株および培養条件
この研究において使用した全ての株はATCCまたはDSMZのいずれかから購入した。この研究において使用した株は、ラクトバシラス・クリスパタス(DSM 20584)、ラクトバシラス・ジェンセニー(DSM 20557)、ラクトバシラス・ガセリ(DSM)、ラクトバシラス・ラムノサス(DSM/ATCC)、ビフィドバクテリウム・ロンガム亜種ロンガム、ビフィドバクテリウム・ブレベ、ビフィドバクテリウム・アドレセンティス、フェカリバクテリウム・プラウスニッツイ、ブラウティア・オベウム、ブラウティア・コッコイデス、バクテロイデス・ブルガタス、ドレア(ユーバクテリウム)・フォルミシゲネランス、バクテロイデス・ユニフォルミス(uniformis)、ルミノコッカス(Ruminococcus)・グナバス(gnavus)、ブラウティア・プロダクタ(producta)、クロストリジウム・レプタム(leptum)、クロストリジウム(ブラウティア)・コッコイデス、およびブラウティア(ルミノコッカス)・オベウムであった。
バイオフィルムの形態の細菌の耐性アッセイの前に、酸性度および抗生物質に耐性のプランクトン細菌を決定した。低pHアッセイのために、膣細菌の終夜培養物を希釈して、特有の株のバイオフィルム中の細菌の数に基づいて105~109コロニー形成単位(CFU)/mLを達成した。バイオフィルムの形態の細菌について行った手順と同様に、プランクトン細菌を異なる酸性度に37℃で1h曝露した。インキュベーションの終わりに、試料を遠心分離(最大速度、2分)し、上清を除去した。細菌ペレットを次にリン酸緩衝食塩水(PBS)×1に再懸濁させた後に、プレーティングおよびCFUのカウントを行った。
単一株(単独培養)
ラクトバシラス種の培養をグリセロールストック(12.5%)から開始し、37℃で、特有の株に基づいて、好気的または嫌気的条件において150~180rpmで、終夜インキュベートした。最終のバイオフィルム培養のために、培養物を0.05のOD600に希釈した。バイオフィルムの形態の細菌の培養物は、WO2016181228A2(参照により全体が本明細書に組み込まれる)に記載されるように得た。
培地-マトリックス溶液からの試料をチューブに移し、次にRTで3分間500rpmで遠心分離した。その後に、上清を廃棄し、PBSを用いてペレットを洗浄して、遠心分離の間に沈殿したプランクトン細菌を除去した。試料を再びRTで3分間500rpmで遠心分離した。遠心分離後に、上清を除去し、各処理(pHまたは抗生物質)について1gのバイオフィルムの形態の細菌を使用した。pH耐性アッセイのために、バイオフィルムの形態の細菌を異なる酸性度:pH 2および3.5(2M HClを使用してPBS×1のpHをそれぞれ2および3.5に調整した)を有する5mLのPBSに1h、37℃で1h、100rpmで曝露した。同じ条件を用いて、5mLのPBS 7(環境pH)中でインキュベートしたバイオフィルムの形態の細菌を対照とした。抗生物質アッセイのために、1grのバイオフィルムの形態の細菌を、特有の株のMIC値より十分に高い3つの逐次的な濃度の抗生物質に曝露した。バイオフィルムの形態の細菌を、抗生物質を含むまたは含まない(後者は対照として使用した)5mLの増殖培地中37℃で24hインキュベートした。
CFUを各耐性アッセイの終わりに決定した。最初に、段階希釈を実行し、細菌をMRS寒天プレート上に3連でプレーティングした。株増殖条件(上記を参照)に基づいて、好気性または嫌気性条件下37℃でプレートを48~72hインキュベートした後にCFUをカウントした。
細菌増殖を最適化するために、小スケール実験および中スケール実験という2種類の実験設計を実施した。小スケール実験から得られた結果に基づいて、本発明者らは中スケール実験のための条件を定義する。手順を簡潔に述べれば、生じた細菌集団の増殖および発達状態を、72時間の総インキュベーション時間の間の24時間のインキュベーション毎に調べた。各日に、培地を新鮮な培地で置き換え、バイオフィルム中の細菌の数を定量化した(以後pH 7での処理)。