Abstract
Description
本出願は、2018年5月10日出願の米国特許仮出願第62/762,549号の利益を主張し、この出願はその全体が参照により本明細書に組み込まれる。

本開示に表される手法は、1つのカップリング部位のみが、多数の活性剤分子を抗体またはアプタマー分子に結合するために必要であるという利点を有する。部位特異的カップリング方法、例えば、酵素カップリング反応を抗体の重鎖のC末端にあるペプチドタグに対して使用することによって、活性剤含有ポリマーは抗体結合界面からはるかに離れて配置される。この手法によって、標的組織の最大親和性が保存され、さらには相対的に均質な産物も得る。選択された連結戦略は、コポリマーと抗体/アプタマーとの間に安定したペプチド結合を形成し、このことは、血流中のADCの高い安定性を確実にする。さらに、最終ステップにおける抗体/アプタマーへの完全に機能化され特徴付けられた活性剤含有コポリマーのカップリングは、感受性の高い結合タンパク質への立体構造応力(conformational stress)を最小限にすることを目的としている。加えて、選択された設計のコポリマーは、同じ分子への2個またはそれより多くの異なる活性剤のカップリングを促進し、併用療法を可能にする。活性剤(がんの文脈において細胞傷害性薬物または毒性ペイロードとも呼ばれる)が標的細胞、例えば腫瘍細胞内に放出され、標的化部分(例えば、抗体またはアプタマー)が分解されると、相対的に小さなコポリマーは腎クリアランスによって身体から除去されると考えられる。
本開示の医薬組成物は、有効量の本開示の活性部分を、1つまたは複数の薬学的に許容される担体または賦形剤と一緒に処方して含む。
銅複合体を介した(S)−6−アクリルアミド−2−アミノヘキサン酸モノマーの合成
L−リジン(14.62g、100mmol)を150mLの脱イオン水に溶解し、約80℃に加熱した。炭酸銅(16.6g、75mmol)を30分間かけて少量ずつ加えた。反応物をさらに30分間撹拌した。高温で深青色の懸濁液をシリカゲルでろ過した。フィルターを少量の水で洗浄した。翌日、リジン銅複合体を含有する合わせたろ液を氷浴で冷却し、100mLのテトラヒドロフラン(THF)を加えた。メチル−tert−ブチルエーテル(TBME)中の塩化アクリロイルの溶液(8.9mL、110mmol)を、1時間にわたって滴下により添加した。pHを10%水酸化ナトリウム溶液の並行滴下により添加により最初に8〜10に維持した。塩化アクリロイル溶液の半分を加えると、生成物が沈殿し始めた。大部分の塩化アクリロイルが添加されたときに、水酸化ナトリウムの添加を遅くして、pHを約6に下げ、反応混合物の温度を室温にした。青色懸濁液をさらに2時間撹拌し、次いでろ過した。フィルターに保持された固体物質を水およびアセトンで洗浄し、次いで乾燥した。収量が6.5gのアクリロイル−L−リジン銅複合体を得た。
(2S)−3−(アクリロイルオキシ)−2−アミノプロパン酸の合成
水(50mL)中のL−セリン(5g、47.6mmol)の溶液を80℃に加熱し、固体炭酸銅(5.79g、26.2mmol)を加えた。溶液を10分間撹拌した。続いて不溶解残留物をろ過により収集し、水(30mL)で洗浄した。合わせたろ液を氷浴で冷却し、KOH(27.1mL、47.6mmol)をゆっくりと加えた。この溶液に、アセトン(30mL)中の塩化アクリロイル(4.52mL、59.5mmol)の混合物を滴下により添加した。次いで反応混合物を撹拌下、4℃で一晩インキュベートした。形成された固体を単離し、水(50mL)/メタノール(50mL)/エチル−tert−ブチルエーテル(50mL)(MTBE)で洗浄し、最後に減圧下で乾燥して、O−アクリロイル−L−セリン−Cu2+複合体(3.8g、10.01mmol、収率42.1%)を得た。続いて、複合体中の銅を実施例1に記載された手順と類似した手順により除去した。収量が1.43g(45%)のアクリロイル−L−セリンを白色の粉末として得た。この化合物の同一性をNMRおよびLC−MS分光法により検証した。
(2S)−3−(アクリロイルオキシ)−2−アミノブタン酸の合成
6mLのトリフルオロ酢酸(TFA)を含む反応容器を、氷浴で冷却した。続いて、固体L−トレオニン(2.00g、16.79mmol)を加え、混合物を5分間撹拌した。トリフルオロメタンスルホン酸(0.18mL、2.0mmol)、続いて塩化アクリロイル(2.5mL、32.9mmol)を加え、反応混合物を室温で2時間インキュベートした。反応が完了した後、生成物をメチル−tert−ブチルエーテル(MTBE)で沈殿させた。固体を単離した後、生成物をMTBEおよびアセトンで洗浄した。最後にO−アクリロイル−L−トレオニン塩酸塩を減圧下で乾燥して、白色の粉末(収率32%)を得た。この化合物の構造をNMRおよびLC−MS分光法により検証した。
(S)−3−(4−(アクリロイルオキシ)フェニル)−2−アミノプロパン酸の合成
O−アクリロイル−L−チロシン−Cu2+複合体の合成は、実施例1に記載された手順に従って実施した。銅を以下の手順により複合体から除去した。73.15g(140mmol)のO−アクリロイル−L−チロシン−Cu2+複合体を破砕皿中の220mLの2N HClに溶解した。混合物を、Polytron(登録商標)PT3000機器を使用して均質化した。続いて混合物をろ過し、残留物を50mLの2N HClで2回洗浄した。次いで固体化合物をNaOHにより減圧下、40℃で乾燥して、O−アクリロイル−L−チロシン塩酸塩(46.96g、収率63%)を得た。
(S)−2−(4−アクリルアミドフェニル)−2−アミノ酢酸の合成
Boc−4−アミノ−L−フェニルアラニン(2.50g、8.9mmol、Anaspec、Fremont、CA)を25mLのクロロホルムに溶解した。トリエチルアミン(2.47mL、17.8mmol)をこの溶液に加え、混合物を−15℃に冷却した。続いて、クロロホルム中の塩化アクリロイル(0.79mL、9.8mmol)を、撹拌しながら混合物に滴下により添加した。塩化アクリロイルの添加が完了した後、反応混合物をさらに3時間撹拌した。その後、反応混合物をガラスフィルターに通し、保護(S)−2−(4−アクリルアミドフェニル)−2−アミノ酢酸をカラムクロマトグラフィーにより精製し、残留溶媒を蒸発させた。得られた(S)−2−(4−アクリルアミドフェニル)−2−((tert−ブトキシカルボニル)アミノ)酢酸(500mg、1.5mmol)を5mLのジクロロメタン(DCM)に溶解した。トリフルオロ酢酸(TFA)(800μL、10.38mmol)を加え、溶液を室温で1時間撹拌した。その後、溶媒を減圧下で除去し、5mLのDCMを加え、溶媒を再び減圧下で除去した。この手順を数回繰り返した。最後に生成物を3mLのDCMに溶解し、メチル−tert−ブチルエーテル(MTBE)で沈殿させた。固体をガラスフィルターで収集し、真空下で乾燥して、純粋なアクリロイル−4−アミノ−L−フェニルアラニンを15%の収率で得た。この化合物の構造をNMRにより検証した。
(2S)−4−(アクリロイルオキシ)ピロリジン−2−カルボン酸および(R)−3−(アクリロイルチオ)−2−アミノプロパン酸の合成
