ヒト多能性幹細胞(hPSC)は、疾患モデリングおよび細胞置換療法に革命をもたらしている。PSCは、今では患者から(例えば、誘導PSC、またはiPSCとして)日常的に作製され、疾患の原因となる疑いがある遺伝子変異を意のままに変更して、遺伝子型と表現型の関係を詳しく調査することができる。これらの技術は、ゲノムワイドな関連性と因果関係を橋わたししており、疾患関連コンテクストの提供に欠かせないステップであるPSC分化を誘導する能力の益々の高度化と併用されれば、疾患の発症機序に対するかつてない見識をもたらす。疾患モデリングに加えて、PSCを臨床的に意義のある細胞型に誘導して、細胞置換療法に無限の資源を提供することができる。両方のPSC応用は強力であるが、hPSCの維持および分化についての多くの実際の態様には改善の余地がありうる。

4.図面の簡単な説明

解凍後に継代なしに使用するための、解離され凍結保存された多能性幹細胞
概要
ヒト多能性幹細胞(PSC)は、細胞置換療法および疾患モデリングのための無限の細胞源を提供する。それらには並外れた活力(enormous power)があるにもかかわらず、技術的態様が再現性を妨げてきた。ここに記載するのは、ほぼすべてのPSC実験についての主要変数(PSC出発物質の質および量)を排除する、PSCワークフローの単純な改良である。ほとんどの研究所は、PSCを連続的に継代し、拡大後に少量を使用するが、これらの実験の「ジャストインタイム」性は、品質管理が使用前にめったに行われないことを意味する。品質管理の欠如は、PSCの質、無菌性および遺伝的完全性を損なわせることがありえ、これは、得られる結果の交絡因子を生じさせる。CryoPauseと呼ばれる、ここで開示する方法(図7)は、PSCの培養物を解離させて単一細胞にし、これらのPSCを、解凍して直ちに分化などの実験に使用することができる使い捨て凍結保存バイアルとして、貯蔵する。
最新の研究は、播種効率のわずかな低下が観察されたが、生存率、多能性および分化能に差がないことを示す。CryoPauseを規模拡大させることができる。例えば、4段セルファクトリーから2億8000万の細胞バンクを構築することができる。本実施例は、解凍後生存率を有意に低下させることなく1バイアル当たり細胞3000万個ほど高い濃度で細胞を凍結させた。
解凍後の拡大をなくして幹細胞実験の再現性を向上させることができるかどうかを判定した。回復および拡大の迂回には、解凍後の高い生存率が現実的であることが求められる可能性が高い。Essential 8(商標)培地(E8;Chenら、2011年)でのWA09(H9)フィーダーフリー培養を使用する臨床パラダイムを使用した。WA09細胞をAccutaseで処置して単一細胞浮遊液を生成した後、洗浄し、単分散ヒト多能性幹細胞凍結保存用に設計された市販の培地であるFreSR(商標)−Sに再浮遊させた。標準プログラムを使用して速度制御フリーザーで細胞を凍結保存した後、「すぐに使用可能な」アリコートとして液体窒素中で長期保管した(材料および方法を参照されたい)。最初の実験は、驚くほど高い解凍後生存率を実証した。解凍後生存率への、細胞株の影響、凍結保存用培地の影響、および速度制御フリーザーの寄与の影響を詳しく調査し、E8培地でのフィーダーフリー培養を使用したとき、これらの変数のいずれかに対しても明らかな依存性がないことが判明した。異なるPSC株(図5A)、異なる凍結保存用培地(図5B)および従来の緩速冷却(図5C)には、効率のはっきりした差はなかった。主な要因は、初期培養条件であるようだった。旧来のフィーダーベースの方法を使用して拡大させたWA09細胞のほうが、生存率が低かったが、この生存率低下がFreSR(商標)−Sでは軽減されたからである(図5D)。
CryoPause処理細胞の健常性を評定するために、分化が正常に誘導されたであろう播種の翌日に、PSCマーカーを検査した。フローサイトメトリーで測定することができたCryoPause処理細胞と新鮮対照細胞の間の多能性幹細胞マーカーSSEA3、SSEA4、OCT4、SOX2およびNANOGに明らかな差はなかった(図1Cおよび1D)。自発的分化マーカーSSEA1の増加もなかった(図1Cおよび1D)。免疫蛍光分析によってフロー結果を独立して確認し、2集団間に識別可能な差はなかった(図1E)。
