FuGene(Promega)に関して推奨されているように、簡潔に説明すると、トランスフェクションの前日に、24ウェルプレート(平底ウェル、組織培養24ウェルNunc(商標)プレート、ThermoFisher)の半分に、8×104個の細胞/ウェルの密度でFlpIn HEK293細胞を蒔き、もう半分に、完全増殖培地(DMEM+10%のウシ胎仔血清)500μl中に4×104個の細胞/ウェルの密度で蒔いた。2重での単一タンパク質トランスフェクションのために、pcDNA5-FRTベクター0.11ug及びpOG44 DNA(ThermoFisher)0.99ugを、滅菌脱イオン水 総量52μlに添加した。3:1のFugene対DNA比を使用して、FuGene 3.3μlを注意深く添加した。このことを、ピペットの先端がマイクロ遠心分離チューブの側面に触れないように実現した。この溶液を20秒にわたりボルテックスし、スピンダウンさせてマイクロ遠心分離チューブの底部中の全ての溶液を回収した。室温での10分間のインキュベーション後、FlpIn HEK293細胞(より高い密度のもの、及びより低い細胞密度のもの)の1つのウェル当たり複合体25μlを添加し、十分に混合した。細胞を、37℃5%CO2加湿インキュベーターに戻し、48時間後に、細胞を、選択のために50μg/mlのハイグロマイシン(PBS中のThermoFisherハイグロマイシンB)を含む完全培地が入った6ウェルプレートに分けた。1ヶ月後、成功裏にトランスフェクトされた細胞はウェル中で遺伝子座を形成し、この遺伝子座は、典型的には1Lのスピナーフラスコ又は5LのWAVEバイオリアクタに拡大され得、2週間後に培養上清を回収した。
細胞上清を回収し、15分にわたり4000×gで遠心分離して細胞残屑を除去した。上清を0.45μmフィルタ(Sartorius)に通し、精製のための0.1%のアジ化ナトリウム(Sigma)と共に4℃で保存した。CD23-IgGFc融合タンパク質を、AKTA Primeシステム(GE Healthcare)を使用するHiTrap Protein-G HPカラム(GE Healthcare)5mlによる親和性クロマトグラフィーにより精製した。このカラムを、5カラム容積(CV)の洗浄緩衝液(PBS、pH7.4)で平衡化した。このカラムに、ろ過した上清を2ml/分の流速でロードし、このカラムを10CVの洗浄緩衝液で洗浄した。CD23-IgGFc融合タンパク質を0.1Mのグリシン-HCl, pH 2.5で溶出させ、画分2.5mlを、中和のために1MのTris-HCl pH8.6 0.5mlが入ったチューブに回収した。
PBS pH7.4にて0.75ml/分の流速でSuperdex(商標)200 10/300 GLカラム(GE Healthcare)を使用するGilson HPLCシステムで、サイズ排除クロマトグラフィーを実施した。このサイズ排除クロマトグラフィー分析により、凝集がないことが示され、且つ親和性カラムにより精製された生成物は、予測サイズ(約100KDa)の単分散分子からなることを確認した。
好塩基球脱顆粒への生物製剤抗IgEの効果の評価
脱顆粒アッセイを使用して、好塩基球及び肥満細胞等のIgE感受性エフェクター細胞が、細胞質内の顆粒中に保持された細胞内メディエーターを放出する傾向を評価した。エフェクター細胞の表面上のアレルゲン特異的IgEが、その特異的なアレルゲンに環境中で遭遇した場合には、高親和性IgE受容体FcεRIとの間で架橋が起こり、下流のシグナル伝達事象が活性化される。これにより、炎症性メディエーターを含む細胞内顆粒が局所環境中に放出され、この放出により典型的なアレルギー反応が起きる。この反応を抗IgE生物製剤が阻害するか又は増強する可能性を、一連の改変好塩基球脱顆粒アッセイで評価する。
好塩基球脱顆粒アッセイ
