合成抗体ライブラリーをスクリーニングし、ヒトおよび齧歯類αKlothoに対して高親和性を有する抗原結合フラグメント(Fab)を作製した。この新規な抗体sb106を、組換えタンパク質、培養細胞、ならびにヒトおよび齧歯類由来の体液および組織を用いて特性評価した。ヒトおよび齧歯類の血清および尿中のαKlothoレベルは正確に定量することができ、早期ヒトCKDでは、血清および尿両方のαKlothoが劇的に低下していることが示される。sb106抗体は、Klothoのα型に特異的であり、現行の市販のαKlotho検出試薬に比べて、明瞭かつ特異的な方法で患者血清サンプルからαKlothoを上手く捕捉することが最初に知られたものである。細胞において、この抗体はαKlothoを免疫沈降させ、それを免疫細胞化学により標識することができる。動物では、この抗体は、血漿からαKlothoを免疫沈降させるのに効果的である。sb106抗体の、体液から少量のαKlothoを検出する能力は、この抗体を、αKlothoのレベルが異常な疾患の診断のための有価な試薬とする。さらに、sb106抗体は、FGF23により媒介されるシグナル伝達経路を含む生理学的状態および病的状態の研究において研究試薬として有価である。この抗体は、酵素結合免疫吸着アッセイ(ELISA)、近接ライゲーションアッセイ(PLA)、および質量分析に基づく技術などの様々な技術を用いる、血清などのヒトおよび齧歯類サンプル中の可溶性αKlothoに関する特異的アッセイで使用可能である。
ヒトFGFR1cのリガンド結合ドメイン(D142〜R365)は、大腸菌(E.coli)で発現され、封入体からイン・ビトロで再折り畳みされ、公開されている方法[72、73]により精製した。マウスαKlothoの細胞外ドメイン(A35〜K982)は、C末端FLAGタグとともにHEK293細胞で発現させ、αKlothoエクトドメインとFGFR1cリガンド結合ドメインの二元複合体は記載のように作製した[9]。
Sb106は、合成ヒトFabファージディスプレーライブラリー(ライブラリーF)から単離された[74]。結合選択、ファージELISAおよびFabタンパク質精製 は記載のように行った[67、75、76]。簡単に述べれば、96ウェルMaxisorp Immunoplates(Fisher Scientific、ネピアン、ON、カナダ)にて捕捉標的としてαKlotho細胞外ドメインFGFR1cリガンド結合ドメインの二元複合体で複数回パンニングすることによりライブラリーFからのファージを循環させた。5回の選択後、96ウェル形式で増殖させた個々のクローンからファージを生成し、特異的結合クローンを検出するためにファージELISAを行った。結合を示したクローンに対してDNAシークエンシングを行った。競合的結合ELISAは、sb106−ファージを可溶性ヒトαKlothoの希釈系(50〜0.0005nM×1時間)とともにプレインキュベートした後、ヒトαKlothoでコーティングしたELISAプレートに結合させることにより行った。sb106の重鎖可変および軽鎖ドメインをコードする遺伝子をそれぞれ軽鎖またはIgG1重鎖の生産用に設計されたベクターにクローニングし、sb106−IgGを293F細胞(Invivogen、サンディエゴ、CA.USA)から発現させた。FabおよびIgGタンパク質をプロテインAアフィニティーカラム(GE Healthcare、ミシサガ、ON、カナダ)でアフィニティー精製した。
FGFR1cリガンド結合ドメインとマウスαKlothoエクトドメインの二元複合体(αKlotho−FGFR1c複合体と呼称)を公開されているプロトコール[9]によって作製した。N末端ヘキサヒスチジンタグを有する成熟型のヒトFGF23(Y25〜I251)を公開されているプロトコール[73、74、77]により、大腸菌で発現させ、精製した。
