を再開してコントラストの顕著な改善を得ることができる。本方法の使用により、コントラストを10倍まで改善し、音響の陰影を減少させそして必要な造影剤の量を減少させることができ、それによって毒性の減少及び安全性の改善がもたらされる。

コントラスト超音波画像形成における第一の問題は、心臓、腎臓、肝臓その他の解剖学的標的器管における灌流の検出及び定量である。今日までの成功のほとんどは血流中に静脈注射される造影ガス充填微小気泡を用いてなされてきた。
しかしながら、静脈注射された微小気泡の固まりは標的器管まで行くにつれて濃度が希釈され、不可能ではないにしても検出が困難になることがしばしばある。超音波心臓検査法の第一の問題の一つは、心筋灌流の検出である。さらに重要なことは、この固まりは心臓の右側を通って流れそして肺臓を通過するとき、サイズが8ミクロンより大きな微小気泡は肺の毛細管により濾過され除かれることである。
さらに、この気泡は心臓の左側に流入するとき、左心室(LV)でかなり高い収縮圧を受けるため微小気泡の一部が心臓周期の過程で破壊される。気泡の一部は周囲の液体媒質中への拡散という単純なプロセスにより消失する。
最後に、心室内の気泡は放射された超音波の音場をひどく減衰させそして遠位の心筋層上に音響陰影を投射する。従って、微小気泡検出感度の改善のための投与量の増加と心筋ベッドの音響陰影の最小化の必要性との間のトレードオフがある。このすべてが、左心室中の総血量のうちの僅か約4%しか冠状循環に入らないという事実により悪化させられる。この正味の結果として、(最初の固まりの注射からの)小さなサイズの微小気泡の極く一部分だけが心筋の血管系を通過することになる。
本発明は、極く僅かな量の超音波造影剤の検出を可能とし10倍程もコントラストを改善することによりこれらの懸念を緩和し,そして腎臓や肝臓の画像形成と同様に超音波心臓検査法に使用することができる。
本発明(中断された超音波により誘導されるキャビテーション画像形成)は標準的な超音波画像形成技法の改良を含み、そして微小気泡造影剤と組み合わせていかなる超音波画像形成装置とでも使用することができる。
この方法では、造影剤が注入される際には超音波シグナルが作動している(すなわち、変換器が標的器管、例えば心臓の上の適正な位置に保持される)。次いで、超音波造影剤が標的器管の視覚的検出に十分な量だけ末梢から注射される。使用される微小気泡造影剤の標準量はヒトに対して約0.04ml/kgから約0.08ml/kgまでであるが、本発明の方法では0.001ml/kgから0.0025ml/kgという遙かに少ない量で等しく良好に働くことが見いだされている。大きな患者は同等の左心室コントラストを得ようとすれば幾らか高い投与量を必要とするであろうから、投与量の範囲は患者に特異的である。コントラスト超音波心臓検査法の標準的方法は、ワイマン(Weyman),アーサーE.“超音波心臓検査法の原理と実際”Lea and Fibiger,マルベルン,PA(1994第2版)に記載されている。
注射後、超音波シグナルの伝播は造影剤が標的器管を灌流するのに十分な時間だけ妨げられ又は中止される。この時間は一般に標的器管によって変動する。それは造影剤の一部が標的器管に到達するのに十分な程長くなければならないだけである。心臓にとってはこれは造影剤が心筋層に到達するのに十分な時間となるであろう。これは、近似的には約10−120秒、好ましくは20−30秒程度であり、そして患者毎に大きく変えるべきものではない。
他の器管や個々の特定の患者に対するこの中止の時間は、伝播を妨害せずそして注射から標的器管の灌流までの時間を測る標準的超音波画像形成を行うことにより容易に確認することができる。
