Abstract
Description
1.(A)又は(B)に従って糖タンパク質組成物の非フコシル化レベルを制御する方法:
(A)糖タンパク質組成物の非フコシル化レベルを、前記糖タンパク質組成物の基準非フコシル化レベルと比較して、上昇させる方法であって、
糖タンパク質を発現する真核細胞を、非フコシル化レベルが基準非フコシル化レベルである糖タンパク質を発現する前記細胞を培養するために使用されるpH及び/又は温度よりも低い温度及び/又はpHで、培養することを含む方法;又は
(B)糖タンパク質組成物の非フコシル化レベルを、前記糖タンパク質組成物の基準非フコシル化レベルと比較して、低下させるための方法であって、
糖タンパク質を発現する真核細胞を、非フコシル化レベルが基準非フコシル化レベルである糖タンパク質を発現する前記細胞を培養するために使用されるpH及び/又は温度よりも高い温度及び/又はpHで、培養することを含む方法。
2.非フコシル化レベルが基準非フコシル化レベルである糖タンパク質を発現する前記細胞を培養するために使用される温度よりも、温度のみが(A)低いか又は(B)高く、そしてpHが同じである、項目1に記載の方法。

5.真核細胞が哺乳動物細胞である、項目1〜4のいずれか1項に記載の方法。
6.哺乳動物細胞がCHO細胞である、項目5に記載の方法。
7.糖タンパク質が抗体又は抗体断片である、項目1〜6のいずれか1項に記載の方法。
8.pH及び/又は温度の変化が産生段階に限定される、項目1〜7のいずれか1項に記載の方法。
9.項目1〜8のいずれか1項に記載の方法により得られる糖タンパク質組成物。
10.項目9に記載の糖タンパク質組成物及び使用説明書を含むキット。
(a)前記糖タンパク質を発現する細胞を初期温度及び/又はpHで培養することにより得られる糖タンパク質組成物の非フコシル化レベルを、所望の非フコシル化レベルと比較する工程;
(b)前記糖タンパク質を発現する細胞を初期温度及び/又はpHで培養することにより得られる非フコシル化レベルが、所望の非フコシル化レベルより低いか又は高いかを判定する工程;並びに
(c)(i)前記糖タンパク質を発現する細胞を初期温度及び/又はpHで培養することにより得られる非フコシル化レベルが、所望の非フコシル化レベルより低い場合、前記糖タンパク質を発現する細胞を初期温度及び/又はpHよりも低い温度及び/又はpHで培養する工程;あるいは
(ii)前記糖タンパク質を発現する細胞を初期温度及び/又はpHで培養することにより得られる非フコシル化レベルが、所望の非フコシル化レベルより高い場合、前記糖タンパク質を発現する細胞を初期温度及び/又はpHよりも高い温度及び/又はpHで培養する工程。
アダリムマブを発現するCHO細胞は、流加培養で維持された。細胞は、温度とpHを除いて同じ条件下で培養され、温度とpHは次のように変化させた。

Claims (11)
- (A)又は(B)に従って糖タンパク質組成物の非フコシル化レベルを制御する方法:
(A)糖タンパク質組成物の非フコシル化レベルを、前記糖タンパク質組成物の基準非フコシル化レベルと比較して、上昇させる方法であって、
糖タンパク質を発現する真核細胞を、非フコシル化レベルが基準非フコシル化レベルである糖タンパク質を発現する前記細胞を培養するために使用されるpH及び/又は温度よりも低い温度及び/又はpHで、培養することを含む方法;又は
(B)糖タンパク質組成物の非フコシル化レベルを、前記糖タンパク質組成物の基準非フコシル化レベルと比較して、低下させるための方法であって、
糖タンパク質を発現する真核細胞を、非フコシル化レベルが基準非フコシル化レベルである糖タンパク質を発現する前記細胞を培養するために使用されるpH及び/又は温度よりも高い温度及び/又はpHで、培養することを含む方法。 - 非フコシル化レベルが基準非フコシル化レベルである糖タンパク質を発現する前記細胞を培養するために使用される温度と比較して、温度のみを変化させ、pHは同じまま維持される、請求項1に記載の方法。
- 非フコシル化レベルが基準非フコシル化レベルである糖タンパク質を発現する前記細胞を培養するために使用されるpHと比較して、pHのみを変化させ、温度は同じまま維持される、請求項1に記載の方法。
- 非フコシル化レベルが基準非フコシル化レベルである糖タンパク質を発現する前記細胞を培養するために使用されるpH及び温度と比較して、温度及びpHの両方を変化させる、請求項1に記載の方法
- 前記真核細胞が哺乳動物細胞である、請求項1〜4のいずれか1項に記載の方法。
- 前記哺乳動物細胞がCHO細胞である、請求項5に記載の方法。
- 前記糖タンパク質が抗体又は抗体断片である、請求項1〜6のいずれか1項に記載の方法。
- 前記pH及び/又は温度の変化が産生段階に限定される、請求項1〜7のいずれか1項に記載の方法。
- 請求項1〜8のいずれか1項に記載の方法により得られる糖タンパク質組成物。
- 請求項9に記載の糖タンパク質組成物及び使用説明書を含むキット。
- 真核細胞を培養している間、温度及び/又はpHを変化させることを含む、糖タンパク質組成物の非フコシル化のレベルを制御するための方法。