リアルタイムタンパク質−タンパク質相互作用を、Biacore 2000表面プラズモン共鳴(SPR)分光計(Biacore AB/GE Healthcare)を用い、25℃、HBS−EPバッファー(10mM HEPES−NaOH、pH7.4、150mM NaCl、3 mM EDTA、0.005%(v/v)ポリソルベート20)中で測定した。タンパク質は、研究級のCM5バイオセンサーチップ(Biacore AB/GE Healthcare)のカルボキシメチル(CM)デキストランに、それらの遊離アミノ基を介して共有結合させた。タンパク質をバイオセンサーチップ上に、流速50μl 分−1で注入し、各タンパク質注入(180秒)の終了時に、解離をモニタリングするために180秒間、HBS−EPバッファー(50μl 分−1)をチップに流した。タンパク質注入間で、10mM酢酸ナトリウム中2.0M NaCl、pH4.5、または10mMリン酸ナトリウム/カリウム、pH6.5を注入してチップ表面を再生した。データはBiaEvaluationソフトウエアバージョン4.1(Biacore AB/GE Healthcare)で処理した。各タンパク質注入で、対照フローチャネルに関して記録された非特異的SPR応答をサンプルフローチャネルに関して記録された応答から差し引いた。
細胞株: 天然にαKlothoを発現する正常ラット腎(NRK)細胞(ATCC、マナサス、VA、USA)、およびベクター単独、全長膜貫通マウスαKlotho、C末端FLAGタグを有するマウスαKlothoの細胞外ドメイン、または全長マウスαKlothoでトランスフェクトされたHEK293細胞[78]。細胞は37℃、95%空気、5%CO2雰囲気で培養し、10%ウシ胎児血清、ペニシリン(100U/ml)、およびストレプトマイシン(100mg/ml)を添加した高グルコース(450mg/dl)DMEM中で継代培養した。
HEK293細胞をベクターまたは示されているαKlothoプラスミドでトランスフェクトし、ポリリシンで前処理した12ウェルのカバーガラス上に播種し(1.8×105細胞/ml)、一晩増殖させた。細胞を洗浄し(4℃ PBS×3)、3%パラホルムアルデヒドで固定し(4℃×10分)、洗浄し(氷冷PBS×3)、1%BSA(PBS 44℃×10分)でブロッキングし、sb106−Fab(1%BSA、PBS中5ug/ml)とともにインキュベートし、洗浄し(PBS 4℃×5)、抗FLAG−Alexa488(Cell Signaling;1%BSAを含有するPBSで1:400希釈;1時間;20℃)とともにインキュベートし、洗浄し(PBS;4℃×4)、次いで、DAPI(Invitrogen)とともに1滴の退色防止剤を含むスライドガラス上に逆さに置き、暗所、室温で乾燥させた。24時間後、、これらのスライドを−20℃で保存した。画像はWaveFXスピニングディスク型共焦顕微鏡で取得した。
ELISAは、製造者(Immuno−Biological Laboratory、日本)が説明しているように行った。IP−IBアッセイについては、一般に、50μlの血清または尿をKRHバッファー[25 mM Hepes−NaOH(pH7.4)、120mM NaCl、5mM KCl、1.2mM MgSO4、1.3mM CaCl2、1.3mM KH2PO4]で希釈して終容量0.5mlとし、低結合シリコン処理チューブにて、2μgのsb106−Fabとともに一晩4℃でインキュベートした。KRHバッファーで3回予備洗浄した、抗FLAG抗体(50%v/v)をコンジュゲートしたセファロースビーズ(50μL)を加え、インキュベートし、(4℃×2時間)、洗浄した(KRH−チューブ当たり500μl 3回;22℃)。免疫複合体を2×SDSサンプルローディングバッファー(50μl;100℃×3分;4℃×3分;回転)で溶出させ、SDS−PAGEで分画し、ニトロセルロース膜に転写し、抗KL1抗体(KM2076、1:4000または3.1mg/mL、1:10000希釈)および希釈剤(Dako#S3022、カーピンテリア、CA、USA)で一晩(4℃、揺動器)でブロットした。膜を洗浄し(3回、0.1%Tweenを含むTris緩衝生理食塩水;TBS−T)、抗ラットIgG2A(LSBio cat#LS−C59051、5%乳汁/2% ヤギ血清/TBS−T中1:20000×1時間)に曝し、洗浄した(×3 TBS−T)。化学発光については、膜をSuperSignal West Femto Maximum Sensitivity基質(Thermo Scientific、ロックフォード、IL、USA)で覆い、30〜90秒間露光した。130kDのバンドをスキャンし、Adobe Photoshop CS4を用い、既知量のKlothoの内部対照サンプルと比較した。