ヒト四量体(αβγ2)高親和性IgE受容体FcεRIを安定して発現するラット好塩基球性白血病細胞系統RBL-SX38細胞[Dibbern, DA他、J Immunol Methods 2003; 274: 37-45]、(Prof. J-P. Kinet, Harvard University, Boston, MAからの寄贈品)を、様々なIgE媒介性トリガーにより刺激して、β-ヘキソサミニダーゼの放出により測定される脱顆粒を評価した。使用する方法論は、本質的には、Rudman他、Clin Exp Allergy 2011年、41(10): 1400-1413、及びWeigand他、1996年、J. Immunol、157:221-230で説明されているものであり、本明細書で簡単に説明する。
この生物製剤構築物を、FcεRI IgE受容体に既に結合しているIgEの架橋を介してIgE媒介性脱顆粒事象を増強する能力に関して試験した。
RBL-SX38好塩基球性細胞を、上記材料及び方法(好塩基球脱顆粒アッセイ)セクションで説明したように調製して48時間の期間にわたりIgEを負荷した。IgEを負荷した細胞に、生物製剤を4μM~0.016nMの連続希釈範囲で添加し、1時間にわたりインキュベートした。次いで、説明したように上清の試料を採取して処理して、細胞の脱顆粒のシグナルとして放出されるβ-ヘキソサミニダーゼの濃度を評価した。
抗IgEと架橋する対照との1時間のインキュベーション後、RBL-SX38好塩基球性細胞を刺激して、用量依存的反応でβ-ヘキソサミニダーゼを放出させた。これに対して、(同一の実験条件を使用して)生物製剤の濃度を増加させると、いかなるβ-ヘキソサミニダーゼ放出の兆候も見られず、このことは、この生物製剤が単独で好塩基球の活性化又は脱顆粒を増強し得なかったことを示す(図3)。
この研究では、抗IgE生物製剤がIgEに結合し、高親和性IgE受容体FcεRIとの結合を防止し、それにより好塩基球性細胞系統RBL-SX38のIgE依存性脱顆粒を防止する可能性を探る。
RBL-SX38好塩基球性細胞を上記で説明したように播種し、一晩インキュベートした。翌日、200ng/mL(1nM)に設定されたIgEの標準濃度を含むプレミックス溶液を、生物製剤抗IgE構築物の増加する濃度と共に調製した。次いで、このプレミックス溶液を細胞に直ちに添加し、一晩インキュベートした後、上記の脱顆粒アッセイセクションで説明したように、ポリクローナル抗IgEによる刺激プロトコルに供した。
図4に示すデータは、抗IgE生物製剤が用量依存的にIgE媒介性脱顆粒を阻害し得たことを示しており、抗IgE生物製剤が高親和性IgE受容体FcεRIへのIgEの結合をブロックしていることと一致している。
この研究では、抗IgE生物製剤がIgEに結合し、高親和性IgE受容体FcεRIとの結合を防止する可能性を探る。
RBL-SX38好塩基球性細胞を上記で説明したように播種し、一晩インキュベートした。翌日、200ng/mL(1nM)に設定されたAlexaFluor-488標識IgEの標準濃度を含むプレミックス溶液を、生物製剤抗IgE構築物の増加する濃度と共に調製した。次いで、このプレミックス溶液を細胞に直ちに添加し、1時間にわたりインキュベートした後、2回洗浄し、分析のためにFACS緩衝液1mLに再懸濁させた。細胞をAttune NxT Acoustic Focusing Cytometer (Lasers: BRVX) (ThermoFisher)で分析し、データをFlowJoバージョン10.2で分析した。
図5に示すデータは、競合結合研究において、抗IgE生物製剤がIgEに結合して、RBL-SX38細胞上の高親和性IgE受容体FcεRIへのIgEの結合を用量依存的に防止し得たことを示す。生物製剤抗IgEの濃度の増加に伴い、表面FcεRIに結合し得たIgE分子は少なくなり、そのため、この分子が、IgEとその高親和性受容体との結合を阻害する能力が実証された。