これらの化合物の合成を実施例1に記載されたように実施した。(2S)−4−(アクリロイルオキシ)ピロリジン−2−カルボン酸および(R)−3−(アクリロイルチオ)−2−アミノプロパン酸では、出発材料は、それぞれ4−ヒドロキシ−L−プロリンおよびL−システインであった。
(S)−6−アクリルアミド−2−(3−(4−ヒドロキシフェニル)プロパンアミド)ヘキサン酸(AK−フェノール)の合成
DMF(9mL)中のAK(538mg、2.69mmol)およびTEA(443μL、3.18mmol)の溶液に、ボルトンハンター試薬(643mg、2.44mmol)を加えた。反応混合物を室温で一晩撹拌した。次いで反応混合物をろ過し、揮発物をN2ストリーム下で除去した。残留物をSiO2カラムクロマトグラフィーにより精製した。構造をNMR分光法により検証した。
アミノ酸のメタクリル/エチルアクリル/プロピルアクリル誘導体の合成
メタクリル/エチルアクリル/プロピルアクリル誘導体の合成を、対応する酸塩化物、例えば塩化メタクリロイルを実施例1〜7に記載された条件下で使用して実施した。
フルオレセイン修飾共主要モノマーAK−フルオレセイン−V1の合成
20mLの反応容器中で、アクリロイル−L−リジン[実施例1を参照すること](150mg、0.749mmol)、フルオレセインイソチオシアネート(FITC、321mg、0.824mmol)およびトリエチルアミン(0.114mL、0.824mmol)の溶液をDMFで調製した。反応物を、一定撹拌下、暗所において室温で一晩インキュベートした。続いて、溶液を0.4μmのフィルターでろ過して、潜在的な粒子を除去した。その後、残留溶媒を、ロータリーエバポレーターにより真空を適用しながら30℃で除去した。構造を、NMRおよびLC−MSにより検証した(収率98%、純度>95%)。その後、合成されたモノマーを、溶媒としてDMFおよび開始剤としてAIBNを使用するジメチルアクリルアミド(DMA)[90/10mol/mol]との共重合で試験した。重合反応を65℃で6時間実施し、得られたコポリマーを、実施例13に提示されているプロトコールを使用してゲル浸透クロマトグラフィー(GPC)により分析した。
フルオレセイン修飾共主要モノマーAK−フルオレセイン−V2の合成
反応は、実施例9に提示された合成プロトコールに従って出発材料としてフルオレセイン−NHSを用いて実施したが、10mol%過剰のAKを用い、これを反応後に沈殿により除去した。
非切断性リンカーを有するドキソルビシン(DOX)修飾共主要モノマー(AK−DOX−V1)の合成
DMF中のDOX・HCl(200mg、345μmol、1.00当量)およびEt3N(50μL、348μmol、1.01当量)の溶液に、無水コハク酸(36.2mg、362μmol、1.05当量)を加えた。混合物を不活性雰囲気下、室温で30分間撹拌し、次いでNHS(43.7mg、379μmol、1.10当量)、続いてEDC・HCl(69.4mg、362μmol、1.05当量)を加えた。得られた混合物を室温で一晩撹拌し、次いでAK(69.0mg、345μmol、1.00当量)、続いてEt3N(53μL、379μmol、1.10当量)を加えた。反応混合物を再び室温で一晩撹拌した。揮発物をN2ストリーム下で蒸発させ、残留物をSiO2カラムクロマトグラフィーにより精製して、所望の生成物(228mg、276μmol、80%)を得た。
カテプシンB感受性リンカーを有するドキソルビシン修飾共主要モノマー(AK−DOX−V2)の合成
N−メチル−2−ピロリドン(NMP)中のDOX・HCl(86mg、149μmol、1.10当量)、MC−Val−Cit−PABO−PNP(100mg、136μmol、1.00当量)およびN,N−ジイソプロピルエチルアミン(DIPEA)(26μL、149μmol、1.10当量)の溶液を室温で2時間撹拌する。得られた混合物に、AK(28.5mg、142μmol、1.05当量)、続いてDIPEA(26μL、149μmol、1.10当量)を加える。反応混合物を室温で一晩撹拌する。揮発物をN2ストリーム下で蒸発させ、残留物をSiO2でのクロマトグラフィーにより精製して、所望の生成物(109mg、81μmol、60%)を得る。
BOC−Gn−Cellophilの合成の一般手順
ステップ1:RAFT−NHS中間体の合成:
CH2Cl2中の、Tucker et al. (ACS Macro Letters (2017) 6(4): 452-457)に記載されたように合成された2−[[(エチルチオ)チオキソメチル]チオ]−2−メチル−プロパン酸(22.85g、102mmol、1.0当量)、および1−ヒドロキシピロリジニン−2,5−ジオン(1-hydroxypyrrolidinine-2,5-dione)(12.89g、112mmol、1.1当量)の溶液に、EDC・HCl(21.48g、112mmol、1.1当量)を0℃で加えた。反応混合物を室温で16時間撹拌した。次いで反応混合物をN2流下で部分的に(総体積の約半分まで)蒸発させ、AcOEtおよび再蒸留水(ddH2O)で希釈した。二相溶液を分液漏斗に移し、抽出した後、有機相をddH2O、NaHCO3飽和水溶液(3×)、ddH2O(2×)およびブラインで連続的に洗浄した。有機相を乾燥し(Na2SO4)、すべての揮発物を減圧下で除去した。残留物をn−ヘキサンで摩砕し、得られた黄色懸濁液をろ過した。ケーキをn−ヘキサンで洗浄した。黄色の固体を減圧下で乾燥し、得られた中間体(RAFT−NHS)をさらに精製することなく使用した(31.8g、99.0mmol、97%)。すべての分析データは、文献の値と一致していた。Yang et al. (2012) Macromolecular rapid communications 33(22): 1921-6。
CH2Cl2中のRAFT−NHS出発材料(1.22g、3.61mmol、1.0当量)の溶液に、CH2Cl2中のt−ブチル−(2−アミノエチル)カルバメート(0.81g、5.0mmol、1.4当量)およびEt3N(1.0mL、7.2mmol、2.0当量)の溶液を、−10℃で滴下により添加した。反応混合物を室温で12時間撹拌した。有機混合物を、NH4Clの飽和水溶液(2×)、NaHCO3の飽和水溶液(2×)およびブラインで連続的に洗浄した。有機相を乾燥し(Na2SO4)、すべての揮発物を減圧下で除去した。残留物をn−ヘプタンおよびEt2Oの混合物で再結晶させた。黄色の結晶をろ過し、n−ヘプタンで洗浄し、減圧下で乾燥して、次の中間体(RAFT−EDA−BOC、1.26g、3.44mmol、95%)を得た。得られた化合物の構造をMSおよびNMR分光法により検証した。
TFA中のRAFT−EDA−BOC(1.25g、3.41mmol、1.0当量)の冷溶液を60分間撹拌した。次いで反応混合物をMeOHおよびCH2Cl2(1/2)で希釈し、揮発物を部分的に(総体積の2/3を)N2流下で除去した。得られたRAFT−EDA−OTfを黄色の油状物(2.00g、3.29mmol、96%)として単離し、さらに精製することなく次のステップで使用した。得られた化合物の構造をMSおよびNMR分光法により検証した。