線維芽細胞増殖因子受容体(FGFR)1cのリガンド結合ドメインとの組換えαKlothoエクトドメイン複合体に対するファージディスプレー合成Fabライブラリーの複数回のバイオパンニングの後、いくつかの結合ファージを同定した。クローンsb106(図1A)をさらなる特性評価のために選択した。ファージELISA(図1B)において、sb106−ファージはヒトおよびマウス両方のαKlothoと結合し、このことは種交差反応性を示し、また、αKlotho単独またはFGFR1cとの複合体いずれかと結合し、このことはそのエピトープが補助受容体複合体の形成によって隠されていないことを示す。sb106−ファージは、FGFR1c単独、ニュートラアビジン(NAV)またはウシ血清アルブミン(BSA)とは結合しなかった。sb106は、ヒトαKlothoと一桁ナノモルの範囲の親和性で結合する(IC50=1.7nM、図1C)。sb106−Fabはまた、バイオセンサーチップ上に固定化されている二元αKlotho−FGFR1c複合体とも高い親和性で結合し(図7A)、それはFGF23、αKlothoおよびFGFR1cの間の三元複合体の形成に干渉しない(図7B)。
sb106のユニークなCDR配列(図1A)を用い、Fabおよび全長IgGタンパク質の両方を生産した(例えば、Fabは細菌細胞で、IgGタンパク質は293F細胞で生産した)。sb106は、天然条件下でαKlothoに対して反応性が高かった。変性条件下でのマウス、ラット、およびヒト腎組織に対するイムノブロットシグナルは弱かったが、αKlothoを過剰発現するトランスジェニックマウス由来のサンプル[79]では、sb106−Fabは、αKlothoの全長細胞外ドメインに相当するバンドを検出した(図2A)。培養細胞では、sb106−Fabは、天然αKlothoを発現するNRK細胞を用いた変性条件下でのイムノブロットではαKlothoを検出できなかったが、αKlothoでトランスフェクトされたHEK293細胞からの細胞溶解液では過剰発現抗原を検出できた(図2B)。新鮮冷凍非固定ラット副甲状腺組織(およびαKlothoを発現することが知られる他の組織Iを用いた免疫組織化学では、sb106−IgGはαKlothoを検出したが、同じす組織が固定された場合には陰性であり(図2C)、このことはsb106が天然の折り畳みのFGFR1−αKlotho複合体と結合することを示唆する(図1B)。新鮮固定細胞を用いた免疫細胞化学染色では、αKlothoを異種過剰発現するHEK293細胞で明白な染色が得られたが、βKlothoを過剰発現する細胞では見られなかった(図2D)。細胞をポリリシンで処理した12ウェルカバーガラス上に1.8×105/mlで播種し、一晩増殖させた。細胞を氷冷PBSで3回洗浄し、氷上で10分間固定し(3%パラホルムアルデヒド)、冷PBSで3回洗浄し、10分間ブロッキングした(冷PBS中1%BSA)。次に、細胞をsb106−Fab(PBS中1%BSA中5μg/ml)とともに1時間インキュベートした。細胞を冷PBSで5回洗浄し(各2分)、次いで、抗FLAG−Alexa488(Fab軽鎖のC末端はFlagエピトープタグを含む)とともに光から保護しながら1時間インキュベートした(PBS中1%BSA中1:400)。細胞を冷PBSで4回洗浄した(各5分)。次に、カバーガラスを、DAPIとともに1滴の退色防止剤を含むスライドガラス上に逆さに置いた。画像はスピニングディスク型共焦顕微鏡で取得した。αKlothoを過剰発現する細胞であっても、固定の延長はsb106による染色を著しく低下または無効にした。これらのデータは、sb106が天然の折り畳みのヒト、ラット、およびマウスαKlothoと特異的に反応し、変性したαKlothoタンパク質とは反応しないことを示す。
sb106−Fabの可溶性αKlothoを沈降させる能力を、一連の免疫沈降−イムノブロット(IP−IB)アッセイを用いて試験した。sb106−Fabは、全細胞溶解液および馴化細胞培養培地およびαKlotho過剰発現細胞からのαKlothoを捕捉した(図3A)。捕捉されたsb106−Fabを、HEK293細胞において、C末端FLAGタグを有する可溶性αKlothoを用い、抗FLAG抗体の場合と比較した。sb106−Fabおよび抗FLAGは、正確に同じ電気泳動移動度のタンパク質を沈降させた。
