IgE EMPD関連ペプチドの合成及びその製剤の調製
a.IgE EMPD関連ペプチドの合成
IgE EMPDペプチド免疫原構築物の開発成果に含まれる、デザイナーIgE EMPD関連ペプチドの合成方法について記載する。血清学アッセイ、実験室試験研究及び現場研究に有用な小スケール量、ならびに医薬組成物の工業/商業生産に有用な大スケール(キログラム)量でペプチドを合成した。有効なIgE系アレルギーワクチンの使用に最適なペプチド構築物のスクリーニング及び選択のために、長さ約20〜約70アミノ酸の配列を有する広いレパートリーのIgE EMPD関連抗原性ペプチドを設計した。
油中水型エマルション及び無機塩との懸濁液を採用する製剤を調製した。大規模集団によって使用され、かつ予防も投与の目的の一部である医薬組成物を設計するには、安全性も考慮すべき重要な要素になる。臨床試験では医薬組成物の多くで油中水型エマルションをヒトに使用しているが、未だ安全性の理由からミョウバンが医薬組成物で使用される主要アジュバントである。したがって、臨床用途向け調製において、アジュバントとしてミョウバンまたはその無機塩ADJUPHOS(リン酸アルミニウム)が頻繁に使用される。
血清学アッセイ及び試薬
合成ペプチド構築物及びその製剤の機能的免疫原性を評価するための血清学アッセイ及び試薬を以下に詳細に記載する。
以下の実施例に記載の免疫血清試料を評価するためのELISAアッセイを開発した。これを以下で説明する。pH9.5の10mM NaHCO3緩衝液中(特に記載がない限り)2μg/mL(特に記載がない限り)の標的ペプチドIgE EMPD1−52、IgE EMPD1−39、IgE EMPD1−17、IgE EMPD19−38、IgE EMPD7−40など(例えば、配列番号2及び5〜8)100μLにより、96ウェルプレートのウェルを37℃で1時間、個別に被覆した。
pH9.5の10mM NaHCO3緩衝液中(特に記載がない限り)2μg/mL(特に記載がない限り)のThペプチド100μLにより、96ウェルELISAプレートのウェルを、類似するELISA法において37℃で1時間個別に被覆し、上記の通り実施した。種々のIgE EMPDペプチドワクチン製剤を与えたワクチン接種動物の抗体価の決定では、1:100〜1:10,000の10倍段階希釈の血清を試験し、Log10で表される試験血清の力価を、カットオフA450を0.5に設定したA450の線形回帰分析によって算出した。
免疫された宿主における抗IgE EMPD抗体の精密特異性分析は、エピトープマッピングによって決定した。簡潔には、96ウェルプレートのウェルを、ウェルごとに0.1mLあたり0.5μgのIgE EMPD 10merペプチド(配列番号10〜58)で個別に被覆した後、100μLの血清試料(PBSで1:100に希釈)を上記の抗体ELISA法の工程に従って2連の10merプレートウェル中でインキュベートした。IgE EMPDペプチド免疫原構築物のB細胞エピトープ、及び免疫された宿主の免疫血清からの抗IgE EMPD抗体の関連する精密特異性分析を、スペーサーまたはTh配列(配列番号1)のない対応するIgE EMPD1−52ペプチド、または関連性のない対照ペプチドで試験し、追加の反応性と特異性を確認した。
実験的ワクチン接種プロトコールに従って、免疫前及び免疫血清試料を動物対象もしくはヒト対象または動物から採取し、56℃で30分間加熱して血清補体因子を不活化した。ワクチン製剤の投与後、プロトコールに従って血液試料を得て、特異的標的部位(複数可)に対する免疫原性を評価した。段階希釈した血清を試験し、陽性力価を希釈度の逆数のLog10で表した。標的抗原内の目的のB細胞エピトープ配列を指向する高力価抗体を誘発すると同時に、望ましいB細胞応答の増強をもたらすために採用されるヘルパーT細胞エピトープ配列に対する抗体反応性を低度から無視できる程度に維持する能力によって、個々のワクチン製剤の免疫原性を評価した。
