Abstract
Description
a)皮膚のネガティブレリーフを備えたスキンプリントを調製するステップ;
b)14〜60mJ/m2の表面自由エネルギー(γ)を有する材料であるポリマー材料を用いて、スキンプリントを成形し直す(molding back)ステップ;
c)硬化ステップ;
d)14%〜60%のトリグリセリド、2%〜40%の遊離脂肪酸、4%〜30%のワックスエステル、3%〜20%のスクアレン、及び1%〜10%のコレステロールを含み、溶媒不含である脂質組成物を調製するステップ;
e)任意に、脂質組成物を完全に液状化するまで加熱するステップ、
f)液状の脂質組成物をポリマー材料上に塗布するステップ;
g)任意に、均質化のために加熱するステップ;及び
h)任意に、過剰の脂質組成物を拭き取り、均質化のために再度加熱するステップ。
本発明において使用される用語「非生物学的皮膚モデル」は、皮膚表面トポグラフィ、化学、物理化学の側面を再現する、健康な又はダメージを受けた皮膚を表す、非生物学的実験システムを表す。
本発明の第1の主題は、脂質組成物で被覆された、皮膚表面トポグラフィを再現するポリマー材料を含む、非生物学的皮膚モデル(NBSM)であり、ここで、該ポリマー材料は、14〜60mJ/m2の表面自由エネルギー(γ)を有する物質であり;該脂質組成物は、14%〜60%のトリグリセリド、2%〜40%の遊離脂肪酸、4%〜30%のワックスエステル、3%〜20%のスクアレン、及び1%〜10%のコレステロールを含み、ここで、ポリマー材料上の脂質組成物の表面濃度は500μg/cm2〜2500μg/cm2である。

本発明による非生物学的皮膚モデル(NBSM)が開発された。これは、2つの異なる部分で構成される(図2参照)。第1部分は皮膚表面トポグラフィを再現するポリマー材料である。このポリマー材料は、皮膚表面の脂質組成物を模倣する人工皮脂で被覆されている。このNBSMは以下のパラグラフに記載されるようにして調製された。
スキンプリントを、インビボで、生体適合性のシリコンBody Double(登録商標)(Creation Silicone、ジュイ−アン−ジョザ、フランス)を用いて調製し、皮膚のネガティブレリーフを備えたシリコン表面を得た。このスキンプリントを、シリコーンDragonSkin(登録商標)(DragonSkin 20、Creation-Silicone、ジュイ−アン−ジョザ、フランス)を用いて成形し直した(図3参照):キットの2つの成分を等しい量で共に混合した後、Dragonskin(登録商標)を高真空下で10分間かけて脱ガスした。次いで、粘性混合部をBody Double(登録商標)スキンプリントに注ぎ、少なくとも4時間硬化させた。
人工皮脂を表2に記載の組成に従い調製した[11]。全ての計量した脂質をクロロホルム/メタノール(CHCl3/MeOH)2:1中に溶解させ、10g/Lの皮脂濃度にした。該溶液を使用するまで冷蔵庫(T=2−4°C)に保管した。溶媒の蒸発が必要あ場合、ロータリーエバポレーターを使用した(Rotavapor(登録商標)R-300、BUECHI Labortechnik AG、フラヴィル、スイス)。
C1.プロトコル1(比較)
第1のプロトコル(「プロトコル1」と称する)は、20g/Lの濃度でのCHCl3/MeOH 3:7の皮脂溶液を用いた。皮脂溶液をポリマー材料上に、スプレーポンプを介して噴霧(pulverize)した[5,12]。1cm×7cmの表面に、室温で、5cmの距離で、10回の噴霧を行った。調製した表面を、閉じたプラスチックチューブに入れ、溶媒の蒸発速度を遅くし、脂質コーティングの良好な均質性を得た。
