前記Fの免疫グロブリン不変領域は、免疫グロブリンCH1、CH2、CH3、及びCH4ドメインからなる群から選択された1つ〜4つのドメインを含むものであってもよい。
具体的には、前記Fの免疫グロブリン不変領域は、ヒンジ領域、CH2ドメイン及びCH3ドメインを含んでもよいが、特にこれに制限されない。
一方、免疫グロブリンFc領域は、ヒトまたは牛、ヤギ、ブタ、マウス、ウサギ、ハムスター、ラット、モルモットなどの動物起源であってもよく、好ましくは、ヒト起源である。
もう一つの具体例として、前記式(1)のL1及びL2のいずれかまたは両方がペプチド性リンカーである時、L1及びL2は、0個のアミノ酸からなるものであって、(i)XとF、またはFとYはペプチド結合によって結合されたものであり;または(ii)XとF、及びFとYはペプチド結合によって結合されたものであってもよい。
また、前記L2がペプチド性リンカーである場合、L2は(GS)m、(GGS)m、(GGGS)m、または(GGGGS)mであり、mは、1〜10であってもよいが、これに制限されない。
本発明において、前記「非ペプチド性リンカー」は、繰り返し単位が2つ以上結合された生体適合性リンカーを意味し、前記繰り返し単位は、ペプチド結合ではなく、任意の共有結合を介して互いに連結される。
また、前記非ペプチド性リンカーは、一種類の重合体だけでなく、異なる種類の重合体の組み合わせが用いられてもよい。
前記官能基は、アルデヒド基、マレイミド基及びスクシンイミド誘導体で構成された群から選択されてもよいが、これに制限されない。
前記で、アルデヒド基として、プロピオンアルデヒド基またはブチルアルデヒド基を例として挙げてもよいが、これに制限されない。
例えば、一方の末端にはマレイミド基を有してもよく、他方の末端にはアルデヒド基、例えば、プロピオンアルデヒド基、またはブチルアルデヒド基を有してもよい。両末端にヒドロキシ官能基を有するポリエチレングリコールを非ペプチド性重合体として用いる場合には、公知の化学反応によって前記ヒドロキシ基を前記様々な反応基で活性化したり、商業的に入手可能な変形された反応基を有するポリエチレングリコールを用いて本発明の結合体を製造してもよい。
例えば、前記非ペプチド性リンカーは、両末端にプロピオンアルデヒド基を有する非ペプチド性重合体、具体的には、両末端にプロピオンアルデヒド基を有するPEGであってもよいが、特にこれに制限されない。
一つの具体例として、前記製造方法は、(i)(a)生理活性物質であるX、ペプチド性または非ペプチド性リンカーであるL1、及び免疫グロブリンFc領域を含むFが結合された、X−L1−F 、及び(b)脳標的ペプチドであるY、ペプチド性または非ペプチド性リンカーであるL2が結合されたL2−Yを製造する段階;及び(ii)前記(a)X−L1−F及び(b)L2−Yを連結する段階を含む、前記式(1)の脳標的持続型結合体を製造する方法であることを特徴としてもよい。この時、もう一つの具体例として、前記(a)のL1はペプチド性リンカーであり、(b)のL2は非ペプチド性リンカーであることを特徴としてもよい。
本発明を具現する他の一つの様態は、前記脳標的ペプチド、前記脳標的ペプチドをコードする分離されたポリヌクレオチド、前記ポリヌクレオチドを含む発現ベクター、及び前記発現ベクターを含む形質転換体を提供する。
一方、前記脳標的ペプチドは、ペプチドそれ自体、その塩(例えば、前記ペプチドの薬学的に許容可能な塩)、またはそれらの溶媒和物の形態をすべて含む。
前記塩の種類は特に制限されない。ただ、個体、例えば哺乳類に安全かつ効果的な形態であることが望ましいが、特にこれに制限されるものではない。
前記用語、「薬学的に許容される」は、医薬的判断の範囲内で、過度の毒性、刺激、またはアレルギー反応などを引き起こすことなく、目的の用途に効果的に使用可能な物質を意味する。
また、本発明で用いられる用語、「溶媒和物」は、本発明に係るペプチドまたはその塩が溶媒分子と複合体を形成したものをいう。
本発明において、用語、「ベクター」とは、適当な宿主細胞で目的タンパク質を発現できる組換えベクターであって、核酸挿入物が発現されるように動作可能に連結された必須的な調節要素を含む核酸構造物(construct)を意味する。
本発明で前記脳標的ペプチドをコードするポリヌクレオチドは、プロモーターに作動可能に連結されてもよい。
本発明に係るポリヌクレオチドを含む組換えベクターを宿主細胞内に導入することにより、本発明の形質転換体(transformant)を獲得してもよい。
