中枢神経系(CNS)はニューロンとグリア細胞の両方を含み、正常な脳機能を維持するために微妙なバランスを形成している。CNS損傷は、神経細胞の脱落又はグリア瘢痕(glial scar)のいずれかとの関連で研究されることが多い。数十年の研究にもかかわらず、成体哺乳動物のCNSにおける神経損傷後の新たなニューロンの生成は困難である(Cregg et al., 2014 Exp Neurol 253:197-207;He et al., 2016 Neuron 90:437-451;及びYiu et al., 2006 Nat Rev Neurosci 7:617-627)。グリア瘢痕は、コンドロイチン硫酸プロテオグリカン(CSPG)及びリポカリン-2(LCN2)などの神経抑制因子、並びにTNFα及びインターロイキン-1β(IL-1β)などの炎症性サイトカインを蓄積することにより、神経再生において、物理的障害になるだけでなく、化学的障害にもなる(Ferreira et al., 2015 Prog Neurobiol 131:120-136;Koprivica et al., 2005 Science 310:106-110;及びSilver et al., 2004 Nat Rev Neurosci 5:146-156)。
本書は脳内で機能的なニューロンを生成するための方法と材料を提供する。例えば、本書は、脳内(例えば、大脳皮質内)のアストロサイト(例えば、反応性アストロサイト)を機能的なニューロン(例えば、生きた哺乳動物(例えば、ヒト)の脳内に機能的に組み込まれ得るニューロン)に変換するために、NeuroD1ポリペプチドをコードする核酸を使用するための方法及び材料を提供する。場合によっては、本明細書で提供される材料及び方法を、脳損傷を処置するために使用することができる。例えば、NeuroD1が媒介するアストロサイト−ニューロン(AtN)変換は、反応性アストロサイトの機能的なニューロンへの変換、ニューロン:グリア比の再平衡化、損傷した脳組織の修復(例えば、グリア瘢痕組織を神経組織に戻すことによるグリア瘢痕組織の修復)、神経炎症の減弱、血液脳関門の回復、A1アストロサイトの形質転換(例えば、毒性A1アストロサイトの、より害の少ないアストロサイトへの形質転換)、及び/又は毒性M1ミクログリアの量の低下によって、脳損傷を処置するために(例えば、脳損傷の後に)使用することができる。
本書は脳内で機能的なニューロンを生成するための方法と材料を提供する。例えば、本書は、NeuroD1ポリペプチドをコードする核酸を用いて、脳内のグリア細胞(例えば、反応性アストロサイト)を、生きた哺乳動物(例えば、ヒト)の脳内で機能的なニューロンを形成するように誘因する(trigger)ための方法と材料を提供する。本明細書に記載の機能的なニューロンの形成には、脳内の反応性アストロサイトを機能的なニューロンに変換することを含み得る。本明細書で使用される「機能的なニューロン」という用語は、生きた哺乳動物(例えば、ヒト)の脳内に機能的に組み込まれたニューロンを指す。例えば、機能的なニューロンは、グルタミン酸作動性ニューロン及び/又はGABA作動性ニューロンであり得る。場合によっては、本明細書に提供される材料及び方法は、脳損傷を処置するために使用することができる。例えば、NeuroD1が媒介するアストロサイト−ニューロン(AtN)変換は、反応性アストロサイトの機能的なニューロンへの変換、ニューロン:グリア比の再平衡化、損傷した脳組織の修復(例えば、グリア瘢痕組織を神経組織に戻すことによるグリア瘢痕組織の修復)、神経炎症の減弱、血液脳関門の回復、A1アストロサイトの形質転換(例えば、毒性A1アストロサイトの、より害の少ないアストロサイトへの形質転換)、及び/又は毒性M1ミクログリアの量の低下によって、脳損傷を処置するために(例えば、脳損傷の後に)使用することができる。
この実施例では、in vivoにおけるグリア‐ニューロン変換を調べる。マウス運動皮質における重度の刺傷モデルを用いて、損傷脳の微小環境に対する細胞変換の影響を調べた。反応性アストロサイトを死滅させたときの結果とは異なり、反応性アストロサイトをニューロンに変換することで、グリア瘢痕を神経組織に戻した。アストロサイトは神経変換後に枯渇せず、むしろ変換後に再増殖した。反応性アストロサイトにおけるNeuroD1の異所性発現は、A1型毒性アストロサイトをより反応性の低いアストロサイトに変換した。反応性ミクログリアもまた改善され、NeuroD1が媒介するアストロサイト‐ニューロン(AtN)変換後に神経炎症が減弱した。さらに、AtN変換後に、血液脳関門(BBB)は回復し、神経のシナプス結合は再確立した。したがって、本実施例で提供される結果は、NeuroD1が媒介する細胞変換が、損傷後のニューロン:グリア比の再平衡化により、グリア瘢痕を神経組織に戻すことができることを実証する。
