適切な電位感受性タンパク質には、1つ以上のオプシン、電位感受性ドメインを含む1つ以上の他の分子、またはそれらの組み合わせが含まれるが、それらに限定されない。例えば、一実施形態では、電位感受性タンパク質は、QuasAr2、Ace2Nなどの微生物オプシンを含むが、それらに限定されない。別の実施形態では、電位感受性タンパク質は、カタユウレイボヤ(Ciona intestinalis)電位感知ドメイン(CiVSD)、ゼブラフィッシュ(Danio rerio)電位感知ドメイン(DrVSD)、セキショクヤケイ(Gallus gallus)電位感知ドメイン(GgVSD)、またはそれらの組み合わせを含む。

実施例1
この実施例は、QuasAr2とHaloTagとを接続する異なる長さのリンカーを有する様々なQuasAr2−HaloTag電位指示剤の蛍光応答を比較する。異なる長さを形成するために、アミノ酸残基をリンカーから除去した。QuasAr2−HaloTag(配列番号1及び配列番号2)、QuasAr2−HaloTag−4(配列番号3及び配列番号4)、QuasAr2−HaloTag−8(配列番号5及び配列番号6)、QuasAr2−HaloTag−12(配列番号7及び配列番号8)、QuasAr2−HaloTag−16(配列番号9及び配列番号10)、QuasAr2−HaloTag−18(配列番号11及び配列番号12)、QuasAr2−HaloTag−20(配列番号13及び配列番号14)、ならびにQuasAr2−HaloTag−22(配列番号15及び配列番号16)を含む、合計8つの異なるリンカー長を有する電位指示剤が形成された。各電位指示剤の末尾の数字は、そのリンカーから除去されたアミノ酸残基の数を示している。例えば、QuasAr2−HaloTag−4は4個のアミノ酸残基がリンカーから除去された電位指示剤であり、QuasAr2−HaloTag−12は12個のアミノ酸残基がリンカーから除去された電位指示剤である。
これらの電位指示剤の蛍光をASAP1に対して測定した。図4に示すように、それぞれの異なるリンカー長は、異なる大きさの蛍光応答を提供した。
実施例2
この実施例では、培養ラット海馬ニューロンにおいて、JF549で標識された電位指示剤を含む様々なQuasAr2の蛍光を測定した。具体的には、図5A〜Bは、JF549で標識されたQuasAr−HaloTag(配列番号1及び配列番号2)を発現しているラット海馬ニューロンの蛍光(図5A)、及びニューロンのフィールド電極誘導脱分極から生じる電位依存性蛍光の変化(図5B)を示す。図6A〜Bは、JF549で標識されたQuasAr2−HaloTag−16(配列番号9及び配列番号10)を発現しているラット海馬ニューロンの蛍光(図6A)、及びニューロンの自発活動電位から生じる電位依存性蛍光の変化(図6B)を示す。図7A〜Bは、JF549で標識されたQuasAr−cpHaloTag(配列番号17及び配列番号18)を発現しているラット海馬ニューロンの蛍光(図7A)、及びニューロンのフィールド電極誘導脱分極から生じる電位依存性蛍光の変化(図7B)を示す。図8A〜Bは、JF549で標識されたQuasAr2−SNAP−Tag(配列番号19及び配列番号20)を発現しているラット海馬ニューロンの蛍光(図8A)、及びニューロンのフィールド電極誘導脱分極から生じる電位依存性蛍光の変化(図8B)を示す。図9A〜Bは、JF549で標識されたHaloTag−QuasAr2(配列番号21及び配列番号22)を発現しているラット海馬ニューロンの蛍光(図9A)、及びニューロンのフィールド電極誘導脱分極から生じる電位依存性蛍光の変化(図9B)を示す。
実施例3
この実施例では、様々な蛍光色素で標識されたAce2N−HaloTag(配列番号23及び配列番号24)電位指示剤の蛍光を測定した。以下に論じるように、指示剤を培養ラット海馬ニューロン及び生きたゼブラフィッシュ幼体において試験したところ、細胞膜電位の変化は色素の蛍光に変化を生じた。

実施例3

Claims (20)

- 前記蛍光色素がアゼチジン含有ジャネリア蛍光(登録商標)色素を含む、請求項2に記載の電位指示剤。
- 前記ジャネリア蛍光(登録商標)色素が、ジャネリア蛍光色素(登録商標)505、ジャネリア蛍光色素(登録商標)525、ジャネリア蛍光色素(登録商標)549、ジャネリア蛍光色素(登録商標)585、ジャネリア蛍光色素(登録商標)646、及びそれらの組み合わせからなる群から選択される、請求項3に記載の電位指示剤。
- 前記電位感受性タンパク質がオプシンである、請求項1に記載の電位指示剤。
- 前記オプシンが微生物オプシンである、請求項5に記載の電位指示剤。
- 前記微生物オプシンが、QuasAr2、Ace2N、またはそれらの組み合わせからなる群より選択される、請求項6に記載の電位指示剤。

- 蛍光の変化が顕微鏡で観察される、請求項13に記載の方法。
- 前記電位指示剤が、前記電位感受性タンパク質と前記捕捉タンパク質との間にリンカーをさらに含む、請求項13に記載の方法。