IgEは、肥満細胞及び好塩基球の表面上のFcεRIに結合する。多価アレルゲンの存在下では、FcεRIに結合したIgEは、受容体を架橋して、細胞の活性化、及び脱顆粒反応による炎症性細胞メディエーターの放出を増強する。生物製剤抗IgEを、既に予め感作された好塩基球性細胞の脱顆粒反応を防止する能力に関して試験した。
RBL-SX38好塩基球性細胞を上記で説明したように播種し、一晩インキュベートした後、上記で説明したプロトコルに従って200ng/mLのIgE(1nM)を添加し、更に24時間にわたりインキュベートした。次いで、この細胞が入ったウェルに、増加する濃度の生物製剤抗IgE構築物を添加し、1時間にわたりインキュベートした後、上記の脱顆粒アッセイセクションで説明したように、5000ng/mLのポリクローナル抗IgEによる刺激プロトコルに供した。
図6に示すデータは、1時間以内に急速に、適度に過剰(>1nM)な場合でβ-ヘキソサミニダーゼ放出により測定した場合に抗IgE生物製剤がIgE媒介性脱顆粒を用量依存的に防止し得たことを示す。インキュベーション時間を更に長くしても、この構築物で観察された脱顆粒の阻害レベルにさらなる変化はなかった(データは示さない)。
リサイクル及び取り込み細胞アッセイ
材料及び方法:
HEK-mFcRn/β2m細胞及びHEK-hFcRn/β2m細胞の調製
HEK293F(ThermoFisher)細胞は、ヒト胚性腎細胞系統である。細胞を、DMEM+10%のウシ胎仔血清中で維持し、実施例1に従ってFugene(ThermoFisher)トランスフェクション試薬を使用して、マウス又はヒトのFcRn及びβ2mのいずれかで一過性にトランスフェクトし、約48時間後に使用可能であった。HUVEC、HepG2、CACO2、及びHMEC1等の他の細胞系統も、この方法で成功裏にトランスフェクトし得る(図示しない)。

このアッセイプロトコルを、図7に示す。
a. HEK293-FcRn/β2m細胞を播種し、コンフルエント(95~100%の培養密度)まで増殖させ、1つのウェル当たり7.5×105個を24ウェルプレート(Costar)に播種し、培養培地中で2日にわたり培養する。
b.培地を除去し、細胞を2回洗浄し、ハンクス平衡塩溶液(HBSS)緩衝液(pH7.4)中で1時間にわたり飢餓状態にした。
c.目的のタンパク質をHBSS(pH7.4又は6.0)で希釈し、細胞に添加し、4時間にわたりインキュベートして、抗体を取り込ませる。
d.培地を除去し、細胞を氷冷HBSS(pH7.4)で4回洗浄し、その後、新鮮で温かいHBSS(pH7.4)、又はFBSを含まず且つMEM非必須アミノ酸(ThermoFisher)が補充された増殖培地を添加した。
e.試料を新鮮で温かいHBSS(pH7.4)と共にインキュベートし、4時間又は一晩(4時間のインキュベーション)(これにより、リガンドの放出が可能となる)で回収した++。
f.細胞を氷冷HBSS(pH7.4)で徹底的に洗浄し、溶解させる+++。
g.回収した試料を、IgG又はIgEに特異的なELISAで分析する。
+培地を取り出し、溶液中に残存する構築物を読み取る。
++培地を吸引し、溶液/培地中に放出された構築物(リガンド)の量を決定するELISAにより読み取る。
+++細胞溶解物を、ELISAにより、細胞内に取り込まれた構築物のレベルに関して分析する。
全タンパク質溶解物を、プロテアーゼ阻害剤カクテル(Sigma- Aldrich)又は完全プロテアーゼ阻害剤錠剤(Roche)が補充されたCelLytic M細胞溶解試薬(Sigma-Aldrich)又はRIPA溶解緩衝液(ThermoFisher)を使用して得た。この混合物を、10分にわたり氷及び振盪機の上で細胞と共にインキュベートし、続いて10,000×gで15分にわたり遠心分離して細胞残屑を除去した。溶解物中に存在するIgG又はIgEの量の定量を、下記で説明するようにELISAにより行った。
