Abstract
Description
本発明の態様は、ジアシルグリセロールPEG付加物を有効成分として含み、前記ジアシルグリセロールPEG付加物が、以下の構造式を有し、長鎖脂肪酸におけるRの炭素数が11~23の範囲内であり、ポリエチレングリコール鎖におけるnが11~46の範囲内である、表皮内の抗酸化物質の発現増強剤である。
また、本発明の別の態様は、ジアシルグリセロールPEG付加物を有効成分として含み、前記ジアシルグリセロールPEG付加物が、以下の構造式を有し、長鎖脂肪酸におけるRの炭素数が11~23の範囲内であり、ポリエチレングリコール鎖におけるnが11~46の範囲内である、表皮内の抗酸化物質の発現増強方法である。
好ましくは、前記ジアシルグリセロールPEG付加物が、ベシクル状態で表皮内に浸透される。
好ましくは、前記ジアシルグリセロールPEG付加物のベシクルが、20~40nmの範囲内の直径を有する。
好ましくは、前記抗酸化物質が酸化ストレス応答遺伝子であり、前記酸化ストレス応答遺伝子がPPARGである。
好ましくは、前記抗酸化物質が抗酸化酵素であり、前記抗酸化酵素が、NAD(P)Hキノン還元酵素(NQO-1)、カタラーゼ(CAT)、及びヘムオキシゲナーゼ-1(HMOX1)からなる群のうち1つ又はそれ以上である。
好ましくは、前記抗酸化物質が抗酸化タンパクであり、前記抗酸化タンパクがグルタチオンである。
本発明は、ジアシルグリセロールポリエチレングリコール付加物(ジアシルグリセロールPEG付加物)において新たに見出された特性を利用して創作されたものである。ここで新たに見出された特性は、ヒトの表皮内の抗酸化物質の発現を増強する作用である。
・ジミリスチン酸グリセロールPEG-12(25.0℃:GDM12)
・ジステアリン酸グリセロールPEG-12(40.0℃:GDS12)
・ジステアリン酸グリセロールPEG-23(39.8℃:GDS23)
・ジパルミチン酸グリセロールPEG-23(31.2℃:GDP23)
・ジオレイン酸グリセロールPEG-12 (25.0℃:GDO12)
ヒト正常表皮メラノサイトを用いて試料を作製した後、RNAを抽出し、リアルタイムPCRにより、各目的物質(Nrf-2、PPARG、NQO-1、CAT、HMOX1)の遺伝子の発現及び増幅を確認した。PPARGは、タンパク質PPARγの遺伝子コードであり、酸化ストレス応答遺伝子の1つである。
ヒト正常表皮メラノサイト(NHEM:クラボウ製)を3.0×104cells/wellの細胞密度にて、培地(DermaLife(登録商標)M Comp kit培地:クラボウ製)を用いて96穴培養プレートに播種した。続いて、37℃、5%二酸化炭素下で24時間培養した。その後、以下の試料1~4をそれぞれ添加した培地に交換した後、37℃、5%二酸化炭素下で6時間培養した。

図1、図2、図3、図4、及び図5はそれぞれ、試料1~4についてのNrf-2、PPARG、NQO-1、CAT、及びHMOX1の発現量を示すグラフである。
ヒト正常表皮メラノサイトを用いて試料を作製した後、細胞内の抗酸化タンパクであるグルタチオンの量を確認した。
ヒト正常表皮メラノサイト(NHEM:クラボウ製)を2.0×104cells/wellの細胞密度にて、培地(DermaLife(登録商標)M Comp kit培地:クラボウ製)を用いて96穴培養プレートに播種した。続いて、37℃、5%二酸化炭素下で24時間培養した。その後、以下の試料5~9をそれぞれ添加した同種の培地に交換した後、37℃、5%二酸化炭素下で24時間培養した。

