Abstract
Description
「抗体」は、本明細書で使用する場合、免疫グロブリン分子の可変領域に位置する少なくとも1つの抗原認識部位を介して、標的、すなわち「抗原」、例えば、糖質、ポリヌクレオチド、脂質、ポリペプチドなどに特異的に結合することができる免疫グロブリン(Ig)分子を指す。本融合タンパク質の抗体またはその抗原結合タンパク質は、疾患環境および/または特異的細胞サブセットへの免疫サイトカインの標的送達を媒介する。好ましい標的抗原は、他所では低レベルでとどまりながら、罹患組織において過剰発現している抗原である。このような抗原は、長年多数の研究の対象となっているため、当業者に周知である。例えば、本免疫サイトカインの抗体部分は、悪性細胞の表面に過剰発現している抗原(例えば、上皮細胞接着分子、EGFR、IGF−1R、GD2ジシアロガングリオシド、HER2/neu、CD20およびCD30)、ならびに腫瘍および慢性炎症部位において認められる血管新生抗原(例えば、フィブロネクチン、スプライスバリアントEDA/EDBおよびテネイシンCのA1ドメイン)を標的とし得る。
「切断可能ペプチドリンカー」は、本明細書で使用する場合、酵素的に切断可能な(例えば、切断部位において)、抗体、またはその抗原結合フラグメントに共有結合的に結合した、およびサイトカイン、またはそのフラグメントに共有結合的に結合した多価リンカーを指す。本発明によれば、切断可能ペプチドリンカーの加水分解(タンパク質分解切断)時、サイトカイン部分、好ましくはIL−15が放出され、その治療活性を発揮することが可能となる。好ましい実施形態において、切断可能ペプチドリンカーは、免疫サイトカインの一部として組換え発現する。他の複数の実施形態において、切断可能ペプチドリンカーは、例えば、コンジュゲート化学を用いて、リンカーに結合した官能(反応)基を抗体、またはその抗原結合フラグメントと反応させることによって形成されるリンカーである。さらに他の複数の実施形態において、切断可能ペプチドリンカーは、例えば、コンジュゲート化学を用いて、リンカーに結合した官能(反応)基をサイトカイン、またはそのフラグメントと反応させることによって形成されるリンカーである。好ましい実施形態において、切断可能ペプチドリンカーは、サイトカイン、またはそのフラグメントを、抗体、またはその抗原結合フラグメントの重鎖と連結させる。別の実施形態において、切断可能ペプチドリンカーは、サイトカイン、またはそのフラグメントを、抗体、またはその抗原結合フラグメントの軽鎖と連結させる。切断可能ペプチドリンカーは、サイトカイン、またはそのフラグメントを、抗体、またはその抗原結合フラグメントの重鎖および軽鎖のうちの1つのN末端と連結させ得る。切断可能ペプチドリンカーは、サイトカイン、またはそのフラグメントを、抗体、またはその抗原結合フラグメントの重鎖および軽鎖のC末端と連結させることもあり得る。最も好ましくは、切断可能ペプチドリンカーは、サイトカイン、またはそのフラグメントを、抗体、またはその抗原結合フラグメントの重鎖のN末端またはC末端と連結させる。
用語「サイトカイン」は、本明細書で使用する場合、免疫系に対する効果を有する小さな分泌調節タンパク質のファミリーのメンバーを指す。サイトカインは、細胞間情報交換に関与し、多くの細胞機能、例えば、細胞の生存および成長、ならびに多くの遺伝子の発現の誘導を調節する。このため、サイトカインの分泌は、多面的および効率的な様式での免疫シグナル伝達の急速な伝播を可能にする。サイトカインは、リンパ球および/または他の細胞に対する種々の効果を発揮することによって、免疫応答の性質、強度および持続時間を調節する。実際に、サイトカインは通常、炎症誘発性および抗炎症性サイトカインに分類される。いくつかのサイトカインは、癌と戦うために免疫系を動員することもできる(例えば、Floros & Tarhini, Semin Oncol. 42(4): 539-548, 2015参照)。サイトカインは、免疫細胞および非免疫細胞を含む多くの細胞型によって産生され得る。