一旦微小気泡が超音波心臓検査法の標的器管すなわち心筋層に到達すると、超音波シグナルの伝播は再開され、そしてピークの心筋コントラスト画像が得られる。
超音波心臓検査法では、心筋層全体の均一な混濁形成が存在するであろう。数秒のうちに、全心筋層の強度は元の(暗い)強度レベルに均一に低下する。血流がなくコントラストのない心筋ベッドの領域は常に暗いままである。従って、正常に灌流した心筋領域と異常に灌流した心筋領域の間の区別をすることが容易となる。
如何なる理論によっても束縛されることは望まないが、この観察された現象は、放射されている超音波の音場に曝されたときの造影剤微小気泡の一過性の応答の結果として生ずるものと仮定される。この超音波シグナルは微小気泡を収縮させ、又はキャビテーションを起こさせる原因となる。微小気泡が注入されると、それらは超音波の音場に遭遇するとき圧縮させられる。超音波シグナルが中止されると、解剖学的構造中への伝播力は遮断される。放射されている音場がないときは、心筋層又は他の器官内の微小気泡は一定の大きさまで成長する。一部は合体してより大きな散乱剤(scatterers)を形成する。ついで超音波シグナルの反射はより大きなサイズの微小気泡により増強される。これは後方散乱された超音波エネルギーは標的領域の散乱剤の数に直接比例しそして散乱剤の半径の6乗で変化するからである。
超音波系が再開されると、解剖学的構造への伝播力はディスプレー上での画像走査を可能としそしてそれを作りだす。ピークコントラスト画像はこの点、すなわち超音波シグナルがもう一度微小気泡をキャビテーションし始める前の点で得られる。超音波の音場がゼロから立ち上がり定常状態に達するにつれて、微小気泡に対する圧力は増加し、もう一度それらのサイズを減少させる。散乱剤のサイズのこの一過性の減少が心筋強度の一過性の減少を生じさせる原因となる。
超音波伝播の再開の際の強度の増加に対する別の説明は、加えられた超音波の音場が微小気泡や合体した微小気泡の集合を吹き飛ばして無数の微小気泡とし、それにより散乱剤の数を有効に増加させそしてそれにより心筋の混濁形成を増加させるという仮定である。このコントラストは非常に改善されるので、形成される画像は核磁気共鳴映像法のようなより複雑な画像形成法の画像に匹敵する。
本発明の主要な利点は、音響陰影を作ることなく、造影剤微小気泡の極めて少量を用い、非侵襲的方法で標的器管における灌流の感度の良い検出及び定量を可能とすることである。小投与量のため、患者の安全性は高められそして費用も最少化される。
多くの超音波画像形成では、造影剤は画像を作るべき宿主に末梢投与するため薬学的に有効な投与形態で処方される。このような宿主は、イヌやウマなどの他の哺乳動物宿主でも有効に画像形成をすることができるが、一般にヒトが対象である。最も好ましい態様では、造影剤は市販されているアルブミン(ヒト),USP,の溶液(一般に5重量%又は25重量%滅菌水溶液で供給される)、及び市販されているデキストロース,USP,の音波処理された混合液であり、静脈内投与が用いられる。この混合液は周囲の条件、すなわち室内空気、室温、室内圧力の下で音波処理されそして音波処理の間パーフルオロカーボン又は他の市販されている不活性ガス(99.9重量%)と共に灌流される。
好ましい態様においては、本発明は微小気泡造影剤を使用する。この微小気泡は、膜形成性の変性可能なタンパクの皮膜によって安定化される。適当なタンパクとしては、アルブミン、ヒト・ガンマ−グロブリン、ヒト・アポトランスフェリン、ベータ−ラクトース、及びウレアーゼなどの天然に生ずるタンパクが挙げられる。本発明は天然に生ずるタンパクを用いることが好ましいが、合成タンパクも使用することができる。特に好ましいのはヒト・血清アルブミンである。
