特定の実施形態では、モノクローナル抗体はインビボで検証される。一般に、方法は、対象のモノクローナル抗体を疾患または状態のために動物モデルに投与し、モデルの動物の疾患または状態に対するモノクローナル抗体の効果を判定することを含む。本発明のインビボのアッセイはコントロールを含み、この場合適切なコントロールは、モノクローナル抗体の非存在下のサンプルを含む。一般に、複数のアッセイ混合物は、様々な濃度に対する異なる応答を得るために、異なる抗体の濃度で並行して実行される。典型的には、これらの濃度の1つが陰性のコントロールとして機能する。つまり、濃度がゼロまたは検出レベル未満である。

SynVH−SDニワトリの調製
この研究では、トランスジェニックニワトリを遺伝子操作して、ヒト可変領域抗体レパートリーを産生した。医薬品の観点から、ニワトリで単一のVフレームワークを使用すると、最適な製造および開発可能性の特性を有する好ましいフレームワークを選択できるため、利点がある。導入遺伝子は、生殖系列または生殖系列に近いフレームワーク配列を維持しながら、CDRに多様性が集中するように設計できる。戦略は、多様性を組み込むためのニワトリB細胞システムの利点を維持することであり、大きなヒトゲノム断片(複数のヒトV、D、およびJ遺伝子を保有する)を挿入する代わりに、上流のヒトベースの偽遺伝子を含む単一のヒトフレームワークを使用し、ニワトリでは適切に規制されていなくてもよい。単一のヒトのフレームワークがいずれかの潜在的な標的に治療候補を供給するためには、導入遺伝子によって産生される多様性のレベルが十分でなければならない。トランスジェニックニワトリの2つの系統が産生された。1つは多様性を生み出すために完全に遺伝子変換/体細胞超変異に依存する、事前再配置された機能的なVH領域を担持し、もう1つは機能的なVH領域を作り出すために最初にV(D)J再配置を経て、それによりCDR−H3の多様性のレベルを潜在的に増加させ、遺伝子変換/体細胞超変異が続いた。
事前再配置されるVH領域の構築物、SynVH−Cは、先に説明した(18)。V(D)J再配置構築物SynVH−SDは、いくつかの部分の遺伝子合成とそれに続くライゲーションによって調製された。V、D、J領域は次のようにして組み立てた。ヒト生殖系列VH3−23*01遺伝子が、使用された単一のV遺伝子であり、JH6遺伝子が、単一のJ遺伝子であった。組換えシグナル配列とニワトリD遺伝子座由来の介在領域(これらのスペーサーはそれぞれ約100〜200bp)に、24の非冗長のヒトDを隣接させた。ヒトD2ファミリーのシステインコドンは、チロシン(7インスタンス)またはトリプトファン(2インスタンス)をコードするように変異されていた。再配置する要素をニワトリVHプロモーターでクローン化して重鎖の発現を促進し、内因性定常領域に対するスプライシングのためニワトリJ−Cイントロンの短いセクションを含めた。ヒト偽遺伝子は、ヒト生殖系列VH3−23遺伝子から得たFRと、VH3生殖系列遺伝子ファミリーから得たCDR1および2を含んでいた。13の偽遺伝子が設計された。各偽遺伝子間のスペーサー配列は、ニワトリ偽遺伝子領域から得たものであったが、ニワトリV配列自体は含まれていなかった。ニワトリ重鎖遺伝子座を標的としたattP部位に挿入するためattB部位が含まれ(21、22)、後に標的ゲノムのloxP部位との組換えのためloxP部位が含まれていた。SynVH−SD構築物を重鎖attP含有細胞にトランスフェクトした。これらの細胞では、loxP部位は以前にCRISPR媒介ターゲティングによってニワトリVHセグメントの上流に挿入されていた(22)。SynVH−SD導入遺伝子の挿入後、第2のloxP部位がSynVH−SD導入遺伝子によって、第1のloxP部位と同じ向きで、ヒト偽遺伝子アレイのすぐ上流に導入された。