Description
本願は、2018年7月6日に出願された発明の名称が「低ADCC/CDC機能性抗PD−L1完全ヒト型モノクローナル抗体及びその使用」である中国特許出願第201810738565.8号、2018年8月14日に出願された発明の名称が「ヒトプログラムデスレセプターPD−1に対するIgG1サブクラスの低ADCC/CDC機能性抗体」である中国特許出願第201810924249.X号、及び、2019年5月7日に出願された発明の名称が「ヒトCTLA4に対するIgG1サブクラスの低ADCC/CDC機能性抗体」である中国特許出願第201910376852.3号に基づく優先権を主張するものであり、それらの開示内容全体を援用により本願に取り込む。
PD−1とPD−L1に対する抗体はいずれもこの経路を遮断し、T細胞の機能を部分的に回復させ、これらの細胞が腫瘍細胞を殺傷し続けられるようにすることができる。米国FDAは、2016年5月18日に、プラチナ含有化学療法を受ける間/受けた後、または、プラチナ含有化学療法のネオアジュバント療法/アジュバント療法を受けた12ヶ月以内に疾患が進行する局所進行性または転移性の尿路上皮がんの治療に用いられる、ジェネンテック(Genentech)社のAtezolizumab(商品名:Tecentriq)注射液を承認した。しかしながら、抗体依存性細胞傷害(ADCC)および補体依存性細胞傷害(CDC)機能の誘導を回避するために、当該抗体医薬品は、抗体の重鎖に対して脱グリコシル化変異を行ったため、生成物中の多量体の含有量が高くなり過ぎてしまい、薬物の免疫原性が著しく向上し、患者に複数回投与した後、抗薬物抗体(ADA)の産生の発生率が41.5%にも達している。
現在、CTLA4抗体の主な推進方向は、複数の標的に対するより効果的な併用投与であり、例えば、IpilimumabとNivolumab(抗PD−1抗体)とを併用したところ、その抗腫瘍効果が大幅に増加する。そのため、併用投与の分野では、CTLA4抗体の多くの有名な抗体医薬品(PD−1抗体Nivolumab、PD−L1抗体Atezolizumabなど)との併用療法によりその市場シェアが効果的に拡大されている。しかしながら、半数以上の患者においてグレード3〜4の重篤な副作用が現れ、抗CTLA−4抗体の研究開発と使用を制限する最も重要な要因になっている。
今までは、アミノ酸配列を挿入することにより、抗体医薬品における多量体の含有量とその免疫原性を低減させることが報告されていない。
本発明は、オリジナルのIgG1抗体の重鎖の定常領域に一定の長さの柔軟なアミノ酸配列を挿入する、IgG1サブクラス抗体のADCC/CDC機能を低減又は除去する方法を提供する。
(1)本発明は、PyMolソフトウェアを使用して、ジェネンテック(Genentech)社のAtezolizumabの構造を分析し、抗体の可変領域と定常領域との間で比較的柔軟な領域を探し、対応する領域に柔軟なアミノ酸配列を挿入し、対応する配列に対して遺伝子合成、シーケンスを実行し、シーケンスが正しい配列を選択する。
(2)本発明は、既知の抗体の重鎖のグリコシル化情報により、対応する修飾後の配列に対して全遺伝子合成を実行し、合成された遺伝子に対してシーケンスを実行するとともに、シーケンスが正しい配列を選択して次のステップを実行し、軽鎖の可変領域の酵素切断部位をKpn I+BamH Iとし、重鎖の可変領域の酵素切断部位をKpn I+Age Iとし、それぞれ、発現ベクターであるpJH16ベクターに接続し、それと同時に大腸菌DH5αを形質転換して、重鎖、軽鎖キメラ抗体の発現ベクターを得る。構築された抗体のシーケンスと配列の比較を同時に行う。
