FCS中のウシリポソーム/エクソソームは、4℃で18時間180000xgで沈降させた。上澄みを収集し、無血清であるが完全な培地(MEM)に加え、負圧下でフィルター滅菌(0.2μmフィルター、Sartorus)した。
HeLa(ATCC:CCL2)およびHEK293(AMSBIO:SCOO8)細胞の培養および継代は、供給元の説明に従って実施した。顕微鏡検査用の細胞を、1×105細胞/ウェルの密度で滅菌カバースリップに播種した。固定、抗体ハイブリダイゼーションおよび検出については、以前に記載されたように実施した[2]。CD63免疫染色細胞を冷メタノールで固定した。
タンパク質PA83をコードするDNA配列(GenBankアクセッション番号AAF86457およびAAT98414に基づく)は以前に記載されている[2]。LFn−PKRは、BioBasic Inc.(カナダ、オンタリオ州)によって、GenBankアクセッション番号AAY15237(LFnの場合)およびNM_002759(PKRの場合)を使用して合成された。LFn−PKRをコードするオープンリーディングフレームは、[2]およびPCT/GB2014/051918で以前に説明されているように、細菌発現カセットpET151/D(Invitrogen、Paisley、UK)にサブクローニングされた。LFn−黄色ブドウ球菌(Sa)Cas9およびGST−PA63をコードするプラスミドは、親プラスミドとしてpET151細菌発現システムを使用してInvitrogenによって合成された。使用したGST配列はpGEX3xからのものであり、SaCAS9配列はGenbankアクセッション番号CCK74173.1からコドン最適化された(すなわち、配列番号14)。V5エピトープタグおよび6xヒスチジンアフィニティータグの追加により、細菌溶解物からのタンパク質の免疫検出およびアフィニティー精製がなされた。
LFn−PKR−TxRおよびLFn−SaCas9−TxRは、以前に説明された方法を使用して調製された[14]。
細胞を175cm2のTC処理ディッシュに播種し、標準的なインキュベーション条件下、すなわち5%(v/v)CO2中37℃で増殖させた。90%のコンフルエンスで、細胞単層をPBSで3回洗浄した後、エクソソームをロードした。
細胞をPA83(50μg/mL)およびLFn−PKR−TxR(50μg/mL)またはLFn−SaCas9−TxR(50μg/mL)とともに3mLの無血清DMEMで37℃で1時間インキュベートした。 1時間後、10%(v/v)FCSを含む5mLのエクソソームフリーDMEMを最終容量8mLになるように添加し、プレートを37℃で3時間インキュベートした。
LFn−PKR(50μg/mL)は、PA83(50μg/mL)を添加する前に、無血清DMEMでGFP siRNA(50nM)と5分間インキュベートした。次に、混合物を細胞単層に加え、細胞を37℃で1時間インキュベートした。1時間後、10%(v/v)FCSを含む5mLのエクソソームフリーDMEMを最終容量8mLになるように添加し、プレートを37℃で3時間インキュベートした。
イオノマイシン(50μM)を培地に添加し、標準条件下で30分間インキュベートした。次に、培地を収集し、4℃で2分間1,500xgで遠心分離した後、細胞破片を沈殿させた。得られた上澄みを、凍結またはエクソソーム単離の前にフィルター滅菌(0.8μm、Sartorous)しました。エクソソームの分離を、次の3つの方法のいずれかを使用して実行した。
これは[15]から変更された。簡単に説明すると、凍結濾過した馴化培地を氷上で解凍し、EVを沈殿させるために10000xgに30分間さらしました。次に、上澄みを4℃で70分間110 000xgにさらし、ペレットを1mLのPBSに収集した。次に、再懸濁されたペレットを、100000xgで70分間4℃で2回目の沈降にかけた。次に、得られたペレットを1000μLのエクソソームフリー培地に懸濁し、0.2μmフィルター(Sartorus)でろ過し、必要になるまで−20℃で凍結保存した。
