T細胞を活性化するT細胞エピトープの能力(免疫原性スコア)は、2つの補完的な特性の結果である:i)HLAへの親和性およびii)異なるHLAハプロタイプに無差別に結合する能力。免疫原性スコアが最も高いペプチドを選択する目的で、異なる起源のいくつかのT細胞エピトープのインシリコ評価(Epivax)を実施した。予備段階では、異なる起源(キーホールリンペットヘモシアニン−KLH、ジフテリア毒素、インフルエンザウイルス、エプスタインバーウイルスおよびヘルペスウイルス)に由来する10種の異なるペプチドを評価した。予測されるHLA親和性とHLAハプロタイプカバレッジ(haplotype coverage)に基づいて、ヒトで免疫原性が高い可能性があるため、最高の免疫原性スコア(10を超える)を持つペプチドを選択した(選択したペプチド配列を表1に示す)。
ユニバーサルT細胞エピトープペプチドの合成と精製の一般的方法
T細胞ペプチドを、標準的なFmoc化学を使用して、2−クロロトリチル樹脂上の線形固相ペプチド合成(SPPS)によって製造した。標準的なカップリング手順は、3.0当量のアミノ酸とカップリング試薬を使用して、DMF中の3.0当量の塩基の存在下、室温で1時間行った。困難なカップリングシーケンスでは、反応時間を延長してダブルカップリングを実装した。アミノ酸カップリングの完了後、ピリジン中の5.0当量のAc2Oを使用してアセチル化キャッピングステップを導入し、望ましくないペプチド鎖の伸長を中止した。樹脂をDMFで洗浄し、DMF中の20%ピペリジンを5分間使用してFmoc基を除去した。SPPSを終了した後、樹脂からの全体的な脱保護とペプチド切断を、標準的な切断カクテル(TFA/TIS /水)を使用して室温で2時間行った。樹脂を濾別し、TFAで洗浄した。続いて、粗生成物を10倍過剰量の冷イソプロピルエーテル/ヘキサンで沈殿させ、ガラスフリットを使用して固体を濾別し、真空下で乾燥させた。粗ペプチドは、分取用HPLCシステムで溶媒A(水、0.1%TFA)と溶媒B(アセトニトリル、0.1%TFA)のグラジエントを使用して、逆相C18カラムで精製した。純度が90%を超える目的のペプチドを含むHPLCフラクションを一緒にプールし、水で希釈してイオン交換を行った。所望のイオン交換画分を凍結乾燥した。最終ペプチドの同一性と純度を、HPLC−MS分析によって特徴づけ、確認した。
封入されたT細胞エピトープペプチドを含むワクチンを、薄膜法とそれに続く均質化および押し出しによって製造した。まず、それぞれ9:1:7:0.05のモル比のDMPC、DMPG(Lipoid、ドイツ)、コレステロールおよびモノホスホリルヘキサアシルリピドA、3−デアシル合成のまたは3D−(6−アシル)PHADTMを60℃にてエタノール中に可溶化させた(Avanti Polar Lipids、USA)。薄い脂質膜を得るために、エタノールを真空ロータベーパー下で蒸発させた。
脂質フィルムは、これらのバッファーの1つで再水和された(封入されるT細胞エピトープペプチドによって異なる)。
・ 20mM酢酸ナトリウムpH4(Fluka)、0.8mg/mL T細胞エピトープペプチドSAT42を含むMilliQ水中の5%DMSO(Sigma Aldrich)、または
・ 0.1xPBS pH7.4、0.3〜0.4mg/mL T細胞エピトープペプチドSAT43、SAT44、またはSAT47を含むMilliQ水中の5%DMSO(すべてSigma-Aldrich)。
溶液を15分間穏やかに攪拌した。ガラスビーズの存在下でサンプルをさらに激しくボルテックスした。得られた多層ベシクルを10回の凍結融解サイクル(液体窒素と37℃の水浴)にかけ、均質化した後、孔径0.1/0.08μmのポリカーボネートメンブレン(Whatman、UK)に順次押し出した。均質化と押し出しの両方のステップを、EmulsiFlex−C5(Avestin、カナダ)を使用して実行した。押し出されたリポソームを限外濾過によって濃縮し、緩衝液をダイアフィルトレーション(10回交換)によってPBS pH7.4に交換した。得られたリポソームをPBS pH7.4で希釈し、30℃に加熱してからPal1−15を添加した。