IP−IB法がCKD患者の単一施設データベースから血清αKlothoレベルを相対的に決定できるかどうかを判定するために、この方法を試験した。アッセイの直線性ならびに延長されるy切片を調べるために、組換えヒトαKlothoを既知量で添加した。IP−IBは、一定範囲の異なる濃度の組換えαKlothoを添加した正常健常ボランティアおよび病期5CKDの患者からの血清を用いて行った(図4A)。漸増接種される外因性αKlothoとともにシグナルの段階的増強が見られた。CKD患者由来の血清もまた外因性αKlothoの増加とともにシグナルの増強を示したが、任意の所与のαKlotho濃度では、シグナル強度は正常血清より低かった。
αKlothoに関して利用可能な市販のアッセイには、それらの間に一貫した相関がない[46、83]。ELISAに基づく健常ヒトおよびCKD患者における研究[58]からは矛盾した結果が得られている。正常およびCKDにおけるαKlothoの絶対レベルは0.4[47]〜2000pg/ml超[41]までの範囲であり、ほとんどの測定値は100代の半ばから後半である[48、50、55、58〜60、83]。このアッセイに基づけば、αKlothoレベルは糸球体濾過速度(GFR)の低下とともに低くなる[48、52、54、57〜60]、関連は無い[40、41、50、51、53]または上昇すらする[44、47]ことが記載されている。同様に、αKlothoレベルは、年齢とともに変化しない、低下することも報告されている[42、53、58、59]。このことはヒトαKlothoデータの解釈をほぼ不能とし、異なる施設から得られた収集データには価値がない。
付加的CDR配列を表2に示す。相同な突然変異が各アミノ酸位置に導入されたが、これは、各位置に関して、元のアミノ酸が保持されたか、またはそのアミノ酸に最も近い「ホモログ」(例えば、保存的アミノ酸変化)が導入され、新たなFabファージライブラリーが構築されたことを意味する。αKlotho−FGFR1c複合体を抗原として用い、新たなライブラリーを用いて選択を行った。抗原に結合したクローンを単離し、配列決定を行い、表2に示す。結合親和性はSb106と同等またはそれより良好であると思われる。
右心カテーテル法を受けた9名のヒト対象(49.066.2歳)を本試験に登録した。右心カテーテル留置中に、腎上部および腎下部大静脈血液サンプルを得、4℃で遠心分離後すぐに血清を分離し、その後の試験のために−80℃で保存した。血清αKlothoを、本明細書に記載の免疫沈降−イムノブロットアッセイにより決定した。簡単に述べれば、0.1mlの血清を合成抗αKlotho Fab(sb106)で免疫沈降させ、免疫複合体をLaemmliサンプルバッファーで溶出させ、KM2076抗体でイムノブロットを行った。130kDの移動度に基づくオートラジオグラム上の特異的シグナルを、ImageJプログラム(National Institutes of Health(NIH)、ベテスダ、メリーランド州)で定量した。
αKlotho低次形態(kl/kl)マウス、kl/klマウスおよびそれらの野生型(WT)同腹子をテキサス大学サウスウェスタン医療センターの動物研究センターで維持した。当時点で、総てのマウスが129 S1/SVlmJ(129 SV)バックグラウンドで、6〜8週間である。正常Sprague−Dawley(SD)ラット(体重220〜250g)はCharles River Laboratories(ウェルミントン、MA)から購入した。αKlothoクリアランス試験では、ラットに両側腎摘(無腎ラット)または腎臓の手動操作を伴う開腹(シャムラット)を施した。ラットまたはマウスに、用量0.1mg/kg体重の組換えマウスαKlothoタンパク質(rMKl)の標識全長細胞外ドメイン(R&D Systems、ミネアポリス、MN)を静脈内または腹膜内に1回注射した。セクレターゼが血中αKlothoを調節するかどうかを調べるため、塩酸ドキシクリン(Sigma−Aldrich、セントルイス、MO)、25mg/kg/日のα−セクレターゼ阻害剤、および/または2.5mg/kg/日のβ−セクレターゼ阻害剤III(Calbiochem、ビレリカ、MA)を正常WTマウスの腹膜内に、2日間毎日注射し、48時間の時点で血液および腎臓を採取し、血清および腎のαKlothoを決定した。
ラットモノクローナル抗ヒトKlotho抗体KM20761,2を免疫ブロット法および免疫電子顕微鏡に使用し、合成抗αKlotho抗体sb10663を血清Klothoの免疫沈降に用いた。
