Abstract
Description
本稿では、NSCLC(非小細胞肺癌)の治療戦略として、GLDC(グリシンデカルボキシラ-ゼ)mRNAをそれぞれ分解する12個の新規shAON(立体障害アンチセンスオリゴヌクレオチド)を同定した(添付の図1)。GLDCは、NSCLCからの腫瘍開始細胞において高度に発現され、細胞形質転換及び腫瘍形成を促進する(Zhang et al. Cell 2012; Jain et al. Science 2012);触媒能力が不活性なGLDCは、細胞形質転換を促進することができない。shAONは、すべての塩基がホスホロチオエート骨格によって結合した2’-O-メチルで修飾されている化学修飾合成一本鎖RNA分子である。各shAONは、新生したばかりのGLDC mRNAに特異的部位で相補的に結合するように設計され、エクソン認識及びそのスプライシングに重要なRNA結合スプライシングレギュレーターに対して立体障害効果を発揮する。機構的としては、成熟RNAにおいて多数の下流の未成熟終止コドンを生成する特異的なフレーム外エクソン(out-of-frame exon)の排除をshAONが誘導すると、得られるmRNAがナンセンス変異依存分解プロセスを介して分解の標的とされるというものである。
細胞株培養
ヒト肺腺癌上皮細胞株A549を、10%FBS、2mML-グルタミン、及び1%ペニシリン-ストレプトマイシンを補充したDMEM培地中で維持した。正常ヒト胎児肺線維芽細胞MRC-5細胞及びヒト成人肺線維芽細胞HLF細胞を、10%FBS及び1%ペニシリン-ストレプトマイシンを補充したDMEM培地中で維持した。
腫瘍球TS32細胞を、以前に記載されたように調製し(Zhang, WC; et al., 2012)、無処理シャーレ中において、0.4%BSA(Sigma)、20ng/mlEGF、4ng/ml塩基性線維芽細胞成長因子(bFGF)(Invitrogen)、1%ペニシリン-ストレプトマイシン(Gibco)を補充したITS(Sigma)含有DMEM/F12培地中で維持した。新鮮な培地を3日毎に補充した。細胞をAccutase(EMD-Millipore)で分割して、単一細胞懸濁液を得た。
shAONは、我々の特許された計算方法を用いて設計され、Sigma-Aldrich Pte Ltd(シンガポール)によって2’-O-メチルで修飾されたホスホロチオエート骨格(2OMePS)を伴い合成された。スクランブルされたシーケンスを有するshAONが全ての実験に含まれた。
全ての細胞/細胞株についての細胞トランスフェクションの間のトランスフェクション培地として、抗生物質(すなわち、ペニシリン-ストレプトマイシン)を含まない増殖培地を使用した。
shAONによるトランスフェクションの24時間後、細胞を回収し、Qiagen RNeasy Mini Kitを用いて全RNAを抽出し、DNase(AmbionTurbo DNA-Freeキット)で処理してコンタミDNAを除去し、ランダムヘキサマーと共にSuperScript(商標)III Reverse Transcriptase(Invitrogen)を製造業者の説明書に従って使用してcDNAに転写した。
チアゾリルブルーテトラゾリウムブロミド(MTT)アッセイ(Sigma)又はCellTiter-Blue細胞生存アッセイ(Promega)を用いて、96ウェルプレート(100μL培地/ウェル中3.5×103細胞)中で細胞生存率及び増殖の計測を行った。96ウェルプレートにおいて細胞をshAONとリポフェクタミン2000との混合物でトランスフェクトした。MTTアッセイ(接着細胞に使用)では、培地を、MTT(0.5mg/ml)を含むDMEM培地(100μL/ウェル)で置換し、37℃で4時間インキュベートし、次いで100μLのイソプロパノールに交換した。波長570nmのマイクロプレートリーダー(Molecular Devices)を用いて630nmでバックグラウンドを差し引いて吸光度を測定し、生細胞数に対する吸光度の較正曲線を用いて生細胞数に変換した。CellTiter-Blue細胞生存率アッセイ(懸濁細胞の場合)では、Cell-Titer Blue試薬を添加し(11μL/ウェル)、37℃で4時間インキュベートした。励起570nM及び発光600nmのマイクロプレートリーダー(Molecular Devices)を用いて蛍光を測定した。再現性を確実にするために、アッセイを少なくとも2回行い、トランスフェクションを各実験において3連のウェルで行った。
