Abstract
Description

2 mmφゲージサイズでふるいにかけた空気乾燥鎌島土壌100 gと150 mlの蒸留水(DW)とを、250 ml容量の遠心分離管中で、機械式シェーカーで30分間、均一になるまで混合した。次に、7秒静置後、デカンテーションを30秒間で行うことにより、粗大粒子を除去した。デカンテーションによって得られた微粒子を、次の方法で化学洗浄した。すなわち、0.1N NaOH水溶液150 mlで2回洗浄し、2%HF水溶液150 mlで4回洗浄し、0.1N NaOH水溶液75 mlで2回洗浄し、2%HF水溶液75 mlで2回洗浄し、最後に0.1N NaOH水溶液で4回洗浄した(各洗浄は24時間)。次に、得られた残渣を蒸留水で2回洗浄した(各洗浄は12時間)。残渣を含む溶液の中和は、0.1 N HClを使用してpH 7.0±0.2まで行った。得られた残渣を蒸留水で2回洗浄した(各洗浄は12時間)。得られた残渣を、3日間、凍結乾燥した後、セラミック乳鉢と乳棒を使用して細かく粉末化した。このようにして得られた粉末を、固体腐植ヒューミンとして以下の参考例及び実施例で使用した。
水田土集積物を、固体腐植ヒューミン、酢酸、及びペンタクロロフェノールを含有する無機培地で、窒素及び二酸化炭素からなる雰囲気下で培養した。継代と共に酢酸濃度を徐々に減らし、最終的に酢酸を含有しない培地で培養を続け、被検細菌を得た。
被検細菌を、1gの固体腐植ヒューミンを含有する又は含有しない50 mlの嫌気性増殖培地で培養した。
培養液量 50mL、固体腐植ヒューミン添加量 1g より
固体腐植ヒューミン1gが酢酸生成菌に電子供与して微生物反応を起こす能力
10-15μmol/gの酢酸生成、これは
40-60μmol/gの水素ガス供給量に相当する(反応式より、酢酸の4倍)
即ち、1gのヒューミンで40-60μmolの水素ガスを供給したことに相当
4H2+2HCO3 -+2H+⇔CH3COO-+H++4H2O (標準電位(pH7):-279mV)
20-30μmol/gの二酸化炭素固定量に相当する(反応式より、酢酸の2倍)。
メタン生成反応 (標準電位(pH7):-238mV)
4H2+HCO3 -+H+⇔CH4+3H2O
水素は生成するメタンの4倍を消費:120μmol/L相当
ギ酸生成反応
H2+HCO3 -+H+⇔HCOO-+H++H2O
水素は生成ギ酸と等モルを消費:4μmol/L相当。
培地として、NaHCO3に代えてMOPSを使用した培地を使用したこと以外は実施例1(固体腐植ヒューミン使用の場合)と同様に培養した場合、及びMOPSを使用し且つヘッドスペースの雰囲気を窒素のみとしたこと以外は実施例1(固体腐植ヒューミン使用の場合)と同様に培養した場合についても、生成物を分析した。
Claims (13)
- 酢酸生成菌及びメタン生成菌からなる群より選択される少なくとも1種を含む細菌を、無機炭素源及び固体腐植ヒューミンを含有する培地中で培養する工程を含む、二酸化炭素を固定する方法。
- 前記培養が嫌気的条件下で行われる、請求項1に記載の方法。
- 前記培養が独立栄養条件下で行われる、請求項1又は2に記載の方法。
- 前記培養が非電気培養である、請求項に記載の方法。
- 前記酢酸生成菌がFirmicutes門細菌、Proteobacteria門細菌、Bacteroidetes門細菌、Actinobacteria門細菌、Chloroflexi門細菌、Acidobacteria門細菌、及びSpirochaetes門細菌からなる群より選択される少なくとも1種の細菌を含む、請求項1〜4のいずれかに記載の方法。
- 前記固体腐植ヒューミンが土壌の化学洗浄残渣である、請求項1〜5のいずれかに記載の方法。
- 前記化学洗浄がアルカリ洗浄を含む、請求項6に記載の方法。
- 前記洗浄がアルカリ洗浄及び酸洗浄を含む、請求項6又は7に記載の方法。
- 前記工程により酢酸を生成させる、請求項1〜8のいずれかに記載の方法。
- 酢酸生成菌及びメタン生成菌からなる群より選択される少なくとも1種を含む細菌を、無機炭素源及び固体腐植ヒューミンを含有する培地中で培養して二酸化炭素固定を行う工程、並びに前記二酸化炭素固定を経て生成された炭素化合物を回収する工程を含む、炭素化合物の製造方法。
- 前記炭素化合物が酢酸、メタン、及びギ酸からなる群より選択される少なくとも1種を含む、請求項10に記載の製造方法。
- 固体腐植ヒューミンを含有する、請求項1〜9に記載の方法又は請求項10若しくは11に記載の製造方法に用いるための電子供与剤。
- 固体腐植ヒューミンを含有する、請求項1〜9に記載の方法又は請求項10若しくは11に記載の製造方法に用いるための培地。
