Abstract
Description

したがって、当該技術分野において、種々のC1エステラーゼ媒介性症候の治療に適する改良型の組換えヒトC1エステラーゼインヒビターが依然として必要とされている。
特定の実施形態では、例えば、以下が提供される:
(項目1)
配列番号1または配列番号2に対して少なくとも約50%、55%、60%、65%、70%、75%、80%、85%、90%、95%、96%、97%、98%、99%、または100%同一であるアミノ酸配列を有する精製された組換えヒトC1エステラーゼインヒビター(rhC1−INH)を含む組成物であって、前記精製された組換えヒトC1エステラーゼインヒビターが、血漿由来ヒトC1エステラーゼインヒビターと同様のまたはそれよりも長い半減期を有する、前記組成物。
(項目2)

本発明により産生されたrhC1−INHタンパク質またはポリペプチドは、シアル酸含有量及びグリカンマップなどの固有の特徴を有する。一部の実施形態では、組換え発現C1−INHタンパク質またはポリペプチドは、血漿由来C1−INHのグリコシル化プロファイルと同様のグリコシル化プロファイルを有する。一部の実施形態では、組換え発現C1−INHタンパク質またはポリペプチドは、組換え発現C1−INHタンパク質が血漿由来C1−INHの半減期と同様の半減期を提示するようなグリコシル化プロファイルを有する。一部の実施形態では、組換え発現C1−INHタンパク質またはポリペプチドは、組換え発現C1−INHタンパク質が血漿由来C1−INHの半減期よりも長い半減期を提示するようなグリコシル化プロファイルを有する。

本発明に適したrhC1−INHタンパク質を任意の使用可能な手段によって産生することができる。例えば、rhC1−INHタンパク質を、rhC1−INHタンパク質コード核酸を発現するように操作された宿主細胞系を利用することにより組換え産生することができる。別の方法として、または加えて、rhC1−INHタンパク質を、内因性遺伝子を活性化することによって産生してもよい。別の方法として、または加えて、rhC1−INHタンパク質を、化学合成によって部分的にまたは完全に調製してもよい。

特定の実施形態では、宿主細胞は、細胞培養用に選択される特定の条件下での特定の好ましい特質または増殖に基づいて、細胞株生成のために選択される。こうした特質は、樹立株(すなわち、特性決定された市販の細胞株)の既知の特徴及び/もしくは形質に基づいて、または実験による評価を通して確認され得ることを当業者は認識するであろう。一部の実施形態では、細胞株は、支持細胞層で増殖する能力に関して選択され得る。一部の実施形態では、細胞株は、懸濁液で増殖する能力に関して選択され得る。一部の実施形態では、細胞株は、細胞の接着単層として増殖する能力に関して選択され得る。一部の実施形態では、こうした細胞を、任意の組織培養容器または好適な接着基質で処理された任意の容器と共に使用することができる。一部の実施形態では、好適な接着基質は、コラーゲン(例えばコラーゲンI、II、II、またはIV)、ゼラチン、フィブロネクチン、ラミニン、ビトロネクチン、フィブリノーゲン、BDマトリゲル(商標)、基底膜マトリクス、デルマタン硫酸プロテオグリカン、ポリ−D−リジン及び/またはそれらの組み合わせからなる群から選択される。一部の実施形態では、接着宿主細胞は、特定増殖条件下で選択及び修飾されて、懸濁液中で増殖することができる。接着細胞を修飾して懸濁液中で増殖させるこうした方法は、当該技術分野で既知である。例えば、増殖培地から動物血清を経時的に徐々に除去することによって、細胞を懸濁液培養で増殖するように調節することができる。