(3)上記の発現ベクターに対して、プラスミドの大量精製を行い、QiagenのEndoFree Plasmid Maxi Kitを選択する。
(4)選択されたプラスミドの組み合わせを最適化し、CHO細胞で一過性トランスフェクション発現を実行し、発現された抗体に対して親和性とEC50の検出を実行し(詳細は図2を参照)、検出結果により、組み合わせを選択して安定トランスフェクションを実行する。
(5)本発明では、以上の検出結果に基づいて、対応する組み合わせを選択し、エレクトロトランスフェクションにより安定株を構築し、同時に、MTXを用いて抗体の発現の程度をスクリーニングし、加圧後の安定株に対してモノクローナル抗体のスクリーニングを行い、最終的に抗体の収量の高い安定株を選択して、その後の実験に使用する。
(6)本発明によって作製されるモノクローナル抗体のマウス体内における免疫反応は、Atezolizumabより弱い。
本発明は、上記の軽鎖の可変領域と重鎖の可変領域を備える組み合わせから、スクリーニングによりH2L0モノクローナル抗体の配列を得る。当該抗体は、本来の、ヒトPD−1に対する結合親和性を維持しながら、PD−1とPD−L1との結合を特異的に遮断でき、ADCC機能とCDC機能の活性を低減させる。そして、本発明のモノクローナル抗体は、IgG1サブクラスのモノクローナル抗体であるため、IgG4サブクラスであるオリジナルのモノクローナル抗体Pembrolizumabに比べて、生産プロセスがより成熟で、難度が低く、抗体の耐重合性、発現量及び安定性が全て向上している。
本発明は、治療量の本発明の抗体を治療が必要な対象に投与することを含む、免疫細胞の活性を高める方法を提供する。前記方法は、癌の治療に使用でき、また、免疫力低下疾患の治療にも使用できる。本発明は、また、第2の治療剤又は治療形態をさらに与えることを含む。
本発明は、腫瘍細胞の殺傷又は免疫力変性疾患の治療における、上記IgG1サブクラスの低ADCC/CDC機能性CTLA4モノクローナル抗体の使用を提供する。
「抗体」とは、所望の生物学的活性(例えば、リガンドとその受容体との結合の阻害、またはリガンドによって誘導される受容体のシグナル伝達の阻害)を示す任意の形態の抗体を指す。したがって、前記抗体は最も広い意味で使用され、モノクローナル抗体、ポリクローナル抗体、および多重特異性抗体を含むが、これらに限定されるものではないことは明確である。
「Fc」領域は、抗体のCH1ドメインとCH2ドメインを含む2つの重鎖フラグメントである。2つの重鎖フラグメントは、2つ以上のジスルフィド結合とCH3ドメインの疎水性相互作用により結合される。
本発明では、PyMolソフトウェアを使用して、ジェネンテック(Genentech)社のAtezolizumabの構造を分析し、抗体の可変領域と定常領域との間で比較的柔軟な領域を探し、対応する領域に柔軟なアミノ酸配列を挿入して、抗体の可変領域の抗原への結合によって生じるメカニカルストレスの伝達を遮断し、抗体医薬品の重鎖の定常領域とFc受容体および/または補体との結合部位を十分に露出させず、抗体医薬品と、NK細胞、マクロファージ及び好中球等のIgG Fc受容体を発現するキラー細胞との結合または補体との結合を弱め、ADCCとCDCを誘導できないようにするか、又はADCCとCDCを誘導するシグナルを低減した。