簡単に説明すると、清澄化および濾過された細胞培養培地の量を推定し、これに、0.5容量の単離試薬を加えた。この調製物を4℃で一晩放置した。次に、混合物を10,000xgで1時間4℃で遠心分離した。次に、得られたペレットを最終容量1000μLのエクソソームを含まない培地に懸濁し、必要になるまで−20℃で凍結保存した。
これは、QIAgenexoEasyキットを製造元の仕様に従って使用して実行された。簡単に説明すると、8mLのXPBバッファーを細胞培養試薬と混合し、分離後、エクソソーム調製物から溶出(XE)バッファーを除去するための追加のステップを追加した。これは、溶出液を110000xgで70分間4℃で遠心分離することによって達成された。次に、得られたペレットを1000μLのエクソソームを含まない培地に懸濁し、必要になるまで−20℃で凍結保存した。
ビシンコニン酸アッセイ(BCA)アッセイは、ビシンコニン酸キット(BCA−1)(Sigma Aldrich、Dorset UK)の仕様に従って実施し、保存前の最終的なエクソソームサンプルのタンパク質濃度を測定した。さらに、μlite(BioDrop Inc.)装置を使用して、メーカーの推奨プロトコルに従って、それぞれOD260およびOD280でのDNAおよびタンパク質濃度を測定した。
リポソーム/エクソソームの顕微鏡による視覚化は、等量のエクソソーム調製物を、Cy5フルオロフォア(カタログ番号C10046; Invitrogen、ペイズリー、英国)を組み込んだ等量の細胞マスク試薬と混合することによって行った。これにより、Airyscanユニット(Carl Zeiss Ltd、ドイツ)を備えたLSM880レーザー走査型共焦点顕微鏡を使用して、蛍光下でリポソーム/エクソソームのイメージングがなされた。Airyscanユニットの超解像機能により、リポソーム/エクソソームの解像が可能となった。テキサスレッドで標識されたタンパク質がロードされたリポソーム/エクソソームの場合、リポソーム/エクソソームは、LSM880のAiryscanユニット(Carl Zeiss Ltd、ドイツ)の超解像機能を使用して直接視覚化された。どちらの場合も、Plan−Ap℃hromat 63x /1.40開口数オイルDICf/ELYRA対物レンズが使用された。免疫染色は、前述のようにカバースリップ上で増殖させたパラホルムアルデヒド固定または冷(20℃)メタノール固定細胞のいずれかで実施した。
siRNAの輸送を評価するために、GFPに特異的なコントロールsiRNAを購入した。これは、HEK293(SCOO8)細胞で発現された安定して発現された導入遺伝子に対して向けられ、遺伝子活性を報告するための標準として使用された。無色の前駆体から不溶性の青色化合物へのx−galの加水分解に関与する酵素であるベータガラクトシダーゼにインフレームで融合したGFPを過剰発現したHEK293細胞を、620nmで分光光度的に検出した。その結果、ベータガラクトシダーゼを介したX−gal変換を測定することにより、GFPsiRNA活性をモニターすることができた。最後に、ベータガラクトシダーゼ活性は、タンパク質濃度に正規化した後(OD620)、未処理のコントロールのパーセンテージとして表された。
6ウェルプレートを使用して活性を評価した。細胞を5×105細胞/ウェル(HEK293、AMSBIO)で播種し、処理前に5%(v/v)CO2中37℃で24時間インキュベートした。
細胞を2mLの完全培地で希釈した200μLのエクソソーム調製物で処理した。この調製物をフィルター滅菌し(0.2μmフィルター、Sartorus)、その後、細胞と目的の時間(24時間、48時間、72時間)インキュベートした。