種々の細胞培養培地及び条件を使用して、本発明の操作された細胞を用いてrhC1−INHタンパク質を産生することができる。例えば、rhC1−INHタンパク質を血清含有培地または無血清培地で産生することができる。一部の実施形態では、rhC1−INHタンパク質は無血清培地で産生される。一部の実施形態では、rhC1−INHタンパク質は、動物を含まない培地(すなわち動物由来の成分を含まない培地)で産生される。一部の実施形態では、rhC1−INHタンパク質は合成培地で産生される。本明細書で使用される場合、用語「合成栄養培地」は、その化学成分の実質的に全てが既知である培地を意味する。一部の実施形態では、合成栄養培地は、血清、血清由来タンパク質(例えば、アルブミンまたはフェチュイン)、及び他の成分などの動物由来成分を含まない。一部の実施形態では、合成培地は1つ以上のタンパク質(例えば、タンパク質増殖因子またはサイトカインなど)を含む。一部の実施形態では、合成栄養培地は、1つ以上のタンパク質加水分解物を含む。一部の実施形態では、合成栄養培地は、無タンパク質培地であり、すなわちタンパク質、加水分解物、または未知組成物の成分を含有しない無血清培地である。
所望の細胞発現C1−INHタンパク質を、まず当業者に周知の様々な方法のうちのいずれかによって初期培養で増殖することができる。細胞の生存、増殖、及び生存率に寄与する温度及び培地で成長させることによって細胞を増殖する。初期培養容量は任意のサイズであり得るが、最終産生で用いる産生バイオリアクターの培養容量よりも小さいことが多く、細胞を数回継代して培養容量を増加させてから、産生バイオリアクターに播種するのが一般的である。細胞培養液を攪拌または振盪して、培地の酸素化と、細胞に対する栄養物の分散とを増大することができる。別の方法として、または加えて、当該技術分野で周知の専用スパージング装置を用いて、培養物の酸素化を増強及び制御することができる。
典型的には、上述のように産生バイオリアクターに播種されると、細胞培養物は、細胞培養物の生存、増殖、及び生存率に寄与する条件下で初期増殖期の状態で維持される。本発明によると、産生バイオリアクターは、タンパク質の大規模産生に適切な任意の容量であり得る。後述の「バイオリアクター」についての小節を参照されたい。
一部の実施形態では、細胞は産生期に入る状態になると、培養条件は、関心対象の組換えタンパク質の産生を最大化するように変えられ得る。こうした培養条件の変更は、移行期に行われるのが典型的である。一部の実施形態では、こうした変更は、温度、pH、オスモル濃度、及び培地などを含むが、これらに限定されない幾種の培養条件のうちの1つ以上の変更であり得る。一実施形態では、培養のpHを変化させる。例えば、培地のpHを増殖期から産生期に向けて増加させるか、または減少させてもよい。一部の実施形態では、このpHの変化は急速である。一部の実施形態では、pHを長期間にわたって徐々に変化させる。一部の実施形態では、pHの変化は重炭酸ナトリウムの添加によって調節される。一部の実施形態では、pHの変化は、移行期の開始時に始まり、続く産生期の間も維持される。
本発明によれば、細胞培養が移行期を伴ってまたは伴わずに所望の細胞密度及び生存率に到達すると、細胞培養は、続く産生期の間、細胞培養物の生存及び生存率に寄与しかつ商業的に十分なレベルでC1−INHタンパク質を発現させるのに適正な培養条件下で維持される。
一部の実施形態では、産生期の間に、細胞培養液に栄養物か、または細胞により枯渇または代謝された他の培地成分を補充することが有益であるか、または必要になり得る。例えば、細胞培養液に栄養物か、または細胞培養の間に枯渇されたと見られる他の培地成分を補充することは有利である可能性がある。別の方法として、または加えて、産生期に先立って細胞培養液に補充することが有益であるか、または必要になり得る。非限定的な例として、酸化還元調節因子、増殖調節因子(例えば、ホルモン及び/または他の増殖因子)、グリコシル化調節因子、特定イオン類(例えばナトリウム、塩化物、カルシウム、マグネシウム、マンガン、リン酸塩)、緩衝液、ビタミン、ヌクレオシドもしくはヌクレオチド、微量元素(通例、極めて低最終濃度で存在する無機化合物)、アミノ酸、脂質、ペプトン、酵母抽出物、植物加水分解物、大豆加水分解物、血清、脂質補助剤、ヌクレオチド糖、ヌクレオチド糖前駆体、アンモニア、グルコサミン、ウリジン、シアル酸前駆体、N−アセチルマンノサミン、グルコース、ガラクトース、アミノ酸(例えば、グルタミン、システイン、イソロイシン、ロイシン、トリプトファン、バリン、アスパラギン、アスパラギン酸、グルタミン酸)、糖(例えば、グルコース、ガラクトース、シアル酸)、またはエネルギー源を、細胞培養液に補充することが有益であるか、または必要になり得る。
