b)マイクロニードルを生物学的障壁内に挿入することと、
c)薬剤を、マイクロニードルを通して生物学的障壁内に送達することであって、送達される薬剤の量または体積が、シグナル増幅器システムから受容されるシグナルによって決定される、送達することとを含む、方法もまた本明細書に開示される。
本出願全体を通して使用される用語は、当業者にとって通常の典型的な意味で解釈されるべきである。しかしながら、本出願人は、以下の用語に、以下に定義される特定の定義が与えられることを要求する。
埋め込みなしでの血糖値(BGL)のグルコース応答制御のための、膵臓β細胞からのインスリン分泌を調節するための無痛マイクロニードル(MN)パッチプラットフォームが本明細書に開示される。図1Aおよび図1Bに示されるように、この戦略は、生(細胞に基づく)及び合成グルコース応答システム(L−S GRS)の両方を統合して、外部に位置付けられたβ細胞カプセルがグルコースシグナルを感知し、低侵襲的な様式でMNを通してインスリンを分泌することを可能にする。初期設計では、細胞カプセルを、架橋ヒアルロン酸(HA)から作製されたMNパッチと統合した(図1A)。高血糖状態下では、グルコースがMNを通して拡散し、アルギン酸マイクロゲル中に被包されたβ細胞と相互作用して、インスリン分泌を促進し得ることが予想された。しかしながら、グルコースの限定された拡散のために、パッチは高血糖状態に対して効果的に応答せず、インスリン分泌のわずかな増加が検出された。細胞応答を効果的に引き起こすために、本明細書に報告されるMNマトリックスは特に、合成「グルコースシグナル増幅器(GSA)」を含有する(図1B)。この革新的なGSAは、3つの酵素、つまりグルコースオキシダーゼ(GOx)、α−アミラーゼ(AM)、及びグルコアミラーゼ(GA)を捕捉する自己組織化重合体ナノサイズ小胞を備える。GOxは、酸素の存在下でグルコースをグルコン酸へと変換する。AMは、α−アミロースを二糖類及び三糖類へと加水分解し、これは、GAによってグルコースへと更に変換される(N.Gurung et al.Process Biochemistry 2003,38,1599、L.Kandra,Journal of Molecular Structure:THEOCHEM 2003,666−667,487)。
材料
言及されない限り、全ての化学物質は、Sigma−Aldrichから購入した。ヒアルロン酸ナトリウム(HA、300kDaの分子量)はFreda Biochem Co.,Ltd.(Shandong,China)から購入し、カスタム合成ペプチド(CGRGDS、MW594.31)はCeltek Bioscience,LLC.(Franklin,TN)から購入し、抗インスリン抗体(ab181547)及びヤギ抗ウサギIgG H&L(FITC)(ab6717)はAbcamから購入し、皮膚貼付外科接着剤はMedline Industries,Inc.から購入した。
アミド形成を通して6−(2−ニトロイミダゾール)ヘキシルアミンを化学的に共役させることによって、低酸素感受性ヒアルロン酸(HS−HA)を合成した。最初に、6−(2−ニトロイミダゾール)ヘキシルアミンを合成して、HAのカルボン酸基と反応させた。簡潔には、NI(0.15g、1.3mmol)をDMF中に溶解させ、これにK2CO3(0.28g、2.0mmol)を添加した。その後、6−(Boc−アミノ)ヘキシルブロミド(0.39g、1.4mmol)を溶液に滴加し、80℃で4時間撹拌した。濾過を通して固体不純物を反応混合物から分離し、メタノールで洗浄した。蒸発によって固体生成物を残渣溶媒から得、これを脱イオン(DI)水中に懸濁させ、その後、酢酸エチルで抽出した。有機層を回収し、硫酸ナトリウムで濃縮した。生成物を氷上でメタノール中に再溶解させた。メタノール中5mLの1.25M HClを溶液に添加し、室温(RT)で24時間撹拌した。その後、回転蒸発器を使用して溶媒を除去して、アミン官能化NIを得た。