CH2Cl2中のBOC−G3(697mg、2.41mmol、1.0当量)、1−ヒドロキシベンゾトリアゾール水和物(HOBt水和物)(92.0mg、600μmol、0.25当量)およびEDC・HCl(485mg、2.53mmol、1.05当量)の溶液を、不活性雰囲気(N2)下、0℃で30分間撹拌した。この溶液に、CH2Cl2中のRAFT−EDA−OTf(917mg、2.41mmol、1.0当量)の溶液およびDIPEA(2.13mL、12.5mmol、5.2当量)を連続的に滴下により添加した。反応混合物を0℃で1時間、次いで室温で一晩撹拌した。反応混合物をCH2Cl2で希釈し、有機混合物を、NH4Cl飽和溶液(3×)、NaHCO3飽和溶液、ddH2Oおよびブラインで連続的に洗浄した。有機相を収集し、乾燥し(Na2SO4)、揮発物を減圧下で部分的に(総体積の2/3を)除去した。得られた溶液にEtOAcを加えた。次いで、得られた曇った溶液を冷蔵庫に一晩保管して、黄色の懸濁液を得て、それをろ過し、ケーキを冷EtOAcで洗浄した。黄色の固体を減圧下で乾燥して、BOC−G3−RAFT剤(396mg、736μmol、31%)を得た。得られた化合物の構造をMSおよびNMR分光法により検証した。
ジオキサン中のDMA(192μL、1.86mmol、10当量)の溶液に、BOC−G3−RAFT剤(100mg、186μmol、1.0当量)およびAIBN(6.1mg、37μmol、0.20当量)を連続的に加えた。反応混合物を60℃で6時間撹拌した。次いで反応生成物をn−ヘキサンに沈殿させた。薄黄色の懸濁液をろ過し、得られたケーキをn−ヘキサンで洗浄し、最後にアセトンに溶解した。次いで揮発物を減圧下で除去して、BOC−G3−DMA−RAFTプレポリマーを黄色の油状物(280mg、186μmol、99%)として得た。得られた化合物の構造をMSおよびNMR分光法により検証した。
ジオキサン中のBOC−G3−DMA−RAFTプレポリマーの溶液を、ジオキサン中のHCl(4M)の溶液で2時間処理する。揮発物をN2流下、室温で除去する。残留物を、さらに精製することなく使用する。得られた化合物の構造をMSおよびNMR分光法により検証する。
ddH2O中のDMA(1.23mL、11.9mmol、70当量)およびAK(272mg、1.36mmol、8当量)の溶液に、BOC−G3−DMAプレポリマー(221mg、170μmol、1.0当量)およびVA044(27.5mg、85μmol、0.4当量)を連続的に加えた。反応混合物を55℃で4時間撹拌した。反応混合物をddH2Oおよびジオキサンで希釈した。この溶液に、ホスフィン酸(50w%、93μL、850μmol、5当量)、TEA(118μL、850μmol、5当量)およびVA044(27.5mg、85μmol、0.5当量)を連続的に加えた。反応混合物を100℃で4時間撹拌した。次いで、得られた混合物をddH2Oで透析し(MWCO 3.5kDa)、濃縮液をフリーズドライして(freeze-dried)、BOC−G3−Cellophilを白色の粉末(1.20g、120μmol、2ステップで71%)として得た。得られた化合物の構造を、NMR分光法および以下のプロトコールを使用するGPCにより検証した。3.33mg/mLのコポリマーの貯蔵溶液を、溶出緩衝液(0.05%(w/v)NaN3を含有する脱イオン水)で調製し、0.45μmシリンジフィルターでろ過した。続いて、0.4mLの貯蔵溶液をGPC装置(1260 Infinity LC−System、Agilent、Santa Clara、CA)のポートに注入した。クロマトグラフィーを、溶出緩衝剤により0.5mL/分の一定流速で実施した。コポリマーの試料を、55℃の外部カラムオーブンに設置されたSupurema3カラムシステム(プレカラム、1000Å、30Å;粒径5μm;PSS、Mainz、Germany)で分離した。コポリマーをRI(屈折率)およびUV検出器により分析した。較正曲線(10ポイント)を、プルラン標準を使用して確立した。特徴付けされたコポリマーの分子量を、この標準を参照することにより推定した。
BOC−Gn−PEGx−Cellophilの合成
BOC−G3−PEG11−Cellophilの合成は、上記の一般手順(実施例13:ステップ1〜4および7)に従って達成した。ステップ2は、出発材料としてBOC−PEG11−CH2CH2−NH2を使用して実施して、RAFT−PEG11−BOCを生じ、ステップ3はEtOAc中のHClを使用して実施した。
BOC−Gn−Cellophil−(フルオレセイン)8またはBOC−Gn−PEGx−Cellophil−(フルオレセイン)8の合成
NaHCO3(0.1N)の水溶液中のBOC−G3−Cellophil(1.0当量)またはBOC−G3−PEG11−Cellophil(1.0当量)(調製には、実施例13、ステップ7を参照すること)の溶液に、DMSO中のFITC(16当量)の溶液をゆっくりと加えた。得られた反応混合物を室温で16時間撹拌し、NaHCO3の水溶液(0.1N)、次いでddH2Oで透析した(MWCO 3.5kDa)。濃縮液をフリーズドライして、暗橙色の粉末(収率:80〜92%)を得た。この化合物の構造をNMRおよびGPCにより検証した。
トリグリシンおよびトリグリシン−(PEG)23誘導体の合成を、同じプロトコールを使用して達成した。
H2N−Gn−Cellophil−(フルオレセイン)8またはH2N−Gn−PEGx−Cellophil−(フルオレセイン)8の合成
EtOH中のBOC−G3−Cellophil−(フルオレセイン)8(1.0当量)またはBOC−G3−PEG11−Cellophil−(フルオレセイン)8(1.0当量)の溶液に、EtOH中のHCl(約1.25N)の溶液を加えた。得られた反応混合物を室温で2時間撹拌し、水(2×)で透析した(MWCO 3.5kDa)。濃縮液をフリーズドライして、暗橙色の(静電)粉末(99%)を得た。この化合物の構造をNMRおよびGPCにより検証した。
カテプシンB感受性ドキソルビシン含有Cellophilの合成
ステップ1 H−Lys(Alloc)−OHの調製
H2O(100mL)中のL−リジン塩酸塩(1.0当量)および塩基性炭酸銅(II)(1.1当量)の溶液を30分間還流させた。還流の際に形成された固体を熱時ろ過により除去した。ろ液を0℃に冷却し、固体重炭酸ナトリウム(3.1当量)の添加によりpH9に調整した。クロロギ酸アリル(1.5当量)を1時間かけて滴下により添加し、その間、溶液を0℃で撹拌した。この添加の際に、反応混合物を固体重炭酸ナトリウムの添加によりpH9に維持した。反応混合物を室温に温め、一晩撹拌した。反応の際に形成された青色の固体生成物を、ろ過により定量収率で収集した。
Lys(Alloc)の固体銅塩をH2O(250mL)に懸濁し、2当量のチオアセトアミド(2.0当量)を加えた。アルカリ性懸濁液を50℃で3時間撹拌し、その間に固体がゆっくりと溶解した。続いて、溶液を2M HClでpH2に酸性化し、5分間沸騰させた。沈殿したCuSをろ過により除去した。ろ液を真空下で約60mLに濃縮し、その時点でLys(Alloc)の塩酸塩が白色の固体(79%)として沈殿し、それをろ過により回収した。