第2のプロトコル(「プロトコル2」又は本発明のプロトコルと称する)は、いずれの溶媒も使用しない純粋な皮脂を使用した。人工皮脂溶液から溶媒を蒸発させた。得られた脂質混合物を、完全に液化するまで、70℃のオーブンに入れた。堆積中の温度差を避けるために、ポリマー材料も70℃のオーブンに15分間入れた。細い鉛筆を使用して、液状の脂質フィルムをシリコーン支持体上に塗布した。次いで、被覆された支持体を、70℃で均質化させるために、オーブン中に5分間戻した。吸収紙を使用して過剰の脂質を拭き取り、被覆されたシリコーンを、再び、オーブン中に70℃で5分間入れた。上記のようにして調製された人工皮膚モデルを、室温まで数分で冷却させた。
実験例1においてコーティングプロトコル2(=NBSMプロトコル2)を用いて調製された、本発明の非生物学的皮膚モデルは、ここで特徴づけられ、以下と比較される:
・ヒトの生きている外植片;
・インビボ測定;
・市販の非生物学的皮膚モデル:Vitroskin(登録商標)(IMS、Inc.、Milford、CT)(=VS);及び
・実験例1でコーティングプロトコル1(=NBSMプロトコル1)を用いて調製した比較NBSM
A1.エクスビボ皮膚(ヒトの生きている皮膚外植片):
ヒトの生きている皮膚の外植片は、BIO-EC(ロンジュモー、フランス)から提供され、これは、29歳の白人女性の腹部領域の形成外科から、彼女の同意を得て得られた。皮膚外植片を調製するために、まず、メスを用いて皮下脂肪を除去し、次いで、皮膚(真皮及び表皮)を直径約2cmの円に切断した。調製した外植片を、BIO−ECの外植片培地(BEM)を用いて生存条件に置き、37℃で、5%CO2のインキュベータ内に保持した。BEMの半分を、2日ごとに、新しいものに取り換えた。8つの外植片を表面自由エネルギーの測定に使用した。これらを接触角の測定まで、インキュベータ中に保持した。表面上に存在する残留BEMを除去するために、測定前にこれらを静かに拭いた。
21歳の白人女性の前腕にて、同意を得て、インビボ測定を行った。分析された皮膚は、洗浄せずに2時間保持し、測定前に処理を行わなかった。
市販のVitroskin(登録商標)(IMS, Inc.、ミルフォード、CT)を特徴付け、研究に使用した。これは、物理化学研究のための非生物学的皮膚モデルに関する参考である。Vitroskin(登録商標)(VS)は、プロテイン及び脂質で作られた合成皮膚モデルであり、イオン強度、pH、トポグラフィ及び臨界表面張力などの皮膚表面特性を模倣する。このNB皮膚モデルは、インビトロSPF(Sun Protection Factor、紫外線防御指数)測定のために、又は、エモリエント拡散の研究のために、皮膚置換物として使用される。これは使用前に、IMSにより開発された標準化プロトコルに従い水和させる必要があった。VSの断片を、下部に注入された350gの水/グリセリン85:15混合物を含有する標準的な密閉水和チャンバー内に入れ、16〜24時間、室温で置いた[13]。
B1.光学顕微鏡法:
表面観察は、カメラを備え、Leica Application Suiteソフトウェアによって制御された光学顕微鏡(DMLP/DC 300、Leica Microsystems、ウェッツラー、ドイツ)を用いて行った。全ての写真は、50倍の倍率で、透過モードを用いて得た。
Spectrumソフトウェアに接続されたFT−IR分光計スペクトル(PerkinElmer, Inc.、Waltham、Massachusetts、USA)を使用した。ATRモード(ZnSe結晶)を使用して、各測定ごとに4つのスペクトルを測定した。振動の範囲は、4000〜650cm−1であった。
VH−Z100Rレンズを用いるキーエンスマイクロスコープVHX-1000(Keyence Corporation TSE、大阪、日本)を300倍の倍率で使用した。3D画像を透過モードで記録し、1600×1200ピクセルサイズを得るように編集した。
