Abstract
Description
本願は、2010年1月7日に出願された米国仮特許出願第61/293,030号明細書、および2009年11月18日に出願された米国仮特許出願第61/262,373号明細書の35U.S.C.§119(e)下での優先権を順に主張する、2010年11月18日に出願された国際出願PCT/US10/57235号明細書の国内段階移行である、2012年5月18日に出願された米国特許出願第13/510,801号明細書の35U.S.C§120下での一部継続出願である、米国特許出願第15/075,008号明細書に対する優先権を主張する。前述の参考文献のすべてが、ここでそれら全体が参照により援用される。
本明細書で用いられるとき、用語「生体試料」は、対象、好ましくはヒト対象からの体液、細胞または組織を含む検体を指す。試料はまた、天然にまたは人工的方法(例えば外科的手段)により、前悪性病変の1つまたは複数の悪性細胞と接触状態になっている体液であり得る。
本明細書に記載の免疫複合体は、抗体を脂質、炭水化物、タンパク質、または他の分子と連結する公知の方法により調製され得る。例えば、本明細書に記載の結合分子は、免疫複合体を形成するため、本明細書に記載の担体(例えば、脂質、ポリマー、リポソーム、ミセル、またはナノ粒子)の1つ以上とコンジュゲートされ得、また免疫複合体は、共有結合、非共有結合、またはその他の何れかにより、治療薬または診断薬に組み込み得る。さらに、本明細書に記載の結合分子の何れかはさらに、本明細書に記載の1つ以上の治療薬または診断薬、または追加的な担体とコンジュゲートされ得る。一般に、1つの治療薬または診断薬が各結合分子に付着されてもよいが、2つ以上の治療薬または診断薬が同じ結合分子に付着され得る。さらに、治療薬は同じである必要はなく、異なる治療薬であり得る。
本発明は、マトリプターゼ抗体または免疫複合体またはマトリプターゼ抗体もしくは免疫複合体を含む組成物のマトリプターゼを発現する細胞を含む悪性腫瘍を治療するための一次組成物としての使用を包含する。悪性腫瘍は、限定はされないが、固形腫瘍、非ホジキンリンパ腫、ホジキンリンパ腫、多発性骨髄腫、B細胞悪性腫瘍および/またはT細胞悪性腫瘍を含む。固形腫瘍は、メラノーマ、がんおよび肉腫からなる群から選択され、がんは、腎がん、肺がん、腸管がん、胃がんおよびメラノーマからなる群から選択される。B細胞悪性腫瘍は、B細胞リンパ腫の無痛性形態、B細胞リンパ腫の侵襲性形態、慢性リンパ性白血病、急性リンパ性白血病、および多発性骨髄腫、B細胞障害および他の疾患からなる群から選択される。特に、本明細書に記載の組成物は、様々な自己免疫ならびにB細胞リンパ腫の無痛性形態、B細胞リンパ腫の侵襲性形態、慢性リンパ性白血病、急性リンパ性白血病、多発性骨髄腫、およびワルデンストレームマクログロブリン血症を治療するために特に有用である。該治療方法は、抗体または免疫複合体を含む組成物を治療有効量でかかる治療を必要とする哺乳動物に投与するステップを含む。
マトリプターゼに対する抗体(「抗マトリプターゼ抗体」)は、当該技術分野で公知であり、また例えば、Lin C.Y.,et al.,J.Biol.Chem.,1999,274(26):18237−18242に開示された方法により作製され得る。マトリプターゼに対する抗体は、潜在型マトリプターゼに特異的な抗体および活性化マトリプターゼに特異的な抗体、ならびに潜在型および活性化マトリプターゼの双方を認識する抗体を含む。