培地を廃棄し、細胞単層を冷却したPBSで3回注意深く洗浄した後、500μLのRIPAバッファー(R0278−50ML、Sigma Aldrich)を各ウェルに添加した。氷上での15分間のインキュベーション期間の後、各ウェルからの細胞溶解物を10回吸引し、標識されたエッペンドルフにデカントした。21,000xgで4℃で10分間遠心分離した後、上澄みを新しいエッペンドルフに移し、ペレットを廃棄した。次に、10μLのライセートを96ウェルプレートの100μLの2%BCA試薬に加え、37℃でインキュベートした。残りの400μLの上澄みを12μlのX−gal(DMSO中50mM)(R0404、ThermoFisher)と混合し、100μL/ウェルで96ウェルプレートに移した。X−Gal変換は、37℃に設定された分光光度計を使用して、620nmで経時的にアッセイされた(読み取りは5時間にわたって15分ごとに行われた)。
ウエスタンブロッティングおよび免疫検出は、製造元の指示に従ってミニテトラセル装置(BioRad)を使用して実行された。タンパク質の分離には、10%(w/v)アクリルアミドゲルを使用し、200Vで60分間泳動した。ニトロセルロースメンブレンへの転写は、400mMで60分間行った。0.1%(v/v)トゥイーン20試薬を含むPBS中の5%(w/v)脱脂粉乳溶液を使用して、45分間ブロッキングを行った。抗体のハイブリダイゼーションは、メーカーが提案する抗体希釈液を使用して、振とう条件下で37℃で60分間3mLで実施した。HRP標識二次抗体の検出は、強化されたECL試薬(Pierce、ThermoFisher Scientific)を使用して、製造元の指示に従って実行された。ゲルおよびブロットは、広範囲の染色済みタンパク質マーカー(Invitrogen)を実行することによって較正された。エクソソームタンパク質のTCA沈殿は、0.6倍量のTCAをエクソソーム調製物に添加することによって実行された。次に、これを4℃で30分間インキュベートした。次に、調製物を21000xgで4℃で10分間沈降させ、ペレットをアセトンで2回、同様に4℃で洗浄した。得られたペレットをLaemmliバッファーに溶解し、ウエスタンイムノブロッティングを行い、非還元条件下で抗LAMP2特異的一次抗体(DHSB、アイオワ大学、アイオワ州、米国);または、メーカーが提案する希釈液を使用したテキサスレッド特異的一次抗体(Vectorlabs)のいずれかでプローブした。
図1に示すように、Texas redでラベル付けされたカーゴ(つまり、Texas Redでラベル付けされたLFn−PKR)は、HeLa細胞内の管腔内構造に局在する。これらの管腔内構造はエクソソーム免疫マーカーCD63(LAMP3としても知られている)に対して陽性であるため、この管腔内シグナルは多小胞エンドソーム、すなわち後期エンドソーム内に存在し得る。これは、テキサスレッドで標識されたLFn−PKRが、PA83で細胞に添加されると、細胞に添加されてから3時間後にMVE/MVB内の管腔内膜を優先的に標識し得ることを示している。
図2は、Hela馴化培地から分離され、Cy5−Cell Maskで染色され、Airyscan検出器を使用して視覚化されたリポソーム/エクソソームが、エクソソームにほぼ適切なサイズ(60〜120 nm)であることを示す本発明者の発見を示しています。このシステムの解像度の限界は、x−yプレーンでは120nMであることに注意のこと。exoEasyキットから単離されたリポソーム/エキソソームも、プローブとしてLAMP2を使用したイムノブロッティング分析の対象となり、予測されるように、バンドはエキソソーム調製物内にほぼ適切な分子量で見られる。これは、exoEasyキットを使用して単離されたリポソーム/エクソソームがほぼ正しいサイズであるだけでなく、予測されるように十分に特徴付けられたエクソソーム免疫マーカーも含んでいたことを意味する。
