本発明は、腫瘍または癌を治療するための方法および薬剤、特に、DNA依存性タンパク質キナーゼ触媒サブユニット(DNA-dependent protein kinase catalytic subunit)(DNA−PKcs)を阻害することにより腫瘍または癌を治療するための方法および薬剤に関する。
腫瘍は、複数の因子が役割を果たし、関与し、多段階で発症する疾患である。化学療法は、腫瘍の治療に重要な役割を果たす。しかしながら、腫瘍細胞は、化学療法薬に対して耐性を生じやすく、これが癌患者の治療に重大な影響を及ぼす。アルキル化剤、白金錯体およびトポイソメラーゼ阻害剤などの慣用化学療法薬は、標的としてDNAを用い、DNA損傷を引き起こしてアポトーシスを誘導することにより腫瘍細胞を死滅させる。従って、DNA損傷の修復は、化学療法薬に対する腫瘍細胞の耐性の重要な理由となると考えられる。化学療法薬により引き起こされるDNA損傷には、塩基の損傷、一本鎖切断(single-strand break)(SSB)、二本鎖切断(double-strand break)(DSB)などを含む多くの形態があるが、このうち、DSB傷害が最も深刻である。哺乳動物細胞では、DSB損傷の修復には、相同組換え(Homologous Recombination)(HR)と非相同末端結合(Nonhomologous End Joining)(NHEJ)の2つの主要な形態がある。HR修復プロセスは、鋳型として姉妹染色分体を用いて損傷したDNA二本鎖を正確に修復するが、細胞周期のS期およびG2期においてのみである。HRに比べ、NHEJは、細胞周期のいずれのステージでも実行可能なより保存的な修復機構である。この修復は相同鋳型を必要とせず、タンパク質の相互作用を用いて損傷した末端を直接接続するものであり、哺乳動物細胞におけるDSB修復の主要な機構である。NHEJの基本プロセスは二本鎖切断後に起こるものであり、Ku70/Ku80がDSB傷害部位の末端に結合し、次いで、KuがDNA−PKcsを動員してDNA−PK複合体を形成し、この複合体がNHEJ修復機構を開始する。DNA−PKcsは、自己リン酸化と下流タンパク質のリン酸化を介して修復シグナルを伝達する。DNA末端処理タンパク質(ヌクレアーゼ、ポリメラーゼなど)は、崩れた末端を連結に適した構造へと処理し、最後に、リガーゼIV−XRCC4−XLF複合体が連結機能を遂行する。それらの中で、DNA−PKcsは、DNA−PK複合体の触媒サブユニットであり、ホスファチジルイノシトール−3−キナーゼ様キナーゼ(phosphatidylinositol-3-kinase-like kinase)(PIKK)ファミリーのメンバーであり、NHEJ修復に中枢的役割を果たす。DNA−PKcsの阻害がDNA傷害薬に対する腫瘍細胞の感受性を著しく高め得ることを示した研究がある(Elaine Willmore, et al. Blood. 2004;103: 4659-4665, Yan Zhao, et al. Cancer Res 2006; 66(10): 5354-62)。DNA−PK阻害剤もまた継続的に開発され、いくつかのDNA−PK阻害剤は臨床試験に入っている。腫瘍を治療するための、化学療法薬と組み合わせたDNA修復阻害剤の使用は有望な分野である。

本発明において、エンドスタチンは、腫瘍細胞に直接作用し、化学療法薬に対する腫瘍細胞の感受性を調節し得ることが分かる。イン・ビトロ細胞学試験において、エンドスタチンは、P53欠損腫瘍細胞の活性および細胞コロニー形成能に対する化学療法薬の阻害効果を有意に増強し、かつ、化学療法薬により引き起こされる腫瘍細胞のアポトーシスを促進し得ることが分かる。一方、マウス異種移植腫瘍モデルでは、エンドスタチンは、P53欠損非小細胞肺癌異種移植片に対する化学療法薬の増殖阻害効果を有意に増強し、担癌マウスの生存を延長し得る。