shAONトランスフェクションの24時間後、細胞を回収し、DMEM培地中の0.4%のノーブル寒天に再懸濁し、固化した底層(DMEM培地中の0.6%の貴寒天)を含む96ウェルプレート上にプレーティング(5×103細胞/ウェル)した。各ウェルを100μLの増殖培地で覆い、標準培養条件下で1週間インキュベートした。細胞をCellTiter Blueで染色し、蛍光マイクロプレートリーダーを用いてコロニー計数を行った。
Haltプロテア-ゼ阻害剤カクテルを含有するM-PER溶解緩衝液を用い、4℃で10分間穏やかに振盪することで、細胞からタンパク質を抽出した。細胞残屑を、最高速度(12,000g?)で20分間、4℃で遠心分離することによって除去し、上清のタンパク質濃度を、BioRad protein assayを使用して、製造業者の指示に従って測定した。製造業者の説明書に従って調製した8%Bis-Tris SDS-PAGEゲル(Bio-rad)に、各試料からの16μgのタンパク質をロードし、次いで、Bio-radニトロセルロースメンブレンに移した。PBST中の5%乳で1時間ブロッキングした後、メンブレンを3%乳/PBST中のウサギ抗ヒトGLDCポリクローナル抗体(1:2000)と共に4℃で一晩インキュベートし、続いて、3%乳/PBST中の西洋ワサビペルオキシダーゼ結合抗ウサギIgG(1:4000)と共に室温で3時間インキュベ-トした。ウェスタンブロッティングのためのロード対照としてβ-アクチンを使用し、第1及び第2の抗体としてマウス抗ヒトβ-アクチン抗体(1:1000)及び西洋ワサビペルオキシダ-ゼ結合抗マウスIgG(1:4000)をそれぞれ使用した。Supersignal West Pico化学発光基質(Thermo Scientific)を用いたECLシステムによって可視化した。
4~6週齢のNOD.Cg-Prkdcscid Il2rgtm1Wjl/SzJマウスに、無血清培地及びMatrigel(BD)(1:1)中の腫瘍球の単細胞懸濁液を皮下移植した(500,000細胞/マウス)。shAON50mg/kgを週3回腹腔内投与した。6週間の注射後にマウスを屠殺し、腫瘍を抽出し、秤量した。Micro Smash MS-100(4500rpmで3×45秒)中の0.5mmガラスビーズ(Sigma-Aldrich)を用いた細胞を破壊した後、Qiagen RNeasy Mini Kit及びM-PER溶解緩衝液をそれぞれ使用して、全RNA及びタンパク質を腫瘍試料から抽出した。
GLDC標的エクソンの効率的かつ特異的なスキップにつながるshAONの選択
GLDC転写物中の25個のエクソンのうち、5個のフレーム外エクソン(エクソン7、8、13、15及び16)を、shAONを設計するための我々の標的として選択した。各々が個々にスキップされるとき、得られる転写物は標的エクソンの下流にシフトしたリーディングフレームを有し、スキップされた転写物中に複数のPTCを生成する。結果として、標的遺伝子は、転写物が活発に分解されるナンセンス変異依存分解(NMD)経路の組み合わせを介してダウンレギュレートされるか、又は、切り縮められた非機能的遺伝子産物の翻訳を導くかのいずれかである。著者らが以前に開発し、検証した計算法を用いて、20個のshAON(4個のshAON/エクソン)を設計した。スクランブルされた配列を有する1つのshAONを、ネガティブコントロールとして全ての実験に含めた。これらのshAONは安定性及び配列特異性を増加させるために、ホスホロチオエート骨格及び2’-O-メチルリボース修飾(2OMePS)を有するものとして合成した。