Cre雌鶏に繁殖することで、loxP部位間のすべてのDNAが削除され、これには、ニワトリのVH遺伝子とD遺伝子、およびトランスフェクションの間に使用される選択可能マーカーが含まれ、図1に示す構造に至った。軽鎖については、この研究で使用されたすべてのトランスジェニックニワトリが、構築物SynVK−CKから得たヒトV−カッパ軽鎖を発現した(18)。これらのトリの軽鎖と重鎖は、ヒトの可変領域とニワトリの定常領域からなっていた。重鎖遺伝子座では、導入遺伝子はすべてのケースでヘテロ接合性であり、他の対立遺伝子がノックアウトされ、遺伝子型IgHSynVH/IgHJH−KOが得られた。軽鎖遺伝子座では、遺伝子型は常にIgLSynVK−CK/IgLVJC−KOであった。
2つの異なるヒト重鎖導入遺伝子を有するニワトリ由来のヒトVH配列を分析した。これらの導入遺伝子の1つであるSynVH−Cには、事前再配置された機能的なV領域(18)が含まれ、他方のSynVH−SDには、B細胞で再配置される生殖系列V、D、およびJセグメントが含まれていた(図1)。SynVH−CのV領域はヒトライブラリーのスクリーニングから取得され、再配置されたVH3−23/D1/JH4領域を含有しており、これは生殖系列VH3−23遺伝子に対する9つのフレームワーク(FR)の変化が伴っていた。SynVH−SD構築物には、単一の生殖系列V遺伝子、VH3−23、ヒトDエレメントすべて、および単一のJH6遺伝子が含まれていた。D要素は、高度に保存された組換えシグナル配列を含む、ニワトリD遺伝子座からの介在配列によって分離された。構築物双方の他のすべての非コード配列(プロモーターとイントロン)は、最適な転写および転写後の調節のためのニワトリ重鎖遺伝子座に由来した。導入遺伝子の構築物は、遺伝子ターゲティングとインテグラーゼを介した挿入の組み合わせを介して、内因性重鎖遺伝子座に挿入された(18、22)。両方の導入遺伝子のヒトV領域は、内因性の下流のニワトリ定常領域にスプライスしている。
6羽のSynVH−Cと3羽のSynVH−SDのトリから得たサンプルは、鳥の番号順に列挙されている。固有のヌクレオチドおよびタンパク質配列の数と、2回以上(2X深度)配列された固有のペプチド配列の数が示される。最後の列には、上位1000の配列で表される総配列数の読み取りデータの割合が提示されている。
9羽のトリのVH領域の分析は、発現された配列が予想どおり完全にヒト型であるか、任意のニワトリ型配列を含んでいるかどうかの判定から開始した(表2)。SynVH−SDのV領域はすべてヒト型の配列であった。これは、SynVH−SDの挿入の上流で内因性のニワトリV領域が削除されたためである。意外にも、SynVH−C導入遺伝子の配列の約5%がニワトリVH領域であった。これらの配列は、CDR3およびヒトJ配列に融合されたニワトリCDR1〜2およびFR1〜3で構成されていた。これ以外の可能性はない。なぜならば、トランスジェニックニワトリのゲノムに存在する唯一のJH配列はSynVH導入遺伝子のヒトJであるためである。CDR−H3はヒト型のように思われる、なぜならば、ニワトリCDR−H3にあると予想される非標準的なシステインがないためである(17)。SynVHの発現された抗体レパートリーにニワトリVH配列の2つの潜在的な源が存在していた。ニワトリ偽遺伝子からの遺伝子変換であり、これは依然ヒト偽遺伝子の上流に存在している。または二次再配置型メカニズムによるニワトリの機能的なVによるヒトの機能的なVの遺伝子置換であり、これにおいてニワトリV遺伝子はV遺伝子の3’末端における潜在的な組換えシグナル配列を介してヒトV遺伝子で再配置した(23〜26)。