ジオキサン(30mL)中のFMOC−Val−OSu(1.0当量)の室温で激しく撹拌した溶液を、水(30mL)中のLys(Alloc)−OH(1.1当量)およびNaHCO3(2.1当量)の溶液と合わせた。温度を、最初の30分間に冷水浴を使用して25℃未満に維持した。混合物を室温で14時間撹拌し続けた。混合物を水(50mL)で希釈し、次いで15%クエン酸でpH3に酸性化した。得られた懸濁液を酢酸エチル(3×100mL)で抽出し、合わせた有機層を水およびブラインで洗浄し、乾燥し、蒸発させて、オフホワイトの固体を得た。固体をTHFに溶解し、次いでメチルtert−ブチルエーテル(MTBE)を加え、ろ過した後に純白な固体(80%)を得た。
THF(15mL)中のFMOC−Val−Lys(Alloc)−OH(1.0当量)およびPABOH(1.1当量)の室温で撹拌した溶液を、EEDQ(1.1当量)で処理した。16時間後、混合物を30℃で蒸発乾固し、残留物をMTBEで再結晶させて、濃黄色(deep yellow)の生成物(84%)を得た。
CH2Cl2(35ml)中のFMOC−Val−Lys(Alloc)−PABOH(1.0当量)を室温でジエチルアミン(50ml)により処理した。混合物を短時間超音波処理し、室温で4時間撹拌した。溶媒を蒸発させ、残留物をCH2Cl2でフラッシュし、シリカのクロマトグラフィーにかけて、生成物を無色の泡状物(69%)として得た。
CH2Cl2(5ml)中のH−Val−Lys(Alloc)−PABOH(1.1当量)およびDIEA(1.1当量)を室温でCH2Cl2(2ml)中のMC−NHS(1.1当量)により処理した。混合物を室温で一晩撹拌した。酢酸エチル(60ml)を加え、混合物を水およびブラインで洗浄し、乾燥し、蒸発させて、所望の生成物(96%)を得た。
THF(15mL)中のMC−Val−Lys(Alloc)−PABOH(1.0当量)の室温で撹拌した溶液を、PNPクロロホルメート(1.2当量)および乾燥ピリジン(1.5当量)で処理した。反応物を、HPLC分析が混合物に抽出物が存在しないことを示すまで、一晩撹拌した。混合物をEtOAc(50mL)で希釈し、クエン酸(50mL、H2O中10%)で酸性化した。有機相を水(50mL)およびブライン(25mL)で洗浄し、硫酸ナトリウムで乾燥し、溶媒を蒸発させて、薄黄色の固体を得た。固体をシリカゲルでのフラッシュクロマトグラフィー(20:1のDCM/MeOH)により精製して、純粋な化合物を、黄色を帯びた固体(58%)として得た。
NMP(8mL)中のMC−Val−Lys(Alloc)−PABO−PNP(1.0当量)およびDOX・HCl(1.1当量)を室温でEt3N(1当量)により処理した。その後、混合物を暗所で3日間インキュベートした。次いで、混合物を10%の2−プロパノール/酢酸エチル(100mL)で希釈し、水(3×100mL)およびブライン(50mL)で洗浄し、乾燥し、蒸発させて、濃橙色(deep orange)の油状物を得た。これをシリカゲルでのフラッシュクロマトグラフィーにより精製して、純粋な生成物を赤色固体(92%)として得た。
THF(7mL)中のMC−Val−Lys(Alloc)−PABC−DOX(1.0当量)を、アルゴン下、室温で、Pd(PPh3)4(0.03当量)、酢酸(2.5当量)および水素化トリブチルスズ(1.5当量)で処理した。混合物を暗所において室温で1時間撹拌し、その最中に橙色の固体が形成し始めた。混合物をエーテル(25mL)で希釈し、続いてエーテル中の1M HCl(2mL)を添加した。得られた懸濁液を短時間超音波処理し、次いでろ過した。橙色の固体をエーテルで繰り返し洗浄し、次いで5:1のDCM:MeOHに溶解した。これに、Celite(7g)を加え、次いで溶媒を蒸発させた。得られた固体をCelite(登録商標)に吸着させ、Celiteカラム(DCM:MeOHの100:1のスラリーから作製)にドライロードした(dry-loaded)。カラムを、DCM:MeOHの100:1の混合物、続いてDCM:MeOHの10:1の混合物で溶出した。所望の生成物を橙色の固体(30%)として得た。
異なるアミノ酸配列を有するリンカー誘導体MC−Ala−Lys−PABC−DOX、MC−Val−Cit−PABC−DOXを、シトルリンリンカーがAlloc保護基を必要としないこと以外は上記に記述された手順と同じ手順に従って合成した。
続いて、上記に記述されたリンカードキソルビシンコンジュゲートを、以下の一般手順を使用してCellophilコポリマー(実施例13、ステップ7)にカップリングした。
DMF中のCellophilコポリマー(1.0当量)およびEt3N(12当量)の溶液に、リンカードキソルビシンコンジュゲート(10当量)の溶液を不活性雰囲気下、室温で滴下により添加した。反応混合物を室温で一晩撹拌した。最後に混合物をddH2Oで希釈し、得られた懸濁液をろ過した。次いで、ろ液をddH2O(2×10L)で透析し(MWCO 3.5kDa)、濃縮液を凍結乾燥して、赤色の易流動性粉末(収率60〜70%)を得た。得られた化合物の構造をNMR分光法により検証した。
CellophilコポリマーからのドキソルビシンのカテプシンB媒介放出。
ヒト肝臓カテプシンB(Merck、MW約27500)(5単位)を400μLの酢酸塩緩衝液(50mMの酢酸塩+1mMのEDTA、pH5.0)に溶解した。10μLの酵素溶液を、390μLの活性化溶液(5mMのジチオトレイトール、100mMのリン酸ナトリウム緩衝剤、5mMのEDTA、100mMのNaCl、0.01%のBrij58、pH6.0)と共に37℃で約30分間インキュベートした。その間、20μLの実施例17のポリマーリンカーDOXコンジュゲート(1μmol)を、1473μLの活性化溶液に加え、37℃でインキュベートした。32μLの活性化酵素(0.01U)を基質溶液に加え、反応物を37℃でインキュベートした。遊離DOXの放出を、HPLCおよび光度計測定によりモニターした。MC−Val−Lys−PABC−PNP−DOXが最短の半減期を呈し、次にMC−Ala−Lys−PABC−DOXおよびMC−Val−Cit−PABC−DOXであった。
ドキソルビシン含有共主要モノマー(AK−DOX−V1およびAK−DOX−V2)の共重合。
ドキソルビシンを含むCellophilコポリマーの合成は、RAFT剤としてBOC−Gn−RAFT中間体、BOC−Gn−DMA−RAFTプレポリマー、RAFT−PEGX−BOC、または対応するBOC脱保護試薬(実施例13、ステップ6に記載されたプロトコールに従って得た)(1.0当量)およびドキソルビシン含有共主要モノマー(8.0当量)(AK−DOX−V1またはAK−DOX−V2)を使用して、共主要モノマーの共重合についての一般手順(実施例13、ステップ7)により達成される。
AK−フェノールの共重合
