Abstract
Description
とりわけ化粧分野において、ケラチン物質をケアする為の様々な化合物が提案されてきている。
このタイプのバラの木は、花弁の数がより多いという点で、花弁の数が、一般的に該メイチボン品種とは異なり、サイズもより大きく、既に記述されている通りグレープフルーツの特徴的なノートを有する、より強い香りを放つ花を有する。
該花弁の色は、一般的に白色であるが、該Delgramaue品種はライラック色の花弁を有するという点で、花弁の色が、一般的に該デルグラマウエ品種とは異なり、より活力があり、「バラの黒点病」として知られている病気に対して、より耐性が高い。
主な実施態様に従って、本発明は、該バラの木が、品種メイチボンと品種デルグラマウエとを交配することによって得られたハイブリッドであることを特徴とする、バラの木の抽出物に関する。
本発明に従う抽出物は、とりわけ、皮膚、爪又は粘膜(口腔内頬側、頬骨、歯肉、生殖器、結合組織)に適用する為の、任意の化粧組成物に製剤化されうる。本発明の組成物は、保持される投与方法に応じて、普通に使用される提示形態のいずれかでありうる。
少なくとも1種の油、例えば揮発性油、とりわけ揮発性炭化水素系油、及び/又は
少なくとも1種の脂肪物質、例えば25℃で固体である脂肪物質
を含みうる。
品種メイチボンと品種デルグラマウエとを交配することによって得られた、本発明に従うハイブリッドのバラの木の抽出物は、任意の既知の手段によって得られうる。
a)品種メイチボンと品種デルグラマウエとを交配することによって得られたハイブリッドのバラの木の全体又は一部を、アルコール溶媒、特にエタノール溶媒、を含む少なくとも1つの浴に50℃未満の温度で浸出して、アルコール混合物を得る工程と、
b)任意的に、該アルコール混合物を濾過して、アルコール花浸出液を収集する工程と、
c)該アルコール混合物又は該アルコール花浸出液の、超臨界CO2による抽出を実施して、該抽出物を得る工程と
を含みうる。
a)品種メイチボンと品種デルグラマウエとを交配することによって得られたハイブリッドのバラの木の該花、花頂及び/又は葉を摘む工程と、
b)工程a)で用意された該花、花頂及び/又は葉を、アルコール溶媒、特にエタノール溶媒、を含む少なくとも1つの浴に50℃未満の温度で浸出して、アルコール混合物を得る工程と、
c)任意的に、該アルコール混合物を濾過して、アルコール花浸出液を収集する工程と、
d)該アルコール混合物又は該アルコール花浸出液の、超臨界CO2による抽出を実施して、該抽出物を得る工程と
を含みうる。
品種メイチボンと品種デルグラマウエとを交配することによって得られた、本発明に従うハイブリッドのバラの木の抽出物は、該瘢痕化及び再上皮化現象を促進し、且つ強化する。
白色バラの超臨界CO2抽出物の生成
バラのハイブリッド品種は、母系品種(Tchaikovsky(商標)Meichibon)及び父系品種(La Rose du Petit Prince/Rose Synactif by Shisheido(商標)Delgramaue)の2つの品種を制御交配することによって得られる。
a)バラの花、花頂及び/又は葉が、摘まれる工程と、
b)該新しく摘まれた花、花頂及び/又は葉が、エタノール溶媒を含む少なくとも1つの浴に50℃未満の温度、例えば室温、で浸出されて、アルコール混合物(この場合、エタノール混合物)が得られる工程と、
c)該アルコール混合物が濾過されて、アルコール花浸出液が収集される工程と、
d)該アルコール花浸出液の、超臨界CO2による抽出(図1中の「CO2sc抽出」)が、45℃及び150バールの圧力において実施されて、該抽出物が得られる工程と、
e)該抽出物が、中程度の真空(100~500mbar)下で、60℃を超えない温度で濃縮される工程と
を含む。
ケラチノサイトでの移動及び再上皮化試験
A.材料及び方法
A1.ケラチノサイトの培養及び処置
ケラチノサイトが、96ウェルプレート中の培養培地に播種され、次に移動分析にかけられた(参照名Platypus Oris(商標)Collagen I Coated Plate)。このプレートにおいて、該ウェルは、コラーゲン溶液で飽和され、各ウェルの中心にカバーが置かれ、この領域の細胞接着を予防し、従って人工創傷(移動領域)が形成された。該細胞の接着後、該カバーは廃棄され、該細胞はカルセイン-AMで標識された。30分間のインキュベーションの後、画像が撮られ(T0)、次に、該培地は、該試験抽出物又は該参照物(EGF)を含有する又は含有していない(対照)アッセイ培地で置き換えられた。該細胞がインキュベーションされ、画像分析が、14時間(T14)、18時間(T18)及び24時間(T24)の時点で動態学的に実施された。T14の時点で該画像が撮られる前に、該細胞は、再びカルセイン-AMで標識され、30分間インキュベーションされた。全ての実験条件が、n=3で実施された。
該細胞の移動領域は、0時間、14時間、18時間及び24時間のインキュベーションの後に、高分解能イメージングシステム、INCell Analyzer(商標)2200自動化顕微鏡(GE Healthcare)(4倍レンズ)を用いてモニタリングされ、人工創傷の表面積は、ソフトウェアImage Jで分析された。該人口創傷の表面積(細胞が存在しない中央面積)が、T0時点において、並びに14時間、18時間及び24時間のインキュベーションの後に測定された。該創傷の被覆をモニタリングし、定量する為に、14時間、18時間及び24時間のインキュベーションの後の該損傷の測定は、T0において測定された初期表面積と関連付けられた。該移動に対する該化合物の効果が、未処置対照と比較された。該結果が、下記の表1~表3に示されている。
該対照条件下では、該正常ヒト表皮ケラチノサイト(NHEK:normal human epidermal keratinocytes)の移動は、中庸(moderate)であり、14時間のインキュベーション後、創傷表面積の平均被覆度は41%であった。次の数時間以内に、NHEKの移動が増大して、24時間のインキュベーション後に平均被覆度は55%に達した。