図6は、試料5~9におけるタンパク質当たりの総グルタチオン量を示すグラフである。図6によれば、表皮メラノサイトをGDS23で処理することによって、高い抗酸化力を有するタンパクであるグルタチオンの細胞内における産生が促進されることが確認された。グルタチオンは、グルタチオン産生酵素により生成され、そしてグルタチオン産生酵素は、Nrf-2及び/又はPPARγにより誘導される。したがって、GDS23によりNrf-2及び/又はPPARγが発現増強されることによって、結果的にグルタチオンの産生が促進されることが確認された。
紫外線による紅斑反応は、短波長紫外線(UVB)(290~320nm)によって引き起こされる紫外線損傷であり、サンバーンと呼ばれる。このサンバーンは、活性酸素及びプロスタグランジンE2によって引き起こされるとされている。そこで、表皮内の抗酸化物質の発現を増強するジアシルグリセロールPEG付加物による紫外線損傷の軽減効果を示すために、UVB照射と細胞生存率との関係を確認する試験を行った。
正常ヒト表皮角化細胞(NHEK:クラボウ製)を2.0×104cells/wellの細胞密度にて、KG2培地(クラボウ製)を用いて96穴培養プレートに播種した。続いて、37℃、5%二酸化炭素下で24時間培養した。その後、以下の試料10~15、16~19、及び20~24をそれぞれ添加した同種の培地に交換した後、37℃、5%二酸化炭素下で24時間培養した。試料10、16、及び20は、コントロール(無添加)である。試料11~15、17~19、及び21~24では、ジアシルグリセロールPEG付加物の種類及び/又は濃度(質量%、溶媒はHBSS(-))がそれぞれ異なる。

次に、ジアシルグリセロールPEG付加物による紫外線損傷の軽減効果を示すために、紫外線損傷を引き起こす炎症性サイトカインであるプロスタグランジンE2及びインターロイキン1-αの産生率を確認する試験を行った。
正常ヒト表皮細胞(クラボウ製)を4.0×104cells/wellの細胞密度にて、KG2培地(クラボウ製)を用いて48穴培養プレートに播種した。続いて、37℃、5%二酸化炭素下で24時間培養した。その後、以下の試料25~29、30~34、及び35~39を添加したKB2培地に交換した後、37℃、5%二酸化炭素下で24時間培養した。

図10及び図11は、試料25~39についてのプロスタグランジンE2及びインターロイキン1-αの産生率をそれぞれ示すグラフである。
近年大気汚染物質であるPM2.5による健康被害の懸念が報告されている。大気汚染物質の人体への悪影響は、活性酸素が発生して各細胞及び臓器に損傷を与えることである。この大気汚染物質損傷に対抗するために、本来ヒトが兼ね備えている体内の抗酸化機能を強化すれば、大気汚染物質への暴露による健康被害を軽減できると考えられる。そこで、表皮内の抗酸化物質の発現を増強するジアシルグリセロールPEG付加物による大気汚染物質損傷の軽減効果を示すために、大気汚染疑似物質であるDPE(Diesel Particle Extracts)への接触と、細胞のタンパク量との関係を確認する試験を行った。
正常ヒト表皮細胞(クラボウ製)を2.0×104cells/wellの細胞密度にて、KG2培地(クラボウ製)を用いて96穴培養プレートに播種した。続いて、37℃、5%二酸化炭素下で24時間培養した。その後、以下の試料40~46をそれぞれ添加したKB2培地に交換した後、37℃、5%二酸化炭素下で24時間培養した。

図12は、試料40~46についてのタンパク量の測定結果を示すグラフである。DPE無添加群では、試料40のコントロールと、試料41~46のジアシルグリセロールPEG付加物添加群との間に大きな差がない。すなわち、ジアシルグリセロールPEG付加物の添加は、細胞のタンパク量に影響を与えていないこと、すなわちジアシルグリセロールPEG付加物による細胞傷害性がないことを示している。
次に、大気汚染疑似物質であるDPE(Diesel Particle Extracts)への接触と、細胞傷害を引き起こす炎症性サイトカインであるプロスタグランジンE2及びインターロイキン1-αの産生率を確認する試験を行った。