A549細胞における特異的エクソンスキップの誘導に対する3つの選択されたshAONの時間及び用量応答効果を、続けてデンシトメトリー分析を行うPCRによって評価した。図2aに示されるように、3つのshAONの全ては、3.5~7nMの範囲のIC50という低用量で特異的エクソンスキップを誘導し得る。10nMのshAONを用いた時間経過実験によると(図2b)、エクソンスキップの誘導に対するそれらの効果は時間と共に減少したが、トランスフェクションの72時間後にも依然としてGLDC転写物の40%超においてスキップを誘導することができた。
肺癌細胞は高レベルのGLDCに依存することが見出されている(Zhang WC, et al, Cell. 2012 Jan 20;148(1-2):259-72)。shAONは細胞にトランスフェクトされた場合、GLDCの効果的なノックダウンを誘導し得るので、本発明者らは癌細胞の増殖及び増殖に対するそれらの効果を試験した。A549細胞生存率を、種々の濃度の選択されたshAONでのトランスフェクションの0、24、48、72及び120時間後にMTTアッセイを用いて測定し、未処理細胞、リポフェクタミン2000又はスクランブルされたshAONでトランスフェクトされた細胞を含むいくつかのネガティブコントロールと比較した。shAON誘導GLDCノックダウンは、トランスフェクション後120時間まで細胞増殖を阻害するようであった(図3a及び図3b)。細胞増殖の阻害は5nMのshAONによる35%阻害から、30nMのshAONによるほぼ100%阻害まで増加した。図3cに示すように、10nMのshAONでトランスフェクトした場合、2つの非癌細胞、HLF及びMRC-5の増殖は阻害されないようであったが、A549細胞では細胞増殖の約70%が阻害された。
癌幹細胞(CSC)又は腫瘍開始細胞(TIC)は腫瘍増殖を駆動及び持続する能力を有し、インビトロ培養で増殖させた場合、球体形成によって単離及び濃縮(enrich)され得る。そのため、本発明者らは、腫瘍開始細胞について高度に濃縮された腫瘍球細胞の増殖に影響を及ぼすことにおけるGLDC shAONの効果を試験した。図4aに示すように、GLDC shAONはGLDC転写物中の特異的標的エクソンのスキップを効果的に誘導することができ、スキップ効率は51~93%の範囲であり、IC50は65nM~220nMの範囲である(図4a)。ウェスタンブロッティングを用いて測定したタンパク質レベルでは、GLDCタンパク質発現も効果的に阻害された(図4b)。腫瘍球細胞における効果的なエクソンスキップ及びGLDCノックダウンを誘導するためには、A549細胞におけるよりも多量のshAONが必要であった。
GLDCの過剰発現は、非小細胞肺癌において見出されたばかりでなく、他のタイプの癌においても報告された(Zhang WC, 2012)。我々はqPCRを用いて2つのMLL白血病細胞株におけるGLDC発現を評価し、GLDCがそれらの両方において過剰発現され、一方、非癌細胞のGLDCの発現は約600倍少ないことを確認した(図5a)。そのため、本発明者らは、GLDC shAONがこれらの2つのMLL細胞株において有効であり得るかどうかを試験した。図5bに示されるように、GLDC shAONは両方のMLL細胞株においてGLDC転写物の50%超において特定のエクソンのスキップを誘導し、本発明者らの設計したshAONが他のタイプの癌にも有効であり得ることを示した。
in vivo実験におけるshAON 7Dの効果を試験するために、免疫不全マウスに腫瘍球細胞を移植し、続いてshAON 7Dの腹腔内注射を行った。図6に示されるように、7D及びNC処置マウス由来の腫瘍は、それぞれ0.325g及び0.820gの中央値、すなわち腫瘍増殖の60%阻害と計量された(P値=0.000676(対応なしt試料検定)及び0.00391(対応なしWilcoxon順位和検定))。腫瘍試料20点(対照群からの12点及びshAON 7D群からの8点)のRNA分析では、7D処置がすべての処置された腫瘍試料においてGLDCエクソン7又はエクソン7及び8(軽度)のスキップを引き起こしたが、NC処置された腫瘍試料においてGLDCエクソンスキップはほとんど観察されなかったことが示された。