本発明は、rhC1−INHを産生するのに有用なバイオリアクターも提供する。バイオリアクターは、例えば、灌流型、バッチ型、流加培養型、反復バッチ型、または連続型(例えば、連続攪拌槽リアクターモデル)であり得る。バイオリアクターは、培地(例えば、合成栄養培地)を収容するように設計及び構成されたされた少なくとも1つの容器を含むのが一般的である。容器は、新鮮栄養培地を容器に流し入れるように設計及び構成された少なくとも1つの導入口を含むのが一般的である。容器はまた、廃棄培地を容器から流し出すように設計及び構成された少なくとも1つの排出口を含むのが一般的である。一部の実施形態では、容器は、容器内の単離された細胞が少なくとも1つの排出口から廃棄培地と共に排出される程度を最小化するように設計及び構成された、少なくとも1つのフィルタをさらに含んでもよい。バイオリアクターは、細胞増殖に適した条件を維持するように設計された1つ以上の他の構成要素を備えていてもよい。例えば、バイオリアクターは、栄養培地を容器内で循環または混合させるように設計及び構成された1つ以上の循環装置または混合装置を備えていてもよい。典型的には、rhC1−INHを発現するように操作された単離された細胞は、栄養培地に懸濁される。したがって、ある場合には、循環装置は、単離された細胞が確実に栄養培地中に懸濁されたままになるようにする。細胞を基体に付着させる場合がある。細胞を、栄養培地に懸濁された1つ以上の基体(例えば、マイクロビーズ)に付着させる場合がある。バイオリアクターは、容器から細胞懸濁液の試料を得るための1つ以上のポートを含んでもよい。バイオリアクターは、ガス(例えば、空気、酸素、窒素、二酸化炭素)含有量、流量、温度、pH、溶存酸素レベル、及び攪拌速度/循環速度などの条件を含めた培養条件を、モニタリング及び/または制御するための1つ以上の構成要素を伴って構成されていてもよい。
本発明の特定の実施形態では、実施者が、増殖細胞培養液の特定の条件を定期的にモニタリングすることが有益であるかまたは必要であると認識する場合がある。細胞培養条件をモニタリングすることにより、実施者は、細胞培養が組換えポリペプチドもしくはタンパク質を準最適なレベルで産生しているかどうか、または培養が準最適な産生期に入ろうとしているのかどうかを判定できるようになる。特定の細胞培養条件をモニタリングするために、分析用に培養液の少量アリコートを採取する必要がある。
様々な方法を用いて、本明細書に記載の種々の方法によって産生されたC1−INHタンパク質を精製または単離することができる。一部の実施形態では、発現C1−INHタンパク質は培地中に分泌されるため、精製プロセスの第1の工程として、例えば遠心分離または濾過によって細胞及び他の固形物を除去することができる。別の方法として、または加えて、発現C1−INHタンパク質は宿主細胞の表面に結合している。一部の実施形態では、ポリペプチドまたはタンパク質を発現する宿主細胞を精製用に溶解する。哺乳類宿主細胞の溶解は、ガラスビーズによる物理的破壊及び高pH条件への曝露を含めた当業者に周知の多くの手段によって成し遂げることができる。
本開示はまた、下流プロセスにおいて非標的タンパク質混入物または不純物を低減する方法を提供する。一実施形態では、下流プロセスは、rhC1−INHのプロセスである。非標的タンパク質混入物として、例えば、混入DNA、RNA、宿主細胞由来タンパク質(HCP)、及び/もしくはウイルス、ならびに/または標的タンパク質ではない任意の他の物質が含まれる。一実施形態では、非標的タンパク質混入物を、rhC1−INH下流プロセスに追加の精製工程を導入することによって低減する。追加の精製工程を伴わないrhC1−INHの1つの下流プロセスの例を図14に示す。
