(ClpPに特異的なRNAi剤又は遺伝子編集剤のような)ClpPの阻害剤を個体に投与することを有する方法が提供されており、ClpPの阻害剤は、(例えばタンパク質及び/又はmRNAのレベルで検出され得るように)ClpPの活性レベル及び/又は発現レベルを低下させる。このような方法は、ClpPの発現レベル及び/若しくは活性レベルを低下させる方法、並びに/又は肥満及び/若しくは糖尿病の個体を処置する方法とみなされ得る。(例えばタンパク質及び/又はmRNAのレベルでの)ClpPの活性レベル及び発現レベル、並びにこのようなレベルを測定する方法の説明は、本明細書の他の箇所に見出され得る。

本明細書に記載されているスクリーニング法又は処置法の一又は複数で使用されてもよいClpPの小分子阻害剤は公知である。例えば、(A2-32-01として知られており、A2-32-01及びA2-32 として本明細書で互換的に称される)(3RS,4RS)-3-(non-8-en-1-yl)-4-(2-(ピリジン-3-yl)エチル)オキセタン-2-oneのようなβ−ラクトンは、バクテリアのClpP及び哺乳類のClpPの両方の阻害剤として識別された。例えば、Cole等著,2015, Cancer Cell 27, 864-876を参照し、この開示全体があらゆる目的のために参照によって本明細書に組み込まれている。
ある実施形態では、本方法(例えば本明細書に記載されているようなスクリーニング法又は処置法)は、薬剤(例えば試験薬、候補薬剤、ClpPの発現レベル及び/又は活性レベルを低下させるClpP阻害剤、例えばClpPの発現を特異的に低下させるRNAi剤又は遺伝子編集剤など)を個体に投与するステップを有する。状況に応じて、個体は、あらゆる好適な種(例えば哺乳類、齧歯類、マウス、ラット、非ヒト霊長類、ヒトなど)とすることができる。例えば、場合によっては、試験薬をマウスに投与する。場合によっては、(例えば主スクリーニング法によって識別されるような)候補薬剤を、マウス、ラット、非ヒト霊長類又はヒト(例えば肥満、糖尿病の個体、インスリン感受性が低下した個体、耐糖能が低下した個体など)に投与する。場合によっては、本明細書に記載されているようなClpPの発現レベル及び/又は活性レベルを低下させるClpP阻害剤を、マウス、ラット、非ヒト霊長類又はヒトに投与する。
上述された本主題の実施形態を含む態様が、単独で、又は一若しくは複数の他の態様又は実施形態と組み合わせることで有益であってもよい。上記の記載を制限することなく、本開示の1〜26と番号付けされたある非制限態様が以下に提示される。本開示を読むと当業者に明らかであるように、個々に番号付けされた態様の各々は、上記又は以下の個々に番号付けされた態様のいずれかと使用されてもよく、又は組み合わせられてもよい。これは、態様のこのような全ての組合せのサポートを提供することが意図されており、以下に明示的に提供される態様の組合せに限定されない。
(a) 哺乳類細胞を試験薬と接触させて、
(b) 接触させた後、基準値に対する哺乳類細胞のClpPの発現レベル及び/又は活性レベルを測定して、
(c) 前記試験薬が前記基準値に対して前記発現レベル及び/又は前記活性レベルを低下させたと判断し、
(d) 前記試験薬を肥満及び/又は糖尿病を処置するための候補薬剤として識別することを特徴とする方法。
個体の脂肪組織の量を減らす、個体の体重増加を防止する若しくは低下させる、個体のインスリン感受性を高める、及び/又は個体の耐糖能を高めるのに有効な量でClpPの阻害剤を前記個体に投与することを特徴とする方法。