本実施例で使用したCNBr活性化セファロース4Bは、Sigma社から購入した。まず、説明書に従い、エンドスタチンおよびウシ血清アルブミン(BSA)をそれぞれCNBr活性化セファロース4Bに結合させ、A549細胞溶解液とともにインキュベートした。プルダウンされたタンパク質に対してSDS−PAGE電気泳動を行い、クーマシーブルーで染色した。質量分析により、DNA−PKcsを、エンドスタチンと相互作用したタンパク質として同定した(図1A)。次に、免疫ブロット試験により、H1299細胞およびA549細胞の両溶解液中のDNA−PKcsタンパク質が、エンドスタチンが結合したCNBr活性化セファロース4Bによって沈降可能であることが実証された(図1B)。この2者間の相互作用をさらに確認するために、H1299細胞およびA549細胞を、エンドサイトーシスのためにエンドスタチンとともに2時間インキュベートした。細胞溶解液にDNA−PKcsの抗体を混合した後、プロテインA/Gビーズと同時インキュベートした。免疫ブロットアッセイにより、エンドスタチンはDNA−PKcsによって共沈降し得ることが確認され、これにより、2者間の相互作用が証明された(図1C)。
本実施例では、DNA−PKcsの既知の基質であるAKTおよびP53を、DNA−PKcs経路に対するエンドスタチンの阻害効果を確認するための検出指標として使用した。腫瘍細胞(非小細胞肺癌H1299細胞およびA549細胞を含む)を6つの処理群に分けた。第1群:陰性対照;第2群:エトポシド(10μM、1時間)処理;第3群:エンドスタチン(10μg/ml、1時間)処理;第4群:エンドスタチン(10μg/ml、エトポシドの1時間前に添加)とエトポシド(10μM、1時間)による併用処理;第5群:NU7026(10μM、1時間)処理;第6群:NU7026(10μM、エトポシドの1時間前に添加)とエトポシド(10μM、1時間)による併用処理、なお、NU7026はDNA−PKcs阻害剤であった。免疫ブロットアッセイ結果(図2、p−DNA−PKcs(S2056)は、DNA−PKcsのリン酸化レベルを表し、S2056は、そのリン酸化部位を表し;p−AKT(S473)は、AKTのリン酸化レベルを表し、S473は、そのリン酸化部位を表し;p−P53(S15)は、P53のリン酸化レベルを表し、S15は、そのリン酸化部位を表す)は、対照群に比べて、エトポシド処理は、DNA−PKcsシグナル伝達経路のレベルを活性化し;エンドスタチン処理は、DNA−PKcsシグナル伝達経路のレベルに影響を及ぼさず;エンドスタチンとエトポシドの組合せは、エトポシドによるDNA−PKcsシグナル伝達経路の活性化を阻害したことを示した。NU7026処理は、DNA−PKcsシグナル伝達経路のレベルに影響を及ぼさず;NU7026とエトポシドの組合せは、エトポシドによるDNA−PKcsシグナル伝達経路の活性化を阻害した。
本実施例では、DSBs損傷の程度を表すためにγH2AXのレベルを使用した。腫瘍細胞(H1299細胞およびA549細胞を含む)を8つの処理群に分けた。第1群:陰性対照、細胞を0時間で回収した;第2群:細胞をエトポシド(10μM)処理1時間の後に回収した;第3群:エトポシド(10μM)処理1時間後に、それを新鮮培地に置き換え、細胞を2時間後に回収した;第4群、エトポシド(10μM)処理1時間後に、それを新鮮培地に置き換え、細胞を5時間後に回収した;第5〜第8群はまずエンドスタチン(10μg/ml)で1時間前処理し、その後、処理はそれぞれ第1〜第4群に相当した。免疫ブロットアッセイ結果は、エトポシド処理細胞によりもたらされたDNA損傷が第1〜第4群において培地から薬物を除去した後に徐々に修復され、γH2AXレベルは5時間にベースラインに戻ったことを示した。第5〜第8群では、エンドスタチンの併用はγH2AXレベルの低下を遅延させ、これは細胞におけるDSBs損傷の修復が遅延されたことを証明した(図3)。