細胞培養上清中のIgG-Fc濃度を、下記の方法を使用するELISAにより決定した。
a.最初に、捕捉抗体であるヤギ抗マウスIgG(Sigma)を、炭酸-炭酸水素緩衝液で1μg/mLの最終濃度まで希釈した。
b.次に、このコーティング溶液100μLを、MaxisorpTM 96ウェルプレートの各ウェルに添加し、4℃で一晩インキュベートした。
c.一晩のインキュベーション後、このウェルからコーティング溶液を除去し、各ウェルに、ブロッキング緩衝液である2%のSkim Milk/PBS+0.5%のTween(登録商標)20(PBS-T) 200μLを添加した。プレートを2時間にわたりインキュベートし、次いでウェルをPBS-T 250μLで2回洗浄した。
d.次に、IgG標準を、50%の培養培地(細胞培養培地と同一)及び50%のPBS-T/1%のSkim Milk (アッセイ緩衝液)で400ng/mLまで希釈し、各ウェルの最終体積が50μLになるように、2重で0.78ng/mLまでウェルプレート中において1:2で連続希釈した。
e.残りのウェルに、アッセイ緩衝液25μLと、細胞培養に由来する上清又は希釈された上清25μLとを入れた。
f.標準及び試料を2時間にわたりインキュベートした後、ウェルをPBS-T 250μLで4回洗浄した。
g.次に、二次抗体であるヤギ抗マウスIgG-HRP(ThermoFisher)をアッセイ緩衝液により1:1000で希釈し、この溶液50μLを各ウェルに添加した。2時間のインキュベーション期間後、ウェルをPBS-T 250μLで4回洗浄した。
h.次に、各ウェルに、OPD 5mgを1×Stable Peroxidase Substrate Buffer 10mLに希釈することにより調製した基質50μLを添加した。この基質を15分にわたりインキュベートし、この反応を、各ウェルへの1MのHCl 50μLの添加により停止させた。
i.各ウェルの吸光度を、492nmの吸光度及び650nmの基準波長減算を使用して、Flurostar Omega (BMG Labtech) Spectrophotometerを使用して決定した。標準曲線フィッティングを、標準曲線上の最低6点を使用する重み付けなしの4パラメータ曲線フィットにより、GraphPad Prism(著作権)ソフトウェアを使用して実施した(Findlay and Dillard 2007)。

IgEのみが細胞に取り込まれてリソソーム分解を受けるか又はFcRnによるか若しくは潜在的に同等のリサイクル及び回収経路によりリサイクルされる能力を、Grevy's他2018. Table 2(表3、表4)は、HEK293-mFcRn/β2mリサイクルアッセイにおいて、IgEを含まず生物製剤抗IgEのみの濃度の増加を評価したことを示す。この構築物は、0.01~5.00nMの生物製剤濃度では、生物製剤の93%超が、4時間のインキュベーション期間内に、トランスフェクトされたHEK293細胞により培地から取り込まれるという、FcRn飲食作用輸送機序による迅速な取り込みを示した。1nMのIgEの存在下では、1~2000nMの生物製剤抗IgEの存在下である場合に4時間のインキュベーションウィンドウ(incubation window)内で細胞培養培地からIgEが完全に除去された。1nM未満の濃度では、IgEが生物製剤抗IgEを超えた場合に、0.01nM、0.05nM、及び0.5nMの生物製剤抗IgEと共に4時間にわたりインキュベートした際に、それぞれ、IgEの8%、5%、及び1%が残存していた(Table 2(表3、表4))。