以下の実施例は、本発明をどのように作製して使用するかについての完全な開示及び説明を当業者に提供するために記載されるのであって、本発明の範囲を制限することを意図するものではなく、以下の実験が、行われた全て又は唯一の実験であると表すことを意図するものでもない。使用された数値(例えば、量、温度等)に関して正確性を確保するために努力がなされたが、ある程度の実験誤差及び偏差は考慮に入れられるべきである。別途記載のない限り、部は重量部であり、分子量は重量平均分子量であり、温度は摂氏であり、圧力は大気圧であるか又は大気圧に近い圧力である。
以下の方法を、本明細書に記載されている実施例で利用した。
全てのマウスを無菌の動物設備で12時間の明/暗サイクルで維持した。マウスを、全ての代謝研究のために遺伝子型によって別々に収容した。高脂肪食(HFD )による肥満の研究のために、HFD (21重量%、脂肪からの42%kcal、0.2 %の総コレステロール;TD.01064、Harlan-Teklad )を動物に特定期間、与えた。麻酔をイソフルラン(生存処置)又はアベルチン(終末処置)で導入した。処置は全て、UCSF動物実験委員会によって承認され、NIH ガイドラインに従った。様々な分析のための動物の試料サイズを以前の文献に基づいて選択した。動物を遺伝子型によってグループ化し、ランダム化を使用しなかった。全ての行動試験に関して、全ての動物IDは試験者に対して盲検化された。試験者は他の分析のためには盲検化されなかった。
凍結ClpP+/- 精子を回復してClpP+/- マウスを生成した。ClpP-/- マウスを3世代以上、C57BL/6 遺伝的背景に戻し交配した。ClpP-/- マウスは生殖力がなかったので、ClpP+/- マウスを互いに繁殖させて3つの異なる遺伝子型(WT、ClpP+/- 、ClpP-/- )の同腹子を生成して使用し、系統を維持した。
ClpP遺伝子を相同の長腕(5’末端,約6Kb)及び相同の短腕(3’末端,約3Kb)と隣接させることにより、ClpP-cKO構築体を生成した。今後取り除くために、ClpPエキソン1〜3を含む領域の両端にLoxP部位を追加した。F3部位によって隣接したピューロマイシン耐性遺伝子を、3’末端loxP部位の目前に挿入した。標的とされたClpP対立遺伝子を保有するマウスES細胞クローンを選択して、マイクロインジェクションのために使用した。生じたキメラをFlp マウスと繁殖させて、標的とされたClpP対立遺伝子を保有する子孫を生成した。その後、異型接合のClpP-cKOマウスを繁殖させて、同型接合のClpP-cKOマウスを生成した。ClpP-cKOマウスをC57BL/6 遺伝的背景で生成した。同型接合のClpP-cKOマウスを使用して系統を維持した。
4時間の絶食後のマウスの体組成を、PIXImus2スキャナ(GE Healthcare Lunar)を用いて二重エネルギーX線吸収測定法(DEXA)によってイソフルラン麻酔下で解析した。
マウスを遺伝子型によって別々に収容した。ケージ毎に、4日間毎日午前10:00 に食料の重量を測った。開始時間の食料の重量から24時間後の食料の重量を引くことにより、ケージ毎の毎日の食料摂取量を算出した。毎日の食料摂取量を動物数又は体重のいずれかによって正規化した。
マウスは24時間、絶食して、その後に自由摂食に戻った。マウスの体重を絶食の前後に測定した。食料摂取量及び体重増加量を、8時間及び24時間の再摂食期間後にモニタした。
マウスの深部体温を齧歯類直腸検温器で得た。
血糖を血糖値測定器及びグルコース試験紙(Free Style Lite)を用いて測定した。血漿インスリンをインスリンElisa キット(Crystal Chem)を用いて定量化した。