第2に、1−エチル−3−(3−ジメチルアミノプロピル)カルボジイミド(EDC)及びN−ヒドロキシスクシンイミド(NHS)の存在下で、6−(2−ニトロイミダゾール)ヘキシルアミンをHAに共役させた。簡潔には、0.24gのHA(分子量:約300kDa)を水中に溶解させ、これにEDC(0.56g、3.4mmol)及びNHS(0.39g、3.4mmol)を添加し、室温で15分間撹拌した。その後、DMF中6−(2−ニトロイミダゾール)ヘキシルアミン(0.18g、0.85mmol)を混合物に添加し、室温で24時間反応させた。DI水及びメタノールの1:1の混合物に対して24時間、DI水に対して48時間透析を完全に実行した。その後、凍結乾燥によってHS−HAを得、1H NMR(Varian Gemini2300)によって特性評価した。紫外可視吸光度によって、グラフト度を20%と決定した。
メタクリル化無水物(MA)と反応させることによって、二重結合でヒアルロン酸(HA)を修飾した。2グラムのHAを100mLの蒸留(DI)水中に低温室内で一晩溶解させ、その後、1.6mLのメタクリル化無水物(MA)を滴加した。5N NaOHを添加し、4℃で24時間連続的に撹拌することによって、反応溶液をpH8〜9に維持した。その後、m−HAをアセトン中に沈殿させ、エタノールで3回洗浄し、その後、DI水中に溶解させた。DI水に対する48時間の透析後、凍結乾燥によって精製されたm−HAを87.5%の収率で得、1H NMR(Varian Gemini2300)によって特性評価した。標準的な逆重畳積分アルゴリズムを実行して、緊密な間隔のピークを分離した後、5.74及び6.17ppm(メタクリル酸プロトン)でのプロトンピーク下面積比を、1.99ppm(HAのN−アセチルグルコサミン)でのピークと比較することによって、修飾度(DM)を約15%であると計算した。
MitoXpress(Cayman Chemical)を製造業者のプロトコルに従って使用することによって、酸素消費速度(OCR)を決定した。簡潔には、10μLのMitoXpressプローブを含有する、0、100、または400mg/dLのグルコースを有する200μLの5mg/mLのGSA溶液を、96ウェルプレート内に置き、380/650nmの励起/発光波長、37℃で、マイクロプレートリーダーでプレートを測定した。30μs及び70μsの遅延時間ならびに30μsのゲート時間で、2つの強度読み取りを取得することによって、各試料ウェルを5分間毎に反復して測定した。各試料ウェルの得られたTR−F強度シグナルを、以下、τ=(70−30)/ln(F1/F2)(式中、F1及びF2は70μs及び30μsの遅延時間でのTR−F強度シグナルである)の通りリン光寿命(μs)τ値に変換した。結果として得られる寿命τの増加は、各試料中の酸素濃度を反映する。
GSAのグルコース応答能力を評価するために、GSAを500μLのPBS緩衝液(NaCl、137mM;KCl、2.7mM;Na2HPO4、10mM;KH2PO4、2mM;pH7.4)中で100μmのNADPH及び5μg/mLのチトクロムcレダクターゼとともにインキュベートした。最初に、α−アミロースをエタノールで洗浄し、その後PBS溶液(10mg/mL)に添加した。様々な量の45%グルコース溶液(Corning)を添加して、様々なグルコース濃度(0mg/dL、100mg/dL、及び400mg/dL)の最終溶液に到達した。質量流量計の制御によって、21%の酸素濃度を有する容器内で、混合物を37℃でインキュベートした。所定の時間間隔で、14,000×gで10分間の遠心分離(10kDaの分子質量カットオフ、Millipore)によって、放出された酵素をGSA懸濁液から分離した。GSA内に被包された残渣酵素の濃度、及びGSAから分離した放出された酵素を、Coomassie Plusタンパク質アッセイを使用して試験した。Infinite200PRO多モードプレートリーダー(Tecan Group Ltd.)