捕捉抗体である抗ヒトIgE、HP6061(Abcam)と検出抗体であるビオチン標識抗ヒトIgE、HP6029(Abcam)を使用したサンドイッチELISAでhuIGHEノックインマウスのヒトIgEレベルを測定した。簡潔には、HP6061を被覆用緩衝液(15mM Na2CO3、35mM NaHCO3、pH9.6)中100ng/ウェルで96ウェルプレートに固定し、4℃で一晩インキュベートした。被覆したウェルを200μL/ウェルのアッセイ用希釈液(PBS中0.5%のBSA、0.05%のTween−20、0.02%のProClin 300)で室温にて1時間ブロッキングした。プレートを200μL/ウェルの洗浄用緩衝液(0.05%のTween−20を含むPBS)で3回洗浄した。精製したU266 IgEを使用して、5%マウス血清を含むアッセイ用希釈液における標準曲線(2倍段階希釈で0〜800ng/mLの範囲)を作成した。50μLの希釈血清(1:20)と標準物質を被覆したウェルに加えた。室温で1時間インキュベートを実施した。全てのウェルを吸引し、200μL/ウェルの洗浄用緩衝液で6回洗浄した。捕捉されたヒトIgEを100μLの検出抗体溶液(アッセイ用希釈液中50ng/mlのビオチン標識HP6029)と共に室温で1時間インキュベートした。その後、結合したビオチン−HP6029を、ストレプトアビジンポリ−HRP(1:10,000希釈、Thermo Pierce)を使用して1時間検出した(100μL/ウェル)。全てのウェルを吸引し、200μL/ウェルの洗浄用緩衝液で6回洗浄した。最後に、ウェルを100μL/ウェルのNeA−Blue TMB基質(Clinical Scientific Products)で発色させ、100μL/ウェルの1M H2SO4の添加によって反応を停止させた。SoftMax Proソフトウェア(Molecular Devices)を使用して、4パラメータロジスティック曲線近似を生成することにより標準曲線を作成し、試験した全試料のIgE濃度を算出するために使用した。Prismソフトウェアを使用したデータ比較にはスチューデントのt検定を使用した。

動物におけるIgE EMPDペプチド免疫原構築物及びその製剤によって誘発される抗体の機能的特性の評価
免疫された宿主からの免疫血清または精製抗IgE EMPD抗体を、組換え可溶性IgE EMPDタンパク質への結合能、及びmIgE.FcL(CH2からCMまで、EMPDを含む)またはmIgE.FcS(CH2からCMまで、EMPDなし)のいずれかをコードする組換えDNAをトランスフェクトしたRamos細胞株への結合能について試験した。
Ramos細胞株は、American Type Culture Collection(ATCC、Manassas,VA)から購入し、10%の加熱不活化FBS(Invitrogen)、4mMのL−グルタミン、25mMのHEPES、及び1mMのピルビン酸ナトリウム(Invitrogen;完全RPMI培地)を補充したRPMI1640培地(Invitrogen,Carlsbad,CA)中で増殖させた。Ramos細胞に、mIgE.FcLまたはmIgE.FcSをコードする組換えDNAをトランスフェクトした。mIgE.FcLを発現するRamos細胞に、EMPDを含めた、CH2から細胞質ペプチドに及ぶmIgEε鎖である長鎖アイソフォームのセグメントをコードするDNAセグメントをトランスフェクトした。mIgE.FcSを発現するRamos細胞に、EMPDを除いた、CH2から細胞質ペプチドに及ぶmIgEε鎖である短鎖すなわち通常アイソフォームのセグメントをコードするDNAセグメントをトランスフェクトした。mIgE.FcLまたはmIgE.FcSを発現するRamos細胞の安定なトランスフェクタントを、400mg/mlのZeocin(Invitrogen)を補充した完全なRMPI1640培地中に維持した。
組換え可溶性IgE EMPDタンパク質を発現するFlp−In CHO細胞に、ヒトIgG1のFc部分及びヒト膜結合型IgEのIgE EMPD、γ1−em67をコードするDNAセグメントをトランスフェクトした。安定なFlp−In CHOトランスフェクタントを、10%の加熱不活化FBS(Invitrogen)、4mMのL−グルタミン、25mMのHEPES、及び1mMのピルビン酸ナトリウム(Invitrogen;完全IMDM培地)を補充したIMDM培地(Invitrogen,Carlsbad,CA)中に維持した。γ1−em67タンパク質を、製造業者の指示に従い、プロテインAセファロース(GE Healthcare)を使用して培養培地から精製した。
種々の免疫血清からのポリクローナルIgGを、製造業者の指示に従い、プロテインAセファロース(GE Healthcare)を使用して精製した。