グルコース及びピルビン酸塩に関して一晩絶食後、又はインスリンに関して4時間絶食後、糖負荷試験、インスリン耐性試験及びピルビン酸塩耐性試験をグルコース(2g/kg)、インスリン(0.5 単位/kg)又はピルビン酸塩(2g/kg)の腹腔内注射によって行った。血糖レベルを注射前及び注射後の様々な時点で測定した。グルコース刺激性のインスリン放出を評価するために、マウスを一晩(16時間)絶食させ、その後、グルコース(2g/kg)を腹腔内に注射した。血液試料を注射前及び注射後の様々な時点で尾静脈から収集した。
ClpP-/- マウスの一般的な神経学的行動プロファイルを握力試験、傾斜試験及びテールサスペンション試験によって評価した。これらのマウスの運動機能をロータロッド試験によって判断した。不安レベルを高架式十字迷路によって決定した。活性レベルをオープンフィールド試験によって調べた。これらのマウスの学習及び記憶をモリス水迷路を使用して検査した。聴覚機能をプレパルス抑制試験によって試験した。
組織(肝臓、内臓脂肪、腓腹筋、肩甲骨間褐色脂肪及び膵臓)を一晩、4%のパラホルムアルデヒド(pH 7.4)に固定してパラフィンに包埋し、7〜8μmで連続的に薄片に切り分けた。標準的なヘマトキシリン・エオシン染色を行った。
脂肪組織切片のヘマトキシリン・エオシン染色を行って、画像をimage J を用いて解析した。
WTマウス又はClpP-/- マウスをEM固定剤で潅流した。肝臓組織を収集して、2日間EM固定剤に固定した。組織を処理して、電子顕微鏡画像をJEOL JEM-1230 透過電子顕微鏡を使用して収集した。ミトコンドリアの数、面積及び丸さをImage J ソフトウェアによって13,600の倍率で測定した。
ClpP-cKOマウスをap2-Cre マウスと繁殖させることにより、脂肪細胞特異的ClpPノックアウトマウスを生成した。Cre+ マウスは脂肪組織のClpPタンパク質レベルを大幅に低下させ、Cre-同腹子は代謝分析の対照として機能した。
AAV8.2-CMV-Cre及び対照AAV8.2を利用した。これらのウイルスを5×109 ゲノムコピー/グラム(gc/グラム)体重の用量で尾静脈を通して3〜5ヶ月のClpP-cKOマウスに注射した。注射されたマウスの体重を週に1回測定した。血糖レベル及び血漿インスリンレベルを注射前及び注射の3週間後に測定した。糖負荷試験をこれらのマウスに注射の4週間後に行った。これらのマウスにHFD を注射の6週間後に与えた。その後、HFD に反応して体重増加が起こり、HFD を2週間与えた後に糖負荷試験を行った。
組織又は細胞を、プロテアーゼ阻害剤の混合物と共に低界面活性剤のバッファ(50mMトリス/HCl、pH 8.0、150 mM NaCl 、0.1 %SDS、0.5 %ノニデットP-40、0.5 %デオキシコール酸ナトリウム)に溶解した。溶解物バッファにホスファターゼ阻害剤を更に含めた。溶解物を10分間13,000rpm で遠心分離してペレットを除去した。業者のプロトコルに従って、InvitrogenのNuPAGE bis-tris システムを用いてSDS-PAGEを行った。標準的なウェスタンブロットプロトコルを、IRDye で標識した二次抗体(Li-cor)と共に使用した。Li-cor画像システムを使用してウェスタンブロット画像をスキャンした。
二次元蛍光ディファレンスゲル電気泳動解析のために、ClpP-/- マウス、ClpP+/- マウス又はWTマウスの器官を提出した。質量分析解析のために最も劇的に変化したスポットをゲルからカットしてタンパク質を識別した。これは同一の会社によって行われた。
新生の幼体から収集された皮膚組織からマウスの繊維芽細胞を調製した。皮膚組織を小片にカットして、100 mmの細胞培養皿に皮膚を上にして置いた。5〜10分後、10mlの繊維芽細胞培地(10%のFBS を含むDMEM)を細胞培養皿に徐々に加えた。埋込物を、加湿された37℃、5%CO2 の培養器で培養した。繊維芽細胞は最終的に培養中7〜10日後に増殖した。マウスの繊維芽細胞を多くの継代に亘って同一の培地で維持した。