CryoPause処理細胞と対照細胞の間にほとんど差がないこと見いだすことができたため、CryoPause処理細胞のin vitro分化能を評定した。対照およびCryoPause処理細胞を二重SMAD阻害に曝露して、WA09 hESCを神経運命に誘導した(Chambersら、2009年)。OCT4およびPAX6を経時的に測定して、神経分化の効率および動態を評定した。神経誘導のタイミングにも程度にも差がなかった(図2A〜2C、および2K)。中内胚葉運命に誘導されたCryoPause処理細胞もまた、分化の程度および動態に関して区別不能であった(図2D〜2F、および2L)。これらの細胞の両方の運命は、相対的に未熟であり、したがって、作製がより容易であるため、CryoPause処理細胞からの中脳ドーパミンニューロンの作製を試みた(Barkerら、2015年)。WA09 CryoPause処理細胞は、臨床的に適合する「標準操作手順」から、FOXA2/TH二重陽性の有糸分裂後中脳ドーパミンニューロンを効率的に生成し、対照細胞に由来するニューロンと同様の遺伝子発現プロファイルを示す(図2Iおよび2J)。
CryoPause処理細胞は、通常は解凍の24時間後に挙動するため、細胞を解凍直後に遺伝子操作することができるかどうかを調査した。解凍直後のCryoPause処理WA09細胞のヌクレオフェクション効率を新鮮対照細胞と比較した。GFPプラスミドでのヌクレオフェクションの24時間後、蛍光顕微鏡検査は、両方の条件でGFP+細胞を明示し、フローサイトメトリー定量は、>85%がGFPを発現したことを明示した(図3A〜3Cおよび4A〜4B)。直ちに解凍したCryoPause処理細胞は、形質導入の24時間後に蛍光顕微鏡検査により示される通り、EmGFPを発現するセンダイウイルスベクターによる形質導入にも成功することができた(図3D)。CP由来のセンダイ形質導入サブクローンを、少なくとも15継代(試験した最長)にわたって増殖させることができた。
新規方法(CryoPause)は、ほとんどの多能性幹細胞ベースの応用にとって非常に重要な変数(分化またはゲノム改変前の多能性細胞の性質)を、排除すると言える。凍結保存hPSCは、使用前に回復、拡大および継代を必要とすることは、一般に認知されている。本実験は、解離させたヒト多能性幹細胞を、CryoPause処理しなかった対応する「新鮮」細胞と比較して、解凍後生存率をほとんど損なうことなく、播種効率のわずかな低下で、単一細胞浮遊液として凍結保存することができることを実証した。最新のデータは、このパラダイム変化を可能にする技術的駆動因子が、多数の凍結保存パラダイム(LiuおよびChen、2014年)を使用してE8で拡大させた細胞に関して回復向上が観察された、培養システムであることを示す。
ヒト多能性幹細胞維持
WA09(H9)および960.1B iPSC株を、DMEM/F12(Thermo Fisher、#11330032)で1:50希釈したGeltrex(Thermo Fisher、#A1413202)上のEssential 8(商標)(E8、Thermo Fisher、#A1517001)培地で、最初に維持し、短時間(3分)の0.05%トリプシン−EDTA(Thermo Fisher、#25300054)処置、その後、剥離(コロニー構造を維持するために)、そして新たなE8培地での2回の洗浄を使用して、3〜4日ごとにクラスターとして継代させた。細胞を、24時間、10μM Y−27632で再度播種した。30〜55継代の間の細胞を使用し、異常な核型は見られなかった。
