本開示は、一般に、医学、感染症および免疫学の分野に関する。さらに具体的には、本開示は、エンテロウイルス(enterovirus)D68に結合するヒト抗体、ならびにそのような抗体を使用してエンテロウイルスD68感染を診断および治療する方法に関する。
エンテロウイルス-D68(EV-D68)は、ピコルナウイルス(Picornaviridae)科エンテロウイルス属のポジティブセンス一本鎖RNAウイルスである。この属内の他の一般的なヒト病原体には、ポリオウイルス(poliovirus)、エコーウイルス(echovirus)、コクサッキーウイルス(coxsackievirus)、ライノウイルス(rhinovirus)、およびエンテロウイルス-A71などの他の番号が付けられたエンテロウイルスが含まれる(Zell et al.,2017)。これらのウイルスは、構造的および遺伝的に類似しているが、新生児敗血症、心筋炎、灰白髄炎、髄膜炎、気道感染および手足口病を含む多種多様な小児疾患を引き起こす。EV-D68は、ライノウイルスと同様に、主に、酸に不安定であり、33℃で最もよく複製する呼吸器病原体である(Oberste et al.,2004)。これらの類似性は、EV-D68の株がライノウイルス87として最初に報告されたことさえあるような類似性である(Blomqvist et al.,2002)。
したがって、本開示によれば、対象のエンテロウイルスD68(EV-D68)感染を検出する方法であって、(a)該対象から得られた試料と、それぞれ表3および表4のクローンと組にした(clone-paired)重鎖CDR配列および軽鎖CDR配列を有する抗体または抗体断片とを接触させる工程、ならびに(b)該抗体または抗体断片を該試料中のEV-D68抗原に結合させることによって、該試料中のEV-D68を検出する工程を含む方法が提供される。試料は、体液、例えば、血液、痰、涙液、唾液、粘膜または血清、精液、子宮頸部分泌物または膣分泌物、羊水、胎盤組織、尿、滲出液、濾出液、組織擦過物、呼吸器飛沫またはエアロゾル、糞便などであり得る。検出は、例えば、ELISA、RIA、ラテラルフローアッセイ、ウエスタンブロットなどを含み得る。方法は、工程(a)および(b)を再度行い、第1のアッセイと比較してEV-D68抗原レベルの変化を決定する工程をさらに含み得る。
上述のように、エンテロウイルスD68(EV-D68)は、世界中で主に小児の呼吸器疾患のアウトブレイクを引き起こし得る。急性弛緩性脊髄炎、灰白髄炎様神経筋脱力症候群のクラスターは、EV-D68の呼吸器アウトブレイクと同時に起こることが多い。血清疫学的試験では、2~5歳を超えるほぼすべての人の血清が抗EV-D68中和抗体を含有することが見出されており、EV-D68が小児期の遍在性病原体であることを示唆している。ただし、体液性免疫応答が対象とするウイルスエピトープの知識は、関連ウイルスの以前の試験から推測されるにすぎない。本明細書において、本発明者らは、一部が強力に中和する、EV-D68に対する64個のヒトモノクローナル抗体を生成した。これらは、この病原体に対する最初のヒトmAbであると考えられ、本発明者らは、EV-D68感染の検出、治療および予防にそれらを使用することを提案する。本開示のこれらおよび他の局面は、以下に詳細に説明される。
エンテロウイルス68(EV68、EV-D68、HEV68)は、エンテロウイルスであるピコルナウイルス科のメンバーである。エンテロウイルス68は、1962年にカリフォルニア州で初めて単離され、かつては希少と考えられたが、21世紀には世界的に急増傾向にある。急性弛緩性脊髄炎と呼ばれるポリオ様障害を引き起こすことが疑われている。