本開示の抗マトリプターゼ抗体、例えばM69は、治療薬または診断薬、例えば、DOX、アウリスタチン(限定はされないが、MMAE/MMAFを含む)に共有結合され、ひいては免疫複合体を形成してもよい。免疫複合体のリンカーに基づく技術の概要については、Jain N et al.Current ADC Linker Chemistry,Pharm Res.2015 Nov;32(11):3526−40(ここでその全体が参照により援用される)に見出される。かかる免疫複合体中の共有結合は、切断可能連結部分、例えばリソソーム内部のカテプシンBによって切断可能なVal−Citリンカーを含んでもよい。市販のVal−Citリンカーは、下記の実施例10で用いるため、修飾された。他の切断可能連結部分は、カテスピン(Cathespin)Bによってやはり切断可能なPhe−Lysリンカーを含んでもよい。最も簡単な切断可能連結部分の一部が、還元的(すなわち細胞内)環境下で切断可能なジスルフィド(S−S)橋を含む。しかし、Val−Citリンカーなどの切断可能連結部分は、例えば、無差別な切断を受け得るジスルフィド架橋よりも高い特異性をもたらし、ひいては優れた選択肢を提示するが、任意のかかる切断可能連結部分は、本発明の範囲内で検討されるべきである。本発明に適し得る切断可能連結部分の概要が、Leriche et al.,Cleavable linkers in chemical biology Bioorg Med Chem.2012 Jan 15;20(2):571−82(ここでその全体が参照により援用される)に提示される。あるいは、リンカーは、切断不能であってもよい。切断不能なリンカーは、切断可能なリンカーよりも多様であり、細胞内環境下で切断に抵抗性を示す任意の連結部分を含んでもよい。例えば、時として 興味深い具体的な切断不能なリンカーは、FDA承認免疫複合体トラスツズマブエムタンシン(商品名Kadcyla)中に見出されるSMCCリンカーを含み、M69−DOX免疫複合体を調製するため、下記の実施例2においても探索される。
MMAEを含むアウリスタチンは、非常に強力な細胞毒のクラスを表す。理論によって拘束されることを望まないが、MMAEのようなアウリスタチンが、適切な細胞分裂にとって重要である、チューブリンの重合を遮断することにより細胞分裂を阻害すると考えられる。したがって、MMAE、MMAF、およびアウリスタチンPEなどの化合物は、有糸分裂を阻害するための「抗有糸分裂剤」として分類される。かかる細胞毒の効力は顕著である。MMAEは、例えばDOXよりも100〜1000倍強力であるが、DOXは本発明における使用に適する。MMAEは、この強力な毒性にもかかわらず、単独での薬剤としての投与に適さない。しかし、細胞傷害性ペイロードは、マトリプターゼ抗体、例えばM69などの抗体にカップリングされるとき、特定の悪性細胞に対して特異的であることができる。
多発性骨髄腫およびB細胞リンパ腫細胞におけるマトリプターゼの発現
ヒト造血悪性腫瘍の24の細胞株パネルを、マトリプターゼおよびその内因性阻害剤HGFアクチベーターインヒビター1すなわちHAI−1に対するウエスタンブロッティングにより分析した(図1)。細胞可溶化物を、24の造血がん細胞(レーン2〜14)およびT−47D乳がん細胞(レーン1)から調製した。等量のタンパク質をSDS−PAGEにより分離した。マトリプターゼおよびHAI−1のレベルは、指定通りに抗マトリプターゼmAbおよび抗HAI−1 mAbを用いるイムノブロット分析により評価した(図1Aおよび1B)。
DOX−免疫複合体の調製
マトリプターゼに対するモノクローナル抗体(M24またはM69、CY Lin,University of Maryland,Baltimore,Md.から入手;Lin C.Y.,et al.,J.Biol.Chem.,1999,274(26):18237−18242;Chen,Y.W.,et al.J.Biol.Chem.,2010,285(41):31755−31762)を、幾つかのマイナーな変更を伴う、Protein−Protein Couplingキット(Invitrogen)を製造業者の使用説明書に従って用いることにより、DOX(Sigma)にコンジュゲートした。簡潔に述べると、DOXを、クロスリンカーSMCC(4−(N−マレイミドメチル)シクロヘキサン−1−カルボン酸スクシンイミジル)と1:3〜1:1.5(DOX:SMCC)の間のモル比(MR)で、常に硬化しながら室温で1時間反応させた。約200mgのM24 mAbをSPDP(3−(2−ピリジルジチオ)−プロピオン酸スクシンイミジル)と室温で1時間反応させ、その後、遠心フィルター装置(Ultrafree,Millipore)を用いてリン酸緩衝液を交換した。