Table 3(表5、表6)中のデータは、オマリズマブがHEK293- hFcRn/β2mトランスフェクト細胞により効率的に取り込まれ、その結果、添加後4時間では上清中にほとんど残存していないことを示す。緩衝液の交換及びさらなる4時間のインキュベーション後に、オマリズマブの50~84%が細胞培地中に回収され、残りは細胞内で保持された。

固定化を、カルボキシメチル化センサーチップ表面(CM5チップ、GE Healthcare)への直接アミンカップリングにより実施した。各CM5チップのカルボキシメチル化デキストラン表面を、脱イオン水中における1:1の比での0.1MのN-ヒドロキシスクシンイミド(NHS)及び0.4Mの1-エチル-3-(3-ジメチルアミノプロピル)-カルボジイミド(EDC)の420秒間注入により活性化した。NHS/EDC溶液は、チップ上に存在する遊離カルボキシル基と反応して反応性のスクシンイミドエステルを生成し、このスクシンイミドエステルは、タンパク質の表面上に露出したリシン残基と反応し得、そのため表面上に固定される。タンパク質を、所望のレベルの固定化が達成されるまで、60~300秒パルスで、10mMの酢酸ナトリウムpH5.0中の10μg/mlの濃度にてNHS/EDC活性化表面全体に注入した。あらゆる残余の活性カルボキシメチル化基を、600秒にわたりチップ全体に注入した1MのエタノールアミンpH8.5でブロックした。参照細胞を、タンパク質の代わりに表面全体に緩衝液を注入した以外は同一の手順を使用して調製した。全ての固定化を、20μl/分の流速により25℃で実施した。
SPR実験を実施して、IgE-Fcとα-Cε4 Fabとの相互作用へのderCD23の効果を決定し、定常状態分析を実施して、この効果をKD及びBmaxの観点で定量化した。アミンカップリングを使用してα-Cε4 Fab CM5センサー表面を調製し、模擬的なアミン共役表面を参照減算対照として使用した。次いで、80nMのIgE-Fcを注入して、固定された1:1 α-Cε4 Fab/IgE-Fc複合体を生成した。短い解離フェーズを開始するためのα-Cε4 Fab共役表面全体への短いSPR緩衝液注入後、4000nM~31nMのderCD23の2倍連続滴定を1:1 α-Cε4 Fab/IgE-Fc複合体全体に注入した。次いで、derCD23注入後に解離フェーズが続き、α-Cε4 Fabにより捕捉されたIgE-Fc表面が再生された(図8)。注入を、10mMのHEPES、pH7.4、150mMのNaCl、4mMのCaCl2、及び0.005%(v/v)の界面活性剤P-20(GE Healthcare)のランニング緩衝液中において25μl.分-1の流速で実施した。全ての実験を2重に実行し、BIACore T200機器(GE Healthcare)を使用して再現性が高い結果が得られ、単相動力学的フィッティング(monophasic kinetic fitting)により1.8×10-6MのKDが得られた。
α-Cε4 Fabにより捕捉されたIgE-FcへのCD23の結合の相互作用、及び3種の抗IgE生物製剤とα-Cε4 Fabにより捕捉されたIgE-Fcとの間の相互作用に関する結合曲線を評価した。この実験での試験物は生物製剤抗IgE分子であり、この生物製剤抗IgE分子は、CD23単量体の対を含むが、IgE結合成分(CD23単量体)とFcRn結合成分(IgGFc)との間のリンカーの長さは、リンカーなし(抗IgE0)から(G4S)リンカー配列の3回の反復(抗IgE3)又は4回の反復(抗IgE4)まで変化した。1:1 α-Cε4 Fab/IgE-Fc複合体を、CM5センサー表面上に固定した。4000nM~31nMの抗IgE分子の2倍連続滴定を、α-Cε4 Fabにより捕捉されたIgE-Fc全体に注入し、解離フェーズ(図9a A~C)が続き、表面が再生された。次いで、SPR反応の変化を使用して、α-Cε4 Fabにより捕捉されたIgE-Fcの抗IgE分子に結合する能力を測定した。二相動力学的モデルを使用するデータフィッティングにより、KD1 1~2×10-6M及びKD2 1~4×10-8Mという2つのKD値が得られ、いずれの場合も、リンカーがより長いとKDがより低濃度であった。
