Abstract
Description
1.分野
本発明は、一般に、免疫学の分野に関する。より詳細には、免疫チェックポイント遮断療法の結果として生じる自己免疫疾患病因論の動物モデル、および上記動物モデルを使用して、上記疾患病因論を軽減する因子をスクリーニングする方法に関する。
T細胞活性化は、自己を非自己から区別しかつ自己免疫を防止する能力を維持しながら、外来抗原に対して迅速かつ高感度な応答を生じることを可能にする、精巧に調節された生物学的プロセスである。この注目すべきプロセスの根底にある重要な概念は、多数の別個のシグナルが、ナイーブT細胞を十分に活性化する、または初回刺激するために要求されることである。これらのきっかけは、T細胞レセプター(TCR)による同種抗原認識(シグナル1)およびCD28陽性共刺激(シグナル2)が挙げられる。CD28陽性共刺激は、専門の抗原提示細胞によって提供されることから、これは、強いT細胞活性化のために細胞外部の要求を強いる。その次の重要な工程は、陰性共刺激(negative co−stimulation)であり、これは、シグナル1および2の阻害を通じてT細胞活性化を弱めるフィードバック調節機構である。CTLA4およびPD−1は、別個の分子機構を通じてT細胞活性化を減弱する主要な陰性共刺激分子である。CTLA4は、CD28陽性共刺激の競合的阻害を通じてT細胞活性化を減弱するが、PD−1は主に、ホスファターゼSHP2を介して、近くのT細胞レセプター(TCR)シグナル伝達を阻害するように作用する(Chemnitzら, 2004; Krummel & Allison, 1996; Parryら, 2005; Walunasら, 1996)。近年の証拠は、PD−1がまた、CD28陽性共刺激の阻害をもたらす(Huiら, 2017)こと、および関連して、CD28シグナル伝達がPD−1遮断に対する効果的な応答のために必要とされる(Kamphorstら, 2017)ことを示唆する。これは、CD28を減弱することが、PD−1およびCTLA4媒介性T細胞調節の共有される機構であり得ることを示唆する。
ヒト患者においてチェックポイント遮断によって誘導される自己免疫を再現する動物モデルが、本明細書で提供される。1つの実施形態において、マウスであって、そのゲノムが(i)Ctla4遺伝子のヘテロ接合性機能喪失および(ii)Pdcd1遺伝子のホモ接合性機能喪失を含むマウスが提供される。1つの局面において、上記マウスは、C57BL/6J遺伝子バックグラウンドを有する。いくつかの局面において、上記マウスは、雌性マウスである。いくつかの局面において、上記マウスは、雄性マウスである。
T細胞活性化は、陰性共刺激を含む広い範囲の機構を介して厳密に調節されるが、個々の分子メディエーターが機能的に相互作用する程度は、不明確なままである。CTLA4およびPD−1は、別個の分子機構および細胞機構を利用するT細胞活性化の重要な負の調節因子である。CTLA4およびPD−1は、ともにT細胞の陰性共刺激分子であり、その機能は、T細胞活性を減弱することである。これらの分子は、別個の分子機構を利用して、これらの機能を実行する。これらの分子機構は、大部分は別個の細胞シグナル伝達経路によって媒介される(Weiら, 2018)。CTLA4の主な機能は、B7リガンドの結合(B7−1/CD80、B7−2/CD86)に関して競合することであり、これは、CD28陽性共刺激および下流のPI3K/AKTシグナル伝達における低減をもたらす。PD−1の主な機能は、そのリガンドPD−L1/PD−L2への結合の際に、近くのT細胞レセプターシグナル伝達を阻害することである;しかし、CD28シグナル伝達の阻害はまた、PD−1の顕著な機能として報告されている(Huiら, 2017)。抗体媒介性遮断は、リガンド結合を防止することによって、これらの機能を阻害する。シグナル伝達能力のこの喪失および結論としての下流の生物学的事象は、Ctla4およびPdcd1の機能喪失型アレルの組み合わせを経る遺伝的手段によってモデル化され得る。

以下の実施例は、本発明の好ましい実施形態を示すために含められる。以下の実施例の中で開示される技術が、本発明の実施において十分に機能することが本発明者らによって発見された技術を表し、従って、その実施のために好ましい態様を構成すると考えられ得ることは、当業者によって認識されるべきである。しかし、当業者は、本開示に照らして、多くの変更が、開示される具体的実施形態の中で行われ得、本発明の趣旨および範囲から逸脱することなく、同様のまたは類似の結果をなお得ることができることを認識するべきである。