図13及び図14は、試料47~53についてのプロスタグランジンE2及びインターロイキン1-αの産生量をそれぞれ示すグラフである。
ハイドロキノン(HQ)は、世界中の美容医療機関で処方されるクリームや軟膏剤などの皮膚外用剤において一般的に用いられている物質である。ハイドロキノンによる皮膚の美白効果は1950年代から報告されているが、副作用の問題から日本では敬遠する医師が多い。日本では2001年まで、医師の管理下でのみ処方が許されていたが、同年の規制緩和以降、化粧品へのハイドロキノンの配合が許されるようになった。しかし、未だハイドロキノンの美白に対する明確な作用機序及び安全性の情報は少なく、その副作用である皮膚の乾燥、紅斑、接触性皮膚炎などが課題である。副作用の要因となる細胞傷害性は、以下の化学反応式に示すように、ハイドロキノンが酸化されることで生じるベンゾキノン(BQ)によってその傷害性及び毒性が強くなると考えられる。ハイドロキノンとベンゾキノンの酸化還元反応は、平衡反応である。
先ず、ハイドロキノンとベンゾキノンの細胞傷害性の違いを確認するための試験を行った。
<試験方法>
正常ヒト表皮メラノサイト(NHEM:クラボウ製)を2.0×104cells/wellの細胞密度にて、培地(DermaLife(登録商標)M Comp kit培地:クラボウ製)を用いて96穴培養プレートに播種した。続いて、37℃、5%二酸化炭素下で24時間培養した。その後、ハイドロキノン(HQ)又はベンゾキノン(BQ)を所定の量(0μM、62.5μM、125μM、250μM、500μM)で含む同種の培地に交換した後、37℃、5%二酸化炭素下で24時間培養した。培養後、ニュートラルレッドアッセイによって、細胞生存率を測定した。無添加のコントロール(N.C.)の細胞生存率を100%として、HQ又はBQを添加した各試料の細胞生存率を算出した。
図15は、ハイドロキノンとベンゾキノンの細胞傷害性に関する試験結果を示すグラフである。ハイドロキノンは250μMでも細胞傷害性がないが、ベンゾキノンは250μMで細胞傷害性(細胞毒性)を示すことが判明した。したがって、ベンゾキノンはハイドロキノンより毒性及び傷害性が大きいといえる。
次に、ハイドロキノンをベンゾキノンに酸化させずに維持した場合の細胞傷害性を確認するための試験を行った。
<試験方法>
ハイドロキノンとベンゾキノンの酸化還元反応は平衡反応であり、ハイドロキノンは容易にベンゾキノンへ酸化されやすい。そこで、以下の化学反応式で示すように、ピロ亜硫酸ナトリウム(食品等でも一般的に用いられる酸化防止剤)をハイドロキノンと併用して培地へ溶解させることでハイドロキノンの酸化を抑止し、前項と同様の試験方法で細胞傷害性の評価を行なった。コントロール(N.C.)は無添加試料であり、それ以外は、全てピロ亜硫酸ナトリウムを0.5μM含み、ハイドロキノンの量が異なる試料である。
図16は、ハイドロキノンの細胞傷害性に関する試験結果を示すグラフである。ピロ亜硫酸ナトリウムのみの試料は、コントロールと差はなかった。ピロ亜硫酸ナトリウムとハイドロキノンを含む試料では、ハイドロキノンが2000μMまで細胞傷害性を示さなかった。それに対し、上述した図15では、ハイドロキノンが500μMで細胞傷害性を示しており、ハイドロキノンの酸化が起こっていたと考えられる。この結果から、細胞内でハイドロキノンを酸化させなければ、細胞傷害性を軽減でき、ベンゾキノンに起因する副作用も抑制できると考えられる。
ジアシルグリセロールPEG付加物による表皮内の抗酸化物質の発現増強作用によって、ハイドロキノンの酸化抑制すなわち細胞傷害性抑制の効果が得られるかを確認するための試験を行った。
ヒト正常表皮メラノサイト(NHEM:クラボウ製)を2.0×104cells/wellの細胞密度にて、培地(DermaLife(登録商標)M Comp kit培地:クラボウ製)を用いて96穴培養プレートに播種した。続いて、37℃、5%二酸化炭素下で24時間培養した。その後、以下の試料54~58を添加した培地に交換した後、37℃、5%二酸化炭素下で6時間培養した。

