4. 100μLの氷冷ISTDを7つの別々の1.5mL円錐バイアルに分注し、0、10、20、40、60、90および120分間の時点につきそれらを標識する。
6. ゼロ時点として標識された円錐バイアルに50μLのMAM溶液を加える。残りのMAM溶液をインキュベーター(37℃)に5分間留置し、ミクロソーム(8.8μL)を添加して反応を開始し、時間を記録する。
8. 100μLの上清を取り、Spin−X HPLCフィルターチューブ(Corning Incorporated、カタログ番号8169)に移し、13,000rpmで5分間遠心分離し、この溶液から5μLを取って、2mLガラスオートサンプラーバイアル(Microsolv、カタログ番号95025−WCV)中、495μLのLCMSグレードのメタノール(Fischer scientific、CAS番号67−56−1)で希釈する。

[00233] 細胞株の培養および手順:ヒト肝臓肝細胞癌(HEPG2)およびヒト胚性腎293(HEK293)細胞株を購入し(ATCC)、75cm2のフラスコ(CellStar)で培養した。細胞は、非必須アミノ酸(Hyclone、#SH30238.01)、10mM HEPES(Hyclone、#SH302237.01)、5×106単位のペニシリンおよびストレプトマイシン(Hyclone、#SV30010)ならびに10%の熱不活性化ウシ胎仔血清(Gibco、#10082147)を添加した、DMEM/高グルコース(Hyclone、#SH3024301)培地で培養された。0.05%トリプシン(Hyclone、#SH3023601)を用いて細胞を回収し、PBSで洗浄し、滅菌白色光学底384ウェルプレート(NUNC、#142762)に分注した。3時間後、小分子溶液を、100nLピンツール(V&P Scientific)を備えたTecan Freedom EVO液体ハンドリングシステムで移した。対照(E)−10−(ブロモトリフェニルホスホラニル)デシル4−(4−(tert−ブチル)フェニル)−4−オキソブタ−2−エノアート(DMSO中400μM、陽性対照)およびDMSO(陰性対照)。細胞を48時間インキュベートした後、CellTiter−Glo(商標)を添加し、発光ベースの細胞生存性アッセイ(Promega、Madison、WI)を行った。全ての発光読み取りは、Tecan Infinite M1000プレートリーダーで行った。このアッセイは、3回の独立した操作で、4連で実施した。データを対照に対して正規化し、非線形回帰(GraphPad Prism)によって分析した。
実施例26:オキサジアゾールの薬力学的評価
[00235] MP−III−080およびMP−III−085の抗不安様作用をガラス玉覆い隠しアッセイを用いてマウスで評価した。10mg/kgでは、オキサジアゾールMP−III−080は溶媒と比較してガラス玉覆い隠しを有意に減少させた。30mg/kgでは、MP−III−085およびMP−III−080はガラス玉覆い隠しを有意に減少させた。しかしながら、30mg/kgのMP−III−080で処置したマウスは、鎮静の兆候が弱かった。これらの化合物の抗不安対運動副作用の潜在性を評価するために、回転ロッド(ロータロッド)上のマウスを用いた運動感覚研究をガラス玉覆い隠しについて研究したのと同じマウスで行った。雄、NIHスイスウェブスターマウス(n=10)に、試験の30分前に溶媒または試験化合物(10または30mg/kg)のいずれかを腹腔内注射した。