本発明は、本明細書に記載のrhC1−INHタンパク質と生理学的に許容される担体または賦形剤とを含有する医薬組成物をさらに提供する。担体及びrhC1−INHタンパク質は、無菌であり得る。製剤は、投与様式に適したものであるべきである。
本明細書に記載のrhC1−INHタンパク質(または本明細書に記載のrhC1−INHタンパク質を含有する組成物または薬剤)は、任意の適正な経路により投与される。一部の実施形態では、rhC1−INHタンパク質またはそれを含有する医薬組成物は、全身に投与される。全身投与は、静脈内、皮内、頭蓋内、鞘内、吸入、経皮(局所的)、眼内、筋肉内、皮下、筋肉内、経口、及び/または経粘膜的投与であり得る。一部の実施形態では、rhC1−INHタンパク質またはそれを含有する医薬組成物は、皮下に投与される。本明細書で使用される場合、用語「皮下組織」は、皮膚直下の疎性不規則性結合組織の層として定義される。例えば、皮下投与は、組成物を、以下に限定されないが、大腿部、腹部、殿部、または肩甲部を含めた領域に注射することによって行われ得る。一部の実施形態では、rhC1−INHタンパク質またはそれを含有する医薬組成物は、静脈内に投与される。一部の実施形態では、rhC1−INHタンパク質またはそれを含有する医薬組成物は、経口投与される。一部の実施形態では、rhC1−INHタンパク質またはそれを含有する医薬組成物は、頭蓋内に投与される。一部の実施形態では、rhC1−INHタンパク質またはそれを含有する医薬組成物は、鞘内に投与される。所望の場合には、2つ以上の経路を同時に用いることができる。
一部の実施形態では、組成物は、治療有効量で、及び/または特定の所望の結果(例えば、HAEなどの補体媒介性慢性疾患の予防)と相関する投与レジメンに従って投与される。
一部の実施形態では、rhC1−INHタンパク質は、補体媒介性疾患の治療に現在用いられている1つ以上の既知の治療剤(例えば、コルチコステロイド)と併用して投与される。一部の実施形態では、既知の治療剤(複数可)は、その標準的もしくは承認済投与レジメン及び/またはスケジュールに従って投与される。一部の実施形態では、既知の治療剤(複数可)は、その標準的もしくは承認済投与レジメン及び/またはスケジュールと比較して変更されたレジメンに従って投与される。一部の実施形態では、こうした変更されたレジメンは、1つ以上の単位用量が異なる(例えば減少または増加)という点で、及び/または投与頻度が異なるという点で(例えば、単位投与間の1つ以上の間隔を長くして低頻度にするか、または間隔を短くして高頻度にしているという点で)、標準的または承認済投与レジメンとは異なっている。
一部の実施形態では、本発明で提供される組換えタンパク質は、補体媒介性障害(例えば、NMOSD、AMR、またはHAE事象)に関連する急性発作に適している。これらの発作は長い場合もあり、短い場合もある。一部の実施形態では、疾患または障害は慢性的である。一部の実施形態では、本発明の組成物及び方法は予防的に用いられる。本明細書に開示される組成物及び方法を用いて治療され得る補体媒介性疾患の例として、以下に限定されないが、遺伝性血管性浮腫、抗体関連型拒絶反応、視神経脊髄炎関連疾患、外傷性脳損傷、脊椎損傷、虚血性脳損傷、火傷、中毒性表皮壊死症、多発性硬化症、筋萎縮性側索硬化症(ALS)、パーキンソン病、脳卒中、慢性炎症性脱髄性多発ニューロパチー(CIDP)、重症筋無力症、多巣性運動ニューロパチーが挙げられる。
ヒト血漿由来C1−INHのアミノ酸配列を用いて、CHO最適化ヌクレオチド配列を作製した。続いて、この配列をpXLG6発現ベクターに挿入し、CHO細胞に形質移入した。
懸濁液培養と無血清増殖条件とに適合したCHO細胞株を親細胞株として用いて、組換えヒトC1−INHを安定的に発現する細胞株を生成及び発生させた。
スケーラブルな産生プロセスの開発用にクローン/培地のロバストな組み合わせ(例えば、高容積産生性と所望のシアリル化度)を見つけ出すために、40個の市販培地の培地スクリーニングをクローン番号1及びクローン番号31を用いて実施検討した。