本発明では、米国メルク(Merck)社のPembrolizumabを操作した。まず、その重鎖のサブクラスをIgG4からIgG1に変更し、PyMolソフトウェアを使用して構造を分析し、抗体の可変領域と定常領域との間で比較的柔軟な領域を探し、対応する領域に柔軟なアミノ酸配列を挿入して、対応する配列に対して遺伝子合成、シーケンスを実行し、シーケンスが正しい配列を選択し、スクリーニングしてモノクローナル抗体H1L0を得た。本発明は、オリジナルのIgG1サブクラス抗体の重鎖のCDR3領域とCH2領域との間に一定の長さの柔軟なアミノ酸配列を挿入することを試み、挿入された柔軟なアミノ酸配列は、抗体の可変領域の抗原への結合によって生じるメカニカルストレスの伝達を遮断し、抗体の重鎖の定常領域とFc受容体および/または補体との結合部位を十分に露出させず、抗体と、NK細胞、マクロファージ及び好中球といったIgG Fc受容体を発現するキラー細胞との結合または補体との結合を弱め、ADCCとCDCを誘導できないようにするか、又はADCCとCDCを誘導するシグナルを低減した。柔軟なアミノ酸配列は、その両端のドメインの空間的コンフォメーションによる相互干渉を防止または低減するための、側鎖の小さいアミノ酸からなる一定の長さのC−ペプチドであり、その配列は、GGSGGS、GSGGSGG、GSGGSGGG、GGGGSGGG、GSGSG、GGSGG、GGS、GGSGSおよびGGGSを含むが、これらに限定されるものではない。本発明の方法に従って操作された重鎖のアミノ酸配列は、配列番号7が示すとおりである。
本発明では、米国ブリストルマイヤーズスクイブ(BMS)社のipilimumabを操作し、PyMolソフトウェアを使用して構造を分析し、抗体の可変領域と定常領域との間で比較的柔軟な領域を探し、対応する領域に柔軟なアミノ酸配列を挿入して、対応する配列に対して遺伝子合成、シーケンスを実行し、シーケンスが正しい配列を選択し、スクリーニングしてモノクローナル抗体H1L0を得た。本実施例は、モノクローナル抗体ipilimumabのIgG1サブクラス抗体の重鎖のCDR3領域とCH2領域との間に、一定の長さの柔軟なアミノ酸配列を挿入することを試み、挿入された柔軟なアミノ酸配列は、抗体の可変領域の抗原への結合によって生じるメカニカルストレスの伝達を遮断し、抗体の重鎖の定常領域とFc受容体および/または補体との結合部位を十分に露出させず、抗体と、NK細胞、マクロファージ及び好中球といったIgG Fc受容体を発現するキラー細胞との結合または補体との結合を弱め、ADCCとCDCを誘導できないようにするか、又はADCCとCDCを誘導するシグナルを低減した。柔軟なアミノ酸配列は、その両端のドメインの空間的コンフォメーションによる相互干渉を防止または低減するための、側鎖の小さいアミノ酸からなる一定の長さのC−ペプチドであり、その配列は、GGSGGS、GSGGSGG、GSGGSGGG、GGGGSGGG、GSGSG、GGSGG、GGS、GGSGSおよびGGGSを含むが、これらに限定されるものではない。操作後の重鎖のアミノ酸配列は、配列番号10が示すとおりである。
本発明は、対応する修飾後の配列に対して全遺伝子合成を実行し、合成された遺伝子に対してシーケンスを実行するとともに、シーケンスが正しい配列を選択して次のステップを実行し、軽鎖の酵素切断部位をEcoR I+Hind IIIとし、重鎖の酵素切断部位をEcoR I+Hind IIIとし、それぞれ、発現ベクターpEE6.4及びベクターpEE12.4に接続し、それと同時に大腸菌DH5αを形質転換して、抗体の重鎖、軽鎖の発現ベクターを得た。構築された抗体のシーケンスと配列の比較を同時に行い、操作後のCTLA4モノクローナル抗体と元の抗体Ipilimumabの重鎖のアライメント結果を図13に示す。
本発明における抗体のADCC機能を評価するために、本発明では、標的タンパク質(PD−L1、PD−1、CTLA4)過剰発現細胞に対するFcR−TANK細胞の殺傷能力を検出することにより、抗体のADCC活性を評価した。