免疫された各動物からの精製ポリクローナル抗体が(a)組換えγ1−em67タンパク質(上記)に結合する相対活性をELISAによって、または(b)mIgE.FcLを発現するRamos細胞に結合する相対活性を蛍光フローサイトメトリー分析によって調べた。96ウェルマイクロプレートはNalge NUNC International製であり、光学測定用は平底(Cat.442404)、細胞インキュベート用はV底(Cat.249570)を使用した。光学密度をVersaMaxマイクロプレートリーダー(Molecular Devices)で読み取った。蛍光染色剤をBD FACSCanto IIサイトメーター(DB Biosciences)で検出し、結果データを付属のFACSDivaソフトウェアで取得した。ELISAとFACSからの結合データをPrism 6ソフトウェアにインポートして定量分析を行った。より具体的には、ウェルあたり0.1mL中2×105細胞のアリコートをV底マイクロプレートに加え、遠心分離し、液体を廃棄した。種々の濃度の抗体試料100μLアリコートと共に細胞を氷上で1時間インキュベートした。細胞を1回洗浄し、300gで5分間遠心分離し、100μLのヤギF(ab)2抗種特異的IgG Fc−FITC(250ng/mL)と共に氷上で30分間染色した。細胞を1回洗浄し、遠心分離後に液体を廃棄した。各ウェルに200μLアリコートの結合用緩衝液を加え、微量希釈チューブに移し、フローサイトメトリー分析を行った。試料あたり10,000細胞の注入量を基準として、結合強度(蛍光強度の幾何平均、GeoMFI)をFACSで読み取った。
mIgE.FcL(5×105細胞/mL)を安定的に発現するRamos細胞を、完全RPMI1640培地中の精製免疫抗体または対照抗体と共に37℃で1時間インキュベートした。次に、最終濃度10μg/mLの二次抗体、モルモットIgGのFcに特異的なヤギF(ab´)2(Jackson ImmunoResearch Laboratories,West Grove,PA)で細胞を処理し、37℃でさらに24時間インキュベートした。細胞のアポトーシスの程度を以下の方法で分析した。アネキシンVを用いたアッセイでは、細胞を染色溶液に15分間、暗所、室温で再懸濁することでホスファチジルセリン(PS)の曝露を測定した。染色液には、1/200に希釈したFITC標識アネキシンV(Biovision,Mountain View,CA)、ならびに10mMのHEPES/NaOH(pH7.4)、140mMのNaCl、及び5mMのCaCl2を含む緩衝液中2.5μg/mlのヨウ化プロピジウム(PI)が含有されてた。染色細胞をFACSCanto IIフローサイトメーター(BD Biosciences,San Jose,CA)で分析した。アネキシンV陽性及びPI陰性と定義されるアポトーシス細胞の比率をドットプロット分析で取得した。
RBC溶解用緩衝液(Thermo Fisher Scientific Inc.)を使用して、赤血球の低浸透圧ショックを繰り返すことにより、Balb/cマウス(雌、6〜8週齢)の脾臓から脾臓リンパ球を単離した。赤血球を除去した後、脾臓リンパ球を3×106細胞/mLで、50μMの2−ME及び100U/mLの組換えヒトIL−2(PeproTech,Inc)を補充した完全RPMI培地中にて3日間培養した。mIgE.FcLを発現するRamos細胞(標的細胞)をPBS/0.1%BSA中のCFSE(Invitrogen)により37℃で10分間標識した。低温完全RPMI1640培地で3回洗浄した後、105細胞/mLに細胞を調整した。完全RPMI培地200μl中標識細胞20,000個のアリコートを、対応する免疫血清から精製したポリクローナルIgG抗体10μg/mLにより37℃で30分間被覆した後、IL−2活性化脾臓リンパ球(エフェクター細胞)とE/T比30で混合した。24時間のインキュベート後、全細胞を2.5μg/mLの7アミノアクチノマイシンD(7−AAD,Invitrogen)により氷上で15分間染色した後、Becton Dickinson FACS Canto IIフローサイトメーター(BD Biosciences)で分析した。生存標的細胞は、ドットプロット分析でCFSE陽性及び7−AAD陰性と定義された。所定のE/T比での溶解標的細胞の比率は、100×[(抗体非依存性対照中の生存標的細胞の比率−試料中の生存標的細胞の比率)/抗体非依存性対照中の生存標的細胞の比率]であった。
