Abstract
Description
近年、きわめて少数の新しい治療法が開発されているが、固形腫瘍に罹患した患者の生存率を高め、彼らの生活の質を改善するためにはさらなる改善が依然として必要である。
1つの有望な治療法は抗体薬物複合体であり、ここで、細胞を殺す能力を有する高度に毒性の実体は、疾患の部位に毒素を送達させる(shuttles)標的結合実体に結合される。
本出願人は過去に、いわゆるATAC、すなわち、アマニチン毒素が高度に標的特異的な抗体に結合されている、抗体標的化アマニチン複合体(Antibody Targeted Amanitin Conjugates)を開発した。
α−アマニチンは、RNAポリメラーゼIIIにわずかしか結合せず、そして、RNAポリメラーゼIに結合しない。
RNAポリマーゼIIに結合するとき、アマニチンはRNAとDNAに対する、酵素のトランスロケーションを阻害し、そして、したがって、転写速度が1000倍以上低下し、最終的には、細胞のアポトーシスをもたらす。
これは、6'−デオキシ位を有するアミノ酸4(上記の図において=R4);および、S−デオキシ位を有するアミノ酸8を有するアマトキシンを含む。
リンカーは、PAB Val Alaリンカーである。
このような血液学的悪性腫瘍は、本明細書では「液性腫瘍」とも呼ばれる。
しかしながら、出願人は、液性腫瘍よりも、ATACaのより困難な標的となる固形腫瘍の治療にはまだ改善の余地があることを認識している。
したがって、本発明の1つの目的は、固形腫瘍の新しい治療選択肢を提供することである。
本発明のさらなる1つの目的は、固形腫瘍におけるATACの効力を改善することである。
これらおよびさらなる目的は、本発明の独立請求項に記載の方法および手段によって達成される。
従属請求項は、特定の実施形態に関連する。
本発明は、固形腫瘍の治療のための標的化されたアマトキシン複合体を提供する。
本発明を詳細に説明する前に、本発明は、そのようなデバイスおよび方法は変化し得るので、説明されたデバイスの特定の構成要素部分に限定されず、説明された方法のプロセスステップに限定されないことを理解されたい。

これらの構築物の重要な特徴の1つは、分子のアミノ酸4(Trp)のインドール環の4位の−OR4置換基である。
これは、4位に−OR4を有さないが、代わりに水素置換基を有する構造的に類似の結合部分−毒素複合体と比較した場合に、とりわけ真実である。
興味深いことに、液性腫瘍の治療においては、PCT出願PCT/EP2017/084431で開示されているように、両者の変異体が同等の有効性を有していたので、そのような明白な相違は観察できなかった。
この理由のために、切断/壊滅(immolation)は、6−OH−Trpを送達する。
適切な切断可能なリンカーとしては、例えば、細胞内プロテアーゼ(例えば、リソソーマルプロテアーゼまたはエンドソーマルプロテアーゼ)によって切断可能なペプチドリンカーが挙げられる。
例示的な実施形態では、リンカーは、
バリン−シトルリン(val−cit)、
バリン−アラニン(val−ala)、
フェニルアラニン−リジン(phe−lys)リンカー、またはマレイミドカプロニック「バリン−シトルリン−p−アミノベンジルオキシカルボニル(mc−Val−Cit−PABA)リンカー」などのジペプチドリンカーであり得る。
このようなリンカーは、例えば、米国特許第6214345号に開示されている。
スルホ−smcc複合化はスルフヒドリル(チオール、−SH)と反応するマレイミド基を介して起こり、一方、そのスルホ−NHSエステルは、(リジンおよびタンパク質またはペプチドN末端に見られるように)第一級アミンに対して反応性である。
他の適当なリンカーとしては、特定のpHまたはpH範囲で加水分解可能なリンカー、例えばヒドラゾンリンカーが挙げられる。
自壊性リンカー(self immolative linker)の一例は、p−アミノベンジルアルコール(PAB)であり、これは、しばしば、芳香族アミン基を介して、そしてカルバメート基を介して、毒素の第一級または第二級アミンに、バリン−シトルリン(val−cit)またはバリン−アラニン(val−ala)のような切断可能なジペプチドに結合される。