実施例1〜3で構築されたベクターに対して、プラスミドの大量精製を行い、QiagenのEndoFree Plasmid Maxi Kitを選択し、選択したプラスミドの組み合わせを最適化しながら、CHO細胞を使用して一過性トランスフェクション発現を実行し、見かけ結合親和性の測定に、タンパク質によるELISA実験を使用した(EC50値として報告される)。改良型抗体と先発医薬品である抗体の結合能力を比較した。
この方法は、CHO−PD−L1−CD3L細胞株及びJurkat−PD−1−NFAT細胞株の2つの細胞株に基づくものである。CHO−PD−L1−CD3L細胞株は、PD−L1とanti−CD3−scFvを安定的に発現し、標的細胞とする;Jurkat−PD−1−NFAT細胞株は、PD−1とルシフェラーゼ(luciferase)を安定的に発現し、ルシフェラーゼ遺伝子は、NFAT要素(転写因子)によって制御され(IL−2プロモーター)、エフェクト細胞とする。PD−1とPD−L1の結合により、CD3の下流のシグナル伝達を遮断し、ルシフェラーゼの発現を阻害できる。PD−1抗体またはPD−L1抗体を添加すると、このような遮断(Block)効果が逆転し、ルシフェラーゼの表現が可能になるため、蛍光シグナルが検出される。具体的な実験手順は次のとおりである。
2、細胞播種:CHO−PD−L1−CD3L細胞を96ウェルプレートに50000cells/wellで播種し、37℃、5%CO2の条件で12〜14時間培養した。その後、培地を除去してから、100,000個のJurkat−PD−1−NFAT細胞を含む50μLのアッセイ培地(2%FBSを含むRPMI1640培地)を各ウェルに加えた。
3、抗体状態スクリーニング:本発明の操作後の抗体または先発医薬品を10μg/mLの濃度から1:3の比率で段階希釈した(アッセイ培地を使用した)。その後、希釈した抗体を各ウェルに加え、37℃、5%CO2で6時間インキュベートした。
4、100μLの蛍光基質(Promega Bio−GloTM Luciferase Assay)を添加し、フルオロメーター(SpectraMax M5)で相対ルシフェラーゼ活性単位を計算した。
ELISA法により、本発明のモノクローナル抗体のCTLA4のCD80またはCD86との結合の阻害効率を検出した。4℃で一晩置いて、hCD80又はhCD86を酵素標識プレートに固相化し、3%BSA含有PBSとともに室温で1時間インキュベートすることにより、非特異的結合部位をブロックした。コーティング後、前記プレートをPBSで洗浄し、抗CTLA4抗体とBiotin−CTLA4の混合希釈液(Biotin−CTLA4は1μg/mLの固定濃度であり、本発明の抗CTLA4抗体を段階希釈し、最高濃度は1μg/mLであり、最低濃度は0である。)を調製し、固相化タンパク質とともに25℃で1時間インキュベートした。結合後、前記プレートをPBSで3回洗浄し、25℃で1/8000に希釈したペルオキシダーゼ標識アビジンを含む結合緩衝液により25℃で1時間インキュベートし、再度洗浄した後、TMBで発色させた。ELISAの結果を図16に示し、本発明のCTLA4モノクローナル抗体がCTLA4のCD80またはCD86との結合を阻害できることを示した。
マウスをそれぞれ改良型抗体H−1L0及び先発医薬品Atezolizumabで免疫して、マウスにおいて産生したADAの感染価を評価した。Balb/cマウスを用いて群分けし、実験を行い、群あたり5匹とした。具体的な免疫方法は次のとおりである。1)基本免疫:抗原とフロイント完全アジュバントを同体積で混合して完全に乳化し、注射箇所を分け、皮下注射した。2)追加免疫:追加免疫では、抗原とフロイント不完全アジュバントのエマルジョンを使用した。Balb/cマウス1匹あたりの総注入量を70μgとした。