図17は、試料54~58についての細胞生存率の測定結果を示すグラフである。ハイドロキノン300μMとジアシルグリセロールPEG付加物を含む試料56~58は、試料54のコントロールとほぼ同じ細胞生存率が得られた。したがって、試料56~58では、ハイドロキノンが酸化せずに維持されていると考えられる。
ジアシルグリセロールPEG付加物による表皮内でのハイドロキノンの酸化抑制効果を確認するために、上記(8)よりもハイドロキノンを増量した試験を行った。
上記(8-1)と同じ試験方法により、細胞生存率を測定した。ただし、培地に添加する試料として、以下の試料59~62を用いた。試料59は、コントロール(ハイドロキノン及びGDS23の両方無添加)である。試料60は、別のコントロール(ハイドロキノン400μMのみ添加)である。試料61は、400μMのハイドロキノン及び500μMのGDS23を含む。試料62は、400μMのハイドロキノン、500μMのGDS23、スクワラン、及びコレステロールを含む。

図18は、試料59~62についての細胞生存率の測定結果を示すグラフである。試料61を、図17の試料56と比較すると、ハイドロキノンが増量されたために細胞生存率が低下している。しかしながら、試料61も、ハイドロキノンのみを含む試料60よりも高い細胞生存率を示している。
Claims (12)
- 前記ジアシルグリセロールPEG付加物が、ジミリスチン酸グリセロールPEG-12(GDM12)、ジステアリン酸グリセロールPEG-12(GDS12)、ジステアリン酸グリセロールPEG-23(GDS23)、ジパルミチン酸グリセロールPEG-23(GDP23)、及びジオレイン酸グリセロールPEG-12(GDO12)からなる群から選択される、請求項1に記載の表皮内の抗酸化物質の発現増強剤。
- 前記ジアシルグリセロールPEG付加物が、溶液状態で表皮内に浸透する、請求項1又は2に記載の表皮内の抗酸化物質の発現増強剤。
- 前記ジアシルグリセロールPEG付加物が、ベシクル状態で表皮内に浸透する、請求項1又は2に記載の表皮内の抗酸化物質の発現増強剤。
- 前記ジアシルグリセロールPEG付加物のベシクルが、20~40nmの範囲内の直径を有する、請求項4に記載の表皮内の抗酸化物質の発現増強剤。
- 前記抗酸化物質が酸化ストレス応答遺伝子であり、前記酸化ストレス応答遺伝子がNrf2である、請求項1~5のいずれかに記載の表皮内の抗酸化物質の発現増強剤。
- 前記抗酸化物質が酸化ストレス応答遺伝子であり、前記酸化ストレス応答遺伝子がPPARGである、請求項1~5のいずれかに記載の表皮内の抗酸化物質の発現増強剤。
- 前記抗酸化物質が抗酸化酵素であり、前記抗酸化酵素が、NAD(P)Hキノン還元酵素(NQO-1)、カタラーゼ(CAT)、及びヘムオキシゲナーゼ-1(HMOX1)からなる群のうち1つ又はそれ以上である、請求項1~5のいずれかに記載の表皮内の抗酸化物質の発現増強剤。