安全性試験、免疫原性試験、毒性試験、及び有効性試験に使用する動物
a.モルモット:
雌雄の成熟ナイーブ成体Duncan−Hartleyモルモット(300〜350g/BW)で免疫原性試験を実施した。実験には群あたり少なくとも3匹のモルモットを使用した。Duncan−Hartleyモルモット(8〜12週齢;Covance Research Laboratories,Denver,PA,USA)に関わるプロトコールを、IACUC申請の承認下で、委託動物施設及び責任者であるUnited Biomedical,Inc.(UBI)で実施した。
雌雄の成体サル(Macaca fascicularis、約4齢;Joinn Laboratories,Suzhou,China)における免疫原性試験及び反復投与毒性試験を、IACUC申請の承認下で、委託動物施設及び責任者であるUBIで実施した。
C57BL/B6遺伝的背景の相同遺伝子ターゲティングにより、IGHG1遺伝子をヒトIGHE遺伝子に置換したマウス系統は、ヒトの分泌型及び膜結合型のIgEを発現する(Lu,el al.,2015)。hIGHEマウスは、マウスIGHG1転写要素の調節制御下でヒトIgEを発現し、ヒト調節要素の選択的RNAスプライシングによってヒト膜結合型IgEを発現する。8〜10週齢という早期に、血液中に血清IgEが検出された。一次/記憶免疫応答の予防モデル、及び感作/リコール免疫応答の治療モデルには、混合hIGHE×Balb/c背景の幼若仔(10〜12週齢)を使用した。いずれの試験も、IACUC申請の承認下で、委託動物施設(National Health Research Institute,Taiwan)及び責任者であるUBIで実施した。
モルモット、トランスジェニックノックインマウス、及びカニクイザルにおける、IgE EMPDペプチド構築物の最終製品を選択する免疫原性評価のためのワクチン製剤
各実験に使用される医薬組成物及びワクチン製剤について以下でさらに詳細に説明する。簡潔には、各試験群に指定された製剤は一般に、種類の異なるスペーサー(例えば、ペプチド構築物の溶解度を増強するεKまたはKKK)によってIgE EMPDペプチドのセグメントが結合されたあらゆる種類のデザイナーIgE EMPDペプチド構築物と、デザイナーペプチド構築物のN末端側でIgE EMPDペプチドセグメント(複数可)と結合される麻疹ウイルス融合タンパク質及びB型肝炎表面抗原に由来する2組の人工Tヘルパーエピトープを含む無差別的ヘルパーT細胞エピトープの変形とを含んでいた。100超のデザイナーIgE EMPDペプチド構築物を最初にモルモットでIgE EMPD1−52との相対免疫原性について、さらにIgEを保持するB細胞(Ramos細胞株)上の膜IgEとの交差反応性について評価した。IgE EMPDペプチド構築物は、指定したような可変量のペプチド構築物で、ヒトワクチン用途の承認済み油であるSeppic Montanide(商標)ISA 51との油中水型エマルションに調製するか、または無機塩ADJUPHOSまたはALHYDROGEL(ミョウバン)と混合して調製した。ワクチンは通常、IgE EMPDペプチド構築物を約20〜800μg/mLの水に溶解することにより調製し、Montanide(商標)ISA 51を用いて油中水型エマルション(1:1体積)に製剤化するか、または無機塩ADJUPHOSもしくはALHYDROGEL(ミョウバン)(1:1体積)と製剤化した。ワクチン製剤を室温で約30分間維持し、免疫前にボルテックスで約10〜15秒間混合した。
IgE媒介型アレルギー性疾患の治療のためのIgE EMPD1−39ペプチド免疫原構築物を組み込んだ多成分ワクチン製剤の設計原理、スクリーニング、同定、機能的特性の評価、及び最適化
図2A及び図2Bは、IgE EMPDの標的化によりmIgE B細胞が減少する原理を示している。IgEは、分泌型IgEと膜結合型IgE(mIgE)という2つの形態で表される(図2A、それぞれ左と右)。分泌型IgEは、FcRIによって好塩基球及びマスト細胞の細胞表面に捕捉されるが、mIgEはB細胞受容体(BCR)の一部としてIgE関与型B細胞にのみ存在する。mIgEの細胞外膜近位ドメイン(EMPD)は、mIgE B細胞にのみ存在する、CH4ドメインと膜貫通領域との間の67アミノ酸のペプチドセグメント(配列番号1)である。IgE EMPDの独自性により、mIgEを保持するB細胞を標的とする誘引部位が与えられた。IgE EMPDの標的化によるmIgE B細胞の減少により、新しいIgE分泌型形質細胞へと分化する前に、アレルゲン特異的IgE生成の抑制が可能になる(図2B)。生存期間が限定される既存のIgE分泌型形質細胞は徐々に消滅し、その結果、総IgE及びアレルゲン特異的IgEは徐々に低下する。