CryoPause処理細胞バンクを生成するために、E8培地で成長させたPSCを37℃インキュベーターにおいて30分間、Accutase(Innovative Cell Technologies、#AT−104)で解離させた。細胞を2容量(細胞:Accutaseに対して)のE8で洗浄し、遠心分離して細胞をペレットにした(200×g、室温、5分間)。上清を吸引し、すすぎを繰り返した。最後に、細胞をFreSR−S(商標)(Stem Cell Technologies、#05859)に別段の指示がない限り細胞1000万個/mLで再浮遊させた。FreSR−S:細胞混合物を予冷したクライオチューブに添加し、その後、次のプログラムを使用して速度制御フリーザーで凍結させた:

Human Pluripotent Stem Cell Sorting and Analysis Kit(BD Biosciences、#560461)およびHuman Pluripotent Transcription Factor Analysis Kit(BD Biosciences、#560589)を製造業者のプロトコール通りに使用して、BD FACS Aria IIIで幹細胞マーカーを定量した。免疫蛍光染色のために、次の一次抗体を使用した:NANOG(1:200、BD Biosciences、#560482);OCT4(1:200、Santa Cruz Biotechnology、#sc−9081);SOX2(1:100、R&D Systems、#AF2018)。適切なAlexa Fluor結合二次抗体(Thermo Fisher)を1:400で使用した。
PluriTestは、全ゲノムトランスクリプトームマイクロアレイデータ分析に基づく。簡単に言うと、以前に記載された手順(Mullerら、2011年;Williamsら、2011年;Mullerら、2012年)と同じ手順を利用した。Qiagen RNeasy単離キットを製造業者(Qiagen、Hilden、Germany)の説明書に従って使用して、培養条件(対照およびCryoPause、1サンプル当たり細胞1×106個)ごとに2回の生物学的反復実験からRNAを単離した。Illumina HT12v4マイクロアレイを製造業者の説明書に従ってハイブリダイズした(Mullerら、2011年;Williamsら、2011年;Mullerら、2012年)。得られた生データをR/Bioconductor lumi−package(Duら、2008年;Linら、2008年;Mullerら、2011年)で処理した。スキャナー技術の変化およびIlluminaによるハイブリダイゼーションプロトコールの改良のため、近年のPluriTestの結果に、より低い多能性およびより高い新規性スコアを示す傾向があることは、認知されている(Bernhard Schuldt、University Hospital Schleswig−Holstein、Kiel、Germany、私信)。この技術的偏りを考慮したとしても、両方(対照サンプルおよびCryoPauseサンプル)は、両方の生物学的反復実験で経験的多能性およびより高い新規性スコア閾値に合格する。
シークエンシングライブラリー作製のために、1μgの高品質ゲノムDNAを、NEXTFlex Bisulfite−Seq Kit(Bioo、#5119−01)で、製造業者の説明書に従って使用した。非メチル化ラムダDNAを1%で加えて、バイサルファイト(bisulfate)転化レベルを評定した。12サイクルのPCRを行った。サンプルをHiseq 2500 Rapid mode Paired End 125にかけ、サンプルごとに平均1億4000万リードを生成した。
FASTQファイルをbcl2fastq(V2.17)により生成し、Illuminaのchastity filterに基づいてパスフィルターリードについてフィルタリングした。シークエンシングアダプターを、FLEXBAR(V2.4)(Dodtら、2012年)、参照ヒトゲノムhg19とのBismark(V0.14.4)(Kruegerら、2011年)およびBowtie2(V2.2.5)(Langmeadら、2012年)を使用するゲノムアラインメント、によりトリミングし、塩基ごとのCpGメチル化指標をカスタムPERLスクリプト(Garrett-Bakelmannら、2015年)で計算した。