サイトカインの例としては、とりわけ、インターロイキン、リンホカイン、モノカイン、インターフェロン、コロニー刺激因子、およびケモカインが挙げられる。「サイトカイン」は、本明細書で使用する場合、IL−1、IL−2、IL−3、IL−4、IL−5、IL−6、IL−7、IL−8、IL−9、IL−10、IL−11、IL−12、IL−13、IL−15、IL−16、IL−17、IL−18、IL−19、IL−20、IL−21、IL−22、IL−23、IL−24、IL−26、IL−28、IL−29、IL−33、IL−36、IL37、IL−38、IFN−α(IFN−α1/13、IFN−α2、IFN−α4、IFN−α5、IFN−α6、IFN−α7、IFN−α8、IFN−α10、IFN−α14、IFN−α16、IFN−α17、およびIFN−α21を含む)、IFN−β、IFN−γ、IFN−λ、TNF−α、TNF−β、TGF−β1、M−CSF、G−CSF、GM−CSF、ならびにCXL10のいずれか1つであり得る。本発明によれば、サイトカインは、好ましくは、ICCに連結すると不活性化または減弱され、プロテアーゼによるICCの切断後にのみ活性化した。
(i)以下と融合している抗体または抗原結合フラグメント、
(ii)切断可能ペプチドリンカー、および
(iii)サイトカイン、好ましくは、IL−15、CXCL10、IL−36、もしくはIFN−α、またはその機能的フラグメント
を含んでなる融合タンパク質に関する。
本発明者らは、本融合タンパク質における使用に好適なリンカーを同定することが可能であることを示した。実験データは、いくつかのリンカーが、in vitroにおいて切断されるか否かに従って識別できることを示している。これらのin vitroでの結果の妥当性は、このように同定されたリンカー総てがin vivoにおいて切断され、それにより、腫瘍の部位で活性サイトカインが遊離されるという事実によって、強調されている。
(i)試験される切断可能ペプチドリンカーを含んでなる、本明細書に記載の融合タンパク質を提供すること;
(ii)前記融合タンパク質を関連するプロテアーゼと接触させること;および
(iii)前記融合タンパク質の切断を検出すること
を含んでなる。
(i)試験されるサイトカインまたはその変異体を含んでなる、本明細書に記載の融合タンパク質を提供すること;
(ii)前記融合タンパク質を関連するプロテアーゼと接触させること;および
(iii)前記サイトカインの活性を検出すること
を含んでなる。
上記の融合タンパク質をコードするヌクレオチド配列を含んでなるポリヌクレオチドも、本明細書において提供される。例えば、上記で定義されるように、本明細書において提供される融合タンパク質または改変融合タンパク質をコードするポリヌクレオチドと、例えば、上記で定義したように、高いストリンジェンシー、中程度または低いストリンジェンシーのハイブリダイゼーション条件下でハイブリダイズするポリヌクレオチドも、本明細書において提供される。
本明細書に記載の本免疫サイトカインを発現させるために、種々の発現系が使用され得る。一つの側面において、このような発現系は、目的のコード配列が産生され、その後に精製され得るビヒクルを表すが、適当なヌクレオチドコード配列で一過性にトランスフェクトされた場合、in situで本発明の抗体を発現し得る細胞も表す。
1.配列
全ICC(項目1.配列参照)をコードする配列を、シグナルペプチド(Thermo Fisher Scientific社)を含むHindIII/BamHI制限部位を用いることにより、pCDNA3.4ベクターにクローン化した。NanoLuc(登録商標)DNA配列を、Promega社から得た。リンカー修飾を、Q5変異誘発(New England Biolabs社)により達成した。Expi293 Expression Medium(Thermo Fisher Scientific社)中で密度2.5×106/mlまで成長させ、ポリエチレンイミン(PEI、Polyscience社、DNA/PEI比:1/4)を用いて1.25μg/mlのDNA(HC/LC:1/1w/w)でコトランスフェクトしたExpi HEK293細胞(Thermo Fisher Scientific社)における一過性タンパク質発現により、融合タンパク質を得た。