究は日増しに徹底させ、細胞免疫、体液免疫及び免疫調節において重要な役割を果たして
いることを発見した。特に、腫瘍免疫治療における作用と臨床的意義は多くの筆者から重
視されている。

胞の生体外長期生存を維持し、そして、その生物学的機能活性を維持し、いくつかの腫瘍
抗原によって活性化されたT細胞はIL-2の作用下で、その腫瘍細胞を殺傷する機能を
増強し、抗腫瘍作用を強化できることと、B細胞の増殖を促進し、分化し、抗体を産生す
ることと、顆粒球-マクロファージコロニー刺激因子及びγ-インターフェロンなどの分
泌を刺激し、リンパ球によるこれらの受容体の発現を促進し、サブ用量の分裂促進因子の
存在下で胸腺の増殖を維持し、IL-2受容体の発現を促進し、生体の免疫調節系に参与
し、免疫のバランス及び安定を維持することとを含む。
AがIL-2薬物であるアルデスロイキン(Aldesleukin)の末期腎臓がんと
悪性黒色腫への応用を許可し、有効率は15%-20%であり、10%~15%近くの患
者の生存期間を5年以上にすることを可能にしたが、臨床での使用量が非常に高いため、
例えば、重篤な低血圧や血管漏出症候群など、IL-2の副作用が非常に大きい。当時、
IL-2治療法を受ける患者は入院して、24時間綿密に観察しなければならず、やや不
用意になると、生命の危険があった。したがって、巨大な副作用でIL-2の使用が制限
され、多くの臨床医はそれを採用することを思いとどまらせている。。
IL-2は、主に抗原で刺激されたCD4+T細胞から産生され、CD8+T細胞、N
K細胞とDC細胞も分泌できる。高用量のIL-2はCD4+T細胞の分化、CD8+T
細胞とNK細胞の増殖を促進し、これらの細胞の殺傷力を増加させることで、生体の免疫
反応を発揮させることができる。逆に、低用量のIL-2は制御性T細胞の増殖を促進し
、免疫系を抑制する作用があるため、低用量のIL-2も自己免疫疾患の治療に用いられ
ることも多い。
-2受容体においては、合わせて3つのサブユニットを有してα、β、γサブユニットを
構成するからである。CD8陽性T細胞とNK細胞などのキラー細胞表面受容体は、主に
βとγサブユニット(IL-2Rβγ)から構成され、該受容体のIL-2に対する親和
力が比較的に低く、高用量のIL-2により活性化させる必要があるが、制御性T細胞表
面の多くは、α、β、γという3つのサブユニットからなる高親和性の抗体(IL-2R
αβγ)であり、低用量のIL-2のほうがこれらを活性化しやすく、それにより免疫抑
制の役割を果たす。
けたが、ずっと免疫治療において重視されなくなり、あまり使用されていなかった。なぜ
かというと、主として以下の原因がある。一つ目は有効率が低く、IL-2のみを使用す
る場合、有効率は15-20%程度しか達成できないことである。二つ目は、IL-2機
能が両面性を有し、低用量のIL-2により制御性T細胞の増殖を促進して免疫抑制を引
き起こし、どのようにこの2つの機能を調和させるかはまだ解決できないことである。三
つ目は、半減期が短く、数分間しかなく、持続的に効果を発揮するには、高用量で投与し
なければならないことである。四つ目は、副作用が重篤であり、過度に活性化された免疫
系が、自己臓器を攻撃して臓器不全を引き起こすおそれがあり、用量範囲が狭いことであ
る。IL-2の治療効果の向上や副作用の減少のために、科学者たちは様々な併用療法、
例えばIL-2+インターフェロン療法、IL-2+リンパ因子活性化キラー(LAK)
細胞療法、IL-2+化学療法などを試したことがあったが、いずれも期待される治療効
果が得られなかった。

なった。したがって、NKTR-214が誕生した。NKTR-214は神秘的なもので
はなく、本質的に20年以上前に発売された従来の薬品IL-2のアップグレード版であ
り、副作用が更に低く、特異性が更に良く(Treg細胞ではなくCD8を活性化しやす
くなる)、免疫系をより効率的に活性化することができる。具体的には、IL-2分子上
に、6つのポリエチレングリコール(PEG)修飾を加えて不活性薬物が形成され、不思
議なことに、腫瘍患者に注射すると、これら6つのPEG修飾は徐々に脱落し、活性のあ
る2-PEGと1-PEGの形式が形成された。