刺傷のマウスモデルとウイルス注射
動物の手順(precedure)は米国国立衛生研究所の動物保護ガイドラインに準拠して実施し、全ての実験プロトコールはペンシルベニア州立大学動物実験委員会(IACUC)の承認を受けた。野生型(WT)C57BL/6J及びFVB/N-Tg(GFAP::GFP) 14Mes/JトランスジェニックマウスはJackson Laboratoryから購入した。マウスを12時間の明/暗サイクルの中で飼育し、十分な食物と水を供給した。両方の性別の成体マウス(20〜30グラム)を3〜6か月齢で実験に動員した。マウス運動皮質は、一部改変して他の所に記載されているように鈍針(外径0.95mm)で損傷された(例えば、Bardehle et al., 2013 Nat Neurosci 16:580-586;Bush et al., 1999 Neuron 23:297-308;及びGuo et al., 2014 Cell Stem Cell 14:188-202を参照)。簡単に説明すると、ケタミン/キシラジン(100mg/kgケタミン;12mg/kgキシラジン)を腹腔内注射により投与した。麻酔下で、正中切開により頭蓋骨とブレグマ(bregma)を露出させたマウスを、定位装置に留置した。運動皮質の頭蓋骨に約1.2mmの穴を掘った(調整:ブレグマと相対的に、前後(AP)+1.0mm;内側‐外側(ML)±1.5mm)。鈍針(0.95mm)を背腹(DV)-1.8mmの深さまで各部位に留置し、3分間維持した(stay still)。刺傷後4又は10日(dps)で、マウスを無作為に、同じ部位へのNeuroD1又は対照のウイルス注射に供した。ウイルス注射の手順は、他の所に記載されているものと同様であり(例えば、Guo et al., 2014 Cell Stem Cell 14:188-202を参照)、5μLのマイクロシリンジ及び34ゲージ針(Hamilton)を用いて、各注射部位に1.5μLのAAV又はレトロウイルスを注入した。0.1mm/分の速度で徐々に針を上方に移動しながら、ウイルス注入速度は0.15μL/分で制御された。注射後、針をさらに3分間維持した後、完全に抜去(withdrawn)した。術後、自由運動が観察されるまで加熱パッド上でマウスを回復させた。マウスを単独飼育し、少なくとも1週間にわたり、毎日注意深くモニターした。
hGFAPプロモーターはpDRIVE-hGFAPプラスミド(InvivoGen Inc.)から入手し、pAAV-MCS(Cell Biolab)のMluIとSacIIの間に挿入してCMVプロモーターを置き換えた。Cre遺伝子は、Addgeneプラスミド番号40591(Dr.Albee Messingから入手)からPCR法によって入手し、pAAV MCSのEcoRI部位とSal1部位の間に挿入してpAAV-hGFAP::Creベクターを作製した。pAAV-FLEX-mCherry-P2A-mCherry及びpAAV-FLEX-NeuroD1-P2A-mCherryベクターを構築するために、NeuroD1又はmCherryをコードするcDNAを、他の所に記載されているように、レトロウイルスコンストラクトを用いたPCR法によって入手した(例えば、Guo et al., 2014 Cell Stem Cell 14:188-202を参照)。NeuroD1遺伝子をP2A-mCherryと融合させ、pAAV-FLEX-GFPベクター(Addgeneプラスミド番号28304)のKpn1部位とXho1部位の間にサブクローニングした。プラスミドコンストラクトはシーケンシングにより確認した。
組み換えAAV血清型(stereotype)9は293AAV細胞(Cell Biolabs)で生成した。簡単に説明すると、三重のプラスミド(pAAV発現ベクター、pAAV9-RC(Cell Biolab)、及びpHelper(Cell Biolab))をポリエチレンイミン(PEI、直鎖状、MW 25,000)によってトランスフェクトした。トランスフェクション後72時間において、細胞を掻きとり(scrap)、遠心分離した。細胞ペレットをドライアイス/エタノールと37℃の水浴とに交互に留置して4回凍結融解をした。AAVライセートを、不連続イオジキサノール勾配(Beckman SW55Tiロータ)中で、54,000rpmで1時間の超遠心分離により精製した。ウイルスを含む層を採取した後、Milliporeアミコンウルトラ遠心フィルターで濃縮した。ウイルス力価は当初(initially)、QuickTiterTM AAV定量キット(Cell Biolabs)で測定した:hGFAP::Creは1.2×10^12 GC/mL、FLEX-NeuroD1-P2AmCherryは1.4×10^12 GC/mL、FLEX-mCherry-P2A-mCherryは1.6×10^12 GC/mLであった。