リポソーム/エクソソームには、黄色ブドウ球菌Cas9、つまりSaCAS9が効果的にロードされている。図3では、LFn−SaCAS9からの赤色シグナルは、PA83およびTexasRedで標識されたLFn−SaCAS9に曝露されたHela細胞からのエクソソーム調製物ではっきりと見られ得る。コントロールリポソーム/エクソソームRwith:PA83なし、カーゴなし、または非転置カーゴ(BSA−Texas Red)は、焦点面を確認するためにセルマスクとインキュベートした場合でも赤い信号を生成しなかった(図3B)。
リポソーム/エクソソームは、小分子を効果的にロードすることがさらに示されている。図4では、小分子であるTexas Redが、3時間後にPA83およびTexasRedで標識されたLFn−PKRまたはPA83に曝露されたHela細胞からのエクソソーム調製物に取り込まれることが示されている。TexasRedでラベル付けされたBSAをネガティブコントロールとして使用した。ここで、テキサスレッドで標識されたLFn−PKRは、リポソーム/エクソソームの細胞マスク陽性集団内で容易に検出され得るが、テキサスレッドで標識されたBSAは検出され得ない。
図5は、PA83およびTexas Red標識LFn−SaCAS9、Texas Red標識LFn−PKR、またはTexasRed標識BSAのいずれかで調製したTCA沈殿リポソーム/エクソソームの結果を示している。PA83 Texas Redでラベル付けされたLFn−SaCAS9およびPA83 Texas Redでラベル付けされたLFn−PKR調製物から、TexasRedがペレットにはっきりと見える。テキサスレッドは、「無処理」またはPA83およびBSA−テキサスレッドの「処理済み」コントロールでは容易に検出され得ない。同様に、Texas Red特異的一次抗体を使用したイムノブロッティングおよび検出後、以前と同じTCA沈殿物から予測された分子量のタンパク質を標識するTexasRedが検出された。
exoEasyキットおよび分画遠心分離を使用して細胞培養培地から単離されたリポソーム/エクソソームの生物活性を図6に示す。ここでは、単位細胞タンパク質あたりのベータガラクトシダーゼ活性の低下が記録され、以下が示された:1)siRNAをリポソーム/エクソソームおよび 2)siRNAをロードしたリポソーム/エクソソームの生物学的活性、すなわち、細胞の第2の集団のサイトゾルにsiRNAを輸送するそれらの能力。
理論的な例として、第1の態様の方法によって生成されたリポソームは、ジカウイルスに感染した患者を治療するために使用される。最初に、生検が患者に対して行われ、患者の多くの細胞が単離される。次に、第1の態様の方法を使用して細胞からリポソームを生成し、抗ジカウイルスsiRNAをロードする。次に、抗ジカウイルスsiRNAを含むリポソームが患者に投与され、エンドサイトーシスを介して特許細胞に取り込まれる。したがって、抗ジカウイルスsiRNAは患者の細胞に存在し、ジカウイルスが患者の体内で増殖する能力は阻害される。
他の理論的な例として、第1の態様の方法によって生成されたリポソームは、FMO5調節性肥満に苦しんでいる患者を治療するために使用される。この例では、リポソームは、第1の態様の方法を使用して培養間葉系幹細胞から誘導され、抗FMO5siRNAがロードされる。次に、抗FMO5 siRNAを含むリポソームが患者に投与され、エンドサイトーシスを介して患者の細胞に取り込まれる。したがって、抗FMO5 siRNAは患者の細胞に存在し、FMO5酵素はダウンレギュレートされ、患者はもはや肥満関連の症状を示さない。
本発明者らは、PA83::LFn融合物を使用してリポソーム/エクソソームにロードした材料としては、低分子量共有結合複合体(すなわち、テキサスレッド)、LFn−PKR::siRNA、LFn−PKRと結合したテキサスレッド、LFn−Gal4::eGFP−Rab5およびLFn−SaCAS9と結合したテキサスレッドが挙げられる。その後、ここに提示されたデータは、LFn融合タンパク質が(LFまたはEFのみではない)選択的なカーゴを、エクソソームと呼ばれる生物学的に誘導されたステルスデリバリーシステムに輸送し得るという新しいアイデアを支持する。本明細書では、最初に、カーゴの過剰発現またはエクソソームの破壊なしにエクソソームのローディングを達成し得る方法論が開示され、この結論を裏付ける証拠が議論されている。