本発明の操作後のPD−L1抗体および先発医薬品である抗体AtezolizumabのCHO安定細胞バンクを同時に構築し、当分野で公知の抗体医薬品製造基準に従って、改良型PD−L1抗体および先発医薬品Atezolizumabの、5gの小規模発現および精製を行った。図6Aおよび図6Bに示すように、得られた抗体に対してHPLC−SECにより比較を行った。先発医薬品Atezolizumab及び改良型PD−L1抗体H−1L0のロード量(50μL)と濃度(60mg/mL)は、いずれも同じであった。図から、ロード量が同じである場合、先発医薬品である抗体Atezolizumabのメインピークの面積が、改良型PD−L1抗体H−1L0に比べて約10%も減少したことが分かった。したがって、先発医薬品Atezolizumabにおける多量体の含有量がより多いため、HPLC−SECのロード中に、多量体がフィルターメンブレンによって除去され、ピーク面積が減少したと判断できる。2つの抗体のHPLC−SECのピーク面積の計算結果を表6に示す。同時に、HPLC−SECの結果は、改良型PD−L1抗体H−1L0のピーク位置が先発医薬品Atezolizumabより前にあることを示しており、抗体のグリコシル化の回復により抗体の分子量が増加し、ピーク位置がより前になったと判断できる。
実施例9で得られた結果に基づいて、改良型PD−L1抗体の糖鎖構造をさらに検証した。主に、当該分野における公知の方法で実行した。詳しくは、抗体のオリゴ糖を消化して収集した後、質量分析によって糖鎖構造を分析・特定した。図7に示すように、特定により、先発医薬品Atezolizumabでは、N型糖鎖修飾が検出されなかったが、改良型PD−L1抗体は正常にグリコシル化修飾されていた。改良型PD−L1抗体の糖鎖構造分析の結果を表7に示す。
本発明の操作後のPD−L1モノクローナル抗体及び先発医薬品である抗体Atezolizumabの安定性を比較検証した。改良型PD−L1抗体及び先発医薬品である抗体Atezolizumabを同時に急速熱処理し、その安定性を検証するための主な操作手順は以下のとおりである。

- オリジナルのIgG1サブクラス抗体の重鎖の定常領域に一定の長さの柔軟なアミノ酸配列を挿入することを特徴とする、IgG1サブクラス抗体のADCC/CDC機能を低減又は除去する方法。
- オリジナルのIgG1サブクラス抗体の重鎖のCDR3領域とCH2領域との間に、一定の長さの柔軟なアミノ酸配列を挿入することを特徴とする、請求項1に記載の方法。
- 前記挿入された一定の長さの柔軟なアミノ酸配列が、抗体の可変領域の抗原への結合によって生じるメカニカルストレスの伝達を遮断し、抗体の重鎖の定常領域とFc受容体および/または補体との結合部位を十分に露出させず、抗体と、NK細胞、マクロファージ及び好中球といったIgG Fc受容体を発現するキラー細胞との結合または補体との結合を弱め、ADCCとCDCを誘導できないようにするか、又はADCCとCDCを誘導するシグナルを低減することを特徴とする、請求項1〜2のいずれか1項に記載の方法。

- 前記モノクローナル抗体が、キメラ型モノクローナル抗体、ヒト化モノクローナル抗体、完全ヒト型モノクローナル抗体、二重特異性抗体又は融合タンパク質であることを特徴とする、請求項6に記載の低ADCC/CDC機能性モノクローナル抗体。