金属イオンは、3つのリガンドを介してタンパク質に配位する。

セリンプロテアーゼは、その構造に基づいて、キモトリプシン様(トリプシン様)またはスブチリシン様の2つの広いカテゴリーに分類される。
このクラスの酵素のプロトタイプメンバーはプロテアソームの触媒サブユニットであるが、アシルトランスフェラーゼは同じ活性部位の形状とメカニズムを収束的に(convergently)進化させた。
一般に、それらは活性部位に2つの高度に保存された(conserved)アスパラギン酸を持ち、酸性pHで最適に活性を示す。

この理由のために、RajiまたはRaji−Lucは、固形腫瘍対液性腫瘍の治療における、クレームされている毒素リンカー複合体の異なる効果を示すための適切なモデルである。
抗体h3/F11のCDR配列は、EP2363486に開示されており、その内容は参照により本明細書に組み込まれる。

本明細書中に記載される実験において、本発明の範囲に該当する4つの標的化アマトキシンリンカー複合体が使用され、これは、式(i)のR4の下で言及されるように、それらが非複合体化またはリンカーに複合体化された、いずれかの6−OH Trp残基を有することを意味する。

2.抗体
以下の抗体を、アマトキシンリンカー複合体に複合させた
トラスツズマブおよびh3/F11は既に上記に記載されている。
chiBCE19は、Luttgauら、2013に実施可能に開示されている。
抗体を、いわゆるチオマブテクノロジー(Thiomab technology)の手段によって、アマトキシンリンカー複合体に複合させた。
この方法では、以下の反応式に示されるように、複合は、毒素リンカー構築物のマレイミド残基の、抗体におけるシステイン残基の遊離SH基への複合によって起こる。
この複合法の原理は、Junutulaら(2008)に開示されており、その内容は、参照により本明細書に組み込まれる。
本実験において使用される抗体は、このような遊離SH基を有するシステイン残基を提供するために、両方のFcドメインにおけるD265C置換を含む。
3.1.細胞生存率測定試験
抗PSMAおよび抗Her2アマトキシン複合体の細胞毒性活性を、標的陽性腫瘍細胞株および化学発光BrdU取り込みアッセイ(Roche Diagnostics)を用いてin vitroで評価した。
用量反応曲線のEC50値は、Graphpad Prism 4.0ソフトウエアによって計算した。
標的陽性細胞に対する抗CD19アマトキシン複合体の細胞毒性活性を、96ウェル組織培養プレートにおいて、CellTiter−Glo 2.0発光細胞生存率アッセイ(Promega、Madison、WI)によって評価した。
データ分析は、GraphPad Prism 7(GraphPad Software、Inc、La Jolla、CA)ソフトウェアで行い、曲線適合をプロット(plot curve fits)し、統計的分析を行った。
LnCap異種移植片モデル:雄CB17 SCIDマウスに、マウス1匹当たり、2.5×106のLNCap前立腺癌細胞を、右側腹部皮下(right flank)に接種した。
平均腫瘍体積、約150 mm3の動物を0日目に群に割り付けた。
同日に、動物に、アマニチンベースの抗PSMA抗体薬物複合体(ADC)を単回静脈内投与し、腫瘍容積および体重を週2回測定した。
平均腫瘍体積、約120 mm3の動物を0日目に群に割り付けた。
同日に、動物に、アマニチンベースの抗Her2抗体薬物複合体(ADC)を単回静脈内投与し、腫瘍容積および体重を週2回測定した。