テトラパルミトイル化ヒトペプチドPal1−15(Bachem AG、スイス)を1%β−OG(Sigma-Aldrich、USA)を含むMilliQ水に溶解した10mM Na2HPO4、pH 11.4に溶解し、30℃のリポソーム溶液に注入し、30分撹拌し、その後、限外濾過による濃縮ステップと、ダイアフィルトレーションによるPBS pH7.4での希釈を行った。次に、得られたリポソームを、0.2μmのポリエーテルスルホン(PES)メンブレンシリンジフィルターに通して滅菌ろ過し、5℃で保存した。
脂質(DMPG、DMPC、コレステロール、3D−(6−アシル)PHADTM(Avanti Polar Lipids、USA))を60℃の加熱キャビネット内で96%EtOHに溶解した。脂質が完全に溶解した後、溶液を孔径0.2μmのフィルターでろ過し、60℃に加熱した注入システムに入れた。詳細には、適切な量のACI−24.043(SAT47)を、超音波処理を利用して室温でEtOHに分散させた(EtOH濃度は、典型的には、最終SAT47溶液の2% v/vである)。ペプチドを完全に分散させた後、His−スクロースバッファー(10mMヒスチジン、250mMスクロース)を添加して、質量で1/50の脂質に対する薬剤比を達成した。SAT47溶液を0.2μmポアサイズフィルター(Sartoscaleフィルター)でろ過してインジェクションバッファーボトルに入れ、40℃に加熱した。リポソームは、脂質/エタノール溶液とインジェクションバッファーが混ざると、インジェクション部位で形成される。リポソーム形成の直後に、EtOH濃度を下げるために10mMヒスチジン、250mMスクロースによるオンライン希釈ステップを行った。中間体リポソームを、室温で孔径100nmのポリカーボネート膜(1パス)を通して押し出した。中空糸膜(MWCO:500kD)を使用した限外/ダイアフィルトレーション(UDF)を実行してEtOHを除去し、バッファーをPBS pH6.9に交換した(10倍の容量交換)。次に、分散バッファー(PBS pH6.9)を使用してSAT47リポソームを総脂質濃度1mg/mLに希釈し、35℃まで温めた。Pal1−15を10mM Na2HPO4 pH11.4バッファー中のベータOGの10% w/v溶液に60℃で溶解し、同じバッファーでさらに希釈して最終濃度を1mg/mLにした。pHを11.4に調整した。クロスフロー注入モジュールを使用してこれら2つの溶液を混合した後、リポソーム懸濁液をさらに35℃で30分間撹拌しながらインキュベートし、Pal1−15を完全に挿入した。中空糸膜(MWCO:500kD)を使用した2番目のUDFステップを実行して、ベータOGを除去し、バッファーを10mMヒスチジン、250mMスクロースに交換した(10倍の容量交換)。生成物を最終容量に濃縮し、0.2μmのAcrodisc mPESシリンジフィルターでろ過した。

異なるT細胞エピトープの封入が成功した後、ACI−24ワクチンと比較して高い免疫原性スコアSAT42、SAT44およびSAT47を有する封入されたT細胞エピトープを含むワクチン(それぞれACI−24.044、ACI−24.046およびACI−24.043ワクチン)の免疫原性を、インビボで試験した。野生型C57BL/6マウスに、ACI−24、ACI−24.044(封入されたSAT42を使用)、ACI−24.046(封入されたSAT44を使用)、およびACI−24.043(封入されたSAT47を使用)を、0、14、および28日目に、合計3回の皮下(sc)接種を行った。−21日目(ACI−24.046)または−7日目(ACI−24、ACI−24.043、ACI−24.044)(プレブリード(prebleed))、7、21、35日目に血液サンプルを採取し、Aβ1−42特異的IgG力価をELISAにより測定した。
4.1 ヒトAβ1−42自己会合のインビトロでの阻害
誘導されたAβ特異的抗体の品質を、Aβ1−42の自己会合/凝集の阻害を測定することによってインビトロでテストした。このアッセイは、マウスの接種前および接種後の血漿が、ヒトAβ1−42の自然な自己会合素因を損なう能力に基づく。
