Abstract
Description
(a)式RN−(P/A)−RC−act[式中、RC−actは、RCのカルボキシ活性化形態であり、RCおよび(P/A)は、調製されるコンジュゲートにおいて定義されるとおりであり、RNは、(P/A)のN末端のアミノ基に付着している保護基である]の活性化P/Aペプチドを、タンパク質薬物とカップリングして、タンパク質薬物と、RNが保護基であるP/Aペプチドとのコンジュゲートを得るステップ、および
(b)任意選択により、ステップ(a)において得られたコンジュゲートに含有されているP/Aペプチドから保護基RNを除去して、タンパク質薬物と、RNが存在しないP/Aペプチドとのコンジュゲートを得るステップ
を含む、プロセスに関する。
本発明によるコンジュゲートに含まれる各P/Aペプチドは、独立に、ペプチドRN−(P/A)−RCである。したがって、本発明のコンジュゲートに含まれるP/Aペプチドの各々について、N末端の保護基RN(存在する場合)、アミノ酸配列(P/A)、およびC末端アミノ酸残基RCは、それらのそれぞれの意味からそれぞれ独立に選択される。したがって、本発明のコンジュゲートに含まれる2つ以上のP/Aペプチドは、同じであってもよく、または互いに異なっていてもよい。好ましくは、コンジュゲートに含まれるP/Aペプチドの全部が同じである。
ペプチドRN−(P/A)−RCに含まれる(P/A)部分は、約7個〜約1200個のアミノ酸残基からなるアミノ酸配列であり、(P/A)中のアミノ酸残基の数の少なくとも80%は、プロリンおよびアラニンから独立に選択され、(P/A)は、少なくとも1個のプロリン残基および少なくとも1個のアラニン残基を含む。
ペプチドRN−(P/A)−RC中のRN基は、存在しないか、またはアミノ酸配列(P/A)のN末端のアミノ基、特にN末端α−アミノ基に付着している保護基である。当然のことながら、RNが存在しない場合、対応するP/Aペプチドは、ペプチド(P/A)−RCである。
ペプチドRN−(P/A)−RC中のRC基は、そのアミノ基を介して(P/A)のC末端のカルボキシ基に結合し、そのアミノ基とそのカルボキシ基との間に少なくとも2個の炭素原子を含むアミノ酸残基である。
本発明によるコンジュゲートにおいて、各P/Aペプチド、すなわち、各ペプチドRN−(P/A)−RCは、P/AペプチドのC末端アミノ酸残基RCのカルボキシ基とタンパク質薬物の遊離アミノ基とから形成されるアミド結合を介して、タンパク質薬物にコンジュゲートしている。タンパク質薬物の遊離アミノ基は、例えば、タンパク質薬物のN末端α−アミノ基または側鎖アミノ基(例えば、タンパク質薬物に含まれるリシン残基のε−アミノ基)とすることができる。タンパク質薬物が複数のサブユニットから構成されている場合、複数のN末端α−アミノ基(すなわち、各サブユニットに1個)が存在してもよい。
本発明のコンジュゲートに含まれるタンパク質薬物は、治療的/薬理学的に活性な任意のタンパク質、すなわち、医薬品として使用するのに適切な、任意のタンパク質とすることができる。「タンパク質薬物」という用語は、本明細書では、「治療用タンパク質」および「治療用タンパク質薬物」と同義で使用される。
本発明はまた、本発明のコンジュゲート(すなわち、タンパク質薬物と2つ以上のP/Aペプチドとのコンジュゲート)および薬学的に許容される添加剤を含む医薬組成物も提供する。加えて、本発明はさらに、医薬品として使用するための前記コンジュゲートまたは前記医薬組成物に関する。
本発明によるコンジュゲートは、当技術分野において公知の方法を使用して調製することができる。特に、以下に記載されているプロセスを使用して、かつ/または実施例に記載されている手順に従って、もしくはそれと同様にして調製することができる。
(a)式RN−(P/A)−RC−act
[式中、RC−actは、RCのカルボキシ活性化形態であり、
RCおよび(P/A)は、調製されるコンジュゲートにおいて定義されるとおりであり、かつ