ングレードを通じて薬物の徐放効果を達成し、それによって耐性を大幅に高めることと、
キラー免疫細胞に対する選択性を高め、微小環境における免疫細胞PD-1の発現量を増
加させるというIL-2の腫瘍治療における2つの課題を解決した。

214は、単回投与後、NKTR-214腫瘍内のAUC値がアルデスロイキンの400
倍近くになった。また、Nektar社が提供したデータによると、NKTR-214は
複数の前臨床動物モデルにおいていずれも持効性抗腫瘍免疫治療効果を示した。腫瘍浸潤
リンパ球モデルにおいて、NKTR-214で処理した後、CD8陽性T細胞とCD4陽
性T細胞との比が、約450:1であり、強力な腫瘍殺傷作用を示した。そのため、NK
TR-214は、免疫刺激因子系抗がん薬に属し、抗体に近い治療効果を有し、T細胞の
成長を刺激することができ、それにより腫瘍に対する殺傷作用を実現する。前臨床研究の
結果から、該薬物は持効性抗腫瘍治療効果を有し、非常に期待性のある抗がん薬物である
ことを明らかにした。

S1:CNBr活性化アガロースゲル(sepharose CNBr)0.8gを秤
量し、12mlの1mMHClを加え、均一になるまで人工でゆっくり混合し、4℃の冷
蔵庫で15min反応させた。
S2:カップリングbufferを調製し、すなわち、0.1M NaHCO3 pH
8.3と0.5M NaClとを十分に混合し、12mlのカップリングbufferを
取って、前のステップで膨潤した充填剤を洗浄した。
S3:IL-2α受容体4mgを4ボトル取り、2mlのカップリングbufferに
溶解し、ステップ2で溶解した充填剤に加えた。
S4:室温下で2時間ゆっくり揺動した。
S5:5倍カラム体積のカップリングbufferで余分な受容体を洗い落とした。
S6:0.1M pH8.0のトリス(ヒドロキシメチル)アミノメタン(Tris-
HCl)を加え、室温2時間放置し、Tris-HClを捨てた。
S7:0.5M NaClを含む5倍カラム体積の0.1M pH4.0酢酸/酢酸ナ
トリウム緩衝液で洗浄し、0.5M NaClを含む5倍カラム体積の0.1M pH8
.0のTris-HCl緩衝液で洗浄した。
S8:ステップ6及びステップ7を3回繰り返した。
S9:0.1M pH8.0のTris-HCをl0.5ml加えた。
S10:sepharose樹脂を5mlの空カラムに注入し、4℃の冷蔵庫で保管し
た。
S1:5ml受容体親和カラムをタンパク精製用クロマトグラフィーシステム(AKT
A Purifier)に装着した。
S2:移動相を調製し、すなわち、A相:5mM PBS、pH7.0;B相:0.2
M NaClを含む0.2M酢酸溶液。
S3:移動相の流量を0.45ml/minに設定した。
S4:A相をUV曲線安定まで平衡化した。
S5:IL-2サンプルをサンプルループでサンプリングし、IL-2上のα受容体結
合部位をIL-2α受容体に結合した。
S6:適量の分子量が20 KDaであるメトキシポリエチレングリコール-プロピオ
ン酸スクシンイミド(M-SPA-20K)をpH8.0、2.5 mMPBに溶解し、
その最終濃度を40mg/mlとし、2mlのM-SPA-20Kを抽出してサンプルル
ープでサンプリングし、UV曲線が安定するまで平衡化した。
S7:ステップ6を3回繰り返した。
S8:グラジエント溶離を行い、クロマトグラムは、図1に示すとおりである。溶出ピ
ークを収集し、限外ろ過濃縮を経て、ドデシル硫酸ナトリウム-ポリアクリルアミドゲル
電気泳動(SDS-PAGE)により検出した。
ゲルろ過クロマトグラフィーの条件:移動相:5mM PBS、流量:0.75 ml
/min、カラム:superdex 200 increase 10/300 GL
とした。
単離手順は以下のとおりである:
S1:クロマトグラフィーシステムをUV曲線安定まで平衡化した。
S2:サンプリング後、溶出ピークを収集した。
S3:収集成分限外ろ過濃縮後、SDS-PAGE測定を行い、その結果が図3と図4
に示されている。
検出結果:SDS-PAGEにより、図4のピーク1はPEG-IL-2で、ピーク2
はIL-2であることが認められた。
図4のピーク1(PEG-IL-2)とピーク2(IL-2)サンプルをそれぞれ収集
し、濃縮後、SPR解析装置を介して親和性テストを行った。受容体タンパクは、それぞ
れIL-2α受容体、IL-2β受容体であり、試験の結果は以下の通りである。
上表の結果が、本発明に係る方法により調製されたPEG-IL-2とIL-2β受容体
との結合力が強いことを明らかにした。