リガンド密度は、SPR実験が固有の親和性又は機能的親和性をどの程度まで測定するかに影響を及ぼす場合がある。高いリガンド密度では、多価分析物が2種以上のリガンドに同時に結合し得る可能性がある。相互作用部位の動力学が同一であり且つ独立している場合には、最初の相互作用は、この部位の固有の親和性に依存することになる。分析物の高い局所濃度のために、その後の部位の会合は有利である。このセットの実験の実施では、IgE-Fcとα-Cε4 Fabとの間で1:1複合体を確実に形成するであろう密度でチップ表面上にα-Cε4 Fabを共有結合的に固定することにより、センサーチップ表面を調製した。3種の異なる濃度のIgE-Fc(80nM、160pM、及び40pM)を、固定されたα-Cε4 Fab捕捉分子全体に注入し、平均分子化間隔がそれぞれ40nm、80nm、及び110nmとなった。IgE-Fcのより低い固定レベルで、抗IgE分子は二価性がより低くなり、単相相互作用が有利となるであろうという仮定に基づいて、平均分子間隔の測定を選択した。様々な濃度の抗IgE分子(4000nM~31nM)を、α-Cε4 Fab/IgE-Fc表面全体に注入した。
図8中のデータは、CD23単量体が比較的低い親和性でIgEに結合し得ることを明確に示す。図9a中のデータは、対として配置されたCD23単量体は、単一の単量体と比較して改善された親和性でIgEに結合し得ること、及びCD23単量体成分とFcRn結合成分との間でのリンカーの導入により結合の改善が示されることを示す。図9bに示されるプロットは、抗IgE分子のそれぞれに関して、固定されたIgE分子間のより大きい間隔は、IgEとの結合により形成されている複合体の解離がより速いこと、及びこの効果は、抗IgE生物製剤のリンカー長の増加により減少することを示す。
IgE媒介性好塩基球脱顆粒を阻害する能力に対するリンカー長が変化する抗IgE生物製剤の評価
アレルゲンの存在下でのIgEと高親和性受容体FcεRIとの結合及びその結果としての結合したIgEの架橋が、肥満細胞及び好塩基球等のエフェクター細胞の活性化を引き起こし、アレルギー反応を引き起こすために炎症性メディエーター(例えばヒスタミン)の放出が引き起こされることが十分に確立されている。この研究では、IgEに結合して好塩基球性エフェクター細胞のIgE媒介性の活性化及び脱顆粒を防ぐために、対として組織されたIgE結合CD23単量体の空間的到達範囲を変更させるリンカーの導入の潜在的効果を調べた。
好塩基球性脱顆粒アッセイ
好塩基球性脱顆粒アッセイのアッセイ方法及び材料は、実施例2で説明された通りであった。この実施例の試験物は、一対のCD23単量体を含むが、IgE結合成分(CD23単量体)とFcRn結合成分(IgGFc)との間のリンカーの長さが、リンカーなし(抗IgE0)から(G4S)リンカー配列の3回の反復(抗IgE3)又は4回の反復(抗IgE4)まで変化する生物製剤抗IgE分子であった。これには、IgE結合成分の空間到達範囲の延長という効果がある。
試験した生物製剤抗IgEのそれぞれは、IgEと、RBL-SX38ヒト好塩基球性細胞系統の表面上で発現されている高親和性IgE受容体FcεRIとの間の相互作用を効率的にブロックすることにより、IgE媒介性脱顆粒を阻害し得た。抗IgE生物製剤のそれぞれの効力及び有効性は異なった。一対のCD23単量体を含むがIgE結合成分(CD23単量体)とFcRn結合成分(IgGFc)との間にリンカーがない生物製剤抗IgE0は、好塩基球のIgE媒介性脱顆粒を部分的に阻害し得たが、有効性は最大でも50%であった。CD23単量体とIgG-Fcとの間のリンカー配列の導入により、有効性及び効力の両方が顕著に増加し、リンカーがG4Sリンカーの3回の反復を含んだ場合には約90%の有効性に達し、リンカー長が(G4S)4反復まで増加した場合には100%の有効性に達した。したがって、観察されたIC50は、リンカー長の増加に伴い効力が増加し、抗IgE0の場合の>300nMからリンカーの付加による10~30nMまで減少することを実証した。