以下、下記実施例により本発明をより詳細に説明する。ただ、下記実施例は、本発明を例示するためのもので、本発明の範囲がこれに制限されるものではない。
免疫グロブリン不変領域を含み、FcRn結合部位を有する生体適合性の物質、脳標的ペプチド及び生理活性物質が、ペプチド性リンカーで連結された脳標的ペプチド及び生理活性ポリペプチドを含む持続性結合体を製造するために、免疫グロブリンFc領域及び脳標的ペプチドが結合された融合タンパク質を製造した。
そこで、脳標的ペプチドの例として、5つのペプチドを選択した(表1)。
前記ヒンジ領域が含まれたIgG4 Fc領域と脳標的ペプチドは、ペプチドリンカーで連結するか、直接融合する方式により、具体的には下記のようなヒンジ領域が含まれたIgG4 Fc領域と脳標的ペプチドとが含まれた融合タンパク質を合成した(Bioneerで合成)(表2)。下記表2−1〜2−6の配列で分かるように、IgG4 Fc領域のC末端に脳標的ペプチドのN末端がペプチドリンカーで連結されるか、直接融合する方式により製造された免疫グロブリンFc領域及び脳標的ペプチドが結合された融合タンパク質を製造した。
合成された免疫グロブリンFc領域と融合された脳標的ペプチドをコードする遺伝子は、両末端にNdeIとBamHIを含んでおり、NdeIとBamHIで切断したpET22b発現ベクターに挿入した。前記免疫グロブリンFc領域と融合された脳標的ペプチドをコードする遺伝子が挿入された発現ベクターをBL21(DE3)菌株に挿入し、免疫グロブリンFc領域と脳標的ペプチドが連結された持続型融合タンパク質を発現させた。
免疫グロブリンFc領域に融合する様々な脳標的ペプチドを探索するために、前記BTP1〜BTP5を含み、追加の脳標的ペプチドを設計して合成した(表3)。
前記脳標的ペプチドが実際に脳標的になるかどうかを確認するために、インビトロ(in vitro)及びインビボ(in vivo)試験を行った。
脳標的ペプチドの血液脳関門(blood brain barrier、BBB)の通過程度を確認するために、通過細胞外に排出(transcytosis)実験を行った。ヒト脳内皮細胞(Human cerebral endothelial cells)であるhCMEC/D3細胞を微多孔膜(microporous membrane)の上で5×104細胞/cm2で10日間培養した。BBBが形成がされたかどうかを確認するために、FITC−デキストラン(70kDa)で浸透実験(penetration test)を行い、タイトジャンクション(tight junction)が形成されることを確認した後、25mMのFITC−BTP5、FITC−BTP11、FITC−BTP12、 FITC−BTP16及びFITC− エキセンジン−4、FITC−P−8(陰性対照区、P−8)をhCMEC/D3の上に置き、2時間、24時間後にFITCの蛍光光度(fluorescence intensity)を測定した(図1)。
その結果、すべてのBTPはエキセンジン−4と陰性対照区(P−8)より高い通過を示した。時間が経つにつれて、BTP5、BTP11、BTP16などの脳標的ペプチドの通過が増加することが確認できた。
前記のような結果は、本発明で確認した脳標的ペプチドが効果的にBBBを通過できることを示唆する。
脳標的ペプチドの血液脳関門(blood brain barrier)の通過程度を確認するために、マウスでの脳分布実験(brain distribution test)を行った。マウスのCCA(common carotid artery)を介して2mg/headで投与した後にマウスの脳を分離した後、断片に切ってFITCの蛍光(fluorescence)を用い、顕微鏡で脳標的ペプチドの血液脳関門の通過程度を確認した(図2)。
CCAを介して注入された脳標的ペプチドは、注入特性上、一方の脳領域に広がるべきであるが、試験結果、脳の一方の領域に広がることを12.5倍率の顕微鏡で観察できた。また、長期記憶に重要な役割をする海馬体(hippocampus)の細胞体が分布する領域にFITCの蛍光が観察されることを200倍率の顕微鏡で確認できた。これにより、脳標的ペプチドが血液脳関門を通過して脳組織に入ることを確認した。
このような結果は、本発明の脳標的ペプチドがインビトロだけでなく、実際の生体内でも脳への標的化が可能であることを示唆するもので、脳標的ペプチドに結合された生理活性物質もBBBを通過して効果的に脳への伝達ができることを示唆する。また、持続型結合体の形態である本発明の結合体は、生体内持続性が増加し、このような持続性が増加するほど脳への標的化がさらに効率的に実行されうることを示唆する。