正常Munich Wistarラット(BW220〜250g)をイナクチン(100mg/kg BW)で麻酔し、標識αKlothoボーラスを頚静脈から注射した(0.1mg/kg BW)。125I標識αKlothoまたは125I標識アルブミンの注射試験では、ボーマン嚢のフリーフロー微小穿刺および近位尿細管による体液回収を、公開されている方法を用いて行った。簡単に述べれば、左腎を露出させ、左尿管に尿回収のためのカテーテル留置を行った。リサミングリーン色素注射後にそれらの特徴的な輪郭により近位尿細管を特定し、ガラスキャピラリーで穿刺した。体液の容量は較正済みの定孔径ガラスキャピラリーで測定した。体液の放射活性は、シンチレーション計数で決定さし、体液容量に対して正規化した。明示された時点で、眼窩後静脈洞から血液を採取し、スポット尿を採取した。採取した尿および血清中の125I標識αKlothoまたは125I標識アルブミンをシンチレーション計数により定量した。種々の臓器のホモジネートを作製し、臓器ホモジネート中の放射活性をシンチレーション計数により測定し、臓器ホモジネート中のタンパク質に対して正規化した。臓器切片(10mm)に対してオートラジオグラフィーを行った。
マウス組換えKlothoタンパク質(0.1mg/kg BW)をkl/klマウスの腹膜内に注射し、注射24時間後にマウスを犠牲にした。腎臓を採取し、大動脈灌流により2.5%パラホルムアルデヒドで固定し、取り出し、4%パラホルムアルデヒド中で後固定した(4℃ 4時間)。超薄冷凍組織切片の免疫金標識を記載ように行った。21の腎皮質に2.3Mスクロースを一晩潤させ、液体窒素中で冷凍し、70〜80nm厚の切片を作製し(Ultramicrotome Reichert Ultracut E;Leica Microsystems、ウェッツラー、ドイツ)、ホルムバールコーティングニッケルグリッドにマウントした。これらの切片をKM2076抗体とともにインキュベートした後、金コンジュゲートプロテインA(10nm金粒子、Sigma−Aldrich)とともに60分間インキュベートした。酢酸ウラニル染色の後、切片をJeol 1200 EX透過型電子顕微鏡(Jeol Ltd.、昭島、日本)で可視化した。
循環αKlothoの生産および運用における腎臓の役割を調べた。直接穿刺による正常ラットの腎上部および腎下部大静脈中の、および右心カテーテル法を受けたヒト対象のαKlothoタンパク質の血清レベル。総ての患者がeGFR≧60ml/分/1.73m2を示した。ラットおよびヒト血清サンプルの両方で、同等の静脈洞αKlothoレベルの腎下部/腎上部増分が見られた。血清αKlothoレベルを周知の腎由来ホルモンである血清エリスロポエチンに対してプロットし、血清エリスロポエチンが上昇すると、および腎下部/腎上部下大静脈からの血清クレアチニン(SCr)が低下し、一方。αKlothoは層化することが見出され、このことは臓が循環中にαKlothoを分泌することを示す。
αKlothoに対する元の抗体試薬sb106(クローン48)、ならびにsb106に由来するCDR変異体は総て、単一の共通エピトープに結合する。検出アッセイを確立するためには、1つの表面/エピトープはタンパク質を単離または捕捉するために使用されるが、検出試薬のためには第2の全く別のエピトープが必要とされるので、異なるエピトープを有する抗体が必要である。よって、sb106とは異なり、全く別のエピトープと結合する抗体を同定するための別の選択戦略を行った。αKlothoに対するこれらの抗体は、ヒトαKlothoの細胞外ドメイン(ECD)に対して、飽和レベルのsb106 Fab(50ug/ml)の存在下で行われる抗体ファージディスプレー選択により同定した。R&D Systems(5334−KL)から購入したタンパク質はアミノ酸E34〜S981をカバーし、C末端に結合された6−Hisタグを有する。
このさらなる抗体のセット内にαKlotho上の少なくとも2つの新たなエピトープが存在する。競合ELISA戦略を用い、エピトープ分類試験を行った。このセット内に多数のクローンが得られたので、予備試験で、sb106(48)が結合するものとは別の2つの全く別のエピトープが明らかになった。これらのエピトープの代表的なもの(4804および4819)を次に、セット内の各クローンの評価のためにsb106(48)とともに使用した(図11)。sb106(48)がエピトープAを表す場合、新規クローンのうち23がエピトープBに分類され、エピトープCに関しては4クローンが見出された(図11および表5に要約)。3つのクローン(4806、4810および4817)については、ファージによる問題のために決定はされなかった。クローン4828は、提示された3つのエピトープのいずれとも競合を示さなかった。
