Abstract
Description
これまで、骨、心臓組織、軟骨、肝臓、肺、神経組織、皮膚及び膵臓組織などの組織を模倣するために、押出技術を用いて多くの研究が行われてきた(非特許文献1)。また、疾患及び薬物放出のインビトロモデリングを行うにも使用されている(非特許文献2)。
3Dバイオプリンティングで使用できるように材料が満たすパラメータは、プリントプロセス、特に、レオロジー特性(粘度、疑可塑性、粘弾性及び降伏強度)及び架橋機構などの物理化学的パラメータの影響を受けるプリンタ適性と、生物医学材料として使用することができる特性、すなわち、その分解速度論が細胞の細胞外マトリックス形成能力と一致しなければならないこと、及び分解生成物が毒にならないことに注目する生体適合性、がある。
ポリマーベースのバイオインクの前述の欠点を解決するために、いくつかの方策が挙げられる。
(1)主に物理的プロセス(イオン相互作用、水素結合又は疎水性相互作用)、化学プロセス(化学反応によって生成された共有結合)、又は両方の組み合わせに基づいて構造を安定化させるゲル化方法(非特許文献3)。
(2)通常、あるインクの構造的欠陥を別のインクの良好なプリンタ適性で補うために、ほぼ全ての既存バイオインクをそのまま混合物にした多成分又はハイブリッドインクの使用(非特許文献4)。
(3)大きな構造にしてプリントをサポートする犠牲インクの使用。
(4)サポートとして合成材料を使用。
現在までに、バイオインク組成物が市販されており、例えば、ゼラチンポリマーをベースとし、別の成長因子を組み合わせたGel4Cell(Bioink Solutions社)、ナノセルロースとアルギン酸をベースにしたCELLINK(CELLINK)、PEG/ゼラチン/ヒアルロン酸及びリン酸カルシウムをそれぞれベースにしたBio127及びBioGel(RegenHU)、Pluronic F127及びメタクリレートをそれぞれベースにしたBio127及びbioGel(Biobot)がある。このタイプのバイオインクの欠点は、これらを形成するゼラチンポリマーが、動物コラーゲンの加水分解から得られ、従って同じでないために、それらの製造バッチ毎の挙動を保証できないことにある。
モノマーB、C、X及びYを含むバイオポリマー、又は
モノマーB、C及びX、並びにモノマーB、C及びYを含む少なくとも2つのバイオポリマーを含むことを特徴とする。
ここで、
Bは、SEQ ID NO:2を含むアミノ酸シークエンス、
Cは、SEQ ID NO:3を含むアミノ酸シークエンス、
Xは、SEQ ID NO:4を含むアミノ酸シークエンス、

モノマーB、C及び少なくともモノマーX又はYを含むバイオポリマーを含む組成物を、バイオインクとして、好ましくはバイオマテリアルの2D又は3Dプリンティングのバイオインクとして使用することを特徴とする。
ここで、
Bは、SEQ ID NO:2を含むアミノ酸シークエンスであり、
Cは、SEQ ID NO:3を含むアミノ酸シークエンスであり、
Xは、SEQ ID NO:4を含むアミノ酸シークエンスであり、

- ELRシークエンス(モノマーB)及びモノマーCを含むモノマーを、シルク(モノマーY)及びジッパー(Xモノマー)シークエンスを含むモノマーと一緒にした組み合わせ;又は、ELRモノマーシルク(モノマーY)をシルクモノマー(モノマーY)と一緒に含むバイオポリマーと、ELRモノマー(モノマーB)とモノマーCをジッパーモノマー(モノマーX)と一緒に含むバイオポリマーとの組み合わせ。これで、モノマーXは、必要なプリンタ適性、すなわち、最初に構造を保持する能力を与え、一方、モノマーYは、経時的な構造の保持に寄与する。
ここで、
Bは、既に述べたように、ELRドメインの繰り返し(SEQ ID NO:7)を作るアミノ酸シークエンスであり、より好ましくは、モノマーBは、SEQ ID NO:2を含む。
Cは、ELRドメインの繰り返しでなるアミノ酸シークエンスであり、より好ましくは、モノマーCは、SEQ ID NO:3を含む。
Xは、“ジッパー”構造モチーフを含むアミノ酸シークエンスであり、より好ましくは、モノマーXは、SEQ ID NO:4を含む。
Yは、エラスチンドメインシークエンスとカイコボンビモリに由来する“シルク”と呼ばれるシークエンスの組み合わせを含むアミノ酸シークエンス(SEQ ID NO:8;GAGAGS)であり、より好ましくは、モノマーYは、SEQ ID NO:5を含む。
ここで、B、C、Dは上述のモノマーであり、
Zは、上で定義したモノマーXとYから選択され、