追加の融合タンパク質を発現するために発現ベクターを製造した。結合された生理活性ポリペプチドの半減期を増加させうる生体適合性物質として、ヒンジ領域が含まれたIgG4 Fc領域を選択し、前記ヒンジ領域が含まれたIgG4 Fc領域と脳標的ペプチドを遺伝子レベルで融合して発現ベクターにそれぞれ挿入した。
前記ヒンジ領域が含まれたIgG4 Fc領域と脳標的ペプチドは、ペプチドリンカーで連結するか、直接融合する方式によって、具体的には下記表4のような配列を有する、ヒンジ領域が含まれたIgG4 Fc領域とそのFc CH3領域のC末端の方向に脳標的ペプチドがペプチド結合で連結された融合タンパク質を合成した(韓国バイオニア社で合成)(表4)。下記表4−1〜4−7の配列から分かるように、IgG4 Fc領域のC末端に脳標的ペプチドのN末端がペプチドリンカーで連結されるか、直接融合する方式により製造された免疫グロブリンFc領域及び脳標的ペプチドが結合された融合タンパク質を製造した。
受容体結合力を増加させるための追加的な融合タンパク質を発現するために発現ベクターを製造した。結合された生理活性ポリペプチドの半減期を増加させうる生体適合性物質として、ヒンジ領域が含まれたIgG4 Fc領域を選択し、前記構築された発現ベクターにリンカーと脳標的ペプチドを遺伝子レベルで部位特異的変異(site-directed mutagenesis)PCR技法を用いて挿入した。具体的には、配列番号プライマー(表5)を用いて、免疫グロブリンFc領域のC末端と脳標的ペプチドのN末端の間、または脳標的ペプチドのC末端とベクターのBamHIが含まれた部位の間にリンカーと脳標的ペプチドを挿入した。前記のPCR技法で2つ以上に複製された脳標的ペプチドを含む融合タンパク質(表6)を発現する発現ベクターを構築した。この融合タンパク質は、表6に示した配列を有するが、例えば、Fc−BTP22結合体は前記表2のBTP5ペプチドの2単位がリンカーを介してFc切片のCH3領域のC末端方向にペプチド結合する。具体的には、下記表6で確認できるように、免疫グロブリンFc領域のC末端と脳標的ペプチドのN末端がペプチドリンカーで連結されるか、直接融合する方式によって製造された。この場合、脳標的ペプチドは、縦列(tandem)に連結される2つ以上の脳標的ペプチドは、1単位の脳標的ペプチドがリンカーで連結される形態である。
pET22bベクター(Novagen社)のT7プロモーター調節下の免疫グロブリンFc領域及び脳標的ペプチドが融合された融合タンパク質の発現を行った。それぞれの構築された発現ベクターでE.coli BL21−DE3(E. coli B F−dcm ompT hsdS(rB−mB−) gal λDE3);Novagen)を形質転換した。形質転換の方法は、Novagen社で推薦する方法に従った。各組換え発現ベクターが形質転換された個々の単一コロニーを取り、アンピシリン(50μg/ml)が含まれた2×ルリアブロス(Luria Broth、LB)培地に接種し、37℃で15時間培養した。組換え菌株の培養液と30%グリセロールが含まれた2×LB培地を1:1(v/v)の割合で混合して、各1mlずつクライオチューブに分注し、−140℃に保管した。これを組換え融合タンパク質の生産のための細胞ストック(cell stock)として使用した。
免疫グロブリンFc領域及び脳標的ペプチドが融合された結合体の発現のために、各細胞ストック(stock)1バイアル(vial)を溶かして、500mlの2×ルリアブロス(LB)培地に接種し、37℃で14〜16時間振とう培養した。OD600の値が4.0以上を示すと培養を終了し、これを種培養液として用いた。5L発酵器(Ependorf、アメリカ)を用いて、種培養液を1.6Lの発酵培地に接種し、初期バッチ(bacth)発酵を開始した。培養条件は、温度37℃、空気量2L/分(1vvm)、攪拌速度650rpm及び20%アンモニア水を用いてpH6.70に維持した。発酵の進行は、培養液内の栄養素が制限された時、追加培地(feeding solution)を添加して、流加培養を行った。菌株の成長は、OD値によってモニタリングし、OD値が120以上で最終濃度0.5mMのIPTGで導入した。培養は導入した後、約23〜25時間までさらに進行し、培養終了後、遠心分離器を使用して組換え菌株を収穫し、使用するまで−80℃で保管した。
