関連出願の相互参照
本出願は、その全体が複製されるかのように参照により本明細書に組み込まれる、John Mansellによって2018年2月2日に出願され、「Compositions and Methods for Treatment of Obesity and Obesity Related Disorders」と題された米国仮特許出願第62/625,688号明細書の優先権を主張する。
本開示は、肥満及び肥満関連障害の治療のための組成物及び方法に関する。さらに具体的には、本開示は、グレリン及びグレリン関連分子を阻害するための組成物及び方法に関する。
肥満(過度の肥満症)は、主要な公衆衛生問題である。2003〜2004年の全国健康栄養調査(NHANES)の結果は、米国成人の推定66%が過体重又は肥満であり、米国小児の17%が過体重であることを示した。過度の肥満症は深刻な問題であり、インスリン抵抗性、脂質異常症、軽度の炎症、ならびに糖尿病、アテローム性動脈硬化症及びある種の癌の発症に何らかの役割を果たす成長因子及び他のホルモンのレベルの変化に関連している。さらに、過度の肥満症は老化の加速に関連しているという証拠が蓄積されつつある。試験では、体重のわずかな減少であっても、心血管リスク因子にプラスの影響を及ぼし、2型糖尿病及び糖尿病関連合併症を発症するリスクの低下と関連していることが示されている。
一態様では、細胞内のグレリンの発現を阻害する方法である。該方法は、細胞にRNAiを導入することを含み得る。該RNAiは、センス鎖及びアンチセンス鎖を含み得る。該アンチセンス鎖は、配列番号1から配列番号42のいずれかを含み得る。前記センス鎖は、前記アンチセンス鎖に相補的であり得る。

Claims (22)
- 細胞内のグレリンの発現を阻害する方法であって、前記細胞にRNAiを導入することを含み、該RNAiが、センス鎖及びアンチセンス鎖を含み、
前記アンチセンス鎖は、配列番号1から配列番号42のいずれかを含み、
前記センス鎖は、前記アンチセンス鎖に相補的である、方法。 - RNAiのアンチセンス鎖及びセンス鎖が、それぞれ19〜25ヌクレオチド長である、請求項1に記載の方法。
- RNAiが少なくとも1つの修飾ヌクレオチドを含む、請求項1に記載の方法。
- RNAiがデンドリマーと結合する、請求項1に記載の方法。
- RNAiがナノ粒子と結合する、請求項1に記載の方法。
- RNAiがsiRNA、マイクロRNA、shRNA又はそれらの組合せである、請求項1に記載の方法。
- RNAiがカプセル化されている、請求項1に記載の方法。
- 細胞がヒト細胞である、請求項1に記載の方法。