Claims (16)
- 結合部位制御可能なPEG化生体分子の製造方法であって、ブロッカーを、生体分子に
おける少なくとも1つの結合部位に結合する工程(1)と、生体分子をPEG化する工程
(2)と、ブロッカーを生体分子から単離する(3)工程とを含み、
前記工程(1)において、生体分子は、ブロッカーと特異的に結合する作用を有する生
体高分子または低分子から選択され、前記生体分子上には、少なくとも1つのブロッカー
の結合部位を有し、
ブロッカーおよび生体分子は、リガンドと受容体、DNAとその相補的DNAまたはR
NA、酵素とその基質、酵素とその競合阻害剤、酵素とその補酵素因子、ビタミンとその
特異的結合タンパク質、糖タンパク質とそれに対応するレクチンなどの組み合わせから選
択され、
好ましくは、前記リガンドと受容体は、ホルモンと受容体、薬物と受容体から選択され
る、結合部位制御可能なPEG化生体分子の製造方法。 - 前記DNAとその相補的DNAまたはRNAは、遺伝子プローブと塩基に相補的な遺伝
子配列から選択され、
前記酵素とその基質は、プロテアーゼとタンパク質、アミラーゼとデンプン、ヌクレア
ーゼと核酸、乳酸デヒドロゲナーゼと乳酸、オキサロ酢酸デカルボキシラーゼとオキサロ
酢酸から選択され、
前記酵素とその競合阻害剤は、コハク酸デヒドロゲナーゼとマロン酸、ジヒドロ葉酸シ
ンターゼとスルフォンアミド類薬物、コリンエステラーゼと有機リン農薬、スルフヒドリ
ル酵素とルイサイトから選択され、
前記酵素とその補酵素因子は、ピルビン酸デヒドロゲナーゼとMn2+、カタラーゼお
よび酸化還元酵素とFe2+/Fe3+から選択され、
前記ホルモンと受容体は、エストロゲンとエストロゲン受容体、アンドロゲンとアンド
ロゲン受容体、鉱質コルチコイドと鉱質コルチコイド受容体、甲状腺ホルモンと甲状腺ホ
ルモン受容体、プロゲステロンとプロゲステロン受容体から選択され、
前記薬物と受容体は、インスリン、インスリン様成長因子、上皮成長因子、血小板由来
成長因子およびリンパ因子とチロシンキナーゼ活性を有する受容体、アドレナリン、ドー
パミン、5-ヒドロキシトリプタミン、M-アセチルコリン、オピオイド類、プリン類、プ
ロスタグランジンおよびポリペプチドホルモン類薬物とG-タンパク質共役受容体から選
択され、好ましくは、前記薬物と受容体は、葉酸と葉酸受容体、ILとIL受容体から選
択され、
前記ビタミンとその特異的結合タンパク質は、ビタミンAとレチノール結合タンパク質
、ビタミンDとビタミンD結合タンパク質、α-トコフェロールとα-トコフェロール輸
送タンパク質、ビタミンKとリポタンパク質から選択され、
前記糖タンパク質とそれに対応するレクチンは、ホースラディッシュペルオキシダーゼ
とコンカナバリンA、レンズマメレクチンとエンドウレクチン、ジャカリン関連レクチン
とガラクトース、ヒユ科レクチンとN-アセチルガラクトサミン、二量体キチン結合レク
チンとN‐アセチルグルコサミンオリゴ糖、DBAレクチンと血液型A物質、ハリエニシ
ダレクチンと血液型O物質2-L-フコースから選択される、ことを特徴とする請求項1
に記載の製造方法。 - 前記薬物と受容体は、IL-2とIL-2受容体から選択され、
IL-2受容体は、IL-2α受容体、IL-2β受容体、IL-2γ受容体を含む、
ことを特徴とする請求項2に記載の製造方法。 - 前記工程(3)において、ブロッカーを生体分子から単離する方法は、遠心分離、沈殿
分離、ろ過分離、泡沫分離、抽出分離、膜分離、クロマトグラフィー分離、電気泳動分離
、グラジエント溶離のうちの1つまたは複数種類の組み合わせを含む、ことを特徴とする
請求項1に記載の製造方法。 - 前記工程(2)において、PEG化生体分子は、以下の式Iで表される構造を有し、
式中、mが、1~12の整数であり、具体的には1、2、3、4、5、6、7、8、9
、10、11、または12であってもよく、
Xは、PEGと生体分子との間の結合基であり、-(CH2)a-、-(CR1R2)
a-、-(CH2)aNH-、-NHCO(CH2)a-、-(CH2)aCONH-、