本発明組成物のバイオポリマーを含み、用いた種々のバイオポリマーのアミノ酸シークエンスをコードする合成ヌクレオチドシークエンスのデザイン及び取得は、WO/2010/092224に記載に従って行った。同様に、バイオポリマーの発現、精製及び特性評価は、WO/2010/092224に記載に従って行った。
得られたバイオポリマーの特性評価は、以下の技術を用いる。
- SDS存在下のアクリルアミドゲル電気泳動(PAGE):バイオポリマーの分子量を概略推測でき、さらに純度も評価できる。
- Q-Star分光計でのMALDI-TOF質量分析:ポリマーの分子量を正確に得る。
- Bruker・ARX300分光計でのプロトン核磁気共鳴(1H-NMR)スペクトル。
- Cary50分光光度計を使用するフーリエ変換赤外分光(FT-IR)スペクトル。
- WATERS2487検出器を備えたWATERS600HPLC勾配システムを用いたUV検出器を備えたHPLCクロマトグラフィー:アミノ酸組成を決定できる。
- Mettler・Toledo822eDSC装置での50mg/ml濃度の材料水溶液の示差走査熱量測定(DSC):ポリマーの逆転移温度が得られる。

バイオポリマー1の理論分子量は93175Daであり、ポリアクリルアミドゲル電気泳動(図1)及びMALDI-TOF質量分析により実験的に推定した分子量は92897Daであった。
バイオポリマー1について得られたHPLC、赤外吸収(IR)及び核磁気共鳴(NMR)スペクトルをそれぞれ図2、3、4に示す。
pH7.8のMQ中でDSC測定により得られた転移温度は19.10℃であり、一方、pH7.65の1xPBS中では14.66℃であった。