追加的に、細菌集団を極端な条件、例えば酸性pH(pH 3.5および2)、女性の膣において一般的なpH(pH 4 5)に近いpH値、ならびに抗生物質(CB、カルベニシリン;CIP、シプロフロキサシン;VAN、バンコマイシン;NVB、ノボビオシン)に曝露することにより粒子上のバイオフィルムの形成および発達を試験した。pH耐性アッセイを1日毎に実行し、抗生物質アッセイを48時間または72時間のインキュベーション後に行った。これらのアッセイからの結果を後に同じ種のプランクトン細菌と比較して、自由生活細菌を上回る生成された細菌集団の技術の利点を示した。追加的に、細菌がマトリックスにより良好に付着することを可能とするために、マトリックスを含む発酵槽へのプランクトン細菌の追加後1日目に、発酵槽を37℃で2時間、静的条件に保った(1時間後の穏やかな混合を伴う)。細菌収率における約1logの変化を細菌プレーティングおよびCFUカウントの技術的エラーの範囲内と考え、有意でない差異とした。試験した細菌株は表1に見られる通りである。
2つまたはそれより多くの細菌株を一緒に共培養する場合、混合物への各細菌株の追加の間の懸濁時間と共に株を粒子:培地混合物に別々に加えた。混合物への細菌株の追加後に、それをよく混合し、30分~1時間後に別の株を加えた。より多くの細菌株が同じ混合物中で共培養される場合にはこの過程を繰り返した。細菌追加の順番は、嫌気性/好気性株または低速/急速増殖株に基づいて決定し、嫌気性または低速増殖株は好気性または急速増殖株の前に加えた。
単一株
実験において使用した各細菌株の培養は-80℃のストック(5%のDMSO)から開始し、嫌気性条件下37℃で終夜インキュベートした。他に記載されなければ、最終のバイオフィルム培養のために培養物を0.05のOD600(FaP、0.03)に希釈した。各実験における単独培養物は、増殖培地(BfA、ビフィドバクテリウム培地または食品グレード+カゼイン;FaP、YCFACまたはRCM)、20%の単独培養技術粒子(MCC:DCP、1:1)および終夜培養からのプランクトン細菌からなる。連続撹拌条件(小スケール、80rpm;発酵槽、300rpm)または静的条件(小スケールのみ)のいずれかを用いて単独培養物を37℃で増殖させた。全ての実験を嫌気性グローブボックス中に保持したが、ビフィドバクテリウム種のための増殖培地は一般に好気性であり、フェカリバクテリウムを用いる実験用は嫌気性であった。連続撹拌を用いた場合、容器を最初に静的条件下で5h放置して細菌をマトリックスに付着させた。全ての単独培養実験は、小スケール(50もしくは60mLの体積)または発酵(3Lの体積)のいずれかの実験室スケール製造において実行した。
少数の細菌株を一緒に共培養する場合、各株について0.03または0.05の初期ODで株を並行して加えた。時間が試験するパラメーターである場合、低速増殖細菌をクラスター中の他の種の24h前に加えた。ODが実験において試験するパラメーターである場合、低速増殖細菌をクラスター中の他の細菌種より10倍多く加えた(初期OD 0.5)。全ての他の実験セットアップおよび分析は、単一株について記載したように行った。
培地-マトリックス溶液からの試料をチューブに移し、次にRTで3分間500rpmで遠心分離した。その後に、上清を廃棄し、PBSを用いてペレットを洗浄して、遠心分離の間に沈殿したプランクトン細菌を除去した。試料を再びRTで3分間500rpmで遠心分離した。遠心分離後に、上清を除去し、各処理(pHまたは抗生物質)について1gの試料を使用した。10mLのPBS×1を用いて3回目の洗浄を行った。遠心分離(500rpm、3分)後に、9mLを除去し、1mLの残存するPBS溶液中のバイオフィルムを高速で1.5分間ボルテックスして、粒子に付着したバイオフィルムから細菌を放出させた。
CFUを決定するために、段階希釈を実行し、特有の寒天プレート上に細菌を3連でプレーティングした。株増殖条件(上記を参照)に基づいて、嫌気性条件下37℃でプレートを48~72hインキュベートした後にCFUをカウントした。
坐剤製剤
坐剤の製剤は、薬学的に許容される賦形剤(油ベースの担体)ならびに/またはプレバイオティクス剤(例えば、クランベリー、アスコルビン酸および抗生物質)中に混合された、上記のように増殖させたバイオフィルムの形態の細菌からなる。使用したバイオフィルムの形態の細菌は、バイオフィルムの形態の凍結乾燥された(乾燥性)または湿潤性の細菌であった。坐剤製剤中にそれぞれ1:5の比の植物性(パーム)バターおよびココアバターを50~52℃の温浴中で融解させた。