これらのニワトリV領域は、導入遺伝子のヒトFR3−CDR3接合部でインフレーム融合し、それによってヒトV遺伝子を削除する二次再配置に由来する可能性が最も高いようである。遺伝子変換よりも遺伝子置換を支持する主な証拠は、シグナルペプチド配列はまたニワトリであったが、偽遺伝子はシグナルペプチド配列を含まず、そのため、ヒトシグナルペプチドをニワトリシグナルペプチド配列に変異させることができなかったということである。さらに、遺伝子変換により、発現されるV領域をニワトリV領域に置き換えることができる場合、上流の偽遺伝子も含むSynVH−SD配列で、同じことが発生することが予想され得る。しかし、V置換はSynVH−SDで観察されなかった。機能的なニワトリVおよびDクラスターは、SynVH−SD導入遺伝子の上流で削除されているため、遺伝子置換の可能性はない。
ニワトリおよびヒトVH領域の数(および各導入遺伝子の全配列のパーセンテージ)が示されている。6のSynVH−Cサンプルと3のSynVH−SDサンプルから得たデータを組み合わせた(合計、SynVH−Cのトリから得た6000の配列およびSynVH−SDのトリから得た3000配列)。最初の列のセットには、ニワトリVHを含む配列が示され、ニワトリシグナルペプチド(Ch sig pep)またはヒトシグナルペプチド(Hu sig pep)を含む配列に分割されている。第2の列のセットには、シグナルペプチドの配列について同様に分割された、ヒトVHを有する配列がある。すべての配列にヒトJHがあった(右側の列)。いくつかの配列にVH欠失があったため、VH領域の総数は配列の総数よりもわずかに少なくなる。
ニワトリの偽遺伝子のアレイは、SynVH−CおよびSynVH−SDの導入遺伝子双方の上流に依然存在している(図1)。SynVH−SDでは、ニワトリの偽遺伝子は、ヒトの機能的なVに非常に近接していてヒトの偽遺伝子のすぐ上流にあるが、SynVH−Cでは、ニワトリのVおよびD遺伝子は、2つの偽遺伝子アレイの間にある。上記のように、FR1に隣接するシグナルペプチド配列はSynVH−SDで遺伝子変換を経ており、そのためニワトリ偽遺伝子によるヒトフレームワークのさらなる遺伝子変換が発生したかどうかを知ることは興味深いことであった。DNAレベルでは、ニワトリの生殖系列のVHセグメントは、導入遺伝子のV遺伝子と全体的に約65%同一である。相同性の最も長いストレッチは11bpであり、1〜6bpのミスマッチがV領域全体に広がっている。このレベルと相同性のパターンが遺伝子変換を可能にするのに十分であるかどうかは不明である。タンパク質の配列の変化につながる遺伝子変換の機能的に適切なレベルを判定するために、ニワトリ残基の置換の長いストレッチに関する証拠を求めてFR1とFR3をタンパク質レベルで分析した(FR2はニワトリとヒトの間で保存され過ぎているため、遺伝子変換事象を明確に検出することができない)。SynVH−CでのヒトFR1およびFR3の非常に低レベルの遺伝子変換(FR1で0.07%、FR3で0.3%、5652配列)、およびSynVH−SDでのヒトFR1およびFR3の非常に低レベルの遺伝子変換(FR1で2%、FR3ではなし)が観察された(表3)。遺伝子変換の最も一般的な例は、配列の約15%で発生したSynVH−SDのシグナルペプチドの変化であった。これは、配列変化が選択されている場合、遺伝子変換がより高い頻度で観察され得ることを示唆している。ヒトFRにおいてニワトリ偽遺伝子により遺伝子が変換される事象が低い頻度であることは、それらがまれな非選択事象であることを示唆している。SynVH−SDのFR1においてわずかに頻度が高い(2%)ことは、遺伝子の5’末端のシグナルペプチド領域で始まり、FR1に続く遺伝子変換事象が継続する結果である可能性がある。別の潜在可能性のある寄与因子は、ニワトリVおよびDクラスターがその導入遺伝子で削除されているため、SynVH−SDのニワトリ偽遺伝子に対してヒトの機能的なVの染色体において物理的近接性があることである可能性がある(図1)。これらのトリに由来する抗原特異的mAb配列(下記を参照)では、どの配列にもニワトリ由来のFR残基は見られず、これらのまれな遺伝子変換由来の配列は選択されていなかったか、抗原結合因子を産生する必要がなかったという考えを裏付けている。
