Abstract
Description
漿膜と、
該漿膜の移植面側に積層された細胞シートと、
からなる、創傷被覆用シートを提供する。
本技術に係る創傷被覆用シートに用いる前記漿膜としては、腹膜を用いることができる。
本技術に係る創傷被覆用シートは、縫合又は吻合された創傷部に用いることができる。
また、本技術に係る創傷被覆用シートは、管腔臓器の創傷部に用いることができる。
この際、本技術に係る創傷被覆用シートは、前記管腔臓器の外壁に移植することができる。
該漿膜の移植面側に積層された細胞シートと、
からなる、創傷治療用シートを、創傷部へ移植することにより創傷を被覆する、創傷被覆方法を提供する。
本技術に係る創傷被覆方法では、前記創傷治療用シートを、創傷部へ縫合することにより移植することができる。
本技術に係る創傷被覆方法に用いる前記漿膜としては、腹膜を用いることができる。
本技術に係る創傷被覆方法を用いれば、縫合又は吻合された創傷部の被覆をすることができる。
また、本技術に係る創傷被覆方法を用いれば、管腔臓器の創傷部の被覆をすることができる。
この際、前記創傷被覆用シートは、前記管腔臓器の外壁に移植することができる。
本技術に係る創傷被覆用シートに用いる漿膜は、本技術の効果を損なわない限り、生体から採取可能な漿膜を、創傷部の位置等に応じて、自由に選択して用いることができる。例えば、腹膜、胸膜、心膜等の漿膜を、本技術に係る創傷被覆用シートに用いることができる。
本技術に係る創傷被覆用シートに用いる細胞シートは、本技術の効果を損なわない限り、一般的な細胞シートを、創傷部の位置等に応じて、自由に選択して用いることができる。より具体的には、細胞培養基材上で培養し、細胞培養基材上から剥離することで得られる1層又は複数層のシート状の細胞群からなるシートを用いることができる。

(1)脂肪由来幹細胞の分離
脂肪由来幹細胞の分離はWatanabe N.らの論文(Watanabe N.,et al.Genetically Modified Adipose Tissue−Derived Stem/Stromal Cells,Using Simian Immunodeficiency Virus−Based Lentiviral Vectors,in the Treatment of Hemophilia B.Hum Gene Ther.2013 Mar;24(3):283-294.)に記載の方法に従った。
細胞播種後3日目に培地交換を行い、5日目に0.25%トリプシンで細胞を剥がした後に10枚の75cm2フラスコに継代した。継代後は2〜3日後で再度継代を行った。2〜3日後に再度0.25%トリプシンで細胞を剥がし、細胞カウントを行った後に、2.3×106個の細胞を2mLの培地に懸濁し、35mm UpCell(登録商標)に播種し、37℃で2日間培養した。2日後に16.4μg/mLアスコルビン酸入り培地で培地交換を行った。更に2日後にアスコルビン酸入り培地で培地交換を行い、細胞シート移植直前に20℃のインキュベーターで20〜30分インキュベートすることで細胞をシート状に回収した。
SD系ラットから、腹膜を採取し、前記で作製した細胞シートと積層させて、創傷被覆用シートを作製した。
SD系ラットの中央背部の筋膜を除去し、筋肉を露出させた。前記で作製した創傷被覆用シートを、背筋に4回縫合することにより移植した。また、比較のために、細胞シートのみ、及び、腹膜のみを背筋に貼付することにより移植した。
手術直後、手術後3日目、及び手術後7日目に、移植後の背筋の様子を観察した。図1は、創傷被覆用シート、細胞シートのみ、及び腹膜のみを移植した直後の背筋の写真である。図2は、創傷被覆用シート、細胞シートのみ、及び腹膜のみを移植した3日後の背筋の写真である。図3は、創傷被覆用シート、細胞シートのみ、及び腹膜のみを移植した7日後の背筋の写真である。
手術後3日目及び7日目に摘出したサンプルをコンパウンドに包埋後、液体窒素で凍結し組織切片を作製した。ヘマトキシリン・エオジン染色、緑色蛍光タンパク質(GFP)染色、カルレチニン染色を行った。図4は、ヘマトキシリン・エオジン染色の結果を示す顕微鏡写真である。図5は、緑色蛍光タンパク質(GFP)染色の結果を示す顕微鏡写真である。図6は、カルレチニン染色の結果を示す顕微鏡写真である。

- 創傷部へ移植することにより、創傷の被覆に用いるシートであって、
漿膜と、
該漿膜の移植面側に積層された細胞シートと、
からなる、創傷被覆用シート。 - 前記漿膜は、腹膜である、請求項1に記載の創傷被覆用シート。
- 前記創傷部は、縫合又は吻合された創傷部である、請求項1又は2に記載の創傷被覆用シート。
- 前記創傷部は、管腔臓器の創傷部である、請求項1から3のいずれか一項に記載の創傷被覆用シート。
- 前記管腔臓器の外壁に移植される、請求項1から4のいずれか一項に記載の創傷被覆用シート。
- 漿膜と、
該漿膜の移植面側に積層された細胞シートと、
からなる、創傷治療用シートを、創傷部へ移植することにより創傷を被覆する、創傷被覆方法。 - 前記創傷治療用シートを、創傷部へ縫合することにより移植する、請求項6に記載の創傷被覆方法。
- 前記漿膜は、腹膜である、請求項6又は7に記載の創傷被覆方法。
- 前記創傷部は、縫合又は吻合された創傷部である、請求項6から8のいずれか一項に記載の創傷被覆方法。