細菌増殖の最適化
実験手順:小スケール、続いて中スケール実験という2種類の実験設計を用いてバイオフィルムの形態の細菌の応用を試験した(図1G)。小スケール実験から得られた結果に基づいて、本発明者らは中スケール実験のための条件を定義する。手順を簡潔に述べれば、バイオフィルムの形態の細菌の増殖および発達状態を、計72hの間の24hのインキュベーション毎に調べた。各日に、培地を新鮮な培地で置き換え、バイオフィルム中の細菌の数を定量化した(以後pH 7での処理)。追加的に、バイオフィルムの形態の細菌を極端な条件、例えば酸性pH(pH 3.5および2)ならびに抗生物質(CB、カルベニシリン;CIP、シプロフロキサシン;VAN、バンコマイシン;NVB、ノボビオシン)に曝露することにより粒子上のバイオフィルムの形成および発達を試験した。pH耐性アッセイを1日毎に実行し、抗生物質アッセイを48hまたは72hのインキュベーション後に行った。これらのアッセイからの結果を後に同じ種のプランクトン細菌と比較して、自由生活細菌を上回るバイオフィルムの形態の細菌の利点を示した。
小スケール実験からの結果は、穏やかな撹拌(100rpm)を伴う好気性条件において、マトリックスとの48hのインキュベーション後の最も高い細菌収率を示した。プランクトン細菌とは対照的に、バイオフィルムの形態の細菌は、撹拌および非撹拌条件についてそれぞれバイオフィルム中の細菌増殖における1~3.8logの低下と共に、pH 2で生存した(図3)。しかしながら、嫌気性条件においてバイオフィルム中の細菌はプランクトン細菌と比較して低pHにおいて何らの利点も示さなかった。
プランクトンLJ
プランクトン細菌は対照条件において約106細胞/mLの細菌収率を生じさせた(pH 7、図6)。対照と比較してpH 3.5において細菌収率において有意な差異は観察されなかった。しかしながら、プランクトン細菌はpH 2への曝露で生存しなかった。
小スケール実験セットアップにおけるバイオフィルムの形態のLJ細菌の増殖は両方の撹拌条件(70rpmおよび130rpm)において類似していた(図7)。バイオフィルムの形態の細菌の増殖において経時的な減少があった。24hのインキュベーション後に107細胞/mLの最大細菌収率が観察され、インキュベーションの3日目に最低細菌収率(約105細胞/mL)が記録された。バイオフィルムの形態の細菌をpH 3.5に曝露した場合、インキュベーションの1日目を除いて、対照(pH 7)と比較して細菌数において有意な差異はなかった。しかしながら、バイオフィルムの形態の細菌は最低pH処理(pH 2)で生存しなかった。一般に、バイオフィルム中の細菌の増殖はそれらの自由生活形態とは有意に異ならなかった。撹拌なしがバイオフィルムの形態の細菌(RD206)のより良好な収率を結果としてもたらすことができるかどうかを試験するために実験を行ったことが留意されるべきである。結果は両方の撹拌条件と比較して細菌増殖における約1logの減少を示した。
小スケール実験に類似して、バイオフィルムにおける細菌収率は約107CFU/mLの最大増殖に達したが、増殖は3日全てのインキュベーションの間に安定なままであった(図8)。さらに、2つの撹拌速度の間に有意な差異は観察されなかった。
プランクトンLCr
プランクトンLCrは、低pH処理に曝露し、抗生物質濃度を増加させた場合にプランクトンLJに類似した結果を呈した(図10)。pH 3.5への曝露は対照(pH 7)と比較して細菌細胞に有意に影響しなかった一方、pH 2において細菌は生存しなかった。
バイオフィルムの形態のLCr細菌が定期的に撹拌されたか否かに関わりなく、LCrを用いた小スケール実験は5×105細胞/mLの最大細菌収率および約104細胞/mLの最低細菌収率を生じさせた(図11)。両方の処理において、バイオフィルムの形態の細菌の増殖は、マトリックスとのインキュベーションでの3日後に最も高かった。驚くべきことに、中等度の酸性条件(pH 3.5)下で、細菌収率における約1~2logの増加が処理にかかわらず観察された。それにもかかわらず、pH 2においてバイオフィルム中の細菌の数は2~4log減少したか、または完全に減少した。この酸性条件でそれらの生存においてパターンを決定することはできず、pH2に対するそれらの耐性は以下の中スケール実験において再び調べられる。
