本発明はキトサンとカルニチンをソバ、ウドン、パン、ビスケット、ラーメン、クッキー及び惣材などの一般食品に配合したダイエット食品、さらには、キトサンとカルニチンを錠剤、ソフトカプセルなどの健康食品に配合したダイエット食品あるいはキトサンとカルニチンを含む錠剤、ソフトカプセル等のダイエット食品に関し、キトサンとカルニチンの併用により優れたダイエット効果を有し、キトサンのエグ味が生じないという利点を有する。

【従来の技術】
キトサンは甲殻類、昆虫類及び菌類に広く存在するキチンを脱アセチル化したD−グルコサミンのポリマーである。キトサンはその難消化性からいわゆる食物繊維の効果が期待され、特にアミノ基をもつために、消化管内で胆汁酸を吸着しコレステロ−ルの吸収阻害並びに胆汁酸の再吸収阻害効果により、優れた血清コレステロールの低下作用を有する他、便秘改善、腸内細菌叢の変化が報告されている。また、近年、キトサンのダイエット効果が注目され、キトサンとアスコルビン酸を同時に摂取することにより、すぐれたダイエット効果を有することが報告されている。

【発明が解決しようとする課題】
しかしながら、キトサンとアスコルビン酸を同時に摂取するとキトサンが溶解しキトサン独特の強いエグ味が生じるため、一般食品に添加することは難しい。また、錠剤に加工してもキトサンが水分を吸収しすばやく崩壊するため、口の中でエグ味を呈することがある。また、糖衣加工を施してもキトサンは糖衣加工時に水分を吸収するため、糖衣層にひび割れを生じ錠剤の保存性が悪い。また、ソフトカプセルは錠剤に比較してキトサンが十分量添加できないため粒数を多く摂取する必要がある。

このように、キトサンとアスコルビン酸の同時摂取はすぐれたダイエット効果を有するにもかかわらず、食品に添加する際に味の問題点があるため、使用方法に制限があった。これらの事より、味に問題のない安全なキトサンを使用したダイエット食品が望まれていた。

【課題を解決するための手段】
本発明者らは種々検討した結果、キトサンとカルニチンを併用することにより、優れたダイエット効果を有し、キトサンのエグ味が生じないダイエット食品が得られることを見出し、本発明を完成させた。

即ち、本発明は、
(1)キトサンとカルニチンを含有することを特徴とするダイエット食品、
(2)ダイエット食品が製剤である上記(1)項記載の食品、
(3)製剤が錠剤、顆粒製剤、粉末製剤又はカプセル製剤である上記(2)項記載の食品、
に関する。
本発明のダイエット食品は、ダイエット効果を有し、キトサンのエグ味が生じない。

【発明の実施の形態】
本発明でキトサンとカルニチンを配合する一般食品は何でも良く、例えばソバ、ウドン、パン、ビスケット、ラーメン、クッキー、惣材等があげられる。
本発明で用いられるキトサンとしては、カニ、エビの甲皮、昆虫の甲殻、イカ、キノコ及び糸状菌等の細胞壁より得られた物等があげられるが、好ましくはカニ、エビの甲皮より得られたものがよい。キトサンは、グルコサミン、アセチルグルコサミンを成分とし、酸性の水に溶解するもので、平均分子量は通常1万以上あればよいが、好ましくは、平均分子量10万以上で、回転粘度が20mPa・s以上であり、より好ましくは、平均分子量が80万以上で、回転粘度が100mPa・s以上である。また、キトサンの粒度は特に制限はないが、好ましくは30メッシュパス以下であり、より好ましくは80メッシュパス以下の細かいものが良い。

また、本発明で使用するカルニチンは天然抽出品、合成品いずれでも良く、また、カルニチンを多く含む羊、牛、ブタ等の食用動物の筋肉等を加水分解したエキスもしくはその乾燥粉末を直接使用してもよい。

本発明のダイエット効果が得られるキトサンとカルニチンの比率は特に制限はないが、好ましくはキトサン100部に対してカルニチン0.2部〜200部であり、より好ましくは1部〜100部である。また、食用動物の加水分解物を使用する場合はそのカルニチン含有量によって調整する必要がある。例えば、カルニチン2%含有の加水分解物を使用する場合はキトサン100部に対して10部〜10000部が好ましく、50部〜5000部がより好ましい。
本発明でのキトサンとカルニチンの食品への配合方法は、いかなる方法でも良いが、好ましくは粉末又は水で懸濁した後に、食品原材料中に配合するか又は一般食品に添加するのが良い。また、その配合割合は、いかなる割合でも良いが、キトサンとカルニチンの合計物として1食あたり0.1〜5gの範囲が好ましい。