あるいは、mAbは、セファデックスG50を用いるゲル濾過により、遊離SPDPから分離することができる。次に、DOXマレイミド誘導体をチオール化M24またはM69 mAbに室温で3時間コンジュゲートし、その後、リン酸緩衝液で透析し、遊離DOXを除去した。セファデックスG50を用いるゲル濾過は、調製規模が1gより多いタンパク質である場合に非カップリングDOXを除去するためにも用いることができる。DOXに対するタンパク質のモル比は、蛍光およびタンパク質濃縮によりDOXを測定することによって判定する。
多発性骨髄腫細胞に対するM24−DOXの細胞傷害性
免疫複合体の細胞傷害性効果を評価するため、MM細胞を様々な濃度のコンジュゲートされたDOXで48時間処理した。免疫複合体は、細胞増殖を用量依存的様式で阻害した。免疫複合体のEC50は、5μg/ml(タンパク質)であった。この細胞傷害性効果は、200nMの遊離DOXでの処理の場合でも認められた。5μg/mlのM24−DOXの濃度が蛍光強度に基づくと250nMの遊離DOXに等しいことから、それ故、コンジュゲートされたDOXの効力は、遊離薬剤と類似する。細胞の非修飾抗体への曝露は、細胞増殖に対する効果を有せず、M24−DOXに誘導される細胞傷害性効果がDOX活性を通じて媒介されることを示した。OPM−2およびRPMI−8226を含む2つの他のMM細胞株もまた、24ウェルプレート内のMM細胞(1×105/ウェル)がM24−DOXの存在下または不在下で異なる濃度で48時間治療された場合の図2で示される通り、M24−DOXの細胞傷害性活性について試験した。細胞数は、Vi−Cellカウンター(Beckman)によって計数した。
M24−DOXの核局在化
DOXの免疫複合体のMM.1S細胞への内部移行を実証するため、蛍光顕微鏡を用いて、様々な期間での細胞の薬剤への曝露後のM24−DOXの細胞内局在化を追跡した。様々な期間でのMM.1S細胞のM24−DOXへの曝露後、細胞を固定し、染色した。免疫複合体を蛍光顕微鏡により可視化した。DAPIを核染色に用いた。
骨髄由来間葉系幹細胞(MSC)のM24−DOXに対する耐容性
マトリプターゼの発現が欠如するヒト骨髄由来MSCを、骨髄由来間葉系間質細胞(5×104/ウェル、24ウェルプレート)が異なる濃度のM24−DOXまたは遊離DOXで4日間処理された場合の図4で示される通り、M24−DOXの細胞傷害性選択性を判定するための対照として用いた。細胞数は、Vi−Cellカウンターによって測定した。
活性マトリプターゼの標的化
DOXはまた、酵素の活性のある2本の鎖に特異的に結合する別のマトリプターゼmAb(M69)とコンジュゲートした。活性マトリプターゼレベルが大部分の乳腺腫瘍に見出された一方、プロテアーゼの活性型は乳腺の正常組織内でかろうじて検出可能である(図5)。これらのデータは、M69−DOXと称されるM69とのDOXコンジュゲートが、正常組織への毒性と関連して、活性化マトリプターゼを発現する腫瘍を標的化する中でさらなる選択性をもたらす。上皮組織がマトリプターゼを発現するとき、M24−DOXの健常組織への毒性が懸念される。例えば、がん細胞内のルイスY抗原を標的化する、BR96と称される以前のDOXコンジュゲートは、かなりの胃腸(GI)毒性が原因で第II相試験において失活した(Tolcher,A.W.,et al.,J.Clin.Oncol.,1999,17:478−484)。上皮組織内でのマトリプターゼの潜在型の発現と対照的に、このプロテアーゼの活性型のレベルは、免疫組織化学によりほぼ検出不能である。したがって、M69−DOXは、上皮に対する毒性はあまりない。
サリドマイドによるM69−DOXの増強された細胞傷害性効果