各々のトリから得たヒト型配列で観察された遺伝子変換事象の数が提示されている。遺伝子変換は、ニワトリの配列である長い領域の配列(つまり、FR1全体)として定められる。ヌクレオチド配列ではなく、タンパク質の配列を分析した。分析されたヒト型配列の数は右に示されている。
ND:判定されていない。ニワトリFR2は、ヒト導入遺伝子と1つ(SynVH−SD)または2つ(SynVH−C)のアミノ酸のみが異なり、遺伝子変換を明確に評価することはできなかった。
*V領域のエクソンにおいてコードされたシグナルペプチドの部分とFR1の最初の3つのアミノ酸であり、最初のエクソンにおいてコードされた部分ではない。
CDR3の長さの多様性は、IMGTの位置105〜117の2つの遺伝子型で分析された。SynVH−C配列では、範囲は3〜22アミノ酸で、平均は11.56±2.06であった(図3)。SynVH−Cの生殖系列のCDR−H3の長さは、これが事前再配置されているため、11のコドンの固定した長さである。また、長さのいずれかの変化は、配列が削除または挿入された遺伝子変換または体細胞超変異の結果であり得る。55%が11残基よりも長い長さであるが、配列のわずか27%が11未満の長さであることから、これらのメカニズムはCDRの長さを増加させる可能性がより高い。導入遺伝子SynVH−SDの再配置では、CDR−H3の平均の長さがわずかにより長く、分布がより広い(11.89±2.68アミノ酸、範囲3〜21。どちらの長さのデータのセットも正規分布に適合しない)ものであった(図3)。長めのCDR−H3はSynVH−SDでより頻繁にあり、SynVH−Cの3.6%と比較すると、長さ15残基以上のCDRの23%であった。単一のDが使用されている場合、SynVH−SDの仮説上の長さの範囲は、コード配列のチューイングバックがない場合、16〜23コドンになる。これは、ほとんどの配列がV(D)J組換え中にチューイングバックすることまたは遺伝子変換/体細胞超変異から長さが減少することを示す。mAb配列が制限されており、1つの抗原に対してのみであるが、CDR−H3の長さは、SynVH−SDについてはより長い長さに偏っており、一方でSynVH−Cの頻度は、バルク配列と一致しているように思われる(図3)。
CDRのアミノ酸含有量をSynVH−CおよびSynVH−SDのデータセットで分析した(表4)。ニワトリV領域の配列は削除され、ヒトの配列のみ分析した。これらのデータセットは免疫されたトリに基づいているため、未処理のレパートリーを表すものではないが、2つの導入遺伝子によって産生されたレパートリーの特性は、免疫原であるヒトプログラニュリン(PGRN)がすべてのトリで同じであるため、互いに比較できる。CDR−H1およびH2(IMGT定義が使用された)の場合、長さに関係なく、すべての配列が含まれていた。

IMGTのCDR名称が使用された。CDR−H3の場合、12〜15残基のCDRの長さのアミノ酸の頻度が計算および平均化され、VおよびJの遺伝子によってコードされた残基が削除された。6羽のSynVH−Cのトリ(n=5652)および3羽のSynVH−SDのトリ(n=3000)から得たヒトの配列のみを分析した。