次いで、2mMのバルプロ酸(VPA、Sigma−Aldrich社)を、トランスフェクションの3時間後に加えた。産生された融合タンパク質を含有する上清を、トランスフェクションの6日後に回収した。タンパク質を、プロテインA−セファロース上でのアフィニティークロマトグラフィーにより精製し、25mMクエン酸ナトリウム、150mM NaCl、6%サッカロースpH5.5に対する一晩の透析により処方した。実現した構築物のいくつかを、図1に要約する。
2.1.材料と方法:
総ての精製ICCを、非還元加熱条件下および還元加熱条件下でSDS−PAGEにより特徴付けた。c9G4−PVGLIG−hIL15、NHS76−PVGLIG−hIL15およびH16/L16−PVGLIG−hIL15のSDS−PAGEでの移動を、図22に示す。SDS PAGEから推定された見かけのICC分子量は、理論計算で予想されたものと一致していた。各ICCに対して少なくとも80%が、完全ICC(H2L2)であった。ICCの単量体含量を、サイズ排除クロマトグラフィーにより決定した。
MSクロマトグラムピークに関して、m/zスペクトルを抽出し、m/zスペクトルデコンボリューション後に質量を決定し、脱グリコシル化ICCおよび脱グリコシル化fc/2の計算質量を、実験質量と比較した。
文献からの5つのリンカーを、PDL1 Ab配列上の4つの異なる部位:N末端HC、N末端LC、C末端HCおよびC末端LCと融合した場合のMMP−9に対する基質として評価した。非切断可能と記載された「GIVGPL」リンカー(Chau et al. Bioconjug Chem (2004) 15(4):931-41)を、陰性対照として使用した。構築物および結果の要約を、図7に示す。
総容量20μlでの20mMトリスpH7.5、10mM CaCl2および100mM NaClを含有するアッセイ緩衝液中の40ngの組換えMMP−9またはMMP−2(モル比25:1)の存在下で、2μgの精製ICCをインキュベートした。サンプルを、穏やかな撹拌下(300rpm)で、30℃で2.5時間インキュベートした。ローディング緩衝液+還元剤(4倍XTサンプル緩衝液および20倍還元剤、BioRad社)を加え、サンプルを90℃で5分間加熱することにより、切断反応を停止させた。
3.2.1.抗PDL1抗体および2つの異なる分子:IL2およびNanoLuc(登録商標)のHCまたはLC C末端融合体における異なるリンカーによる切断性:
結果は図4に示す。

図4、5および6の結果を、図7に要約する。プロテアーゼによる正確な切断を可能にした、2つの融合部位:Ab重鎖のN末端およびC末端が同定された。
4.1.材料と方法
12nM MMP−9を含有するおよび含有しない濃度100μg/mlの緩衝液(50mMトリスpH7.5、150mM NaCl、20mM CaCl2)、血清、血漿、および新鮮血に、ICCを添加した。100μlのアリコートを、Protein LoBind Tube(Eppendorf社)中にて+37℃でインキュベートした。24時間後、サンプルをオーブンから取り出し、サンプルの処理および分析まで、−80℃で保存した。
4.2.1 ICCにおけるMMP−9切断部位の検証
MMP−9の非存在下では、抗PDL1−PVGLIG−NanoLuc(登録商標)および抗PDL1−GIVGPL−NanoLuc(登録商標)の両方に関して、切断されたフラグメントは認められず、このため、両リンカーとも、濃度12nMの緩衝液中で安定であった。
結果を図9に示す。MMP−9の添加ありまたはなしで、マウスヘパリン血漿、マウス、カニクイザル、ヒト血清およびマウスヘパリン全血において、類似のプロファイルがT0およびT24にて得られた。MMP−9は、試験した総ての生体液中で阻害され、LC−MSプロファイルの例を図9に示した。
