【発明の属する技術分野】
本発明はコンタクトレンズの洗浄液に関し、特に含まれる微生物由来の蛋白質
分解酵素の安定性が向上され、その結果、高い洗浄力を有するコンタクトレンズ
用洗浄液に関するものである。

【従来の技術】
従来より、コンタクトレンズに付着した蛋白質汚れを除去するため蛋白質分解
酵素を用いた洗浄方法が広く普及している。これらは、蛋白質分解酵素を含有す
る錠剤もしくは顆粒を精製水もしくは生理的食塩水で溶解して用いる。また近年
液体化された酵素による洗浄液として、特開平1−180515号公報や特開平
1−318099号公報に開示された洗浄液等が提案されている。これらの洗浄
液は基本的に、界面活性剤と蛋白質分解酵素とグリセリン等の水に混和性の有機
液体と水との混合液からなるものである。グリセリン等の水に混和性の有機液体
を添加することで、液体でありながら一定期間酵素活性を安定化している。そし
て、前述の洗浄液は、一般的に使用時において、酵素と有機液体を含有する洗浄
原液を水等により希釈して作成し、コンタクトレンズ保存ケース中に入れると共
にコンタクトレンズを浸漬して洗浄するものである。

【発明が解決しようとする課題】
しかしながら、上記洗浄液においても、酵素活性の経時的な低下は避けられず
、さらなる酵素活性の安定化が望まれている。
本発明は、このような課題を解決するためになされたものであり、本発明の目
的は酵素活性がより安定化されたコンタクトレンズ用洗浄液を提供することにあ
る。

【課題を解決するための手段】
本発明の第1の態様は、微生物由来の蛋白質分解酵素、プロピレングリコール
及びホウ砂を含むことを特徴とするコンタクトレンズ用洗浄液に関する。
本発明の第2の態様は、上記第1の態様の洗浄液にホウ酸をさらに含む洗浄液
である。ホウ酸をさらに含むことで酵素の活性をさらに向上させることができる
という利点がある。
本発明の第3の態様は、上記第1または第2の態様の洗浄液であって、実質的
に界面活性剤を含有しない洗浄液である。実質的に界面活性剤を含有しないこと
で、酵素の活性をさらに向上させることができるという利点がある。

以下、本発明を詳細に説明する。
本発明のコンタクトレンズ用洗浄液は、微生物由来の蛋白質分解酵素、プロピ
レングリコール及びホウ砂を必須成分として含有する。
蛋白質分解酵素は、コンタクトレンズに付着した汚れを除去するためのもので
ある。コンタクトレンズに付着する汚れとしては、例えば、蛋白質、脂質、多糖
類等が知られており、蛋白質分解酵素により、主な汚れである蛋白質を分解して
除去することができる。
蛋白質分解酵素としては、微生物由来の蛋白質分解酵素を用いる。微生物由来
の蛋白質分解酵素としては、例えばナガセ生化学工業(株)のビオプラーゼ、天
野製薬(株)のプロテアーゼNアマノ、ノボ・インダストリー・ジャパン(株)
のアルカラーゼ、エスペラーゼ及びサビナーゼ等が使用出来る。これら微生物由
来の蛋白質分解酵素は単独又は2種以上の組み合わせで用いることができる。

また、微生物由来の蛋白質分解酵素に、脂質分解酵素(例えば、リバーゼ)や
多糖類分解酵素(例えば、アミラーゼ、セルラーゼ及びテキストラナーゼ)等を
併用することもできる。さらに、パパイン、プロメライン等の植物由来の蛋白質
分解酵素を併用しても良い。但し、植物由来の蛋白質分解酵素はシスティンやチ
オ尿素等の還元剤が必要であり、その還元剤が空気中の酸素により酸化されやす
いため、併用の際には酵素活性が低下し易いことを考慮すべきである。
本発明の洗浄剤に用いられる酵素の使用量は、0.01〜50W/V%の濃度
が適当であり、好ましくは0.05〜30W/V%である。0.01W/V%未
満では、後述のプロピレングリコール及びホウ砂の作用があっても充分な酵素活
性の安定性が得られず、また50W/V%を超えても安定性の顕著な上昇は認め
られない。

本発明の洗浄剤に含有されるプロピレングリコールは、前述した酵素活性を安
定化するために必須である。プロピレングリコールに、例えばグリセリン、エチ
レングリコール、ポリエチレングリコール、ソルビトール等を1種又は2種以上
組み合わせて用いることができる。プロピレングリコールの濃度は、5〜50W
/V%が適当であり、好ましくは10〜40W/V%である。5W/V%未満で
は、酵素活性の安定化が充分に得られず、また50W/V%を超えても酵素活性
の安定化作用に顕著な上昇は認められない。

本発明の洗浄液は、さらに、酵素の安定化剤としてホウ砂を含有する。プロピ
レングリコールとホウ砂とは、5W/V%以上のプロピレングリコールとプロピ

本発明の洗浄剤では、プロピレングリコールとホウ砂以外に、ゼラチン、カゼ
イン、エタノール等の他の安定化剤を併用することもでき、プロピレングリコー

ン及びカゼインのそれぞれの濃度は高い程よいが、ゼラチンは高濃度で液状にな
りにくいので1〜10W/V%が適している。カゼインは更にそれ以上の高濃度

大きくなるため、0.5〜10W/V%の濃度範囲であることが適当である。好

を持つものがあるので、その様な酵素を使用する場合には添加することができな
い。

本発明の洗浄液は、コンタクトレンズを洗浄する際に適宜希釈液で希釈して用
いる。希釈液は、例えば、精製水、生理食塩水、専用の希釈液等を挙げることが
できる。
ここで、洗浄液を希釈液により希釈するのは使用直前がよい。その理由は、前
記洗浄液に含有される酵素は、希釈されると、洗浄液中の酵素活性を経時的に安
定にする成分の濃度が低下することにより、その安定性が低下するからである。
それ故、コンタクトレンズの洗浄を行う際、できるだけその直前に、例えばコン
タトレンズケースのような容器中で本発明の洗浄液と希釈液とを混合することに
より希釈した方がよい。
本発明の洗浄液は、例えば、15倍〜30倍程度に希釈して用いることができ
る。