各ペプチド免疫原構築物または免疫療法生成物には、その特異的疾患メカニズム及び治療介入に必要となる標的タンパク質(複数可)に基づいた固有の設計重点と手法が必要とされる。設計の判断材料とする標的には、疾患経路に関与する細胞タンパク質、または病原体に由来するいくつかのタンパク質が関与し得る感染作用物質を含み得る。研究から商品化までの過程は非常に長期にわたり、通常は完了までに10年以上を要する。
医薬組成物に組み込むための最も強力なペプチド構築物を生成するには、IgE EMPD B細胞エピトープペプチド(配列番号5〜8)(表1)、及び種々の病原体に由来する無差別Tヘルパーエピトープまたはさらに設計された人工Tヘルパーエピトープ(配列番号59〜87)(表2)の広いレパートリーから、モルモットにおける免疫原性試験のためのIgE EMPDペプチド免疫原構築物を作製した。
IgEは、分泌型IgEと膜結合型IgE(mIgE)という2つの形態で表される。分泌型IgEは、FcRIによって好塩基球及びマスト細胞の細胞表面に捕捉されるが、mIgEはB細胞受容体(BCR)の一部としてIgE関与型B細胞にのみ存在する。mIgEの完全長細胞外膜近位ドメイン(EMPD)は、mIgE B細胞にのみ存在する、CH4ドメインと膜貫通領域との間の67アミノ酸のペプチドセグメント(配列番号1)である。生存期間が限定される既存のIgE分泌型形質細胞は徐々に消滅し、その結果、総IgE及びアレルゲン特異的IgEは徐々に低下する。モルモットの免疫原性について試験した多数のペプチド免疫原構築物のうち、一連のIgE EMPD由来ペプチド免疫原構築物(配列番号88〜93)を作製して、IgE EMPD1−52(配列番号2)に由来する代表的なIgE EMPD B細胞エピトープペプチドと、代表的なThエピトープペプチドUBITh(登録商標)1(配列番号72)とを組み込み、表4に示す、プレート被覆したIgE EMPD1−39(配列番号5)ペプチドを使用してモルモットにおける免疫原性試験を行った。3方向の6種のIgE EMPDペプチド免疫原構築物で高免疫原性が認められた。そのうち、UBITh(登録商標)1 Thエピトープペプチドは、C末端側(配列番号88及び91)もしくはN末端側(配列番号89、90、及び92)のいずれかで、またはスペーサーリンカーのεKをもつIgE EMPD B細胞エピトープペプチドのC末端側とN末端側の両方で(配列番号93)IgE EMPD B細胞エピトープペプチドに結合されている。構築物設計において高免疫原性を可能にするために、より長鎖のリンカーεK−KKK(配列番号129)を含むIgE EMPDペプチド免疫原構築物も採用した(例えば配列番号94〜97)。
表2には、マウス、ラット、モルモット、ヒヒ、マカクなどからの多種において、B細胞エピトープの免疫原性を増強する相対力価について試験した合計29の異種Thエピトープ(配列番号59〜87)が列挙されている。
図6A〜6Cは、IgE EMPD免疫原構築物(配列番号88〜97)で免疫した動物群ごとにプールした、モルモット血清からの精製ポリクローナル抗体による、mIgE.FcLまたはmIgE.FcSを発現するRamos細胞株に由来するB細胞への結合を示している(サイトフルログラフ染色の方法については、実施例2を参照)。プロテインAクロマトグラフィーによってモルモット血清から精製したポリクローナル抗IgE EMPD抗体を10μg/mLで使用した。
図7は、mIgE.FcLを発現するRamos細胞の細胞表面上で、用量依存的にIgE EMPD免疫原構築物(配列番号88〜93)で産出されるポリクローナル抗IgE EMPD抗体の種々の調製物によりアポトーシスが誘導されることを示す。プロテインAクロマトグラフィーによってモルモット血清から精製したポリクローナル抗IgE EMPD抗体を2倍段階希釈により1000から62.5ng/mLまで希釈した。ヒト化抗IgEモノクローナル抗体であるXolair(登録商標)を陽性対照として使用した。ポリクローナル抗IgE EMPD抗体の各組のEC50を、4パラメータロジスティック曲線近似による非線形回帰によって算出した。