SynVH−CおよびSynVH−SDからの各CDR−H3(IMGTの位置105〜117)の平均疎水性は、アミノ酸疎水性の正規化されたKyte−Doolittleスケールに基づいて計算された(34、35)。長さ12〜15の残基のCDR−H3からのデータを組み合わせた(図6)。これらの値の平均は、SynVH−CとSynVH−SDの両方に対してスケールの親水性の側にあり、範囲はヒトとマウスのレパートリーに対して以前に報告されたものと同様に表れている(36〜38)。SynVH−SDのCDR−H3の平均値は、SynVH−Cと比較してわずかに疎水性にシフトしている(疎水性の値の平均の差は、2群で有意であった(対応のないKolmogorov−Smirnov検定、p<0.0001))。
低頻度の非標準的なシステイン残基が、SynVH−CおよびSynVH−SD配列のV領域全体に散在し、単一の不対のシステイン、またはジスルフィド架橋を形成する潜在可能性のあるシステインの対として見出された(表5)。合計123の不対の個々のシステインが、2つの導入遺伝子の8652のVH配列で見出された(表5)。これらの残基は、SynVH−CのFR1を除いて、CDR−H3を含むSynVH−CとSynVH−SDの両方のすべてのFRおよびCDRで見出された。不対のシステインの頻度は、WTニワトリで観察された頻度よりも低かった(SynVHとWTのCDR1、FR2、およびCDR2で、0.08%、0.13%、0.17%対2.1%、0.8%、2.4%(17))。
番号は、各々のトリから得た上位1000の固有の配列で、単一の不対のCys残基または2つの潜在的に対のCys残基を含む固有の配列の数を示す(ヒトV領域のみを分析した;SynVH−Cに対して合計n=5652、SynVH−SDに対してn=3000)。括弧内には、潜在的なループの配列が示されている。他の唯一の可能性のあるジスルフィドループは、FR2およびFR3にCys残基を含む単一のSynVH−C配列であった。FR2とFR4では、非標準のCysのすべてのインスタンスは、Trp−>Cysの変更のケースであり、単一のヌクレオチド置換によって発生する可能性がある。3つ以上の非標準システインのインスタンスは観察されなかった。
NGSのデータの源である脾臓リンパ球は、免疫化され、抗原特異的抗体の産生に使用されたトリに由来する。6羽のSynVH−Cおよび3羽のSynVH−SDのトリを、検証する免疫原であるヒトのプログラニュリン(PGRN)で免疫した。抗原特異的mAbのパネルは、GEMアッセイで脾臓の細胞をスクリーニングすることにより、各々のトリから特定された(18、39)。ELISAにより、mAbをPGRNへの結合として確認し、重鎖および軽鎖のV領域の配列が得られた。これらの全トリの軽鎖は、ヒトV−カッパ導入遺伝子によってもたらされた(18)。トリは他の対立遺伝子に軽鎖と重鎖のノックアウトがあり、重鎖と軽鎖の両方の導入遺伝子がヘテロ接合性であった。したがって、トリにおいてヒトV領域抗体のみが産生されている。
ニワトリの遺伝子変換/体細胞超変異システムは、宿主に防御免疫を与えることができる多様なレパートリーを生み出すように発展した。組み合わせたメカニズムに由来する多様性の欠如は、CDR−H3レパートリーの制限とは思われない。ヒト型可変領域をコードする2つの導入遺伝子が導入されたとき、一方は再配置ができないが、遺伝子変換および/または体細胞超変異によってその配列多様性を純粋に生じることを余儀なくされ、他方は再配置を経て、すべてのヒトDを選択して組み合わせの多様性を作りだしたが、アミノ酸の多様性のレベルは類似していた。データセットは免疫化されたトリからのものであるため、免疫化からの特定の残基にいくらかの偏りがある可能性があるが、2つの導入遺伝子は、異なる多様性の生成の様式であるにもかかわらず、同様に振る舞った。CDR1と2、およびFRでは、アミノ酸の頻度を両方の導入遺伝子のヒト偽遺伝子に見出される残基に関連付けることができたが、CDR3の場合はできなかった。