Geltrex被覆皿の上の10μM Y−27632を含むE8培地に単一PSCを細胞100,000個/cm2で播種した。24時間後、細胞をE8中0.5μMカンプトテシン(Sigma、#C9911)で1時間、37℃で処置した。次いで、細胞をホスホヒストンH2A.X(1:300、Millipore、#05−636)に対して染色し、適切なAlexa Fluor結合二次抗体を1:400で使用した。画像を獲得し、eHarmonyソフトウェアを使用してPerkin−Elmer Operettaで分析した。
Chambersら、2009年の派生物を使用した。神経誘導前に、Geltrex被覆皿の上の10μM Y−27632を含むE8培地(1:30)に単一PSCを細胞200,000個(新鮮対照)または400,000個/cm2(CryoPause)で播種した。24時間後、培地を除去し、100nM LDN189193および10μM SB431542(LSB)を含むEssential 6(商標)(E6)培地(Thermo Fisher、#A1516401)を添加した。細胞に3日間、E6培地+LSBを供給した後、3回の供給の間に段階的にN2培地(下記参照)に移行し、2日ごとに新たな培地と交換した。神経変換効率をOCT4(BD Biosciences、#560186)およびPAX6(BD Biosciences、#562249)についてのフロー分析により定量した。OCT4およびPAX6(1:200、BD Biosciences、#561462)対する抗体を使用して、培養物を染色もした。Kriksら、2011年の改良版を使用して、中脳ドーパミンニューロンを作製した。簡単に言うと、上記の通りGeltrex上で成長させたWA09細胞を、SHHおよびWNTシグナルでの二重SMAD阻害に付した後、最終的に回収した。回収後、Kriksら、2011年(準備中の原稿)の中で見つけられる同じ増殖因子および小分子を含有する中脳ドーパミン支持培地を添加した。チロシンヒドロキシラーゼ(1:1000、Thermo Fisher、#P21962)およびFOXA2(1:200、R&D Systems、#AF2400)に対する抗体を使用して、培養物を染色した。適切なAlexa Fluor結合二次抗体を1:400で使用した。

単一PSCを上記の通り播種した。24時間後、培地を除去し、5μM CHIR99021(Stemgent #04−0004−10)を含むE6培地を添加して、中内胚葉を生じさせた(Lamら、2014年)。5μM CHIR99021を含むE6を、最長4日間、24時間ごとに交換した。変換効率をOCT4(BD Biosciences、#560186)およびブラキウリ(R&D Systems、#IC2085A)についてのフロー分析により定量した。OCT4およびブラキウリに対する抗体(1:40、R&D Systems、#AF2085)を使用して培養物を染色し、適切なAlexa Fluor結合二次抗体を1:400で使用した。

NSGマウス(Jackson Laboratories)をin vivo研究に使用し、MSKCC Institutional Animal Care and Use CommitteeおよびResearch Animal Resource Centerにより承認されたガイドラインに従ってケアした。8週齢雌マウスの側腹部に、300万個のH9細胞を、1:2でHEPES緩衝HBSSに混合したMatrigel(商標)(BD Biosciences)とともに皮下注射した。マウスを罹患/死亡の兆候について毎日観察し、体重を少なくとも週に2回評定した。ノギスを使用して週に2回、腫瘍を測定し、長さ×幅2×0.52という式を使用して体積を計算した。研究の最後に、腫瘍を10%ホルマリンで固定し、処理し、パラフィンに包埋し、切片を作製し、ヘマトキシリンおよびエオシン(H&E)で染色した。顕微鏡スライドを病理医が再調査した。
CryoPause処理の前に、Nunc(商標)Cell Factory(商標)System、4トレイ層(Thermo Fisher、#140004)を使用して、大型バンクを拡大させた。継代後の最初の2日間、細胞に500mLのE8培地を供給し、3日目に600mLを供給した。単一細胞浮遊液を生成するために、140mLのAccutaseを30分間、37℃で添加し、次いで、トレイを100mLの培地で洗浄した。