図8は、IgE EMPDの異なる免疫原構築物(配列番号88〜93)によって生成されるポリクローナル抗IgE EMPD抗体の種々の調製物が、エフェクター/標的比30で、mIgE.FcLを発現するRamos細胞に対してADCCを誘導できたことを示している。プロテインAクロマトグラフィーによってモルモット血清から精製したポリクローナル抗IgE EMPD抗体を10μg/mLで使用した。IL−2刺激マウス脾臓細胞をエフェクター細胞として使用した。マウス抗IgEモノクローナル抗体5D5分泌型を陽性対照として使用した。
多様な遺伝的背景の患者を治療するための医薬組成物を設計する場合、多様な遺伝的背景をもつ最大限の集団を対象範囲とする設計が可能であることが重要である。MVFまたはHBsAgに由来する無差別Tヘルパーエピトープは、このような免疫原性の増強を提供する最も強力なものの一つであるため、このような2つのヘルパーTエピトープを含むペプチド構築物の組合せを設計することで、相乗的な免疫原性効果が可能になる。同じB細胞エピトープをもつ2つのペプチド免疫原構築物の混合物は、それぞれの個々のペプチド構築物によって誘発される免疫応答と比較した場合、相当な免疫応答を誘発すると予測される。
IgE EMPDペプチド免疫原構築物を含むIgE−EMPDワクチンの設計は、機能的及び構造的標的である、IgE−EMPDの中央領域にあるC18−C39の特別なループ構造を中心とした。この構造に基づく設計は、免疫原性標的として天然細胞外ループ構造を保持することを目的としている。
IgE EMPDペプチド免疫原構築物のB細胞エピトープを限定的に指向することが認められた集中型抗体反応
標的B細胞エピトープペプチドのそれぞれの担体タンパク質への化学的結合によって、そのようなB細胞エピトープペプチドを指向する免疫応答を増強するために使用される担体タンパク質(例えば、キーホールリンペットヘモシアニン(KLH)またはジフテリアトキソイド(DT)及び破傷風トキソイド(TT)タンパク質などの他の担体タンパク質)はすべて、免疫される宿主において、増強する担体タンパク質を指向する抗体の90%超を誘発し、標的B細胞エピトープを指向する抗体の10%未満を誘発することはよく知られている。
予防的モデルと治療的モデル両方の免疫療法アレルギーワクチンが遺伝子改変ノックインマウスの血清IgEレベルに与える効果
HuIGHE×Balb/cノックインマウスのF1子孫である、ヒトIgEを発現する遺伝子操作マウスを使用した概念実証試験において、配列番号88及び配列番号93のIgE EMPDペプチド免疫原構築物を選択し、一次及び二次/記憶応答でのIgE生成を評価した。
カニクイザルにおけるプロトタイプの免疫療法アレルギーワクチン製剤の免疫原性評価による用量及び製剤試験
a.全体目標:
本試験の目的は、選択したIgE EMPDペプチド免疫原構築物、配列番号88による筋肉内免疫付与が、20週間にわたり免疫原性に与える効果を評価することであった。ヒト試験の実施前に、プロトタイプのIgE−EMPDペプチドワクチン製剤及び投薬計画の免疫原性を評価するため、動物モデルとしてカニクイザルを選択した。代表的なペプチド免疫原構築物(配列番号88)を、2つの一般的に使用される製剤に製剤化した。第1の製剤では、配列番号88のIgE EMPDペプチド免疫原構築物をCpGで安定化免疫刺激性複合体にした後、Montanide(商標)ISA51と混合して油中水型エマルションを形成した(試験パートA)。第2の製剤では、配列番号88のIgE EMPDペプチド免疫原構築物をCpGで安定化免疫刺激性複合体にした後、ADJUPHOSで懸濁製剤を形成した(試験パートB)。この包括的な免疫原性試験では、各製剤で、30μg、100μg、300μg、1000μg/用量からなる4通りの投薬を評価した。