ヒトのClpPのためのレンチウイルス過剰発現ベクタを、Life TechnologiesのpLenti7.3TOPO TAクローニングキットを用いて構築した。レンチウイルスを、Life TechnologiesのViraPower (商標)レンチウイルスパッケージングミックスを用いてパッケージング化した。空のベクタ(対照)又はヒトのClpP cDNA のいずれかを保有するレンチウイルスをClpP-/- 繊維芽細胞に加えて、更なる実験の前に48時間、培養した。
レンチウイルスshRNA 構築体を得た。レンチウイルスを、Life TechnologiesのViraPower (商標)レンチウイルスパッケージングミックスを用いてパッケージング化した。空のベクタ(対照)又は異なるshRNA のいずれかを保有するレンチウイルスをClpP-/- 繊維芽細胞に加えて、更なる実験の前に48時間、培養した。
マウスの繊維芽細胞を黒いきれいな底の96ウェルプレートにウェル当たり10,000で播いた。24時間、培養した後、細胞をOPTI-MEM培地に一晩培養した。細胞をH2O2で様々な用量で4時間処置した。その後、Alamar blue 試薬(Invitrogen)をウェルに(1:10の比率で)加えた。2時間の培養後、蛍光強度を590 nmの放射(560 nmの励起)で測定した。
マウスの繊維芽細胞をコンフルエンスまで繊維芽細胞成長培地で維持した。分析の前日、繊維芽細胞をトリプシン処理して、96ウェルSeahorse培養プレートにウェル当たり40,000細胞で播いた。分析の前に培養培地を、Seahorse Bioscience によって提供されているCO2 非依存性培地に変えた。その後、細胞をCO2 無しで0.5 〜1時間37℃で培養した。業者のプロトコルに従って、XF Cell mito-stress test kit及びXF Extracellular Flux Analyzer(Seahorse Bioscience )を用いて酸素消費速度(OCR )分析を行った。
マウスの血漿のGHレベル及びレプチンレベルを、Crystal Chem, Inc のマウスGH ELISAキット及び超高感度マウスインスリンELISA キットによって決定した。
使用された抗体は、抗-ClpP (ウサギ,Novus Biologicals )、抗-ClpX (SDI ,特注)、抗-Akt(ウサギ,Cell Signaling)、抗−ホスホ−Akt (ウサギ,Cell Signaling)、抗-Grp75(ウサギ,Cell Signaling)、抗-TRAP1(マウスmAb ,AbCam)、抗-LRPPRC (ウサギ,Proteintech Group)、抗-LonP (ウサギ,Sigma-Aldrich)、抗-OAT (マウス,Abcam )、抗-SDH2 (ウサギ,aric antibodies )、抗-ATP6V1A(ウサギ,Proteintech Group)、抗-CPS1 (マウス,Lifespan)、抗-Hsp70(ウサギ,Cell Signaling)、抗-GAPDH(マウスmAb ,Millipore )、抗Hsp60 (ウサギ,Abcam )、抗-VDAC (マウスmAb ,EMD Chemical)及び抗アクチン(ウサギ,Sigma-Aldrich )であった。
特に指定されていない限り、データを平均±SDとして示した。統計解析は全てPrism 6 ソフトウェア(GraphPad)を使用して行った。平均の差を、t検定、one-way ANOVA 又は反復測定ANOVA 、その後にボンフェローニ事後検定又はテューキー−クレーマー事後検定によって評価した。全ての場合で、P<0.05を統計的に有意であるとみなした。