音波処理された微小気泡から作られた静脈音響造影剤が知られており(例えば、アルブネクス、モレキュラー・バイオシステムズ社)、本発明でも使用することができるが、薬学的に許容される糖類、例えばデキストロース、とタンパク質、例えばアルブミン、との混合物を含む音波処理された水溶液を用いることが好ましい。一般に、音波処理は空気雰囲気下で行われる。特に好ましい態様では、薬学的に許容される投与形態で容易に入手可能なデキストロースが好ましい糖類でありそしてヒト・血清アルブミンが好ましいタンパクである。好ましい態様には、5〜50重量%のデキストロースと2〜10重量%のヒト・血清アルブミンの2倍〜8倍希釈液が含まれる。本発明の他の糖溶液の例は、単糖類の水溶液(例えば、ヘキソース、デキストロース又はフルクトースなどの式C6H12O6を持つもの又はこれらの混合物)、2糖類水溶液(例えば、スクロース、ラクトース又はマルトースなどの式C12H22O11をもつもの又はこれらの混合物)、又は多糖類水溶液(例えば、アミロース又はデキストランなどの式(C6H10O5)nを持つ可溶性澱粉、ここでnは約20と約200の間の全整数である、又はこれらの混合物)である。超音波エネルギーによる音波処理は、デキストロース−アルブミン溶液内で流体中の粒状物又はガスの部位でキャビテーションを惹起する。これらのキャビテーション部位はついには共鳴し、合体せず安定な小さな微小気泡(約4〜約7ミクロンのサイズのもの)を形成する。一般に、音波処理の条件としては、約5ミクロンと約6ミクロンの間の微小気泡が約4×108mより高い濃度で形成される条件が好ましい。
音波処理されたデキストロース・アルブミン微小気泡の平均サイズは約5ミクロンと約6ミクロンの間にある。これは良いサイズである。その理由は、微小気泡の半径が、その微小気泡の内部ガスと外部ガスの間に存在する拡散勾配のためまだ液体中に存在する時間の関数として減少することが観察されているからである。微小気泡のサイズの増加は血液内での微小気泡の存続に対して重要な効果を有する。それは肺臓を通過するために十分なサイズでもなければならない。それは平均直径が0.1ミクロンより大きく10ミクロンより小さくなければならない。アルブミン微小気泡のサイズは肺臓通過のためには理想的(5ミクロンと6ミクロンの間)であるから、アルブミン被覆微小気泡の静脈注射の後に生ずる左心室及び心筋のビデオ強度の有意の喪失の主な理由は、左心室腔への輸送の間に起こる静脈注射後の微小気泡内のガスの有意の拡散によるものである。空気よりも低い血液溶解性をもち100グラム/モルよりも大きな分子量をもつパーフルオロカーボンガスのような不活性ガスで増強された音波処理デキストロース・アルブミンは、音波処理されたアルブミン単独よりも有意に高い左心室及び心筋のビデオ強度を生ずる。
高い表面張力のために、微小気泡内の窒素ガス及び酸素ガスの濃度は血液中のそれよりも遙かに高い、従って、静脈注射の後、このガスの血液中への迅速な拡散が生ずる。ワイブル(Wible)ら(サーキュレーション,88:I-401,1993)は、吸入される窒素の画分を減らすことにより血液内の窒素の分圧を減少させると、この拡散過程を加速させることができることを証明した。このより低い窒素の外部濃度は、室内空気を吸入しながら音波処理されたアルブミンの同じ静脈注射により形成された左心室ビデオ強度の喪失を招く。ガス気泡からヒト血液中へ酸素が迅速に拡散することも観察された(ヤング(Yang)ら,J Biomech 3:275,1971を見よ)。