サリドマイドは、多発性骨髄腫治療において治療的であり、サリドマイドおよびデキサメタゾンの組み合わせは、多発性骨髄腫を有する患者を治療するための最も一般的な治療計画の1つであり、応答速度は最大60〜70%である(Denz,U.,Eur.J.Cancer.,2006,42:1591−1600;Gieseler,F.,et al.,Thromb.Haemost.,2008,99:1001−1007)。サリドマイドなどの現行のMM治療薬と組み合わせたDOXコンジュゲート双方の有効性を試験した。M69−DOX単独は、M24−DOXほどに大して強力でなかった。意外にも、M69−DOX治療による細胞増殖の阻害がサリドマイドにより有意に増強された一方で、M24−DOXの阻害活性は改変されなかった(図6A〜B)。サリドマイド単独がMM細胞増殖に対する効果を有しなかったことから、サリドマイドと組み合わされたM69−DOXによるこの増強された阻害は、相乗的であるように思われた(図6A)。したがって、これらの試験では、サリドマイドが増強された増殖の阻害をもたらしたことが示される。
MM細胞内でサリドマイドによって誘導されるマトリプターゼ活性化
サリドマイドおよびM69−DOXコンジュゲートとともに生じた細胞傷害性における増加を試験するため、活性化マトリプターゼの発現に対するサリドマイドの効果を測定した。MM細胞内でのマトリプターゼの活性化は、50μMのサリドマイドでの24時間治療後、著しく増強された(図8)。サリドマイドは、少なくとも部分的にはVGEF産生の阻害を通じて、MMと骨髄間質細胞との間の相互作用に干渉することにより血管新生抑制活性を発揮することが知られている。サリドマイドは、MM細胞の細胞増殖に対する効果をほとんど有しないように思われる一方(図7A〜B)、マトリプターゼを活性化するその能力は重要な新規の知見であり、これは、M69−DOXコンジュゲートにより活性マトリプターゼ陽性骨髄腫細胞を標的化することによる、MM細胞のサリドマイド治療に対する応答を利用するための新規な手法を示唆する。
活性マトリプターゼをインビボで標的化するM69−DOXによる乳腺腫瘍増殖の阻害
M69−DOXがインビボで腫瘍増殖を抑制する有効性を評価するため、ヒト乳房異種移植腫瘍モデルを用いた。乳腺腫瘍(MDA−MB468)を有するNOD/重症複合免疫不全マウスを、20または10mg/kgの何れかのM69−DOXで週2回治療するとともに、2mg/kgのDOXで治療した。未治療対照群におけるマウスの腫瘍サイズは3週目に著しく増加し、何れかの用量のM69−DOXで治療し、DOXで治療したマウスにおける腫瘍増殖は、全治療経過の間に有意に消失した(図8)。単一のmAbがDOXの7つの分子と付着し、M69−DOXの10mg/kgでの用量は、投与したDOXの0.035mg/kgに等しい。これらの結果は、乳腺腫瘍をインビボで治療するために活性化マトリプターゼを標的化するDOXコンジュゲートの強力な効力および有効性を実証する。
M69−DOXまたはDOXの何れかで治療したマウスの平均重量は、未治療対照群からの場合と比べて減少が顕著でなく(15%未満の減少)、これはかかる用量でのM69−DOXが十分な耐容性を示したことを示す(図10)。M69−DOXの20mg/kgでの用量は注目すべき毒性をもたらさず、これは薬剤用量が最大耐容用量に達するように漸増され得ることを示す。
コンジュゲート:M69−MMAEを形成するためのMMAEのM69 mAbへの放出可能リンカーを介したカップリング
本明細書に記載の通り、免疫複合体を形成するため、MMAEをM69にカップリングさせた。放出可能リンカー技術は、Bioconjugate Chem.2002,13:855−869;Blood 2003,102:1458−1465;Nature Biotech.,2003,21:778−784(これらのすべてはここでそれら全体が参照により援用される)に記載の、オリジナルなカテプシンB切断可能リンカーであるSeattle Geneticsのバリン−シトルリン−PABAリンカーに基づいた。バリン−シトルリン−p−アミノベンジルアルコールカルバメートリンカーは、(i)2段階のペグ化、(ii)銅を含まないクリックケミストリーを介したコンジュゲーション、および(iii)システインスルフヒドリル−マレイミドカップリングに代わるmAbのアシル化リジン側鎖により修飾した。