血清抗体ウイルス中和能の測定値は、ポリクローナル抗体レパートリー全体の活性を反映する。ただし、ウイルスに対する体液性応答を完全に理解するためには、個々の抗体が結合する特異的ウイルスエピトープを定義し、特異的エピトープへの抗体結合が疾患を防御するかどうかを決定することが必要である。かつて、ライノウイルス-B14に対して惹起されたマウスモノクローナル抗体の試験によって、エンテロウイルス属のウイルスに対する4つの中和免疫エピトープ(Nim)が同定された(Rossmann et al.,1985)。EV-D68特異的モノクローナル抗体エピトープの試験はこれまで行われていない。ただし、EV-D68の結晶構造の決定から、ライノウイルスとの構造相同性の観察によって、EV-D68ビリオン上の4つのNimの可能性のある位置が示唆された(Liu et al.,2015a)。米国(Zhang et al.,2015)または日本(Imamura et al.,2014)からのEV-D68の近年のヒト単離株の表面タンパク質のアミノ酸配列とFermon参照ウイルス株のものとを比較する試験では、Nimおよび近くの隣接残基における変異が、特に、結晶構造が無秩序である、キャプシドタンパク質VP1のBCループおよびDEループに生じていることが示唆されている(Liu et al.,2015a)。これらのVP1多型は、既存の体液性免疫を回避することによってウイルスの病原性の増大に寄与する可能性がある。マウスモノクローナル抗体は市販されているが(供給元には、GeneTex、ThermoFisherおよびSigmaが含まれる)、免疫原として使用された特異的ウイルス表面タンパク質を列挙することを除けば、これらの抗体のエピトープに関する情報は入手できない。注目すべきことに、これらの市販の抗体のいくつかは、他のエンテロウイルス属ウイルスの免疫原から生成され、これは、EV-D68に結合するヘテロタイプ抗体の能力を示唆している。この可能性をさらに裏付けるように、2つのマウスmAbが40の異なるエンテロウイルス種に交差反応したが、興味深いことに、EV-D68は、それらが反応しなかった試験された唯一のウイルスであった(Miao et al.,2009)。他のタイプのエンテロウイルスによって誘導される抗体によるEV-D68のヘテロタイプ分子認識は、幼児期には、観察された普遍的なEV-D68血清有病率に寄与する可能性がある。EV-D68に対するヒト抗体エピトープの優れた特異性と、そのような抗体のタイプ特異性または幅広さとを理解するには、自然感染によって誘導されるクローン化ヒトモノクローナル抗体を用いた試験が必要であろう。
インビトロで抗体によるウイルスの中和を測定することは、典型的には、抗体の抗ウイルス機能を特性評価するための最も迅速で再現性のある方法である。ただし、この試験様式は、補体活性化、または細胞表面Fc受容体との係合など、インビボでのみ作用する、抗体のFc部分によって媒介されるエフェクター機能を測定しない。その結果、インビトロ中和能の大きさは、インビボで観察されるウイルス複製の阻害または疾患に対する保護のレベルと完全に相関しない可能性がある((Lu et al.,2018)に概説される)。抗体のFc媒介エフェクター機能の多くは、完全な保護効果を媒介するために自然免疫系または適応免疫系のFc受容体担持細胞を必要とする。前臨床試験では、体液性免疫の効果を評価するのに感染の小動物モデルが必要である。
EV-D68感染の現在のマウス試験の主な限界には、麻痺および死を誘導するための非生理学的な接種経路の使用、およびヒトではEV-D68感染の主な症状である呼吸器疾患の欠如がある。コットンラット(アラゲコトンラット(Sigmodon hispidus))(Patel et al.,2016)およびフェレット(ムステラ・プトリウス・フロ(Mustela putorius furo))(Zheng et al.,2017a)の両方について、各々の単一の試験では、EV-D68の鼻腔内接種が鼻および肺内でウイルスの複製をもたらし、肺内で炎症誘発性自然免疫分子転写物の増加を誘導することが実証された。接種したフェレットの約25%では、上気道感染(咳、鼻汁および鼻乾燥)と一致する臨床症状も発現した。試験では、筋肉または中枢神経系組織内のEV-D68の存在について評価されておらず、明らかな四肢の衰弱または麻痺も認められなかった。以下にさらに記載される所見では、コットンラットのワクチン接種は、いくつかのレベルの既存の体液性免疫が呼吸器感染に対して完全に防御せず、実際に有害であり得ることを示した(Patel et al.,2016)。