4種のフィードについて、細胞増殖及びC1−INH発現への影響を評価するために検討した。これらは、表1に記載のFeed3、PW2、Pep1510、及びPep4601である。細胞生存率及び力価を7日目と10日目とに各条件について測定した。Feed3とPep1510との混合物(Feed3:Pep1510(v/v)=9:1)が、実験培養条件可で力価と生存率の両方を最大化した。
クローン番号31によるC1−INHの大規模産生を最適化するために、いくつかの産生を実行し種々のパラメータを評価した。培養液のフィードに至適な混合物は、1容量のHyPep1510(本明細書では「Pep1510」と称される)(Sheffield Bioscience社、カタログ番号5X59053、水中200g/l)に対して、9容量のimMEDIAte ADVANTAGE(本明細書では「Feed3」と称される)(Sigma社(SAFC)、カタログ番号8383C)であることを見出した。PowerCHO1培地と共にFeed3+Pep1510混合物を用いることで、10日目におよそ1.3g/l(90%の生存率)の収率、また14日目に約2.0g/l(87%の生存率)の収率を得た。PCO2及びオスモル濃度を最小化するために、pHをHCl/炭酸塩で制御した。空気と5%CO2の混合ガスを0.03vvmで連続的に吹き込み、pHを7.2に維持した。
細胞により発現したrhC1−INHを精製するプロセスには、収穫物の濃縮、それに続く溶媒界面活性剤処理(SD)を用いた濃縮された収穫物のウイルス不活性化、3種のクロマトグラフィー下流精製工程、ウイルスフィルタ低減工程、及び最終濃縮/透析濾過工程が含まれ得る。図2は、rhC1−INH精製の例示的なプロセスを示す。
C1−INHのインタクトな分子量のおよそ50%がグリカンである。6つのN結合型部位(セルピンドメインに3つ(Asn216、Asn231、Asn330)及びN末端ドメインに3つ(Asn3、Asn47、Asn59))と、8つのO結合型部位(N末端ドメインに全て(Ser42、Thr25、Thr26、Thr49、Thr61、Thr66、Thr70、Thr74))とがある。
混合モード順相/陰イオン交換クロマトグラフィー(NP/AE)によるN−グリカン分析法(NGA)を用いることで、分類されたグリカンを、主に電荷(シアル酸を含むため)に基づいて、さらにはサイズ、オリゴ糖組成、及び結合に基づいて分離することによって、rhC1−INHのN結合型グリカン含有物プロファイルを得た。溶出ピークをシアル酸含有量に基づき大まかに「電荷クラスター」に分類したが、電荷クラスター内のピークは、サイズ、オリゴ糖組成、または結合という点で様々である。蛍光検出器を備えた高速液体クロマトグラフィー(HPLC)システムを用いてグラジエント分離を行った。得られたクロマトグラムを積分することにより、シアル酸含有量に基づいて、相対的なN−グリカン分布の再現性の高い定量が可能であった。エラー!参照元が見つかりません。図7は、シアル酸含有量に基づいて分類されたピーク群を持つrhC1−INHプロファイルの例を表す。観測されたピーク群の帰属を、分取したピーク画分のオフラインMALDI−TOF質量分析で確認した(図8)。
親水性相互作用液体クロマトグラフィー(NGA−HILIC)を用いたN−グリカン分析法(NGA)により、主にグリカンサイズに基づいて、さらには単糖含有量、及び結合に基づいて、C36のN結合型グリカン含有物の高分解能プロファイルを得た。蛍光検出器を備えた高速液体クロマトグラフィーシステムを用いてグラジエント分離を行った。得られたクロマトグラムを積分することにより、相対的なN−グリカン分布の再現性の高い定量が可能であった。
O−グリカン分析をORELA遊離試薬(Ludger社)を用いて行った。ORELA試薬を用いて化学的にグリカンを遊離した結果として、アスパラギン結合型(N結合型)グリカン、及びセリン/トレオニン結合型(O結合型)グリカンから構成されるグリカン試料を得る。ORELA試薬を用いて生成した評価試料の分析結果を、PNGaseFを用いて生成した試料と比較して、全グリカンプロファイルをN結合型(単独)グリカンプロファイルと比較できるようにする。したがって、付加的なグリカンピークは、rhC1−INHのO結合型グリコシル化部位からの寄与を表す。この分析を親水性相互作用クロマトグラフィー(HILIC)を用いて行う。