ADCアセンブリの全体設計を図18に示す。アジド−PEG4−Val−Cit−PABA−MMAE構築物の化学合成は、図19に示す通りであった。本設計により、このリンカープラットフォームが、DBCOおよびN3を用いる銅を含まないクリックケミストリーを用いて改善され、重要な薬剤負荷段階を非常に穏やかな条件下、化学量論的に制御された様式で実施することができた。システイン残基の一時的に還元された水硫基を用いる代わりに、リジン側鎖を用いて、リンカー−薬剤リガンドを抗体の表面上にコンジュゲートした(図11)。この手法がシステイン間のジスルフィド橋に影響しないことから、抗マトリプターゼ抗体は、コンジュゲーション手順の間に構造的にインタクトな状態を維持し、ミスフォールド/解離した抗体鎖による活性低下を伴う課題を排除する。さらに、リジン側鎖は、mAbの表面上で容易に利用可能であることから、Cys−Cysジスルフィド橋を還元する必要がなく、mAbの構造的不安定化およびマレイミドコンジュゲーション後の酸化時の不適正なジスルフィド結合形成に関する課題を排除した。適切な分析方法(HR−MALDI−TOF質量分析)により、免疫複合体の直近のバッチが工業標準を満たしていることが示された(図11)。極めて疎水性の薬剤分子であっても使用を促進するクリックケミストリーの両前駆物質上のコンジュゲートの薬剤−リンカー部分においてPEGスペーサーを用いることにより、さらなる進歩が達成された。PEG鎖は、mAbと薬剤とを分離しておく程に十分に長いことで、非特異的な薬剤および抗体相互作用の確率を低減し、またリンカーのPEG5−PEG4立体配置が、銅を含まないクリックケミストリーによりもたらされる四環系の疎水性を補償した。選択した細胞傷害性化合物は、モノメチルアウリスタチンE、すなわちMMAEであった。PEG化リンカーの可動性および長さは、MMAEを切断および放出するため、カテプシンBに高度な接近性をもたらす。コンジュゲーション反応は、MALDI−TOF質量分析により監視し、各mAb分子への3.5の薬剤の連結に対応する7000Daの平均分子量の増加を示した。
M69−MMAE免疫複合体のインビトロ細胞傷害性
実施例10に記載のようなM69−MMAE免疫複合体のインビトロ細胞傷害性を評価するため、TNBC(MDA−MB−468、MDA−MB−231およびBT549)、前立腺(DU145およびPC3)、膵臓(PANC1およびMiaPacal)、NSCL(H322およびH1299)、卵巣(OVCAR5)、胃(AGS)、およびマントル細胞リンパ腫(Mino、Jeko、MaverおよびZ138)がんなどの腫瘍細胞を、指定通りの様々な濃度でのM69−MMAE免疫複合体に曝露した。免疫複合体への持続的曝露の72時間後、以下のようなMTS(Promega)アッセイにより細胞傷害性を評価した。つまり、MTS比色分析(Promega製CellTiter 96)を製造業者のプロトコルに従って実施した。がん細胞を96ウェルプレートに8000細胞/ウェルで蒔き、段階的滴定でM69−MMAEに37℃で72時間曝露した。未治療対照ウェルに関して、パーセント生存度を免疫複合体濃度に対してプロットした。各試験の結果は、5通りの測定の平均である。これらの細胞に対するM69−MMAEのIC50の値を下の表2にまとめた。指定腫瘍細胞に対するM69−MMAEのIC50は、上記のように96ウェルプレートを用いるMTS比色分析法により測定した。IC50値は、ADCが3.5MMAE/mAbの化学量論比を有することを考慮して、ADCの平均(μg/mL)±標準偏差およびMMAE濃度に相当するnMとして表した。
M69−MMAE免疫複合体を含むインビボ異種移植試験