本実施例では、腫瘍細胞(H1299細胞およびA549細胞を含む)を8つの処理群に分けた。第1〜第4群では、細胞を対照siRNAでトランスフェクトし、最後の4群では、細胞を、DNA−PKcsを特異的にノックダウンしたsiRNAでトランスフェクトした。DNA−PKcsのsiRNA配列は:フォワード5’−CAGGGUUUAAUCAGAAUAUTT−3’、リバース5’−AUAUUCUGAUUAAACCCUGTT−3’である。siRNAトランスフェクション法は、トランスフェクション試薬リポフェクタミン2000の説明書に従って行った。次に、第8群では、細胞を薬物で処理した。第1群および第4群:対照群;第2群および第5群:エンドスタチン(10μg/ml、16時間)処理;第3群および第6群:エトポシド(H1299では10μM、A549では1μM、16時間)処理;第4群および第8群:エンドスタチン(10μg/ml、エトポシドの1時間前に添加)とエトポシド(H1299では10μM、A549では1μM、16時間)の併用。細胞を回収し、アネキシンV−FITC/PIアポトーシス検出キットの説明書に従って操作を行った。アポトーシスはフローサイトメトリーにより検出した。A549細胞はP53野生型細胞であり、H1299細胞はP53欠損細胞であった。結果は、A549細胞は化学療法薬に対する感受性がより高く、同程度のアポトーシスが生じた場合、H1299細胞のエトポシド濃度はA549細胞のエトポシド濃度よりはるかに高かったことを示した。これに基づけば、エンドスタチンの付加は、エトポシドにより誘導されるアポトーシスをさらに促進すると考えられる(図4)。DNA−PKcsをノックダウンした後には、このエトポシドにより誘導されるアポトーシス促進におけるエンドスタチンの役割は減弱した。
まず、レンチウイルスキット(Shanghai Genepharma社から購入)を用いてP53安定過剰発現H1299細胞株(H1299−P53)およびP53安定ノックダウンA549細胞株(A549−NC)を構築した。A549細胞においてP53をノックダウンするために使用したshRNAにより標的化されるP53配列は、5’−GACTCCAGTGGTAATCTAC−3’であった。ウイルストランスフェクションおよび安定細胞株の構築はキットの説明書に従って行った。構築された腫瘍細胞(H1299−NC、H1299−P53、A549−NC、A549−shP53)を96ウェルプレート(1000細胞/ウェル)に播種し、8つの処理群に分けた。第1群〜第4群では、種々の濃度のエトポシド(0μM、0.1μM、1μM、10μM)を含有する培地を各ウェルに加えた。第5群〜第8群は、まず、10μg/mlのエンドスタチンを含有する培地で1時間前処理し、次に、種々のエトポシド濃度を含有する培地を、第5群〜第8群のエトポシド終濃度がそれぞれ0μM、0.1μM、1μM、および10μMとなるように加えた。12時間の処理後、薬物含有培地を通常培地に置き換え、細胞をさらに72時間培養した。細胞活性を、CCK8キットの説明書(同仁堂、東京、日本)に従って測定した。結果は、P53欠損腫瘍細胞では(図5AおよびD)、エンドスタチンは腫瘍細胞に対するエトポシドの毒性を増強し得ることを示した。正常なP53機能を有する腫瘍細胞では(図5BおよびC)、エトポシドは、腫瘍細胞の活性を有意に阻害することができ、エンドスタチンは、エトポシドの阻害効果を有意には増強しなかった。
腫瘍細胞(H1299−NC、H1299−P53、A549−NC、A549−shP53)を4つの処理群に分けた。第1群:陰性対照;第2群:エンドスタチン(10μg/ml、16時間)処理;第3群:エトポシド(1μM、16時間)処理;第4群:エンドスタチン(10μg/ml、エトポシドの1時間前に添加)とエトポシド(1μM、16時間)による併用処理。処理後、細胞をトリプシンで消化し、計数し、6ウェルプレートに1000細胞/ウェルで再播種した。細胞を通常培地で14日間培養して細胞コロニーを形成させた。細胞コロニー計数値の結果は、P53欠損腫瘍細胞では(図6AおよびD)、エンドスタチンが腫瘍細胞コロニー形成能に対するエトポシドの阻害効果を増強し、腫瘍細胞の生存力を低下させ得ることを示した。正常なP53機能を有する腫瘍細胞では(図6BおよびC)、エトポシドは、腫瘍細胞のコロニー形成能を有意に阻害することができ、エンドスタチンは、エトポシドの阻害効果を有意には増強しなかった。