例示的なrhC1−INHタンパク質の構造の特徴を、種々の生理化学的方法を用いて明らかにした。1次構造及び翻訳後修飾を、脱N−グリコシル化したrhC1−INHのペプチドマッピングLCMS、MS/MSにより評価した。この方法により、野生型C1−INH(E165Q)とは単一のアミノ酸が異なっている、予測されたアミノ酸配列が確認された。O結合型グリカンをこの方法により同定し、LCに基づくグリカンマッピングによって別途確認した。
rhC1−INHタンパク質の活性を、TECHNOCHROM(登録商標)C1−INH試薬キット(Technoclone社、オーストリア、ウィーン)などの発色診断キットを用いて評価することができる。市販の診断キットを、国際標準と対照してかつ参照標準に対してrhC1−INHの効力を測定するために、完全曲線発色アッセイに改良した。
一実施形態では、150mMのグリシンと、50mMのソルビトールと、50mMのリン酸ナトリウム緩衝液(pH7.1)との中に70mg/mLのrhC1−INHを含有する水溶液を調製した。タンパク質濃度は、rhC1−INHの溶解性及び安定性プロファイルに基づき、最大の臨床用量を提供するように選択された。50mMのリン酸ナトリウム(pH7.1)は、温度ストレス下で安定性を持つことから選択された。150mMのグリシン及び50mMのソルビトールは、至適なタンパク質安定性だけでなく、静脈内及び皮下投与用の適正な等張性も得られるように選択された。
ウサギに血漿由来C1−INHまたはrhC1−INHを注射した。rhC1−INHは、10〜20%の中性グリカン、約26%のモノシアリル化グリカン、約35%のジシアリル化グリカン、約17%のトリシアリル化グリカン、及び約5%のテトラシアリル化グリカンというグリコシル化プロファイルを有していた。ウサギPK試験の結果を図13に示す。
14日ラット毒性/TKと14日回復期間試験において、150mMのグリシンと、50mMのソルビトールと、50mMのリン酸ナトリウム緩衝液(pH7.1)との中に70mg/mLのrhC1−INHを含有する水溶液として製剤化されたrhC1−INHを、ラットに500U/kg/日、1000U/kg/日、または2000U/kg/日で投与した。各群は10匹のラットを含んだ。全ての動物が生存した。rhC1−INHに関連して及ぼされる、体重、摂食量、または臨床病理学パラメータ(血液学、凝固、血清化学、及び尿分析)への影響はいずれも見られなかった。rhC1−INHに関連した眼科的所見、巨視的観察、臓器重量の変化、または組織学的変化はいずれも見られなかった。抗薬物抗体を検出したが、非中和性であり、曝露には影響を及ぼさなかった。無毒性量(NOAEL)は、評価した最大投与量である2000U/kg/日であった。
本実施例では、組換えヒトC1インヒビタータンパク質(rhC1−INH)を精製するための混入物(例えば、DNA及び/または宿主細胞由来タンパク質)低減工程の開発について詳細に述べる。現行の下流プロセスの概要を図14に示す。
UV分光光度法によるタンパク質含有量の算出
馴化培地を含まない全試料のタンパク質濃度を、測定された280nmでの吸収と、吸光係数ε280=0.514mL/(mg×cm)とを用いて算出した。
TME−0499−01(チャイニーズハムスター卵巣細胞タンパク質(CHOP)ELISA−第3世代)
CHOのDNA分析をまずPico Greenにより行った。この方法は、超高感度蛍光核酸染色法である。dsDNAを含み得る試料をまず希釈して、使い捨てのキュベットでTE緩衝液の最終容量を1.0mlにする。実験試料の希釈を行って、特定の混入物(例えば、NaCl)の干渉の影響を減らす。続いて、Quant−IT PicoGreen試薬の1.0ml希釈標準水溶液をキュベットに加え、遮光しながら2〜5分間インキュベートする。試料の蛍光を分光蛍光光度計で標準フルオレセイン波長(励起波長:約480nm、発光波長:約520nm)にて測定する。未知試料のシグナルを、同時にアッセイしたdsDNA標準と比較することによって正確に定量する。また、CHO DNA分析をqPCRによって行った。