バイオフィルム中の細菌収率は小スケール実験と比較して中スケール実験においてわずかにより高かった(約1~2log)(図12)。しかしながら、pH 3.5への曝露の場合のバイオフィルムの形態の細菌における増加は小スケール実験セットアップにおいて観察されたものと同等であった。バイオフィルムの形態のLCr細菌の高い生存が、最低pH処理に曝露されば場合に、インキュベーションの第1および第3の後に記録された。両方の実験セットアップにおいて、このpH処理においてバイオフィルムの形態の細菌は48h後に消滅した。
プランクトンLG
酸性pHに曝露された場合のプランクトンLGの結果はプランクトンLI、LJおよびLCrとは異なった(図14)。pH 3.5への曝露はプランクトンLGの数をわずかに減少させるが、pH 2でのインキュベーションは非常に低い数における細菌の生存を結果としてもたらした。
バイオフィルムの形態のLCrおよびLJ細菌に類似して、試験した2つの撹拌速度の間で有意な差異は観察されなかった(図15)。最も高い細菌収率は107細胞/mLであった。48h時に、酸性pHへの曝露後にバイオフィルムの形態の細菌は高い生存能を示し、バイオフィルム中の細菌の数はpH 3.5と対照との間で異ならなかった一方、pH 2において接種された場合に細菌において70rpmおよび130rpmでそれぞれ1.3および1.6logのみの低下があった。この結果は、バイオフィルム細胞の性能はプランクトン細胞のそれよりも優位であることを指し示し、後者において細胞はpH 2で完全に消滅した。
2つの撹拌条件を比較した場合、インキュベーション時間にかかわらずバイオフィルム包埋細菌細胞の数において有意な差異は検出されなかった(図16)。さらには、両方の処理において、最低pH処理への曝露後にバイオフィルム細胞の高い生存が観察された。このpH処理での細胞生存能における減少は(130rpmでの48hのインキュベーション後に)3.2log以下であった。pH2での相対的に大きい生存がまた他の実験において観察された(図22~図24を参照)。pH3.5において、バイオフィルムの形態のLG細菌の生存は、70rpmと比較して130rpmで増殖された場合にわずかにより良好なようである。しかしながら、pH2において、2つの撹拌速度の間でバイオフィルムの形態の細菌の生存において差異は検出されなかった。
プランクトンLRh
プランクトンLRh細胞は、増加酸性度に曝露された場合、このプロジェクトにおいて記載される全ての細菌株に類似した応答を示し、pH 3.5に曝露された対照試料およびバイオフィルムの形態の細菌の間で差異は検出されなかった一方、細胞はpH 2において消失した(図18)。
バイオフィルム包埋細菌の最大数は約1010CFU/mLであった(図19)。全体的に、70rpmを経験したバイオフィルムの形態のLRh細菌の増殖は、130rpmを経験したバイオフィルムの形態の細菌よりもわずかに良好なようであり、これはpH 2での48h後のそれらの高い生存(5.8log)およびインキュベーションの3日目における(pH 7と比較して)pH 3.5でのそれらのわずかに向上した生存において表される。追加的に、72hのインキュベーションにおいてバイオフィルム包埋細菌の数における小さい減少がある。
以前の小スケール実験とは異なり、バイオフィルム包埋細菌の平均数は107CFU/mLを越えなかった(図20)。この差異は、バイオフィルムの形態の細菌からの懸濁された細菌の分離を向上させるためにプロトコールに含めた追加の洗浄ステップに起因した。この時点以降の全ての実験設計においてこの洗浄ステップを用いた。経時的に、バイオフィルムの形態のLRh細菌の増殖速度は一定のままであった。しかしながら、この実験において、バイオフィルムの形態のLRh細菌はインキュベーションの2日目の終わりに低pH処理に対してより良好な耐性を示すようであり、2log以下の低減がpH 2で観察された。