前記実施例7で発現させた免疫グロブリンFc領域及び脳標的ペプチドが融合された融合タンパク質を可溶性の形態に変えるために、細胞を破砕してリフォールディングした。各培養液200mlに該当する細胞ペレットを200mlの溶解緩衝液(50mMトリス(pH9.0)、1mM EDTA(pH8.0)、0.2M塩化ナトリウム及び0.5%トリトンX−100)に再浮遊した。マイクロフルイダイザー(microfluidizer)プロセッサーM−110EH(AC Technology Corp. Model M1475C)を用いて、15,000psiの圧力で実行し、細胞を破砕した。破砕された細胞溶解物を12,200gで4℃で30分間遠心分離して上澄み液を捨てて、200mLの洗浄緩衝液(0.5%トリトンX−100及び20mMトリス(pH8.0))に再浮遊した。12,200gで4℃で30分間遠心分離してペレットを200mLの洗浄緩衝液(20mMトリス(pH8.0))に再浮遊した後、同様の方法で遠心分離した。ペレットを取って、100mLの緩衝液(8Mグアニジン−HCl、50mMトリス(pH8.0))に再浮遊して、常温で2時間攪拌した。2時間後、12,200gで23℃で30分間遠心分離した後、上澄み液を取った後に2.5mMジチオトレイトール(Dithiothreitol)を添加した後、37℃で30分間攪拌した。可溶化された免疫グロブリンFc領域及び脳標的ペプチドが融合された融合タンパク質のリフォールディング(refolding)のために、ここに3.5Lの緩衝液(3Mウレア、1mMシステイン、0.160Mアルギニン、50mMトリス(pH9.2)、20%グリセロール)を蠕動ポンプ(peristaltic pump)を用いて1mL/分の流速で入れながら4℃で36時間撹拌した。免疫グロブリンFc領域及び脳標的ペプチドが融合された融合タンパク質内の免疫グロブリンFc領域は、二量体(dimer)で発現する。
リフォールディングが終わった試料を12,200gで4℃で30分間遠心分離した後、上澄み液を取って0.22μmのフィルター(satorius)でフィルタリングした後、試料のpHを50%HClを用いてpH7.5に調整した。10mMトリス(pH7.0)を緩衝液で平衡化したプロテインA−セファロース(GE healthcare社)カラムに試料を結合させた後、100mMクエン酸ナトリウム(pH3.3)と10%グリセロール緩衝液を用いて、免疫グロブリンFc領域及び脳標的ペプチドが融合された結合体タンパク質を溶出した。
溶出された試料に0.6M硫酸アンモニウムを挿入した後、10mMトリス(pH7.5)と0.6M硫酸アンモニウム緩衝液で平衡化されたPhenyl HP(GE healthcare社)カラムに0.6M硫酸アンモニウムを挿入した試料を結合させた後、10mMトリス(pH7.5)緩衝液を用いて濃度が0%から100%になるように6カラム容量の直線濃度勾配で免疫グロブリンFc領域及び脳標的ペプチドが融合された融合タンパク質を溶出した。
免疫グロブリンFc領域と脳標的ペプチドが連結されている融合タンパク質の血液脳関門(blood brain barrier)の通過程度を確認するために、マウスで脳分布実験(brain distribution test)を行った。マウスの静脈内注射(intravenous)を介して、10mg/kgで投与した後、2時間後にマウスの脳を分離した後、免疫グロブリンFc領域と脳標的ペプチドが連結されている融合タンパク質の濃度を測定して、血液脳関門の通過程度を確認した(図3)。
その結果、免疫グロブリンFc領域と脳標的ペプチドの血液脳関門の通過程度は、免疫グロブリンFc領域より血清に比べて脳で高濃度で測定され、高い通過程度を示した。免疫グロブリンFc領域に同一種類の脳標的ペプチドが2つ以上連結されている融合タンパク質の場合、1つだけ連結された時より脳で高い濃度で測定され、高いT/S ratioを確認できた。
このような結果は、本発明の免疫グロブリンFc領域と脳標的ペプチドが融合された融合タンパク質が実際の生体内でも効果的にBBBを通過して、脳への伝達が可能であることを示唆する。また、持続型結合体の形態である本発明の融合タンパク質は、生体内の持続性が増加し、このような持続性が増加するほど脳への標的化がさらに効率的に実行されうることを示唆する。
以下では、本発明の免疫グロブリンFc領域と脳標的ペプチドが融合された持続型融合タンパク質に生理活性物質が連結された場合も、効果的にBBBを通過して脳に伝達されて、脳で生理活性を示すことを確認する実験を追加で行った。