CryoPause処理細胞にヌクレオフェクトするために、Amaxa(商標)Cell Line Nucleofector(商標)Kit V(Lonza、#VCA−1003)を使用した。WA09 CryoPause処理細胞を解凍し、洗浄した。100μLのSolution Vを22.2μLのSupplement 1と混合し、500万個のCryoPause処理細胞を100μLのこの混合物に添加した。10μlのGFP対照プラスミドを反応物に添加した後、プログラムB−016(Lonza Nucleofector(商標)2bデバイス)を用いてヌクレオフェクトした。ヌクレオフェクト細胞を、上記の通りGeltrex上の10μM Y−27632を含むE8に添加した。生細胞を図3Aおよび4Bのために撮像し、Accutaseでの培養物の処置、洗浄、および0.1%BSAを含むPBSへの再浮遊の後、BD FACS Aria IIIで蛍光を測定することにより、蛍光を発する細胞の数を判定した。フローデータをFlowJoで分析し、免疫蛍光画像をAdobe Photoshopで補正した。
CryoPause処理WA09細胞を解凍し、洗浄し、10μM Y−27632を補給したE8培地に再浮遊させ、EmGFPを発現するセンダイウイルスベクター(CytoTune(商標)EmGFP、ThermoFisher Scientific、#A16519)を感染多重度5で添加した。形質導入CP細胞をE8に再度播種して拡大させた。形質導入細胞を少なくとも15継代にわたって拡大させることができた。

WA09ヒト胚性幹細胞を凍結保存し、解凍し、解凍後直ちにそれらの細胞に、実施例1により説明したように3.4kb GFPプラスミドをヌクレオフェクトした。凍結保存しなかった新鮮非凍結対照WA09細胞に同様にヌクレオフェクトし、ヌクレオフェクションの24時間後にフローサイトメトリーを使用してGFP発現を分析した。非トランスフェクトWA09細胞も陰性対照として使用した。図8Aにより示される通り、CryoPause処理細胞は、異なる小分子DNA取込みパターンを示した:全体としては、小プラスミドからGFPを発現した細胞は若干少なかったが、より明るく発現した。これは、CryoPauseによって大量のDNAを取り込む細胞の能力がより高くなることを示唆する。細胞の全プール内のより小さい構築物に関するより低いパーセンテージが、DNA力価測定の結果でありうる:すなわち、大量のプラスミドを取り込む細胞は、より少ないDNAを通常は取り込むであろう集団内の細胞のためのものをほとんど残さない。
Claims (39)
- 解離された細胞の凍結集団と凍結保存用培地とを含む組成物であって、前記凍結集団内の細胞の濃度が、少なくとも細胞約100万個/mlである、組成物。
- 異種核酸がトランスフェクトされた細胞を含む組成物であって、解離された細胞の凍結集団および凍結保存用培地を解凍することにより得られる細胞にトランスフェクトすることにより調製され、前記凍結集団内の細胞の濃度が、少なくとも細胞約100万個/mlである、組成物。
- 前記凍結集団内の細胞の濃度が、少なくとも細胞約500万個/mlである、請求項1または2に記載の組成物。
- 前記凍結集団内の細胞の濃度が、少なくとも細胞約3000万個/mlである、請求項1〜3のいずれか一項に記載の組成物。
- 前記凍結集団内の細胞の濃度が、少なくとも細胞約5000万個/mlである、請求項1〜4のいずれか一項に記載の組成物。
- 前記細胞が、哺乳動物細胞である、請求項1〜5のいずれか一項に記載の組成物。
- 前記哺乳動物細胞が、多能性幹細胞(PSC)である、請求項6に記載の組成物。
- 前記多能性幹細胞が、誘導多能性幹細胞(iPSC)または胚性幹細胞(ESC)である、請求項7に記載の組成物。
- 凍結細胞を含む組成物を調製する方法であって、
培養培地で培養された細胞の集団を解離させるステップ、