本発明者らは、使用したプラスミドを除いて、上記のようにPA83を使用して条件付き致死性カーゴLFn−ジフテリア毒素A鎖(DTA)をエクソソームにロードしたが、すなわち、そのプラスミドはAddgene(pET−15b LFn−DTA、Addgene番号11075)(https://www.addgene.org/11075/)からのものであった。
LFnDTA−配列番号15(タンパク質)および配列番号16(DNA)−をHeLa細胞とともに10μg/mlの濃度において、ならびに50μg/mlの配列番号2(PA83)を5%(v/v)CO2を含む加湿雰囲気において37℃で4時間、インキュベートすることによって、エクソソームをロードした。この後、細胞をPBSで洗浄し、無血清培地で5μMイオノマイシン(Sigma化学会社のカタログ番号I9657−1MG)とともにインキュベートした。次に、前述のように分画遠心分離を使用して、すぐに馴化された培地からエクソソームを単離した。
エクソソーム調製物の半分に、5μlの細胞培養トリプシン/EDTA(TE)バッファー(ThermoFisher Scientificカタログ番号25200056)をエキソソームに添加し、PBSで容量を100μlに調整した。調製物の残りの半分に、PBSを100μlに加えた。次に、エクソソーム調製物を、エクソソーム調製物に含まれるLFnDTAの量を大幅に超える5μgのLFnDTAを消化するのに十分であることがすでに実証されている条件下で60分間インキュベートした。次に、エクソソームをトリプシンコントロール(非毒性であることが判明している)を含むHeLa細胞の培養物に添加し、24時間後に細胞生存率をアッセイした。結果は、未処理のコントロールに対して正規化されたDTA活性(%)として表された(エクソソームを含むLFnDTAは細胞の約45%を殺した)。
図8を参照すると、PA83(すなわち配列番号2)を使用して最初にカーゴ(すなわちLFn−ジフテリア毒素A鎖(DTA)−配列番号15および配列番号16)がロードされたエクソソームは、その後、それらが外部酵素(すなわちトリプシン)の活性から保護されていることを示すために特徴付けられた。これらのデータは、子宮頸癌などの癌の治療に使用され得るLFnDTAをエクソソームに正常にロードし得ることを示している。
この実施例では、本発明者らは、動的光散乱AKA光子相関分光法を使用して、分画遠心分離を使用して単離されたエクソソームおよび細胞外小胞(EV)のサイズを特徴付けた。
HeLa細胞のエクソソームに、タンデムダイマートマトに特異的なプレハイブリダイズしたホスホロチオエート−ホスホジエステルハイブリッドアンチセンスオリゴヌクレオチド(ASO)を200pMol/ml(総ASO)−配列番号17および配列番号18の濃度においてロードした。次の表は、ホスホロチオエートコード(Thermofisher Webサイトから取得)を説明している。
馴化培地は、最初に1.5kxgで2分間の遠心分離によって除去され、0.8ミクロンのフィルターでろ過された。フロースルーを10000xgで30分間4℃で遠心分離した。最後に、エクソソームは、4℃で70分間110000xgで沈降させることにより単離され、PBSに再懸濁され、4℃で70分間110,000xgで再沈降された。エクソソームはそれまで4℃で保存されていた。細胞培養に使用する場合、エクソソームを必要な量の無血清培地で希釈し、細胞とインキュベートする前に0.8μフィルターで再度ろ過した。
図9を参照すると、50μg/ml PA83(配列番号2)および50μg/ml LFnPKR(配列番号13)を使用して200pMol/ml抗タンデムダイマートマト(TdTom)アンチセンスオリゴヌクレオチド(ASO)をロードすると、エクソソームのサイズが予測されることが示された。