実施例10に記載のようなM69−MMAE免疫複合体を用いてパイロット異種移植試験を実施した。マウスコンジュゲートを10mg/kgで週3回腹腔内投与したヌードマウスにおける毒性試験(N=6)を、キメラM69−MMAEコンジュゲートを試験する前に実施した。体重減少または毒性の徴候がなく、これは構築物が安定であり、正常組織を標的にしなかったことを示す。この情報に基づき、ヒトトリプルネガティブ乳腺腫瘍MDA−MB−468を有するマウスを、M69−MMAE免疫複合体で治療した。図13は、重要なことにコンジュゲートがこのトリプルネガティブ乳がんに対して強力な抗がん活性を有する一方、動物において体重減少または毒性の他の証拠が認められなかったことを示し、これは遊離薬剤がコンジュゲートから循環中に有意に放出されなかったことを示す。これがマウスにおけるマウス抗体であることから、マトリプターゼに対する抗体応答であれば想定されないことになる。図14は、対照マウスおよびM69−MMAE免疫複合体で治療されたマウスにおける中央値腫瘍体積(mm3単位)の結果を示し、腫瘍サイズにおける有意な増加が、M69−MMAEで治療されたマウスと比べて対照群において認められた。
前立腺がん細胞は酸性細胞外pH下でM69−MMAEに対する感受性がより高い
マトリプターゼの活性化がアシドーシスにより誘導される。したがって、活性化マトリプターゼを発現するがん細胞は、抗原レベルの増加に起因する酸性条件下で、実施例10に記載のようなM69−MMAE免疫複合体の細胞傷害性効果に対する感受性がより高い。これは、DU145およびPC3前立腺がん細胞双方がpH6.5のHCl酸性培地下で免疫複合体に対する感受性が高くなるという実験により証明された。IC50は、DU145およびPC3細胞において各々、8から1μg/ml、および6.8から2μg/mL未満に有意に低下した(図16)。培地の炭酸緩衝化によりCO2インキュベーター内で1時間後にpHが漸増したことから、酸性条件への曝露は約30分間だけ維持した。
Taxatere抵抗性前立腺がん細胞(PC3R)はM69−MMAEに対する感受性を示す
化学療法抵抗性がん細胞に対する実施例10に記載のようなM69−MMAE免疫複合体のインビトロ有効性を判定するため、taxtere抵抗性細胞をコンジュゲートで治療した。PC3R細胞は、有糸分裂阻害に対する抵抗性に反して、親PC細胞のMMAEコンジュゲートに対する感受性と同様であった(図17)。これは、M69−MMAEが化学療法抵抗性前立腺がんを有する患者にとっていかに有効な治療であるかを強調する。
Claims (25)
- 抗マトリプターゼ抗体またはその抗原結合断片、および細胞毒を含む、マトリプターゼを発現するがん細胞を選択的に標的化する免疫複合体において、前記細胞毒がモノメチルアウリスタチンE(MMAE)、モノメチルアウリスタチンF(MMAF)、およびアウリスタチンPEからなる群から選択されることを特徴とする免疫複合体。
- 請求項1に記載の免疫複合体において、前記抗マトリプターゼ抗体がM69モノクローナル抗体またはその抗原結合部分を含むことを特徴とする免疫複合体。
- 請求項1乃至2の何れか一項に記載の免疫複合体において、リンカーをさらに含むことを特徴とする免疫複合体。
- 請求項3に記載の免疫複合体において、前記リンカーが切断可能連結部分を含むことを特徴とする免疫複合体。
- 請求項4に記載の免疫複合体において、前記切断可能連結部分が、カプテシン(Capthesin)Bによって切断可能なVal−Cit連結部分またはPhy−Lys連結部分を含むことを特徴とする免疫複合体。
- 請求項3乃至5の何れか一項に記載の免疫複合体において、前記リンカーが前記抗マトリプターゼ抗体上のリジン残基に共有結合されることを特徴とする免疫複合体。
- 請求項3乃至6の何れか一項に記載の免疫複合体において、前記リンカーがPEGを有することを特徴とする免疫複合体。
- 請求項1乃至2の何れか一項に記載の免疫複合体において、リンカーをさらに含み、かつ前記リンカーが第1の連結成分および第2の連結成分を含むことを特徴とする免疫複合体。
- 請求項8に記載の免疫複合体において、前記第1の連結成分が前記抗マトリプターゼ抗体上のリジン残基に共有結合されることを特徴とする免疫複合体。
