Raji細胞に対するIMP321の結合を決定するための蛍光標識細胞分取(FACS)アッセイの使用の評価
FACSアッセイを実施して、Raji細胞に対するIMP321の結合を決定した。100%、75%、および50%のMHCクラスII結合活性を有するIMP321試料を試験した。100%活性を有する試料は、所定の濃度で既知のMHCクラスII結合活性を有する基準試料であった。75%および50%の活性を有する試料は、基準試料の希釈によって調製された。
Raji細胞に対するIMP321の結合を決定するためのMeso Scale Discovery(MSD)アッセイの使用の評価
この実施例は、Raji細胞に対するIMP321の結合を決定するためのメソスケールディスカバリー(MSD)アッセイの評価を記載する。
ELISAプレートに対するIMP321の非特異的結合の評価

この実施例は、カゼインブロッキング緩衝液を使用して異なる播種密度のRaji細胞に対するIMP321の結合を決定するためのMSDアッセイの評価を記載する。
Raji細胞に対するIMP321の結合を決定するためのELISAアッセイの使用の評価
この実施例は、Raji細胞に対するIMP321の結合を決定するための細胞に基づく直接ELISAおよび細胞に基づくトランスファーELISAの能力の評価を記載する。
バイオレイヤー干渉法(BLI)を用いたLAG−3タンパク質誘導体IMP321の製剤の結合活性を測定するための細胞に基づくアッセイ
IMP321は、MHCクラスII分子に対して高い親和性を有するLAG−3タンパク質の可溶性組換え誘導体である。この実施例は、BLIを使用して、MHCクラスII発現Raji細胞に対するIMP321の結合活性を測定するための細胞に基づくアッセイを記載する。アッセイは簡単かつ迅速であり、そして基準標準物質と試料との間の比較を可能にする。

1.すぐ使用できるRaji細胞の調製
1)液体窒素フリーザーからNバイアルのRaji細胞を取り出し、37℃のウォーターバスで素早く解凍する。
2)バイアルの内容物を無菌的に、およそN×9mLのRaji細胞増殖培地を含む滅菌遠心分離チューブに移す。静かにピペッティングしてよく混ぜる。
3)細胞を300×gで5分間遠心する。結合アッセイ希釈液に細胞を再懸濁し、細胞計数器または血球計でそれらを計数する。
4)1mLあたり4.0E6〜8.0E6細胞に細胞密度を調整するために十分な量の結合アッセイ希釈液に一定量の細胞ストック懸濁液を加え、使用するために氷上に保つ。
注:1)精度を確保するために逆ピペッティングを使用する。
2)泡沫と泡の発生を避けるか最小限に抑えるために、穏やかにボルテックスする。
1)基準標準物質:
1.1)必要に応じてIMP321基準標準物質のバイアルを解凍する。2〜8oCで保存する。有効期限は解凍日から7日間である
1.2)IMP321基準標準物質を製剤緩衝液中で約1.0mg/mLに希釈する。新鮮なものを調製し、新鮮なものを使用する。ブランクとして製剤緩衝液を用いて分光光度的にタンパク質濃度を決定する。
1.3)測定されたタンパク質濃度に基づいて、RMを希釈して以下に記載されるような適切な濃度に標準曲線を調製する。ボテックス(votex)により希釈液を混合する。
2)対照の準備
2.1)対照は、上記ステップ1.3で調製したチューブCからの基準試料の独立した希釈物である。上記の表に記載されているようにさらに希釈する。ボテックスにより希釈液を混合する。
2.2)対照に希釈C−Jを使用する。
3)試料の調製
3.1)タンパク質濃度に基づいて、IMP321試料をアッセイ希釈液で約1.0mg/mLに希釈する。新鮮なものを調製し、新鮮なものを使用する。
3.2)上記表に記載されるように、標準曲線を作成するためにさらに希釈して適切な濃度にする。ボテックスにより希釈液を混合する。
3.3)試料に希釈C−Jを使用する。曲線の直線部分および上部および下部のプラトーを含めるために、必要に応じて追加の濃度を使用することができる。