pCAG::GFP-IRES-GFPのレトロウイルスベクターを入手した。マウスNeuroD1配列を上記ベクターに挿入してpCAG::NeuroD1-IRES-GFPベクターを作製した(Guo et al., 2014 Cell Stem Cell 14:188-202)。レトロウイルス粒子をパッケージングするために、標的ベクターを、水疱性口炎ウイルス糖タンパク質(VSV‐G)ベクターと共に、gpgヘルパーフリーヒト胎児腎(HEK)細胞に、PEIによってトランスフェクトした。レトロウイルス粒子の力価は約107粒子/mLと決定された。
脳試料は、他の所に記載されているように採取した(例えば、Guo et al., 2014 Cell Stem Cell 14:188-202を参照)。簡単に説明すると、麻酔のために、動物に2.5%アベルチンを注射した。人工脳脊髄液(ACSF)による経心潅流を実施し、全身的に血液を洗い流した。次に、脳を切り出し、免疫組織化学での使用のためには、脳を4%パラホルムアルデヒド(PFA)中で4℃で一晩、後固定した。RNA抽出での使用のためには、損傷部位周辺の脳組織(約1.5×1.5mm四方)を切り出し、-80℃で急速冷凍した。
固定後、Leica-1000ビブラトームを用いて40μm切片で脳組織を切断した。脳切片をリン酸緩衝生理食塩水(PBS)で3回洗浄後、PBS中の2% Triton X-100で1時間透過処理した。次に、脳切片をPBS中の5%正常ロバ血清及び0.3% Triton X-100で1時間ブロッキングした。一次抗体をブロッキング緩衝液に添加し、脳切片とともに4℃で一晩インキュベートした。一次抗体をPBSで3回すすいだ後、室温(RT)で2時間、二次抗体のインキュベーションをした。PBSで洗浄後、脳切片をDAPI(Invitrogen)を含む退色防止封入溶液でスライドガラス上に封入した。画像は共焦点顕微鏡(Olympus FV1000又はZeiss LSM800)で取得した。抗体特異性を確保するため、左右対照(side-by-side control)としての免疫染色には二次抗体のみを用いたが、目立った(distinct)シグナルは検出されなかった。
脳損傷後の増殖性アストロサイトの標識のためには、GFAP-GFPトランスジェニックマウスを用い、1 dps〜4 dps又は10 dpsまでの間、BrdU(BrdU標識試薬、Invitrogen)の腹腔内注射を、0.1ml/10gの用量で毎日行った。変換後に新しく生成したアストロサイトを特徴付けるためには、BrdUを7〜14 dpiの間毎日投与した。DNA変性のために、固定した脳切片を37℃で30分間、2M HClによる処理に供した。PBSで5回洗浄後、脳切片を2% Triton-PBSで1時間透過させ、ブロッキング緩衝液(PBS中5%正常ロバ血清及び0.3% triton)中で、室温でさらに1時間インキュベートした。一次抗体をブロッキング溶液に混合し、脳切片と共に4℃で一晩インキュベートした。
一連の冠状切片を採取し、組織の損傷の解析を行った。針損傷の中心を採取し、ゼロ点に設定した;損傷部位の前方と後方の2切片(200μm間隔)も選択した。脳切片をまずキシレン中に5分間留置し、その後アルコール(水中95%、70%、及び0%)で3分間ずつ一連の段階的な水和を行った。試料を60℃で8分間、クレシルバイオレート緩衝液(NaAc緩衝液中0.121mg/mLクレシルバイオレット酢酸、pH 3.5)に移した。完了すると、染色液をすすぎ、アルコール(0%、70%、95%、及び100%)で30秒間ずつ連続して脱水した。最後に、試料をキシレン中で1時間清澄化し(clear)、DPXマウンタント(Sigma-Aldrich)中に封入した。画像はOlympus BX61顕微鏡を用いて取得した。