でA595を検出し、酵素含有量を酵素溶液の標準曲線で較正した。14,000×gで10分間の遠心分離によって、pH7.4で遠心分離濾過(10kDaの分子質量カットオフ、Millipore)によってグルコースを更に分離した。グルコース(GO)によって、グルコース濃度を決定した。簡潔には、試料を適切に希釈し、その後、37℃でアッセイ試薬を30分間添加した。2.0mLの12N H2SO4を各管に添加することによって、30〜60秒間隔で反応を停止した。試薬ブランクに対して、540nmで吸光度を測定した。標準曲線からグルコース濃度を計算した。GSA溶液の紫外可視吸収をプロットするために、設定時間に330nmのAで強度を測定した。天然AA及びGAならびにGSA(1μm)から放出されたAA及びGAの遠紫外CDスペクトルをCD分光計(Aviv)によって分析した。
Blueacre Technology Ltd.からの10個の均一なシリコン鋳型を使用して、マイクロニードル製作を実行した。レーザーアブレーションによって機械加工された20×20個のマイクロニードル錐体空洞のアレイで、各鋳型を特性評価した。ニードル空洞は、基部で400μmの側辺長さ、800μmの高さ、及び先端部で5μmの側辺長さを有した。鋳型をプラズマ清浄した後、50μLのGSA溶液を鋳型表面上に堆積させた。その後、真空(600mmHg)処理を5分間続けて、GSA溶液がMN空洞内に流動し、望ましい粘度を達成することを可能にした。その後、鋳型を、2000rpmのHettich Universal32R遠心分離機に20分間移して、GSAを先端部領域に圧縮した。GSA層が完全に乾燥した後、0.5mLの10mg/mLのα−アミロース溶液の第2の層を鋳型に供給し、同じプロセスを使用して製作した。その後、MBA(2重量%)及び光開始剤(0.5重量%)と混合した300μLのm−HA溶液(4重量%)を鋳型表面にピペッティングし、その後、真空及び遠心分離の組み合わせを続けた。m−HA層が乾燥し、真空条件下で鋳型空洞から明白な気泡が生じなくなるまで、このプロセスを3〜4回反復した。MNパッチの背部について、4cm×10cmの銀接着テープ片を2cm×2cmの鋳型基部プレートのまわりに適用し、準備した微細鋳型リザーバーに3mLのHA(5重量%)溶液を添加し、25℃で乾燥させた(真空乾燥器内で一晩)。乾燥が完了した後、30秒間の紫外照射(365nmの波長)下、インサイチュ重合を通してマイクロニードルパッチを架橋させた。結果として得られる生成物を慎重に鋳型から分離し、自家製アプリケーターに適合させた。最終MNは、密閉した6ウェル細胞培養プレート内、4℃で1週間貯蔵され得る。
MTS30G引っ張り試験機でニードルをステンレス鋼プレートに押し付けることによって、応力−歪みゲージでMNの機械的強度を測定した。初期ゲージを、10.00Nを細胞充填容量として、MN先端部とステンレス鋼プレートとの間を2.00mmに設定した。MNに向かって移動する上ステンレス鋼プレートの速度を、0.1mm・s−1に設定した。ニードルが曲がり始めるときの、MNの破損力を記録した。
マウス膵島細胞腺腫細胞株6(MIN6)細胞は、Dr.Michael McManus(University of California,San Francisco)が快く提供した。使用した培養培地は、37℃及び5%のCO2の、500mLの培地当たりウシ胎仔血清(15%)、ペニシリン/ストレプトマイシン(1%)、及び2.5uLの島メルカプトエタノール(Biorad)を有するDMEM高グルコース培地であった。培地を3日毎に交換し、細胞を60%の培養密度で継代培養した。MIN6細胞株の第32〜38の継代培養を使用した。