- 前記モノクローナル抗体が、PD−L1、PD−1、CTLA4、TNF−α、PCSK9、NGF、C5、Aβ、IL−6、IL−17A、IL23A、HGF、CMET、Notch1、CCL11、IL6R、IL−31R、IL−1B、IL−20、CD40、CD47、DKK1/2、TIGIT、4−1BB、IGF−1R、LL4、PDGFR2、HER3、IGF1/2、RANKL、sclerostin、GCGR、CGRP、ANGPTL3、IL−13、IL−4R、CSF−1R、TFPI、FCGRT、CD47のモノクローナル抗体であることを特徴とする、請求項6または7に記載の低ADCC/CDC機能性モノクローナル抗体。
- IgG1サブクラス抗体のADCC/CDC機能の低減又は除去における、請求項8に記載の低ADCC/CDC機能性モノクローナル抗体の使用であって、前記モノクローナル抗体が、オリジナルのIgG1サブクラス抗体の重鎖のCDR3領域とCH2領域との間に、一定の長さの柔軟なアミノ酸配列を挿入して得られるものであり、前記柔軟なアミノ酸配列が、両端のドメインの空間的コンフォメーションによる相互干渉を防止または低減するための、側鎖の小さいアミノ酸からなる一定の長さのC−ペプチドであり、その配列が、GGSGGS、GSGGSGG、GSGGSGGG、GGGGSGGG、GSGSG、GGSGG、GGS、GGSGSおよびGGGSを含むが、これらに限定されるものではないことを特徴とする、使用。
- IgG1サブクラス抗体のADCC/CDC機能の低減又は除去における、柔軟なアミノ酸配列の使用。
- 前記柔軟なアミノ酸配列が、オリジナルのIgG1サブクラス抗体の重鎖の定常領域に挿入されることを特徴とする、請求項10に記載の使用。
- 前記柔軟なアミノ酸配列が、オリジナルのIgG1抗体の重鎖のCDR3とCH2との間に挿入されることを特徴とする、請求項11に記載の使用。
- 前記柔軟なアミノ酸配列の挿入部位が、GG(138/139)、SS(177/178)、SG(178/179)、SS(184/185)、SSS(191/192/193)、LL(235/236)、GG(237/238)を含むが、これらに限定されるものではないことを特徴とする、請求項10〜12のいずれか1項に記載の使用。
- 前記柔軟なアミノ酸配列が、GGSGGS、GSGGSGG、GSGGSGGG、GGGGSGGG、GSGSG、GGSGG、GGS、GGSGSおよびGGGSを含むが、これらに限定されるものではないことを特徴とする、請求項13に記載の使用。
- 抗体の安定性の向上/抗体の免疫原性の低下/抗体の半減期の延長における、柔軟なアミノ酸配列の使用。
- 前記柔軟なアミノ酸配列が、オリジナルのIgG1抗体の重鎖の定常領域に挿入され、好ましくは、前記柔軟なアミノ酸配列が、オリジナルのIgG1抗体の重鎖のCDR3とCH2との間に挿入され、より好ましくは、前記柔軟なアミノ酸配列が、GGSGGS、GSGGSGG、GSGGSGGG、GGGGSGGG、GSGSG、GGSGG、GGS、GGSGSおよびGGGSを含むが、これらに限定されるものではないことを特徴とする、請求項15に記載の使用。
- オリジナルのモノクローナル抗体Atezolizumabの重鎖の定常領域に、一定の長さの柔軟なアミノ酸配列が挿入され、抗体の可変領域の抗原への結合によって生じるメカニカルストレスの伝達が遮断され、モノクローナル抗体の重鎖の定常領域とFc受容体および/または補体との結合部位が十分に露出せず、モノクローナル抗体と、NK細胞、マクロファージ及び好中球といったIgG Fc受容体を発現するキラー細胞との結合または補体との結合が弱められ、ADCCとCDCを誘導できなくなり、又はADCCとCDCを誘導するシグナルが低減されたことを特徴とする、低ADCC/CDC機能性抗PD−L1モノクローナル抗体。