ヨウ素反応性モノマーを含むCellophilコポリマーの合成は、RAFT試薬としてBOC−G3−RAFT中間体、BOC−G3−DMA−RAFTプレポリマー、RAFT−PEG11−BOC、または対応するBOC脱保護試薬(実施例13、ステップ6に記載されたプロトコールに従って得た)(1.0当量)および共主要モノマーとしてAK−フェノール(8.0当量)を使用して、共主要モノマーの共重合についての一般手順(実施例13、ステップ7)によって達成した。ddH2Oの代わりに、0.1MのNaHCO3を溶媒として使用した。所望の生成物の構造をNMR分光法により検証した。
ヨウ素反応性ポリマー担体のヨウ素化
PBS緩衝液(pH=7.4)中の実施例20のAK−フェノールコポリマー(70mg、7.0μmol)の溶液に、NaI(7.86mg、52μmol)およびクロラミンT(14.8mg、52μmol)を連続的に加えた。次いで反応混合物を室温で30分間撹拌し、その後、Na2S2O5水溶液(0.3M)で希釈した。次いで、得られた溶液を10LのddH2Oで透析し、濃縮液をフリーズドライした。所望の生成物の構造をNMR分光法により検証した。
ソルターゼ媒介反応を使用するHer2+モデル抗体へのH2N−G5−Cellophil−(フルオレセイン)8のカップリング
H2N−G5−Cellophil−(フルオレセイン)8(実施例16)を、以下の一般手順を使用して、Her2抗原に対する完全ヒトモノクローナル抗体にコンジュゲートすることができる。重鎖のC末端でソルターゼモチーフ(LPETG)およびヘキサヒスチジンタグ(His6)により遺伝子修飾されたHer2モノクローナル抗体[10μM]を、H2N−G5−Cellophil−(フルオレセイン)8[100μM]と共に、0.62μMのソルターゼAの存在下、50mMのHepes、150mMのNaCl、5mMのCaCl2、pH7.5中において、25℃で3.5時間インキュベートする。反応は、25mMリン酸ナトリウム(pH7.5)で平衡にしたProtein A HiTrapカラム(GE Healthcare)に通過させることによって停止させる。充填したカラムを5カラム体積(CV)の緩衝剤で洗浄する。結合したコンジュゲートを5CVの溶出緩衝液(0.1Mのクエン酸、pH2.8)で溶出し、1CVの画分を、25%(v/v)の1M Tris塩基を含む管に収集して、溶液を中和する。タンパク質含有画分をプールし、続いて製造会社の使用説明書に従ってNAP−25カラム(GE Healthcare)を使用して、緩衝剤交換により10mMのコハク酸ナトリウムpH5.0、100mg/mLのトレハロース、0.1%(w/v)のポリソルベート20に配合する。カップリング反応の成功は、4〜20%勾配のトリスグリシンゲルでのSDS−PAGE、ならびに抗His6一次抗体およびホースラディッシュペルオキシダーゼコンジュゲート二次抗体を使用するウエスタンブロット(WB)分析により定量的に検証する。WBシグナルの検出は、増強化学発光(ECL)キット(Pierce(商標)、ECL Western Blotting Substrate)を使用して実施する。非修飾抗Her2−LPETG−His6抗体は対照としての機能を果たす。抗His6抗体の消滅は、ソルターゼ反応が完了したことを示す。定量的分析では、サイズ排除クロマトグラフィーを実施する。薬物の抗体に対する比(DAR)は、残存非修飾抗体(UV検出波長280nm)のピーク強度の比較により計算する。
H2N−G3−Cellophilコポリマーの毒性探索
本開示のポリマー担体は生物分解性ではない。したがって、担体が健康な組織に対して無害であることを実証することが重要であった。本開示のポリマー担体(ペイロードなし)の毒性探索研究を実施した。簡潔には、HepG2細胞を、96ウェル黒色壁透明底ポリスチレンプレートに1ウェルあたり100μLで平板培養した。試験化合物は、DMAおよびAK(90/10mol%)を含むH2N−G3−Cellophilコポリマー(12kDa)であり、実施例13、ステップ7に記載された手順により調製した。HepG2細胞に、試験化合物を、0.04〜100μMの濃度で投与した。37℃での72時間のインキュベーションの終了時に、適切な色素または抗体を培養物に加えた。次いでプレートを、自動蛍光細胞画像機(ArrayScan(登録商標)、Thermo Scientific Cellomics)を使用してスキャンした。
以下の8個の細胞健康パラメーター(CHP)を評価した。
この研究は、100μMまでの濃度のG3−CellophilコポリマーへのHepG2細胞の曝露が、試験CHPに対して効果を有さなかったことを明らかにした。このことは、本開示のコポリマーの高い生体適合性を実証している。
Cellophil−(フルオレセイン)n−ADCのがん細胞特異性
実施例22のHER2抗体機能化G5−Cellophil−(フルオレセイン)8コポリマーの、その標的細胞への親和性を、SKBR3およびMDA−MB−468がん細胞系を使用した実験で検査する。SKBR3細胞は、ヒト上皮増殖因子受容体2を過剰発現し(HER2+)、一方、MDA−MB−468細胞は、その受容体を発現しない(HER2−)。結合は、以下の簡潔なプロトコールを使用してFACS(=蛍光活性化細胞選別)により評価する。

腫瘍細胞特異的アプタマーDML−7によるCellophil−(フルオレセイン)8の機能化
ステップ1:1−((3−アジドプロピル)アミノ)−2−メチル−1−オキソプロパン−2−イルエチルカルボノ−トリチオエートの合成:
CH2Cl2(44mL)中の2,5−ジオキソピロリジン−1−イル2−(((エチルチオ)カルボノチオイル)チオ)−2−メチルプロパノエート(2.4g、7.47mmol、1.0当量)およびEt3N(1.249mL、8.96mmol、1.2当量)の溶液に、CH2Cl2(15mL)中の3−アジドプロパン−1−アミン(0.879mL、8.96mmol、1.2当量)の溶液を室温および不活性雰囲気下で60分間かけて滴下により添加した。反応混合物を室温で一晩撹拌した。最後に、反応混合物を、HCl水溶液(1M、3×20mL)、ddH2O(2×25mL)およびNaHCO3飽和水溶液(20mL)で連続的に洗浄した。有機相を乾燥し(Na2SO4)、揮発物を減圧下で除去した。残留した橙色の油状物(2.25g、7.34mmol、98%)を、さらに精製することなく使用した。標記化合物の構造をNMR分光法により検証した。

フルオレセインを含むCellophilコポリマーの合成は、RAFT試薬としてRAFT−DMA−N3プレポリマーおよび共主要モノマーとしてAK−フルオレセイン−V2(8.0当量)を使用して、共主要モノマーの共重合についての一般手順(実施例13、ステップ7)を介して達成することができる。
転移性前立腺がん細胞への特異性が知られているアプタマーDML−7の修飾形態(Duan et al. (2016) Oncotarget 7(24): 36436)を、固相(Sigma Aldrich、Gillingham、UK)に合成した。