2重量%のアルギン酸の水溶液を12000rpmで遠心分離して、いかなる不純物も除去した。アルギン酸(120mg)溶液を、pH5.0の酢酸緩衝液中1−エチル−3−(3−(ジメチルアミノ)プロピル)カルボジイミド(EDC)/N−ヒドロキシスクシンイミド(NHS)(50mg/30mg)と30分間混合して、アルギン酸上のカルボニル基を活性化させ、その後、追加の1,6−ジアミノヘキサン(60mg)と4時間混合した。混合物を2−プロパノール(IPA)中に沈殿させて、未反応のジアミンを除去した。アルギン酸−アミン誘導体を、重炭酸ナトリウム溶液(50mM、pH=8.5)中、遊離チオールのCys残基を有するCGRGDSのペプチド配列(ペプチド対アルギン酸の重量比は0.02:1であった)と4℃で4時間反応させた。脱イオン水で5日間の広範な透析を介して、ペプチド修飾アルギン酸を精製(3500Daの分子質量カットオフ、Millipore)し、0.22μmのフィルターを通して滅菌し、凍結乾燥させた。MIN6細胞をトリプシン処理し、1%のIV型コラーゲンで補助した2%のアルギン酸−RGD培養培地中(2×106個の細胞/mL)で懸濁させた。鈍い先端部である22ゲージの金属ニードルが結合した1mLのシリンジ内に、混合物を移した。シリンジポンプを備えるエレクトロスプレーシステム内に、シリンジを置いた。エレクトロスプレーシステムの正電極をニードルに接続し、負電極を50mLの20mM BaCl2を有する金属受容容器に接続した。高電位(8kV)下、5cmの受容容器までの作動距離で、溶液を0.155mL/分の流量でスプレーした。液滴(50〜65uL)を押し出した後、内相を滅菌BaCl2(200mM)溶液中で5分間ゼラチン化した。その後、回収したカプセルを滅菌NaCl溶液(150mM)で3回濯いでから、12ウェル培養プレート内のDMEM培地に移した。均一な数の約100個のカプセルを回収し、各実験群のバルクハイドロゲルに更に導入して、処理間の比較を可能にした。1分間の紫外照射(波長:365nm)を介して、DMEM(2%、重量%、DMEM)中m−HAとN,N’−メチレンビスアクリルアミド(MBA2%、重量%)及び光開始剤(Irgacure2959、0.05%、重量%)とを光重合することによって、HAハイドロゲルを架橋させた(図15Aおよび図15B)。
細胞クラスターの形態的特徴を分析するために、Olympus IX70多パラメータ顕微鏡下で膵臓β細胞カプセルを観察した。膵臓β細胞カプセルの平均直径を楕円に適合させ、ImageJソフトウェアの粒子分析法を使用して画像を調節することによって分析した。
SEMを使用して、MNパッチの形態を特性評価した。両面粘着炭素タブを使用して、マイクロニードルをそれらの基部とともにSEM試料スタブに結合した。EmTech Turbo EMスパッタコーターを使用して、試料に7nmの厚さの金−パラジウム層をコーティングした。Analytical Instrumentation Facility,North Carolina State Universityで、FEI Verios460L電界放出走査電子顕微鏡(FESEM)で撮像を実行した。カルシウムAM染色した膵臓β細胞カプセルを、ローダミン標識GSAを充填したMNパッチの背部に位置付けた。Olympus IX70多パラメータ蛍光顕微鏡によって、L−S GRSの側面図から蛍光画像を取得した。
静的グルコース刺激インスリン分泌(GSIS)アッセイを介して、被包された細胞の機能性評価を試験した。実験の2〜3日前、96ウェルプレート当たり2×105個のMIN6細胞を播種した(Corning Costar)。2D細胞培養と同様に、グルコース処理の1日前、3Dカプセル内の同じ量の細胞を96ウェルプレート内で培養した(10個/各ウェル)。0.1%のBSAで補助したクレブス・リンゲル(KRB)緩衝液(128mmのNaCl、4.8mmのKCl、1.2mmのKH2PO4、1.2mmのMgSO4、2.5mmのCaCl2、10mmのHEPES、0.1重量容量%)を事前に調製した。37℃及び5%のCO2のKRB緩衝液BSA中で、細胞を2時間インキュベートし、その後、同じ条件で100mg/dLまたは400mg/dLのグルコースとともに45分間インキュベートした。マウスインスリンELISAキット(Alpco.)を使用して、細胞によって分泌されたインスリンの量を定量化した。KRBを含有する400mg/dLのグルコース中のインスリン分泌の量を、KRBを含有する100mg/dLのグルコース中で分泌されたインスリンに対して正規化し、インスリン分泌インデックスとして表現した。