ICCと連結した場合またはMMP9による切断後のhIL15の生物活性を評価および比較するために、IL−15受容体二量体化の試験を行った。IL15は、その活性型において、その2つの受容体サブユニットIL2Rβ/IL2Rγの二量体化を介してシグナルを伝達する。
5.1.1.材料および試薬
組換えヒト組換えプロMMP−9を、R&D Systems社から購入し、50mMトリス、150mM NaCl、10mM CaCl2、0.05% Brij−35(w/v)、pH7.5を含有する緩衝液中の1mM酢酸4−アミノフェニル水銀(APMA)により活性化させた。次いで、Zeba(商標)Spin Desalting Columns(ThermoFisher Scientific社)を用いてAPMAを除去し、APMA不含MMP−9を、必要になるまで−80℃で直ちに保存した。組換えヒトIL15を、PeproTech社から購入した。PathHunter(登録商標)U2OS IL2Rβ/IL2Rγ/(IL2Rα)Dimerisation Bioassay(DiscoverX、Eurofins社)を用いて、IL15活性をモニタリングした。このアッセイは、2つの受容体サブユニットIL2RβおよびIL2RγのIL15誘導二量体化の検出を可能にする。
3つのIL15ベースの免疫サイトカイン:NHS76−PVGLIG−hIL15、H16/L16−PVGLIG−hIL15およびc9G4−PVGLIG−hIL15の活性を評価した。組換えhIL15およびhMMP−9の効果も、同じ手順を用いて評価した。50mMトリス、150mM NaCl、10mM CaCl2 pH7.5を含有するアッセイ緩衝液中で、組換えhMMP−9の存在下(+MMP−9)または非存在下(−MMP−9)で、全サンプルを2時間インキュベートした。切断効率をSDS−PAGE分析により制御し、サンプルを、処理するまで−20℃で直ちに保存した。U2OS IL2Rβ/IL2Rγ/IL2Rα細胞を、切断ICCまたは非切断ICCのいずれかおよび対照により、6時間処置した。次いで、検出試薬を加え、化学発光強度を、マイクロプレートリーダー(Infinite M1000Pro、Tecan社)で記録した。データ解析は、Prism7.01ソフトウェア(GraphPad)で行った。
3つの試験したICCに関するhIL15誘導受容体二量体化の結果を、hIL15濃度の関数として図10に示す。
6.1.材料と方法:
6.1.1.マウスの移植およびICCの注入
6週齢の免疫適格BALB/cマウスを、総てのin vivo評価に使用した。それらを滅菌フィルタートップケージに収容し、無菌条件下で飼育し、フランスおよび欧州のガイドラインに従って操作した。
投与後0、3、6、24および48時間に動物を屠殺し、血液サンプルを、心臓穿刺によりNa−ヘパリン加チューブに回収した。血液サンプルを4℃にて1500gで15分間遠心分離し、血漿を96ウェルU底プレートに回収した。血漿サンプルを、分析の前に凍結した(−80℃)。
腫瘍を採取し、液体窒素中で急速凍結し、−80℃で保存した。
血清サンプルおよび腫瘍サンプルを、ウエスタンブロットにより分析した。各in vivo実験につき、ウエスタンブロットを3回行った。
ウエスタンブロットのデンシトメトリー分析を、Image Lab(Bio−Rad社)を用いて行う。
6.2.1 マウス血清におけるICCのin vivo安定性
ICCの安定性を、時間の関数としてマウス血清において評価した。図11は、ICC注入後3時間、6時間および24時間に、ウエスタンブロットにより得た結果を示す。循環ICCの量は、試験した各ICC構築物に関して、時間の関数として劇的に減少する。理論に拘束されるものではないが、この結果は、腫瘍におけるICCの分離および/または自然のin vivoでのAb分解により、最も容易に説明される。
ICCの安定性を、時間の関数としてマウス腫瘍サンプルにおいて評価した。図14は、ICC注入後3時間、6時間および24時間に、ウエスタンブロットにより得た結果を示す。
2つのICCの挙動を、時間の関数として、RENCA移植マウスにおける血漿サンプルと腫瘍サンプルにおいて比較した。