坐剤製剤の評価
坐剤の製剤は、薬学的に許容される賦形剤(油ベースの担体)および/またはサプリメント中に混合されたバイオフィルムの形態の細菌からなる。バイオフィルムの形態の細菌を、(72hのインキュベーションの終わりに)凍結乾燥(乾燥性)粉末またはバイオフィルムの形態の湿潤性細菌のいずれかとして使用した。坐剤中のバイオフィルムの形態の細菌の生存の安定性アッセイを6か月の持続期間にわたり月に1回行った。各月に、各製剤からの1つの坐剤をPBS(×1)に融解し、CFUをカウントするために細菌をプレーティングした(分析方法を参照)。
添加剤。高い膣pHは膣病原体の増加と関連付けられ、膣の酸性度は、乳酸菌が増殖するために都合のよい環境を作りながらの嫌気性病原体の定着に対する保護的機構であると長く理解されてきた。(小スケールの部分として)バイオフィルムLPの形態の細菌に対する異なる添加剤(クランベリーおよびアスコルビン酸)の効果の調査。ここで、本発明者らは2つの酸性化剤、クランベリーおよびアスコルビン酸を調べ、前者はまた、UTI感染症の反復性の予防および/または低減において役割を有することが示唆される。目的は、坐剤組成物中にバイオフィルムの形態の細菌と共にこれらの添加剤の1つを加えることである。そのようにして、バイオフィルムの形態の細菌の生存および増殖に対するそれらの効果を試験した。クランベリー粉末(300mg)へのバイオフィルムの形態のLP細菌の曝露後に、バイオフィルムの形態の細菌の生存および/または増殖に対する有意な効果は記録されなかった(図27)。
少数の小実験の後に、油ベースの担体の基本的な製剤はそれぞれ1:5の比の植物性(パーム)バターおよびココアバターの他に、混合物の均質性を補助するための数滴のレシチンを含んだ。次に、本発明者らは、バイオフィルムの形態の細菌と比較して油ベースの担体の体積を低減させて、坐剤中のバイオフィルムの形態の細菌の量を増加させることを試みた。担体に対するバイオフィルムの形態の細菌の2つの比、それぞれ1:5および1:10を試験した(図24)。結果は、2つの比の間でバイオフィルムの形態の細菌の増殖において差異を示さず、そのため坐剤組成物中のバイオフィルムの形態の細菌の量を上昇させることが許容された。

共培養されたバイオフィルム
2つの細菌株を用いる共培養
LJおよびLGは嫌気性/低速増殖細菌として同定された一方、LRhは好気性/急速増殖細菌として同定された。LJまたはLGプランクトン細胞をLRhと共に接種する場合、それらは株の追加の間の約1時間のギャップと共にLRhの前に加えた。単独培養としてのそれらの成長と比較してバイオフィルム成長(最大1log)および発達(pHに対する耐性)において有意な差異は記録されなかった(図27)。バイオフィルム包埋細菌の数は106~107CFU/mLであった。
この実験において本発明者らは以下の細菌の組み合わせを接種した:LRh-LJ-LGまたはLRh-LJ-LG-LCr(以後それぞれ「組み合わせA」および「組み合わせB」;図7)。LRh、LJおよびLGを一緒に培養した場合、各株についてバイオフィルム中の細菌の数は107~108CFU/mLであった。細菌株をFG1ブロス中に接種し、それゆえMRSを用いて行った以前の共培養実験と比較してより多くの数のバイオフィルム細胞とした。pH 3.5において、全ての細菌株は培養時間にかかわらず高い耐性を呈した。しかしながら、pH 3.5に対するLJ細菌集団の耐性はインキュベーション時間と共に減少した。pH 2において、LGの生存はpH 3.5におけるその生存とは有意に異ならなかったが、LJおよびLRhは不良な生存率を示した。これらの結果は、細菌を単独で増殖させた以前の実験からの発見にしたがった。これらの株は互いにバイオフィルム集団を阻害も抑制もしないことをこれらの結果は指し示す。
インキュベーションの始めにおけるマトリックス:培地の混合物への細菌追加の順序を試験するために、本発明者らは、より弱い/低速増殖株および次により強い/急速増殖株の追加を反対の組み合わせ(最初により強いおよび次により弱い)と比較する。これは、より弱い株によるバイオフィルムの形成に対する発酵に加えられる株の順序における利益があるかどうかを示す。