6炭素原子スペーサーおよび反応性アミノ基を付加して、機能化を容易にした。その後、アプタマーの凍結乾燥粉末を(緩衝剤)により室温で2時間再水和した。続いて、溶液を95℃で10分間加熱し、次いで、一晩かけて室温に冷まして、正確な三次元立体構造を得た。
DNAアーゼ無含有PBS緩衝剤中のDML−7−[C6]−NH2(1.0当量、ステップ5で調製)の溶液に、ジベンゾシクロオクチン−N−ヒドロキシスクシンイミジルエステル(1.5当量)を加える。反応混合物を、アプタマーアミンの完全な変換が観察(LC−MS)されるまで室温で混合する。続いて、Cellophil−(フルオレセイン)8−N3(2.0当量)を加え、アルキン−アプタマー中間体の完全な変換が観察されるまで反応させる。次いで混合物を、溶出剤として水(+0.01%のNaN3)を用いるセミ分取(semi-preparative)GPCにより精製する。所望の生成物を含有する精製画分を、脱塩カラム(PD10、Thermo Fisher Scientific、German)で脱塩する。続いて、所望の生成物を凍結乾燥して、白色の粉末を得る。
オキシム機能化ポリマー担体の合成
ステップ1:tert−ブチル(6,6−ジメチル−7,12−ジオキソ−4−チオキソ−3,5−ジチア−8,11−ジアザトリデカン−13−イル)オキシカルバメート(RAFT−EDA−オキシム−BOC)の合成
CH2Cl2(40mL)中の2−(((tert−ブトキシカルボニル)アミノ)オキシ)酢酸(486mg、2.54mmol、1.0当量)の溶液に、HOBt水和物(467mg、3.05mmol)およびN1−((エチルイミノ)メチレン)−N3,N3−ジメチルプロパン−1,3−ジアミン塩酸塩(511mg、2.67mmol、1.05当量)をN2下、0℃で加えた。得られた溶液を0℃で30分間撹拌した。最後に、2−(2−(((エチルチオ)カルボノチオイル)チオ)−2−メチルプロパンアミド)エタンアミニウム2,2,2−トリフルオロアセテート(966mg、2.54mmol、1.0当量)およびN−エチル−N−イソプロピルプロパン−2−アミン(2,24mL、13.2mmol、5.2当量)を連続的に加え、反応混合物を0℃で1時間、次いで室温で一晩撹拌した。すべての揮発物を減圧下で除去し、残留物をEtOAc(100mL)に取った。有機混合物を、NH4Clの飽和水溶液(3×40mL)、NaHCO3の飽和水溶液(3×40mL)、ddH2O(3×40mL)およびブライン(40mL)で連続的に洗浄した。有機相を乾燥し(Na2SO4)、すべての揮発物を減圧下で除去した。得られた黄色の固体残留物をEt2O(40mL)に懸濁した。懸濁液をろ過し、フィルターケーキをEt2O(2×20mL)、ddH2O(3×20mL)およびEt2O(3×20mL)で洗浄し、真空下で乾燥した。生成物を黄色の粉末(900mg、2.00mmol、収率79%)として単離した。標記化合物の構造をNMR分光法により検証した。

Cellophil−(フルオレセイン)8−オキシムを、文献(Dong et al. (2017) Angew Chem Int Ed 56: 1273)に記載されているように、酸化(NaIO4)ポリクローナル抗体IgG(アルデヒドIgG)に共有結合的にカップリングさせることができる。所望の生成物の構造をMSにより検証することができる。
モデルタンパク質へのCellophil−(フルオレセイン)8−N3の結合
ステップ1:mCherryモデルタンパク質のアルキン機能化:
脱ガスPBS緩衝剤、pH7.5中の実施例25のシステイン担持mCherryモデルタンパク質(1.0当量)の溶液に、過剰量のトリス(2−カルボキシエチル)ホスフィン(TCEP)(100当量)を不活性雰囲気下で加える。得られた溶液を十分に混合し、DMSO中のジベンゾシクロオクチン−マレイミド(DBCO−マレイミド)の脱ガス溶液を不活性雰囲気下で加える前に、20分間放置する。得られた反応混合物を室温で一晩撹拌した。次いで混合物を、溶出剤として水(+0.01%のNaN3)を用いるセミ分取GPCにより精製する。所望の生成物を含有する精製画分を、脱塩カラムを使用して脱塩して、mCherry−DBCOを得る。
PBS緩衝剤、pH7.5中のmCherry−DBCO(1.0当量)およびCellophil−(フルオレセイン)8−N3(2.0当量、実施例26、ステップ4で得た)の溶液を、室温で16時間撹拌する。次いで混合物を、製造会社のプロトコールに従ってNi2+NTAアフィニティークロマトグラフィー(ニッケル−NTAアガロース、Thermo Fisher Scientific、Germany)により精製する。(mCherryはヘキサ−ヒスチジンタグを含む。)所望の生成物を含有する精製画分を、脱塩カラムを使用して脱塩して、mCherry−Cellophil(フルオレセイン)8を得る。得られたタンパク質ポリマーコンジュゲートは、実施例25に提示されたプロトコールおよび対照としてCys−mcherry−LPETG−His6(実施例25を参照すること)を使用して、SDS−PAGEにより分析することができる。クーマシー染色ゲルにおいて観察された対照と比較した生成物のサイズの増加は、タンパク質へのポリマーのカップリングが成功したことを示している。
アザマイケルライゲーション戦略を利用したアミン修飾NH2−Gn−Cellophil−(DOX)8によるHer2+抗体(トラスツズマブ)における未変性リジン残基の機能化
トラスツズマブ−Cellophil−(DOX)16の合成は、抗体の軽鎖にカップリングしているアミン求核剤としてNH2−Gn−Cellophil−(DOX)8を使用し、Bernades, G. J. L.(J Am Chem Soc (2018) 140: 4004-)により開発されたプロトコールに従って達成される。これは、抗体分子1個あたり平均して16個のドキソルビシン分子を含むADC複合体をもたらす。トラスツズマブ(PBS中20mg/ml)はCarbosynth、UKから得た。
トラスツズマブ−Cellophil−(DOX)16の抗がん有効性
トラスツズマブ−Cellophil−(DOX)16(実施例29を参照すること)の抗がん有効性は、以下のように検証することができる。
フルオレセイン修飾共主要モノマーを使用するクリック反応性アジドCellophil(フルオレセイン)8の合成
AK−フルオレセイン(実施例10)を、以下のプロトコールを利用して、アジド修飾RAFT剤(1−((3−アジドプロピル)アミノ)−2−メチル−1−オキソプロパン−2−イルエチルカルボノトリチオエート)によるRAFT重合において共重合した。
発癌性タンパク質を標的にするヨウ素負荷Cellophil抗体コンジュゲートの合成
広範囲の癌遺伝子に対するモデル抗体ポリマーコンジュゲートを生成するために、タンパク質BMI−1を標的にする市販の抗体を選択した。BMI−1(ポリコームリングフィンガー(polycomb ring finger)癌遺伝子)は、成人造血幹細胞、ならびに成人末梢および中枢神経系神経幹細胞の効率的な自己再生細胞分化にとって必要である。BMI−1は、細胞周期阻害遺伝子であるp16およびp19を調節する癌遺伝子であると報告されている。BMI−1の過剰発現は、膀胱、皮膚、前立腺、乳房、卵巣、結腸直腸などのいくつかのタイプのがん、ならびに血液系悪性腫瘍において重要な役割を果たすと思われる(Lessard J et. al. (2003). Nature 423 (6937): 255−60. doi:10.1038;Molofsky AV et. al. (2005). Genes Dev. 19 (12): 1432−7. doi:10.1101/gad.1299505)。
