Abstract

本発明は、例えば、以下の項目も提供する。

(a)SAMHD1阻害活性を保持するVpxタンパク質またはVprタンパク質と、(b)抗原をコードする外因性ポリヌクレオチドと、(c)DC−SIGNを発現する細胞に優先的に結合する複数のエンベロープ糖タンパク質とを含む偽型レンチウイルスベクター粒子を含む組成物であって、マンノシダーゼ阻害剤の不在下で調製された同じ偽型レンチウイルスベクター粒子の対照組成物と比較して、より高度にマンノシル化されている、組成物。

EndoHによる処理後の前記エンベロープ糖タンパク質の分子量が、(a)エンドグリコシダーゼにより処理されないエンベロープ糖タンパク質と、(b)PNGase Fにより処理されたエンベロープ糖タンパク質との間の距離の約70%以上シフトしている、項目6に記載の組成物。
(項目8)
EndoHによる処理後の前記エンベロープ糖タンパク質の分子量が、(a)エンドグリコシダーゼにより処理されないエンベロープ糖タンパク質と、(b)PNGase Fにより処理されたエンベロープ糖タンパク質との間の距離の約90%以上シフトしている、項目6に記載の組成物。
(項目9)
前記組成物中の前記エンベロープ糖タンパク質のうちの少なくとも30%が、マンノシダーゼ阻害剤の不在下で調製された偽型レンチウイルスベクター粒子の対照組成物中の同じアミノ酸配列(複数を含む)を有する対照糖タンパク質と比較して、増加した量のEndoH感受性グリカンを有する、項目1〜8のいずれか一項に記載の組成物。

ステリカップフィルター(Millipore)を使用して浄化した。ベクターは、1Lの遠心分離ボトル中、16,000gで、5時間回転させることによって濃縮した。それぞれのリットル採取物からのペレットは、1mLのHBSS中で再懸濁したか、または−80℃で保存するためにアリコートされたか、または1mLのベンゾナーゼ処理用緩衝液(50mM Tris−HCL pH7.5、1mM MgCl2、5% v/v Sucrose)中で再懸濁した。ベンゾナーゼヌクレアーゼを、最終的に、250U/mLで添加し、トランスフェクションからのいかなる使い残しのプラスミドを分解するために、4℃で一晩インキュベートした。ベンゾナーゼで処理したベクター調製物を、スクロースクッション(上部に30% スクロース、下部に5% スクロース)を使用して再濃縮し、超遠心分離機中、116,000gで、4℃で1.5時間遠心分離した。ベクターペレットは、1mL HBSS中で再懸濁し、アリコートし、−80℃で保存した。小規模なベクターの産生については、293T細胞を、2.5E5細胞/プレートで、10cmプレート中で播種し、ベクターの産生を比較する場合に、gag/polプラスミドの量を変えることを除いては、上記と同様に、PEIを使用したが、6μgのベクターゲノムプラスミドおよび3μgの残りのプラスミドを用いて、翌日トランスフェクトした。小規模なトランスフェクションは、正確さのために三連で行われた。5時間後、培地を、5% 血清、L−グルタミン、および抗生物質を含有する4mLのDMEM培地と交換した。ベクターは、トランスフェクションから2および3日後に採取し、0.45μm フィルターを使用して浄化した。ベクターは、−80℃で保存した。
