各GLP-2ペプチボディを、ナノ示差走査蛍光定量法(NanoDSF)を用いて融解温度を決定することによって試験した。NanoDSFは、温度傾斜を用いた、ある範囲の温度にわたるタンパク質安定性の測定である。トリプトファンの安定性は、350nmでの蛍光の330nmでの蛍光に対する比に反映されるように、蛍光によって測定される。このアッセイから、1つまたは複数の融解温度が決定される。特定の状態のタンパク質は融解温度を有すると理解されているため、観察される溶融温度の数は、異なる状態の数を反映する。
GLP-2ペプチボディのEC50を、DiscoverXのcAMP Hunter(商標)eXpress GLP2R CHO-K1 GPCRアッセイを使用してinvitroで評価した。cAMP Hunter(商標)アッセイは、酵素フラグメント相補性(EFC)に基づく。EFCアッセイでは、酵素ドナーをcAMPに融合させる。GLP-2Rの活性化による細胞内cAMPの増加は、抗体のED-cAMPと競合する。非結合ED-cAMPは、酵素アクセプターを補完して活性なβガラクトシダーゼを形成し、それがその後に発光シグナルを生成する。
