Abstract
Description
本開示は、バイオセンサー、バイオセンサーを含む検出デバイス、生体分子を検出する方法、及びバイオセンサーを製造する方法に関する。
汎用性及び低コストの点から、紙ベースの診断の用途に対する関心は高い。しかし、定量的な診断テストを紙形式で実施することは非常に困難であり、分析感度も懸念事項である。対照的に、シリコンフォトニックデバイスについて、注目すべき感度を有する一方で多重(多検体)検出能力を可能にするということが実証されている。シリコンフォトニクスはコストが重大な懸念事項である。

バス導波路1050に結合された一対のリング共振器1040、1045を有する説明したバイオセンサー1010の1つの実施態様において、ニトロセルロースメンブレン1060上の2つの場所、1062、1064のうちの1つに捕捉抗体の斑点をつける。これは、紙への単純な吸着または共有結合のいずれかによって行ってもよい。他方の領域1064は、対照分子(たとえば、抗フルオレセイン抗体)によって機能を持たせるか、または空白のままにして参照ゾーンを形成する。リング共振器1045と位置関係が合うように、抗体フォトニックチップ上にニトロセルロースメンブレンを配置する(図8)。ニトロセルロースメンブレン/フォトニックの「サンドイッチ」を、対象とする試料にさらした後、好適な潜伏期間の後に洗浄ステップを行う。
説明したバイオセンサーの別の実施態様では、ニトロセルロースメンブレン上に捕捉抗体の斑点をつける。メンブレンを試料にさらし、洗浄し、そして任意的に乾燥させることを、フォトニックチップと接触するように置く前に行う。参照を、メンブレンの空白領域によって、または抗フルオレセインなどの非反応性の抗体スポットと比べることによって行う。
ニトロセルロースメンブレンと接触するように置いたときにリング共振器が機能するか否か、及びその感度がリング共振器単体に匹敵するか否かを、ナノピュア水及びショ糖溶液を用いて評価した。図9A〜図9Bに、ナノピュア水(左の塊)またはショ糖溶液(右の塊)で飽和したメンブレンに対して収集したスペクトルを示す。図9Aにおいて、ナノピュア水スペクトルは1550.75nmにおいて塊になった共振波長を示し、5%ショ糖は1551.30nmにおいて示し、平均の共振波長シフトは0.559nm(σ=0.013nm)である。図9Bにおいて、ナノピュア水スペクトルは1548.85nmにおいて塊になった共振波長を示し、5%ショ糖は1549.45nmにおいて示し、平均の共振波長シフトは0.662nm(σ=0.039nm)である。
メンブレン上でのタンパク質のバルク吸着に起因する信号を観察できるか否かを次に評価した。図10に、ナノピュア水に浸漬させたニトロセルロースメンブレンと、ナノ孔水に浸漬させた1%BSAブロックを伴う500μg/mlのα−CRP抗体を伴ったニトロセルロースメンブレンとのスペクトルを示す。結果として得られる共振波長シフトは0.06nmである。これによって、多孔材料シートは、捕捉分子及び標的分子を(これらが捕捉されたときに)フォトニック検知デバイス上に、共振挙動を変えて出力光の中に検出可能な変化を形成できるように、適切に送出できることが確認される。
ニトロセルロースの細片を用いてリング共振器にタンパク質溶液を送出した。異なる濃度のウシ血清アルブミン(BSA)の5マイクロリットル試料をニトロセルロース細片に付与して、リング共振器を介して吸い上げさせた。濃度に依存する共振周波数の変化を観察した(図11)。デバイスのバルク屈折率感度は90.8nm/RIUと測定された(既知のショ糖溶液によって)。160nm/RIUという高いチップ感度が測定されたため、検出感度を大幅に高められる可能性がある。
リング共振器を用いてラテラルフローアッセイの結果を検出できる否かをテストするために、ヒト絨毛性ゴナドトロピンに対する市販のラテラルフローアッセイをリング共振器のバンクにわたって置いた。図12に、陽性対照バンド(斜線領域で示す)下のリングに対して、より強いシフトが観察されたことを示す。2つの別個の共振測定値(FSR、自由スペクトル領域)を表す2つのデータ点が、各リングに対して示されている。
Claims (20)
- 基材と、前記基材上または前記基材内に形成された、光曝露時に光信号を生成するのに適した3次元構造体とを含む、フォトニック検知デバイスと、
光信号を生成するのに適した前記3次元構造体を覆う多孔材料シートであって、1つ以上の捕捉分子を含む、前記多孔材料シートと、
前記フォトニック検知デバイスに接続された光学的に透明なカバー層であって、前記多孔材料シートは、前記カバー層と、前記3次元構造体を含む前記フォトニック検知デバイスの一部との間にある、前記カバー層と
を含む、バイオセンサー。 - (i)前記カバー層と前記フォトニック検知デバイスの前記一部との間の前記多孔材料シートを圧縮する、クランピングメカニズム、または
(ii)前記光学的に透明なカバー層の一部を前記フォトニック検知デバイスの前記基材に直接接続する、接着層