3.4.1.10mgのchiBCE19−D265CへのHDP 30.2060の複合化
PBS緩衝液中の10mgのThiomab chiBCE19−D265Cを、HDP 30.2060への結合のために使用する。
#抗体溶液を、1mM EDTAに調整する:
*2mLの抗体溶液(10.0mg)+20μLの100mM EDTA、pH 8.0
*抗体量:10mg= 6.8×10-8モル
#2mLの抗体溶液(6.8×10-8 mol)+54.5μLの50mM TCEP溶液(2.72×10-6 mol)
#シェーカー機上で、37℃で、3時間インキュベートする。
#Slide−A−Lyzer Dialysis Cassette 20'000 MWCO中、2.0 I 1x PBS、1mM EDTA、pH 7.4中、4℃での2回の連続透析、
最初の透析は約4時間、2回目の透析は一晩。
#Amicon Ultra Centrifugal Filters 50'000 MWCOを用いて、約4.0mLに濃縮する。
#約2mLの抗体溶液(6.8×10-8モル)+27.2μLの新鮮な50mM dhAA溶液(1.36×10-6モル)
#シェーカー機上で、室温で、3時間インキュベートする。
#70μLのDMSO中に0.7mgのHDP 30.2060を可溶化する=10μg/μL
*約2mLの抗体溶液(=9.5mg:6.46×10-8モル)+50.9μL HDP 30.2060(=509μg:3.88x10-7 mol)。
*室温で1時間インキュベートする。
*16μLの100mM N−アセチル−L−システイン(1.62×10-6モル)を加えてクエンチする。
*室温で15分間(または4℃で一晩)インキュベートする。
*1×PBS、pH 7.4で平衡化したPD−10カラムで各反応混合物を精製する。パラフィルム上でブラッドフォード試薬を用いてタンパク質含有画分を同定し、タンパク質含有画分を合わせる。
*2.0 I PBS、pH 7.4およびSlide−A−Lyzer Dialysis Cassettes 20'000 MWCO中、各抗体溶液を4℃で一晩透析する。
蛋白質濃度を5.0mg/mL(3.4×10-5 M)に調整し、濾過により滅菌状態にする。4℃で保存する。
JIMT−1は乳管腺癌(breast ductal adenocarcinoma)由来の固形がん細胞株である。
結果を図1に示す。
30.1699、
30.2371、
30.2060および
30.2347
は、同等の効力を示し、一方、Trp変異体30.2115はわずかに低減した効力を示し、Trp変異体30.2183は、強く低減した効力を示す。
すべての場合において、6'−ヒドロキシ−Trp変異体(ここで、式(i)のR4は、Hであるか、または前記アマトキシンを標的結合部分に連結するための反応性基Yを有するリンカーである)は、Trp変異体(ここで、アマニチンのTrp残基における6位は、H残基で置換されている)よりも高い細胞毒性能力を示した。
「S」は、式(i)のアマニチンにおけるR5が存在しないことを意味し、
「6−OH−W」は、OR4を有する式(i)に示される、実施形態を指し、
「W」は、アマニチンのTrp残基の6位がH残基で置換されている本発明に従わない実施形態を指し、そして、
「AA4/AA1」は、リンカーが結合するアマニチンのアミノ酸残基を意味する。
結果を図2に示す。
30.1699、
30.2371、
30.2060および
30.2347
は、強力な腫瘍縮小活性を示し(図2A)、一方、Trp変異体30.2115および30.2183は有意に低減した腫瘍縮小活性を示す(図2B)。
Fcドメインに、D265C置換を有する、抗PSMA抗体h3/F11に結合した、上述の毒素リンカー構築物を用いて処理されたLNCaP細胞を用いて、細胞生存率アッセイを行った。
LNCaPは左鎖骨上リンパ節転移(left supraclavicular lymph node metastasis)に由来する固体前立腺腺癌細胞株である。
結果を図3に示す。

30.2060および
30.2347
は同等の効力を示し、一方、Trp変異体30.2115および30.2183は、強く低減した効力を示す。
ほぼ同じパターンが全ての細胞株で観察された。
すべての場合において、6'−ヒドロキシ−Trp変異体(ここで、式(i)のR4は、Hであるか、または前記アマトキシンを標的結合部分に連結するための反応性基Yを有するリンカーである。)は、Trp変異体(ここで、アマニチンのTrp残基の6位がH残基で置換されている)よりもわずかに高い細胞毒性能力を示す。
「S」は、式(i)のアマニチンにおけるR5が存在しないことを意味し、
「6−OH−W」は、OR4を有する式(i)に示される、実施形態を指し、
「W」は、アマニチンのTrp残基の6位がH残基で置換されている本発明に従わない実施形態を指し、そして、

「Nfb」は、>50%の残存細胞生存率を有する本格的な細胞毒性の能力がないことを意味する。

結果を図4に示す。

は強力な腫瘍縮小活性を示し(図4A、B)、一方、Trp変異体30.2115および30.2183は有意に低減した腫瘍縮小活性を示す(図4C)。
細胞生存率アッセイは、FcドメインにD265C置換を有する抗CD19抗体chiBCE19に結合した、上記の毒素−リンカー構築物で処理したRaji細胞を使用した。
Rajiはバーキットリンパ腫(Burkitt's lymphoma)由来の細胞株である。
30.1699、
30.2371、
30.2060および
30.2347、ならびに
Trp変異体30.2115および30.2183は同等の効力を示す。
ほぼ同じパターンが全ての細胞株で観察された。
「S」は、式(i)のアマニチンにおけるR5が存在しないことを意味し、
「6−OH−W」は、OR4を有する式(i)に示される、実施形態を指し、
「W」は、アマニチンのTrp残基の6位がH残基で置換されている本発明に従わない実施形態を指し、そして、