-(CH2)aCO-、-CO(CH2)a-、-(CH2)aCONH(CH2)a-
、-(CH2)a-S-S-(CH2)a-、-(CH2)aCOO(CH2)a-、-
(CH2)a-S-(CH2)a-から選択される1つ又は複数の組み合わせであり、
aが、0~10の整数であり、
R1およびR2は、独立して、-H、C1-6のアルキル基、-OR´、-NHR´、
-N(R´)2、-CN、-F、-Cl、-Br、-I、-COR´、-COOR´、-
OCOR´、-CONHR´、または-CON(R´)2から選択される1つ又は複数の
組み合わせであり、
R´は、-H、C1-6のアルキル基、-F、-Cl、-Brまたは-Iから選択され
、
前記PEGは、直鎖、Y型、多分岐ポリエチレングリコール残基であり、例えば、モノ
メトキシポリエチレングリコール(mPEG)、直鎖両末端PEG、Y型PEG、4-A
rm分岐鎖PEG、6-Arm分岐鎖PEGまたは8-Arm分岐鎖PEGを含み、
PEGの分子量が、1~100KDaである、ことを特徴とする請求項1に記載の製造
方法。 - 前記aが、0~5の整数であり、
R1およびR2は、独立して、H、C1-3のアルキル基、-OH、C1-3のアルコ
キシ基、-NH2、-F、-Cl、-Brおよび-Iから選択される1つ又は複数の組み
合わせであり、
R´は、-H、C1-3のアルキル基から選択され、
前記PEGは、一般式IIまたはIIIで表される構造を有する直鎖ポリエチレングリコー
ル残基であり、
式中、pおよびqは、独立して、1~2280の整数から選択され、
または、前記PEGは、一般式IVまたはVで表される構造を有するY型ポリエチレング
リコール残基であり、
式中、iおよびhは、独立して、1~1140の整数から選択され、
または、前記PEGは、一般式VIで表される構造を有する多分岐ポリエチレングリコー
ル残基であり、
式中、kが、1~760の整数であり、jが、3~8の整数であり、
Yは、エンドキャップ基であり、H、C1-6アルキル基(具体的には、メチル基、エ
チル基、n-プロピル基、イソプロピル基、n-ブチル基、イソブチル基、tert-ブ
チル基、n-ペンチル基、n-ヘキシル基などであってもよい)、C3-6シクロアルキ
ル基(具体的には、シクロプロピル基、シクロブチル基、シクロペンチル基、シクロヘキ
シル基などであってもよい)、C6-10アリール基(具体的には、フェニル基、ナフチ
ル基などであってもよい)、-L-Tから選択され、
Lは、酸素(O)と末端基Tとの間の結合基であり、-(CH2)b-、-(CR3R
4)b-、-(CH2)bNH-、-NHCO(CH2)b-、-(CH2)bCONH
-および-CO(CH2)b-から選択される1つ又は複数の組み合わせであり、bが、

IL-2をIL-2α受容体に結合する工程(1)と、
PEG化し、すなわちPEGをIL-2に結合する工程(2)と、
IL-2をIL-2α受容体から単離する工程(3)とを含む、ことを特徴とする請求項
1~10に記載の製造方法。 - 結合部位制御可能なPEG化IL-2の製造方法であって、
IL-2α受容体親和カラムを調製する工程(1)と、
IL-2を親和カラムにてIL-2α受容体に結合する工程(2)と、
PEG化し、すなわちPEGをIL-2に結合する工程(3)と、
IL-2をグラジエント溶離によりIL-2α受容体から単離する工程(4)とを含む、
ことを特徴とする請求項11に記載の製造方法。 - 請求項11または12に記載の製造方法により製造された結合部位制御可能なPEG化
IL-2と、薬学的に許容される担体とを用いて製造された薬物組成物。 - 疾患の治療および/または予防のための薬物の製造における、請求項11または12に
記載の製造方法により製造された結合部位制御可能なPEG化IL-2と、薬学的に許容
される担体とを用いて製造された薬物組成物の使用。 - 前記疾患は、腫瘍、自己免疫疾患、ウイルス性疾患または細菌性疾患である、ことを特
徴とする請求項15に記載の使用。