窒素と酸素は両方ともこれらの濃度勾配を横切って迅速に拡散するが、窒素は酸素よりもよりゆっくりと血液に溶解するようにみえる。窒素は空気の主要成分であるから、微小気泡を横切る窒素濃度勾配を減少させると、静脈注射の後の左心室及び心筋のビデオ強度を改善する。微小気泡を音波処理の間、窒素のそれよりも低い血液溶解性及び/又は微小気泡拡散性持ちそして約0.300lb/ft3よりも大きなガス密度を持つ無毒のガスに曝すと、音波処理されたデキストロース・アルブミン中の微小気泡のサイズ及び安定性が増大し、微小気泡の血液中への溶解性及び拡散性が減少する。適当なガスは37℃で気体でありそして無毒なガスである。造影剤として有用な不溶性ガスは、パーフルオロメタン、パーフルオロエタン、パーフルオロプロパン、パーフルオロブタン、パーフルオロペンタン等などのパーフルオロカーボンガス、又は六フッ化イオウを含むが、これらに限定されるものではない。好ましい態様では、このガスはパーフルオロプロパン(C3F8)又はパーフルオロブタン(C4F10)である。微小気泡のパーフルオロカーボンガスの含量は、イン・ビボで微小気泡ガスの血液溶解性及び血液拡散性を低下させるのに十分な量である。一般に、微小気泡中の不溶性ガスの有効最少量は、合体せずに肺循環を確実に通過する微小気泡を生ずる量である。これは静脈注射後における心臓の左心室の心筋層の混濁形成により証明され、そして超音波心臓検査法により視覚的に区別することができる。
心筋画像形成に加えて、本発明の造影剤は、腎臓及び肝臓の画像形成にも有用である。こうして、本発明の別の態様は心筋、腎臓又は肝臓の混濁形成のための方法を提供する。好ましい方法は本発明の音響造影剤を製造すること、該音響造影剤を静脈注射により宿主に注入すること、そして後により詳細に論ずるように適当なドプラー装置又は超音波反響装置を用いて該宿主に対する音響造影研究を行うことを含む。
最も好ましい態様の一つでは、造影剤は、5%ヒト血清アルブミンを5%デキストロースで3倍希釈し音波処理したものを含むパーフルオロブタン−増強・音波処理デキストロース・アルブミン溶液である。音波処理の間、該溶液は約80秒間パーフルオロプロパンと共に灌流され、付随的に音波処理の間に少なくとも約5秒間パーフルオロブタン又はパーフルオロプロパンに曝される。これは微小気泡ガスの溶解性及び拡散性を低下させる。過剰の溶液を音波処理用シリンジ中で少なくとも約120±5分間室温に置いて澄明にし、得られる微小気泡を濃縮する。過剰の溶液を追い出し、濃縮された微小気泡を滅菌シリンジに移し、そして哺乳動物中に静脈注射する。
超音波心臓検査法に本発明の方法を用いると、末梢静脈内投与の際、より高度の心筋の混濁形成、心臓内の縁の描出、及び左側超音波ドプラーシグナルの検出の増強を得ることができる。さらに、本発明の方法は微小気泡の僅かな投与をも容易に検出する程感度を増加させ、続いて静脈注射の後の肝臓及び腎臓の超音波映像化を可能とする。
以下の実施例は、例示の目的のみでなされるものであり、本発明を制限する意図は全くないものである。他の態様が実用可能でありそして本発明に含まれるものであることは当技術分野に熟練せる者にとって明らかであろう。
実施例1
造影剤の調製
アルブミン(ヒト)USP,5%溶液(本明細書では以下、“アルブミン”と呼ぶ)及びデキストロースUSP,5%溶液(本明細書では以下、“デキストロース”と呼ぶ)は市販品から入手した。音波処理に用いた音波処理装置はヒート・システム・超音波処理装置モデルXL2020(ヒート・システム社、ファーミングデール、ニューヨーク)であった。1/2インチ・ホーン型変換器は共振圧電装置であった。1/2インチ音波発生用ホーンチップは音波処理それぞれの前に滅菌した。
試料の音波処理