各抗体に関する親和性評価は、表面プラズモン共鳴(SPR)によって決定した。Fabを、抗H+L抗体を用いて固定化し、αKlothoの希釈系を注入した。結合曲線を図12に示し、結合親和性(KD)を表5に要約する。親和性は240pM〜8.7nMの範囲である。
抗体を、ヒト患者尿サンプルからFabとしてαKlothoを免疫沈降させるそれらの能力に関してアッセイした(図13)。正常ボランティアからの50ulのヒト尿サンプルを、4℃で一晩、1ug/mlのFab(sb173−203、sb106)を含む400ulのKRHバッファー内でインキュベートした。次に、50ulのM2抗FLAGセファロースビーズを加え、4℃で2時間インキュベートした(Fab軽鎖のC末端はFLAGエピトープタグを含む)。これらのビーズを500ulのKRHバッファーで3回洗浄し、結合したタンパク質を100mM DTTを含有する2×LDSサンプルローディングバッファーで溶出させた。イムノブロットは、市販の一次ラット抗Klotho抗体(KM2076)、次いで、可視化のための標準抗ラットIgG二次抗体で行った。Super Signal West Femto基質を化学発光基質として使用した。半数を超えるクローンがsb106と同じかより良好に挙動した。
新規エピトープ(Bでは4808、およびCでは4831)を呈する選択クローンを全長IgGにサブクローニングし、発現させ、精製し、さらにsb106(48)とともに特性評価を行った。全長IgGを1ug/mlで固定化し(捕捉)、1%BSAを含有するバッファーでブロッキングした後、捕捉バッファー(Tris緩衝生理食塩水 pH7.4+0.1%BSA+0.05%Tween20)中20ngのビオチン化αKlotho(aa34〜981)、20ngのビオチン化αKlotho(aa1〜549)のいずれかとともに、または捕捉バッファー単独(BSA)で、室温で1時間インキュベートした。PBS+0.05%Tween20で洗浄した後、捕捉されたビオチン化αKlothoを、HRP−ストレプトアビジン試薬を用いて検出した。比色定量HRP試薬は450nmでの吸光度の測定を可能とする。3つのIgGは総て、予想通り、溶液からαKlothoを捕捉したが(図14)、4831だけがαKlothoの末端切断型のECDを検出でき、sb106とのエピトープ認識の違いをさらに特徴付けた。
ヒトおよびマウスαKlothoに対する交差反応性を評価するために、ヒトおよびマウスαKlothoの両方を用いてFab ELISAを行った(図16)。Fab(5ug/ml)を、固定化したαKlothoヒト、マウス、または陰性対照としてのニュートラアビジン(NA)およびSBAとともにインキュベートした。結合してないFabを洗い流した後、結合したFabを、HRPコンジュゲート抗Flag抗体(Fabはそれらの軽鎖の末端にFalgタグを有する)を用いて検出した。比色定量HRP試薬は、450nmでの吸光度の測定を可能とする。
Fabタンパク質(クローン4804〜4824、4826〜4834)をELISAプレート(Maxisorb)に15ug/mlの濃度で、室温で1時間吸着させた後、PBS+0.5%BSAで、室温で1時間ブロッキングした。このプレートをPBS+0.05%Tween 20で洗浄した後、2ug/mlの以下のタンパク質:αKlotho(Fcダイマー)、aKlotho−FGFR1c複合体(Fc複合体)、Fc単独、またはPBSとともに室温で1時間インキュベートした。結合してない抗原をPBS+0.05%Tween 20の6回の洗浄洗い流した後、これらのウェルをヤギ抗マウス(mG2a、Jackson ImmunoResearch Laboratories)とともに室温で30分間インキュベートした。結合してない抗マウス抗体をPBS+0.05%Tween 20の6回の洗浄で洗い流した後、これらのウェルを抗ヤギHRP試薬とともに室温で30分間インキュベートした。結合してない抗ヤギHRP抗体をPBS+0.05%Tween 20の6回の洗浄で洗い流した後、比色定量HRP試薬(TMB基質および停止溶液)を用い、450nmで吸光度を測定した。