1)バイオセンサーをPBS中で少なくとも10分間水和する。
2)アッセイプレートを準備する。黒色のポリプロピレン製マイクロプレートで、以下の試料プレートマップに従って、ウェル当たり200μLのPBS、アッセイ希釈液、AD中のIMP321の滴定、またはRaji細胞をそれぞれ適切なウェルに移す。
4)データを保存する場所とファイル名を入力する。
5)GOをクリックしてアッセイを実施する。
1)Octet Data Analysisソフトウェアで、分析するデータフォルダをロードする。
2)加工タブで、結合ステップを選択する。次に「選択したステップの定量化」をクリックする。
3)濃度情報を適宜入力する。
4)結果タブで、結合速度式としてR平衡(Req)を選択する。この式は、実験中に生成された結合曲線に適合し、平衡時の応答を出力シグナルとして計算する。
5)結合速度を計算するをクリックする。結果は自動的に表に表示される。
6)レポートを保存ボタンをクリックしてMSエクセルレポートファイルを生成する。
7)4パラメータロジスティック曲線近似プログラムであるSoftMax Proを使用して、ug/mLで表されるIMP321濃度に対する結合速度(nm)により標準曲線または試料曲線を作成する。例を図10に示す。
8)基準標準物質と試料のEC50比を用いて試料の相対結合効力を計算する。
以下の基準をすべて満たす場合、アッセイは有効である。
1)すぐ使用できるRaji細胞生存率>=60%
2)対照の相対活性は80%〜120%以内である。
3)対照のシグナル対バックグラウンド比(パラメータD/パラメータA)>=2。
4)平行度(比較可能性):標準との勾配比は0.8から1.4の間である。
5)アッセイ対照の結果が上記の基準を満たさない場合、アッセイは無効とみなされる。
1)臨床試料については、試料についての報告可能な値は、以下に詳述するように、2つまたは3つの有効かつ独立したアッセイ結果の平均として定義される。
差異%は次のように計算される。
絶対値(アッセイ1の結果−アッセイ2の結果)/平均値(アッセイ1の結果、アッセイ2の結果)×100%
2)2つのアッセイの差異%が20%以下の場合、2つのアッセイの平均結果を報告する。
3)2つのアッセイの差異%が20%を超える場合、1つの追加の有効なアッセイを実行する。
4)3試料アッセイのCVが25%以下の場合、3アッセイの平均結果を報告する。
5)3試料アッセイのCVが25%を超える場合、報告可能な値はない。再試験計画との不一致を起こす。
6)試料の報告可能な値がCOAに記載されている仕様を満たしていない場合は、再試験計画との不一致を起こす。
次のように試料の再試験を実行します。
1)3つの有効で独立したアッセイで試料を再試験する
2)3試料アッセイのCVが25%以下の場合、3アッセイの平均結果を報告する。
3)3試料アッセイのCVが25%を超える場合、報告可能な値はない。
4)再試験の結果がCOAに記載されている規格外(OOS)である場合、結論は不合格である。
BLIアッセイにおける溶液中のRaji細胞に対する固定化IMP321の特異的結合の決定
実施例6に記載のBLIアッセイを使用して、溶液中の異なる濃度(8E6/mL、4E6/mL、2E6/mL、1E6/mL)のRaji細胞に対する固定化IMP321の結合を決定した。Jurket細胞を陰性対照として使用した。得られた結合曲線および解離曲線を図11(a)に示す。図11(b)は、異なるRaji細胞濃度について得られた結合シグナルのグラフを示す。結果は、結合シグナルがRaji細胞の濃度に依存すること、すなわち、Raji細胞の濃度が高いほど、高い結合速度およびより高い上部プラトーが得られることを示している。同じアッセイにおいて、Jurket細胞の特異的結合は観察されなかった。
BLIアッセイにより測定したIMP321結合活性と既知の結合効力との相関