本発明者らは、活性ASOをロードしたエクソソームがそれらの薬理学的活性を保持するかどうかを試験した。
HeLa細胞を200pMol/ml抗TdTomASO(配列番号17および配列番号18)および50μg/ml七量体(PA83 -配列番号2)、または強制八量体PA83変異体(すなわち25μg/ml−配列番号7(PA83 D512K)および25μg/ml配列番号8(PA83 K245G;R252N))および50μg/ml配列番号13(LFnPKR)とともにインキュベートすることにより、エクソソームをロードした。分離後、これらのASOをロードしたエクソソームは、GFP融合β−ガラクトシダーゼを標的コードし、HEK293細胞(ASMBIOのカタログ番号SC008)において過剰発現されるタンデムダイマートマトをバイシストロン的に発現するmRNAに対してアンチセンス活性を示した。HEK細胞(ウェルあたり約1x106)を次の方法で処理した:57μg[OD280で測定された総タンパク質]の七量体::ASOエキソソームプレップまたは42μg[OD280で測定された総タンパク質]の八量体::ASO。
図11を参照すると、ASOがロードされたエクソソームは薬理活性を有することが示されている。これらのデータは、この方法論を使用してASOをエクソソームにロードし得ること、およびエクソソームが以下の(i),(ii),(iii)および(iv)であることを示している:(i)融合能力がある;(ii)ASOを含む;(iii)ASOが活性である;および、(iv)エクソソームを使用してASOを輸送し得る。これらのデータは、PAの変異体を使用してエクソソームをロードし得ることも示している。
エクソソームは、50nMステルスリポーター抗GFPsiRNA(Invitrogenカタログ番号12935−145)、50μg/ml PA83(配列番号2)および50μg/ml LFnPKR(配列番号13)を無血清培地に再懸濁してロードした。エクソソームは、以前と同様に、すなわち、タンパク質::siRNA混合物との4時間のインキュベーション、および無血清培地中の5μMイオノマイシンとの30分間のインキュベーションの後に、単離された。すべてのインキュベーションは37℃で実施し、以前と同様にインキュベーションの間に細胞をPBSで洗浄した。 QiagenのexoEasyキット(カタログ番号76064)を使用してエクソソームを単離した。エクソソームをPBSで洗浄し、以前と同様に(すなわち、4℃で70分間110000xg)沈降させた後、500μlのPBSに再懸濁した。エクソソームを完全培地を使用して希釈し、GFP融合ベータガラクトシダーゼおよびタンデムダイマートマト(ASMBIOのカタログ番号SC008)を安定して発現するHEK293細胞に添加した。次に、細胞溶解物中のベータガラクトシダーゼ活性を、OD620でのX−gal変換を経時的に測定し、変換を総細胞溶解物タンパク質濃度に正規化することによってアッセイした。
本明細書に記載のリポソームの態様および実施形態の利点は、ILV逆融合の前に、Atx関連カーゴをロードしたILVをリポソーム(例えば、エクソソーム)として隔離することによって、先行技術に固有の1つまたは複数の問題に対処しようとすることにある。さらに、Atx PA63と同じ機能を実行するが、組換え膜貫通配列を含む膜貫通配列の使用についても対処する。

Claims (43)
- リポソームを調製する方法であって、少なくとも1つの細胞を、以下の(i)および(ii):
(i)孔形成タンパク質、またはその孔形成ドメインもしくは変異体もしくは断片;
(ii)生物活性ペイロード分子に付着していてもよいシャトルタンパク質、
と接触させることを含み、