「nfb」は、>50%の残存細胞生存率を有する、本格的な細胞毒性の能力がないことを意味する。
(X%)は、残存生存率を示す。
Rajiはバーキットリンパ腫由来の細胞株であり、Nalm−6は急性リンパ芽球性白血病(ALL)由来の細胞株である。
結果を図6のカプランマイヤー曲線に示す。
全ての場合において、6−OH Trp(30.1699)を有する変異体は、Trp変異体30.2115よりも低い力価を有することが判明した。
このアプローチは、固形腫瘍の表現型を模倣する腫瘍を導く。

Claims (17)
- リンカーが切断可能なリンカーおよび/または自壊性リンカーである、請求項1に記載のアマトキシン−リンカー構築物。
- リンカーが、以下からなる群から選択される少なくとも1つの薬剤によって切断可能である、前記請求項のいずれか一項に記載のアマトキシン−リンカー構築物:
・システインプロテアーゼ
・メタロプロテアーゼ
・セリンプロテアーゼ
・トレオニンプロテアーゼ、および(または)
・アスパラギン酸プロテアーゼ。 - リンカーが、以下からなる群から選択されるモチーフを含む、前記請求項のいずれか1項に記載のアマトキシン−リンカー構築物。
・Val Ala
・Val Cit
・Val Lys
・Val Arg
・Phe Lys Gly Pro Leu Gly
・Ala Ala Pro Val
・β−グルクロニド、および/または
・β−ガラクトシド。 - リンカーが非切断可能性リンカーである、請求項1に記載のアマトキシン−リンカー構築物。
- 前記固形腫瘍が、そのアマトキシンがインドールの6'位に前記−OR4置換基を含まない、結合部分−毒素複合体に対して抵抗性である、上記請求項のいずれか1項に記載のアマトキシン−リンカー構築物。
- (i)固形腫瘍と診断された、
(ii)固形腫瘍に罹患している、または
(iii)固形腫瘍を発症する危険性がある、ヒトまたは動物の被験体の治療において使用するための結合部分−毒素複合体であって、以下を含む複合体:
(a)請求項1〜9のいずれか1項に記載のアマトキシン−リンカー構築物、および
(b)標的結合部分;および
ここで、リンカーは、前記アマトキシンおよび前記標的結合部分を連結する。 - 標的結合部分が、抗体、抗体断片、抗体ベースの結合タンパク質、または抗体模倣物であり、それらの全てが標的結合特性を保持する、請求項10に記載の使用のための複合体。
- 前記標的結合部分が、以下からなる群から選択される少なくとも1つの標的に結合する、請求項10〜11のいずれか1項に記載の使用のための複合体:
・Her2、および/または
・PSMA。 - 固形腫瘍が、以下からなる群から選択される少なくとも1つである、請求項10〜12のいずれか1項に記載の使用のための複合体:
a)肉腫,
b)芽腫、および/または
c)癌。 - 抗体またはその断片もしくは誘導体が、操作されたシステイン残基を含む、請求項10〜13のいずれか1項に記載の使用のための複合体。
- 前記システイン残基が、EU番号付けシステムによる、重鎖118Cys、重鎖239Cys、および重鎖265Cysからなる群から選択される、請求項14に記載の使用のための複合体。
- 前記固形腫瘍が、そのアマトキシンがインドールの6'位に前記−OR4置換基を含まない、結合部分−毒素複合体に対して抵抗性である、請求項10〜15のいずれか1項に記載の使用のための複合体。
- (i)固形腫瘍と診断された、
(ii)固形腫瘍を患っている、または
(iii)固形腫瘍を発症する危険性がある、ヒトまたは動物の被験体の治療において使用するための医薬組成物であって、請求項10〜16のいずれか1項に記載の複合体を含む、医薬組成物。