本発明でのキトサンとカルニチンの錠剤への配合方法は、いかなる方法でも良いが、好ましくは粉末状のキトサンとカルニチンを必要により賦形剤等の通常使用される配合剤と混合して打錠するか又は配合剤と練合して造粒後、乾燥して打錠する方法が好ましい。また、キトサンとカルニチンをそれぞれ別工程で造粒し、混合して打錠しても良い。また、その配合割合は、いかなる割合でも良いが1日に摂取する錠剤量あたりキトサンとカルニチンの合計量として0.1〜5gの範囲が好ましい。

本発明でのキトサンとカルニチンの顆粒製剤、粉末製剤又はカプセル製剤への配合方法は、いかなる方法でも良いが、キトサンとカルニチンとその他の配合剤の粉末から常法により造粒して顆粒製剤とするのが好ましい。また、キトサンとカルニチンをそれぞれ別工程で造粒し、混合しても良い。粉末製剤ではキトサンとカルニチンを粉末のまま配合するのが好ましい。また、カプセル製剤ではそのままカプセルに封入するか又は造粒して顆粒としてこれを乾燥後配合するか、オイル等で懸濁し、カプセルに封入するのが好ましい。また、それらの配合割合は、いかなる割合でも良いが、1日摂取量あたりキトサンとカルニチンの合計量として0.1〜5gの範囲が好ましい。
錠剤、顆粒製剤、粉末製剤、カプセル剤等の製剤は、キトサン及びカルニチンの他に、必要に応じ滑沢剤、賦形剤、崩壊剤等、通常使用する配合剤を含むことができ、又、他の健康食品の有効成分として使用されているものであって、本発明の目的の達成に障害とならない成分を含むことができる。

次に、本発明を実施例により説明するが、本発明はこれに限定されるものではない。

試験例
本発明の優れた効果を動物実験で確認した。動物実験は「実験動物の飼育及び保管等に関する基準」(昭和55年3月総理府告示第6号)等を厳守して行なった。試料組成を表1に示す。使用したキトサンは回転粘度150mPa・s 、脱アセチル化度92%で80メッシュパスの粉末を用いた。カルニチンは金剛化学(株)製のL−カルニチンを用いた。また、カルニチン含有ペプタイドとしてゼネックス(株)製の羊ペプタイド(商品名SBMB粉末)を用いた。
試験飼料はショ糖を糖質源とした標準飼料(実験例1)を対照としてキトサン添加群はセルロースの代りにキトサンを5%添加し、カルニチンは0.44%をショ糖に置き換えて添加した。カルニチン含有羊ペプタイドはカゼインに置き換えて20%添加した。このペプタイドはカルニチンを2.2%含有するので、試料中にはカルニチンが0.44%含まれることになる。
実験は、体重70−80gの4週齢Sprague-Dawley系雄ラット(日本クレア社)を、標準飼料で7日間予備飼育した後、1群6匹づつ6群に分け、表1に示した実験飼料をそれぞれ投与して9日間飼育した。飼料及び水は自由に摂取させた。9日目の12時より絶食し、10日目にネンブタ−ル麻酔下で開腹し、心臓穿刺により採血した後、脂肪組織(後腹壁ならびに副睾丸周辺組織)を摘出し重量を測定した後、内蔵を摘出した。結果に関しては、1元配置分散分析を行ない、各水準の有意差の検定にはTukey 型の多重比較を用いた。

結果
試験結果を表2に示す。全ての試験群で成長、及び内蔵摘出屠体重に差はみられなかった。しかしながら、脂肪組織の生体重比では、実験例1に比較して、キトサンとカルニチンを同時に与えた実験例4とキトサンと羊ペプタイドを与えた実験例6で有意な低下がみられた。一方、キトサン単独投与群(実験例2)およびカルニチン単独投与群(実験例3)、羊ペプタイド単独投与群(実験例5)はコントロールとの間に有意な差は見られなかった。このことは、キトサンとカルニチンの同時摂取が、体の筋肉組織の減少を伴わず体組織の脂肪を取り除く効果があることを示している。また、すべての試験区で血液生化学検査及び剖検において異常は認められなかった。