典型的なサイズのタンパク質を生理活性物質として含む脳標的持続型結合体の例として、イデュルスルファーゼ(約72kDa)を含む脳標的持続型結合体を製造した。この脳標的持続型結合体の前駆体として、まずイデュルスルファーゼにリンカーとしてポリエチレングリコールが連結されたモノPEG化結合体を次のように製造した。
イデュルスルファーゼ(アイルランドShire社)を適正濃度まで濃縮した後、10Kブチル−ALD(2)PEG(両末端の水素がそれぞれブチルアルデヒド基に改質されている10kDaポリエチレングリコール、日本のNOF社)をイデュルスルファーゼのN末端にPEG化させるため、イデュルスルファーゼ:10Kブチル−ALD(2)PEGのモル比を1:10になるようにして、イデュルスルファーゼ濃度を10mg/mLとし、4℃で約2時間反応させた。この時、0.1Mリン酸カリウム(pH6.0)緩衝溶液に20mM濃度のNaBH3CN(sodium cyanoborohydirde)還元剤を添加して反応させた。反応液は、リン酸ナトリウム(pH6.0)とNaCl濃度勾配を利用したSource 15Q(GE、米国)カラムを用い、モノPEG化結合体であるイデュルスルファーゼ−10K PEGを精製した。
次に、イデュルスルファーゼ−10kDa PEG結合体を脳標的ペプチドを含むFc切片に連結させて脳標的持続型結合体を製造した。実施例12で精製したイデュルスルファーゼ−10K PEGと脳標的融合タンパク質の代表例であるFc−BTP22のモル比が1:10になるようにして、イデュルスルファーゼ濃度を10mg/mLとし、25℃で15時間反応させた。この時、反応液はダルベッコリン酸緩衝液(DPBS)と還元剤として10mM NaBH3CNを添加した。
反応が終結した後、反応液をリン酸ナトリウム緩衝液(pH6.0)とNaCl濃度勾配を用いてSource 15Q(GE、米国)カラムに適用し、硫酸アンモニウムとリン酸ナトリウム(pH6.0)の濃度勾配を用いてSource 15ISO(GE、米国)に適用し、目的とする脳標的持続型結合体であるイデュルスルファーゼ−10K PEG−Fc−BTP22を精製した。これはイデュルスルファーゼ−Fc−BTP22と混用される。
典型的なサイズのペプチドを生理活性物質として含む脳標的持続型結合体の例として、40個のアミノ酸残基からなるGIP(glucose-dependent insulinotropic polypeptide)受容体刺激活性のある人工GIP誘導体の脳標的持続型結合体を製造した。この持続型結合体の前駆体として、まず人工GIP誘導体のリンカーとしてポリエチレングリコールが連結されたモノPEG化結合体を次のように製造した。
10k MAL−PEG−ALD(一末端の水素原子が3−[N−(3−N−マレイミド−1−オキソプロリル)アミノ]プロピル基、反対側末端の水素原子が3−オキソプロピル基(プロピルアルデヒド基)に改質されている10kDaポリエチレングリコール、日本のNOF社)をこのGIP誘導体のC末端システインにPEG化させるために、GIP誘導体:PEGのモル比を1:1.3になるようにして、GIP誘導体の濃度を3mg/mLとし、4℃で約1時間反応させた。この時、50mM Tris−HCl(pH7.5)+45%イソプロパノール溶媒で反応させた。反応液は、クエン酸ナトリウム緩衝液(pH3.0)+45%エタノールとKCl濃度勾配を用いたSP HP(GE、米国)カラムを用い、モノPEG化結合体であるGIP誘導体−10K PEGを精製した。
次に、GIP誘導体−10K PEG結合体を脳標的ペプチドを含むFc切片に連結させて脳標的持続型結合体を製造した。実施例14で精製したGIP誘導体−10K PEGとFc−BTP22のモル比が1:2になるようにして、全体反応濃度を4.55 mg/mLとし、4℃で16時間反応させた。この時、反応液は、DPBSと還元剤として20mM NaBH3CNを添加した。
反応が終結した後、反応液はリン酸ナトリウム緩衝液(pH7.5)と硫酸アンモニウム濃度勾配を用いて、Source 15ISO(GE、米国)カラムに適用し、同じ緩衝液と同じカラムに再び適用して、目的とする脳標的持続型結合体であるGIP誘導体−10K PEG−Fc−BTP22を精製した。これはGIP誘導体−Fc BTP22と混用される。