ここで、前記孔形成タンパク質、またはその孔形成ドメインもしくは変異体もしくは断片は、少なくとも1つの前記細胞のリン脂質二重層を介して孔を作製し、かつ、前記シャトルタンパク質は、前記孔形成タンパク質、またはその孔形成ドメインもしくはその変異体もしくは断片と相互作用し、それによって前記細胞内に内在化され、それによってリポソームを作製し、この際、前記シャトルタンパク質に生物活性ペイロード分子がロードされていてもよい、方法。 - 前記リポソームを前記細胞から単離することをさらに含む、請求項1に記載の方法。
- 前記リポソームが小胞を含む、請求項1または2に記載の方法。
- 前記リポソームが、細胞外小胞(EV)、細胞内小胞、または管腔内小胞(ILV)を含む、請求項1〜3のいずれか1項に記載の方法。
- 前記リポソームがエキソソームを含む、請求項1〜4のいずれか1項に記載の方法。
- 前記リポソームが、10nm〜500nmの間、または20nm〜400nmの間、または30nm〜300nmの間、または40nm〜200nmの間、または50nm〜150nmの間、または60nm〜120nmの間の平均直径を有する、請求項1〜5のいずれか1項に記載の方法。
- 前記細胞が、哺乳動物またはヒトの細胞であってもよい生物学的細胞を含む、請求項1〜6のいずれか1項に記載の方法。
- 前記孔形成タンパク質、またはその孔形成ドメインもしくは変異体もしくは断片が、非毒性タンパク質を含むか、または非毒性タンパク質に由来する、請求項1〜7のいずれか1項に記載の方法。
- 前記孔形成タンパク質、またはその孔形成ドメインもしくは変異体もしくは断片が、炭疽菌またはリシンに由来する、請求項1〜8のいずれか1項に記載の方法。
- 前記孔形成タンパク質、またはその孔形成ドメインもしくは変異体もしくは断片が、炭疽菌病原性因子保護抗原(PA)に由来する、請求項1〜9のいずれか1項に記載の方法。
- 前記孔形成タンパク質、またはその孔形成ドメインもしくは変異体もしくは断片が炭疽菌83または炭疽菌63に由来する、請求項1〜10のいずれか1項に記載の方法。
- 前記孔形成タンパク質、またはその孔形成ドメインもしくは変異体もしくは断片が、配列番号1、2、5〜10のいずれか1つに記載のアミノ酸配列またはその変異体もしくは断片を含むか、または実質的にそれらからなる、請求項1〜11のいずれか1項に記載の方法。
- 前記シャトルタンパク質が、好ましくは生物活性ペイロード分子の、予備形成タンパク質の細孔を通る輸送を容易にするように構成される、請求項1〜12のいずれか1項に記載の方法。
- 前記シャトルタンパク質が弱毒化された毒素タンパク質を含む、請求項1〜13のいずれか1項に記載の方法。
- 前記シャトルタンパク質が炭疽菌由来の致死因子(LF)または浮腫因子(EF)である、請求項1〜14のいずれか1項に記載の方法。
- 前記シャトルタンパク質が、配列番号11に記載のアミノ酸配列またはその変異体もしくは断片を含むか、または実質的にそれらからなる、請求項1〜15のいずれか1項に記載の方法。
- 前記シャトルタンパク質がリンカータンパク質を含む、請求項1〜16のいずれか1項に記載の方法。
- 前記弱毒化された毒素において、炭疽菌致死因子タンパク質毒素の1つまたは複数の毒性ドメインII〜IVであってもよい少なくとも1つの毒素ドメインが、前記リンカータンパク質によって置き換えられている、請求項17に記載の方法。
- 前記リンカータンパク質が核酸結合ドメインを含む、請求項17に記載の方法。
- 前記核酸結合ドメインがサッカロミセス・セレビシエGAL4である、請求項19に記載の方法。
- 前記シャトルタンパク質が、配列番号12または13に記載のアミノ酸配列またはその変異体もしくは断片を含むか、または実質的にそれらからなる、請求項1〜20のいずれか1項に記載の方法。
- 前記生物活性ペイロード分子が、細胞サイトゾル内、核内、細胞小器官または小胞もしくは液胞などの細胞内構造内、細胞表面脂質膜内あるいは細胞内脂質膜内で活性である治療的に活性な分子である、請求項1〜21のいずれか1項に記載の方法。