Claims (23)
- a)それぞれがCD23のC型レクチンドメインを含み、各単量体がIgEに結合することができる、少なくとも2つの単量体;及び
b)胎児性Fc受容体(FcRn)に結合することができる実体
を含むタンパク質構築物であって、前記タンパク質構築物がリンカーを含み、前記リンカーが、CD23のC型レクチンドメインを含む前記単量体を、FcRnに結合することができる前記実体に連結するために使用される、タンパク質構築物。 - 前記構築物が2つの単量体、又は2つより多い単量体、好ましくは4つ若しくは6つの単量体を含む、請求項1に記載のタンパク質構築物。
- 前記CD23のC型レクチンドメインが、配列番号1のV159~P290(配列番号6)、又は配列番号1のC160~C288(配列番号7)、又は配列番号1のF170~L277(配列番号8)、又はそれに対して少なくとも80%の同一性を有する配列を含むか、又はそれに対応する、請求項1又は請求項2に記載のタンパク質構築物。
- 前記CD23のC型レクチンドメインが、配列番号1のS156からA292(配列番号15)、好ましくは配列番号1のE133からA292(配列番号10)を含むか、若しくはそれに対応する、又は配列番号1のS156からC288(配列番号31)、若しくはその断片、若しくはそれに対して少なくとも80%の同一性を有する配列を含むか、若しくはそれに対応する、請求項1から3のいずれか一項に記載のタンパク質構築物。
- 前記CD23のC型レクチンドメインが、配列番号1のS156からE298(配列番号13)、好ましくは配列番号1のE133からE298(配列番号11)、又はその断片、又はそれに対して少なくとも80%の同一性を有する配列を含むか、又はそれに対応する、請求項1から4のいずれか一項に記載のタンパク質構築物。
- 前記CD23のC型レクチンドメインが、配列番号1のS156からS321(配列番号9)、好ましくは配列番号1のE133からS321(配列番号12)、又はその断片、又はそれに対して少なくとも80%の同一性を有する配列を含むか、又はそれに対応する、請求項1から5のいずれか一項に記載のタンパク質構築物。
- 各単量体が0.1~3μMの親和性でIgEに結合する、請求項1から6のいずれか一項に記載のタンパク質構築物。
- FcRnに結合することができる前記実体が、Fc領域、好ましくはIgG-Fc領域、若しくはその断片若しくはバリアント、又はアルブミン若しくはその断片若しくはバリアント、又はIgG抗体若しくはアルブミンに対する結合タンパク質、若しくはFcRnに対する結合タンパク質を含む、請求項1から7のいずれか一項に記載のタンパク質構築物。
- FcRnに結合することができる前記実体が、IgG1-Fc領域若しくはその断片若しくはバリアント、又はヒト血清アルブミン若しくはその断片若しくはバリアント、又はIgG1抗体若しくはヒト血清アルブミンに対する結合タンパク質を含む、請求項8に記載のタンパク質構築物。
- 前記結合タンパク質が、抗体若しくは抗体断片、好ましくはsdAbを含むか、又は非免疫グロブリンベースの単一ドメイン結合タンパク質、好ましくはフィブロネクチン若しくはフィブロネクチンベースの分子、アフィマー、アンキリンリピートタンパク質、リポカリン、ヒトAドメイン、ブドウ球菌プロテインA、チオレドキシン、ガンマBクリスタリン、又はユビキチンベースの分子を含む、請求項8又は請求項9に記載のタンパク質構築物。