- 請求項8乃至9の何れか一項に記載の免疫複合体において、前記第1の連結成分が前記第2の連結成分にトリアゾール部分を介して結合されることを特徴とする免疫複合体。
- 請求項8乃至10の何れか一項に記載の免疫複合体において、前記第2の連結成分が切断可能連結部分を含むことを特徴とする免疫複合体。
- 請求項11に記載の免疫複合体において、前記切断可能連結部分が、カプテシンBによって切断可能なVal−Cit連結部分またはPhy−Lys連結部分を含むことを特徴とする免疫複合体。
- 請求項8乃至12の何れか一項に記載の免疫複合体において、前記第1の連結成分がPEGを有することを特徴とする免疫複合体。
- 請求項8乃至13の何れか一項に記載の免疫複合体において、前記第2の連結成分がPEGを有することを特徴とする免疫複合体。
- 請求項8乃至14の何れか一項に記載の免疫複合体において、前記細胞毒が前記第2の連結成分に結合されることを特徴とする免疫複合体。
- マトリプターゼを発現する細胞を含む悪性腫瘍を治療する方法において、請求項1乃至15の何れか一項に記載の免疫複合体を含む組成物を治療有効量でかかる治療を必要とする対象に投与するステップを含むことを特徴とする方法。
- 請求項16に記載の方法において、前記悪性腫瘍が血液学的悪性腫瘍を含むことを特徴とする方法。
- 請求項17に記載の方法において、前記血液学的悪性腫瘍が、急性リンパ性白血病(ALL)、急性骨髄性白血病(AML)、慢性リンパ性白血病(CLL)、小リンパ球性リンパ腫(SLL)、慢性骨髄性白血病(CML)、急性単球性白血病(AMOL))、ホジキンリンパ腫、非ホジキンリンパ腫、バーキットリンパ腫(BL)、びまん性大細胞型B細胞リンパ腫(DLBL)、マントル細胞リンパ腫(MCL)、および多発性骨髄腫(MM)からなる群から選択されることを特徴とする方法。
- 請求項16に記載の方法において、前記悪性腫瘍が上皮悪性腫瘍を含むことを特徴とする方法。
- 請求項19に記載の方法において、前記上皮悪性腫瘍が、前立腺腫瘍、乳腺腫瘍、脳腫瘍、腎腫瘍、肺腫瘍、結腸腫瘍、膀胱腫瘍、皮膚腫瘍、甲状腺腫瘍、卵巣腫瘍、および中皮腫からなる群から選択されることを特徴とする方法。
- 請求項16乃至20の何れか一項に記載の方法において、マトリプターゼを活性化する免疫調節剤を治療有効量で同時投与するステップをさらに含むことを特徴とする方法。
- 請求項21に記載の方法において、前記免疫調節剤がサリドマイドまたはサリドマイド類似体を含むことを特徴とする方法。
- 細胞毒にリンカーを介して共有結合される、抗マトリプターゼ抗体またはその抗原結合部分を含む免疫複合体において、前記細胞毒が、モノメチルアウリスタチンE(MMAE)、モノメチルアウリスタチンF(MMAF)、およびアウリスタチンPEからなる群から選択され、かつ前記リンカーが、
前記抗マトリプターゼ抗体またはその抗原結合部分の上に位置するリジン残基を介して、前記抗マトリプターゼ抗体またはその抗原結合部分に結合された第1の連結成分;および
切断可能連結部分を含む第2の連結成分において、トリアゾール部分を介して前記第1の連結成分に、かつ前記切断可能連結部分を介して前記細胞毒に結合されている、第2の連結成分を含み、
ここで前記切断可能連結部分は、カプテシンBによって切断可能なVal−Cit連結部分またはPhy−Lys連結部分を含み、
ここで前記第1の連結成分は、前記抗マトリプターゼ抗体またはその抗原結合部分の上の前記リジン残基と前記トリアゾール部分との間の前記アミド結合間に約3〜約7の間のポリ(エチレングリコール)(PEG)単位を含み、かつ
ここで前記第2の連結成分は、前記トリアゾール部分と前記切断可能連結部分との間に約3〜約7の間のポリ(エチレングリコール)(PEG)単位を含む、ことを特徴とする免疫複合体。 - 請求項23に記載の免疫複合体において、前記抗マトリプターゼ抗体またはその抗原結合部分がM69またはその抗原結合部分を含むことを特徴とする免疫複合体。
- 請求項24に記載の免疫複合体において、前記細胞毒がモノメチルアウリスタチンE(MMAE)を含むことを特徴とする免疫複合体。