RNは、(P/A)のN末端のアミノ基に付着している保護基である]
の活性化P/Aペプチドを、タンパク質薬物とカップリングして、タンパク質薬物と、RNが保護基であるP/Aペプチドとのコンジュゲートを得るステップ、および
(b)任意選択により、ステップ(a)において得られたコンジュゲートに含有されているP/Aペプチドから保護基RNを除去して、タンパク質薬物と、RNが存在しないP/Aペプチドとのコンジュゲートを得るステップ、
を含む、プロセスも提供する。
本発明はまた、式RN−(P/A)−RC−actの活性化P/Aペプチドであって、RNは、(P/A)のN末端のアミノ基に付着している保護基であり、(P/A)は、約7個〜約1200個のアミノ酸残基からなるアミノ酸配列であり、(P/A)中のアミノ酸残基の数の少なくとも80%は、プロリンおよびアラニンから独立に選択され、(P/A)は、少なくとも1個のプロリン残基および少なくとも1個のアラニン残基を含み、RC−actは、活性化カルボキシ基を有し、そのアミノ基を介して(P/A)のC末端のカルボキシ基に結合し、そのアミノ基とその活性化カルボキシ基との間に少なくとも2個の炭素原子を含むアミノ酸残基である、活性化P/Aペプチドにも関する。
次の定義は、特に他の意味が指定されない限り、本明細書の全体で適用される。
Ac−P/A#1(40)ペプチド(配列番号:1)(TFA塩、純度98%;Peptide Specialities Laboratories、Heidelberg、Germany)35mgを、無水DMSO(99.9%;Sigma−Aldrich、Taufkirchen、Germany)1268μLに溶解した。P/Aペプチドの、その末端のカルボン酸基を介しての化学的活性化を実現するために、500mM TBTU(CAS#125700−67−6;Iris Biotech、Marktredwitz、Germany)のDMSO溶液214μLを添加し、混合した後に、DIPEA(99.5%、biotech.Grade、Sigma−Aldrich)18μLを添加した。全混合物を短時間ボルテックスし、25℃で20分間インキュベートした(図1を参照)。この設定において、ペプチド濃度は7.14mMであり、DIPEAとTBTUとAc−P/A#1(40)との間のモル比は10:10:1であった。
Pga−P/A#1(20)−Ahxペプチド(TFA塩、純度98%;Almac Group、Craigavon、UK)(配列番号:9)3mgを、435mM TBTUのDMSO溶液37.3μlに溶解した。P/Aペプチドの、その末端のカルボン酸基を介しての化学的活性化は、DIPEA2.7μLをペプチドの溶液に添加し、ボルテックスすることによって開始した。この設定において、ペプチドの濃度は40.6mMであり、DIPEAとTBTUとPga−P/A#1(20)−Ahxとの間のモル比は10:10:1であった。25℃で10分インキュベートした後に、表1に従って、混合物をEppendorf(商標)チューブに入れ、DMSOで希釈した。各Eppendorf(商標)チューブは、最終的に15μLの体積の希釈ペプチド溶液を含有していた。
凍結乾燥した組換えバチルス−ファスティディオスス(Bacillus fastidiosus)ウリカーゼ(Sigma−Aldrich、カタログ番号94310、配列番号:17)をPBSに溶解し、Slide−A−Lyzer(商標)透析カセット(MWCO 10.000;Thermo Fisher Scientific、Waltham、MA)を使用して、4℃で終夜PBSに対して透析し、低分子量夾雑物を除去した。
P(R)=Pmax×R/(R1/2+R)
Pmaxはカップリングされたペプチドの最大(漸近)平均数に相当し、R1/2は、カップリングされたペプチドの最大半量数のカップリング比に相当する。