腫瘍細胞(H1299−NC、H1299−P53、A549−NC、A549−shP53)を4つの処理群に分けた。第1群:陰性対照;第2群:エンドスタチン(10μg/ml、16時間)処理;第3群:エトポシド(1μM、16時間)処理;第4群:エンドスタチン(10μg/ml、エトポシドの1時間前に添加)とエトポシド(1μM、16時間)による併用処理。培養培地に浮遊しているアポトーシス細胞および接着細胞を回収し、これらの細胞を、0.05mg/mlのヨウ化プロピジウム(PI)を含有する500μlのPBSに懸濁させ、室温、暗所で15分間染色し、フローサイトメトリー(BD FACS Calibur)により検出した。PI陽性細胞がアポトーシス細胞を表す。結果は、P53欠損腫瘍細胞では(図7AおよびD)、エンドスタチンは、腫瘍細胞のアポトーシスを促進するエトポシドの能力を増強し得ることを示した。正常なP53機能を有する腫瘍細胞では(図7BおよびC)、エトポシドは、腫瘍細胞のアポトーシスを有意に促進することができ、エンドスタチンは、エトポシドのアポトーシス誘導効果を有意には増強しなかった。
本実施例では、エンドスタチンが化学療法に対するP53欠損腫瘍細胞の感受性を高め得ることを確認するために別の化学薬剤ドキソルビシンを使用した。操作プロセスは実施例5と同じであった。結果は、P53欠損腫瘍細胞(H1299−NCおよびA549−shP53)では(図8AおよびD)、エンドスタチンは、腫瘍細胞に対するドキソルビシンの毒性を増強し得ることを示した。正常なP53機能を有する腫瘍細胞(H1299−P53およびA549−NC)では(図8BおよびC)、ドキソルビシンは、腫瘍細胞の活性を有意に阻害することができ、エンドスタチンは、ドキソルビシンの阻害効果を有意には増強しなかった。
本実施例では、P53状態の異なる他種の腫瘍細胞の化学療法薬感受性に対するエンドスタチンの調節機能を確認するために、別の腫瘍種(黒色腫)の2つの細胞株、MDA−MB−435S細胞株(P53変異型)およびA375細胞株(P53野生型)を使用した。操作プロセスは実施例5と同じであった。結果は、MDA−MB−435S細胞では(図9AおよびD)、エンドスタチンは、腫瘍細胞に対するエトポシドの毒性を増強し得ることを示した。A375細胞では(図9BおよびC)、エトポシドは、腫瘍細胞の活性を有意に阻害することができ、エンドスタチンは、エトポシドの阻害効果を有意には増強しなかった。
DNA−PKcsはカスパーゼ−3の基質であり、活性化されたカスパーゼ−3はDNA−PKcsを切断してそれを分解することを示した研究がある(T. Itoh et al. Journal of dermatological science, 2001; 25, 72-77, K. M. Henkels et al. Cancer research, 1997; 57, 4488-4492)。本実施例では、腫瘍細胞(H1299−NC、H1299−P53、A549−NC、A549−shP53)を6つの処理群に分けた。第1群:陰性対照;第2群:エンドスタチン(10μg/ml、16時間)処理;第3群:エトポシド(10μM、16時間)処理;第4群:エンドスタチン(10μg/ml、エトポシドの1時間前に添加)とエトポシド(10μM、16時間)による併用処理;第5群:z−DEVD−fmk(10μM、エトポシドの1時間前に添加)とエトポシド(10μM、16時間)による併用処理;第6群:z−DEVD−fmk(10μM)とエンドスタチン(10μg/ml)(エトポシドの1時間前に添加)、次いで、エトポシド(10μM、16時間)による併用処理、なお、z−DEVD−fmkは、カスパーゼ−3阻害剤であった。免疫ブロットアッセイは、DNA−PKcs、PARP、γH2AXおよびカスパーゼ−3シグナル伝達経路を検出した。