種々のレベルのRaji細胞結合効力を有する基準標準物質から希釈したIMP321の試料をBLIアッセイに用いて、アッセイにより測定された結合活性が試料の既知の結合効力と相関するかどうかを決定した。結果を以下の表に示す。図13は、BLIアッセイによって測定された結合効力パーセンテージ対それらの予想される効力のプロットを示す。
MHCクラスII結合活性を測定するためのBLIアッセイにおける凍結細胞の使用
実施例6に記載のBLIアッセイを実施して、培養または凍結保存溶液から得られた溶液中のRaji細胞に対する固定化IMP321の結合を比較した。溶液中の培養Raji細胞に対する異なる濃度の固定化IMP321の結合について得られた結合シグナルのプロットを図14(a)に示す。溶液中の予め凍結したRaji細胞に対する異なる濃度の固定化IMP321の結合について得られた結合シグナルのプロットを図14(b)に示す。
インプロセス試料試験
実施例6に記載のBLIアッセイを実施して、IMP321の様々な異なる製剤のMHCクラスII結合活性を決定し、CCL4放出アッセイによって決定されるように製剤の生物活性を比較した。
ストレスを受けたIMP321試料のBLIアッセイ試験および細胞に基づくCCL4放出アッセイとの相関
実施例6に記載のBLIアッセイを用いて、異なる処理(PNGaseでの処理による脱グリコシル化、37℃での保存、1%または0.1%過酸化水素処理による酸化、pH3.0、3.6、または3.1の酸による処理、pH9.2、9.75のアルカリによる処理、または光への曝露)にさらされたIMP321試料のMHCクラスII結合活性を測定した。結果を図15〜24に示す。
図15(b)は、Raji細胞に対する異なる濃度の固定化IMP321または脱グリコシル化IMP321の結合についてのBLIアッセイによって得られた結合シグナルのプロットを示す。
図16は、Raji細胞に対する、異なる濃度の、固定化した、IMP321または不適切に(37℃で12日間)保存したIMP321の結合シグナルのプロットを示す。図16(a)に示される結果はCCL4放出を測定する細胞に基づくアッセイによって得られ、そして図16(b)に示される結果はBLIアッセイによって得られた。
図17は、Raji細胞に対する、異なる濃度の、固定化した、IMP321または不適切に(37℃で1ヶ月間)保存したIMP321の結合シグナルのプロットを示す。図17(a)に示される結果はCCL4放出を測定する細胞に基づくアッセイによって得られ、そして図17(b)に示される結果はBLIアッセイによって得られた。
図18は、Rajiの細胞に対する、異なる濃度の、固定化した、未処理のIMP321または酸化IMP321(1%の過酸化水素による)の結合について、CCL4放出を測定する細胞に基づくアッセイ(図18a)またはBLIアッセイ(図18b)によって得られたシグナルのプロットを示す。
図19は、Rajiの細胞に対する、異なる濃度の、固定化した、未処理のIMP321または酸化IMP321(0.1%の過酸化水素による)の結合について、CCL4放出を測定する細胞に基づくアッセイ(図19a)またはBLIアッセイ(図19b)によって得られたシグナルのプロットを示す。

Claims (24)
- リンパ球活性化遺伝子−3(LAG−3)タンパク質、またはその断片、誘導体もしくは類似体を含む製剤のMHCクラスII結合活性を決定する方法であって、バイオレイヤー干渉法(BLI)を用いてMHCクラスII分子に対する前記LAG−3タンパク質、断片、誘導体、または類似体の結合を決定することを含む、方法。
- MHCクラスII発現細胞上に存在するMHCクラスII分子に対する前記LAG−3タンパク質、断片、誘導体または類似体の結合を決定するステップを含む、請求項1に記載の方法。
- 前記LAG−3タンパク質、断片、誘導体または類似体がBLIプローブの試薬層に固定化されており、かつ、前記MHCクラスII発現細胞が溶液中にある、請求項2に記載の方法。