- 前記リンカーが、ペプチドリンカーである、請求項1から10のいずれか一項に記載のタンパク質構築物。
- 構築物の部分a)の各単量体とIgEとの前記結合、及び/又は構築物の部分b)とFcRnとの前記結合が、エンドソーム条件に感受性である、請求項1から11のいずれか一項に記載のタンパク質構築物。
- 構築物の部分a)とIgEとの前記結合が、pH7.4と比較してpH6.0若しくは6.5において減少するか、又は血清カルシウムレベルと比較してエンドソームカルシウムレベルにおいて減少する、請求項12に記載のタンパク質構築物。
- 構築物の部分b)とFcRnとの前記結合が、pH7.4と比較してpH6.0若しくは6.5において増加するか、又は血清カルシウムレベルと比較してエンドソームカルシウムレベルにおいて増加する、請求項12又は請求項13に記載のタンパク質構築物。
- 少なくとも2つの単量体が、個々の単量体の結合親和性の合計と比較して、IgEとの結合のアビディティーの増加をもたらす、請求項1から14のいずれか一項に記載のタンパク質構築物。
- 請求項1から15のいずれか一項に記載のタンパク質構築物をコードするヌクレオチド配列を含む1つ若しくは複数の核酸分子;又は
そのような核酸分子を含む1つ若しくは複数の発現ベクター;又は
前記発現ベクター、核酸分子、若しくは請求項1から15のいずれか一項に記載のタンパク質構築物を含む、1つ若しくは複数の宿主細胞。 - 請求項1から15のいずれか一項に記載のタンパク質構築物を生成する方法であって、(i)請求項16に記載の、発現ベクターのうちの1つ若しくは複数又は核酸配列のうちの1つ若しくは複数を含む宿主細胞を、コードされたタンパク質構築物の発現に適した条件下で培養する工程;及び場合により(ii)宿主細胞から又は増殖培地/上清から発現されたタンパク質構築物を単離又は取得する工程を含む方法。
- 請求項1から15のいずれか一項に記載のタンパク質構築物を生成する方法であって、(i)IgE Fcが固定化されたアフィニティーマトリックスを、前記構築物がアフィニティーマトリックス上のIgE Fcに結合する条件下で、請求項1から15のいずれか一項に記載の構築物と接触させる工程;及び(ii)構築物がアフィニティーマトリックス上のIgE Fcに結合しなくなる条件下で、アフィニティーマトリックスから構築物を溶出させる工程を含む方法。
- 工程(i)において、そのような条件が、血清カルシウム又はpHレベル、好ましくは1から2mMのカルシウムレベル、又はpH7.4若しくは約pH7.4のpHに対応する条件である;及び/又は、工程(ii)において、そのような条件が、エンドソームカルシウム又はpHレベル、好ましくは3~30μMのカルシウムレベル又はpH5.0から6.5若しくは約pH5.0から6.5のpHに対応する条件である、請求項18に記載の方法。
- 請求項1から15のいずれか一項に記載のタンパク質構築物又は請求項16に記載の1つ若しくは複数の核酸分子若しくは発現ベクターを含む、組成物、好ましくは薬学的に許容される組成物。
- 療法における使用、好ましくは抗IgE療法における使用、又はIgE関連の疾患若しくは状態の治療若しくは予防における使用のための、請求項1から15のいずれか一項に記載のタンパク質構築物、又は請求項16に記載の1つ若しくは複数の核酸分子若しくは発現ベクター。
- 抗IgE療法又はIgE関連の疾患又は状態の治療又は予防における使用のための医薬又は組成物の製造における、請求項1から15のいずれか一項に記載のタンパク質構築物、又は請求項16に記載の1つ若しくは複数の核酸分子又は発現ベクターの使用。
- IgE関連の疾患又は状態の治療又は予防の方法であって、それを必要とする患者に、治療有効量の請求項1から15のいずれか一項に記載のタンパク質構築物又は請求項16に記載の1つ若しくは複数の核酸分子若しくは発現ベクターを投与する工程を含む方法。