被包の6時間後、細胞を4℃で4%のパラホルムアルデヒド中に固定し、その後、OCT化合物(Sakura Finetek)中に包埋させ、ドライアイス上、イソペンタン浴内で急速凍結させた。凍結した細胞カプセルを切片化(5μmの厚さ)し、顕微鏡スライド上に載せ、−80℃で貯蔵した。インスリンを免疫蛍光で染色するために、スライドを2回洗浄し、0.1%のTriton X100溶液を使用して30分間透過処理し、その後、1%のウシ血清アルブミン(BSA)溶液を使用して1時間遮断した。遮断後、1/200の希釈度のインスリン抗体に対してモノクローナルである一次ウサギ(abcam181547)を一晩4℃で適用し、その後、洗浄し、1/400の希釈度の抗体ヤギ抗ウサギIgG(Alexa Fluor(登録商標)488)(abcam150077)とともにインキュベートした(緑色)。スライドを3回洗浄し、細胞透過性染料DAPIを適用して細胞核を染色し、カバーガラスで被覆した。Olympus IX70多パラメータ蛍光顕微鏡を使用して試料を撮像し、ImageJソフトウェアを使用して加工した。
血液循環系のラボオンチップ模擬実験として、マイクロ流体技術デバイスを使用して、MNからのインスリンの放出を実行した。グルコース応答能力を動的に評価するために、放出培地がニードル先端部を通して流動する間(様々なグルコース濃度を有するpH7.4のKRB)、MNパッチをマイクロ流体チャネルの開いた中心部に置いた。2つの別個のシリンジポンプ(Harvard Apparatus PHD2000、Holliston,MA)で、注入及び撤去速度を50μL/分に設定した。マウスインスリンELISAキット(Alpco.)を使用して、インスリン放出速度を定量化した。Infinite200PRO多モードプレートリーダー(Tecan Group Ltd.,Switzerland)で、450nmでのインスリン含有量を測定し、インスリン標準曲線で較正した。
糖尿病治療のためのMNパッチのインビボ有効性を、STZ誘導成体糖尿病マウス(雄C57B6、Jackson Lab,U.S.A.)で評価した。動物研究プロトコルは、Institutional Animal Care and Use Committee at North Carolina State and University and University of North Carolina at Chapel Hillによって承認された。Clarity GL2Plusグルコースメータ(Clarity Diagnostics,Boca Raton,Florida)を使用して、血漿当量グルコースをマウスの尾静脈血液試料(約3μL)から測定した。それらのBGLを投与前の2日間監視し、投与前に全てのマウスを一晩絶食させた。各群の5匹のマウスを、GRSを有しない空MNを含有するMN(GRSなし)、L−GRSとのみ統合されたMN(L−GRS)、S−GRSとのみ統合されたMN(S−GRS)、L−S−GRSと統合されたMN(L−S GRS)、L−S−GRSと統合されているが、S−GRS中にGOxを有しないMN(L−S GRS(GOxなし))、及びL−S−GRSと統合されているが、S−GRS中にα−アミロースを有しないMN(L−S GRS(AMなし))で皮下治療するために選択した。自家製アプリケーターによって、5N/パッチで10分間、MNパッチを背側皮膚に適用した。皮膚貼付外科接着剤を使用して、持続した放出のためにパッチを皮膚に固定した。12mm×12mm×5mmのカスタマイズPDMS鋳型を、パッチの上に被覆した。