Abstract
Description
以下の実施例では、様々な量のカバノキ樹皮抽出物を有する異なるナノ分散物を用いた。ナノ分散物をPVA溶液と混合し、混合物を電界紡糸した。実施例1〜4の異なる混合物を以下に示す。
本発明の実施形態の優れた活性を示すために、ブタのエクスビボ創傷治癒アッセイを実施した。簡潔に言うと、屠畜場(食用)から得られたブタの耳を虐殺後に実験室に直接運んで、洗浄し消毒した。その後、耳のひだから6mmのパンチ生検片を採取し、脂肪および皮下組織を除去した。したがって、7.1mm2の中央領域において表皮および真皮上層を除去することによって創傷を生じさせた。次に、そのように形成したエクスビボの創傷治癒モデルを、培養皿内のガーゼ上に真皮を下にして配置し、ヒドロコルチゾン、2%のウシ胎児血清、ペニシリンおよびストレプトマイシンを添加したダルベッコ改変イーグル培地によって気液界面で培養した。4cm2の電界紡糸マット/5μlのオレオゲルを、傷をつけた後、直ちに適用し、このモデルを37℃および5%二酸化炭素(CO2)中で48時間にわたって培養した。さらなる工程は急速冷凍を含み、創傷治癒モデルの中央部のクライオスタット部分を、顕微鏡でルーラーを用いて識別し、評価中に創傷の全長をチェックすることにより、ヘマトキシリン−エオジンで染色した。創傷治癒プロセス(表皮再形成)は、顕微鏡で創縁と再生成された表皮の先端部との間の距離を測定することにより評価した。
a)さらなる実験では(結果は図2に図示せず)、実施例5による調製物を、カバノキ樹皮抽出物を本質的に含まない調製物と比較する。驚いたことに、カバノキ樹皮抽出物を含まない調製物で得られた結果は、カバノキ樹皮抽出物を有するものに匹敵する。
Claims (15)
- 少なくとも1種のリン脂質で安定化されたナノ分散物を含有する組成物。
- 前記ナノ分散物は、カバノキ樹皮抽出物をさらに含有することを特徴とする、請求項1に記載の組成物。
- 前記ナノ分散物は、医薬として許容可能なポリマー担体を組み込まれた電界紡糸繊維内に含有されることを特徴とする、請求項1または2に記載の組成物。
- 前記リン脂質は、ホスファチジルコリン、ホスファチジルエタノールアミン、ホスファチジルイノシトール、ホスファチジン酸(phosphatic acid)、およびホスファチジルセリン、またはそれらの組み合わせからなる群から選択されることを特徴とする、請求項1〜3のいずれか1項に記載の組成物。
- 前記ナノ分散物は、高せん断乳化もしくは超音波乳化、または高圧均質化もしくはマイクロ流体化によって調製されることを特徴とする、請求項1〜4のいずれか1項に記載の組成物。
- 前記ナノ分散物の粒子の少なくとも95%は、1μm未満の粒径を有することを特徴とする、請求項1〜5のいずれか1項に記載の組成物。
- 前記ナノ分散物中のカバノキ樹皮抽出物は、重量パーセントで計算された下記の成分:
74〜85重量%のベツリン、
1.0〜4.0重量%のルペオール、
3.0〜5.0重量%のベツリン酸、
0.3〜2.8重量%のエリトロジオール、
および
2〜13重量%の、特にオレアノール酸、ベツリン酸、ベツリン酸のメチルエステルのようなカバノキ樹皮に由来する他の成分を含有し、すべての成分は合計で100重量%となることを特徴とする、請求項2〜4のいずれか1項に記載の組成物。 - 前記ナノ分散物は、0.1重量%〜5.0重量%未満のカバノキ樹皮抽出物を含有することを特徴とする、請求項1〜7のいずれか1項に記載の組成物。
- 前記ナノ分散物は、乳化剤としての前記リン脂質および水を、任意選択でカバノキ樹皮抽出物とともに、医薬として許容可能な油と混合することと、次に、ナノ分散物を得るために、混合物の高せん断乳化もしくは超音波乳化または高圧均質化を実施することとによって調製され、それにより、粒子液滴の少なくとも95%が1μm未満の直径を有することを特徴とする、請求項1〜8のいずれか1項に記載の組成物。
- 前記組成物は、医薬として許容可能なポリマー担体を組み込まれた電界紡糸繊維内に含有されることを特徴とする、請求項1〜9のいずれか1項に記載の組成物。
- ポリマーは、ポリビニルアルコール、ポリ(グリコリド)、ポリラクチド−ポリグリコリド−コポリマー、ポリウレタン、ポリビニルアルコール/酢酸セルロース、絹フィブロイン、絹/キトサン、ゼラチン、コラーゲン/キトサンを含む群から選択されることを特徴とする、請求項10に記載の組成物。
- 請求項10または11に記載の電界紡糸繊維を含有する組成物であって、電界紡糸ナノ繊維は、前記電界紡糸繊維の総重量に基づいて計算された、1.0〜60重量%の請求項1〜9のいずれか1項に記載のナノ分散物を含有することを特徴とする、組成物。
- 電界紡糸繊維内に含有されるナノ分散物の製造方法であって、前記方法は、
a)リン脂質、油および水、ならびに任意選択でカバノキ樹皮抽出物の、ナノ分散物を調製することと、
b)工程a)において得られた前記ナノ分散物をPVAポリマー溶液と配合することと、
c)その混合物を電界紡糸してフリースを形成することとを含む、方法。 - 創傷の処置に使用するための請求項1〜12のいずれか1項に記載の組成物。
- 創傷の処置に使用するための噴霧剤またはフォームの形態にある請求項1〜12のいずれか1項に記載の組成物。