EV-D68の実験的ワクチン候補は未だ臨床開発に入っていないが、酵母細胞(Zhang et al.,2018a)もしくは昆虫細胞(Dai et al.,2018)内で作製されたウイルス様粒子(VLP)ワクチン、またはβ-プロピオラクトン不活化EV-D68(Zhang et al.,2018)のいずれかをマウスに接種した初期試験は有望性を示している。VLPワクチンは、その後の致死的な腹腔内攻撃感染からマウスを保護する抗体応答を誘導した。これらのワクチン候補の各々では、ナイーブマウスへのワクチン免疫血清の受動的移入は、致死的な腹腔内攻撃感染からの保護に十分であった。VLPベースのワクチンの存在し得る欠点には、これらの合成構築物が粒子の表面上のあらゆる立体配座感受性構造を完全に再現するかどうかという不確実性がある。他のウイルスに対する多くの強力なヒトウイルス中和抗体は、厳しい立体配座制約を有する複雑な四次抗原部位を認識する(Crowe,JE,2017)。VLP上のヒト抗体エピトープの完全性は、これらのエピトープが未知であるため、現在評価することができない。これらのマウスワクチン試験の有望性をさらに抑えるものには、生きたEV-D68またはUV不活化EV-D68のいずれかを筋肉内接種し、その後、同種の生ウイルスを鼻腔内負荷したコットンラットにおける所見があり、この所見では、ナイーブ動物の感染と比較して、炎症の増強が肺に見られた(Patel et al.,2016)。具体的には、UV不活化ウイルスワクチン接種は、肺または鼻腔内でウイルス複製を制限せず、得られたサイトカインシグネチャーを、生ウイルスワクチン接種からの免疫で見られるバランスのとれたTh1表現型およびTh2表現型ではなく、Th2表現型に向けてゆがめた。呼吸器合胞体ウイルス(Karron,R,2018)および麻疹(Strebel et al.,2018)に対する不活化ウイルスワクチン候補では、炎症および疾患の増強が認められている。これらの矛盾した所見を考慮すると、EV-D68ワクチン候補によって引き起こされる防御または増強の免疫相関をさらに慎重に試験することが必要である。
「単離された抗体」は、その天然環境の成分から分離および/または回収されたものである。その天然環境の汚染成分は、抗体の診断的使用または治療的使用を妨げる材料であり、酵素、ホルモン、および他のタンパク質性溶質または非タンパク質性溶質を含み得る。特定の態様では、抗体は、(1)ローリー法によって決定した場合、抗体の95重量%超、最も具体的には99重量%超まで、(2)スピニングカップシークエネーターを使用してN末端もしくは内部アミノ酸配列の少なくとも15残基を得るのに十分な程度まで、または(3)クーマシーブルーもしくは銀染色を使用した還元条件もしくは非還元条件下でのSDS-PAGEによって均一になるまで精製される。単離された抗体は、抗体の天然環境の少なくとも1つの成分が存在しないため、組換え細胞内にインサイチューで抗体を含む。ただし、通常、単離された抗体は、少なくとも1つの精製工程によって調製される。
EV-D68に結合するモノクローナル抗体は、いくつかの用途を有することが理解される。これらには、EV-D68感染の検出および診断、ならびにEV-D68感染の治療に使用するための診断キットの製造が含まれる。これらの状況では、そのような抗体を診断剤もしくは治療剤に連結するか、それらを競合アッセイにおいて捕捉剤もしくは競合剤として使用するか、またはそれらに追加の薬剤を付着させることなくそれらを個別に使用してもよい。抗体は、以下にさらに説明されるように、変異または修飾され得る。抗体を調製し、特性評価するための方法は、当技術分野において周知である(例えば、Antibodies:A Laboratory Manual,Cold Spring Harbor Laboratory,1988;米国特許第4,196,265号を参照)。
本開示による抗体は、第1の例では、それらの結合特異性によって定義され得る。当業者に周知の技術を使用して所与の抗体の結合特異性/親和性を評価することによって、当業者であれば、そのような抗体が本特許請求の範囲内に入るかどうかを決定することができる。例えば、所与の抗体が結合するエピトープは、抗原分子内に位置する3個以上(例えば、3、4、5、6、7、8、9、10、11、12、13、14、15、16、17、18、19、20)のアミノ酸の単一の連続配列からなり得る(例えば、ドメイン内の線状エピトープ)。あるいは、エピトープは、抗原分子内に位置する複数の非連続アミノ酸(またはアミノ酸配列)からなり得る(例えば、立体構造エピトープ)。