結果(図10に示されるように、p−DNA−PKcs(S2056)は、DNA−PKcsのリン酸化レベルを表し、S2056は、そのリン酸化部位を表す)は、対照群に比べて、P53欠損腫瘍細胞(図10AおよびD)および正常なP53機能を有する腫瘍細胞(図10BおよびC)の両方で、エンドスタチンは、上記のシグナル伝達経路レベルに影響を及ぼさず;P53欠損腫瘍細胞では、エトポシドは、上記のシグナル伝達経路に有意な効果を示さず;エトポシドに比べて、エンドスタチンとエトポシドの組合せは、γH2AXの有意な増大を伴って、カスパーゼ−3の活性化ならびにDNA−PKcsおよびPARPの分解を有意に促進し;z−DEVD−fmkとエトポシドの組合せは、上記のシグナルレベルに有意な効果を示さず;z−DEVD−fmkは、エンドスタチンとエトポシドの共同作用をある程度逆転させ得ることを示した。正常なP53機能を有する腫瘍細胞では、エトポシド単独により処理およびエンドスタチンとエトポシドによる併用処理はどちらも有意なカスパーゼ−3の活性化、DNA−PKcsおよびPARPの分解、ならびにγH2AXの増大を示し;z−DEVD−fmkとびエトポシドの組合せは、エトポシドの効果をある程度逆転させ;z−DEVD−fmkとエンドスタチンとエトポシドの組合せは、z−DEVD−fmkがエトポシドの効果を逆転させ、DNA−PKcsのタンパク質レベルを回復させ、エンドスタチンとエトポシドの共同作用をある程度反映したことを示した。
腫瘍細胞(H1299−NC、H1299−P53、A549−NC、A549−shP53)を4つの処理群に分けた。第1群:陰性対照;第2群:エンドスタチン(10μg/ml、16時間)処理;第3群:エトポシド(1μM、16時間)処理;第4群:エンドスタチン(10μg/ml、エトポシドの1時間前に添加)とエトポシド(1μM、16時間)による併用処理。細胞を上記のように処理した後、それらの細胞の全RNAを抽出し、それらの細胞のBAX、NOXA、PUMA、およびP21のmRNAレベルでの相対的変化をリアルタイムPCRにより検出した。使用したプライマーは下表に示す。
腫瘍細胞(H1299−NC、H1299−P53、A549−NC、A549−shP53)を、6〜8週齢のヌードマウス(Beijing Weitong Lihua Experimental Animal Technology Co., Ltd.)に皮下接種した。担癌ヌードマウスを各群6〜8個体として4群に無作為に分け、異なる薬物処置を行った。第1群:陰性対照;第2群:エンドスタチン(20mg/kg、腹腔内注射、1日1回)処置;第3群:エトポシド(2mg/kg、腹腔内注射、2日毎に1回)処置;第4群:エンドスタチン(20mg/kg、腹腔内注射、1日1回)とエトポシド(2mg/kg、腹腔内注射、2日毎に1回)による併用処置。腫瘍体積を1日おきに測定した。投与2〜4週間後に、マウスを犠牲にし、さらなる使用のために腫瘍組織を摘出した。結果は、P53欠損マウス腫瘍モデルでは(図12AおよびD)、対照群に比べて、エンドスタチンの腫瘍抑制率は19.6%(H1299−NC、19日目)および37.1%(A549−shP53、27日目)であり;エトポシドの腫瘍抑制率は28.0%(H1299−NC、19日目)および39.3%(A549−shP53、27日目)であり;エンドスタチンとエトポシドの組合せは、腫瘍増殖の阻害に共同作用を示し、腫瘍抑制率は66.9%(H1299−NC、19日目)および65.8%(A549−shP53、27日目)であったことを示した。正常なP53機能を有するマウス腫瘍モデルでは(図12BおよびC)、対照群に比べて、エンドスタチンの腫瘍抑制率は33.8%(H1299−P53、19日目)および40.9%(A549−NC、27日目)であり;エトポシドの腫瘍抑制率は、30.0%(H1299−P53、19日目)および34.0%(A549−NC、27日目)であり;エンドスタチンは、腫瘍増殖に対するエトポシドの阻害効果を増強できず、組合せの腫瘍抑制率は37.7%(H1299−P53、19日目)および32.2%(A549−NC、27日目)であった。