7.1.材料と方法:
7.1.1 材料と方法
組換えヒトIL15を、PeproTech社から購入した。
・NHS76−PVGLIG−IL15;
・H16/L16−PVGLIG−IL15;および
・c9G4−PVGLIG−IL15
3つのIL15ベースの免疫サイトカイン、すなわち、NHS76−IL15、H16/L16−IL15、およびc9G4−IL15の活性を、マウスおよびヒトCD3+T細胞を活性化するそれらの能力に関して評価した。hIL15+/−MMP9、MMP9および個々の抗体の効果も、同じ手順を用いて評価した。
マウスT細胞を活性化するICCの能力を、対照と比較した。T細胞活性化を、CD69またはCD25のT細胞発現により測定した。図23AおよびBは、NHS76−PVGLIG−IL15およびc9G4−PVLGIG−IL15は、MMP−9の存在下でCD69およびCD25の発現を誘導するが、MMP−9の非存在下では誘導しないことを示している。このため、ICCが非切断である場合、マウスT細胞の活性化は観察されない。実験の妥当性を確認するために、陰性対照(抗体単独、MMP−9単独、緩衝液単独)または陽性対照(IL15およびIL15+MMP−9単独)も試験した。
ICCの一部である場合またはMMP9による切断後のhIL36γの生物活性を評価および比較するために、IL36γ活性を、ヒト類表皮癌A431細胞培養物におけるCXCL8産生を測定することにより、アッセイした。このアッセイは、IL36γシグナル伝達経路はCXCL8分泌を引き起こし得るという所見に基づいている。
8.1.1.材料および試薬
組換えヒト組換えプロMMP−9を、R&D Systems社から購入し、50mMトリス、150mM NaCl、10mM CaCl2、0.05% Brij−35(w/v)、pH7.5を含有する緩衝液中の1mM酢酸4−アミノフェニル水銀(APMA)により活性化させた。次いで、Zeba(商標)Spin Desalting Columns(ThermoFisher Scientific社)を用いてAPMAを除去し、APMA不含MMP−9を、必要になるまで−80℃で直ちに保存した。組換えヒトIL36γを、R&D Systems社から購入した。IL8を、Human IL8 DuoSet ELISA(R&D Systems社)を用いて投与した。
IL36 γベースの免疫サイトカイン(H16/L16−PVGLIG−IL−36)の活性を評価するために、扁平上皮癌A431細胞を、ICCまたは異なる対照分子で24時間インキュベートした。50mMトリス、150mM NaCl、10mM CaCl2 pH7.5を含有するアッセイ緩衝液中で、組換えhMMP−9の存在下(+MMP−9)または非存在下(−MMP−9)で、全サンプルを試験した。次いで、細胞培養培地を回収し、hIL8をELISAにより投与した。組換えhIL36γおよびH16/L16抗体(+/−hMMP−9)の効果も、同じ手順を用いて評価した。データ解析は、Prism7.01ソフトウェア(GraphPad)で行った。
ICCおよび対照に関するA431培養物におけるhIL8産生の結果を、図26に示す。データは、hIL36γ濃度の関数としてのhIL8発現レベルに対応するO.D(450nm)で表している。
ICCと連結した場合またはMMP9による切断後のhIFNα2a生物活性を評価および比較するために、IFNα2a活性を、細胞ベースのルシフェラーゼレポーターバイオアッセイを用いてアッセイした。
9.1.1.材料および試薬
組換えヒト組換えプロMMP−9を、R&D Systems社から購入し、50mMトリス、150mM NaCl、10mM CaCl2、0.05% Brij−35(w/v)、pH7.5を含有する緩衝液中の1mM酢酸4−アミノフェニル水銀(APMA)により活性化させた。次いで、Zeba(商標)Spin Desalting Columns(ThermoFisher Scientific社)を用いてAPMAを除去し、APMA不含MMP−9を、必要になるまで−80℃で直ちに保存した。組換えヒトIFNα2aを、Pbl Assay Science社(rhIFNα2a)から購入した。