表9は、rhC1−INH発生用のプロセス中間体及び最終バルクの既存のDNAレベルを示す。データは、DNAレベルがプロセスでの最初のカラム工程の後に検出限界値未満であることを示した。しかしながら、最終バルクの濃度が70g/Lまでになると、全ての試験で測定できるDNAレベルとなった。
POROS XS溶出物を、AEX吸収体を適用するのに至適であるとして特定した。POROS XS溶出物は、rhC1−INHのpIが約2.7〜3.6と低いことを考慮すると、Gigacap Q溶出物に対するpH7.5と比べてpH6で、AEX吸収体の負荷としてより好適であると考えられた。AEX膜吸収体の理想形態は、負に帯電した混入物を結合させる一方で、rhC1−INHを装置を貫通させて流すものである。pHが低い実行条件により、混入物を結合させつつ、タンパク質が膜に結合する可能性を減じた。導電性はPOROS XS溶出物を選択した別の理由である。高負荷導電率は一般的にAEX膜クロマトグラフィーに望ましくないが、これは、混入物の膜への親和性が減少するからである。POROS XS溶出物の導電率は、GigacapQ溶出物よりもわずかに低いのが通例であり、それ故にこの点からさらに好適である。さらに、GigacapQ溶出物は50mMのリン酸塩という二価の負に帯電した緩衝液で処理される。高いリン酸塩濃度条件は、その高イオン強度と複数の負電荷とが陰イオン交換クロマトグラフィーと干渉する可能性があるため、極めて悪い条件と考えられる。
Sartobind Q膜、Sartobind STIC膜、及びNatrix膜に対してフロースルーモードでAEX吸収体工程を実施できるかどうかを判定するために、プロファイルを実行させた。
産物の試験を、Sartobind Q吸収体とSartobind STIC吸収体とを比較して行った。Sartobind QとSartobind STICとを、平衡状態かつ予想されるPOROS XS溶出条件に合わせた洗浄条件で評価した。AEX膜吸収体は使い捨てになるように設計されているが、これらの実験に高導電性洗浄及びストリップを含めることで、低イオン強度で膜が保持した材料の定量を可能にしている。これらの産物試験の実験条件を以下の表10に示す。
Sartobind QとSartobind STICとを、平衡状態でかつ予想されるPOROS XS溶出条件に合わせた洗浄条件で評価した。これらの実験に高導電性洗浄及びストリップを再度含めることで、低イオン強度で膜が保持した材料の定量を可能にしている。
GigacapQの後の膜の性能を評価するために、追加の実験を行った。
要約すると、この実施例は、rhC1−INH下流プロセスの新規なDNAクリアランス工程を特定することを目的とした一連の実験について詳細に述べている。陰イオン交換クロマトグラフィーはDNAクリアランス工程としての一選択肢であるが、これは、DNAの極めて低いpIにより、ほぼ全てのpHでDNAが陰イオン交換リガンドに結合するためである。しかしながら、rhC1−INHの低いpIにより、rhC1−INHも陰イオン交換膜に結合する可能性もある。Sartobind Q、Sartobind STIC、及びNatrixのAEX膜吸収体の適正を、現行のrhC1−INHプロセスの第2のカラム工程の後に評価した。Natrixは、rhC1−INHがその膜に結合し、その結果、この膜に対して低収率になったことから、初期に検討から除外された。Sartobind Q及びSartobind STICをDNAスパイク試験でさらに評価し、Sartobind Qが、STICと比較して優位な収率とDNAクリアランスとを有することから選択された。さらなる実験によって、Sartobind Qは、収率及びDNAクリアランスに関して優れた性能を有することが示された。
