本明細書で提供される組み換え分泌シグナルに作動可能に連結されているタンパク質をコードするポリヌクレオチド配列を含む発現構築物が本明細書で提供される。組み換え分泌シグナルは通常、タンパク質のN末端に作動可能に連結されている。

本明細書で提供される発酵は、本明細書で提供される組み換え宿主細胞、及び組み換え宿主細胞を成長させるのに適する培養培地を含む。

種々の組み換えベクターでGS115(NRRL Y15851)のHIS+誘導体を形質転換することにより、シルク様タンパク質を分泌するピキア・パストリス(コマガタエラ・ファフィ(Komagataella phaffii))組み換え宿主細胞を生成した。
種々の組み換えベクターでGS115(NRRL Y15851)のHIS+誘導体を形質転換することにより、α−アミラーゼまたは緑色蛍光タンパク質のいずれかを分泌するピキア・パストリス(コマガタエラ・ファフィ)組み換え宿主細胞を生成した。
Claims (32)
- 発現構築物であって、組み換え分泌シグナルに作動可能に連結されているタンパク質をコードするポリヌクレオチド配列を含み、前記組み換え分泌シグナルがリーダーペプチド及びシグナルペプチドを含み、前記リーダーペプチドが天然pro−αMF(sc)、または配列番号1と少なくとも80%のアミノ酸配列同一性を有する機能的変異体であり、前記シグナルペプチドがpre−αMF(sc)を含まない、前記発現構築物。
- 前記機能的変異体が、1つまたは2つの置換アミノ酸を含む天然pro−αMF(sc)である、請求項1に記載の発現構築物。
- 前記機能的変異体が、配列番号2である、請求項1に記載の発現構築物。
- 前記機能的変異体が、配列番号1と少なくとも85%、少なくとも90%、少なくとも95%、または少なくとも99%のアミノ酸配列同一性を有する、請求項1に記載の発現構築物。
- 前記シグナルペプチドが表1から選択されるか、または、表1から選択されるシグナルペプチドと少なくとも80%のアミノ酸配列同一性を有する機能的変異体である、請求項1に記載の発現構築物。
- 前記機能的変異体が、表1から選択されるシグナルペプチドと少なくとも85%、少なくとも90%同一、少なくとも95%、または少なくとも99%のアミノ酸配列同一性を有する、請求項5に記載の発現構築物。
- 前記組み換え分泌シグナルが表2から選択されるか、または、表2から選択される組み換え分泌シグナルと少なくとも80%のアミノ酸配列同一性を有する機能的変異体である、請求項1に記載の発現構築物。
- 前記組み換え分泌シグナルが、表2から選択される組み換え分泌シグナルと少なくとも85%、少なくとも90%、少なくとも95%、または少なくとも99%のアミノ酸配列同一性を有する、請求項1に記載の発現構築物。
- 前記ポリヌクレオチド配列が、複数のコピーで存在する、請求項1に記載の発現構築物。
- 前記ポリヌクレオチド配列が、プロモーターに作動可能に連結されている、請求項1に記載の発現構築物。
- 前記プロモーターが、ピキア・パストリス(Pichia pastoris)のpGCW14プロモーターである、請求項10に記載の発現構築物。
- 前記プロモーターが、ピキア・パストリスのpGAPプロモーターである、請求項10に記載の発現構築物。
- 前記プロモーターが、誘導性プロモーターである、請求項10に記載の発現構築物。
- 前記タンパク質が、シルクまたはシルク様タンパク質である、請求項1に記載の発現構築物。
- 前記タンパク質が、配列番号17を含む、請求項14に記載の発現構築物。
- 前記タンパク質が、配列番号17の複数のコピーを含む、請求項14に記載の発現構築物。
- 請求項1〜16のいずれか1項に記載の発現構築物を含む組み換えベクター。
- 前記発現構築物が、複数のコピーで存在する、請求項17に記載の組み換えベクター。
- 前記ベクターが、PARSを含む、請求項17に記載の組み換えベクター。
- 請求項1〜16のいずれか1項に記載の発現構築物を含む組み換え宿主細胞。
- 酵母細胞である、請求項20に記載の組み換え宿主細胞。
- 出芽酵母細胞である、請求項21に記載の組み換え宿主細胞。
- メチロトローフ酵母細胞である、請求項21に記載の組み換え宿主細胞。
- ピキア種である、請求項22に記載の組み換え宿主細胞。
- ピキア・パストリスである、請求項23に記載の組み換え宿主細胞。
- 前記発現構築物が、前記組み換え宿主細胞のゲノム内に安定して組み込まれている、請求項20に記載の組み換え宿主細胞。
- 前記発現構築物が、前記組み換え宿主細胞において染色体外に維持されている、請求項20に記載の組み換え宿主細胞。
- 前記組み換え宿主細胞により産生される前記タンパク質の総収量の少なくとも30重量%の分泌収量である、前記発現構築物に含まれる前記ポリヌクレオチド配列によりコードされる前記タンパク質を産生する、請求項20に記載の組み換え宿主細胞。
- 請求項20〜28のいずれか1項に記載の組み換え宿主細胞及び培養培地を含む、発酵。
- 前記組み換え宿主細胞により産生される前記発現構築物に含まれる前記ポリヌクレオチド配列によりコードされる前記タンパク質の総収量の少なくとも30重量%を分泌タンパク質として含む、請求項29に記載の発酵。
- 前記発酵の前記培養培地が、前記発現構築物に含まれる前記ポリヌクレオチド配列によりコードされる前記タンパク質を少なくとも0.5g/L含む、請求項29に記載の発酵。
- タンパク質を産生するための方法であって、
(a)培養培地中で請求項20〜28のいずれか1項に記載の組み換え宿主細胞を培養して、請求項29〜31のいずれか1項に記載の発酵を得るステップ、及び
(b)前記培養培地から前記タンパク質を抽出するステップ
を含む、前記方法。