哺乳類ClpXP プロテアーゼの生理的役割を理解するために、ClpPノックアウト(ClpP-/- )マウスモデルをジーントラップ法によって生成した(図7,パネルa)。定量ポリメラーゼ連鎖反応(qPCR)(不図示)及びウェスタンブロットにより、異なる器官にClpP遺伝子が効率的に除去されたことが確認された(図7,パネルb〜i)。ClpP-/- の幼体は生存可能に正常なメンデル比で生まれた。肝臓及び筋肉の形態的評価により、6ヶ月の同型接合(ClpP-/- )又は異型接合(ClpP+/- )のノックアウトマウスの異常がないことが明らかになった(図8)。一般的な神経学的行動、運動機能(ロータロッド試験及び泳ぐ速度試験)、不安(高架式十字迷路試験)、又は学習及び記憶(モリス水迷路試験)でClpP-/- 同腹子及びWT同腹子に差がなく(図9)、正常な神経学的プロファイルを示した。
低いレプチンレベルはWTマウスの摂食行動及び脂肪堆積をもたらす。より低い血清レプチンレベルと一致して、全てのグループのマウスで1日当たり動物毎に同様の食料摂取量であった(図2、パネルa)が、体重に正規化された食料摂取量は、WTマウス及びClpP+/- マウスよりClpP-/- マウスで著しく多かった(図2、パネルb)。従って、ClpP-/- マウスはより多くの食料を消費しながら、体重を増やさず脂肪を減らし、より多くのエネルギーを消費したか、又は栄養を適切に利用しなかったことを示唆している。
ClpP-/- マウスの血糖レベル及び血漿インスリンレベルを解析した。血糖レベルは、ClpP+/- マウス及びWTマウスよりClpP-/- マウスで著しく低かった(図3,パネルa)。血漿インスリンレベルも、自由摂食及び絶食の両方の条件下でWT対照よりClpP-/- マウスで著しく低く、ClpP+/- マウスの血漿インスリンレベルは中間であった(図3,パネルb〜c)。低いインスリンレベルは、形態データに基づき、膵臓欠損による可能性が低かった(図11,パネルb)。従って、ClpP-/- マウスは、非常に低いインスリンレベルを有するにもかかわらず正常な血糖レベルを維持した。HFD の2ヶ月後、WTマウス及びClpP+/- マウスの血糖レベルは大幅に上昇した。対照的に、ClpP-/- マウスは、HFD で遥かに低い血糖レベル及びインスリンレベルを維持した(図3,パネルd〜e)。糖負荷試験及びインスリン耐性試験によって、ClpP-/- マウスのインスリン感受性が高められたことが更に明らかになった(図3,パネルf〜h)。ClpP-/- マウスのピルビン酸塩耐性の上昇傾向が、有意性に達していないが検出された(図3,パネルi)。インスリン感受性の上昇の裏付けとして、インスリンシグナル経路の主成分であるp-AKT のより高いレベルが、ClpP-/- マウスの筋肉及び繊維芽細胞で見つかった(図3,パネルj〜l)。更に、ClpP-/- 繊維芽細胞は、WT繊維芽細胞より培養物のIGF によるAKT 活性化に更に反応し易かった(図3,パネルm)。これらのデータは、ClpPの除去がマウスのインスリン感受性を高めるという結論を強く裏付けている。
ClpPの下流エフェクタを識別するために、二次元(2D)蛍光ディファレンスゲル電気泳動(2D-DIGE )を使用して、異なるClpP遺伝子型を有するマウスの様々な器官のタンパク質プロファイルを比較した(図15)。その後、質量分析を使用して、ClpP-/- マウスの様々な器官の最も劇的に変化したタンパク質を識別した(図20)。ウェスタンブロットによる検証によって、6つの可能性のあるClpXP 下流エフェクタ(TRAP1 、Grp75 、LRPPRC、ClpX、OAT 及びLonP;図21)が確認され、これらのタンパク質レベルは多くの器官で大幅に上昇した(図4,パネルa〜h及び図16)が、mRNAレベルは、ほとんどの場合で同一のままであった(図22)。