VDJノックアウトの調製
ガイドRNAの設計:ニワトリの機能的重鎖Vの上流159bp領域をガイドRNAの設計のためにMITサーバーで分析した。4つのガイドRNAが選択され、合成されて、以下の野生型Cas9ヌクレアーゼ(Horizon)を含むGE6ベクターに個別にクローニングされた:gRNA1、AAATCATTAATCAACCCGAC(配列番号43);gRNA2、AACACGACTCCGGGCCTAGA(配列番号44);gRNA3、TGATTAATTGGGCGCCCGTC(配列番号45);gRNA4、ATTTAATGGCCGTCTAGGCC(配列番号46)。これらのgRNAには、予測される標的外の部位はほとんどなかった。いずれも公知のコード配列に存在していなかった。EGFPに特異的なgRNA5を含むコントロール構築物も調製した(AAGTTCGAGGGCGACACCC(配列番号47))。

Claims (27)

(ii)FR1、CDR1、FR2、CDR2、およびFR3配列を含む可変ドメインのコード配列を含む生殖系列ヒトVHセグメント、
(iii)ヒトDクラスター、
(iv)単一のヒトJセグメント、
(v)定常領域をコードする、(b)(ii)の前記生殖系列ヒトVHセグメントの上流の複数の配列、
(vi)それぞれが(b)(ii)の前記機能的なヒトVHセグメントと実質的に同じFR1、FR2およびFR3配列と、偽遺伝子ごとに異なるCDR1およびCDR2配列とを含む構造FR1−CDR1−FR2−CDR2−FR3の複数の偽遺伝子
を含み、
前記改変されたIgH遺伝子座は、前記ニワトリでV(D)J組換えを経て、
前記複数の生殖系列ヒトVH偽遺伝子は、V(D)J組換え後、遺伝子変換によってヌクレオチド配列を前記生殖系列ヒトVHセグメントに供与し、
前記ニワトリは多様な免疫グロブリン重鎖を含む抗体を産生する、
トランスジェニックニワトリ。 - (b)(vi)の前記CDR1およびCDR2の配列が、(b)(ii)の前記機能的なヒトVHセグメントと同じファミリーにある異なるヒトVHセグメントのCDR1およびCDR2をコードする、請求項1に記載のトランスジェニックニワトリ。
- (a)の隣接する内因性ニワトリV−D−J領域全体の長さが、10〜12kbの範囲である、請求項1または2のいずれかに記載のトランスジェニックニワトリ。
- (b)(i)の前記プロモーターが、ニワトリ免疫グロブリン重鎖遺伝子プロモーターである、請求項1から3のいずれかに記載のトランスジェニックニワトリ。
- (b)(ii)の前記生殖系列ヒトVHセグメントならびに(vi)の前記CDR1およびCDR2配列が、前記VH3ファミリー、前記VH1ファミリーまたは前記VH4ファミリーに由来する、請求項1から4のいずれかに記載のトランスジェニックニワトリ。
- 前記Dクラスターにおいて、システインの任意のコドンが、別のアミノ酸をコードするように変異されている、請求項1から5のいずれかに記載のトランスジェニックニワトリ。
- 前記Dクラスターにおいて、システインの任意のコドンが変異して、チロシンまたはトリプトファンをコードする、請求項6に記載のトランスジェニックニワトリ。
- 前記Dクラスターの前記介在配列がニワトリ由来である、請求項1から7のいずれかに記載のトランスジェニックニワトリ。
- (b)(v)の前記複数の偽遺伝子が少なくとも10の前記偽遺伝子を含む、請求項1から8のいずれかに記載のトランスジェニックニワトリ。
- (b)(v)の前記偽遺伝子が、(b)(ii)の前記生殖系列ヒトVHセグメントに対して逆向きである、請求項1から9のいずれかに記載のトランスジェニックニワトリ。