カップリング生成物の詳細な特徴付けでは、IdeS消化抗体CellophilコンジュゲートのLC−MS分析を、以下のプロトコールを使用して実施した。
mTGタグ(=NH2−PEGn)−RAFT−BOCによるCellophil(DMAn/AKm)コポリマーの合成
以下の手順は、放射性同位体を結合するために共有結合キレート剤により機能化されうるCellophilコポリマーの合成を説明している。ここに提示されているコポリマー(mTGタグ)−DMA30/AK8は、一般合成手順を例示する機能を果たす。コポリマーのサイズおよびコポリマーに含まれている機能化部位の数は、用いるモノマーのモル比を変えることで変更することができる。
無水物DOTA/NHS−DOTAによるCellophil[BOC−NH−PEG5−(DMA30/AK8)]コポリマーの機能化
ddH2O中のCellophil BOC−NH−PEG5−(DMA30/AK8)(20mg、3.45μmol)の溶液に、DMSO中の無水物DOTA(25mg、36μmol)またはNHS−DOTA(27mg、36μmol)の溶液を加え、35℃で24時間撹拌し、次いでこの溶液に3M HClを加え、0℃に1時間加熱した。次いで、得られた混合物をddH2Oで透析し(MWCO 3.5kDa)、濃縮液をフリーズドライして、NH2−PEG5−(DMA30/AK−DOTA8)を得た。得られた化合物の構造をNMR分光法により検証した。
Cellophil[DBCO−NH−PEG5−(DMA30/AK−DOTA8)]の合成。
乾燥DMSO(1.5mL)中のCellophil NH2−PEG5−(DMA30/AK−DOTA8)(3.6μmol)、DBCO−NHS(18μmol)およびトリエチルアミン(7.2μmol)の溶液を25℃で24時間撹拌した。次いで、得られた混合物を0.1Mの炭酸アンモニウムで透析し(MWCO 3.5kDa)、濃縮液をフリーズドライして、Cellophil DBCO−NH−PEG5−(DMA30/AK−DOTA8)を得た。反応をGPCで追跡し、得られた化合物の構造をNMR分光法により検証した。
Her2受容体過剰発現がん細胞を標的にする診断薬および治療薬のための放射標識トラスツズマブ−[Cellophil−(DOTA4]2コンジュゲートの合成
腫瘍診断では、原発性腫瘍またはその転移の検出限界が患者の生存率にとって重要であり、それは後期腫瘍が多くの場合に不十分な予後に関連するからである。がん細胞の検出と同様に後の治療における放射標識腫瘍組織特異的抗体の使用は、放射線医学(radio medicine)にとって潜在的に有望な方法である。しかし、この種の手法は低い信号対雑音比が妨げになっており、それは、わずかな放射性同位体のみが標的化部分/抗体に結合されうるという事実、および目的の放射性同位体が短い(通常、抗体の半減期より短い)半減期を有するという事実に起因している。したがって、放射性同位体のカーゴを増やすことがきわめて望ましい。この実施例には、改善された腫瘍細胞の検出および治療のために放射標識抗体Cellophilコンジュゲートが記載されている。
シクロアルキンCellophil(DOTAn)−CO2Hの合成
本開示のコポリマーをクリック反応性シクロアルキン基、例えばDBCOにより機能化するため、クリック反応性部分を、RAFT基の除去の際に組み込むことができる。
ステップ1−シクロアルキン開始剤の合成
DBCO修飾開始剤を、Ulbrichおよび同僚のプロトコール(Polym. Chem., 2014 5, 1340)に従って合成する。
ステップ2−RAFT−Cellophil−CO2Hコポリマーの合成:
RAFT−Cellophil−CO2Hコポリマーの合成は、実施例13に記載された一般的プロトコール(ステップ6〜7のエチルRAFT試薬から出発する)に従って達成される。
ステップ3−シクロアルキンCellophil(DOTAn)−CO2Hコポリマーの合成:
DMSO/ddH2O(1/1)中のRAFT−Cellophil−CO2Hの溶液に、シクロアルキン含有開始剤(20当量)を一度に加える。反応混合物を封止し、黄色が消滅するまで(4時間)70℃で加熱する。そのうえ、反応の進行をHPLCによって追跡する。得られた溶液を室温に冷却し、pHを、p−NCS−Bz−DOTA−GA(Chematech)またはDOTA−NHS(コポリマーにおける反応性アミノ基1つあたり2当量)を加える前に8に調整する。混合物をddH2O(MWCO:5000Da)で透析し、濃縮液を凍結乾燥し、NMR分光法およびSECにより特徴付ける。
Her2受容体過剰発現がん細胞を標的にする治療薬のための放射標識トラスツズマブ−[Cellophil−(DOTA4]4コンジュゲートの合成
実施例33および35に提示された手順により合成されたDBCO機能化Cellophilポリマーを、製造会社のプロトコールに従って使用した市販の酵素ベース修飾キット(SiteClick(商標)Antibody Labeling System、Thermo−Fisher−Scientific、Waltham、USA)により、重鎖1つあたり2つのアジド基で機能化されている(抗体1つあたり最大4つのアジド基の付加をもたらす)IgGタイプのがん細胞特異的抗体(Her2+がん細胞を標的にするトラスツズマブ(登録商標))にコンジュゲートした。
mTgタグCellophilからの、Her2受容体過剰発現がん細胞を標的にする診断薬および治療薬のための放射標識トラスツズマブ−[Cellophil−(DOTA8]2コンジュゲートの合成
実施例34のCellophilコポリマーを、基質としてコポリマーのNH2−PEG5基を使用して、トランスグルタミナーゼ媒介反応により、トラスツズマブのようなモノクローナル抗体に直接カップリングすることができる。このため、実施例36のプロトコールを使用するが、NH2−PEG4−アジドをNH2−PEG5−DMA30/AK−DOTA8に置き換え、抗体の40倍のモル過剰量で使用して16個のキレート剤を有する抗体、すなわち、トラスツズマブ−[Cellophil−(DOTA8]2を生成することが例外である。
TCO−Tzクリック化学による標的化部分へのカップリングのための、テトラジン機能化Cellophilコポリマーの合成
ステップ1:テトラジン−Cellophilコポリマーの合成:
乾燥DMSO(1.5mL)中の実施例34のCellophilコポリマー(3.6μmol)、2,5−ジオキソピロリジン−1−イル2−(4−(6−メチル−1,2,4,5−テトラジン−3−イル)フェニル)アセテート(18μmol)およびトリエチルアミン(7.2μmol)の溶液を、25℃で24時間撹拌した。次いで、得られた混合物を0.1Mの炭酸アンモニウムで透析し(MWCO 3.5kDa)、濃縮液をフリーズドライして、テトラジン−Cellophil[DMA30/AK−DOTA8]を得た。反応をSECにより追跡し、得られた化合物の構造をNMR分光法により検証した。