従って、ClpPは、これらのタンパク質のレベルを転写後に制御し得る。タンパク質レベルの著しい上昇が、体重の減少無しで胎生19日齢の幼体(不図示)で更に検出され、これらのタンパク質の蓄積が、観察された表現型の結果ではなく、原因であり得ることを示唆している。同一のタンパク質のレベルの上昇がマウスの繊維芽細胞で更に検出され(図4,パネルi〜j)、これらのレベルは、その後、マウスのClpPを過剰発現させることにより低下し(図4,パネルk〜l)、ClpPが細胞自律的にこれらのタンパク質レベルを特異的に調節することを示唆している。
ミトコンドリアの数及び機能をClpP-/- マウスで解析した。ClpP-/- 肝臓切片の電子顕微鏡(EM)研究によって、ClpPの枯渇がミトコンドリアの数を増加させて、肝実質細胞のミトコンドリアの形態を変えたことが示された(図5,パネルa〜b及び図17,パネルa〜b)。(フィールド当たりの総ミトコンドリア面積によって測定された)ミトコンドリアの質量は、ClpP-/- 肝実質細胞で更に著しく増加した(図5,パネルc及び図17,パネルa〜b)。ClpP-/- ミトコンドリアは、恐らくClpP-/- ミトコンドリアのタンパク質含有量の増加により、WTと比較してより密なマトリクスを有した(図5,パネルa及び図17,パネルa〜b)。ClpP-/- 肝実質細胞及びWT肝実質細胞のミトコンドリアのサイズ分布は同様であった(図17,パネルc)が、丸さがより低いClpP-/- ミトコンドリアの割合がより高く(図17,パネルd)、より多くのClpP-/- ミトコンドリアの形状が細長かったことを示唆している。
ClpPによって調節されたミトコンドリアのエフェクタがClpP-/- 繊維芽細胞で見られたミトコンドリアの機能の向上を媒介したか否か、またClpPによって調節されたミトコンドリアのエフェクタのどれがClpP-/- 繊維芽細胞で見られたミトコンドリアの機能の向上を媒介したかを判断するために、レンチウイルスshRNA を使用してClpP-/- 繊維芽細胞のミトコンドリアのエフェクタの各々をノックダウンした。この後、機能分析を行った(図18,パネルa〜d)。LRPPRC及びClpXではなくTRAP1 又はGrp75 をノックダウンすることにより、H2O2細胞毒性に対するClpP-/- 繊維芽細胞の耐性が消失又は低下した(図5,パネルh〜i及び図18,パネルe〜f)。LRPPRC又はTRAP1 のいずれかをノックダウンすることにより、ClpP-/- 細胞の呼吸容量の増加が消失した一方、ClpX又はGrp75 のレベルの低下はこのような影響を及ぼさなかった(図5,パネルj)。これらのデータは、エフェクタレベル、特にTRAP1 、Grp75 及びLRPPRCを制御することによりClpPがミトコンドリアの機能を調節することを示唆している。これらのタンパク質は、別個の経路を介してミトコンドリアの機能に影響を及ぼすか、又は同一の経路で相乗効果をもたらす場合がある。
多くの組織は、異なるメカニズムによってインスリン感受性に影響を及ぼし得る。ClpPコンディショナルノックアウトマウス(ClpP-cKO)を使用して、ClpP-/- マウスの表現型の基礎となる一又は複数の組織特異的なメカニズムを更に分析した。このマウスモデルでは、今後取り除くために、ClpPエキソン1〜3を含むマウスゲノム領域に隣接させるためにLoxP部位を挿入した。特に肝臓のClpPを除去するために、AAV-CMV-Cre をClpP-cKOマウスに尾静脈を介して注射した。免疫染色により検出された肝臓のClpPレベルは、AAV が注射された対照マウスと比較して、Cre が注射されたマウスで劇的に低下した(図6,パネルa)。