RNA抽出はMacherey-NagelヌクレオスピンRNAキットを用いて行った。RNA濃度と純度はナノドロップで測定した。cDNA合成のためには、500ngのRNAをQuanta Biosciences qScript cDNA supermixと混合し、25℃で5分間、42℃で30分間、85℃で5分間インキュベートし、4℃で保持した。完了すると、cDNAをRNase/DNaseを含まない水で5倍に希釈した。リアルタイムqPCR用のプライマーは、Applied Biosystems Primer Expressソフトウェアを用いて設計し、IDTで合成した。qRT-PCRの各反応のために、6.25μLのSYBR Green Supermix (Quanta Biosciences PerfeCTa、ROX)、2μLのcDNA、及び3.75μLの水をよく混合し96ウェルプレート(Applied Biosystem Inc)に充填(load)した。Gapdhを内部対照として用いた。全ての比較は、健常マウス由来の損傷なしの皮質組織に対して行った。倍数変化の計算には比較Ct法を用いた。
マウスを麻酔し、上記のようにACSFで潅流した後、PBS中0.5mg/mLのスルホ-NHS-LC-ビオチンを15mL加えた。免疫組織化学のためには、一次抗体インキュベーションの後、脳切片をFITCストレプトアビジン(1:800;PBS + 0.3% triton +2.5%正常ヤギ又はロバ血清で希釈)と共に室温で1時間インキュベートし、続いて通常の封入手順を行った。
脳切片記録は、他の所に記載されているように実施した(例えば、Guo et al., 2014 Cell Stem Cell 14:188-202を参照)。簡単に説明すると、NeuroD1-AAV注射の1ヵ月後に、マウスを2.5%アベルチンで麻酔し、次いでNMDGベースの切断溶液((mMで):93 NMDG、93 HCl、2.5 KCl、1.25 NaH2PO4、30 NaHCO3、20 HEPES、15グルコース、12 N-アセチル-L-システイン、5 アスコルビン酸ナトリウム、2 チオ尿素、3 ピルビン酸ナトリウム、7 MgSO4、0.5 CaCl2;pH 7.3〜7.4、300 mOsmo、95% O2 / 5% CO2でバブリング)で潅流した。300μm厚の冠状切片には、AAV注射された皮質領域周辺を、ビブラトーム(VT1200S、Leica、ドイツ)を用いて室温で切断した。切片を採取し、酸素添加NMDG切断溶液中で、33.0±1.0℃で10〜15分間インキュベートした。次に、切片を、95% O2 / 5% CO2で連続してバブリングした保持溶液((mMで):92 NaCl、2.5 KCl、1.25 NaH2PO4、30 NaHCO3、20 HEPES、15 グルコース、12 Nアセチル-L-システイン、5 アスコルビン酸ナトリウム、2 チオ尿素、3 ピルビン酸ナトリウム、2 MgSO4、及び2 CaCl2)に移した。脳切片は、室温で少なくとも0.5時間、保持溶液中で回復させた。パッチクランプ記録のために、単一切片を記録チャンバーに移し、95% O2 / 5% CO2で飽和させた33.0±1.0℃の標準ACSF((mMで):124 NaCl、2.5 KCl、1.25 NaH2PO4、26 NaHCO3、10 グルコース、1.3 MgSO4、及び2.5 CaCl2)で連続潅流した。活動電位とイオン電流を記録するためには、ホールセルでの記録をピペット溶液((mMで):135K‐グルコネート、10 KCl、5 Na‐ホスホクレアチン、10 HEPES、2 EGTA、4 MgATP、及び0.3 Na2GTP含有;KOHでpH 7.3に調整、280〜290 mOsm)を用いて実施した。活動電位を引き出すために、電流クランプモデル(model)下で脱分極電流を注入した。自発性興奮性シナプス後電流(sEPSC)及び自発性抑制性シナプス後電流(sIPSC)を記録するためには、ピペット溶液は以下を含んだ(mMで):120 Cs‐メタンスルホン酸塩、10 KCl、10 Na‐ホスホクレアチン、10 HEPES、5 QX-314、1 EGTA、4 MgATP、及び0.3 Na2GTP;KOHでpH 7.3に調整、280〜290 mOsm。細胞膜電位はsEPSC記録のためには-70mV(イオンチャネル型グルタミン酸受容体の反転電位)、sIPSC記録のためには0 mV(GABAA受容体の反転電位)でそれぞれ保持した。MultiClamp 700A増幅器でデータを収集し、pCLAMP10ソフトウェア(Molecular Devices)で解析した。
統計解析及び棒グラフにはPrism 6 GraphPadソフトウェアを用いた。2つのデータセットの比較のためには、スチューデントのt検定を行った。3つのデータセットの比較のためには、一元配置又は二元配置分散分析(ANOVA)を実施し、続いて事後検定を行った。統計学的有意性はp<0.05とした。データは平均値±標準誤差で示した。