実施例で得られた脳標的持続型結合体の血液脳関門の通過程度を確認するために、通過細胞外排出(transcytosis)実験を行った。ヒト脳内皮細胞(Human cerebral endothelial cells)であるhCMEC/D3細胞を微多孔膜(microporous membrane)の上で5×104細胞/cm2で10日間培養した。BBBが形成されたかどうかを確認するために、FITC−デキストラン(70kDa)で浸透実験(penetration test)を行い、タイトジャンクション(tight junction)が形成されたことを確認した後、25mMの脳標的持続型結合体をhCMEC/D3の上に置き、2時間、4時間後にELISA kitを用いてタンパク質の濃度を測定した。
その結果、GIP誘導体−10K PEG−Fc−BTP22は、対照群と比較して、血液脳関門の通過効果を示し、時間の経過とともに通過率も高くなることが確認できた(図4)。イデュルスルファーゼ−10K PEG−Fc−BTP22に対しても同様の血液脳関門の透過性が見られた(図示せず)。
前記のような結果は、本発明で確認した脳標的持続結合体が効果的にBBBを通過することができることを示唆する。
イデュルスルファーゼ−10K PEG−Fc−BTP22結合体の製造に伴うイデュルスルファーゼの酵素活性(specific activity)の変化があるかどうかを測定するために、インビトロ酵素活性実験を行った。イデュルスルファーゼの酵素基質として知られている4-Methylumbelliferyl α-L-Idopyranosiduronic Acid-2-sulfate Sodium Salt(4MU−α−IdopyraA−2)をイデュルスルファーゼβ及びIDS−Fc−BTP22と37℃で4時間反応させた後、二次反応酵素であるα−イズロニダーゼと再び37℃で24時間反応させた。最終的に生成された4−メチルウンベリフェロン(4-Methylumbelliferone;4MU)の蛍光を測定することにより、前記物質に対する酵素活性を測定した(図5)。
その結果、結合体を形成しない遊離イデュルスルファーゼとイデュルスルファーゼ−10K PEG−Fc−BTP22の酵素活性がそれぞれ23.4±1.94、10.8±1.39nmol/分/μgで示された。ここで酵素活性の値はイデュルスルファーゼの換算重量(μg)当たりの比活性(Specific activity)で計算したが、つまり、時間当たり酵素反応の数を遊離イデュルスルファーゼの場合は重量で、イデュルスルファーゼ−10K PEG−Fc−BTP22の場合はこの結合体内でイデュルスルファーゼ部分が占める換算重量で割った値である。結論として、イデュルスルファーゼに比べてイデュルスルファーゼ−10K PEG−Fc−BTP22の酵素活性が46.6%で、一部が減少したが、酵素を本発明の脳標的持続型結合体に製造したときにも、その結合体で有意な酵素活性が示されることを確認した。
前記で製造された持続性結合体は、Fc領域に結合された脳標的ペプチドによって血液脳関門の通過が可能であり、結合された生理活性物質の生理活性が維持されて、持続性かつ安定性が改善された持続性結合体を提供して、新しいBBB通過が可能な治療薬の開発プラットフォームを提供できる。

細胞表面抗原が、CD2、CD3、CD4、CD5、CD7、CD11a、CD11b、CD18、CD19、CD20、CD23、CD25、CD33、CD38、CD40、CD45、及びCD69からなる群から選択され、
抗体断片類が、scFv、Fab、Fab’、F(ab’)2及びFdからなる群から選択される、請求項5に記載の脳標的持続型結合体。 - 前記GLP−1受容体アゴニストが、天然型エキセンジン−4、天然型エキセンジン−4のN末端アミン基が除去されたエキセンジン−4、天然型エキセンジン−4のN末端アミン基がヒドロキシル基に置換されたエキセンジン−4、天然型エキセンジン−4のN末端アミン基がジメチル基に修飾されたエキセンジン−4、天然型エキセンジン−4のN末端アミン基がカルボキシル基に置換されたエキセンジン−4、天然型エキセンジン−4の一番目のアミノ酸(ヒスチジン)のα炭素が除去されたエキセンジン−4;及び前記エキセンジン−4の12番目のアミノ酸(リジン)がセリンに置換されたエキセンジン−4及び前記エキセンジン−4の12番目のアミノ酸(リジン)がアルギニンに置換されたエキセンジン−4からなる群から選択される、請求項6に記載の脳標的持続型結合体。
