前記第1の架橋ヒアルロン酸粒子及び第2の架橋ヒアルロン酸粒子の重量比は、1:0.5以上ないし2.5未満であり、
前記第1の架橋ヒアルロン酸粒子及び非架橋ヒアルロン酸の重量比は、1:0.1以上ないし1.2以下である皮膚注入用組成物を提供する。
(b)pHが7±1であり、
(c)粘度が40,000ないし90,000cPであり、
(d)浸透圧が平均0.325Osmol/kg±10%であり、
(e)押出力が30N以下であり、
(f)弾性値は、75ないし85であり、

以下、本発明を実施例を通じて詳細に説明する。下記の実施例は、本発明を例示するものに過ぎず、本発明の範囲が下記の実施例に限定されるものではない。本実施例は、本発明の開示が完全なようにし、本発明が属する技術分野で通常の知識を有する者に発明の範疇を完全に知らせるために提供されるものであり、本発明は、請求項の範疇によって定義されるだけである。
(1)粘度測定
大韓薬典一般試験法のうち、粘度測定法によって粘度を測定した。
具体的に、試料組成物500ulを粘度測定器(DV3T、Brookfield)のサンプルカップにロードし、CP−52のサンプルカップに装着した後、スピンドルの回転速度を2rpmに設定して粘度を測定した。
万能材料試験機(T0−101−161、TestOne)を用いて圧縮試験を行った。
具体的に、試料組成物を注射器に充填し、前記注射器に27G1/2インチの注射針を装着した後、治具にセットした。その後、速度50mm/min、変位25mmを設定した後、圧縮試験を行った。
レオメーター(rheometer)を使用して流動学的特性を測定した。
具体的に、平行板の間に試料を置いて平行板を振動回転させながら加わる力に対して、抵抗する力と消失する力を測定して、試料の貯蔵弾性係数(G’)、損失弾性係数(G”)、phase angle(位相角)を測定した。

(1)第1の架橋ヒアルロン酸粒子の製造
ヒアルロン酸ナトリウム(sodium hyaluronic acid)10g、精製水81g、1M NaOH(1M水酸化ナトリウム)9gを真空状態で400rpmで粒がなく透明な状態のゲルになるまで攪拌した。その後、架橋剤(BDDE、ブタンジオールジグリシジルエーテル)0.5gを追加して攪拌した。攪拌が完了した後、容器を密封して、80rpm、50℃の条件で1時間架橋反応させた。以後、27℃の条件で16時間放置してゲルを製造した。
ヒアルロン酸ナトリウム(sodium hyaluronic acid)20g、精製水117g、1M NaOH(1M水酸化ナトリウム)13gを、真空状態で400rpmで粒がなく透明な状態のゲルになるまで攪拌した。その後、架橋剤(BDDE、ブタンジオールジグリシジルエーテル)1gを追加して攪拌した。攪拌が完了した後、容器を密封して80rpm、50℃の条件で1時間架橋反応させた。以後、27℃の条件で16時間放置してゲルを製造した。
ヒアルロン酸ナトリウム(sodium hyaluronic acid)2gを精製水100gに添加し、攪拌して2%の非架橋ヒアルロン酸を製造した。
前記(1)で製造した第1の架橋ヒアルロン酸粒子、(2)で製造した第2の架橋ヒアルロン酸粒子、及び非架橋ヒアルロン酸を下記表1の含量(g)及び含量比で混合してヒアルロン酸組成物を製造した。
(1)ヒアルロン酸組成物の製造_比較製造例2−1ないし2−10
第1の架橋ヒアルロン酸粒子、第2の架橋ヒアルロン酸粒子、及び非架橋ヒアルロン酸をすべて含む組成物の比較例として、第1の架橋ヒアルロン酸粒子、第2の架橋ヒアルロン酸粒子、及び非架橋ヒアルロン酸の含量(g)及び含量比を下記表1のようにしてヒアルロン酸組成物を製造し、その他の条件は、製造例と同様にした。
比較製造例2−1ないし2−10のヒアルロン酸組成物の粘度、押出力、及び粘弾性の測定結果を下記表2及び3に示した。