重度の刺傷モデルにおける、NeuroD1が媒介する高い効率のアストロサイト‐ニューロン変換
反応性グリア細胞は、単一の転写因子NeuroD1(Guo et al., 2014 Cell Stem Cell 14:188-202)によって、マウス脳内で直接的に機能的なニューロンに変換することができる。グリア瘢痕によって軸索再生が促進(aid)されるか、そしてアストロサイトの機能的なニューロンへの変換によって何らかの有害な作用が生じるかを明らかにするため、成体マウス(3〜6か月齢、両方の性別を含む)において重度の刺傷モデルを確立し、損傷領域の微小環境に対するNeuroD1が媒介するAtN変換の影響を調べた。具体的には、鈍針(外径0.95mm)を用いてマウス運動皮質に重度の刺傷を作製したところ、損傷部位において、反応性アストログリア増殖症と共に顕著な組織の脱落が誘導された(図1A)。予想された通り、刺傷後10日(dps)で刺傷領域におけるアストロサイトの数が有意に増加した(図1A、右の棒グラフ)。これは、細胞分裂中のブロモデオキシウリジン(BrdU)の取り込みによって示される、神経損傷後のアストロサイトの内的な増殖能力と一致する(図1B:BrdU+アストロサイト、38.7±2.5%;n = 4匹のマウス、10 dps)。
高い効率のAtN変換によって、ニューロン変換後のアストロサイトの枯渇が生じるかどうかを明確にするためにGFAP染色を実施したところ、対照群と比較してNeuroD1群のGFAPシグナルの全体的な低下が観察された(図4C)。予想外なことに、GFAPシグナルが低下しただけでなく、アストロサイト形態がNeuroD1群において顕著な変化を示し、対照群と比較して肥はん突起がはるかに少ないことが示され、(図4C、左の画像の矢尻)NeuroD1変換領域のアストロサイトの反応性が低くなったことが示唆された。定量的解析によって、対照群では刺傷後にGFAPシグナルが有意に(5〜10倍)増加したが(図4C、黒色の棒)、NeuroD1感染領域では有意に(半分に)減少することが認められた(図4C、灰色棒)。NeuroD1群におけるGFAPシグナルの減少は、反応性アストロサイトのニューロンへの変換と一致していた。一方、NeuroD1感染領域においてGFAPシグナルが有意なレベルで検出されることから、反応性アストロサイトが変換後に枯渇しないことが示唆された。アストロサイトは内的な増殖能力を有するため、神経変換によって残りのアストロサイトの増殖が誘因されるかどうかを調査した。この考えを試験するために、対照群とNeuroD1群の両方で細胞増殖をモニターするため、細胞分裂の間にDNAに取り込まれ得るBrdUを7 dpi〜14 dpi(4 dpsでウイルス注射)の間毎日注射した(図4D、左の模式図)。興味深いことに、NeuroD1群におけるBrdU標識アストロサイトの数が対照群の3倍以上であることが分かった(図4D、右パネル及び定量用棒グラフ)。BrdU標識アストロサイトの多くはNeuroD1変換ニューロンに隣接しており(図4D、矢尻)、アストロサイトはAtN変換後に自己再生できることが示唆された。したがって、アストロサイトはAtN変換により枯渇せず、むしろ再増殖した。
脳機能はニューロンとグリア細胞の間の微妙なバランスに依存している。神経損傷の後、ニューロンは死滅するが、グリア細胞は増殖し、損傷領域におけるニューロン:グリア比の変化が導かれる。これは重度の刺傷モデルに明らかに反映され、健常なニューロン(NeuN+細胞)の数は損傷後に顕著に減少したが、アストロサイト(GFAP+/s100b+)の数は顕著に増加した(図4E)。興味深いことに、NeuroD1が媒介するAtN変換の後、NeuN+ニューロンは顕著に増加したが、アストロサイトの数は減少し(図4E)、損傷から明確に逆転した。定量的解析によって、マウス運動皮質のニューロン:アストロサイト比は休止条件で約4:1(4つのニューロン対1つのアストロサイト)であることがわかった(図4E、右の棒グラフの白色の棒)。刺傷後には、ニューロン:アストロサイト比は1未満に低下した(図4E、黒色の棒)。NeuroD1変換後には、ニューロン:アストロサイト比は14 dpiで2.6に戻った(図4E、灰色の棒)。このようなニューロン:アストロサイト比の有意な逆転は、損傷脳の機能的回復に関与し得る。