様々な態様では、発現の改善、交差反応性の改善またはオフターゲット結合の減少などの様々な理由のために、同定された抗体の配列を操作することを選択することができる。修飾された抗体は、標準的な分子生物学的技術による発現、またはポリペプチドの化学合成を含む、当業者に公知の任意の技術によって作製され得る。組換え発現のための方法は、この文献中の他の箇所で扱われる。以下は、抗体操作のための関連する目標技術の一般的な説明である。

抗体薬物コンジュゲート、すなわちADCは、感染症を有する人々を治療するための標的療法として設計された新しいクラスの非常に強力な生物医薬品である。ADCは、不安定な結合を有する安定な化学リンカーを介して、生物学的に活性な細胞傷害性/抗ウイルス性のペイロードまたは薬物に連結された抗体(全mAbまたは抗体断片、例えば、一本鎖可変断片、すなわちscFv)から構成される複合分子である。抗体薬物コンジュゲートは、バイオコンジュゲートおよびイムノコンジュゲートの例である。
二重特異性T細胞エンゲージャー(BiTE)は、抗癌薬として使用するために研究されている人工二重特異性モノクローナル抗体のクラスである。それらは、感染細胞に対する宿主の免疫系、さらに具体的にはT細胞の細胞傷害活性を導く。BiTEは、Micromet AGの登録商標である。
特定の態様では、抗体は、細胞内での作用に適した組換え抗体であり、そのような抗体は「イントラボディ」として知られている。これらの抗体は、様々な機構によって、例えば、細胞内タンパク質輸送を変化させること、酵素機能を妨害すること、およびタンパク質-タンパク質相互作用またはタンパク質-DNA相互作用を遮断することによって標的機能を妨害し得る。それらの構造は、多くの点で、上述の一本鎖抗体および単一ドメイン抗体の構造を模倣するか、またはそれらと並行する。実際、単一転写物/一本鎖は、標的細胞内の細胞内発現を可能にし、細胞膜を通過するタンパク質をさらに実現可能にする重要な特徴である。ただし、追加の特徴が必要とされる。

A. 製剤および投与
本開示は、抗EV-D68抗体およびそれを生成するための抗原を含む薬学的組成物を提供する。そのような組成物は、予防的または治療的に有効な量の抗体もしくはその断片、またはペプチド免疫原と、薬学的に許容される担体とを含む。特定の態様では、用語「薬学的に許容される」は、連邦政府もしくは州政府の規制機関によって承認されていること、または動物、さらに具体的にはヒトに使用するために米国薬局方もしくは他の一般に認識されている薬局方に記載されていることを意味する。用語「担体」は、治療薬とともに投与される希釈剤、賦形剤またはビヒクルを指す。そのような薬学的担体は、滅菌液体、例えば、水および油、例えば、石油、動物起源のもの、植物起源のもの、または合成起源のもの、例えば、落花生油、大豆油、鉱油、ゴマ油などであり得る。水は、薬学的組成物が静脈内投与される場合、特定の担体である。生理食塩水ならびにデキストロース水溶液およびグリセロール水溶液も、特に注射液用の液体担体として使用することができる。他の好適な薬学的賦形剤には、デンプン、グルコース、ラクトース、スクロース、ゼラチン、麦芽、米、小麦粉、チョーク、シリカゲル、ステアリン酸ナトリウム、モノステアリン酸グリセロール、タルク、塩化ナトリウム、乾燥スキムミルク、グリセロール、プロピレン、グリコール、水、エタノールなどが含まれる。
抗体依存性細胞媒介性細胞傷害(ADCC)は、免疫エフェクター細胞による抗体被覆標的細胞の溶解をもたらす免疫機構である。標的細胞は、Fc領域を含む抗体またはその断片が、一般にFc領域のN末端であるタンパク質部分を介して特異的に結合する細胞である。「抗体依存性細胞媒介性細胞傷害(ADCC)が増加/減少した抗体」とは、当業者に公知の任意の好適な方法によって決定した場合、ADCCが増加/減少した抗体を意味する。
補体依存性細胞傷害(CDC)は、補体系の機能である。CDCは、免疫系の抗体または細胞が関与することなく病原体の膜を損傷することによって病原体を死滅させる、免疫系におけるプロセスである。3つの主要なプロセスがある。3つはいずれも、致死的なコロイド浸透圧膨潤、すなわちCDCを引き起こす1つまたは複数の膜攻撃複合体(MAC)を病原体に挿入する。これは、それによって抗体または抗体断片が抗ウイルス効果を有する機構の1つである。