デキストロースで1:3(3倍)に希釈されたアルブミンの16mlずつを35ccの“モノジェクト”シリンジ(ベクトン・ディキンソン・アンド・カンパニー、ラザーフォード、ニュージャージー)中にとり、80±1秒間音波処理を行った。ついで、35mlシリンジの“リュール−ロク(Leur-Lok)”をストップコックに付ける。デキストロース・アルブミン溶液を約7〜約10秒間手で混合した後、シリンジの先端からプランジャーを除いた。滅菌した音波処理用ホーンを次いでシリンジの開口端中に入れアルブミン・デキストロース溶液の表面まで降ろした。溶液はホーンチップの位置に置かれそして手でこの位置に保ちながら、20,000Hzの振動数、210Wの出力で80±1秒間連続的に音波処理を行って安定な微小気泡溶液を調製した。
試料のガス還流
音波処理の間、デキストロース・アルブミン混合液はパーフルオロブタンガス又はパーフルオロプロパンガス(市販品のグレード、99.9重量%)に曝した。このガスは汚染を防止するため0.22μMフィルター(ミクロン・セパレーションズ社、ウエストボロウ、マサチューセッツ)を通して無菌シリンジ中へ引き込んだ。音波処理の間、パーフルオロカーボンガスの5mlを80秒の間にわたってストップコックを通して溶液中に手動で注入し、この溶解性のより小さなガスを含む微小気泡を形成させた。この処方に従って調製されたパーフルオロブタン−増強音波処理デキストロース・アルブミン(BESDA)又はパーフルオロプロパン−増強音波処理デキストロース・アルブミン(PESDA)の総容量は25±2ミリリットルであった。これらの試料は、次いで、静脈注射に使用された。
微小気泡分析
微小気泡のサイズ及び純度は血球計算法を用いて測定した。溶液中に合体した微小気泡が存在するときは、微小気泡の顕微鏡検査を行った。微小気泡濃度はクールター・カウンターを用いて計測した。次の諸条件のうちの一つに該当するときは、造影剤は使用から排除された。すなわち、微小気泡の平均サイズが4.0ミクロンから6.0ミクロンであること、合体した微小気泡又は糸状体が光学顕微鏡により検出されたこと、又は微小気泡の平均濃度が0.8×109微小気泡/ミリリットルよりも小さいか又は1.5×109微小気泡/ミリリットルよりも大きいこと。10ミクロンよりも大きな微小気泡の数が試料中で4%よりも多いときも、その試料は排除された。
試料はすべて注射時まで35ミリリットルのシリンジ中で貯蔵された。試料はすべて製造後36時間以内に投与された。試料はすべて薄層状の流動ずきん(laminar flow hood)中で調製された。
実施例2
開胸イヌの調製
いずれも6歳の雑種犬(15〜30キログラム)をペントバルビタールナトリウム(30ミリグラム/キログラム静脈注射)で麻酔し、インキュベートし、そして陽圧人口呼吸器を用い初めは室内空気で通気した。無菌条件下で左開胸を行いそして心膜を切開した。超音波造影剤の静脈投与及び圧力監視のため、19ゲージの末梢静脈ラインに加えて、8本のフレンチ・カテーテルを大腿の動脈及び静脈中に挿入した。1本の大腿静脈鞘から7Fバルーンを先端に装着したサーモダイリューション・カテーテルをX線透視装置を用いて肺動脈中に挿入し、肺動脈圧及び心拍出量を測定した。7Fのピグテール・カテーテルを左心室内に挿入して、超音波造影剤それぞれの注射後の圧力(左心室収縮期及び拡張終期の圧力)を測定した。
適当な外科的処置に続いて、市販の超音波スキャナー(ヒューレット・パッカード社、アンドーバー、マサチューセッツ)に連結させた3.5メガヘルツ超音波変換器を温かい水浴中に設置した。水浴は前方心外膜表面を覆う。変換器はクランプに載せ水浴中に沈められた。左心室及び右心室の最適安定短軸投影が心室中乳頭筋レベルで得られるように調節された。次いでこれらの画像が静脈注射後の左心室腔及び心筋の造影剤取り込み評価するために使用された。
実施例3
超音波心臓検査法のための、遅延された超音波−誘導キャビテーション画像形成対従来の超音波画像形成