- 前記生物活性化合物の分子量が1Da〜10MDaの間である、請求項1〜22のいずれか1項に記載の方法。
- 前記生物活性分子が:
(i)小分子、タンパク質、ヌクレオチド、DNAまたはDNA構築物、プラスミド、RNAまたはRNA構築物、mRNA、miRNA、ガイドRNA、snRNA、siRNA、アンチセンスオリゴヌクレオチド(ASO)、あるいは、
(ii)タンパク質もしくは酵素またはその断片、ヌクレアーゼ、あるいは抗体またはその抗原結合断片などの大分子、
ある、請求項1〜23のいずれか1項に記載の方法。 - 前記生物活性ペイロード分子が、ヌクレアーゼであってもよく、好ましくはCas9、Cpf1、TALEN、またはジンクフィンガーヌクレアーゼであるゲノム編集ツールを含む、請求項1〜24のいずれか1項に記載の方法。
- 前記生物活性分子が、配列番号14に記載のアミノ酸配列またはその変異体もしくは断片を含むか、または実質的にそれらからなる、請求項1〜25のいずれか1項に記載の方法。
- 請求項1〜26のいずれか1項に記載の方法により得られた、または得られ得るリポソーム。
- 管腔を取り囲むリン脂質二重層と、孔形成タンパク質またはその孔形成ドメインもしくは変異体もしくは断片と、シャトルタンパク質と、を含む、リポソーム。
- リポソームが請求項1〜26のいずれか1項で定義される通りのリポソームである、請求項28に記載のリポソーム。
- 治療または診断に使用するための、請求項27〜29のいずれか1項に記載のリポソーム。
- 疾患の治療、予防、または改善に使用するための、請求項27〜29のいずれか1項に記載のリポソーム。
- 治療される疾患が肥満、より好ましくはFMO5調節性肥満である、請求項31に記載のリポソーム。
- 治療される疾患が、男性型脱毛症(AGA);にきび;酒さ様皮膚炎;および前立腺癌からなる群から選択されるプロスタグランジンD2調節性疾患である、請求項31に記載のリポソーム。
- ジカ熱(またはジカウイルス病)、エボラウイルス病、後天性免疫不全症候群(ヒト免疫不全ウイルス)、Stat3応答性癌、P53欠損癌、ウイルス介在性子宮頸癌(すなわち、ヒトパピローマウイルス)、家族性高コレステロール血症、デュシェンヌ型筋ジストロフィー、脊髄性筋萎縮症、クローン病、または炎症性疾患、特に細胞内接着分子−1(ICAM−1)の過剰発現に関係する腸の炎症性疾患を治療、予防、または改善するために使用される、請求項31に記載のリポソーム。
- リポソームを作製するために使用される細胞が、治療される対象から得られたものである、請求項30〜34のいずれか1項に記載のリポソーム。
- 健常な細胞が前記対象の幹細胞株から、または前記対象の標的組織から得られ、問題の臨床状態の治療に適した治療用化合物がロードされるリポソームの作製に使用される前に、培養で増殖される、請求項35に記載のリポソーム。
- 前記対象から得られた細胞が、患者の生検からのものであってもよい健常な細胞を含まない、請求項35に記載のリポソーム。
- 請求項27〜29のいずれか1項に記載のリポソーム、および使用説明書を含むキット。
- ゲノム編集技術で使用するための、請求項27〜29のいずれか1項に記載のリポソーム。
- 請求項27〜29のいずれか1項に記載のリポソームに(i)ガイドRNAおよび/または(ii)ヌクレアーゼまたはヌクレアーゼをコードする遺伝子構築物をロードすること、ならびにロードされた前記リポソームを遺伝子編集療法において使用することを含む、ゲノム編集方法。
- 前記ヌクレアーゼが、Cas9またはCpf1、またはTALENまたはジンクフィンガーヌクレアーゼを含む、請求項40に記載の方法。
- プロテインキナーゼR(PKR)に付着した弱毒化毒素タンパク質を含む、シャトルタンパク質。
- 前記タンパク質が、配列番号11に記載のアミノ酸配列、またはその断片もしくは変異体を含むか、または実質的にそれらからなる、請求項42に記載のシャトルタンパク質。