Claims (33)
- 配列番号1~配列番号12、配列番号26、配列番号28、及び、配列番号52から選択されるいずれか1つの配列を含み、
GLDCのmRNA前駆体又は成熟mRNAのエクソンのスキップを誘導することによって、GLDCの発現を阻害して、細胞周期停止又はアポトーシスを誘導する、
グリシンデカルボキシラーゼ(GLDC)の活性を阻害するための単離されたアンチセンスオリゴヌクレオチドであって、前記アンチセンスオリゴヌクレオチドはGLDC RNAのエクソン、イントロン又はエクソン-イントロン境界の標的領域に特異的にハイブリダイズし、前記標的領域が、エクソン7、エクソン8、エクソン13、エクソン15、及びエクソン16からなる群より選択されるいずれか1つである、アンチセンスオリゴヌクレオチド。 - 前記オリゴヌクレオチドが、活性、又はmRNA前駆体、成熟mRNA及びタンパク質の発現レベルのうち1つ以上を調節する、請求項1に記載のアンチセンスオリゴヌクレオチド。
- 前記オリゴヌクレオチドが、GLDCの発現を阻害するか、又はその発現産物を改変する、請求項2に記載のアンチセンスオリゴヌクレオチド。
- GLDCのmRNA前駆体又は成熟mRNAの標的領域に特異的にハイブリダイズし、1種以上のRNA結合スプライシング調節因子に対して立体障害効果を発揮する、請求項1~請求項3のいずれか1項に記載のアンチセンスオリゴヌクレオチド。
- 立体障害によりmRNAの翻訳を妨げる、請求項4に記載のアンチセンスオリゴヌクレオチド。
- 修飾されたポリヌクレオチド骨格を含む、請求項1~請求項5のいずれか1項に記載のアンチセンスオリゴヌクレオチド。
- 前記修飾されたポリヌクレオチド骨格が、少なくとも1つのポリヌクレオチドの糖を置きかえた修飾部分を含む、請求項6に記載のアンチセンスオリゴヌクレオチド。
- 前記修飾部分がホスホロジアミデートモルホリノオリゴマー(PMO)、ペプチド結合ホスホロジアミデートモルホリノオリゴマー(PPMO)、及び非ペプチドデンドリマーオクタグアニジン部分標識モルホリノオリゴマーからなる群より選択される、請求項7に記載のアンチセンスオリゴヌクレオチド。
- 前記修飾されたポリヌクレオチド骨格が少なくとも1つの修飾ヌクレオチド間結合を含む、請求項6~請求項8のいずれか1項に記載のアンチセンスオリゴヌクレオチド。
- 前記修飾ヌクレオチド間結合が修飾されたホスフェートを含む、請求項9に記載のアンチセンスオリゴヌクレオチド。
- 前記修飾されたホスフェートが、非架橋酸素原子を置換する硫黄原子、ホスホネート、ホスホロチオエート、ホスホジエステル、ホスホロモルホリデート、ホスホロピペラジデート、及びホスホロアミデートからなる群より選択される、請求項10に記載のアンチセンスオリゴヌクレオチド。
- リボ核酸、デオキシリボ核酸、DNAホスホロチオエート、RNAホスホロチオエート、2’-O-メチル-オリゴリボヌクレオチド、2’-O-メチル-オリゴデオキシリボヌクレオチド、2’-O-ヒドロカルビルリボ核酸、2’-O-ヒドロカルビルDNA、2’-O-ヒドロカルビルRNAホスホロチオエート、2’-O-ヒドロカルビルDNAホスホロチオエート、2’-F-ホスホロチオエート、2’-F-ホスホジエステル、2’-メトキシエチルホスホロチオエート、2-メトキシエチルホスホジエステル、デオキシメチレン(メチルイミノ)(デオキシMMI)、2’-O-ヒドロカルビMMI、デオキシメチルホスホネート、2’-O-ヒドロカルビルメチルホスホネート、モルホリノ、4’-チオDNA、4’-チオRNA、ペプチド核酸、3’-アミデート、デオキシ3’-アミデート、2’-O-ヒドロカルビル3’-アミデート、ロックド核酸(Locked Nucleic Acids)、シクロヘキサン核酸、トリサイクルDNA、2’-フルオロ-アラビノ核酸、N3’-P5’ホスホロアミデート、カルバメート結合、ホスホトリエステル結合、及びナイロン骨格修飾からなる群より選択される一つ又は複数の修飾を有する骨格を含む、請求項1~請求項11に記載のアンチセンスオリゴヌクレオチド。