C1.前進接触角測定:
接触角測定は、PGPlusソフトウェアに接続された携帯型ゴニオメーターPGX+(ScanGaule、Gravigny、フランス)を用いて行った。このゴニオメーターは、画像を得るための高解像度カメラ、液滴を視覚化するためのミラーに関連した特定の照明システムを備える。液滴を堆積させるために使用するシリンジは、0.77mmの内部直径を有する。測定は室温(20±1.5℃)で行った。前進接触角θAは、液滴堆積が増加する際に、液滴ベースライン変位の前に測定される、接触角の最大値である。小さい液滴が表面に堆積し、5回の連続した液滴の追加によってその体積が増加した。最終的な液滴は約7μlの体積に達した。各滴下の追加後に5枚の画像を撮影した。実験の合計時間は、約20秒であるため、蒸発又は浸透は無視される。ソフトウェアプログラムを使用して、滴の両側から接触角を算出した。その場合、θAは、これら5枚の画像のうち、滴の両側の平均から得た最大値であった。各液体の前進接触角測定を、各研究モデルについて少なくとも3回行った。
表面自由エネルギー計算を行うために、次の3つの参照液体を使用した:超純水(25℃で18MΩ.cm−1の抵抗率)、ジヨードメタン(分析グレート、99%純分)及びホルムアミド(分析グレート、99%純分)(Sigma Aldrich、Saint-Louis、Missouri)。これらの表面自由エネルギー成分値を表5に報告する。
固体表面の表面自由エネルギーを測定するために、最新のアプローチの1つがVan Oss及び協力者らによって開発された[1,16]。彼らは、固体及び液体表面自由エネルギーは2つの成分に分けられることを提案する:ファンデルワールス相互作用(Keesom、Debye、London)に対応する、Lifschitz-ファンデルワールス成分(γLW)、及び、ルイス概念:
Van Ossによれば、界面表面自由エネルギーは次のように表される:
結果は、プロトコル1は、プロトコル2よりも、エクスビボヒト皮膚に近い特性に達するにより効率的でないことを明らかに示す。確かに、プロトコル1を用いて調製した表面は、エクスビボ皮膚(0.5±0.5mJ/m2)と比較して、より高い酸成分(3.1±2.0mJ/m2)を示すことが観察できる。その場合、プロトコル1について決定したLW成分(19.8±3.1)はエクスビボで得たものよりも低い。プロトコル2は、皮膚物理化学を模倣するのに非常に適している。さらに、プロトコル2に溶媒が含まれていないことは、環境への配慮に間違いなく有利であり、また、皮脂で被覆されたポリマー材料の溶解のリスクを防止する。さらに、光学顕微鏡は、皮脂の分布がより均質であり、プロトコル2で、脂質の結晶化が制限されたことを示す。これらの全ての理由から、第2のプロトコルが選択され、残りの研究に適用された。
物理化学的研究に使用した化粧品原料及びエマルション:
この研究に使用した化粧品グレード原料をその物理化学特性と共に表6に示す。シリコーンはEvonik Goldschmidt(Essen、ドイツ)から供給された;エステルはStearinerie Dubois(Boulogne-Billancourt、フランス)から提供された;イソヘキサデカンはIMCD Group(Cologne、ドイツ)から提供された;アラガン油はOlvea Group(Saint-Leonard、フランス)から供給された。
接触角測定を実験例2パートC1及びC2に記載されるように行った。
50μLのそれぞれの化粧品原料及びエマルション(表6及び7)を、40回転で10cm2の表面積に手動で広げた。水接触角測定は、塗布の1分後及び3分後に3回行った。