IFNα2a活性を、GloResponse(商標)ISRE−luc2P/HEK293(Promega社)を用いてモニタリングした。ISRE−luc2P/HEK293細胞株は、HEK293細胞内に安定して組み込まれたISREの制御下のホタルルシフェラーゼ遺伝子を含有する。IFNα2aのその受容体への結合は、JAK経路の活性化をもたらし、それにより、ルシフェラーゼ遺伝子のISRE依存的転写を促進する。次いで、発光を、基質の添加時に検出し、ルミノメーターで定量化することができる。
3つの試験したICCに関するhIFNα2a活性アッセイの結果を、hIFNα2a濃度の関数として図27に示す。各化合物またはICCのEC50を、表10に示す。
ICCと連結した場合またはuPAによる切断後のhIL15の生物活性を評価および比較するために、IL15活性の生物学的試験を用いた(Promega IL15 Bioassay)。
10.1.1.材料および試薬
以下の組換えウロキナーゼ(=uPA)を、Abcam社から購入した:ヒト用ab92767。組換えヒトIL15を、PeproTech社から購入した(200−15)。
評価する構築物を、uPAでPBS中にて、37℃で24時間インキュベートする。
IL15細胞を、切断ICCもしくは非切断ICCのいずれかまたは対照で、6時間処置した。次いで、検出試薬(Bio−Glo(商標)ルシフェラーゼアッセイ試薬)を加え、化学発光強度を、マイクロプレートリーダー(Mithras、Berthold社)で記録した。データ解析は、Prism7.01ソフトウェア(GraphPad)で行った。
ICCおよび対照に関するhIL15活性の結果を、図28に示す。データは、STAT−5活性化の関数としてのhIL15活性レベルに対応する相対発光量(RLU)で表している。
ICCと連結した場合またはuPAによる切断後のhIFNαの生物活性を評価および比較するために、IFNα活性の生物学的試験を用いた(GloResponse ISRE−luc2P)。
11.1.1.材料および試薬
組換えヒトウロキナーゼ(=uPA)を、Abcam社から購入した(ab92767)。組換えヒトIFNαを、PBL社から購入した(11101−02)。IFNα活性を、GloResponse ISRE−luc2P(Promega CS190701)を用いてモニタリングした。これは、ルシフェラーゼレポーター遺伝子luc2P(Photinus pyralis)の転写を駆動するインターフェロン刺激応答エレメント(ISRE)を用いてIFNα刺激または阻害を測定するように設計された、生物発光HEK293細胞ベースアッセイである。IFNαがその受容体に結合すると、受容体媒介経路シグナル伝達が、基質の添加時に検出でき、かつルミノメーターで定量化することができる発光を誘導する。
評価する構築物を、uPAでPBS中にて、37℃で24時間インキュベートする。切断効率をSDS−PAGE分析により制御し、サンプルを、処理するまで−20℃で直ちに保存した。
細胞表面上のIGF1R結合部位を飽和させるために、ISRE−luc2P/HEK293細胞を、抗IGF1R抗体10μgで1時間前処置した。
ICCおよび対照に関するhIFNα活性の結果を、図29Aおよび29Bに示す。データは、ISRE活性化の関数としてのhIFNα活性レベルに対応する相対発光量(RLU)で表している。
図29Aから分かるように、rhIFNαおよびrhIFNα+uPAは、発光を高度に活性化し、このため、実験形式が検証された。

12.1.1.材料および試薬
組換えヒトウロキナーゼ(=uPA)を、Abcam社から購入した(ab92767)。組換えヒトCXCL10を、Peprotech社から購入した(300−12)。CXCL10活性を、PathHunter eXpress CXCR3 CHOK1 β−アレスチンGPCRアッセイ(DiscoverX 93−0271E2)を用いてモニタリングした。これは、CXCR3受容体に対するCXCL10の結合により誘導されたβ−アレスチン動員を測定するように設計された、生物発光CHO細胞ベースアッセイである。