本開示の抗体は、抗体コンジュゲートを形成するために少なくとも1つの作用物質に連結され得る。診断剤または治療剤としての抗体分子の効果を高めるために、少なくとも1つの所望の分子または部分を連結または共有結合または複合化することが従来から行われている。そのような分子または部分は、限定されることなく、少なくとも1つのエフェクター分子またはレポーター分子であり得る。エフェクター分子は、所望の活性、例えば細胞傷害活性を有する分子を含む。抗体に付着したエフェクター分子の非限定的な例には、毒素、抗腫瘍剤、治療用酵素、放射性核種、抗ウイルス剤、キレート剤、サイトカイン、成長因子、およびオリゴヌクレオチドまたはポリヌクレオチドが挙げられる。対照的に、レポーター分子は、アッセイを使用して検出され得る任意の部分として定義される。抗体にコンジュゲートされたレポーター分子の非限定的な例には、酵素、放射性標識、ハプテン、蛍光標識、リン光分子、化学発光分子、発色団、光親和性分子、着色粒子または着色リガンド、例えばビオチンが挙げられる。
なおさらなる態様では、本開示は、EV-D68およびその関連抗原の結合、精製、除去、定量および他の一般的な検出のための免疫検出方法に関する。そのような方法は従来の意味で適用され得るが、別の用途は、本開示による抗体を使用してウイルス内の抗原の量または完全性(すなわち、長期安定性)を評価することができる、ワクチンおよび他のウイルスストックの品質管理およびモニタリングである。あるいは、方法は、適切な/所望の反応性プロファイルについて様々な抗体をスクリーニングするために使用され得る。
イムノアッセイは、その最も単純かつ直接的な意味で、結合アッセイである。特定の好ましいイムノアッセイは、当技術分野において公知の様々なタイプの酵素結合免疫吸着測定法(ELISA)およびラジオイムノアッセイ(RIA)である。組織切片を使用した免疫組織化学的検出も特に有用である。ただし、検出はそのような技術に限定されず、ウエスタンブロッティング、ドットブロッティング、FACS分析なども使用され得ることは容易に理解されるであろう。
ウエスタンブロット(あるいは、タンパク質イムノブロット)は、組織ホモジネートまたは組織抽出物の所与の試料中の特定のタンパク質を検出するために使用される分析技術である。それは、ポリペプチドの長さ(変性条件)によって、またはタンパク質の3-D構造(天然/非変性条件)によって、天然または変性タンパク質を分離するためにゲル電気泳動を使用する。次いで、タンパク質はメンブレン(典型的にはニトロセルロースまたはPVDF)に転写され、そこで標的タンパク質に特異的な抗体を使用してプローブ(検出)される。
ラテラルフローイムノクロマトグラフィーアッセイとしても知られているラテラルフローアッセイは、専用の高価な装置を必要とせずに試料(マトリックス)中の標的分析物の存在(または非存在)を検出することを意図した単純な装置であるが、読み取り装置によって補助される多くの実験室ベースの用途が存在する。典型的には、これらの検査は、家庭用検査、ポイントオブケア検査、または実験室での使用のいずれかのための低資源医療診断として使用される。広く普及している周知の用途は、家庭用妊娠検査である。
本開示の抗体はまた、免疫組織化学(IHC)による試験のために調製された新鮮凍結組織ブロックおよび/またはホルマリン固定パラフィン包埋組織ブロックの両方と併せて使用され得る。これらの微粒子標本から組織ブロックを調製する方法は、様々な予後因子の以前のIHC試験で首尾よく使用されており、当業者に周知である(Brown et al.,1990;Abbondanzo et al.,1990;Allred et al.,1990)。

材料および方法