NeuroD1が媒介する変換後にアストロサイトが持続する(persist)か明らかにし、それらが対照群における反応性アストロサイトと異なるかどうかを評価するために、ニューロン変換前の初期段階である3 dpiにおいて、A1アストロサイト及び神経炎症に関する様々な遺伝子のRT-PCR解析を実施した。損傷なし(NI)の群と比較して、刺傷によってGfapなどの汎反応性アストロサイト遺伝子の37倍のアップレギュレーションが引き起こされ(図6A)、NeuroD1群で有意に減弱した(一元配置分散分析、シダック検定、** P<0.01、n = 4対)。損傷後の反応性アストロサイトに関連する神経炎症マーカーであるLcn2は、刺傷後に700倍増加したが、NeuroD1群では劇的に低下した(図6A、一元配置分散分析、シダック検定、*** P<0.001、n = 4対)。さらに、Gbp2やSerping1などのA1アストロサイトに特徴的な遺伝子は刺傷後に300〜900倍アップレギュレートされたが(一元配置分散分析、シダック検定、*** P<0.001、n = 4対)、NeuroD1処置後には大きく減弱した(図6B)。NeuroD1感染後の3 dpiでは、アストロサイトはまだ完全にニューロンに変換されていないため、この著しい変化は予想外であった(図7A、B)。3 dpiでの免疫染色により、NeuroD1が実際に感染アストロサイトで発現していることが確認された(図7B:mCherry+細胞の87.4±2.5%がNeuroD1+;mCherry+細胞の92.8±2.8%がGFAP+;n = 6匹のマウス)。それにもかかわらず、神経変換の前にA1アストロサイトは既に阻害又は形質転換されていた。さらに、NeuroD1処置により、Anax2、Thbs1、Gpc6、Bdnfなどの神経機能を補助するアストロサイト遺伝子が増加したようであった(図6C)。このRT-PCRの結果の検証として、NeuroD1の過剰発現がRT-PCR解析によって確認された(図8A)。NeuroD1感染後にGFAP発現が減少したにもかかわらず、A2アストロサイト遺伝子は変化しなかった(図8B、C)。
次に、ミクログリアに対するAtN変換の影響を調べた。損傷なしの脳の休止ミクログリア(図9A、上段)と比較して、刺傷で誘導された反応性ミクログリアは肥はん性でアメーバ状形状であった(図9A、中段)。BrdU標識(図10A‐C)で示されたように刺傷後はミクログリア、アストロサイトともに増殖性が高かったが、刺傷後の成体マウス皮質では新生ニューロンは検出されなかった(図10D)。しかし、NeuroD1感染領域では、ミクログリア形態は戻り、網目状突起を有する休止ミクログリアに近かった(図9A、下段)。このような形態的変化は、3 dpiという早い時期に始まり(図9B)、NeuroD1感染アストロサイトに接触したミクログリアは、mCherry感染アストロサイトに接触したミクログリアと比較して、反応性がはるかに低かった。RT-PCR解析によって、刺傷後にサイトカインであるTNFαとIL-1bはいずれも有意に増加したが、NeuroD1群ではいずれも減弱することが明らかになった(図9B、棒グラフ、一元配置分散分析の後、トゥーキー(Turkey)検定、* P<0.05、*** P<0.001、n = 4対)。AtN変換中のこのようなサイトカインの劇的な減少によって、NeuroD1群でミクログリアの反応性が低かった理由が説明できるかもしれない。これに一致して、iNOSに対して免疫陽性であった毒性M1ミクログリアは、対照群と比較してNeuroD1群で有意な減少を示した(図9C、スチューデントのt検定、*** P<0.001、n = 4対)。iNOS標識M1ミクログリアの低下は、NeuroD1感染後の3 dpiで検出されたような毒性A1アストロサイトの低下と合致しており、アストロサイトとミクログリアの間の密接な相互作用が示唆された。7 dpiでは、対照群と比較して、NeuroD1群におけるiNOSの低下はさらに顕著であり、Iba1シグナルの減少をも伴っていた(図9D、二元配置分散分析、** P<0.01、*** P<0.001、3 dpi及び7 dpiにはn = 6対、14 dpiにはn = 5対)。加えて、刺傷によって、マクロファージ及び単球、並びにいくつかの反応性ミクログリアのマーカーであるCD68の発現レベルの著しい増加が誘導された(図9E)。しかし、NeuroD1感染領域では、CD68の発現レベルが有意に減弱していた(図9E、棒グラフ、二元配置分散分析、** P<0.01、*** P<0.001、n = 6対)。総合すると、これらの結果から、アストロサイト‐ニューロン変換に伴い、毒性M1ミクログリアが低下し、神経炎症が緩和されたことが示唆された。