pH6.14のMQ中でのDSC測定により得られた転移温度は20.08℃であり、一方、pH6.40の1xPBS中では16.92℃であった。

pH6.48のMQ中でのDSC測定により得られた転移温度は17.58℃であり、pH6.02の1xPBSでは14.92℃であった。

pH6.59でMQ中でのDSC測定により得られた転移温度は20.84℃であり、pH7.24で1xPBSでは17.26℃であった

バイオポリマー6の理論分子量は126393Daであり、ポリアクリルアミドゲル電気泳動(図21)とMALDI-TOF質量分析法(図22)により実験的に推定した分子量は125857Daであった。
バイオポリマー6に対してのIR及びNMRスペクトルをそれぞれ図23及び24に示す。
pH7.50でMQ中でのDSC測定により得られた転移温度20.42℃であり、pH7.50で1xPBSでは17.41℃であった。
表2に記した種々のバイオポリマー、又はこれらの混合物を用いた3Dプリンティングは、それらの各々の逆転移温度を考慮して行われる。この転移温度は、本発明のバイオポリマー組成物の特定の性状と共に、単純な温度変化によって潜在的にバイオインクをゲル化させる。
バイオポリマー4(重量%80%)+前硬化バイオポリマー5(20重量%);バイオポリマー4(重量%60%)+前硬化バイオポリマー5(重量40%)及びバイオポリマー4(重量%20%)+前硬化バイオポリマー5(80重量%);バイオポリマー4(重量60%)+前硬化バイオポリマー5(40重量%)の割合のプリントは、プリントされた混合物で最高のPr値を示し、故にこの混合物は3Dプリンティングに最適と考えることができる。前硬化バイオポリマー5の割合が高いときに最も低い値が得られる。
バイオインクとして使用すべく本発明の組成物をレオロジー分析し、機械的特性を分析した。プリンタ適性がより大きいことを示したこれら組成物は、バイオポリマー4、バイオポリマー6、前硬化バイオポリマー5及びバイオポリマー4(60重量%)+前硬化バイオポリマー5(40重量%)を含む組成物に対応して選んでいる。
次に、本発明に記載した組成物の選ばれた流体中での安定性を、本実施例の場合に1xPBS中で分析した。本発明に記載した組成物でプリントされた構造がインビトロ培養、又は組織モデルに使用されるであろうことから、その構造は、長期間水性媒体中で安定した状態に保たれねばならない。この例では、シリンダーは、直径6mm、高さ1.5cmであり、プリント操作は、直径0.25mmのノズルを通して行った。評価した組成物は、バイオポリマー4、前硬化バイオポリマー5、バイオポリマー6、又はバイオポリマー4(60重量%)+前硬化バイオポリマー5(重量40%)の混合物を含む組成物であった。
本発明の組成物のバイオインクとしてのプリンタ適性、及び経時安定性を評価した後、ヒト線維芽細胞株HFF-1を用いてそれらの細胞毒性と細胞生存率を評価した。この目的に、直径5mm、高さ1mmで横向きに正方形空隙1mmがある多孔質円形格子をプリントしている。
次に、バイオポリマー6と共にプリントしたときにヒト線維芽細胞HFF-1によって示される細胞生存率及び形態を評価する。
Claims (27)
- モノマーB、C、X及びYを含むバイオポリマーを含む、又は、
モノマーB、C及びX、並びにモノマーB、C及びYをそれぞれ含む少なくとも2つのバイオポリマーを含むことを特徴とする組成物。
ここで、
Bは、SEQ ID NO:2を含むアミノ酸シークエンス、
Cは、SEQ ID NO:3を含むアミノ酸シークエンス、
Xは、SEQ ID NO:4を含むアミノ酸シークエンス、
Yは、SEQ ID NO:5を含むアミノ酸シークエンスである。 - 前記バイオポリマーは、さらに、モノマーDを含み、
前記モノマーDは、細胞結合性アミノ酸シークエンスであることを特徴とする請求項1に記載の組成物。 - 前記モノマーDは、
RGD(SEQ ID NO:9)、LDT(SEQ ID NO:27)、SEQ ID NO:10、SEQ ID NO:17、SEQ ID NO:18、SEQ ID NO:19、SEQ ID NO:28、及びSEQ ID NO:29からなる群から1つ選ばれるシークエンスを含むことを特徴とする請求項2に記載の組成物。 - 前記モノマーDは、さらにSEQ ID NO:6を含むことを特徴とする請求項2又は3に記載の組成物。
- 前記バイオポリマーは構造(I):[(Bb-Cc)-Zz]n-Dd、
の構造を有する請求項1乃至請求項4のいずれかの組成物であって、
前記組成物は、
ZがSEQ ID NO:4である構造(I)の第1のバイオポリマーと、
ZがSEQ ID NO:5である構造(I)の第2のバイオポリマーとを含むことを特徴とする請求項1乃至4のいずれか1項に記載の組成物。
ここで、
B、C、Dは、請求項1乃至請求項4で定義しており、
Zは、請求項1で定義したモノマーX及び/又はYから選択され、
bは、5~15の値であり、
cは、50~70の値であり、
zは、1~5の値であり、
nは、1~5の値であり、
dは、0~3の値である。 - 前記第1のバイオポリマーは、前記組成物中に40~60重量%の濃度であり、
前記第2のバイオポリマーは、前記組成物中に60~40重量%の濃度であることを特徴とする請求項5に記載の組成物。 - 前記バイオポリマーは、構造(II):Z1z-[(Bb-Cc)-Z2z]n-Dd、
の構造を有することを特徴とする請求項1乃至4のいずれか1項に記載の組成物。
ここで、
B、C、Dは、請求項1乃至請求項4で定義しており、
Z1は、SEQ ID NO:4又はSEQ ID NO:5を含むアミノ酸シークエンスであり、
Z2は、SEQ ID NO:5 又はSEQ ID NO:4を含むアミノ酸シークエンスであり、
好ましくは、Z1は、SEQ ID NO:4を含むアミノ酸シークエンス、Z2は、SEQ ID NO:5を含むアミノ酸シークエンスであり、
b、c、z、n及びdは、請求項5で定義している。 - bが10の値、cが60の値、zが1の値、nが2の値、dが0又は1の値を有することを特徴とする請求項5乃至7のいずれか1項に記載の組成物。
- 前記バイオポリマーは、SEQ ID NO:16から選ばれる、
又は、SEQ IDNO:14とSEQ IDNO:15のバイオポリマーの組み合わせから選ばれることを特徴とする請求項2に記載の組成物。 - 前記組成物は、さらに、細胞、生理活性分子、活性成分、又はそれらの組み合わせを含むことを特徴とする請求項1乃至9のいずれか1項に記載の組成物。
- 前記組成物は、ヒドロゲルの形態であることを特徴とする請求項1乃至10のいずれか1項に記載の組成物。
- 請求項1乃至11のいずれか1項に記載の組成物のバイオポリマーのアミノ酸シークエンスをコードするヌクレオチドシークエンスを含むことを特徴とする核酸。
- 請求項12に記載の核酸をトランスフェクトしたことを特徴とする単離細胞。
- バイオインク、好ましくはバイオマテリアルの2D又は3Dプリンティング用のバイオインクとして使用することを特徴とする請求項1乃至11のいずれか1項に記載の組成物の使用。
- 臓器及び/又は組織を得るサポートの調製のため、又は活性成分若しくは細胞をカプセル化するための使用であって、
請求項1乃至11のいずれか1項に記載の組成物、
請求項12に記載の核酸、
又は請求項13に記載の細胞を使用することを特徴とする使用。 - 請求項1乃至11のいずれか1項に記載の組成物を含むことを特徴とするバイオインク。

- モノマーB、C、及び少なくともモノマーX又はYを含むバイオポリマーを含む組成物を、バイオインクとして、好ましくはバイオマテリアルの2D又は3Dプリンティングのバイオインクとして使用することを特徴とする組成物の使用。