- 前記MHCクラスII発現細胞が、少なくとも1E6/mL、好ましくは少なくとも4E6/mLまたは8E6/mLの密度で存在する、請求項3に記載の方法。

- 前記MHCクラスII発現細胞が、凍結保存溶液から得られた、解凍されてすぐ使用できる細胞である、請求項2〜7のいずれか1項に記載の方法。
- 複数の異なる濃度の前記LAG−3タンパク質、断片、誘導体、または類似体について、前記MHCクラスII分子への前記LAG−3タンパク質、断片、誘導体、または類似体の結合速度を決定するステップと、前記結合速度についての用量反応曲線を生成するステップとを含む、請求項1〜8のいずれか1項に記載の方法。
- LAG−3タンパク質、またはその断片、誘導体もしくは類似体の基準試料のMHCクラスII結合活性を、前記製剤の前記LAG−3タンパク質、断片、誘導体、または類似体の結合を決定するために使用したのと同じ条件下で、BLIを使用して前記MHCクラスII分子に対する前記基準試料の前記LAG−3タンパク質、その断片、誘導体、もしくは類似体の結合を決定することによって決定するステップと、前記基準試料について決定された前記MHCクラスII結合活性を前記製剤について決定された前記MHCクラスII結合活性と比較するステップとをさらに含む、請求項1〜9のいずれか1項に記載の方法。
- 前記基準試料の前記MHCクラスII結合活性が100%に設定される、請求項10に記載の方法。
- 前記基準試料が、そのMHCクラスII結合活性を低下させるように処理されているLAG−3タンパク質、またはその断片、誘導体もしくは類似体を含む、請求項10または11に記載の方法。
- 前記基準試料の前記LAG−3タンパク質、断片、誘導体または類似体が、脱グリコシル化されているか、37℃で少なくとも12日間保存されているか、酸化されているか、酸もしくはアルカリ処理により変性されているか、または少なくとも5日間光に曝露されている、請求項12に記載の方法。
- 試薬層を含む、LAG−3タンパク質、またはその断片、誘導体もしくは類似体のMHCクラスII結合活性を決定するためのBLIプローブであって、前記試薬層に前記LAG−3タンパク質、またはその断片、誘導体もしくは類似体が固定化されている、BLIプローブ。
- 前記試薬層に対する前記MHCクラスII発現細胞の非特異的結合を最小限に抑えるために、前記試薬層がブロッキング試薬で前処理されている、請求項14に記載のプローブ。
- 前記ブロッキング試薬がアルブミン、好ましくはBSAを含む、請求項15に記載のプローブ。
- LAG−3タンパク質、またはその断片、誘導体もしくは類似体のMHCクラスII結合活性を決定するためのキットであって、前記LAG−3タンパク質、またはその断片、誘導体もしくは類似体が固定化されている試薬層を有するBLIプローブと、MHCクラスII発現細胞とを含む、キット。
- 前記試薬層に対する前記MHCクラスII発現細胞の非特異的結合を最小限に抑えるために、前記BLIプローブの前記試薬層がブロッキング試薬で前処理されている、請求項17に記載のキット。
- 前記ブロッキング試薬がアルブミン、好ましくはBSAを含む、請求項18に記載のキット。
- 前記MHCクラスII発現細胞が凍結細胞である、請求17〜19のいずれか1項に記載のキット。
- 前記細胞がRaji細胞である、請求項17〜20のいずれか1項に記載のキット。
- 前記細胞が少なくとも1E6/mL、好ましくは少なくとも4E6/mLまたは8E6/mLの密度で存在する、請求項17〜21のいずれか1項に記載のキット。
- LAG−3タンパク質、またはその断片、誘導体、もしくは類似体を含む基準試料をさらに含む、請求項17〜22のいずれか1項に記載のキット。
- 前記基準試料の前記MHCクラスII結合活性が既知である、請求項23に記載のキット。