凍結乾燥した組換えバチルス−ファスティディオスス(Bacillus fastidiosus)ウリカーゼ(Sigma−Aldrich、カタログ番号94310、配列番号:17)をPBSに溶解し、サイズ排除クロマトグラフィーにより、PBSで平衡化したSuperdex(商標)200 increase 10/300カラム(GE Healthcare)で四量体として精製した。
1−ヒドロキシベンゾトリアゾール(HOBt)、4−ニトロフェニル(pNP)またはペンタフルオロフェニル(PFP)のいずれかでエステルとして活性化されたPga−P/A(20)−Ahxペプチドを調製するために、それぞれの活性化用に、Pga−P/A#1(20)−Ahxペプチド(TFA塩、純度98%;Almac Group、Craigavon、UK)(配列番号:9)10mgを、150mM DIPEAのDMF溶液360μlに溶解した。次いで、P/Aペプチドの、その末端のカルボン酸基を介しての化学的活性化は、TBTU、4−ニトロフェニルトリフルオロアセテート(Sigma−Aldrich)またはペンタフルオロフェニルジフェニルホスフィネート(Sigma−Aldrich)の、DMF中150mM溶液360μlを、それぞれペプチド/DIPEA溶液に添加し、ボルテックスすることによって開始した。この設定において、ペプチドの濃度は7.5mMであり、DIPEAとカップリング試薬とPga−P/A#1(20)−Ahxとの間のモル比は10:10:1であった。pNP活性エステルの形成は、50mM 4−(ジメチルアミノ)ピリジン(Sigma−Aldrich)のDMF溶液22μlの添加によって促進された。25℃で20分インキュベートした後に、各混合物の72μlのアリコートを取り出した。ジエチルエーテル500μlを添加して活性ペプチドを沈殿させた。遠心分離(13.500×g、4℃)した後に、上清を除去し、沈降物をジエチルエーテル500μlで洗浄し、真空エバポレーター(SpeedyDry RVC2−18 CDplus、Martin Crist Freeze Dryers、Germany)を使用して乾燥し、−20℃で、例えば14日間保管した。
Pga−P/A#1(40)−Ahxペプチド(Almac Group、Craigavon、UK)(配列番号:18)またはPga−P/A#3(40)−Ahxペプチド(Peptide Specialities Laboratories)(配列番号:19)の各々3.2mgずつをDMSO3.5μlに溶解し、500mM TBTUのDMSO溶液18.5μlを添加した。P/Aペプチドの、その末端のカルボン酸基を介しての化学的活性化は、DIPEA1.6μLをペプチドの溶液に添加し、ボルテックスすることによって開始した。この設定において、ペプチドの濃度は17.35mMであり、DIPEAとTBTUとPga−P/A#1(40)−Ahx(またはPga−P/A#3(40)−Ahx)との間のモル比は10:10:1であった。実施例2と同様に、25℃で10分インキュベートした後に、混合物をEppendorf(商標)チューブに入れ、DMSOで希釈して、酵素:ペプチドの質量比、1:1、1:3、1:6および1:10を実現した。各Eppendorf(商標)チューブは、最終的に25μLの体積の希釈した活性ペプチド溶液を含有していた。
Pga−P/A#1(40)−Ahxペプチド(Almac Group、Craigavon、UK)(配列番号:18)またはPga−P/A#3(40)−Ahxペプチド(Peptide Specialities Laboratories)(配列番号:19)の各々3.2mgずつをDMSO3.5μlに溶解し、500mM TBTUのDMSO溶液18.5μlを添加した。P/Aペプチドの、その末端のカルボン酸基を介しての化学的活性化は、DIPEA1.6μLをペプチドの溶液に添加し、ボルテックスすることによって開始した。この設定において、ペプチドの濃度は17.35mMであり、DIPEAとTBTUとPga−P/A#1(40)−Ahx(またはPga−P/A#3(40)−Ahx)との間のモル比は10:10:1であった。実施例2と同様に、25℃で10分インキュベートした後に、混合物をEppendorf(商標)チューブに入れ、DMSOで希釈して、酵素:ペプチドの質量比、1:1、1:3、1:6および1:10を実現した。各Eppendorf(商標)チューブは、最終的に25μLの体積の希釈した活性ペプチド溶液を含有していた。