AO、IHD、CPS及びPDCの接触角(表6)を、各研究モデルについて、3回測定した。滴下堆積プロトコルは、前述のものと同じであった。
エモリエント拡散の研究:
接触角測定は、皮膚上のエモリエントの拡散特性の研究に使用した機器方法の一部である。エモリエントの拡散は、化粧品の感覚的品質だけでなく、その有効性、とりわけサンスクリーンについて、重要な目的である。確かに、日焼け止め防止係数は、一方はエマルション中に存在するソーラーフィルターによるものであり、他方では、皮膚表面に製品が拡散された後の残留フィルムの均質性によるものである。したがって、フィルム形成及び均質性に影響する他のパラメータの中で、拡散が最も重要であるように思われる[21,22]。化粧品原料の拡散特性を評価するために、4つの原料:CPS、PDC、AO及びIHDを用いて、ここで研究される異なる表面上で、接触角測定を行った。NBSMプロトコル2、VS及びインビボで得られた結果を、図10に示す。一般的な観点から、4つの異なる原料を使用して測定された接触角は、NBSMプロトコル2及びインビボ皮膚との間で、非常に類似しているように見える。反対に、VSにおける異なるエモリエントについて測定された前進接触角は、インビボ及びNBSMプロトコル2表面の両方と比較して、より低い。これらの結果は、本研究において特別に開発されたNBSM表面が、インビボ皮膚と同様に、製品と相互作用することを意味する。唯一の違いは、NBSMプロトコル2よりも、インビボ皮膚とより親和性を示すIHDに関する。これは、NBSMと比較してより高い皮膚LW成分であるため、無極性IHDとのより高い相互作用を引き起こすことで説明できる。しかし、このNBSMは、VSと比較した場合に、エモリエント拡散を評価するに効果的なサポートを構成することは間違いない。確かに、VSについて測定したより低い接触角は、各原料が、インビボ皮膚よりもVSと、より親和性であることを示す。使用した参照液体の毒性のために、インビボ皮膚の表面自由エネルギーを測定することができないため、得られたデータを、エクスビボ値と比較することにした。さらに、文献データは、個人間の差に内在する大きな変動を示す。その結果、化粧品原料の拡散は、エクスビボ皮膚及びNBSMプロトコル2と比較したとき、VSについての表面自由エネルギー(γVS=35.8mJ/m2)がより高いために、高められる。水の存在は、慣行的に表面自由エネルギー値を高くする傾向があるため、VSの水和状態がこの結果の原因であり得る。これらの結果によれば、NBSMプロトコル2は、局所塗布後の皮膚表面に存在する残留フィルムを研究するための適切なツールとして興味深く思われる。化粧品残留フィルムの物理化学特性を以下に示す。
皮膚上の原料またはエマルションの残留フィルムの特徴付けは、その均質性、組成及び安定性が皮膚表面特性に大きく影響し、その結果、化粧品活性成分及び製品の有効性に大きく影響するため、非常に興味深いトピックである。残留フィルムの物理化学を研究するために、VS、NBSMプロトコル2及びインビボで、上記のプロトコルに従い一連の化粧品を塗布後、2つの異なる時間(1分及び3分)で水接触角を測定した。結果を図11に示す。化粧品原料で表面処理した1分後に測定した水接触角は、次のように分類される:インビボ皮膚及びNBSMプロトコル2の両方について、θ水/PDC<θ水/AO<θ水/IHD<θ水/CPSであり、一方、VSについて、θ水/IHD<θ水/PDC≒θ水/AO θ水 /CPSである。製品塗布前、θ水 インビボ=93,2±7.8°、θ水 NBSM=114,2±5.8°及びθ水 VS=104.2±11.8°。

Claims (15)
- 脂質組成物で被覆された皮膚表面トポグラフィを再現するポリマー材料を含む非生物学的皮膚モデルであって、