1)嫌気性/低速増殖株を最初におよび次に好気性/急速増殖株を加えること、LCr→LJ→LG→LRh;
2)より強い/急速増殖株を最初におよび次に遅いより弱い/低速増殖株を加えること、LRh→LG→LJ→LCr。
粒子サイズおよび粒子に対する細菌の親和性

細菌集団増殖に対する動物および非動物培地の効果
LG、LRh、LCrおよびLJの細菌集団増殖および発達状態に対する異なる動物ベースおよび非動物ベースの増殖培地の効果を評価した(図32A~図32C)。ラクトバシラス種を動物ベースのブロス(以後、動物)ならびに/または非動物ベースのブロス(以後、非動物1および非動物2)のいずれかで増殖させた。
ビフィドバクテリウム・アドレセンティス単独対共培養
本発明者らは、B.ブレベおよびB.ロンガム亜種ロンガムとのそのバイオフィルム共培養増殖と比較して単独培養におけるB.アドレセンティス(BfA)の増殖性能を調べた。BfAは、単独でのその増殖と比較してBLLおよびBfBrと共培養された場合に72h後に増殖の向上を示した(図34)。BfAは、単独でのその増殖と比較してBfBrと共培養された場合に24hおよび72h後に増殖の向上を示した(図35)。
フェカリバクテリウム・プラウスニッツイ単独対共培養
本発明者らは、ブラウティア・オベウム(BlO)、ブラウティア・コッコイデス(BlC)、または両方とのそのバイオフィルム共培養増殖と比較して単独培養におけるF.プラウスニッツイ(FaP)の増殖性能を調べた。FaPは、単独でのその増殖と比較してBlO、BlCまたは両方のいずれかと48h共培養された場合に増殖の向上を示した(図39)。FaPはまた、単独でのその増殖と比較してバクテロイデス・ブルガタス(BaV)、またはBaVおよびドレア(ユーバクテリウム)・フォルミシゲネランス(DoF)のいずれかと共培養された場合に増殖の向上を示した(図40)。
Claims (26)
- 第1の親油性担体ならびにラクトバシラス(Lactobacillus)・クリスパタス(crispatus)およびL.ガセリ(gasseri)、L.ジェンセニー(jensenii)、およびL.ラムノサス(rhamnosus)からなる群から選択される少なくとも1つの追加の細菌種を含む乾燥したバイオフィルムの形態の共培養されたプロバイオティクス細菌、ならびに抗生剤、pH調整剤、または両方を含む第1の剤を含む、組成物。
- 前記乾燥したバイオフィルムの形態の共培養されたプロバイオティクス細菌および前記抗生剤を含む第1の剤が前記組成物内に均質に分散している、請求項1に記載の組成物。
- 前記乾燥したバイオフィルムの形態の共培養されたプロバイオティクス細菌が前記組成物全体の10%~50%(w/w)である、請求項1または2に記載の組成物。
- 前記乾燥したバイオフィルムの形態の共培養されたプロバイオティクス細菌が粒子に付着している、請求項1~3に記載の組成物。
- 前記粒子が、種子、MCC、リン酸二カルシウム、多糖、およびこれらの任意の組み合わせからなる群から選択される、請求項4に記載の組成物。
- 第2の層をさらに含む、請求項1~5のいずれか1項に記載の組成物。
- 前記第2の層が第2の親油性担体、第2の剤または両方を含む、請求項6に記載の組成物。
- 前記乾燥したバイオフィルムの形態の共培養されたプロバイオティクス細菌の放出が前記第2の剤の放出よりも遅い、請求項7に記載の組成物。
- 前記第1の剤および前記第2の剤のいずれか1つが抗生物質である、請求項7または8に記載の組成物。
- 安定化剤、防腐剤、滑沢剤、粘度改変剤、緩衝化剤、脂肪酸、およびこれらの組み合わせをさらに含む、請求項1~9のいずれか1項に記載の組成物。
- 細菌性膣症の治療における使用のための、請求項1~10のいずれか1項に記載の組成物。
- a.フェカリバクテリウム(Faecalibacterium)・プラウスニッツイ(prausnitzii)ならびにブラウティア(Blautia)・オベウム(obeum)、Bl.コッコイデス(coccoides)、バクテロイデス(Bacteroides)・ブルガタス(vulgatus)、およびドレア(Dorea)(ユーバクテリウム(Eubacterium))・フォルミシゲネランス(formicigenerans)からなる群から選択される少なくとも1つの追加の細菌種、