Pga−PAS#1(40)−Ahxペプチド(Peptide Specialities Laboratories)(配列番号:22)2mgを132mM DIPEAのDMSO溶液44μlに溶解した。PASペプチドの、その末端のカルボン酸基を介しての化学的活性化は、500mM TBTUのDMSO溶液11.6μLを添加し、ボルテックスすることによって開始した。この設定において、ペプチドの濃度は10.4mMであり、DIPEAとTBTUとPga−PAS#1(40)−Ahxとの間のモル比は10:10:1であった。全混合物を短時間ボルテックスし、25℃で10分間インキュベートした。

- (P/A)は、プロリンおよびアラニンから独立に選択される15個〜45個のアミノ酸残基からなるアミノ酸配列であり、(P/A)は、少なくとも1個のプロリン残基および少なくとも1個のアラニン残基を含む、請求項1から3のいずれか一項に記載のコンジュゲート。
- (P/A)に含まれるプロリン残基の数の、(P/A)に含まれるアミノ酸残基の総数に対する割合は、≧10%かつ≦70%であり、好ましくは≧20%かつ≦50%であり、より好ましくは≧25%かつ≦40%である、請求項1から4のいずれか一項に記載のコンジュゲート。
- (P/A)は、(i)AAPAおよびAPAPから独立に選択される2つ以上の部分配列、および(ii)任意選択により、プロリンおよびアラニンから独立に選択される、さらに1個、2個または3個のアミノ酸残基からなる、請求項1から5のいずれか一項に記載のコンジュゲート。
- (P/A)は、(i)1つまたは複数の部分配列AAPAAPAP、(ii)任意選択により、1つまたは2つの部分配列AAPA、および(iii)任意選択により、プロリンおよびアラニンから独立に選択される、さらに1個、2個または3個のアミノ酸残基からなる、請求項1から6のいずれか一項に記載のコンジュゲート。
- (P/A)は、(i)配列ASPAAPAPASPAAPAPSAPA、(ii)配列APASPAPAAPSAPAPAAPSA、(iii)配列AASPAAPSAPPAAASPAAPSAPPA、(iv)前記配列のうちのいずれかの断片、または(v)前記配列の2つ以上の組合せからなる、請求項1に記載のコンジュゲート。

- 前記P/Aペプチドがコンジュゲートしている前記遊離アミノ基のうち少なくとも1個は、前記タンパク質薬物のリシン残基のε−アミノ基である、請求項1から15のいずれか一項に記載のコンジュゲート。
- 前記P/Aペプチドがコンジュゲートしている前記遊離アミノ基は、前記タンパク質薬物の任意のリシン残基のε−アミノ基、前記タンパク質薬物または前記タンパク質薬物の任意のサブユニットのN末端α−アミノ基、およびそれらの任意の組合せから選択される、請求項1から16のいずれか一項に記載のコンジュゲート。

- 請求項1から21のいずれか一項に記載のコンジュゲートを調製するプロセスであって、
(a)式RN−(P/A)−RC−act[式中、RC−actは、RCのカルボキシ活性化形態であり、RCおよび(P/A)は、調製される前記コンジュゲートにおいて定義されるとおりであり、RNは、(P/A)のN末端のアミノ基に付着している保護基である]の活性化P/Aペプチドを、タンパク質薬物とカップリングして、前記タンパク質薬物と、RNが保護基である前記P/Aペプチドとのコンジュゲートを得るステップ、および
(b)任意選択により、ステップ(a)において得られた前記コンジュゲートに含有されている前記P/Aペプチドから前記保護基RNを除去して、前記タンパク質薬物と、RNが存在しない前記P/Aペプチドとのコンジュゲートを得るステップ