解離された細胞を凍結保存用培地に浮遊させるステップであって、細胞浮遊液を形成するステップ、および
前記細胞浮遊液を凍結させるステップであって凍結細胞の組成物を形成するステップ
を含み、
凍結細胞の前記組成物が、少なくとも細胞約100万個/mlの濃度を有する、方法。 - トランスフェクト細胞を調製する方法であって、
(i)凍結細胞を含む組成物を、
培養培地で培養された細胞の集団を解離させるステップと、
解離された細胞を凍結保存用培地に浮遊させるステップであって、細胞浮遊液を形成するステップと、
前記細胞浮遊液を凍結させるステップであって、凍結細胞の組成物を形成するステップと
を含む方法であって、凍結細胞の前記組成物が、少なくとも細胞約100万個/mlの濃度を有する、方法により、調製するステップ;および
(ii)組成物(i)からの細胞に異種核酸をトランスフェクトするステップ
を含む方法。 - 前記細胞の集団を解離させるステップが、細胞の前記集団を有効量の細胞解離溶液に曝露することをさらに含む、請求項9または10に記載の方法。
- 前記細胞解離溶液が、酵素不含細胞解離溶液および酵素含有溶液からなる群から選択される、請求項11に記載の方法。
- 酵素含有細胞解離溶液が、1つまたは複数の酵素を含む、請求項12に記載の方法。
- 前記酵素が、コラゲナーゼ、プロテアーゼ、トリプシン、パパイン、ヒアルロニダーゼ、およびDNaseからなる群から選択される、請求項13に記載の方法。

- 前記組成物中の前記凍結細胞の濃度が、少なくとも細胞約3000万個/mlである、請求項9〜18のいずれか一項に記載の方法。
- 前記細胞が、哺乳動物細胞である、請求項9〜19のいずれか一項に記載の方法。
- 前記哺乳動物細胞が、PSCまたはESCである、請求項20に記載の方法。
- 前記PSCがiPSCである、請求項21に記載の方法。
- 細胞を培養するin vitroでの方法であって、
解離された細胞の集団と凍結保存用培地とを含む凍結細胞の組成物を解凍するステップ、および
前記細胞を下流の処置に付すステップ
を含み、前記細胞が、前記下流の処置の前に、指数関数的に拡大されない、方法。 - 前記凍結細胞の組成物が、少なくとも細胞約100万個/mlの濃度を有する、請求項23に記載の方法。
- 前記凍結細胞の組成物が、少なくとも細胞約500万個/mlの濃度を有する、請求項23または24に記載の方法。
- 前記凍結細胞の組成物が、少なくとも細胞約3000万個/mlの濃度を有する、請求項23〜25のいずれか一項に記載の方法。
- 前記細胞が、哺乳動物細胞である、請求項23〜26のいずれか一項に記載の方法。
- 前記哺乳動物細胞が、PSCまたはESCである、請求項27に記載の方法。
- 前記PSCがiPSCである、請求項28に記載の方法。
- 前記下流の処置が、前記細胞を分化させるin vitroでの方法を含む、請求項23〜29のいずれか一項に記載の方法。
- 前記細胞が、多数のドーパミン産生前駆細胞に分化する、請求項30に記載の方法。
- 前記ドーパミン産生前駆細胞が、検出可能なレベルのフォークヘッドボックスタンパク質A2(FOXA2)、LIMホメオボックス転写因子1アルファ(LMX1A)、チロシンヒドロキシラーゼ(TH)またはこれらの組合せを発現する、請求項31に記載の方法。
- 前記iPSCが、神経変性疾患と診断された対象に由来する、請求項29〜32のいずれか一項に記載の方法。
- 前記神経変性疾患が、パーキンソン病である、請求項33に記載の方法。
- 前記解離された細胞の集団が、外来性核酸を発現し、その発現レベルが、凍結されたことのない細胞の集団による前記外来性核酸の発現レベルより高い、請求項1〜8のいずれか一項に記載の組成物。

- 前記解離された細胞の集団の発現レベルが、前記凍結されたことのない細胞の集団の発現レベルより少なくとも約5%高い、請求項35または36に記載の組成物。
- 凍結される細胞の集団に核酸を導入するステップをさらに含む、請求項9〜34のいずれか一項に記載の方法。
- 前記核酸が、トランスフェクションまたはヌクレオフェクションにより導入される、請求項38に記載の方法。