さらに前記希釈液は、界面活性剤や着色剤等を含むものであることもできる。
希釈液に着色剤を添加することで、洗浄液を他のもの例えば保存液と識別するこ
とができ、かつレンズ洗浄後のすすぎが充分か否かを判断することもできる。ま
た着色剤は、レンズに付着している汚染物を着色するため、汚染物が残留してい
ることを示す指標としても作用する。
着色剤としては、水溶性の色素でかつレンズ素材に吸着したり、反応したりし
ない物であって人体に安全な物が望ましい。例えば、食用色素の類が使用可能で
あるが、特にエリスロシン(赤色3号)、タートラジン(黄色4号)、ブリリア
ントブルー(青色1号)等が適当である。その他に例えばフルオレセインやその
塩類等も使用できる。着色剤の使用量は、コンタクトレンズを浸漬する時におい
て0.0001〜5W/V%となる濃度が適当であり、好ましくは、0.001
〜2W/V%である。0.0001W/V%未満の濃度では、洗浄液としての識
別が難しく、5W/V%を超えても識別能の向上は生じない。着色剤は少なくと

着色剤よる色が変化を生じる場合、すなわち色の安定性が維持できない場合があ
る。例えば、赤色系の色素の関しては、ローズベルガルは酸に対して不安定であ
り、またニューコクシンはアルカリに対して不安定である。従って、着色剤は、
それを含有する希釈液の液性及び洗浄液に含有される酵素の至適pHにより適宣
選択される。

また、希釈液に添加することができる界面活性剤としては、特に限定はされな
いが、非イオン系界面活性剤、陰イオン系界面活性剤が有効であり、両者を組み
合わせて併用してもよい。非イオン系界面活性剤としては、例えばポリオキシエ
チレン・ポリオキシプロピレン・ブロックポリマー、ポリオキシエチレン・オク
チルフェニルエーテル、アルキル・アルキロールアミド、グリセリン脂肪酸エス
テル・ポリオキシエチレン・アルキルエーテル等を挙げることができる。
また、陰イオン系界面活性剤としては、ドデシル硫酸ナトリウム、ドデシルベン
ゼンスルフォン酸ナトリウム、アルキル硫酸エーテル、ジオクチル・スルホコハ
ク酸ナトリウム等を挙けることができる。
これらの界面活性剤の化学的洗浄力は、前述の酵素の有する生化学的洗浄効果
と相乗的に作用し、洗浄力をさらに向上させることができる。特に脂質の汚れが
多量に付着しているコンタクトレンズに対しては、界面活性剤を添加した洗浄液
は極めて有効に作用する。界面活性剤の含有率としては、レンズを浸漬する時に
おいて、0.05〜15W/V%となる濃度が適当であり、好ましくは、0.1
〜10W/V%である。0.05W/V%未満では前述の界面活性剤の作用が発
揮されず、15W/V%を超えても、界面活性剤の洗浄力の顕著な上昇は認めら
れない。

防腐剤としては、ソルビン酸塩、サリチル酸塩、パラベン類、グルコン酸クロ
ルヘキシジン等が挙げられるが、洗浄液に菌が増殖するのを防止し、希釈液が長
時間保存できるようにするために用いられる。防腐剤の含有量としては、0.0
01〜1W/V%が適当であり、好ましくは0.005〜0.5W/V%である
。pH調整剤としては、酸(たとえば塩酸、酢酸、クエン酸等)やアルカリ(例
えば水酸化ナトリウム、クエン酸ナトリウム等)を単独で用いてもよいが、好ま

化剤が挙げられる。またその他の無機塩類を用いてもよい。なお、希釈液のpH
は、希釈される洗浄液に含まれる酵素の至適pH及び着色剤の安定が得られるに
適したpHを充分に考慮して調整される。
金属封鎖剤としては、エチレンジアミン四酢酸ニナトリウム(EDTA−2N
a)等が一般的に用いられる。但し、酵素の中には活性中心に金属を持つものが
あるので、その様な酵素を含む洗浄液を希釈するために使用する場合添加するこ
とができない。

40℃で1ヵ月保管した実施例2と比較例2を用いて洗浄力試験を行った。レ
ンズはホーヤ製ソフトコンタクトレンズ(商品名:ホーヤソフト/T40)を用
いた。汚れ液を下記配合により調整し、室温にて3時間レンズを浸漬し乾燥させ
たものを汚れレンズとした。

せた後濁度を測定し、下式により濁度回復率を求めた。濁度の測定には日本電色
製濁度計NDH−1001DPを用いた。

以上の結果から、本発明で得られた洗浄剤は充分な活性安定性を有しているた
め満足できる洗浄力を有しているが、比較例では活性安定性が劣っているため、
洗浄力が低下していることが確認された。

活性の安定性を有しているため、長期にわたり優れた洗浄効果を提供できるとい