- オリジナルのモノクローナル抗体Atezolizumabの重鎖のCDR3領域とCH2領域との間に、一定の長さの柔軟なアミノ酸配列が挿入され、抗体の抗原への結合によって生じる抗体の可変領域と定常領域との間のストレスの伝達が遮断され、前記オリジナルのモノクローナル抗体Atezolizumabの重鎖の全長のアミノ酸配列が、配列番号1が示すとおりであり、その軽鎖の全長のアミノ酸配列が、配列番号2が示すとおりであることを特徴とする、請求項17に記載の低ADCC/CDC機能性抗PD−L1モノクローナル抗体。
- 軽鎖の全長に、配列番号2が示すアミノ酸配列又はその変異体が含まれ、重鎖の全長に、配列番号3〜4のいずれかが示すアミノ酸配列が含まれることを特徴とする、請求項17に記載の低ADCC/CDC機能性抗PD−L1モノクローナル抗体。
- 請求項17〜19のいずれか一項に記載の低ADCC/CDC機能性抗PD−L1モノクローナル抗体をコードする遺伝子。
- 発現カセット、発現ベクター、人工細菌又は細胞である、請求項20に記載の遺伝子を含む生物学的材料。
- 請求項20に記載の遺伝子または請求項21に記載の生物学的材料の下記のいずれかの使用。
(1)PD−L1又はPD−1を標的とする疾患を治療する医薬品の調製における使用、
(2)PD−L1又はPD−1を標的とする疾患の治療における使用、
(3)腫瘍又は免疫力変性疾患の治療における使用。 - 請求項17〜19のいずれか一項に記載の低ADCC/CDC機能性抗PD−L1モノクローナル抗体の下記のいずれかの使用。
(1)PD−L1又はPD−1を標的とする疾患を治療する医薬品の調製における使用、
(2)腫瘍を治療する医薬品の調製における使用、
(3)免疫力変性疾患を治療する医薬品の調製における使用、
(4)PD−L1又はPD−1を標的とする疾患の治療における使用、
(5)腫瘍又は免疫力変性疾患の治療における使用。 - 請求項17〜19のいずれか一項に記載の低ADCC/CDC機能性抗PD−L1モノクローナル抗体を含む医薬品又は検出試薬。
- オリジナルのモノクローナル抗体Pembrolizumabの重鎖の定常領域に、一定の長さの柔軟なアミノ酸配列が挿入され、抗体の可変領域の抗原への結合によって生じるメカニカルストレスの伝達が遮断され、モノクローナル抗体の重鎖の定常領域とFc受容体および/または補体との結合部位が十分に露出せず、モノクローナル抗体と、NK細胞、マクロファージ及び好中球といったIgG Fc受容体を発現するキラー細胞との結合または補体との結合が弱められ、ADCCとCDCを誘導できなくなり、又はADCCとCDCを誘導するシグナルが低減されたことを特徴とする、低ADCC/CDC機能性抗PD−1モノクローナル抗体。
- オリジナルのモノクローナル抗体Pembrolizumabの重鎖のCDR3領域とCH2領域との間に、一定の長さの柔軟なアミノ酸配列が挿入され、抗体の抗原への結合によって生じる抗体の可変領域と定常領域との間のストレスの伝達が遮断され、前記オリジナルのモノクローナル抗体Pembrolizumabの重鎖の全長のアミノ酸配列が、配列番号5が示すとおりであり、軽鎖の全長のアミノ酸配列が、配列番号6が示すとおりであることを特徴とする、請求項25に記載のモノクローナル抗体。
- 軽鎖の全長に、配列番号6が示すアミノ酸配列が含まれ、重鎖の全長に、配列番号7が示すアミノ酸配列が含まれることを特徴とする、請求項25に記載のモノクローナル抗体。
- 請求項25〜27のいずれか一項に記載の低ADCC/CDC機能性抗PD−1モノクローナル抗体をコードする遺伝子。