本発明の方法を用いて音響陰影を減らすこと又は消去することができるかどうかを決定するため、4人のヒト患者について、従来の超音波画像形成と遅延された超音波−誘導キャビテーション画像形成とを比較する試験を行った。この実験では、市販の超音波システム(ヒューレット・パッカード・ソノス1500フェイズド・アレー・イメージング・システム、ヒューレット・パッカード社、アンドーバ、マサチューセッツ)と一緒に供給されたソフトウエアから得られたオン−ライン上デジタル化されたビデオ強度を用いて心筋のコントラストが決定された。ピーク心筋ビデオ強度(PMVI)、音響陰影の持続(AS)、及び心筋コントラストの%(MC)を観察した。結果を以下の表1に示す。
各患者は実施例1のようにして調製した0.0025〜0.005ミリリットル/キログラムの造影剤を静脈注射された。使用したフルオロカーボンガス剤はデカフルオロブタンであった。従来の超音波画像形成に対しては、超音波の伝播は2.5〜2.7MHzで一定であり、画像形成は実験を通じて連続的に行われた。心筋層のピークコントラスト画像は、造影剤の微小気泡が心筋層に到達したときに得られた。
遅延された超音波−誘導キャビテーション画像形成に対しては、超音波の伝播は患者が造影剤を注射されるまで2.5〜2.7MHzに維持された。造影剤の静脈注射の直後に、超音波伝播を20秒〜30秒間中止した。造影剤の微小気泡が心筋層に到達したとき超音波の伝播は2.5〜2.7MHzで再開された。ピークの心筋コントラスト画像は超音波伝播の開始直後そして微小気泡が収縮し始める前に得られた。
この実験の結果は、パーフルオロカーボン−増強音波処理デキストロース・アルブミン造影剤の静脈注射後の従来の画像形成が左心室の後部構造中に高度の音響陰影を生ずることを示す。遅延された超音波−誘導キャビテーション画像形成
は、心筋層の下層の欠陥の画像形成を可能とするのに十分な量だけ音響陰影の量を減少させた。本発明の方法は犬で標準的心筋画像形成とも比較され、従来法のそれの10倍の心筋コントラストの増加が得られた。音響陰影の顕著な減少も見られた。
実施例4
キャビテーション超音波画像形成に対する変換器の振動数と出力の影響
2頭の開胸犬を用い、本発明の方法における変換器の振動数及び変換器の音響出力の影響を明らかにした。
2頭の犬は全部で22回の、0.005〜0.010ミリリットル/キログラム体重の投与量でパーフルオロプロパン−増強音波処理デキストロース・アルブミン造影剤(実施例1のように調製したもの)の静脈注射を受けた。診断のための超音波シグナルは造影剤微小気泡が心筋層に到達した後にのみ伝播され、そしてピーク心筋ビデオ強度(PMVI)及び心筋コントラストの持続時間(dur-MC)を各注射の後に測定した。変換器の振動数及び音響出力は個々の注射の間で変更された。その結果を以下の表2に示す。
この結果から、最良の心筋コントラスト画像形成は変換器の振動数が低くそしてこの現象の間低い音響出力が引き延ばされるときに得られることが示される。

- 微小気泡を有する音響造影剤(echo contrast agent)を用いる心筋、腎臓、又は肝臓の超音波画像形成方法であって、該音響造影剤を静脈注射により人間以外の動物中に注入する工程、該造影剤が標的器管を灌流するのに要する10〜120秒間超音波の伝播を中止する工程、そしてその後に、超音波の伝播を再開する工程を含む方法。
- 該動物が犬でありそして造影剤を注入する該工程が0.005ml/kgから0.010ml/kgまでの量でなされるものである、請求項1記載の方法。
- 該音響造影剤を注入する該工程が、パーフルオロカーボンガス又は100グラム/モルより大きな分子量を持つ他の不溶性ガスをカプセル化した微小気泡を持つ造影剤を注入する工程を含むものである、請求項1記載の方法。