テトラジン−歪みアルキン[4+2]付加環化を利用する蛍光団修飾Cellophil−[DMA30/AK−DOTA8)の合成
乾燥DMSO(0.5mL)中の実施例40のテトラジン修飾Cellophilコポリマー(1.3μmol)および(E)−6−アミノ−9−(2−カルボキシ−5−((5−(((シクロオクタ−4−エン−1−イルオキシ)カルボニル)アミノ)ペンチル)カルバモイル)フェニル)−4,5−ジスルホ−3H−キサンテン−3−イミニウム(1.3μmol)の溶液を、25℃で24時間撹拌した。次いで、得られた混合物を0.1Mの炭酸アンモニウムで透析し(MWCO 3.5kDa)、濃縮液をフリーズドライして、AF色素488クリックCellophilを得た。反応をGPCにより追跡し、得られた化合物の構造をNMR分光法により検証した。蛍光団標識Cellophil誘導体を使用して、例えば、強力な読み取りシグナルが有益である血流内のコポリマーの半減期または腎排出を決定するために、薬力学的研究を実施することができる。
TCO修飾タンパク質へのCellophilテトラジン−[DMA30/AK−DOTA8)のカップリング
実施例40のテトラジン機能化Cellophilコポリマーの溶液(3当量)をPBS(pH7.4)に溶解し、PBS(pH7.4)に溶解したトランスシクロオクテン(TCO)修飾タンパク質[NH2−PEG5−TCOをNH2−PEG5−アジドに置換して実施例36に提示された方法と同様の方法によって調製することができる](2つのTCO基を含む1当量)を撹拌下、室温で3時間かけて滴下により添加する。続いて未反応ポリマーを、100kDaのMWCOを有する膜を使用する透析により除去する。反応の成功をSDS−PAGEおよびHPLCにより追跡する。
AK−DOTAの合成
無水DMF(1mL)中のAK(50mg、250μmol)およびEt3N(104μL、749μmol)の溶液に、DOTA−NHS(HPF6/TFA塩)(200mg、262μmol)を加えた。反応混合物を室温で一晩撹拌し、綿のパッドでろ過した。ろ液をMeCNに沈殿させ、次いでろ過した。ケーキをMeCNで洗浄し、減圧下で乾燥して、白色の粉末(95mg、65%)を得た。この化合物を、重合の際にコポリマーへのキレート剤の指向組み込みに使用することができる。
Claims (19)
- (a)(1)モノマーが少なくとも1個のビニル基を有し、アミノ酸残基を含まないことを特徴とする、1つまたは複数の重合性主要モノマーと、
(2)YおよびZのうちの少なくとも一方がHである式IまたはIIの1つまたは複数の共主要モノマーと、
(3)必要に応じて、式III〜Xのいずれかの1つまたは複数の共主要モノマーと、
(4)ラジカル重合を制御する薬剤と、
(5)フリーラジカル種を生成する開始剤系とを含む反応混合物を重合して、前記重合がコポリマーを生じること、
(b)必要に応じて、前記コポリマーを細胞型特異的または組織型特異的標的化部分で機能化すること、ならびに
(c)ステップ(a)または必要に応じたステップ(b)の後に、活性剤を前記コポリマーにカップリングすること

請求項4に記載の複数の活性剤分子を含むコポリマー。 - 前記RAFT剤が、5〜25単位の単分散スペーサーを含む、請求項4に記載の複数の活性剤分子を含むコポリマー。
- 前記RAFT剤が、細胞型特異的または組織型特異的標的化部分による前記コポリマーの機能化に使用される反応性基を含む、請求項4に記載の複数の活性剤分子を含むコポリマー。
- 前記反応性基が、チオール、アルデヒド、アルキン、アジド、テトラジン、歪みアルケン、アミン、カルボキシル、エステル、ジアジリン、アジ化フェニル、チオエステル、ジアゾ、シュタウディンガー反応性ホスフィノエステル(もしくは、ホスフィノチオエステル)、ヒドラジン、オキシム、アザマイケルライゲーションを実施するためのアクリレート、または酵素カップリング反応に使用することができるモチーフである、請求項7に記載の複数の活性剤分子を含むコポリマー。
- 前記モチーフが、2〜8個のアミノ酸単位を含み、かつソルターゼ媒介カップリング反応を可能にするオリゴ−グリシン、トランスグルタミナーゼ反応性基質、アルデヒドタグ、または自己触媒性インテイン配列である、請求項8に記載の複数の活性剤分子を含むコポリマー。
- 前記RAFT剤のRAFT基が、前記コポリマーの共重合または機能化の後に脱離される、請求項4〜9のいずれかに記載の複数の活性剤分子を含むコポリマー。
- (a)モノマーが少なくとも1個のビニル基を有し、アミノ酸残基を含まないことを特徴とする、1つまたは複数の重合性主要モノマーと、
(b)式III〜Xの1つまたは複数の共主要モノマーと、
(c)必要に応じて、式Iおよび/または式IIの1つまたは複数の共主要モノマーと、
(d)ラジカル重合を制御する薬剤と、
(e)必要に応じて、コポリマーを細胞型特異的または組織型特異的標的化部分で機能化することと、
(f)フリーラジカル種を生成する開始剤系と、
を含む反応混合物の重合によって作製される、複数の活性剤分子を含むコポリマー。 - 前記コポリマーが、5,000ダルトン〜100,000ダルトンの平均分子量を有する、請求項11に記載の複数の活性剤分子を含むコポリマー。
- 前記ラジカル重合を制御する薬剤がRAFT剤である、請求項11または12に記載の複数の活性剤分子を含むコポリマー。
- 前記RAFT剤が、細胞型特異的または組織型特異的標的化部分による前記コポリマーの機能化に使用される反応性基を含む、請求項13に記載の複数の活性剤分子を含むコポリマー。
- 前記反応性基が、チオール、アルデヒド、アルキン、アジド、テトラジン、歪みアルケン、アミン、カルボキシル、エステル、ジアジリン、アジ化フェニル、チオエステル、ジアゾ、シュタウディンガー反応性ホスフィノエステル(もしくは、ホスフィノチオエステル)、ヒドラジン、オキシム、アザマイケルライゲーションを実施するためのアクリレート、または酵素カップリング反応に使用することができるモチーフである、請求項14に記載の複数の活性剤分子を含むコポリマー。
- 前記モチーフが、2〜8個のアミノ酸単位を含み、かつソルターゼ媒介カップリング反応を可能にするオリゴ−グリシン、トランスグルタミナーゼ反応性基質、アルデヒドタグ、または自己触媒性インテイン配列である、請求項15に記載の複数の活性剤分子を含むコポリマー。
- 前記RAFT剤のRAFT基が、前記コポリマーの共重合または機能化の後に脱離される、請求項13〜16のいずれかに記載の複数の活性剤分子を含むコポリマー。
- 有効量の、請求項1〜17のいずれかに記載の複数の活性剤分子を含むコポリマー、および薬学的に許容される担体または賦形剤を含む、医薬組成物。
- 対象に前記医薬組成物を投与することを含む、前記対象のがん、または別の疾患もしくは状態を処置するための、請求項18に記載の医薬組成物の使用。