- (b)(v)の定常領域をコードする前記複数の配列が前記ニワトリにとって内因性である、請求項1から10のいずれかに記載のトランスジェニックニワトリ。
- 前記改変された免疫グロブリン重鎖(IgH)遺伝子座の前記転写産物が、前記Jコード配列のコピーの3’末端を(b)(v)の前記定常領域コード配列のコピーの5’末端に結合するイントロンを含む、請求項1から11のいずれかに記載のトランスジェニックニワトリ。
- 前記ニワトリが、前記改変されたIgH遺伝子座についてホモ接合性である、請求項1から12のいずれかに記載のトランスジェニックニワトリ。
- 前記ニワトリが前記改変されたIgH遺伝子座についてヘテロ接合性であり、前記相同染色体の前記IgH遺伝子座がノックアウトされている、請求項1から13のいずれかに記載のトランスジェニックニワトリ。
- 前記ニワトリが前記改変されたIgH遺伝子座についてヘテロ接合性であり、前記相同染色体の前記IgH遺伝子座が野生型である、請求項1から14のいずれかに記載のトランスジェニックニワトリ。
- 前記ニワトリが前記改変されたIgH遺伝子座についてヘテロ接合性であり、前記相同染色体の前記IgH遺伝子座がその隣接する内因性ニワトリV−D−J領域全体を欠く、請求項14に記載のトランスジェニックニワトリ。
- 前記ニワトリが前記改変されたIgH遺伝子座についてヘテロ接合性であり、前記相同染色体の前記IgH遺伝子座がJ領域を欠く、請求項14に記載のトランスジェニックニワトリ。
- 前記ニワトリによって産生された前記抗体が、前記重鎖CDR1、CDR2およびCDR3領域において多様化されている、請求項1から17のいずれかに記載のトランスジェニックニワトリ。
- その相同染色体の一方または両方においてその隣接する内因性ニワトリV−D−J領域全体を欠く、トランスジェニックニワトリ。
- 請求項1から19のいずれかに記載の前記トランスジェニックニワトリに由来するB細胞。
- (a)請求項1から20のいずれかに記載のトランスジェニックニワトリを抗原で免疫する工程、および
(b)前記ニワトリから、前記抗原に特異的に結合する抗体を得る工程
を含む方法。 - 前記抗体がポリクローナルである、請求項21に記載の方法。
- 前記抗体がモノクローナルである、請求項21に記載の方法。
- (c)前記トランスジェニックニワトリのB細胞を使用してハイブリドーマを調製する工程、および
(d)前記ハイブリドーマをスクリーニングして、前記抗原に特異的に結合する抗体を産生するハイブリドーマを特定する工程
をさらに含む、請求項21から23のいずれかに記載の方法。 - PCRを使用して、前記トランスジェニック動物のB細胞からの前記重鎖および軽鎖可変領域をコードする核酸を増幅し、前記増幅された核酸を使用して組換え抗体を発現させる工程をさらに含む、請求項21から24のいずれかに記載の方法。
- 請求項1から19のいずれかに記載のトランスジェニックニワトリによって産生される抗体。
- 方法であって
(a)前記隣接する内因性ニワトリV−D−J領域全体をニワトリの前記免疫グロブリン重鎖(IgH)遺伝子座から削除する工程、および
(b)構築物を前記遺伝子座に挿入する工程を含み、前記構築物が、
(i)免疫グロブリン重鎖遺伝子プロモーター、
(ii)FR1、CDR1、FR2、CDR2、およびFR3配列を含む可変ドメインのコード配列を含む生殖系列ヒトVHセグメント、
(iii)ヒトDクラスター、
(iv)ヒトJセグメント、および
(vi)それぞれが(b)(ii)の前記機能的なヒトVHセグメントと実質的に同じFR1、FR2およびFR3配列と、偽遺伝子ごとに異なるCDR1およびCDR2コード配列とを含む構造FR1−CDR1−FR2−CDR2−FR3の複数の偽遺伝子