H1299細胞およびA549細胞を活発で増殖能のある状態で培養し、腫瘍の収集、群分け、および投与方法は実施例12と同様とした。25日後に投与を止め、マウスの生存を毎日記録し、マウスの生存曲線をプロットした。結果は、H1299マウス腫瘍モデルでは(図13A)、対照群に比べて、エンドスタチン処置群の生存期間中央値は対照群よりも24.2%長く;エトポシド処置群の生存期間中央値は対照群よりも30.3%長く;エンドスタチンとエトポシドの組合せは、マウスの生存期間の延長に共同作用を示し、生存期間中央値は対照群よりも72.7%長かったことを示した。正常なP53機能を有するマウス腫瘍モデルでは(図13B)、対照群に比べて、エンドスタチン処置群およびエトポシド処置群は、マウスの生存期間をある程度延長することができ、エンドスタチンとエトポシドの組合せは、マウスの生存期間の延長に共同作用を示さなかった。
実施例12で摘出した腫瘍組織を固定および包埋した後に切片とした。腫瘍組織中のPCNA(腫瘍細胞の活発な増殖状態を示すマーカー分子)およびDNA−PKcsタンパク質を免疫蛍光技術により染色し、レーザー共焦顕微鏡(ニコンA1)で観察し、画像を取得した。試験結果を図14に示すが、青はDAPI染色細胞の核を表し、緑はDNA−PKcsタンパク質を表し、赤はPCNAタンパク質を表す。図14Aに示すH1299−NC腫瘍組織を例に挙げれば、左の1段目はDNA−PKcs単一染色を表し、緑の領域はDNA−PKcsタンパク質の発現レベルを表す。左の2段目はDNA−PKcs(緑)と核(青)を重畳した結果を表し、重複部分は藍色で示す。左の3段目はPCNA単一染色を表し、赤の領域はPCNAタンパク質の発現レベルを表し、腫瘍細胞増殖の強度を示す。左の4段目はPCNA(赤)と核(青)を重畳した結果を表し、重複部分を紫で示す。同様に、図14B、C、およびDは、それぞれH1299−P53、A549−NC、およびA549−shP53腫瘍組織におけるPCNAおよびDNA−PKcsの発現を示す。この図のタンパク質染色および画像配置の順序は図12Aと一致する。定量結果は、P53欠損マウス腫瘍モデルでは(図14AおよびD)、対照群に比べて、エンドスタチンおよびエトポシドはどちらも腫瘍細胞の増殖をある程度減弱し、DNA−PKcsのタンパク質レベルを低下させることができ;エンドスタチンとエトポシドの組合せは共同作用的に働くことができ、エトポシド処置群に比べて腫瘍増殖およびDNA−PKcsタンパク質レベルをより有意に阻害し得ることを示した。正常なP53機能を有するマウス腫瘍モデルでは(図14BおよびC)、対照群に比べて、エンドスタチン、エトポシド、およびエンドスタチンとエトポシドの組合せは、腫瘍細胞の増殖を低減し、DNA−PKcsのタンパク質レベルを低下させることができ、エンドスタチンは、腫瘍増殖に対するエトポシドの阻害効果およびDNA−PKcsのタンパク質レベルに対するエトポシドの効果を増強しなかった。
本実施例では、ZBP−エンドスタチンおよびPEG−エンドスタチンによるDNA−PKcs経路の阻害を、DNA−PKcsおよびAKTの活性化レベルを検出することにより確認した。腫瘍細胞(H1299細胞およびA549細胞を含む)を6つの処理群に分けた。第1群:陰性対照;第2群:エトポシド(10μM、1時間)処理;第3群:ZBP−エンドスタチン(10μg/ml、1時間)処理;第4群:ZBP−エンドスタチン(10μg/ml、エトポシドの1時間前に添加)とエトポシド(10μM、1時間)による併用処理;第4群:PEG−エンドスタチン(10μg/ml、1時間)処理;第6群:PEG−エンドスタチン(10μg/ml、エトポシドの1時間前に添加)およびエトポシド(10μM、1時間)による併用処理。免疫ブロットアッセイ結果は、対照群に比べて、エトポシド処理はH1299細胞およびA549細胞においてDNA−PKcsシグナル伝達経路のレベルを活性化し(p−DNA−PKcs(S2056)およびp−AKT(S473)の免疫ブロットアッセイの結果に示されるように、p−DNA−PKcs(S2056)は、DNA−PKcsのリン酸化レベルを表し、S2056は、そのリン酸化部位を表し、p−AKTは、AKTのリン酸化レベルを表し、S473は、そのリン酸化部位を表す);ZBP−エンドスタチンまたはPEG−エンドスタチンとエトポシドの組合せは、エトポシドによるDNA−PKcsシグナル伝達経路の活性化を阻害したことを示した(図15)。