PDMS鋳型の内側では、膵臓β細胞カプセルは、栄養素補給のために、DMEMから作製された架橋m−HAハイドロゲル中に包埋された。MN(S−GRS)群、MN(GRSなし)群について、細胞はデバイス内に組み込まれた。その後、各群の投与されたマウスのBGLを連続的に(5、15、30、及び60分間で、及びその後1時間に1回)監視した。グルコース負荷試験を実行して、MNの投与の2時間後のMNのインビボでのグルコース応答性を確認した。簡潔には、マウスを一晩絶食させ、L−S GRSを投与した。投与の2時間後、マウスのグルコースレベルが最低値に到達したとき、腹腔内注射を介して1.5gのグルコース/kg(45%の滅菌グルコース溶液、corning cellgro)をマウスに与えた。健康なマウスに対するグルコース負荷試験を、対照として実行した。グルコース投与の0、10、20、30、40、60、90、及び120分間後に、尾静脈から採血し、血糖計を使用してグルコースレベルを測定した。同様に、低血糖のリスクを評価するために、2つの群の健康なマウスにMN(L−S GRS)またはMN(GRSなし)を投与したが、グルコース負荷には供与しなかった。
比色分析メチルチアゾリルテトラゾリウム(MTT)アッセイを使用して、細胞増殖分析を実行した。MIN6細胞を、1ウェル当たり5,000個の細胞の密度で96ウェルプレートに播種し、100μLのDMEM中で培養した。その後、プレートを5%のCO2中37℃で12時間インキュベートして、70〜80%の培養密度に到達してから、溶解したMN溶液の連続希釈を添加した。24時間のインキュベーション後、細胞をKRB溶液で洗浄し、100μLの新鮮なFBSを含まないDMEM及び新たに調製した20μLの3−(4,5−ジメチルチアゾール−2−イル)−2,5−ジフェニルテトラゾリウムブロミド溶液(MTT溶液、5mg/mL)とともにインキュベートした。プレートを更に4時間インキュベートした。その後、培養培地を慎重に除去し、その後、150μLのジメチルスルホキシド(DMSO)を添加した。マイクロプレートリーダー(Infinite M200Pro、Tecan,Morrisville,NC,USA)を使用して、10分間以内に、590nm、及び620nmの参照波長でプレートの吸光度を読み取った。
データは、平均±標準偏差として示される。スチューデントt検定またはANOVA検定を使用して、統計学的分析を実行した。P≦0.05で、実験群間と対照群との差は、統計学的に有意であると見なされた。

Claims (82)

前記マイクロニードルアレイの前記基端部と接続されている少なくとも1つのリザーバーであって、前記リザーバーが、薬剤送達システムを備え、前記薬剤送達システムが、前記生物学的障壁をわたって輸送される薬剤、または前記生物学的障壁をわたって輸送される薬剤を生成するための手段、及びレシピエントからの生理学的シグナルを検出するための手段を含む、前記リザーバーと、
シグナル増幅器システムであって、前記レシピエントからの前記生理学的シグナルを増幅することができる構成要素を備える、前記シグナル増幅器システムと、を備える、前記デバイス。 - 前記シグナル増幅器システムが、前記マイクロニードル内に統合される、請求項1に記載のデバイス。
- 前記生物学的障壁をわたって輸送される前記薬剤の体積または量が、前記生理学的シグナルに基づいて変更され得るように、前記薬剤送達システムが、フィードバック構成要素を備える、請求項1または2に記載のデバイス。
- 前記レシピエントからの前記生理学的シグナルが、前記シグナル増幅器システムからの増幅なしでは前記フィードバック構成要素によって検出されるのに不十分である、請求項3に記載のデバイス。
- 前記生物学的障壁リザーバーをわたって輸送される薬剤を生成するための前記手段が、細胞を含む、請求項1〜4のいずれか1項に記載のデバイス。