2.5〜4.0kgの成体カニクイザルを選択し、IgE EMPDペプチド免疫原が免疫原性及びマカク血清IgEレベルに与える効果を評価した。合計20匹のマカクを10群に分け、プラセボ対照動物(n=2)にはアジュバントのみ(Montanide(商標)ISA 51とCpGオリゴデオキシヌクレオチド)または(ADJUPHOSとCpGオリゴデオキシヌクレオチド)を注射した。実験動物に、IgE EMPDペプチド免疫原(配列番号88)を群あたり30、100、300、及び1,000μgの用量で注射した(動物あたりワクチン体積合計500μL;n=群あたり2匹、雄1及び雌1)。0週目、3週目、及び6週目に合計3回の筋肉内免疫付与を施した。0週目、3週目、6週目、8週目、10週目、12週目、14週目、16週目、20週目、及び24週目の免疫原性及び血清IgEレベルについて全マカクを監視した。
0週目、3週目、6週目、8週目、10週目、12週目、14週目、16週目、20週目、及び24週目に全動物から採血した。採血ごとに血清を分離し、γ1−cyno em67 ELISAを使用して抗IgE EMPD抗体価を決定した。プラセボ治療した動物は、検出可能な抗IgE EMPD抗体価がほとんどないか、皆無であった(図18A及び18B)。しかし、3回の免疫付与を受けた動物はすべて、IgE EMPD Bエピトープに対して検出可能なIgG抗体価を示し、8〜12週目にピーク力価が得られた。このような特異的反応性は、24週間の試験全体を通じて用量依存的に維持された(図18A及び18B)。1000μg用量の製剤は、それぞれの製剤でペプチド免疫原がISA51VまたはADJUPHOSアジュバントよりも過剰になるため、いずれの製剤でもγ1−cyno εm67組換えタンパク質に対する抗血清力価が高い、用量あたり0.5mL中300μgが最も至適であることが全動物で認められた。この結果は、ISA51/CpG ODNを含有する製剤が、ADJUPHOS/CpG ODNを含有する製剤と比較して、免疫原性の結果が高いことを示している。IgE EMPD B細胞エピトープに対する抗体反応は、各ペプチド免疫原の追加免疫に伴って増強され、その後、抗IgE EMPD力価は経時的に徐々に低下した。著しく高い抗血清力価が、24週間の試験全体を通じて維持された(図18A及び18B)。
カニクイザルにおける代用免疫療法アレルギーワクチンの有効性実証
a.全体目標:
本試験の目的は、ヒトIgE生成を綿密に模倣した動物モデルであるカニクイザルにおいて、自己mIgEに由来する配列であるIgE EMPDペプチド免疫原による筋肉内免疫付与が、20週間にわたり免疫原性及び血清IgEレベルに与える効果を評価することであった。非ヒト霊長類(例えば、新世界及び旧世界ザル、ならびに類人猿)のmIgEのIgE EMPDは進化的に保存されており、他の種(例えば、齧歯類、ウサギ、及びイヌ)において当該IgE EMPD対応配列は認められなかった。カニクイザルのIgE EMPDのアミノ酸配列(配列番号127)は、ヒトIgE EMPDのアミノ酸配列(配列番号2)と高い配列同一性(90%)を有している。
2.5〜4.0kgの成体カニクイザルを選択し、IgE EMPDペプチド免疫原が免疫原性及びマカク血清IgEレベルに与える効果を評価した。合計12匹のマカクを3群に分け、プラセボ対照動物(n=4、雄2及び雌2)にはアジュバントのみ(Montanide(商標)ISA 51とCpGオリゴデオキシヌクレオチド)を注射した。実験動物に、IgE EMPDペプチド免疫原(配列番号125または126)を動物あたり300μgの用量で注射した(動物あたりワクチン体積合計500μL;n=群あたり4匹、雄2及び雌2)。0週目、3週目、及び6週目に合計3回の筋肉内免疫付与を施した。0週目、3週目、6週目、8週目、10週目、12週目、14週目、16週目、18週目、及び20週目の免疫原性及び血清IgEレベルについて全マカクを監視した。
0週目、3週目、6週目、8週目、10週目、12週目、14週目、16週目、18週目、及び20週目に全動物から採血した。採血ごとに血清を分離し、γ1−cyno em67 ELISAを使用して抗IgE EMPD抗体価の決定を行った。プラセボ治療した動物は、検出可能な抗IgE EMPD抗体価が皆無であった(図19)。しかし、配列番号125または配列番号126のいずれかによる3回の免疫付与を受けた動物はすべて、IgE EMPD Bエピトープに対して検出可能なIgG抗体価を示し、8〜12週目にピーク力価が得られた(図19)。このような特異的反応性は、20週間の試験全体を通じて維持された。加えて、免疫された動物はすべて、20週間にわたって特異的IgM及びIgA抗体価を示した(図20)。