評価する構築物を、uPAでPBS中にて、37℃で1時間インキュベートする。切断効率をSDS−PAGE分析により制御し、サンプルを、処理するまで−20℃で直ちに保存した。
CHO−K1 CXCR3細胞を、切断ICCもしくは非切断ICCのいずれかまたは対照で、1時間30分処置した。次いで、検出試薬を加え、化学発光強度を、マイクロプレートリーダー(Mithras、Berthold社)で1時間後に記録した。データ解析は、Prism7.01ソフトウェア(GraphPad)で行った。
ICCおよび対照に関するhCXCL10活性の結果を、図30A、30Bおよび30Cに示す。データは、CXCR3 β−アレスチン動員の関数としてのhCXCL10活性レベルに対応する相対発光量(RLU)で表している。
組換えhCXCL10および組換えhCXCL10+uPAの両方とも、発光シグナルの強力な活性化をもたらし、これにより、実験形式が検証された。uPAの非存在下では、9G4−SGRS−CXCL10 ICCは、有意な活性を示さなかった。uPAによる同じ分子の前処置は、CXCR3 β−アレスチン動員を介して認められた活性の回復をもたらした(図30A)。
組換えhCXCL10および組換えhCXCL10+uPAの両方とも、発光シグナルの強力な活性化をもたらし、これにより、実験形式が検証された。uPAの非存在下では、NHS76−SGRS−CXCL10 ICCは、有意な活性を示さなかった。uPAによる同じ分子の前処置は、CXCR3 β−アレスチン動員を介して認められた活性の部分的回復をもたらした(図30B)。親NHS76−SGRS−CXCL10の部分的タンパク質分解がおそらく原因で、完全な活性は認められなかった。
組換えhCXCL10および組換えhCXCL10+uPAの両方とも、発光シグナルの強力な活性化をもたらし、これにより、実験形式が検証された。uPAの非存在下では、H16/L16−SGRS−CXCL10 ICCは、有意な活性を示さなかった。uPAによる同じ分子の前処置は、CXCR3 β−アレスチン動員を介して認められた活性の部分的回復をもたらした(図30C)。親H16/L16−SGRS−CXCL10の部分的タンパク質分解がおそらく原因で、完全な活性は認められなかった。

(ii)切断可能ペプチドリンカーが、抗体またはその抗原結合フラグメントの重鎖に対してN末端またはC末端で融合している、
請求項1〜9のいずれか一項に記載の融合タンパク質。 - 療法における使用のための、請求項1〜10のいずれか一項に記載の融合タンパク質。
- 哺乳動物、好ましくは、ヒトにおける癌の処置における使用のための、請求項1〜10のいずれか一項に記載の融合タンパク質。
- 前記使用が、前記哺乳動物の免疫細胞、好ましくは、T細胞または単球の活性化を含んでなる、請求項12に記載の使用のための融合タンパク質。
- 請求項1〜9のいずれか一項に記載の少なくとも1つの融合タンパク質および場合により薬学上許容可能な賦形剤を含んでなる、医薬組成物。
- サイトカインまたはその変異体を選択する方法であって、
(i)試験されるサイトカインまたはその変異体を含んでなる、請求項1〜10のいずれか一項に記載の融合タンパク質を提供すること;
(ii)前記融合タンパク質を関連するプロテアーゼと接触させること;および
(iii)前記サイトカインの活性を検出すること
を含んでなる、方法。 - 切断可能ペプチドリンカーを同定する方法であって、
(i)試験される切断可能ペプチドリンカーを含んでなる、請求項1〜10のいずれか一項に記載の融合タンパク質を提供すること;
(ii)前記融合タンパク質を関連するプロテアーゼと接触させること;および
(iii)前記融合タンパク質の切断を検出すること
を含んでなる、方法。 - 前記方法の工程(iii)が、前記融合タンパク質のサイトカインの活性を測定することを含んでなる、請求項16に記載の方法。