(1)ヒアルロン酸組成物の製造_製造例1−1ないし1−4および比較製造例1−5ないし1−10
第1の架橋ヒアルロン酸粒子、第2の架橋ヒアルロン酸粒子、及び非架橋ヒアルロン酸の含量(g)及び含量比を下記表4のようにしてヒアルロン酸組成物を製造した。
製造例1−1ないし1−4及び比較製造例1−5ないし1−10のヒアルロン酸組成物の粘度、押出力及び粘弾性の測定結果を下記の表5及び6に示した。
(1)ヒアルロン酸組成物の製造_製造例1−1ないし1−2及び製造例1−11ないし1−13
第1の架橋ヒアルロン酸粒子、第2の架橋ヒアルロン酸粒子、及び非架橋ヒアルロン酸の含量(g)及び含量比を下記表7のようにしてヒアルロン酸組成物を製造した。
製造例1−1ないし1−2及び1−11ないし1−13のヒアルロン酸組成物の粘度、押出力、及び粘弾性の測定結果を下記表8及び9に示した。
Claims (15)
- 第1の架橋ヒアルロン酸粒子、前記第1の架橋ヒアルロン酸粒子と粒径が異なる第2の架橋ヒアルロン酸粒子及び非架橋ヒアルロン酸を含み、
前記第1の架橋ヒアルロン酸粒子及び第2の架橋ヒアルロン酸粒子の重量比は、1:0.5以上ないし2.5未満であり、
前記第1の架橋ヒアルロン酸粒子及び非架橋ヒアルロン酸の重量比は、1:0.1以上ないし1.2以下である、皮膚注入用組成物。 - 第1の架橋ヒアルロン酸粒子及び第2の架橋ヒアルロン酸粒子の重量比は、1:0.5以上ないし1.5未満であり、
第1の架橋ヒアルロン酸粒子及び非架橋ヒアルロン酸の重量比は、1:0.1以上ないし0.5未満である、請求項1に記載の皮膚注入用組成物。 - 第1の架橋ヒアルロン酸粒子の平均粒径は、10ないし250μmである、請求項1に記載の皮膚注入用組成物。
- 第2の架橋ヒアルロン酸粒子の平均粒径は、700ないし1500μmである、請求項1に記載の皮膚注入用組成物。
- 第1の架橋ヒアルロン酸粒子、第2の架橋ヒアルロン酸粒子又は非架橋ヒアルロン酸の分子量は、100万ないし150万Daである、請求項1に記載の皮膚注入用組成物。
- 第1の架橋ヒアルロン酸粒子または第2の架橋ヒアルロン酸粒子の架橋度は、10ないし20mol%である、請求項1に記載の皮膚注入用組成物。
- 第1の架橋ヒアルロン酸粒子、第2の架橋ヒアルロン酸粒子及び非架橋ヒアルロン酸は、全体の組成物100重量部対比1ないし10重量部で含まれるものである、請求項1に記載の皮膚注入用組成物。
- 上皮成長因子(EGF)をさらに含む、請求項1に記載の皮膚注入用組成物。
- 上皮成長因子(EGF)は、全体の組成物100重量部対比0.0001ないし0.002重量部で含まれるものである、請求項1に記載の皮膚注入用組成物。
- 麻酔剤の成分をさらに含む、請求項1に記載の皮膚注入用組成物。
- 麻酔剤の成分は、注入用組成物100重量部対比0.1ないし1重量部で含まれるものである、請求項1に記載の皮膚注入用組成物。
- 皮膚注入用組成物は、下記の理化学的特徴を有するものである、請求項1に記載の皮膚注入用組成物。
(a)外観に気泡がなく、無色、透明で、
(b)pHが7±1であり、
(c)粘度が40,000ないし90,000cPであり、
(d)浸透圧が平均0.325Osmol/kg±10%であり、
(e)押出力が30N以下であり、
(f)弾性値は、65ないし90であり、
(g)位相角(Phase angle)は、8ないし30である。 - 皮膚注入用組成物は、粘度が60,000ないし80,000cPである、請求項12に記載の皮膚注入用組成物。
- 皮膚注入用組成物は、押出力が25N以下である、請求項12に記載の皮膚注入用組成物。
- 皮膚注入用組成物は、弾性値が75ないし85であり、位相角が10ないし20である、請求項12に記載の皮膚注入用組成物。