注射の3週間後に、2つのグループのマウスの総体重に有意差はなかった(図6,パネルb〜c)が、Cre が注射されたマウスは対照マウスより体重増加が著しく低かった(図6,パネルd)。Cre が注射されたマウスの血糖レベルも対照マウスの血糖レベルより著しく低かった(図6,パネルf)が、血漿インスリンレベルの差は検出されなかった(図6,パネルe)。Cre が注射されたマウスのインスリン感受性の向上は、食事制限又はHFD を2週間行ったマウスで糖負荷試験によって明らかにされた(図6,パネルg〜h)。ClpP-/- マウスとは異なり、ClpPの肝臓特異的なノックダウンは、HFD による体重増加に影響を及ぼさなかった(図6,パネルi)。これらのデータは、肝臓のClpPレベルが、脂肪含有量に影響を及ぼすことなく、ClpP-/- マウスで観察されたインスリン感受性の調節に関与することを示唆している。
野生型マウスMEF 細胞を、処置の前日に6つのウェルプレートにウェル当たり200,000 の細胞で播いた。細胞を、OPTI-MEMで(本明細書に既に記載されている)10μMの小分子ClpP阻害剤のA2-32-01、AV167 及びAV179 で2日間(48時間)1日に2回処置した。処置毎に古い培地を取り除き、新しい培地をウェルに追加した。48時間の処置後、細胞をClpP基質のウェスタンブロット解析又はqPCR分析のために収集した。
7〜9ヶ月のWTメスの(20匹のマウス)に3週間高脂肪食を与えた。HFD に応じた体重変化及び血糖/血漿インスリンのレベルを測定した。4週目の初めに処置グループ毎にA2-32-01を300 mg/kg 、8日間1日2回腹膜に注射した。対照グループについては、トウモロコシ油(賦形剤)を注射した。マウスをHFD で維持した。毎日、体重変化を追跡した。A2-32-01注射の8日目に血糖レベルを測定し、糖負荷試験を適用してインスリン感受性を測定した。インビボA2-32-01処置の最初の7日間の体重データが図28に示されており、賦形剤対照グループと比較して2日目〜7日目で体重の著しい減少を示す。インスリン感受性を反映する糖負荷試験を8日目に行った。そのデータが図29に示されており、賦形剤対照マウスと比較してA2-32-01で処置されたマウスでインスリン感受性の有意な向上を示す。
実施例9のインビボ解析を継続し、A2-32-01注射の9日目に血液及び多くの器官(肝臓/筋肉/脳)を収集する。ミトコンドリアをマウスの肝臓から分離し、ClpP活性を、例えば本明細書に記載されているようにペプチド基質を使用して試験する。ウェスタンブロット解析を行って、ClpP基質レベルを検出する。

Claims (26)
- 肥満及び/又は糖尿病を処置するための候補薬剤を識別する方法であって、
(a) 哺乳類細胞を試験薬と接触させて、
(b) 接触させた後、基準値に対する哺乳類細胞のClpPの発現レベル及び/又は活性レベルを測定し、

- 前記哺乳類細胞はエクスビボにあることを特徴とする請求項1〜3のいずれか一項に記載の方法。
- 前記哺乳類細胞はインビボにあることを特徴とする請求項1〜3のいずれか一項に記載の方法。
- 前記哺乳類細胞は肝実質細胞であることを特徴とする請求項1〜6のいずれか一項に記載の方法。
- 接触させる際に、前記試験薬をマウスに投与することを特徴とする請求項2に記載の方法。
- 前記基準値を得るためにステップ(a) で接触させる前に前記マウスのClpPの発現レベル及び/又は活性レベルを測定することを特徴とする請求項8に記載の方法。
- ステップ(d) の後、識別された前記候補薬剤を、肥満及び/又は糖尿病を患う個体に投与することを特徴とする請求項1〜9のいずれか一項に記載の方法。