第2のハイブリッドのバラの木の抽出物との比較研究
本研究において、本発明に従って、超臨界CO2による抽出によって得られたハイブリッドのバラの木メイチボン×デルグラマウエの抽出物は、別のハイブリッドのバラの木Rosa×Centifoliaのプロピレングリコール/水抽出物と比較される。
A1.試験された抽出物
該超臨界CO2抽出物は、実施例1において既に詳説されている通りに得られる。
該試験されたケラチノサイトは、5%CO2下、37℃で培養されたNHEK細胞(Bioalternatives K593;第3継代)である。該細胞培養培地は、0.25ng/mLの表皮成長因子(EGF:Epidermal Growth Factor)、25μg/mLの下垂体抽出物(EP)及び25μg/mLのゲンタマイシンを補充された、ケラチノサイト-SFM培地である。試行培地は、EGFもEPも含まないことを除いて、前述されたものと同じである。
該プロトコールは、実施例1において定義されている通りである。
該試験された細胞は、試行培地中24時間インキュベーションされたNHEK細胞に対応する。該細胞は、MTTアッセイ(テトラゾリウム塩)及び顕微鏡での形態学的研究の両方において評価される。
該対照条件下では、該正常ヒト表皮ケラチノサイト(NHEK)の移動は、中程度であり、14時間のインキュベーション後、創傷表面積の平均被覆度は30%であった。次の数時間以内に、NHEKの移動が増大して、24時間のインキュベーション後に平均被覆度は37%に達した。10ng/mLにおけるEGFは、NHEKの移動を有意に刺激し、この効果は、14時間、18時間、及び24時間のインキュベーションの後に観察される(該未処置試料と比較して、それぞれ221%、223%、208%)。この結果は、期待された結果であり、該研究の正当性を立証する。

- 前記バラの木の抽出物が、前記バラの木の全体又は一部のアルコール混合物の超臨界CO2での抽出によって得られることを特徴とする、請求項1又は2に記載のバラの木の抽出物。
- アルコール混合物は、前記バラの木の全体又は一部を、アルコール溶媒を含む少なくとも1つの浴中で50℃未満の温度で浸出した後に得られたものであることを特徴とする、請求項1~3のいずれか1項に記載のバラの木の抽出物。

- 皺、小皺、しなびた皮膚、皮膚の弾力性及び/又は張り及び/又は密度の喪失、皮膚の色つやの輝きの低下、皮膚の紙のような外観、皮膚のたるみ、並びに皮膚のしなびた外観から選択される化粧的徴候を処置及び/又は予防する為の、請求項6に記載の化粧的使用。
- ケラチン物質をケアする為の化粧的方法であって、それを必要としている個体に、少なくとも1つのバラの木の抽出物を活性剤として投与することからなる少なくとも1つの工程を含み、該バラの木が、品種メイチボンと品種デルグラマウエとを交配することによって得られたハイブリッドであることを特徴とする、前記化粧的方法。