- 発現カセット、発現ベクター、人工細菌又は細胞である、請求項28に記載の遺伝子を含む生物学的材料。
- 請求項28に記載の遺伝子または請求項29に記載の生物学的材料の下記のいずれかの使用。
(1)PD−L1又はPD−1を標的とする疾患を治療する医薬品の調製における使用、
(2)PD−L1又はPD−1を標的とする疾患の治療における使用、
(3)腫瘍又は免疫力変性疾患の治療における使用。 - 請求項25〜27のいずれか一項に記載の低ADCC/CDC機能性抗PD−1モノクローナル抗体の下記のいずれかの使用。
(1)PD−L1又はPD−1を標的とする疾患を治療する医薬品の調製における使用、
(2)腫瘍を治療する医薬品の調製における使用、
(3)免疫力変性疾患を治療する医薬品の調製における使用、
(4)PD−L1又はPD−1を標的とする疾患の治療における使用、
(5)腫瘍又は免疫力変性疾患の治療における使用。 - 請求項25〜27のいずれか一項に記載の低ADCC/CDC機能性抗PD−1モノクローナル抗体を含む医薬品又は検出試薬。
- オリジナルのモノクローナル抗体Ipilimumabの重鎖の定常領域に、一定の長さの柔軟なアミノ酸配列が挿入され、抗体の可変領域の抗原への結合によって生じるメカニカルストレスの伝達が遮断され、モノクローナル抗体の重鎖の定常領域とFc受容体および/または補体との結合部位が十分に露出せず、モノクローナル抗体と、NK細胞、マクロファージ及び好中球といったIgG Fc受容体を発現するキラー細胞との結合または補体との結合が弱められ、ADCCとCDCを誘導できなくなり、又はADCCとCDCを誘導するシグナルが低減されたことを特徴とする、低ADCC/CDC機能性抗CTLA−4モノクローナル抗体。
- オリジナルのモノクローナル抗体Ipilimumabの重鎖のCDR3領域とCH2領域との間に、一定の長さの柔軟なアミノ酸配列が挿入され、抗体の抗原への結合によって生じる抗体の可変領域と定常領域との間のストレスの伝達が遮断され、前記オリジナルのモノクローナル抗体Ipilimumabの重鎖の全長のアミノ酸配列が、配列番号8が示すとおりであり、軽鎖の全長のアミノ酸配列が、配列番号9が示すとおりであることを特徴とする、請求項33に記載の低ADCC/CDC機能性抗CTLA−4モノクローナル抗体。
- 軽鎖の全長に、配列番号9が示すアミノ酸配列が含まれ、重鎖の全長に、配列番号10が示すアミノ酸配列が含まれることを特徴とする、請求項34に記載の低ADCC/CDC機能性抗CTLA−4モノクローナル抗体。
- 請求項33〜35のいずれか一項に記載の低ADCC/CDC機能性抗CTLA−4モノクローナル抗体をコードする遺伝子。
- 発現カセット、発現ベクター、人工細菌又は細胞である、請求項36に記載の遺伝子を含む生物学的材料。
- 請求項36に記載の遺伝子又は請求項37に記載の生物学的材料の下記のいずれかの使用。
(1)CTLA−4を標的とする疾患を治療する医薬品の調製における使用、
(2)CTLA−4を標的とする疾患の治療における使用、
(3)腫瘍の治療又は免疫力の向上における使用。 - 請求項33〜35のいずれか一項に記載の低ADCC/CDC機能性抗CTLA−4モノクローナル抗体の下記のいずれかの使用。
(1)CTLA−4を標的とする疾患を治療する医薬品の調製における使用、
(2)腫瘍を治療する医薬品の調製における使用、
(3)免疫力を強化させる医薬品の調製における使用、
(4)CTLA−4を標的とする疾患の治療における使用、
(5)腫瘍細胞又は免疫力変性疾患の治療における使用。 - 請求項33〜35のいずれか一項に記載の低ADCC/CDC機能性抗CTLA−4モノクローナル抗体を含む、医薬品又は検出試薬。