該ポリマー材料は14〜60mJ/m2の表面自由エネルギー(γ)を有する材料であり、
該脂質組成物は、脂質組成物の総重量に対して、14重量%〜60重量%のトリグリセリド、2重量%〜40重量%の遊離脂肪酸、4重量%〜30重量%のワックスエステル、3重量%〜20重量%のスクアレン、及び1重量%〜10重量%のコレステロールを含み、
かつ、ポリマー材料上の脂質組成物の表面濃度は500μg/cm2〜2500μg/cm2である、
非生物学的皮膚モデル - 24mJ/m2〜45mJ/m2、有利には26mJ/m2〜33mJ/m2の表面自由エネルギー(γ)を有する、請求項1に記載の非生物学的皮膚モデル。
- 0.0〜−2.5、有利には−0.2〜−1.7、より有利には−0.5〜−0.9の歪度係数を有する、請求項1又は2に記載の非生物学的皮膚モデル。
- 脂質は結晶化されていない、請求項1〜3のいずれかに記載の非生物学的皮膚モデル。
- 以下のステップ:
a)皮膚のネガティブレリーフを備えたスキンプリントを調製するステップ;
b)14〜60mJ/m2の表面自由エネルギー(γ)を有する材料であるポリマー材料を用いて、スキンプリントを成形し直すステップ;
c)硬化ステップ;
d)14%〜60%のトリグリセリド、2%〜40%の遊離脂肪酸、4%〜30%のワックスエステル、3%〜20%のスクアレン、及び1%〜10%のコレステロールを含み、溶媒不含である脂質組成物を調製するステップ;
e)任意に、脂質組成物を完全に液状化するまで加熱するステップ、
f)液状の脂質組成物をポリマー材料上に塗布するステップ;
g)任意に、均質化のために加熱するステップ;及び
h)任意に、過剰の脂質組成物を拭き取り、均質化のために再度加熱するステップ
を含む、非生物学的皮膚モデルの調製方法。 - 脂質組成物を調製するステップd)は、脂質を溶媒中に溶解させるステップd1)及び全ての溶媒を蒸発させるステップd2)を含む、請求項5に記載の方法。
- ステップe)、g)及びh)の加熱温度は、互いに独立に、少なくとも25℃、有利には65℃〜200℃、より有利には65℃〜100℃である、請求項5又は6に記載の方法。
- ステップf)の前に、ポリマー材料をステップe)における脂質組成物と同じ温度にする、請求項5〜7のいずれかに記載の方法。
- 過剰の脂質組成物拭き取るステップh)は吸収によって行われる、請求項5〜8のいずれかに記載の方法。
- ポリマー材料上へ液状脂質組成物を塗布するステップf)は、ハケ塗り、スピンコーティング又はディップコーティングによって行われる、有利にはハケ塗り又はスピンコーティングによって行われる、請求項5〜9のいずれかに記載の方法。
- ポリマー材料上の脂質組成物の表面濃度は500μg/cm2〜2500μg/cm2である、請求項5〜10のいずれかに記載の方法。
- 得られる非生物学的皮膚モデルは、24mJ/m2〜45mJ/m2、有利には26mJ/m2〜33mJ/m2の表面自由エネルギー(γ)を有する、請求項5〜11のいずれかに記載の方法。
- 得られる非生物学的皮膚モデルは、0.0〜−2.5、有利には−0.2〜−1.7、より有利には−0.5〜−0.9の歪度係数を有する、請求項5〜12のいずれかに記載の方法。
- 化粧品性能を評価するための、又は、皮膚表面特性における汚染の効果を評価するための、請求項1〜4のいずれかに記載の非生物学的皮膚モデル、又は、請求項5〜13のいずれかに記載の方法で得られる非生物学的皮膚モデルの使用。
- 皮膚の物理化学を評価するための、又は、皮膚の表面自由エネルギー(γ)への脂質組成物又は皮膚トポグラフィの影響を評価するための、請求項1〜4のいずれかに記載の非生物学的皮膚モデル、又は、請求項5〜13のいずれかに記載の方法で得られる非生物学的皮膚モデルの使用。