前記マイクロニードルアレイの前記基端部と接続されている少なくとも1つのリザーバーであって、前記リザーバーが、薬剤送達システムを備え、前記薬剤送達システムが、前記生物学的障壁をわたって輸送される薬剤、または前記生物学的障壁をわたって輸送される薬剤を生成するための手段、及び生理学的シグナルを検出するための手段を含み、前記生物学的障壁をわたって輸送される前記薬剤の体積または量が、前記生理学的シグナルに基づいて変更され得るように、前記薬剤送達システムが、フィードバック構成要素を備える、前記リザーバーと、を備える、前記デバイス。 - 前記生物学的障壁リザーバーをわたって輸送される薬剤を生成するための前記手段が、細胞を含む、請求項19に記載のデバイス。
- 前記細胞が、ヒト細胞である、請求項20に記載のデバイス。
- 前記細胞が、膵臓β細胞または幹細胞分化ヒト膵臓細胞である、請求項19〜21のいずれか1項に記載のデバイス。

a)マイクロニードルパッチを前記対象に提供することであって、前記マイクロニードルパッチが、
i)各々が基端部及び先端部を有する複数のマイクロニードルと、
ii)前記マイクロニードルの前記基端部が結合または統合される基板と、
iii)前記マイクロニードルアレイの前記基端部と接続されている少なくとも1つのリザーバーであって、前記リザーバーが、薬剤送達システムを備え、前記薬剤送達システムが、生物学的障壁をわたって輸送される薬剤、または前記生物学的障壁をわたって輸送される薬剤を生成するための手段、及びレシピエントからの生理学的シグナルを検出するための手段を含む、前記リザーバーと、
iv)シグナル増幅器システムであって、前記レシピエントからの前記生理学的シグナルを増幅することができる構成要素を備える、前記シグナル増幅器システムと、を備える、前記提供することと、
b)前記マイクロニードルを前記生物学的障壁内に挿入することと、
c)前記薬剤を、前記マイクロニードルを通して前記生物学的障壁内に送達することであって、送達される薬剤の量または体積が、前記シグナル増幅器システムから受容されるシグナルによって決定される、前記送達することと、を含む、前記方法。 - 前記生物学的障壁リザーバーをわたって輸送される材料を生成するための前記手段が、細胞を含む、請求項31に記載の方法。
- 前記細胞が、膵臓β細胞または幹細胞分化ヒト膵臓細胞である、請求項32に記載の方法。
- 前記リザーバーが、細胞生成物を送達するのに使用され得る材料マトリックスで構成される、請求項31に記載の方法。

- 治療を必要とする対象における疾患を治療する方法であって、
a)マイクロニードルパッチを前記対象に提供することであって、前記マイクロニードルパッチが、
i)各々が基端部及び先端部を有する複数のマイクロニードルと、
ii)前記マイクロニードルの前記基端部が結合または統合される基板と、
iii)前記マイクロニードルアレイの前記基端部と接続されている少なくとも1つのリザーバーであって、前記リザーバーが、薬剤送達システムを備え、前記薬剤送達システムが、生物学的障壁をわたって輸送される薬剤、または前記生物学的障壁をわたって輸送される薬剤を生成するための手段、及び生理学的シグナルを検出するための手段を含み、前記生物学的障壁をわたって輸送される前記薬剤の体積または量が、前記生理学的シグナルに基づいて変更され得るように、前記薬剤送達システムが、フィードバック構成要素を備える、前記リザーバーと、を備える、前記提供することと、
b)前記マイクロニードルを前記生物学的障壁内に挿入することと、
c)前記薬剤を、前記マイクロニードルを通して前記生物学的障壁内に送達することであって、送達される薬剤の前記量または体積が、前記生理学的シグナルに基づく、前記送達することと、を含む、前記方法。 - 前記生物学的障壁リザーバーをわたって輸送される材料を生成するための前記手段が、細胞を含む、請求項50に記載の方法。
- 前記細胞が、膵臓β細胞または幹細胞分化ヒト膵臓細胞である、請求項51に記載の方法。
- 前記リザーバーが、細胞生成物を送達するのに使用され得る材料マトリックスで構成される、請求項52に記載の方法。