0週目、3週目、6週目、8週目、10週目、12週目、14週目、16週目、18週目、及び20週目に全動物から採血した。採血ごとに血清を分離し、定量的マカクIgE ELISAを使用して血清IgEの測定を行った。プラセボ群での基礎IgEレベルは、生存局面の期間中に変化した。配列番号125を投与した群で観察されたIgEの減少は統計的に有意であったが、配列番号126を投与した群でも監視期間中にIgE減少の傾向を示した(図19)。
代用モデルにおいて、IgE EMPDペプチド免疫原が、成体カニクイザルの血清試料の自己mIgEに対する免疫原性及び血清IgE濃度に与える効果を評価した。本概念実証試験では、各群4匹の動物にCpG ODNとMontanide(商標)ISA 51との混合物をプラセボ対照として投与し、8匹の成体カニクイザル(n=群あたり4匹)には、CpGオリゴデオキシヌクレオチド(CpG ODN)で独自の免疫刺激性複合体(ISC)に複合化し、Montanide(商標)ISA 51アジュバントで製剤化した300μgのマカクIgE EMPDペプチド免疫原、配列番号125または126のいずれかで0週目、3週目、及び6週目に免疫付与した。配列番号125及び126はそれぞれ、ヒトIgE EMPD免疫原、配列番号93及び88の対応物である。CpG ODN/Montanide(商標)ISA 51製剤を用いた2つのマカク由来免疫原は、全動物において強力な抗IgE EMPD IgG抗体反応をもたらした(図19)。さらに、全動物がIgE EMPDに対するIgM抗体及びIgA抗体を発現した(図20)。マカクの基礎血清IgEの減少傾向も観察された(図21)。注射部位の副作用は認められなかった。本試験は、UBITh(登録商標)T細胞エピトープが増強された、自己由来の配列である合成IgE EMPDペプチド免疫原(配列番号125及び126)が、強力な抗IgE EMPD抗体反応を誘発して、IgE生成の抑制及びマカクの基礎血清IgEレベルの減少傾向をもたらすことを実証した。
20週間にわたりIgE EMPDペプチド免疫原(配列番号125及び126)またはプラセボ対照を筋肉内経路でカニクイザルに3回注射した。動物は全体的に良好な忍容性を示し、免疫寛容が破壊された。免疫されたマカクはすべて、IgE EMPD構築物の対応するB細胞エピトープに対する強力かつ持続的なIgG(最大105)及びIgA(最大104)抗体価の発現と共に、一過性の特異的IgM抗体を発現した。あらゆる応答者に基礎IgEレベルの減少が観察された。これらの結果は、抗体が膜結合型IgEを発現するB細胞を標的とし、引き続きIgE生成を抑制させるという抗IgE−EMPD抗体の作用モードを裏付けている。

- 前記ペプチド免疫原構築物が、配列番号88〜95、98〜124、及び130からなる群から選択される、請求項1に記載のIgE EMPDペプチド免疫原構築物。
- 配列番号1または配列番号2のIgE EMPD配列からの約20〜約40アミノ酸残基を含むB細胞エピトープ、
配列番号59〜87からなる群から選択されるアミノ酸配列を含むTヘルパーエピトープ、ならびに
アミノ酸Lys−、Gly−、Lys−Lys−Lys−、(α,ε−N)Lys、及びε−N−Lys−Lys−Lys−Lys(配列番号129)からなる群から選択される任意選択の異種スペーサーを含む、IgE EMPDペプチド免疫原構築物であって、前記B細胞エピトープが直接、または前記任意選択の異種スペーサーによって、前記Tヘルパーエピトープに共有結合している、前記IgE EMPDペプチド免疫原構築物。 - 前記B細胞エピトープが、配列番号5、6、8、及び9からなる群から選択される、請求項5に記載のIgE EMPDペプチド免疫原構築物。
- 前記Tヘルパーエピトープが、配列番号59〜87からなる群から選択される、請求項5に記載のIgE EMPDペプチド免疫原構築物。
- 前記任意選択の異種スペーサーが、(α,ε−N)Lys、またはε−N−Lys−Lys−Lys−Lys(配列番号129)である、請求項5に記載のIgE EMPDペプチド免疫原構築物。
- 前記Tヘルパーエピトープが前記B細胞エピトープのアミノ末端に共有結合している、請求項5に記載のIgE EMPDペプチド免疫原構築物。
- 前記Tヘルパーエピトープが前記任意選択の異種スペーサーによって前記B細胞エピトープのアミノ末端に共有結合している、請求項5に記載のIgE EMPDペプチド免疫原構築物。
- 請求項1〜10のいずれかに記載のペプチド免疫原構築物を含む組成物。