病理学的Aβに結合するC57BL/6マウスで誘導された抗体の特異性を評価するために、Aβ1−42−オリゴマー特異的IgG応答をELISAによって調べた。プレートを、前述のように4℃で一晩調製した10μg/mlのオリゴマーにてコーティングした(Adolfsson、2012)。0.05%ツイン20/PBSで洗浄し、1%BSA/0.05%ツイン20/PBSでブロッキングした後、血漿の系列希釈液をプレートに加え、37℃で2時間インキュベートした。洗浄後、プレートをアルカリホスファターゼ(AP)結合抗マウスIgG抗体(Jackson ImmunoResearch, カタログ番号115-055-164, PA, USA)とともに37℃で2時間インキュベートした。最終洗浄後、プレートをAP基質(pNPP)とともに2.5時間インキュベートし、ELISAプレートリーダーを使用して405nmで読み取った。結果を、市販の抗体(6E10, Biolegend, UK, カタログ番号803002)の系列希釈を参照して表す。

- 各ユニバーサルT細胞エピトープが、長さが30アミノ酸以下、長さが20アミノ酸以下、または長さが10〜20アミノ酸以下である、請求項1から3のいずれか一項に記載のリポソームワクチン組成物。

- リンカーが少なくとも2つのアミノ酸を含み、所望によりリンカーがアミノ酸VVRまたはPMGAPを含むか、本質的になるか、またはそれからなっていてもよい、請求項8に記載のリポソームワクチン組成物。
- ユニバーサルT細胞エピトープが以下から選択される、請求項1から9のいずれか一項に記載のリポソームワクチン組成物:
a.ジフテリア毒素と破傷風毒素のユニバーサルT細胞エピトープの組み合わせ
b.エプスタインバーウイルスと破傷風毒素のユニバーサルT細胞エピトープの組み合わせ
c.エプスタインバーウイルス、破傷風毒素、キーホールリンペットヘモシアニンのユニバーサルT細胞エピトープの組み合わせ;または
d.インフルエンザ血球凝集素、ジフテリア毒素、破傷風毒素、およびエプスタインバーウイルスのユニバーサルT細胞エピトープの組み合わせ。 - ユニバーサルT細胞エピトープを含むペプチドが、配列番号1(SAT42)、配列番号2(SAT43)、配列番号3(SAT44)、配列番号4(SAT47)から選択されるアミノ酸配列を含む、本質的になる、またはそれからなる、請求項1から10のいずれか一項に記載のリポソームワクチン組成物。
- ユニバーサルT細胞エピトープが、配列番号5(SAT6)、配列番号6(SAT13)、配列番号7(SAT15)、配列番号8(SAT17)から選択されるアミノ酸配列を含む、本質的になる、またはそれからなる、請求項1から10のいずれか一項に記載のリポソームワクチン組成物。
- 以下を含むリポソームワクチン組成物:
a.Aβのアミノ酸1〜15を含む、本質的になる、またはそれからなる、リポソームの表面に提示されるテトラパルミトリル化β−アミロイド(Aβ)由来ペプチド抗原
b.リポソーム内に封入されたユニバーサルT細胞エピトープを含むペプチドであって、配列番号3(SAT44)および配列番号4(SAT47)から選択されるアミノ酸配列を含む、本質的になる、またはそれからなるペプチド
c.アジュバント。 - アジュバントがリポソームの一部を形成する、請求項1から13のいずれか一項に記載のリポソームワクチン組成物。
- アジュバントが、少なくとも部分的に、リポソームの表面に表示される、請求項1から14のいずれか一項に記載のリポソームワクチン組成物。
- アジュバントがモノホスホリルリピドA(MPLA)を含む、請求項1から15のいずれか一項に記載のリポソームワクチン組成物。
- ユニバーサルT細胞エピトープを含むペプチドが30から60アミノ酸の長さである、請求項1から16のいずれか一項に記載のリポソームワクチン組成物。
- 対象におけるアミロイドベータ関連疾患または状態に関連する症状を治療、予防、該症状に対する防御免疫応答を誘導、あるいは該症状を軽減する際に使用するための、請求項1から17のいずれか一項に記載のワクチン組成物。