- 前記受容体媒介輸送による経路が、インスリン受容体(insulin receptor)、トランスフェリン受容体(transferrin receptor)、低密度リポタンパク質受容体(low density lipoprotein receptor)、低密度リポタンパク質受容体関連タンパク質(Low density lipoprotein receptor‐related protein)、レプチン受容体(leptin receptor)、ニコチン性アセチルコリン受容体(nicotinic acetylcholine receptor)、グルタチオン輸送体(Glutathione transporter)、カルシウム依存性カリウムチャネル(calcium-activated potassium channel)、及びRAGE(receptor for advanced glycation endproducts)、及び前記受容体のリガンド及び前記受容体またはリガンドに結合する抗体からなる群から選択されたいずれか一つを介して受容体媒介輸送の経路によって血液脳関門を通過する、請求項4に記載の脳標的持続型結合体。
- 前記L1が、FのN末端の領域に連結されて、L2が、FのC末端の領域に連結されたものである、請求項1に記載の脳標的持続型結合体。
- 前記式(1)のF−L2−Yが、

Fa及びFbが、それぞれのヒンジ領域、CH2ドメイン、及びCH3ドメインを含む免疫グロブリンFc領域を含む一本のポリペプチド鎖であって、Fa及びFbが、ヒンジ領域でジスルフィド結合で互いに連結されて、これにより前記結合体が、Fc切片を含み、Faが、L1と共有結合で連結され、
それぞれのBTPa1、......、BTPanが、互いに同じか異なる脳標的ペプチドであり、
それぞれのBTPb1、......、BTPbn’が、互いに同じか異なる脳標的ペプチドであり、
それぞれのL2a1、......、L2anが、互いに同じか異なるペプチド性リンカーであり、
それぞれのL2b1、......、L2bn’が、互いに同じか異なるペプチド性リンカーであり、
この時、n及びn’が、それぞれ独立して自然数である。 - 前記L1が、FaのN末端に連結されたものであり、L2a1及びL2b1が、それぞれFa及びFbのC末端に連結されたものである、請求項10に記載の脳標的持続型結合体。
- n=n’であって、それぞれL2a1=L2b1、......、L2an=L2bn’とBTPa1= BTPb1、......、 BTPan= BTPbn’の条件を満足する、請求項11に記載の脳標的持続型結合体。
- 前記L1が、XのN末端アミン基、リジンの側鎖に位置するアミン基、またはシステインの側鎖に位置する−SH基(チオール基)に連結されたものである、請求項10に記載の脳標的持続型結合体。
- 前記生理活性物質であるXが、2個〜1000個のアミノ酸からなるポリペプチドである、請求項10に記載の脳標的持続型結合体。
- 前記n及びn’が、1〜5である、請求項10に記載の脳標的持続型結合体。
- 前記L1が、0.5〜100kDaのサイズである非ペプチド性リンカーである、請求項10に記載の脳標的持続型結合体。
- 前記非ペプチド性リンカーが、ポリエチレングリコールである、請求項16に記載の脳標的持続型結合体。
- 前記L2が、ペプチド性リンカーであって、(GS)m、(GGS)m、(GGGS)m、または(GGGGS)mであり、mが、1〜10である、請求項1に記載の脳標的持続型結合体。
- 前記L1及びL2のいずれかが、ペプチド性リンカーであって、もう一つが、非ペプチド性リンカーである、請求項1に記載の脳標的持続型結合体。
- 前記L1が非ペプチド性リンカーであり、L2がペプチド性リンカーである時、ペプチド性リンカーが、0〜1000個のアミノ酸を含むものである、請求項1に記載の脳標的持続型結合体。
- 前記L1及びL2のいずれかまたは両方が非ペプチド性リンカーである時、非ペプチド性リンカーが、ポリエチレングリコール、ポリプロピレングリコール、エチレングリコール−プロピレングリコール共重合体、ポリオキシエチル化ポリオール、ポリビニルアルコール、多糖類、デキストラン、ポリビニルエチルエーテル、生分解性高分子、脂質重合体、キチン類、ヒアルロン酸、脂肪酸、高分子重合体、低分子化合物、ヌクレオチド及びこれらの組み合わせからなる群から選択されたものである、請求項1または10に記載の脳標的持続型結合体。
- 前記L1及びL2のいずれかまたは両方がペプチド性リンカーである時、前記XとF、またはFとYが、共有化学結合、非共有化学結合、またはこれらの組み合わせでL1及びL2によって互いに結合され、L1及びL2が、0〜1000個のアミノ酸を含むものである、請求項1に記載の脳標的持続型結合体。
- L1及びL2のいずれかまたは両方がペプチド性リンカーである時、L1及びL2が、0個のアミノ酸からなるものであって、