- 前記個体はマウス、非ヒト霊長類又はヒトであることを特徴とする請求項10に記載の方法。
- 識別された前記候補薬剤を前記個体に投与した後、インスリン感受性、血糖レベル、耐糖能、体脂肪量、脂肪組織の量、白色脂肪組織の量、体脂肪率、体重、内臓脂肪の脂肪細胞サイズ、血漿レプチンレベル、成長ホルモンレベル、基礎エネルギー消費量、筋肉及び/又は繊維芽細胞のリン酸化AKT (p-AKT )レベル、除脂肪率、肝実質細胞のミトコンドリアの数、肝実質細胞のミトコンドリアの質量、肝実質細胞のミトコンドリアの形態、繊維芽細胞の呼吸容量、繊維芽細胞の最大酸素消費速度(OCR )及びH2O2による細胞毒性に対する繊維芽細胞耐性から選択された前記個体の一又は複数の特徴を測定することを特徴とする請求項10又は11に記載の方法。
- 前記試験薬は小分子又はポリペプチドであることを特徴とする請求項1〜12のいずれか一項に記載の方法。
- 定量RT-PCR、マイクロアレイ又はRNA シーケンシングを使用して、RNA 発現レベルであるClpPの発現レベルを測定することを特徴とする請求項1〜13のいずれか一項に記載の方法。
- タンパク質の発現レベルであるClpPの発現レベルを測定し、測定する際に、抗ClpP抗体、質量分析及び/又は酵素結合免疫吸着測定法(ELISA )を使用してClpPタンパク質を検出することを特徴とする請求項1〜13のいずれか一項に記載の方法。
- TNF 受容体関連タンパク質1(TRAP1 )、熱ショックタンパク質ファミリーA(Hsp70 )メンバー9(Grp75 )、ロイシンリッチペンタトリコペプチドリピート含有(LRPPRC)、カゼイン分解性ミトコンドリアマトリックスペプチダーゼシャペロンサブユニット(ClpX)、オルニチンアミノトランスフェラーゼ(OAT )及びlon ペプチダーゼ1(LonP1 )から選択された一又は複数のタンパク質の量の増加を測定することにより、ClpPの活性レベルの低下を測定することを特徴とする請求項1〜13のいずれか一項に記載の方法。
- 複数の試験薬をスクリーニングして、肥満及び/又は糖尿病を処置するための一又は複数の候補薬剤を識別することを特徴とする請求項1〜16のいずれか一項に記載の方法。
- 肥満及び/又は糖尿病の個体を処置する方法であって、
個体の脂肪組織の量を減らす、個体の体重増加を防止する若しくは低下させる、個体のインスリン感受性を高める、及び/又は個体の耐糖能を高めるのに有効な量でClpPの阻害剤を前記個体に投与することを特徴とする方法。 - ClpPの前記阻害剤は、ClpPの発現を特異的に低下させるRNAi剤又は遺伝子編集剤であることを特徴とする請求項18に記載の方法。
- ClpP発現の低下が実質的に肝臓特異的であるように、ClpPの前記阻害剤を投与し、投与される量は、個体のインスリン感受性を高めるのに有効な量であることを特徴とする請求項18又は19に記載の方法。
- ClpPの前記阻害剤は小分子であることを特徴とする請求項18に記載の方法。
- 前記小分子はβ−ラクトンであることを特徴とする請求項21に記載の方法。
- β−ラクトンは、(3RS,4RS)-3-(non-8-en-1-yl)-4-(2-(ピリジン-3-yl)エチル)オキセタン-2-one、又はその薬学的に許容される塩であることを特徴とする請求項22に記載の方法。
- ClpPの前記阻害剤を前記個体の肝臓に直接送達し、投与される量は、個体のインスリン感受性を高めるのに有効な量であることを特徴とする請求項18〜23のいずれか一項に記載の方法。
- 投与する際に局所注射することを特徴とする請求項18〜24のいずれか一項に記載の方法。
- 前記個体のインスリン感受性を測定することを特徴とする請求項18〜25のいずれか一項に記載の方法。