マウス。Ctla4tm1Allマウス(Chambersら, 1997)を、The Jackson Laboratory(021157)から購入したPdcd1ノックアウトマウス(Pdcd1tm1.1Shr)(Keirら, 2007)に交配した。Pdcd1ノックアウトマウスを、この交配の前にC57BL/6Jに一度戻し交配した。得られたF1 Ctla4+/− Pdcd1+/−マウスを、インタークロスして、その2つの遺伝子の野生型および変異アレルの全ての可能な組み合わせを生成した。この第1の育種スキームは、Ctla4+/−(これは、Pdcd1に関して野生型である)およびPdcd1−/−マウス(これは、Ctla4に関して野生型である)の交配から得られるF1マウスを具体的に利用した。この育種スキームは、F1マウスにおけるCtla4およびPdcd1の変異アレルがtransにあり、従って、組換え頻度が計算され得ることを担保する(図9A)。Ctla4とPdcd1との間の遺伝的距離を、この交配における組換え数および全事象を評価することによって計算した。その観察された組換え頻度17.03cMは、Mouse Phenome Database(MPD, RRID:SCR_003212)によって報告されるCtla4とPdcd1との間の16.89センチモルガン遺伝的距離と密接に並んだ(Bogueら, 2018)。これの予測された遺伝的距離は、この育種スキームにおいて観察された組換え頻度と一致した。
Ctla4とPdcd1との間の遺伝的相互作用が存在するか否かを試験するために、Ctla4およびPdcd1(PD−1をコードする)ノックアウトトランスジェニックマウスを交配した。マウスCtla4およびPdcd1は、Mouse Phenome Databaseからの予測に基づいて、16.89cMの遺伝的距離で遺伝的に連鎖する(Bogueら, 2018)。ヘテロ接合性インタークロス育種スキーム(材料および方法を参照のこと)を使用した。これは、単交雑に由来する変異アレルの全ての考えられる並べ替えの生成および17.03cMにおいてCtla4とPdcd1との間で観察された組換え頻度の予測を可能にした。驚くべきことに、Ctla4+/− Pdcd1−/−のうちのおよそ50%が、3ヶ月齢以内に自然に死亡した(図1A)。対照的に、関連するコントロール同腹仔(例えば、Ctla4+/+ Pdcd1−/−およびCtla4+/− Pdcd1+/−)は、明白な表現型を示さず、これらの群において死亡は観察されなかった。Ctla4+/+ Pdcd1−/−マウスにおいて明白な表現型がないことと、以前の観察とは一致する(Nishimuraら, 1999)。さらに、Ctla4−/− Pdcd1+/+マウスの全ての死亡は、CTLA4欠損マウス系統の最初の特徴付けと一致する(Chambersら, 1997; Tivolら, 1995; Waterhouseら, 1995)。興味深いことではあるが、Pdcd1の単一アレル喪失は、CTLA4の喪失によって誘導される致死的リンパ球増殖を加速した(図1A)。これは、Pdcd1遺伝子量が、CTLA4欠損マウスの表現型およびtリンパ球増殖性疾患を改変することを示唆する。これらの観察は、Ctla4+/+ Pdcd1+/−またはCtla4+/− Pdcd1+/−マウスにおいていかなるPD−1 ハプロ不全もないことと対照をなす。
次に、Ctla4+/− Pdcd1−/−マウスの死亡原因を理解し、特異的組織が影響を受けるか否かを調査しようとした。さらに、特定の細胞タイプが、疾患病因論を媒介するか否かを理解しようとした。これらの問題に対処するために、Ctla4+/+ Pdcd1−/−およびCtla4+/− Pdcd1−/−マウスの42の組織を、組織学的に分析した(材料および方法を参照のこと)。炎症を、心臓、膵臓、肺、肝臓、および消化管を含む多数の組織において観察した。特異的病理観察としては、リンパ球性心筋炎、動脈内膜炎、肺血管炎、およびリンパ球性膵炎が挙げられる。最も劇的な組織学的所見のうち、顕著な免疫浸潤が、Ctla4+/− Pdcd1−/−マウスの心臓および膵臓組織において観察された(図3A〜D;図10および図11A)。本質的により軽微であるが、顕著でもあるさらなる病理観察としては、唾液腺、涙腺、ハーダー腺、および腎臓のリンパ球浸潤が挙げられる。他の軽微な観察としては、肝臓変性/壊死、リンパ球性胃炎、および滑膜炎が挙げられるが、これらの所見が遺伝子型と関連する程度は、十分に決定されないままである。興味深いことに、より多くの顕著な組織学的所見が、リンパ器官(例えば、脾臓またはリンパ節)よりむしろ末梢の非リンパ組織において観察された。これは、末梢の免疫学的寛容がCtla4+/− Pdcd1−/−マウスにおいて特異的に突破されることを示唆する。

- 請求項1〜14のいずれか1項に記載のマウスにおいて少なくとも1種の候補因子をスクリーニングするための方法であって、前記方法は、1種またはこれより多くの候補因子を前記マウスに投与する工程を包含する方法。
- 前記少なくとも1種の候補因子を、(i)ホモ接合性野生型Ctla4遺伝子および(ii)Pdcd1遺伝子のホモ接合性機能喪失型アレルを含むマウスにおいてスクリーニングする工程をさらに包含する、請求項18に記載の方法。