本実施例では、腫瘍細胞(H1299細胞およびA549細胞を含む)を8つの処理群に分けた。第1〜第4群では、細胞を対照siRNAでトランスフェクトし、最後の4群では、細胞を、DNA−PKcsを特異的にノックダウンしたsiRNAでトランスフェクトした。DNA−PKcsのsiRNA配列は:フォワード5’−CAGGGUUUAAUCAGAAUAUTT−3’(配列番号5)、リバース5’−AUAUUCUGAUUAAACCCUGTT−3’(配列番号6)である。siRNAトランスフェクション法は、トランスフェクション試薬リポフェクタミン2000の説明書に従って行った。次に、第8群では、細胞を薬物で処理した。第1群および第4群:対照群;第2群および第5群:エンドスタチン(10μg/ml、16時間)処理;第3群および第6群:エトポシド(H1299では10μM、A549では1μM、16時間)処理;第4群および第8群:エンドスタチン(10μg/ml、エトポシドの1時間前に添加)とエトポシド(H1299では10μM、A549では1μM、16時間)の併用。細胞を回収し、アネキシンV−FITC/PIアポトーシス検出キットの説明書に従って操作を行った。アポトーシスはフローサイトメトリーにより検出した。A549細胞はP53野生型細胞であり、H1299細胞はP53欠損細胞であった。結果は、A549細胞は化学療法薬に対する感受性がより高く、同程度のアポトーシスが生じた場合、H1299細胞のエトポシド濃度はA549細胞のエトポシド濃度よりはるかに高かったことを示した。これに基づけば、エンドスタチンの付加は、エトポシドにより誘導されるアポトーシスをさらに促進すると考えられる(図4)。DNA−PKcsをノックダウンした後には、このエトポシドにより誘導されるアポトーシス促進におけるエンドスタチンの役割は減弱した。
まず、レンチウイルスキット(Shanghai Genepharma社から購入)を用いてP53安定過剰発現H1299細胞株(H1299−P53)およびP53安定ノックダウンA549細胞株(A549−NC)を構築した。A549細胞においてP53をノックダウンするために使用したshRNAにより標的化されるP53配列は、5’−GACTCCAGTGGTAATCTAC−3’(配列番号4)であった。ウイルストランスフェクションおよび安定細胞株の構築はキットの説明書に従って行った。構築された腫瘍細胞(H1299−NC、H1299−P53、A549−NC、A549−shP53)を96ウェルプレート(1000細胞/ウェル)に播種し、8つの処理群に分けた。第1群〜第4群では、種々の濃度のエトポシド(0μM、0.1μM、1μM、10μM)を含有する培地を各ウェルに加えた。第5群〜第8群は、まず、10μg/mlのエンドスタチンを含有する培地で1時間前処理し、次に、種々のエトポシド濃度を含有する培地を、第5群〜第8群のエトポシド終濃度がそれぞれ0μM、0.1μM、1μM、および10μMとなるように加えた。12時間の処理後、薬物含有培地を通常培地に置き換え、細胞をさらに72時間培養した。細胞活性を、CCK8キットの説明書(同仁堂、東京、日本)に従って測定した。結果は、P53欠損腫瘍細胞では(図5AおよびD)、エンドスタチンは腫瘍細胞に対するエトポシドの毒性を増強し得ることを示した。正常なP53機能を有する腫瘍細胞では(図5BおよびC)、エトポシドは、腫瘍細胞の活性を有意に阻害することができ、エンドスタチンは、エトポシドの阻害効果を有意には増強しなかった。

天然ヒトエンドスタチンを分子量20kDaのモノメトキシポリエチレングリコールプロピオンアルデヒド(mPEG−ALD)で修飾することにより得られる産物(ここで、それらの結合部位は活性化されたmPEG−ALDアルデヒド基と天然ヒトエンドスタチンのN末端α−アミノ基である)、

- 細胞においてDNA二本鎖切断を誘導する工程において適用される用量が、その治療計画が単独で使用される場合に適用される用量よりも少ない、請求項57に記載の方法。
- 前記腫瘍または癌が非小細胞肺癌または黒色腫である、請求項55〜58のいずれか一項に記載の方法。