試験デザイン。本発明者らは、EV-D68感染に応答してヒトが産生することができるいずれかの抗体を同定しようと試みるためにこの試験をデザインした。したがって、本発明者らは、間接ELISAスクリーニングに生ウイルス単離株を使用して、EV-D68結合抗体を分泌するB細胞を同定し、次いで、それらのB細胞をミエローマ細胞と電気融合して、モノクローナル抗体分泌ハイブリドーマを作製した。次いで、本発明者らは、CCID50、ELISAおよびクライオEMに基づく技術を使用して、インビトロでこれらの個々のmAbの中和および結合特性を特性評価した。本発明者らはインビボ実験を追求して、ヒトを対象とした予防剤および/または治療剤としてのmAb EV68-228の開発を裏付ける前臨床データを生成した。この目的のために、本発明者らは、理論的利益に基づいてAFMを用いてヒトを治療するために広く使用されているヒトIVIGと比較して、EV-D68感染からマウスを保護する際のmAb EV68-228の効果を試験したが、このIVIG治療はこれまで効果的であることは証明されていない。AG129マウス感染モデルの利点は、本発明者らが、感染の呼吸器モデルおよび神経学的モデルの両方を対象として抗体処置の効果を測定することができたことである。
を用いて、ウイルスゲノムのP1領域を包含する3,080bpアンプリコンを作製した。本発明者らは、Pacific Biosciences(PacBio)次世代配列決定プラットフォームを使用して、3つの反復の各々の配列を生成した。各配列決定ランの2,000個のリードを使用して、各変異が観察されたリードの割合を定量した。陰性対照選択mAb選択から得られた全リードの野生型コンセンサス配列と比較して、変異を決定した。
書面によるインフォームドコンセントが得られた後、米国の2014年のアウトブレイク中にEV-D68気道感染が以前に実証されている対象12例が献血し、その中から、本発明者らは末梢血単核球(PBMC)を単離した。対象は、感染時に12~15歳、採血時に16~18歳であった。各対象は、EV-D68関連呼吸器疾患の病歴を有し、AFMの症状を有しなかった(表E)。回収したPBMCをエプスタイン・バーウイルスの接種によってインビトロで形質転換して、抗体を分泌するメモリーB細胞由来リンパ芽球様細胞株(LCL)を生成した。間接ELISAでLCL培養上清を使用して、EV-D68反応性IgGの存在についてスクリーニングした。本発明者らは、2014年の臨床単離株から生成された、EV-D68の実験室増殖生ウイルス調製物に結合したが、同様に調製した非感染細胞上清には結合しなかった抗体を用いて培養物を選択した。EV-D68特異的抗体を分泌するLCLと非分泌ミエローマ細胞株との電気融合後、得られたハイブリドーマ細胞を単一細胞ソーティングして、完全ヒトmAbを分泌するクローンハイブリドーマを生成した(表F)。

- エンテロウイルスD68(EV-D68)に感染した対象を治療するか、またはEV-D68に罹患するリスクがある対象の感染の可能性を低減する方法であって、それぞれ表3および表4のクローンと組にした(clone-paired)重鎖CDR配列および軽鎖CDR配列を有する抗体または抗体断片を前記対象に送達する工程を含む、前記方法。

- 前記抗体または抗体断片が、感染前または感染後に投与される、請求項1~10のいずれか一項記載の方法。
- 前記治療が、肺感染および/もしくは神経感染を治療するか、または治療が、肺感染および/もしくは神経感染を低減する、請求項1~11のいずれか一項記載の方法。
- 送達する工程が、抗体もしくは抗体断片の投与、または抗体もしくは抗体断片をコードするRNA配列もしくはDNA配列もしくはベクターによる遺伝的送達を含む、請求項1~12のいずれか一項記載の方法。
- 対象のエンテロウイルスD68(EV-D68)感染を検出する方法であって、以下の工程を含む方法:
(a)前記対象から得られた試料と、それぞれ表3および表4のクローンと組にした重鎖CDR配列および軽鎖CDR配列を有する抗体または抗体断片とを接触させる工程;ならびに

- 検出がELISA、RIA、ラテラルフローアッセイまたはウエスタンブロットを含む、請求項14~16のいずれか一項記載の方法。
- 工程(a)および(b)を再度行い、第1のアッセイと比較してEV-D68抗原レベルの変化を決定する工程をさらに含む、請求項14~17のいずれか一項記載の方法。
- 抗体または抗体断片が、表1に記載のクローンと組にした可変配列によってコードされる、請求項14~18のいずれか一項記載の方法。
- 前記抗体または抗体断片が、表1に記載のクローンと組にした可変配列に対して70%、80%または90%の同一性を有する軽鎖可変配列および重鎖可変配列によってコードされる、請求項14~18のいずれか一項記載の方法。