脳内でのアストロサイトの機能は、血管と相互作用して、血液脳関門(BBB)に寄与することで、細菌・ウイルス感染を予防し、化学的毒性を低下させることである(Obermeier et al., 2013 Nat Med 19:1584-1596)。健常な脳内では、アストロサイトの終足が血管の周りに巻きつくことにより、BBBが引き締められる(図11A)。損傷なしの脳における均等に分布した血管(内皮マーカーLy6Cで標識)と比較して(図11B、左パネル)、刺傷によって血管は膨張した(図11B、中央パネル)。しかし、NeuroD1処置群では、血管はより低い肥はん形態を示し、健常な脳のそれに近かった(図11B、右パネル)。刺傷後の血管の形態変化に伴って、AQP4シグナルの誤った局在化によって明らかなように、BBBの破壊が認められた。AQP4は水チャンネルタンパク質であり、正常では血管の周りに巻きついた休止状態のアストロサイトの終足に集中する(図11Aを参照)。刺傷後、AQP4シグナルは血管から解離し、代わりに損傷領域全体に分布した(図11C、上段)。NeuroD1処置領域では、AQP4シグナルは血管との再会合を示し、正常な状態に戻った(図11C、下段)。また、NeuroD1処置群ではアストロサイト形態は反応性がはるかに低いように思われた(図11C、下段)。BBBの完全性をさらに評価するために、BBBの破たん後に漏れ出しやすい分子であるビオチンでマウスを潅流した。刺傷後、mCherryだけを発現させた対照群では、損傷領域におけるビオチンの有意な漏出が観察された(図11D、上段)。しかし、NeuroD1群では、ビオチンは主に血管の内側で検出され、実質組織における漏出は有意に減少しており(図11D、下段)、BBBの完全性の回復が示された。総合すると、これらの結果から、NeuroD1が媒介する細胞変換の後、アストロサイトは再度血管と相互作用して、神経損傷によって引き起こされた破壊されたBBBを回復することが示唆された。
NeuroD1が媒介するAtN変換後のグリア環境の実質的な変化と共に、損傷領域における樹状突起の形態、シナプス密度、電気生理学的機能などのニューロン特性を調べた。刺傷によって、樹状突起マーカーであるSMI32で示されるような重度の樹状突起損傷が生じた(図12A、左の画像、緑シグナル)。しかし、NeuroD1感染領域では、樹状突起シグナルSMI32の顕著な増加が検出された(図12A、下の画像)。定量的には、SMI32で標識されたニューロンの樹状突起は刺傷害後に重度に損傷し、損傷なしのレベルの25%にまで減少したが、NeuroD1処置によって、14 dpiに樹状突起シグナルが損傷なしのレベルの50%超にまでレスキューされた(図12A;棒グラフ、二元配置分散分析、*** P<0.001、n = 4対)。樹状突起損傷と一致して、刺傷によって、グルタミン酸作動性シナプスマーカーvGluT1及びGABA作動性シナプスマーカーGAD67によって示されるように、損傷領域におけるシナプスの重度の低下も引き起こされた(図12B、左の画像)。NeuroD1処置後(30 dpi)、損傷領域におけるグルタミン酸作動性及びGABA作動性シナプスの密度はいずれも、対照群と比較して有意な増加を示した(図12B、棒グラフ)。

Claims (51)
- 生きた哺乳動物の脳の大脳皮質においてグリア瘢痕組織を修復する方法であって、前記方法が、ニューロン分化1(neuronal differentiation 1:NeuroD1)ポリペプチドをコードする核酸を前記大脳皮質内のアストロサイトに投与することを含み、前記NeuroD1ポリペプチドが前記アストロサイトによって発現され、前記アストロサイトが前記大脳皮質内で機能的なニューロンを形成し、かつ、前記グリア瘢痕組織が神経組織へと戻る、前記方法。
- 前記大脳皮質において、NeuroD1ポリペプチドをコードする前記核酸の投与後に、グリア線維性酸性タンパク質(Gfap)、リポカリン-2(Lcn2)、及び/又はコンドロイチン硫酸プロテオグリカン(CSPG)の発現が減少する、請求項1に記載の方法。
- 前記大脳皮質において、NeuroD1ポリペプチドをコードする前記核酸の投与後に、アネキシンA2(Anax2)、トロンボスポンジン1(Thbs1)、グリピカン6(Gpc6)、及び/又は脳由来神経栄養因子(Bdnf)の発現が増加する、請求項1に記載の方法。