- アンチセンスオリゴヌクレオチドの活性、細胞内分布、又は細胞への取り込みを増進する1つ以上のコンジュゲートに化学的に連結されている、請求項1~請求項12のいずれか1項に記載のアンチセンスオリゴヌクレオチド。
- 前記オリゴヌクレオチドが、活性、又はmRNA前駆体、成熟mRNA及びタンパク質の発現レベルのうち1つ以上を調節する、請求項1~請求項13のいずれか1項に記載のアンチセンスオリゴヌクレオチド。
- GLDC発現癌患者の治療に使用するための、請求項1~請求項14のいずれか1項に記載のアンチセンスオリゴヌクレオチド。
- 前記患者はさらなる抗癌剤又は治療を施される、請求項15に記載のアンチセンスオリゴヌクレオチド。
- 前記癌が、非小細胞肺癌(NSCLC)、乳癌、卵巣癌、結腸癌、黒色腫、精巣癌、甲状腺癌、神経膠腫、カルチノイド腫瘍及び葉状腫瘍からなる群より選択されるいずれか1つである、請求項15又は請求項16に記載のアンチセンスオリゴヌクレオチド。
- 請求項1~請求項17のいずれか1項に記載のアンチセンスオリゴヌクレオチド及び医薬的に許容されるキャリアを含む医薬組成物。
- そのままで、又は送達剤と複合して、患者への非経口投与に適している、請求項18に記載の組成物。
- 前記キャリアが、ナノ粒子、リポソーム、ウイルスベクター、カチオン性脂質、ポリマー、UsnRNA、及び細胞膜透過ペプチドからなる群より選択される、請求項19に記載の組成物。
- 前記アンチセンスオリゴヌクレオチドが、経口、直腸、経粘膜、腸、筋肉内、皮下、髄内、髄腔内、直接脳室内、静脈内、硝子体内、腹腔内、鼻腔内、又は眼内に投与される、請求項18~請求項20のいずれか1項に記載の組成物。
- 組成物が、癌を有する患者における予後の分類又は決定を補助するための組成物、又は癌を有する患者のための治療戦略を選択するための組成物である、請求項18~請求項21のいずれか1項に記載の組成物。
- 癌を治療するための医薬の製造における、GLDCの活性を阻害する、請求項1~17のいずれか1項に記載のアンチセンスオリゴヌクレオチドの使用。
- 前記オリゴヌクレオチドが、活性、又はmRNA前駆体、成熟mRNA及びタンパク質の発現レベルのうち1つ以上を調節する、請求項23に記載の使用。
- 前記オリゴヌクレオチドが、GLDCの発現を阻害するか、又はGLDCの発現産物を改変する、請求項24に記載の使用。
- 前記癌が非小細胞肺癌(NSCLC)である、請求項23~請求項25のいずれか1項に記載の使用。
- 癌を有する患者における予後の分類又は決定を補助するための方法、又は癌を有する患者のための治療戦略を選択するための方法であって、試料中のGLDCの核酸、タンパク質又は活性のレベルをex vivoで評価することを含み、
前記試料中のGLDCの核酸、タンパク質又は活性のレベルが上昇したレベルである場合、請求項1~請求項17のいずれか1項に記載のアンチセンスオリゴヌクレオチドを含むGLDC活性の阻害剤が、治療レジメンにおいて選択されることを含む方法。 - 前記試料中のGLDCの核酸、タンパク質又は活性のレベルに関する情報を利用して治療レジメンを選択するステップをさらに含む、請求項27に記載の方法。
- 前記試料が前記患者から得られ、前記試料が、癌が疑われるか、癌が発見された組織の試料であるか、又は前記組織に由来する細胞を含む、請求項27又は請求項28のいずれか1項に記載の方法。
- GLDCmRNA前駆体のエクソンスキップをex vivoで誘導する方法であって、請求項1~請求項17のいずれか1項に記載のアンチセンスオリゴヌクレオチド又は請求項18~請求項22のいずれか1項に記載の組成物をex vivoで細胞に送達することにより、GLDCmRNA前駆体のエクソンスキップをex vivoで誘導することを含む方法。
- 前記細胞がヒト細胞である、請求項30に記載の方法。
- 前記ヒト細胞が癌細胞であり、前記癌が、血液悪性腫瘍、肺癌、乳癌、前立腺癌、胃癌、精巣癌、喉頭癌、肝臓癌、子宮癌、結腸直腸癌、黒色腫、神経膠芽腫、肉腫、及び網膜芽細胞腫からなる群より選択されるいずれか1つである、請求項31に記載の方法。