b.ラクトバシラス・クリスパタスならびにL.ガセリ、L.ジェンセニー、およびL.ラムノサスからなる群から選択される少なくとも1つの追加の細菌種、または
c.ビフィドバクテリウム(Bifidobacterium)・アドレセンティス(adolescentis)ならびにBif.ロンガム(Longum)亜種ロンガム(longum)、およびBif.ブレベ(Breve)からなる群から選択される少なくとも1つの追加の細菌種
を含む乾燥したバイオフィルムの形態の共培養されたプロバイオティクス細菌を含む、組成物。 - 経口、膣、直腸、および外用からなる群から選択される送達経路のために製剤化されている、請求項1~12のいずれか1項に記載の組成物。
- 坐剤の形態である、請求項1~13のいずれか1項に記載の組成物。
- それを必要とする対象においてフローラをモジュレートする方法であって、前記対象に治療有効量の請求項1~14のいずれか1項に記載の組成物を投与し、それにより前記対象においてフローラをモジュレートすることを含む、方法。
- 前記フローラが、膣フローラ、腸フローラ、または皮膚フローラである、請求項15に記載の方法。
- 前記モジュレートすることが前記対象の生来のフローラを回復させることである、請求項15または16に記載の方法。
- それを必要とする対象においてディスバイオシス関連状態または腸および代謝性疾患を予防または治療するための、請求項15~17のいずれか1項に記載の方法。
- 前記ディスバイオシス関連状態または腸および代謝性疾患が、細菌性膣症、泌尿生殖器感染症、潰瘍性大腸炎、炎症性腸疾患(IBD)、クローン病、結腸直腸がん、肥満症、およびセリアック病からなる群から選択される、請求項18に記載の方法。
- 請求項1に記載の組成物を調製する方法であって、
a.粒子を含む増殖培地にL.クリスパタスを接種するステップ、
b.前記粒子をステップ(a)からのL.クリスパタスと、L.クリスパタスが前記粒子に付着することを可能とするために好適な条件下でインキュベートするステップ、
c.ステップ(b)の前記粒子に少なくとも1つの追加の細菌種を接種するステップ、ならびに
d.ステップ(c)の前記接種された粒子を、L.クリスパタスおよび少なくとも1つの追加の細菌種を含むバイオフィルムを形成するために好適な条件下で培養するステップ
を含む、方法。 - 前記少なくとも1つの追加の細菌種が、L.ジェンセニー、L.ガセリ、およびL.ラムノサスからなる群から選択される、請求項20に記載の方法。
- 請求項12に記載の組成物を調製する方法であって、
- 粒子を含む増殖培地に、(i)L.クリスパタス、(ii)Bif.アドレセンティス、または(iii)F.プラウスニッツイから選択される第1の細菌を接種するステップ、
- 前記粒子を前記第1の細菌と、前記第1の細菌が前記粒子に付着することを可能とするために好適な条件下でインキュベートするステップ、
- 前記粒子に少なくとも1つの追加の細菌種を接種するステップ、ならびに
- 前記接種された粒子を、(i)L.クリスパタス、(ii)Bif.アドレセンティス、または(iii)F.プラウスニッツイ、および少なくとも1つの追加の細菌種を含むバイオフィルムを形成するために好適な条件下で培養するステップ
を含む、方法。 - 前記第1の細菌がL.クリスパタスである場合、前記少なくとも1つの追加の細菌種が、L.ジェンセニー、L.ガセリ、およびL.ラムノサスからなる群から選択される、請求項22に記載の方法。
- 前記第1の細菌がBif.アドレセンティスである場合、前記少なくとも1つの追加の細菌種が、Bif.ロンガム亜種ロンガム、およびBif.ブレベからなる群から選択される、請求項22に記載の方法。
- 前記第1の細菌がF.プラウスニッツイである場合、前記少なくとも1つの追加の細菌種が、Bl.オベウム、Bl.コッコイデス.Bac.ブルガタス、およびドレア(ユーバクテリウム)・フォルミシゲネランスからなる群から選択される、請求項22に記載の方法。
- 前記第1の細菌がF.プラウスニッツイであり、かつ前記接種するステップが同時に行われる、請求項25に記載の方法。