- 生きた哺乳動物の脳の大脳皮質においてニューロン:グリア比を再平衡化(rebalancing)する方法であって、前記方法が、ニューロン分化1(NeuroD1)ポリペプチドをコードする核酸を前記大脳皮質内のアストロサイトに投与することを含み、前記NeuroD1ポリペプチドが前記アストロサイトによって発現され、前記アストロサイトが前記大脳皮質内で機能的なニューロンを形成し、かつ、前記ニューロン:グリア比が増加する、前記方法。
- 前記ニューロン:グリア比が、アストロサイトの数を減少させること、及び/又はニューロンの数を増加させることによって増加する、請求項4に記載の方法。
- 前記ニューロン:グリア比が、アストロサイトの数を減少させることによって、及びニューロンの数を増加させることによって増加する、請求項4に記載の方法。
- 生きた哺乳動物の脳の大脳皮質において神経炎症を減弱させる方法であって、前記方法が、ニューロン分化1(NeuroD1)ポリペプチドをコードする核酸を前記大脳皮質内のアストロサイトに投与することを含み、前記NeuroD1ポリペプチドが前記アストロサイトによって発現され、前記アストロサイトが前記大脳皮質内で機能的なニューロンを形成し、かつ、前記神経炎症が減弱する、前記方法。
- 前記大脳皮質において、NeuroD1ポリペプチドをコードする前記核酸の投与後に、腫瘍壊死因子α(TNFa)、インターロイキン1ベータ(interleukin 1 beta:IL-1b)、及び/又は指定分類68(cluster of designation 68:CD68)の発現が減少する、請求項7に記載の方法。
- 生きた哺乳動物の脳の大脳皮質において血液脳関門を回復させる(restore)方法であって、前記方法が、ニューロン分化1(NeuroD1)ポリペプチドをコードする核酸を前記大脳皮質内のアストロサイトに投与することを含み、前記NeuroD1ポリペプチドが前記アストロサイトによって発現され、前記アストロサイトが前記大脳皮質内で機能的なニューロンを形成し、かつ、前記血液脳関門が回復される、前記方法。
- 前記大脳皮質において、NeuroD1ポリペプチドをコードする前記核酸の投与後に、血管でAQP4シグナル伝達が増加する、請求項9に記載の方法。
- 生きた哺乳動物の脳の大脳皮質においてA1アストロサイトを形質転換する方法であって、前記方法が、ニューロン分化1(NeuroD1)ポリペプチドをコードする核酸を前記大脳皮質内のアストロサイトに投与することを含み、前記NeuroD1ポリペプチドが前記アストロサイトによって発現され、前記アストロサイトが前記大脳皮質内で機能的なニューロンを形成し、かつ、前記A1アストロサイトがより害の少ないアストロサイトに形質転換される、前記方法。
- 前記大脳皮質において、NeuroD1ポリペプチドをコードする前記核酸の投与後に、グアニル酸結合タンパク質2(Gbp2)及び/又はセルピンファミリーGメンバー1(Serping1)の発現が減少する、請求項11に記載の方法。
- 生きた哺乳動物の脳の大脳皮質において毒性M1ミクログリアの量を低下させる方法であって、前記方法が、ニューロン分化1(NeuroD1)ポリペプチドをコードする核酸を前記大脳皮質内のアストロサイトに投与することを含み、前記NeuroD1ポリペプチドが前記アストロサイトによって発現され、前記アストロサイトが前記大脳皮質内で機能的なニューロンを形成し、かつ、毒性M1ミクログリアの量が低下する、前記方法。
- 前記毒性M1ミクログリアが、NeuroD1ポリペプチドをコードする前記核酸の投与後に、休止ミクログリアの形態を有する、請求項13に記載の方法。
- 前記哺乳動物がヒトである、請求項1〜14のいずれか一項に記載の方法。
- 前記アストロサイトが反応性アストロサイトである、請求項15に記載の方法。
- 前記NeuroD1ポリペプチドがヒトNeuroD1ポリペプチドである、請求項1〜16のいずれか一項に記載の方法。
- 前記大脳皮質が運動皮質である、請求項1〜17のいずれか一項に記載の方法。
- 前記NeuroD1ポリペプチドをコードする前記核酸が、ウイルスベクターの形で前記アストロサイトに投与される、請求項1〜18のいずれか一項に記載の方法。
- 前記ウイルスベクターがアデノ随伴ウイルスベクターである、請求項19に記載の方法。
- 前記アデノ随伴ウイルスベクターがアデノ随伴ウイルス血清型9ウイルスベクターである、請求項20に記載の方法。
- リコンビナーゼをコードする核酸を、前記大脳皮質内のアストロサイトに投与することをさらに含み、前記リコンビナーゼポリペプチドが前記アストロサイトによって発現され、NeuroD1ポリペプチドをコードする前記核酸がリコンビナーゼ標的部位に挟まれている、請求項1〜21のいずれか一項に記載の方法。