(i)XとF、またはFとYが、ペプチド結合によって結合されたものであり;または
(ii)XとF、及びFとYが、ペプチド結合によって結合されたものである、請求項1に記載の脳標的持続型結合体。 - L1及びL2のいずれかはペプチド性リンカーであり、このうち他の一つは非ペプチド性リンカーである時、
ペプチド性リンカーが、0〜1000個のアミノ酸を含むリンカーであり、
非ペプチド性リンカーが、ポリエチレングリコールである、請求項1に記載の脳標的持続型結合体。 - Fが、免疫グロブリンFc領域を含むものである、請求項1に記載の脳標的持続型結合体。
- 前記免疫グロブリンFc領域が、CH1、CH2、CH3、及びCH4ドメインからなる群から選択された1つ〜4つのドメインを含む、請求項25に記載の脳標的持続型結合体。
- 前記免疫グロブリンFc領域が、ヒンジ領域をさらに含むものである、請求項25に記載の脳標的持続型結合体。
- 前記免疫グロブリンFc領域が、IgGのFc領域である、請求項25または10に記載の脳標的持続型結合体。
- 前記免疫グロブリンFc領域が、IgG4 Fc領域である、請求項28に記載の脳標的持続型結合体。
- 前記免疫グロブリンFc領域が、ヒト配列の非糖鎖化IgG4 Fc領域である、請求項29に記載の脳標的持続型結合体。
- L1によってXのN末端及びFのN末端が連結されたものであり、L2によってYのN末端及びFのC末端が連結されたものである、請求項1に記載の脳標的持続型結合体。
- L1の一末端が、Xのリジン残基またはシステイン残基に、L1の他の末端が、FのN末端に連結されたものであり、
L2によってYのN末端及びFのC末端が連結されたものである、請求項1に記載の脳標的持続型結合体。 - 下記段階を含む、請求項1に記載の脳標的持続型結合体を製造する方法:
(i)(a)生理活性物質であるX、ペプチド性または非ペプチド性リンカーであるL1、及び免疫グロブリンFc領域を含むFが結合された、X−L1−F、及び(b)脳標的ペプチドであるY 、ペプチド性または非ペプチド性リンカーであるL2が結合されたL2−Yを製造する段階;及び
(ii)前記(a)X−L1−F及び(b)L2−Yを連結する段階。 - 下記段階を含む、請求項1に記載の脳標的持続型結合体を製造する方法:
(i)(a)生理活性物質であるX、及びペプチド性または非ペプチド性リンカーであるL1が結合された、X−L1及び(b)脳標的ペプチドであるY、ペプチド性または非ペプチド性リンカーであるL2、及び免疫グロブリンFc領域を含むFが結合されたF−L2−Yを製造する段階;及び
(ii)前記(a)X−L1及び(b)F−L2−Yを連結する段階。 - 下記段階を含む、請求項1に記載の脳標的持続型結合体を製造する方法:
(i)脳標的ペプチドであるY、ペプチド性または非ペプチド性リンカーであるL2、及び免疫グロブリンFc領域を含むFが結合されたF−L2−Yを製造する段階;及び
(ii)前記F−L2−YとXとをペプチド性または非ペプチド性リンカーであるL1で連結する段階。 - (a)のL1が、ペプチド性リンカーであり、(b)のL2が、非ペプチド性リンカーである、請求項33〜35のいずれか一項に記載の方法。
- (a)のL1が、非ペプチド性リンカーであり、(b)のL2が、ペプチド性リンカーである、請求項33〜35のいずれか一項に記載の方法。
- 前記L1が、両末端官能基を有する非ペプチド性重合体であって、前記両末端官能基が、それぞれアミン反応基(amine‐reactive group)またはチオール反応基(thiol‐reactive group)であり、前記両末端官能基が、互いに同じ種類であるか、異なる種類である、請求項1に記載の方法。
- 非ペプチド性リンカー及びX及びYが、それぞれの官能基によって連結され、この時、非ペプチド性リンカーの官能基がアルデヒド基、マレイミド(maleimide)基及びスクシンイミド誘導体(succinimide derivative)からなる群から選択されるものである、請求項33〜35のいずれか一項に記載の方法。
- 配列番号2、4、6、8、10、20、22、24、26、28、30、32、34、36、38及び40で表されるアミノ酸配列の中で選択されるいずれか一つのアミノ酸配列からなる、脳標的ペプチド。
- 請求項40に記載の脳標的ペプチドをコードする、分離されたポリヌクレオチド。
- 請求項41に記載のポリヌクレオチドを含む、組換え発現ベクター。
- 請求項42に記載の組換え発現ベクターを含む、形質転換体。
